Petroleum-based packaging materials are nondegradable and unsustainable and thus are harmful to the environment. Renewable packaging films prepared from bio-based raw materials are promising alternatives to petroleum-based packaging materials. In this study, colorless and transparent bio-based films were successfully cast using a solution containing a mixture of arabinogalactan (AG) and poly (vinyl alcohol) (PVA). Vanillin was incorporated into the mixture to endow the films with UV-shielding, antioxidant, and antibacterial properties. The morphological, physical, antioxidant, and antibacterial properties of the blend films were then characterized. At an AG:PVA weight ratio of 1:3, and the vanillin content was 0.15 %, the tensile strength of the AG/PVA/Vanillin (APV) films reached ~28 MPa, while their elongation at break reached ~475 %. The addition of vanillin significantly affected the antioxidant and antibacterial properties of the blend films, which exhibited superb UV barrier capacity. The APV films exhibited extremely low oxygen transmittance, delaying the onset of mold/rot in strawberries and reducing their weight loss. Because of the heat sealability of the blend films, they can be used for encapsulating various substances, such as concentrated laundry liquid. Moreover, the blend films were recyclable and biodegradable. Sodium alginate (SA) is widely used in the food, biomedical, and chemical industries due to its biocompatibility, biodegradability, and excellent film-forming properties. This article introduces a simple method for preparing uniform alginate-based packaging materials with exceptional properties for fruit preservation. The alginate was uniformly crosslinked by gradually releasing calcium ions triggered by the sustained hydrolysis of gluconolactone (GDL). A cinnamaldehyde (CA) emulsion, stabilized by xanthan without the use of traditional surfactants, was tightly incorporated into the alginate film to enhance its antimicrobial, antioxidant, and UV shielding properties. The alginate-based film effectively blocked ultraviolet rays in the range of 400-200 nm, while allowing for a visible light transmittance of up to 70 %. Additionally, it showed an increased water contact angle and decreased water vapor permeability. The alginate-based film was also employed in the preparation of coated paper through the commonly used coating process in the papermaking industry. The alginate-based material displayed excellent antioxidant properties and antimicrobial activity against Escherichia coli, Staphylococcus aureus and Botrytis cinerea, successfully extending the shelf life of strawberries to 7 days at room temperature. In this study, we investigated the structural characterization and biological activities of Bletilla striata polysaccharides (BSPs) for their role as antioxidants and anti-melanogenesis agents in skin healthcare protection. Three neutral polysaccharides (BSP-1, BSP-2, and BSP-3) with molecular weights of 269.121 kDa, 57.389 kDa, and 28.153 kDa were extracted and purified. Their structural characteristics were analyzed by ion chromatography, GC-MS, and 1D/2D NMR. The results showed that BSP-1, which constitutes the major part of BSPs, was composed of α-D-Glcp, β-D-Glcp, β-D-Manp, and 2-O-acetyl-β-D-Manp, with the branched-chain accompanied by β-D-Galp and α-D-Glcp. BSP-1, BSP-2, and BSP-3 can enhance the total antioxidant capacity of skin fibroblasts with non-toxicity. Meanwhile, BSP-1, BSP-2, and BSP-3 could significantly inhibit the proliferative activity of melanoma cells. Among them, BSP-1 and BSP-2 showed more significance in anti-melanogenesis, tyrosinase inhibition activity, and cell migration inhibition. Polysaccharides as the biopolymers are showing various structural and modulatory functions. Effective separation of carbohydrate structures is essential to understanding their function. In this study, we choose an efficient organic acid in combination with recyclable organic solvent three-phase partitioning technology for the simultaneous extraction of polysaccharides from Ampelopsis japonica (AJPs) to ensure the integrity of linear and branched polysaccharide. The monosaccharide composition, glycosidic linkage information, structural and physicochemical analyses and associations with antioxidant activities were extensively analyzed. Synergistic extraction was compared with the conventional hot water extraction method and the results showed that AJPs-HNP exhibited better elastic properties and excellent antioxidant activity. Correlation analysis confirmed that