Abstract: International audience; The induction of plant defense mechanisms by resistance inducers is an attractive and innovative alternative to reduce the use of fungicides on wheat against Zymoseptoria tritici, the responsible agent of Septoria tritici blotch (STB). Under controlled conditions, we investigated the resistance induction in three wheat cultivars with different susceptible levels to STB as a response to a treatment with a sulfur, manganese sulfate, and protein-based resistance inducer (NECTAR Céréales). While no direct antigermination effect of the product was observed in planta, more than 50% reduction of both symptoms and sporulation were recorded on the three tested cultivars. However, an impact of the wheat genotype on resistance induction was highlighted, which affects host penetration, cell colonization, and the production of cell-wall degrading enzymes by the fungus. Moreover, in the most susceptible cultivar Alixan, the product upregulated POX2, PAL, PR1, and GLUC gene expression in both noninoculated and inoculated plants and CHIT2 in noninoculated plants only. In contrast, defense responses induced in Altigo, the most resistant cultivar, seem to be more specifically mediated by the phenylpropanoid pathway in noninoculated as well as inoculated plants, since PAL and CHS were most specifically upregulated in this cultivar. In Premio, the moderate resistant cultivar, NECTAR Céréales elicits mainly the octadecanoid pathway, via LOX and AOS induction in noninoculated plants. We concluded that this complex resistance-inducing product protects wheat against Z. tritici by stimulating the cultivar-dependent plant defense mechanisms.

Abstract: International audience; One way of exploring knowledge representation is to compare expert and non-expert strategies. Expert knowledge representation seems to be acquired through both exposure and intensive formal training, leading to the development of skills and conceptual knowledge related to a specific field. The objective of our study was to dissociate the effects of familiarity and expertise on the development of knowledge representations about wine. Three panels were therefore recruited to study the conceptual representations of Beaujolais red wines among wine,experts and consumers. One panel was composed of unfamiliar consumers from a region without wine production, while the other two were familiar consumers, or experts, both from the Beaujolais region. A free hierarchical sorting task with wine labels was followed by a semi-directed interview. The results of the hierarchical sorting task showed that the three panels formed common groups that were mostly related to the Beaujolais wine taxonomy-based system. However, the interview results regarding criteria and strategies showed that the three panels had very different knowledge representations of Beaujolais wines. Although experts and familiar consumers used top-down strategies based on knowledge, their wine knowledge representations differ. For experts, knowledge representation is flexible and leads to different categorization schemes, while for familiar consumers, knowledge representation is rigid and leads to a single categorization scheme based on the Beaujolais taxonomy. Unfamiliar consumers have fragmented knowledge, leading to the use of bottom-up strategies to compensate for their lack of exposure to wine. This strategy leads to a misrepresentation of Beaujolais wines.

Abstract: International audience; Two kitchen garden soils (A and B) sampled in contaminated areas were amended using phosphates in sustainable quantities in order to reduce the environmental availability of potentially toxic inorganic elements (PTEs) and to favour the availability of alkali, alkali earth and micronutrients. The environmental availability of PTEs was evaluated using a potential plant for revegetation of contaminated soils (ryegrass) and a mixture of low molecular weight organic acids. Despite the highest contamination level of B, the concentration of metals was highest in the ryegrass shoots grown on A for the two harvests. These results correlated well with those obtained using low molecular weight organic acids for Cd, Zn and Cu, whereas this mixture failed to represent the transfer of nutrients due to the presence of biological and physiological mechanisms. The statistical differences between the biomass of ryegrass obtained at the first and the second harvests were attributed to the decrease of available potassium, implicated in the growth and development of plants. Phosphates increased the ratios Zn/Cd, Zn/Pb and Zn/Cu up to 176 ± 48, 38 ± 6 and 80 ± 12, respectively, and made possible the reduction of the concentration of Cd and Pb in the shoots of ryegrass by 22% and 25%, respectively. The concentration of Zn in the shoots of ryegrass from the first and the second harvests grown on soil A were in the range 1050–2000 mg kg−1, making this plant a potential biomass to (i) produce biosourced catalysts for organic chemistry applications in a circular economy concept and (ii) limit human exposure to commercial Lewis acids. Abstract: International audience; Two smelters in the North of France emitted potentially toxic metals for more than a century and today, the resulting contamination represents a risk to human health and affects also the biodiversity. To limit health risks and to improve the soil quality, a study using calcium phosphates (monocalcium phosphate, dicalcium phosphate and a mixture of both salts) and Lolium perenne L was conducted. Through this preliminary investigation, we will try to shed some light about (i) the effects of a sustainable amount of calcium phosphates on the agronomic, biological (microbial and fungi communities) and physiological parameters (chlorophyll a and b, antocyanins, carotenoids) as well as the phytoavailability of potentially toxic metals and nutrients in time, and (ii) the potential use of contaminated biomass from ryegrass as a source of new valorisation ways instead of using it as contaminated compost by gardeners. Although slight variations in pH and significant increases of assimilable phosphorus after adding calcium phosphates were registered, the physiology of plants and the biological parameters were statistically unchanged. The germination of the ryegrass seeds was favoured with calcium phosphates regardless the contamination level of the studied soils. No clear effects of calcium phosphates on the microbial and fungi communities were detected. In contrast, results indicated relationships between the physicochemical parameters of soils, their contamination level and the composition of fungal communities. Indeed, for one of the soils studied, calcium could limit the transport of nutrients, causing an increase in fungi to promote again the transfer of nutrients. Surprisingly, the phytoavailability of Pb increased in the most contaminated soil after adding dicalcium phosphate and the mixture of phosphates whereas a slight decrease was highlighted for Cd and Mn. Although minor changes in the phytoavailability of potentially toxic metals were obtained using calcium phosphates, the ability of ryegrass to accumulate Zn and Ca (up to 600 and 20,000 mg kg−1, respectively) make possible to qualify this plant as a bio ‘ore’ resource. © 2019, Springer Nature Switzerland AG.

Abstract: International audience; Aided phytoremediation was studied for 48 weeks with the aim of reducing extractable and phytoavailable toxic elements and producing potential marketable biomass. In this sense, biomass of ryegrass was produced under greenhouse on two contaminated garden soils that have been amended with two successive additions of phosphates. After the first addition of phosphates, seeds of ryegrass were sown and shoots were harvested twice. A second seedling was performed after carefully mixing the roots from the first production (used as compost), soils and phosphates. Forty-eight weeks after starting the experiments, the concentrations of Cd, Pb, Zn, Cu, Fe, and Mn extracted using the rhizosphere-based method were generally lower than those measured before the addition of phosphates and cultivation (except for Pb and Fe in the most contaminated soil). The concentrations of metals in the shoots of ryegrass from the second production were lower than those from the first (except for Al). The best results were obtained with phosphates and were the most relevant in the lowest contaminated soil, demonstrating that the available metal concentrations have to be taken into account in the management of contaminated soils. In view of the concentration of metals defined as carcinogens, mutagens, and reprotoxics (e.g., Cd, Pb) and those capable to be transformed into Lewis acids (e.g., Zn, Fe), the utilization of ryegrass in the revegetation of contaminated soils and in risk management may be a new production of marketable biomass. The development of phytomanagement in combination with this type of biomass coincided with the view that contaminated soils can still represent a valuable resource that should be used sustainably. © 2019, Springer-Verlag GmbH Germany, part of Springer Nature.

Unmanned aerial systems (UASs) are interesting versatile tools that can provide relevant 3D information, such as sward height (3D structure), or even describe the physical condition of pastures through the use of spectral information. This study aimed to evaluate the potential of UAS to characterize a pasture's sward height and above-ground biomass at a very ﬁne spatial scale. The pasture height provided by UAS products showed good agreement (R2 = 0.62) with a reference terrestrial light detection and ranging (LiDAR) dataset. We tested the ability of UAS imagery to model pasture biomass based on three different combinations: UAS sward height, UAS sward multispectral reﬂectance/vegetation indices, and a combination of both UAS data types. The mixed approach combining the UAS sward height and spectral data performed the best (adj. R2 = 0.49). This approach reached a quality comparable to that of more conventional non-destructive on-ﬁeld pasture biomass monitoring tools. As all of the UAS variables used in the model ﬁtting processwereextractedfromspatialinformation(rasterdata),ahighspatialresolutionmapofpasture biomass was derived based on the best ﬁtted model. A sward height differences map was also derived from UAS-based sward height maps before and after grazing. Our results demonstrate the potential of UAS imagery as a tool for precision grazing study applications. The UAS approach to height and biomass monitoring was revealed to be a potential alternative to the widely used but time-consumingﬁeld approaches. While reachinga similarlevel ofaccuracy to theconventional ﬁeld sampling approach, the UAS approach provides wall-to-wall pasture characterization through very high spatial resolution maps, opening up a new area of research for precision grazing.

Abstract: Members of the genus Burkholderia colonize diverse ecological niches. Among the plant-associated strains, Paraburkholderia phytofirmans PsJN is an endophyte with a broad host range. In spatially structured environment (unshaken broth cultures), biofilm-constructing specialists of P. phytofirmans PsJN colonizing the air-liquid interface arose at high frequency. In addition to forming a robust biofilm in vitro and in planta on Arabidopsis roots, those mucoid phenotypic variants display a reduced swimming ability and modulate the expression of several Microbe Associated Molecular Pattern (MAMPs) including exopolysaccharides (EPS), flagellin and GroEL. Interestingly, the variants induce a low PR1 and PDF1.2 expression compared to the parental strain, suggesting a possible evasion of plant host immunity. We further demonstrated that switching from the planktonic to the sessile form did not involve quorum-sensing genes but arose from spontaneous mutations in two genes belonging to an Iron-Sulfur Cluster: hscA (encoding a co-chaperone protein) and iscS (encoding a cysteine desulfurase). A mutational approach validated the implication of these two genes in the appearance of variants. We showed for the first time that in heterogeneous environment, P. phytofirmans strain PsJN is able to rapidly diversify and co-express a variant that outcompete the wild-type form in free-living and static conditions but not in planta . IMPORTANCE Paraburkholderia phytofirmans strain PsJN is a well-studied plant-associated bacterium known to induce resistance against biotic and abiotic stresses. In this work, we described the spontaneaous appearance of mucoid variants in PsJN from static cultures. We showed that the conversion from wild-type (WT) form to variants (V) correlates with an overproduction of EPS, an enhanced ability to form biofilm in vitro and in planta and a reduced swimming motility. Our results revealed also that these phenotypes are in part associated with spontaneaous mutations in an Iron-Sulfur Cluster. Overall, the data provided here allowed a better understanding of the adaptive mechanisms likely developed by P. phytofirmans PsJN in heterogeneous environment.

Abstract: Concentrating diluted samples is a key step to improve detection capabilities. The wise use of scaling laws shows the advantages of working with sub-microliter-sized samples. Rapid progress in MEMS technologies has driven the design of integrated platforms performing many biochemical operations. Here we report a new concentrator device based on electro-hydrodynamic forces which can be easily integrated into electrowetting-on-dielectric (EWOD) platforms. This approach is label-free and applicable to a wide range of micro-objects. The detection and analysis of two common waterborne parasites, Cryptosporidium and Giardia, is a perfect test case due to their global health relevance. By fully controlling the interplay of the various forces acting on the micron-sized Cryptosporidium parvum and Cryptosporidium muris oocysts, we show that it is possible to concentrate them on the side of a 10 μL initial drop and then extract them efficiently from a droplet of a few hundred nanoliters. We performed a finite element modeling of the forces acting on the parasites' oocysts to optimize the electrodes' shapes. We obtained state-of-the-art concentration factors of 12 ± 0.4 times and 2 to 4 times in the sub-region of the drop and the extracted droplet, respectively, with an efficiency of 70 ± 6%. Furthermore, this device had the ability to selectively concentrate parasites of different species out of a mix. We demonstrated this by segregating C. parvum oocysts from either Giardia lamblia cysts or its related species, C. muris oocysts.

Abstract: Ce projet s’inscrit dans le contexte de recherche de moyens plus durables de protection des cultures. Cela concerne l’agriculture conventionnelle mais également l’agriculture biologique pour lesquelles le remplacement du cuivre et la recherche d’alternatives efficaces aux produits phytosanitaires de synthèse constitue un important challenge. L’objectif principal est d’évaluer l’intérêt d’huiles essentielles dans des stratégies de protection des cultures sur 4 principales maladies des cultures (mildious de la pomme de terre, de la vigne et de la salade, tavelure du pommier) en s’appuyant à la fois sur les connaissances empiriques existantes et sur des recherches scientifiques en laboratoire (in vitro et in vivo) et au champ. Les résultats in vitro montrent sans ambiguïté que toutes les HE testées possèdent une activité fongicide. Cependant, en condition de terrain (sous abri ou en plein champ), le projet n’a pas permis de mettre en évidence des efficacités significatives à 0.2% (v/v) sur les champignons cibles. Des tendances peuvent cependant être rapportées. En outre, vis-à-vis des insectes auxiliaires des cultures, à la concentration de 0.2% dans le sirop de nourrissage des abeilles, aucun effet toxique significativement supérieur à la mortalité naturelle n’a été remarqué. La mortalité sur typhlodrome est quant à elle augmentée par les huiles essentielles. Une évaluation ad hoc de l’impact de ces substances sur les différents compartiments écologiques semble donc nécessaire pour comprendre l’impact des HE sur les productions végétales et leur environnement, This project seeks for sustainable plant protection product based on essential oils for conventional and organic agriculture. It must assess efficacy of 7 essential oils chosen from literature for plant protection against 4 fungal diseases: potato blight, powdery mildew of grape, downy mildew of salad and apple scab. The assessment is based on in vitro and in vivo approaches before field trials. Only pure culture fungi i.e. Phytophthora infestans and Venturia inaequalis could be tested under in vitro conditions.Results showed unambiguous efficacy of all tested essential oils on fungal development in laboratory (Phytophthora infestans and Venturia inaequalis). However, on field trials, at 0,2% v/v, essential oils did not show statistical efficiency but some trends can be exploited. In addition, concerning ecotoxycology, when bees were fed with sugar mixture containing 0,2% of essential oil, no extra mortality beyond natural one has been reported. However, typhlodromus were more impacted by these substances. To conclude, methodology must be developed to include ecological compartments in the scheme of assessment of essential oil on the whole production system.