the antioxidant activity of AJPs was significantly correlated with relative molecular weight, uronic acid content and terminal glycoside linkage molar ratios. The collaborative processing has significantly improved the utilization potential of AJPs and provides a sound theoretical foundation for the effective extraction and separation of polysaccharides. Hemicellulose extracted from ecalyptus APMP pulping waste liquor and undergoes etherification modification to produce carboxymethyl hemicellulose (CMHC). Subsequently, CMHC undergoes esterification reaction with p-hydroxybenzoic acid to synthesize a novel polysaccharide-based preservative known as carboxymethyl hemicellulose p-hydroxybenzoate (P-CMHC). The synthesis conditions of P-CMHC were optimized using the response surface methodology, resulting in an optimal esterification condition that achieved a degree of substitution of 0.232. P-CMHC exhibits excellent antioxidant activity, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical scavenging activities. Additionally, it demonstrates favorable hygroscopic and moisturizing properties. Thiazole blue (MTT) experiments evaluating cell proliferation rate indicate that P-CMHC possesses negligible cytotoxicity, making it a promising, safe, and healthy preservative. Acmella radicans (Asteraceae) is a plant native to America. Despite it having medicinal attributes, studies on its phytochemical properties are scarce, and biotechnological studies do not exist for this species. In this study, we established an adventitious root culture from A. radicans internodal segments in shake flasks with indole-3-butyric acid (IBA), and then elicited it with jasmonic acid (JA) and salicylic acid (SA). The total phenolic content and antioxidant activity were evaluated, and a comparison was made using in vitro plantlets and wild plants. Internodal segments with 0.1 mg/L IBA showed 100% root induction and exhibited better growth after transfer to shake flasks with MS liquid culture medium. JA had a significant effect on biomass increase compared to unelicited roots, mainly with 50 µM JA (28%), while SA did not show significant results. Root elicited with 100 µM (SA and JA) showed a 0.34- and 3.9-fold increase, respectively, in total phenolic content (TPC) compared to the control. The antioxidant activity was also significant, and a lower half-maximal inhibitory concentration (IC 50 ) was observed as the AJ concentration increased. Roots elicited with AJ (100 µM) exhibited high antioxidant activity with DPPH (IC 50 = 9.4 µg/mL) and ABTS (IC 50 = 3.3 µg/mL) assays; these values were close to those for vitamin C (IC 50 = 2.0 µg/mL). The TPC and antioxidant activity of in vitro plants and root cultured in shake flasks showed the lowest values in most cases; even the root cultures without elicitation were better than those of a wild plant. In this study, we demonstrated that A. radicans root culture is capable of producing secondary metabolites, while its production and antioxidant activity can be enhanced using jasmonic acid.

Protein-based coatings are of immense interest due to their rich biological functions. Layer-by-layer (LbL) assembly, as a powerful means of transferring protein functions to the material surface, has received widespread attention. However, the assembly mechanism of protein-based LbL coatings is still far from being explained, not only because of protein structure and function diversity but also characterization limitations. Herein, we monitored in situ the LbL assembly process of tannic acid (TA) and lysozyme (Lyz), a classic pair of polyphenol and protein, by combining quartz crystal microbalance with dissipation monitoring (QCM-D) and spectroscopic ellipsometry (SE). The water content, morphology, mechanical properties, antioxidant activity, and the driving force of TA-Lyz coating engineered under different pH values were analyzed in detail by various techniques. The water content, a key factor in TA-Lyz coatings, increased with increasing assembled pH values, which resulted in a porous morphology, inhomogeneous mechanical distribution, faster assembly growth, and better antioxidant activity in both acellular and cellular levels. In addition, high water content is unfavorable to both entropy and enthalpy changes, and the thermodynamic driving force of TA and Lyz assembly mainly comes from the enthalpy change brought by the noncovalent interaction between TA and Lyz. These results provide new insights into engineering the structure, function, and assembly mechanisms of protein-based coatings.