Abstract: International audience; NonRibosomal peptides (NRPs) are a huge untapped resource of natural products displaying activities with applications in health (i.e. antibiotics) or in biocontrol (especially siderophores and lipopeptides with antifungal activity). NRPs are microbial secondary metabolites produced by enzymatic complexes, so-called non-ribosomal peptide synthetases (NRPSs). These modular assembly lines work step by step to build the peptides, each module adding one monomer to the peptidic chain. Considering the modular organization of NRPSs, and the structural specific features of the NRPs, dedicated bioinformatics tools have been developed with the aim of accelerating the screening for new active metabolites.

Abstract: International audience; Lactococcus lactis is a lactic acid bacterium widely used in cheese and fermented milk production. During fermentation, L. lactis is subjected to acid stress that impairs its growth. The small heat shock protein (sHsp) Lo18 from the acidophilic species Oenococcus oeni was expressed in L. lactis. This sHsp is known to play an important role in protein protection and membrane stabilization in O. oeni. The role of this sHsp could be studied in L. lactis, since no gene encoding for sHsp has been detected in this species. L. lactis subsp. cremoris strain MG1363 was transformed with the pDLlisp18 plasmid, which is derived from pDL278 and contains the hsp18 gene (encoding Lo18) and its own promoter sequence. The production of Lo18 during stress conditions was checked by immunoblotting and the cellular distribution of Lo18 in L. lactis cells after heat shock was determined. Our results clearly indicated a role for Lo18 in cytoplasmic protein protection and membrane stabilization during stress. The production of sHsp in L. lactis improved tolerance to heat and acid conditions in this species. Finally, the improvement of the L. lactis survival in milk medium thanks to Lo18 was highlighted, suggesting an interesting role of this sHsp. These findings suggest that the expression of a sHsp by a L. lactis strain results in greater resistance to stress, and, can consequently enhance the performances of industrial strains.

Abstract: Why do people eat and drink as they do? This complex question is of great interest to both the academic and industrial world. In recent years, a change of consumption has been noticed in several countries with an increase in interest for traditional and craft products. This is the case, for example, for the beer sector, going from a beer category dominated by one style to a wider range of options of industrial beers and to less commercial options such as craft beers. A change in patterns of consumption is usually an indicator that the perception towards the product is changing. In this study, the objective is to understand the impact of gender (men versus women) and type of consumption (craft versus industrial beer) on mental beer representations. Four groups of participants were asked to visually sort a set of beer in the presence of brand and packaging. The results show both similarities and differences in the categorization made by each group of consumers. Overall, participants agreed more on their categorization of industrial beers than they did with craft beers. Gender differences were perceived in the sorting task especially in terms of the number of groups used to sort beers, more groups in men; but also in the words used to describe beers. When comparing the results across women and men, it was seen that the latter sort the beers based on previous knowledge (cognitive dimension) while women rely more on the affective dimension (like – do not like) to sort the beers. An interaction effect was also found between gender and type of consumption which highlights the complex relationship that consumers have towards beers.

Abstract: Product experience is shaped by the interaction between the human systems and the product. Human systems include a sensory system to perceive the surrounding world, an affective system that evokes emotional responses to certain stimuli, and a cognitive system that makes meaning and processes information. We hypothesise that experience is a combination between these three systems rather than a linear continuum of hedonic reactions. In order to test this hypothesis, we conducted a study measuring the experience of drinking craft and industrial beers. A total of 400 consumers were invited to drink beer, rate their liking and select a set of phrases that better described their drinking experience. Results showed no significant difference in expected liking and purchase intention between the eight beers evaluated. However, a difference between beers was observed for the CATA phrases. Cognitive phrases were more frequently checked for craft beers, while sensory, and affective phrases were more frequently checked for industrial beers. A Multiple Factor Analysis for Contingency Tables showed that the sensory and cognitive systems were more related to liking than the affective system.

Abstract: International audience; The Pivot Profile (PP) was proposed recently by Thuillier, Valentin, Marchal, and Dacremont (2015) as an alternative to classical descriptive methods. Its principle is to describe each product by comparing it to a stable reference (called Pivot). While the method seems promising there is little data available and some issues still need to be examined. This paper proposes to evaluate two of these issues: the effects of the similarity within the product space and of the choice of pivot. We compared the pivot profiles obtained for three different sets of beers, varying in their within-set sensory similarity, using different pivots. We found that PP results are more influenced by the within-set similarity than by the choice of the pivot. We suggest that the PP method is more suitable for restricted product spaces in terms of qensory characteristics, and that the creation of a "central product" as the pivot can be a good option when the type of products allows it. However, further studies need to be conducted to assess PP in terms of assessors' repeatability and consensus, as well as to propose alternative statistical analyses that would take into account the individual PP data.

Abstract: In today's market, every product seems to be marked by the label of "experience". It is expected that successful products give the consumer "extraordinary experiences". The research in consumption experience is growing, but much work still needs to be done to understand the food and beverage experience. A qualitative study was conducted using contextual focus groups to explore the building blocks of consumers' drinking experience, of industrial and craft beers. The results show that drinking experience is shaped by our cognitive, sensory or affective systems, especially during the core consumption experience. Elements such as attitudes, consumption habits, and individual versus social consumption, shopping experience and product benefits are also responsible for shaping the experience, but are more relevant during the pre-consumption or post-consumption experience. Gender differences occur more frequently in the affective experience, as women search more for relaxation while men for excitement and stimulation while drinking beer. When comparing industrial users versus craft, in the latter the cognitive and shopping experiences are more relevant. Overall, the results showed that the drinking experience of beers can be studied as a function of the salient human system used during product interaction, and this systems act as the building blocks of the drinking experience of beer. This information can be applied in consumer research studies to further study the experiential differences across products and consumers.

Abstract: International audience; Consumption of craft beer has been changing worldwide; it passed from a rarely known to a common product amongst consumers. In the past years craft beer sales have been growing faster than the dominant lager-style in countries with different beer tradition like Mexico and France. A change in consumption habits is a sign that the representation of the product is changing. It is, therefore, valuable to understand the elements and structure of this new representation. The present study was conducted to understand the impact of culture and consumption habits on beer representation. Three-hundred male consumers (craft and industrial beer users) were interviewed in Mexico and France. Participants were asked to perform: (1) free word association task using "craft beer" as inductor word, (2) ranking task of the evoked words based on their importance, and (3) valence rating task of each evoked word. Each word was characterized by a mean importance value and a frequency of elicitation to identify their role as central or peripheral elements in the representation. Results showed that consumers from same culture share similar social representations of craft beer, even when they have different consumption habits (craft-industrial). Mexican consumers share alcohol, and flavour as central elements in their representations but share only one peripheral element "tradition". French consumers share two central elements taste and party. When comparing consumers across cultures, French and Mexican craft participants share no central elements in their representations towards beer; whereas industrial beer consumers share one element "alcohol". Craft consumers seem to have a more structured social representation, sustained or nourished on consumption habits, whilst the representation of industrial beer consumers seems less resistant, or capable of changing over time. Abstract: Polyphenols are secondary metabolites of plants and are broadly distributed in fruits and vegetables. It is demonstrated that these compounds act as strong antioxidants which determines a variety of their medicinal properties. Wastes of black chokeberry after juice recovery were used to investigate the optimal conditions for maximal yield of polyphenols applying solid-liquid extraction. Parameters such as temperature, medium acidity and type of solvent were varied. Total polyphenol content was assayed according to the Folin-Ciocalteu colorimetric reaction method. The results obtained demonstrated that water and water-ethanol mixtures were suitable for polyphenols extraction from black chokeberry wastes. The increase in temperature, regardless of the solvent, led to increasing polyphenol content in the extracts, reaching 7700 mg/L GAE with 50 % ethanol at ebullition, while acidity had clearly less effect on polyphenols extraction.

Abstract: Escherichia coli strains from swine origin, either susceptible or resistant to colistin, were grown under planktonic and biofilm cultures. After which, they were treated with antibacterial agents including nisin and enterocin DD14 bacteriocins, colistin and their combinations. Importantly, the combination of colistin, enterocin DD14 and nisin eradicated the planktonic and biofilm cultures of E. coli CIP54127 and the E. coli strains with colistin-resistance phenotype such as E. coli 184 (mcr-1(+)) and E. coli 289 (mcr-1(-)), suggesting therefore that bacteriocins from lactic acid bacteria could be used as agents with antibiotic augmentation capability.

Abstract: Based on the exploration of data generated by genome sequencing, a bioinformatics approach has been chosen to identify the biosynthetic pathway of the siderophores produced by Aeromonas species. The amonabactins, considered as a virulence factor, represent a family of four variants of catechol peptidic siderophores containing Dhb, Lys, Gly, and an aromatic residue either Trp or Phe in a D-configuration. The synthesis operon is constituted of seven genes named amoCEBFAGH and is iron-regulated. The cluster includes genes encoding proteins involved in the synthesis and incorporation of the Dhb monomer, and genes encoding specific nonribosomal peptide synthetases, which are responsible for the building of the peptidic moiety. The amonabactin assembly line displays a still so far not described atypical mode of synthesis that is iterative, alternative, and optional. A disruption mutant in the adenylation domain of AmoG was unable to synthesize any amonabactin and to grow in iron stress conditions while a deletion of amoH resulted in the production of only two over the four forms. The amo cluster is widespread among most of the Aeromonas species, only few species produces the enterobactin siderophore.

Abstract: SPE IPM UBSPEIPMUB; Downy mildew, caused by Plasmopara viticola, is one of the most important grape pathogen in Europe and North America. Although the control is traditionally performed with fungicides, the appearance of resistant pathogen populations and the possible adverse effects on human and environment health are spurring the search for alternative means. In the present investigation, two protein hydrolysates of soybean (soy) and casein (cas) origin were successfully tested against P. viticola. On Vitis vinifera cv. Marselan plants, the application of soy and cas reduced the infected leaf surface by 76 and 63%, as compared to the untreated control, respectively. Since both hydrolysates seemed to trigger the plant immunity, we investigated their effect on selected grapevine defense responses. On treated grapevine cell suspensions, a different free cytosolic calcium signature was recorded for each hydrolysate, whereas a similar transient phosphorylation of two MAP kinases of 45 and 49 kDa was observed. These signalling events were followed by transcriptome reprogramming, including the up-regulation of genes encoding pathogenesis-related (PR) proteins and the enzyme stilbene synthase responsible for the biosynthesis of resveratrol, the main grapevine phytoalexin. Liquid chromatography analyses confirmed the production of resveratrol and its dimer metabolites, δ- and ε-viniferins. Overall, soy effect was more pronounced than cas one. Both hydrolysates proved to be able to enhance grapevine immunity against pathogen attack.

Abstract: Soil-crop models are increasingly used as predictive tools to assess yield and environmental impacts of agriculture in a growing diversity of contexts. They are however seldom evaluated at a given time over a wide domain of use. We tested here the performances of the STICS model (v8.2.2) with its standard set of parameters over a dataset covering 15 crops and a wide range of agropedoclimatic conditions in France. Model results showed a good overall accuracy, with little bias. Relative RMSE was larger for soil nitrate (49%) than for plant biomass (35%) and nitrogen (33%) and smallest for soil water (10%). Trends induced by contrasted environmental conditions and management practices were well reproduced. Finally, limited dependency of model errors on crops or environments indicated a satisfactory robustness. Such performances make STICS a valuable tool for studying the effects of changes in agro-ecosystems over the domain explored. STICS v8.2.2 soil-crop model was evaluated over a large and varied dataset using its standard set of parameters.Level of accuracy is 10-50% for plant, soil water and nitrate outputs.Model reproduces well trends arising from contrasted agro-environmental conditions.Errors are weakly dependent on the agro-environmental conditions tested.Model accuracy and robustness is considered good for scenario testing and large scale use within the conditions tested here.

Abstract: We investigated the in vitro effects of four alkyl-galactofuranoside derivatives, i.e., octyl-β- d -galactofuranoside (compound 1), 6-amino-β- d -galactofuranoside (compound 2), 6- N -acetamido-β- d -galactofuranoside (compound 3), and 6-azido-β- d -galactofuranoside (compound 4), on Leishmania donovani . Their mechanism of action was explored using electron paramagnetic resonance spectroscopy (EPR) and nuclear magnetic resonance (NMR), and ultrastructural alterations were analyzed by transmission electron microscopy (TEM). Compound 1 showed the most promising effects by inhibiting promastigote growth at a 50% inhibitory concentration (IC 50 ) of 8.96 ± 2.5 μM. All compounds exhibit low toxicity toward human macrophages. Compound 1 had a higher selectivity index than the molecule used for comparison, i.e., miltefosine (159.7 versus 37.9, respectively). EPR showed that compound 1 significantly reduced membrane fluidity compared to control promastigotes and to compound 3. The furanose ring was shown to support this effect, since the isomer galactopyranose had no effect on parasite membrane fluidity or growth. NMR showed a direct interaction of all compounds (greatest with compound 1, followed by compounds 2, 3, and 4, in descending order) with the promastigote membrane and with octyl-galactopyranose and octanol, providing evidence that the n -octyl chain was primarily involved in anchoring with the parasite membrane, followed by the putative crucial role of the furanose ring in the antileishmanial activity. A morphological analysis of compound 1-treated promastigotes by TEM revealed profound alterations in the parasite membrane and organelles, but this was not the case with compound 3. Quantification of annexin V binding by flow cytometry confirmed that compound 1 induced apoptosis in >90% of promastigotes. The effect of compound 1 was also assessed on intramacrophagic amastigotes and showed a reduction in amastigote growth associated with an increase of reactive oxygen species (ROS) production, thus validating its promising effect.