This research established the optimal conditions for alkali-assisted extraction (AAE) of bioactive polysaccharides from Bletilla striata integrated with response surface methodology (RSM) and the genetic algorithm-artificial neural networks (GA-ANN). In comparison with RSM, the ANN model showed a relatively higher determination coefficient in the global output values (RSM: ANN = 0.9270: 0.9742) performing more satisfactorily in the validation. Under the optimum conditions (52 °C; 167 min, and 0.01 mol/L NaOH), the extraction yields, IC 50 of ABTS, and FRAP value were 29.53 ± 0.97 %, 3.41 mg/mL, and 39.11 μmol Fe 2+ /g, respectively. The results indicated that BSPs-A was mainly composed of glucose and mannose with small amounts of arabinose, galactose, and galacturonic acid, while possessed a molecular weight of about 305.94 kDa (Mw). The structural characterization of BSPs-A was initially characterized by FT-IR, SEM, and Congo red tests, which indicated that BSPs-A possessed a triple helix conformation of typical Bletilla striata polysaccharides. In addition, BSPs-A exhibited excellent antioxidant activity, which was further confirmed by a series of in vitro antioxidant activity assays including DPPH, ABTS, FRAP, and ORAC. This research study explores the fabrication of polyvinyl alcohol (PVOH) and corn starch (ST) with pineapple peel extract (PPE) as a natural antioxidant agent, which is an abundant by-product from the food processing industry via casting method. The effects of PPEs concentration (5%, 10%, 15%, and 20%) on the antioxidant capacity, optical, thermal, mechanical, barrier properties, and changes in PVOH-starch molecular structure of PVOH/ST films were investigated. The results revealed that with the increasing concertation of PPEs, prepared films' thickness and water vapor permeability slightly increased. Elongation at break of PVOH/ST films was also enhanced with PPEs concentration. All PPEs incorporated films exhibited enhanced thermal stability as the degradation occurred above 300 °C. The addition of PPE to PVOH/ST films remarkably increased the antioxidant properties. Finally, prepared PVOH/ST/PPE films demonstrated to be a capable material for developing active biodegradable packaging material due to its proven antioxidant activity and mechanical property, which can be helpful in the packaging of food products that gets spoiled due to oxidation reactions., (Copyright © 2021 Elsevier B.V. An active antioxidant film was fabricated by using polyvinyl alcohol (PVA) and corn starch (ST) and incorporated with and 3, 5, 10, and 20% (v/v) coconut shell extract (CSE) and sepiolite clay (SP) for the first time. The microstructure and physical properties of prepared films were characterized and examined. The addition of sepiolite clay to PVA improved the elongation break 15.57 to 69.24%, and water vapor permeability increased with the addition of CSE. In addition to CSE to films, the antioxidant activity properties of the films were enhanced up to 80%. Further, increasing the amount of CSE slightly affected the color of the active films. The thermal properties of films were enhanced with the addition of CSE and SP. The dispersion of SP in the PVA matrix was affected by an increase in CSE concentration in the PVA matrix. Remarkably, PVA-ST film-based sachets successfully improved the oxidative stability of packaged soybean oil. A facile and environmentally-friendly strategy for increasing antioxidant activity is a crucial issue for value-added lignin and lignin-carbohydrate complex (LCC) as alternative antioxidants. However, the antioxidant activities of lignin and LCC by the traditional solid-liquid extraction (SLE) methods were restricted by the relatively lower solubility induced from high molecular weight (Mw), and the less functional groups including, phenolic hydroxyl and carboxyl. To improve the antioxidantion of lignin and LCC, lithium chloride/dimethyl sulfoxide (LiCl/DMSO) solvent fractionation (LDSF) was conducted to increase the functional groups and reduce Mw, in which LiCl/DMSO acted triple roles as solvent, acid, and metal chloride catalyst for the depolymerization reaction synchronously. The β-O-4' linkages were cleaved to release the phenolic hydroxyl, resulting in decreasing Mw; the hydroxyl of the side-chain of lignin was oxidized into carboxyl. Thus, the lignin (LD-RL) and LCC (LD-LCC) samples from LDSF had a higher syringyl (S)/guaiacyl (G) ratio, phenolic hydroxyl, and carboxyl contents, but less Mw than control groups from SLE. Consequently, they presented more excellent scavenging rates toward DPPH and ABTS radicals, up to 90%. In this study, an alkaline-catalyzed sulfolane/water solvent system was developed for isolating high-purity and antioxidative lignin from willow (Salix matsudana cv. Zhuliu). Optimization of the pretreatment conditions such as temperature, sulfolane/water ratio, and alkaline catalyst (NaOH) dosage were comprehensively investigated for effective lignin extraction from willow. The 44.4% of lignin was recovered from the biomass with 54% of delignification in 50/50 (w/w) sulfolane/water system at 170 °C. As the addition of the alkaline catalyst (NaOH) increased to 4%, the delignification yield was increased up to 94% with about 70% of lignin recovery yield. The recovered lignin was comparatively investigated with its control, milled wood lignin (MWL). The β-O-4 linkages and phenolic hydroxyl were well preserved in the extracted lignin fractions with the sulfolane/water system. Furthermore, excellent radical scavenging ability was observed with the extracted lignins by sulfolane/water pretreatments owing to rich phenolic hydroxyl groups in the lignins. Natural active compounds with antioxidant properties and other potential health benefits, like quercetin (Qu), have aroused wide concern for developing bio-functional products. However, their applications are hindered by their intrinsic poor water solubility and chemical instability. In this paper, a natural antioxidant alpha lipoic acid (ALA) was grafted onto chitosan (CS) to synthesize a novel graft polymer (CS-graft-ALA). In particular, this graft-polymer could self-assemble into spherical nanomicelles in water, with a low critical micelle concentration (CMC) of 0.0076 mg/mL. As a robust and active nanocarrier, the CS-graft-ALA micelles showed high efficiency in encapsulating Qu and dispersing Qu in water. As found, the antioxidant activity of Qu was effectively enhanced when entrapped within CS-graft-ALA micelles. Moreover, CS-graft-ALA micelles could significantly improve the photo-stability and temperature-stability of Qu. Various ferulic acid (FA) dimers occurring in plant cell walls, such as 8-5-, 8-O-4-, 5-5-, and 8-8- coupled dimers, are effective antioxidants and potential antimicrobials. It is necessary to access these diferulates as reference compounds to validate those isolated from plants. 3,6-bis(4-hydroxy-3-methoxyphenyl)-tetrahydrofuro-[3,4-c]furan-1,4-dione, a 8-8-coupled FA dilactone generated from ferulic acid via radical coupling, has been used to synthesize 8-8-coupled FA dimers although few reports investigated the distribution of products and mechanisms involved in the transformation of FA dilactone. In this work, the FA dilactone, obtained from FA by a peroxidase-catalyzed radical coupling, was reacted under various base/acid conditions. Effects of reaction conditions and workup procedures on the distribution of products were investigated by GC-MS. The isolated products from such treatments of FA dilactone were characterized by NMR. New derivatives of FA dimer including 2-(4-hydroxy-3-methoxybenzylidene)-3-(hydroxyl-(4-hydroxy-3-methoxyphenyl)methyl)succinic acid and 2-(bis(4-hydroxy-3-methoxyphenyl)-methyl)-succinic acid were produced from NaOH treatment. Another novel 8-8-coupled cyclic FA dimer, diethyl 6-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-7-methoxy-1,2-dihydronaphthalene-2,3-dicarboxylate was identified in products from FA dilactone treated by dry HCl in absolute ethanol. Puerarin, an important isoflavonoid from the edible root of Pueraria lobata, shows multiple bioactivities but suffers from low bioavailability. In this study, a new whole-cell catalytic method for acylation modification of puerarin was developed. Among the 12 strains tested, Aspergillus oryzae showed the highest catalytic activity and selectively catalyzed acylation of puerarin at the 6″-hydroxyl group. The organic solvents used significantly influenced the catalytic efficiency of the cells. In the green solvent 2-methyltetrahydrofuran, the reaction showed high substrate conversion (92.5%) and regioselectivity (95.8%), with results similar to those with tetrahydrofuran (94.2% and 98.5%, respectively) under optimal conditions. The monoester products showed higher liposolubility in comparison to puerarin, and those with C3-C8 fatty acid chain lengths showed evidently improved antioxidant activity toward erythrocyte hemolysis. Considering the operational stability of the cells and efficiency of the scaled-up reactions, this method is efficient and cost effective, with promising applications in the health food industry.