Abstract: Changes in grassland management induce disturbances that influence both soil functioning and soil fauna. This study aimed at determining the extent to which the composition of a grassland soil nematode community could be altered by a shift of grazing regime and the potential feedback that these alterations could provoke on grassland functioning. Therefore, we monitored the composition of the soil nematode community (i.e., plant-, bacterial- and fungal-feeders, omnivores and carnivores) of mesocosms that were sampled from two contrasted long-term field trials (high vs low grazing treatments) and subsequently subjected to high or no grazing for 2 years. The soil nematofauna responded faster and more strongly to the application of an intensive grazing regime on a previously extensively exploited system than the other way round. The application of an intensive grazing regime induced a significant increase in numbers of bacterial feeders and a decrease of the relative abundance of fungal-feeding nematodes. The nematofaunal community structure was determined by both the past and current grazing regimes throughout the 2 years of monitoring. Observed effects on soil microbivores seemed to reflect the 'immediate' above-ground primary production potential and to follow micro-organism dynamics. On the other hand, observed effects on root-feeding nematodes seemed to reflect the integral effect of past and current grazing regimes on plant community root biomass and quality.

Abstract: International audience; This study reports the in vitro efficacy of a mixture of plant extracts used as antibacterial agents loaded in a dressing to prevent infection in a wound. These natural compounds were extracted from avocado and mango.A mixture of equal proportions of petroleum ether extract from avocado kernels and methanolic extract from mango kernels was effective on the majority of 36 bacterial strains and presented excellent results against S. aureus. The Minimal Inhibitory Concentrations (MIC) measured on Staphylococcus aureus (ATCC 6538) were 0.019 mg/mL for the mixture compared to 0.039 mg/mL for the avocado or mango extract alone. This mixture was incorporated into a wound dressing composed of non-woven fabric and a gel. This gel consists of a plasticizer (starch/glycerin) and an absorbent (CMC/PCD) and is designed to have swelling properties and entrap plant extract. A formulation containing equal proportions of CMC and PCD enabled us to obtain 800% of swelling and a good adhesion of the gel to the non-woven fabric.The final dressing containing the mixture of avocado and mango extracts presented excellent antibacterial activity against S. aureus. Microbiological tests demonstrated that a wound dressing loaded with avocado/mango seed extract induced additional bacterial reduction of 3 Log10 and 2 Log10 against S. aureus and P. aeruginosa respectively but no additional bacterial reduction of C. albicans compared to the control.Our study demonstrated the scope for interest in plant species extracts provided by agricultural co-products in developing a new antibacterial wound dressing.

Abstract: Feces from 142 animals were collected on 15 farms in the region of Brittany, France. Each sample was directly collected from the rectum of the animal and identified with the ear tag number. Animals were sampled three times, at 5, 15 and 22 weeks of age. After DNA extraction from stool samples, nested PCR was performed to amplify partial 18S-rDNA and 60 kDa glycoprotein genes of Cryptosporidium. The parasite was detected on all farms. One hundred out of 142 calves (70.4%) were found to be parasitized by Cryptosporidium. Amplified fragments were sequenced for Cryptosporidium species identification and revealed the presence of C. parvum (43.8%), C. ryanae (28.5%), and C. bovis (27%). One animal was infected with Cryptosporidium ubiquitum. The prevalence of these species was related to the age of the animal. C. parvum caused 86.7% of Cryptosporidium infections in 5-week-old calves but only 1.7% in 15-week-old animals. The analysis of the results showed that animals could be infected successively by C. parvum, C. ryanae, and C. bovis for the study period. C. parvum gp60 genotyping identifies 6 IIa subtypes of which 74.5% were represented by IIaA15G2R1. This work confirms previous studies in other countries showing that zoonotic C. parvum is the dominant species seen in young calves.

Abstract: International audience; Fusarium head blight (FHB) is a cereal disease of major importance responsible for yield losses and mycotoxin contaminations in grains. Here, we introduce a new measurement approach to quantify FHB severity on grains based on the evaluation of the whitened kernel surface (WKS) using digital image analysis. The applicability of WKS was assessed on two bread wheat and one triticale grain sample sets (265 samples). Pearson correlation coefficients between Fusarium-damaged kernels (FDK) and WKS range from r = 0.77 to r = 0.81 and from r = 0.61 to r = 0.86 for the correlation between deoxynivalenol (DON) content and WKS. This new scoring method facilitates fast and reliable assessment of the resistance to kernel infection and shows significant correlation with mycotoxin content. WKS can be automated and does not suffer from the "human factor" inherent to visual scorings. As a low-cost and fast approach, this method appears particularly attractive for breeding and genetic analysis of FHB resistance where typically large numbers of experimental lines need to be evaluated, and for which WKS is suggested as an alternative to visual FDK scorings.

Abstract: International audience; This study presents a miniaturized 3-methyl-2-benzothiazolinone hydrazone hydrochloride-assay (MBTHassay) 1 , optimized for high-throughput measurement of shrimp phenol oxidase activities and the screening of potential inhibitors of post-mortem melanosis. To validate this approach, assay parameters, including limit of detection and dynamic range, were compared to values obtained for DOPAchrome-assay, used here as a reference method. A reliable high-throughput automatized analysis was also performed, under standardized parameters, in order to compare and characterize 40 potential inhibitors of phenol oxidase activities. Among the selected molecules, twenty showed inhibitory action. Enzyme-inhibitor dissociation constant (K i), median inhibitor concentration (IC 50) and inhibition modes could be determined for nine of them. 4-Hexylresorcinol, Nphenylthiourea L-cystein and glutathione displayed K i values lower than sulfite salts (Na 2 S 2 O 5 and Na 2 SO 3) used as positive controls.

Abstract: International audience; Front-face fluorescence spectroscopy technique coupled with chemometric tools was used for predicting the freshness state of a Lebanese dairy product called Qishta, stored up to 20 days. Acid, peroxide and thiobarbituric acid reactive substances (TBARS) values reached no more than 0.93 mg NaOH g−1 fat, 6.22 meq O2 Kg−1 fat, and 0.0313 mg malonaldehyde (MA) kg−1 sample, respectively, throughout the investigated storage time. In parallel, fluorescence emission spectra of tryptophan and riboflavin, and fluorescence excitation spectra of vitamin A were recorded and showed the highest fluorescence intensity for the Qishta samples aged of 20 days and the lowest intensity for the fresh ones. The primary and secondary indicators of lipid oxidation showed that Qishta can be stored for 20 days without any alteration despite the increase in the TBARS after 16 days of storage. Principal component analysis (PCA) applied on riboflavin emission spectra allowed better discrimination between Qishta samples with a clear distinction of those aged 20 days while some overlapping was noticed between samples aged below 16 days. A high correlation (R2 = 0.923) was observed between the peroxide value and the intensity of the riboflavin fluorescence recorded at 460 nm.

Abstract: Particle-based photodynamic therapy (PPDT) holds great promise in theranostic applications. Herein, we demonstrate that PPDT based on gold nanorods coated with an indocyanine green (ICG)-loaded silica shell allows for the inactivation of the Crohn's disease-associated adherent-invasive Escherichia coli strain LF82 (E. coli LF82) under pulsed laser light irradiation at 810 nm. Fine-tuning of the plasmonic structures together with maximizing the photosensitizer loading onto the nanostructures allowed optimizing the singlet oxygen generation capability and the PPDT efficiency. Using a nanoparticle concentration low enough to suppress photothermal heating effects, 6 log10 reduction in E. coli LF82 viability could be achieved using gold nanostructures displaying a plasmonic band at 900 nm. An additional modality of nanoparticle-based photoinactivation of E. coli is partly observed, with 3 log10 reduction of bacterial viability using Au NRs@SiO2 without ICG, due to the two-photon induced formation of reactive oxygen species. Abstract: International audience; Wine supply in the French market is structured in an intricate system of categories based on origin. There is very little knowledge about consumers understanding of this complex category system and the sensory styles behind these categories. This study investigated how assessors with different level of expertise categorized Beaujolais wines from general to more specific levels of categorization (grape variety, appellation, and “lieu-dit”) in both perceptual (wines) and conceptual (wine labels) conditions. Based on the literature on expertise, we expected a stronger effect in the perceptual condition, in particular for the most specific levels of categorization. For each wine categorization level, three sets of 12 wines were tasted by three panels of 60 assessors: a panel of unfamiliar novices with no much exposure to Beaujolais wines; a panel of familiar novices with regular exposure to Beaujolais wines; and a panel of experts from the Beaujolais. In both perceptual and conceptual conditions, assessors were asked to perform a binary sorting task, followed by a verbalisation task. Data were analysed using DISTATIS. At each level, with a few exceptions, a clearer separation was observed between the two categories in the conceptual condition than in the perceptual condition. Although the experts categorized the labels by grape variety, they did not spontaneously categorize the wines in that way. Finally, we observed a clear expertise effect only in the conceptual condition. This result will be discussed in terms of expertise acquisition and categorization processes.

Abstract: International audience; Blood, from slaughterhouses, is an inevitable part of meat production, causing environmental problems due to the large volumes recovered and its low valorization. However, the α137-141 peptide, a natural antimicrobial peptide, can be obtained after hydrolysis of hemoglobin, the main constituent of blood red part. To recover it at a sufficient concentration for antimicrobial applications, a new sustainable technology, called electrodialysis with ultrafiltration membrane (EDUF), was investigated. The α137-141 concentration was increased about 4-fold at a feed peptide concentration of 8% with an enrichment factor above 24-fold. This feed peptide concentration also needed the lowest relative energy consumption. Moreover, this peptide fraction protected meat against microbial growth, as well as rancidity, during 14 days under refrigeration. This peptide fraction was validated as a natural preservative and substitute for synthetic additives against food spoilage. Finally, producing antimicrobial/antioxidant peptide from wastes by EDUF fits perfectly with the concept of circular economy.

Abstract: Dans la recherche de couplages optimaux de catalyseurs, les chercheurs pionniers en catalyse hybride se sont tout naturellement tournés vers l’élaboration de matériaux multi-catalytiques hybrides (MMCH) constitués à partir de co-immobilisation sur un même support de catalyseurs chimiques et biologiques. Cette exécution disciplinaire innovante, située au carrefour de la chimie, la biologie et la science des matériaux, fait appel à de très nombreuses compétences, et sa complexité n’a d’égale que la diversité des propriétés catalytiques que peuvent offrir ces MMCH. En particulier, l’utilisation d’une large variété de supports organiques et inorganiques, rigides et flexibles, a d’ores et déjà permis l’élaboration de quelques déclinaisons remarquables de catalyseurs hybrides démontrant de véritables effets de synergie, avec des rendements et des excès énantiomériques finaux inatteignables en utilisant des catalyseurs de manière isolée, et ce même dans des procédés en 1P1S. Véritable fer de lance de la catalyse hybride, les MMCH concentrent l’essence même de ce domaine, nécessitant communication et collaboration extrêmement étroites entre chimistes, biologistes, chercheurs en science des matériaux, ou encore en modélisation. Son développement sera sans aucun doute dans un futur proche le témoin d’une véritable (r)évolution de la recherche en catalyse vers des thématiques inter-, voire transdisciplinaires. In the search for optimal coupling of catalysts, it is quite naturally that pioneering researchers in hybrid catalysis have turned toward the development of hybrid multi-catalytic materials (HMCMs) constituted from the co-immobilization on the same carrier of chemical and biological catalysts. Such an innovative disciplinary variant, located at the crossroads of chemistry, biology and materials sciences, calls for a wide range of skills ; its complexity is only equaled by the diversity of catalytic properties that HMCMs can offer. In particular, the use of a wide variety of organic and inorganic supports, either rigid or flexible, has already led to the development of some remarkable examples of hybrid catalysts exhibiting real synergy effects, with yields and final enantiomeric excesses that are unreachable when using each catalyst separately, even in 1P1S processes. As the spearhead of hybrid catalysis, HMCMs concentrate the very essence of this field, requiring seamless communication and collaboration between chemists, biologists, materials science researchers and modeling specialists. Abstract: The heterogeneous catalytic partial oxidation of alcohols and aldehydes in the liquid phase usually needs the addition of a homogeneous base, which in turn makes the products&rsquo, recovery cumbersome, and can further induce undesired side reactions. In the present work, we propose the use of novel catalysts based on metallic Au, Pd and bimetallic AuPd nanoparticles embedded in a titanosilicate matrix. The as-prepared catalysts showed good efficiency in the base-free partial oxidation of furfural and 5-hydroxymethylfurfural. Au4Pd1@SiTi catalyst showed high selectivity (78%) to monoacids (namely, 5-formyl-2-furancarboxylic acid and 5-hydroxymethyl-2-furancarboxylic acid) at 50% 5-hydroxymethylfurfural (HMF) conversion. The selectivity even reached 83% in the case of furfural oxidation to furoic acid (at 50% furfural conversion). The performances of the catalysts strongly depended on the Au&ndash, Pd ratio, with an optimal value of 4:1. The pH of the solution was always below 3.5 and no leaching of metals was observed, confirming the stabilization of the metal nanoparticles within the titanosilicate host matrix.

Abstract: The effectiveness of chemical as well as biological catalysts for carrying out chemical reactions on an industrial level is no longer to be demonstrated. Their specificity and selectivity make them essential tools for many processes, and as such, they represent a major key in the establishment of tomorrow's industry. Historically, catalysts have mostly been used in a unitary manner, with processes involving several catalysts most often forming part of multi-stage strategies. However, the last two decades have seen a significant increase in research into multi-catalytic reactions that combine two or more catalysts in order to benefit from collaborative effects. While catalysts of different natures are very rarely used together, the combination of a chemical catalyst and a biocatalyst, known as "hybrid catalysis", is most likely the most promising type of multi-catalytic reaction because of the unprecedented diversity and efficiency allowed. Nevertheless, it remains the most complex to implement, calling on skills from different fields (chemistry, biology, materials, etc.). The prospects it offers are giving riseto pioneering work that is pushing researchers to develop new strategies for overcoming the limitations and inhibitions that inevitably arise when these two types of catalyst are forced to work together.; L’efficacité des catalyseurs chimiques et biologiques pour la réalisation de réactions chimiques au niveau industriel n’est plus à démontrer. Leur spécificité et sélectivité en font des outils essentiels à de nombreux procédés, et ils représentent à ce titre un outil majeur dans l’établissement de l’industrie de demain.Historiquement, les catalyseurs ont été très majoritairement utilisés de manière unitaire, les procédés en faisant intervenir plusieurs s’inscrivant le plus souvent dans des stratégies multi-étapes. Cependant, les deux dernières décennies ont vu un essor de la recherche autour des réactions multi-catalytiques qui combinent deux catalyseurs ou plus afin de bénéficier d’effets collaSi des catalyseurs de natures différentes ne sont que très rarement mis en jeu ensemble, la combinaison d’un catalyseur chimique et d’un biocatalyseur, appelée « catalyse hybride », représente très probablement le type de réaction multi-catalytique le plus prometteur de par la diversité et l’efficacité inédites permises. Elle n’en demeure pas moins la plus complexe à mettre en œuvre, faisant appel à des compétences de domaines différents (chimie, biologie, matériaux, etc.). Abstract: International audience; Freezing is a major environmental limitation that affects biomass and seed productivity in a large number of crop species including legumes. Medicago truncatula is a model molecular‐genetic system for legume biology. A strategy to decipher freezing tolerance after a cold acclimation period in M. truncatula was developed using a quantitative genetic approach. Three main quantitative trait loci (QTL) with additive effects for freezing damage were detected on chromosomes 1, 4, and 6 using a recombinant inbred line population derived from a cross between the freezing‐tolerant accession F83005‐5 and the freezing‐sensitive accession DZA045‐5. The QTL on chromosome 6, named Mt‐FTQTL6, explained 40% of the phenotypic variance. Fine mapping of Mt‐FTQTL6 led to the identification of 20 positional candidate genes. Twelve of these 20 genes belong to the C‐repeat binding factor/dehydration‐responsive element binding factor 1 group of the AP2/EREBP transcription factor family and are tandemly arrayed. Non‐synonymous single nucleotide polymorphism (SNP) mutations, indels, and copy number variations were identified as putative candidate polymorphisms in CBF/DREB1 genes to explain the difference in freezing tolerance between the parental M. truncatula accessions. Abstract: Fungi are notoriously prolific producers of secondary metabolites including nonribosomal peptides (NRPs). The structural complexity of NRPs grants them interesting activities such as antibiotic, anti-cancer, and anti-inflammatory properties. The discovery of these compounds with attractive activities can be achieved by using two approaches: either by screening samples originating from various environments for their biological activities, or by identifying the related clusters in genomic sequences thanks to bioinformatics tools. This genome mining approach has grown tremendously due to recent advances in genome sequencing, which have provided an incredible amount of genomic data from hundreds of microbial species. Regarding fungal organisms, the genomic data have revealed the presence of an unexpected number of putative NRP-related gene clusters. This highlights fungi as a goldmine for the discovery of putative novel bioactive compounds. Recent development of NRP dedicated bioinformatics tools have increased the capacity to identify these gene clusters and to deduce NRPs structures, speeding-up the screening process for novel metabolites discovery. Unfortunately, the newly identified compound is frequently not or poorly produced by native producers due to a lack of expression of the related genes cluster. A frequently employed strategy to increase production rates consists in transferring the related biosynthetic pathway in heterologous hosts. This review aims to provide a comprehensive overview about the topic of NRPs discovery, from gene cluster identification by genome mining to the heterologous production in fungal hosts. The main computational tools and methods for genome mining are herein presented with an emphasis on the particularities of the fungal systems. The different steps of the reconstitution of NRP biosynthetic pathway in heterologous fungal cell factories will be discussed, as well as the key factors to consider for maximizing productivity. Several examples will be developed to illustrate the potential of heterologous production to both discover uncharacterized novel compounds predicted in silico by genome mining, and to enhance the productivity of interesting bio-active natural products.

Abstract: The most common frauds of tuna cans supply chain concern the substitution or mixing of valuable tuna species with cheaper ones, which is strictly forbidden. The objective of the present study was to determine the potential use of front-face fluorescence spectroscopy (FFFS) as a rapid tool to authenticate species in canned tuna: skipjack tuna (Katsuwonus pelamis), yellowfin tuna (Thunnus albacares), Albacore tuna (Thunnus alalunga), and bigeye tuna (Thunnus obesus). Different spectra (tryptophan residues, aromatic amino acids, and nucleic acids (AAA + NA), riboflavin, nicotinamide adenine dinucleotide (NADH), and vitamin A) were recorded on 256 canned tunas, produced at the pilot scale, that were used for the establishment of models. The robustness of the established models was tested on 40 commercial canned tunas. According to the label tunas, the percentage of correct classification reached 75% allowing us to conclude that FFFS may represent a promising tool to be used by both canning industry and governmental control agencies to ascertain correct labeling of canned tuna.

Abstract: International audience; The objectives of this work are to address the prebiotic effects of chicory (Cichorium intybus) together with its possible role in appetite control. We compared nine chicory genotypes in order to determine if variations in the content of metabolites in the roasted roots would lead to modifications in release of satiety hormones and in composition of gut microbiota. To this aim, a 5-week dietary-intervention study was achieved using mice fed with distinct chicory-based preparations. A 16S rRNA gene-based metagenetic analysis of fecal microbiota was performed. In vitro gastrointestinal digestions were performed in order to study the effect of chicory intestinal digests on gut hormone regulation in enteroendocrine cells. Firmicutes/ Bacteroidetes ratio and gut bacterial groups, such as Alloprevotella, Blautia, Alistipes, and Oscillibacter, were found to be modulated by chicory. On the other hand, CCK and GLP-1 satiety hormones were demonstrated to be significantly increased by chicory in vitro.

Abstract: The objective of the present study was to reproduce in 4 months the quality changes that occur in shelf-stable strawberry pudding after 1 year of storage. A full factorial experiment 23 was used to investigate the effect of storage temperature (16 and 30 °C), headspace volume (9.5 and 18 cm3), and agitation (no/yes—pouches turned upside down twice a week) on aging. Sensory, rheological, pH, and fluorescence measurements were performed on samples subjected to accelerated aging tests (up to 4 months) or stored in standard conditions (up to 1 year). Although storage at 30 °C induced a slight decrease in apparent viscosity and thickness—in contrast with samples stored at 16 °C—it showed the best results for most of the studied parameters, regardless of the headspace volume and agitation. Hence, storage at 30 °C could be used for accelerated shelf-life or stability studies of sterilized pudding.

Abstract: International audience; The Arabidopsis thaliana genome contains 58 members of the solute carrier family SLC25, also called the mitochondrial carrier family, many of which have been shown to transport specific metabolites, nucleotides, and cofactors across the mitochondrial membrane. Here, two Arabidopsis members of this family, AtUCP1 and AtUCP2, which were previously thought to be uncoupling proteins and hence named UCP1/PUMP1 and UCP2/PUMP2, respectively, are assigned with a novel function. They were expressed in bacteria, purified, and reconstituted in phospholipid vesicles. Their transport properties demonstrate that they transport amino acids (aspartate, glutamate, cysteine sulfinate, and cysteate), dicarboxylates (malate, oxaloacetate, and 2-oxoglutarate), phosphate, sulfate, and thiosulfate. Transport was saturable and inhibited by mercurials and other mitochondrial carrier inhibitors to various degrees. AtUCP1 and AtUCP2 catalyzed a fast counterexchange transport as well as a low uniport of substrates, with transport rates of AtUCP1 being much higher than those of AtUCP2 in both cases. The aspartate/glutamate heteroexchange mediated by AtUCP1 and AtUCP2 is electroneutral, in contrast to that mediated by the mammalian mitochondrial aspartate glutamate carrier. Furthermore, both carriers were found to be targeted to mitochondria. Metabolite profiling of single and double knockouts shows changes in organic acid and amino acid levels. Notably, AtUCP1 and AtUCP2 are the first reported mitochondrial carriers in Arabidopsis to transport aspartate and glutamate. It is proposed that the primary function of AtUCP1 and AtUCP2 is to catalyze an aspartate(out)/glutamate(in) exchange across the mitochondrial membrane and thereby contribute to the export of reducing equivalents from the mitochondria in photorespiration.

Abstract: International audience; For powder catalyst characterization, Fourier Transform Infrared (FTIR), Raman, and X-Ray Fluorescence (XRF) spectrometers and X-Ray Diffraction (XRD) are available in high-throughput (HT) configurations, for example at the REALCAT platform to sequentially analyse multiple sets of samples. To remove the bottleneck resulting from the use of different sample holders for each equipment, a unique multi-well plate was developed. This paper details the design of such a plate including the selection of the fabrication material and the plate dimensioning based on the study of the 4 different physical interactions between matter and electromagnetic radiations for the aforementioned techniques. This new plate consists of a holder for removable wells enabling the avoidance of cross-contamination between samples. Raman, a focusing technique, has no strict constraint on the plate design. The number of wells, their geometry, spacing and dimensions were adjusted to deal with the constraints of IR optics. The well depth was set according to the XRF maximum penetration depth in the sample. The well diameter was optimized in order to obtain from the X-ray spot size the maximum achievable intensity. Poly-methyl-methacrylate (PMMA) was chosen as the material for the new plate due to its amorphous structure (no peak in XRD analysis) and ease with which it can be cut by a laser. Finally, the flatness of the multi-well plate was validated on the most challenging instrument: XRD. This new plate allows fast sample filling/preparation, requires small quantities of catalyst (50 to 80 mg) in each well and is compatible and convenient for HT experimentation.

Abstract: International audience; The aim of this study was to evaluate the potential of using front face fluorescence spectroscopy (FFFS) to discriminate fresh refrigerated (up to 13 days at 4 °C) sea bass (Dicentrarchus labrax) fillets (F group) from: i) frozen-thawed (3 months at −18 °C), then refrigerated up to 9 days fillets at 4 °C (F-T R group), and ii) refrigerated (up to 9 days at 4 °C), then frozen-thawed (3 months at −18 °C) fillets (R F-T group). The evolution of emission fluorescence spectra after excitation set at 290, 340, and 380 nm, and excitation spectra after emission set at 410 nm were acquired throughout storage. Some traditional measurements based on color, textural, and chemical parameters were performed on the same fillets. Canonical correlation analysis (CCA), applied to the traditional and fluorescence data sets, showed high correlations between the two data sets, since squared canonical coefficients were found higher than 0.8 for canonical variates 1 and 2. This was confirmed by the factorial discriminant analysis (FDA) applied to the concatenated emission spectra recorded after excitation sets at 340 and 380 nm since 72 out of 78 samples were correctly classified. Finally, a multi-block analysis method based on common components and specific weights analysis (CCSWA) was applied to the whole data sets. A reliable discrimination between fresh from frozen/thawed fish was found suggesting that FFFS could be used as a promising tool in routine inspection for differentiating between fresh and frozen-thawed fish.

Abstract: International audience; The potentiality of the front-face fluorescence spectroscopy coupled with chemometric tools to characterize changes in camel milk following thermal treatments in the 55–75 °C temperature range from 0.5 min up to 30 min was studied. Nicotinamide adenine dinucleotide (NADH), fluorescent Maillard reaction products (FMRP) and vitamin A fluorescence spectra were collected on camel milk samples. Using principal component analysis (PCA), the vitamin A spectra allowed some discrimination between milk samples according to heat treatment intensity and holding time. The best results was obtained by applying common components and specific weights analysis (CCSWA) to the three spectral data sets, since a clear differentiation between camel milk samples preheated at 70 and 75 °C from the others was achieved. Apparent activation energy of camel milk determined from NADH, FMRP and vitamin A spectra was of 85.1, 84.2 and 47.3 kJ mol−1, respectively. The spectral patterns of the CCSWA model enabled us to get information about the molecular changes occurring in camel milk fluorophores during heat treatment.

Abstract: National audience; Zymoseptoria tritici, responsible for Septoria tritici blotch, is the most important pathogen of wheat. The control of this parasite relies mainly on synthetic fungicides, but their use is increasingly controversial and searching for alternative management strategies is encouraged. In this context, the biocontrol potential of crude methanolic extracts of eight extremophile plant species from Tunisia, including three xerophytes and five halophytes, against Z. tritici was assessed. Only the extract of Juncus maritimus rhizomes showed significant in vitro antifungal activity. In extremophile plants, the production of secondary metabolites is often influenced by abiotic conditions. Thus, we collected several samples of J. maritimus rhizomes at different vegetative stages, at different periods, and from different substrates to compare their antifungal activities. Our results suggest that the plant environment, especially the substrate of the soil, should be taken into account to identify great sources of natural antifungal products. From the most active sample, a 9,10-dehydrophenanthrene derivative, effusol, absent from other J. maritimus rhizomes extracts, was purified. This product showed a strong antifungal activity against the pathogen, with a minimal inhibitory concentration of 19gmL(-1) and an half-maximal inhibitory concentration of 9.98gmL(-1). This phenanthrene derivative could be a promising biocontrol molecule against Z. tritici.