In this study, 11 dehydrodiferulic acids (DFAs) and 8 diethyl dehydrodiferulates (DEFs) were synthesized and evaluated by Trolox equivalent antioxidant capacity (TEAC) and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) assays for their antioxidant properties to understand the Structure-Antioxidant Activity Relationship (SAR) of these dehydrodiferulates. In both assays, the order of antioxidant activity for all tested ferulic acid dimers were consistent except for 3-(4-Hydroxy-3-methoxy-benzylidene)-5-(4-hydroxy-3-methoxy-phenyl)-3H-furan-2-one (2, 8-8-lactone DC DFA, not occurred naturally) being the best antioxidant by TEAC test. The order of antioxidant activity of diferulic acid ethyl esters, evaluated by both assays, was not consistent; however, TEAC and DPPH assays provided consistent results for certain set of ethyl diferulates. In this study most of dimeric ferulates, with three exceptions, showed higher radical-scavenging efficacy than the monomers. We report a new drug-loaded film for blending poly(L-lactide) (PLLA) with lignin-based functional filler, which enhanced stability and sustained release of trans-resveratrol (trans-RSV). In addition, trans-RSV loaded PLLA/lignin films showed good antioxidant and anticancer effects. Incorporation of the optimized amount of lignin-based filler to PLLA matrix was key to achieve these desired functions. The lignin-based filler (LG-g-PDLA) was synthesized via ring-opening polymerization on alklin lignin with D-lactide. Benifiting from the stereocomplex formation (PLLA and PDLA side chains), similar polyphenolic structures between lignin and trans-RSV, and UV chromophoric groups of lignin, trans-RSV loaded PLLA/lignin-graft-PDLA films (R/P/LGPD) have good performances on mechanical property, uniformity distribution of drug, and light barrier property. The release behaviors of trans-RSV could be controlled by variation of LG-g-PDLA content. The current study focuses on the analysis of in vitro biological activity of extract from bark of Norway spruce (Picea Abies), which can find potential application in food and cosmetic industry and pharmacology. Milled bark was subjected to Soxhlet extraction and supercritical fluid extraction to obtain two ethanol extracts. These extracts were further used to obtain their pre-extracts to n-hexane. It was investigated whether beta-sitosterol exhibits bacteriostatic activity necessary to observe antimicrobial and antifungal activity of methyl dehydroabiatate. This synergic effect and bacteriostatic activity of beta-sitosterol have not been previously reported. The greatest inhibition zone of n-hexane pre-extracts was confirmed in bacterium Pseudomonas aeruginosa (0,9 - 1,5 cm) and yeast Alternaria alternata (0,7 - 1,6 cm). Background: Flavonoid glycosides have many beneficial effects on health, but these bioactivities tend to decrease after oral administration owing to their poor lipophilicity. In this study, a facile whole-cell-based method was developed for selective preparation of monoester or diester of troxerutin, a flavonoid derivative. In addition, the bioavailabilities and antioxidant properties of troxerutin and its acylated derivatives were also investigated in cells., Results: Pseudomonas aeruginosa and Pseudomonas stutzeri cells showed high catalytic efficiency (substrate conversion > 90%) and different preferences for troxerutin, resulting in the production of its monoester (TME) and diester (TDE), respectively. The logP values of troxerutin, TME, and TDE were - 2.04 ± 0.10, - 0.75 ± 0.08, and 1.51 ± 0.05 and their P app values were 0.34 × 10 -6  ± 0.05, 0.99 × 10 -6  ± 0.12, and 1.54 × 10 -6  ± 0.17 cm/s, respectively. The results of hydroxyl radical, ABTS, and ORAC assays indicated that the antiradical activities of acylated derivatives did not exceed that of troxerutin, but showed higher inhibition effects upon 2,2'-azobis(2-amidinopropane) dihydrochloride-induced erythrocyte hemolysis than that of troxerutin (P < 0.05)., Conclusion: A facile and efficient whole-cell biocatalysis method was developed to synthesize troxerutin-acylated derivatives, markedly enhancing the bioavailability and antioxidant activities of troxerutin in cells. Additionally, the mechanism underlying the observed difference in the antioxidant activities of troxerutin and its esters was ascribed to both their free radical scavenging abilities and distribution on the cell membrane surface.

The papers have a porous structure which can be a suitable medium for the growth of bacteria. Therefore, in the sanitary papers, creation of a suitable antibacterial property is necessary. In this way, the plant extracts were prepared using solid-liquid extraction method from the Rosmarinus officinalis, Olea europaea, Mentha spicata and Punica granatum. Relatively, the total phenol and flavonoid contents were determined according to colorimetric method. Their antioxidant activity was evaluated using DPPH method. The extracts were sprayed on the surface of the handsheets, and then their antibacterial activity was investigated. The results showed that extracts from the P. granatum has the highest phenol (5.82%) and antioxidant activity (71.13%), while the extract from the R. officinalis had the highest flavonoid (4.23%) and antibacterial activity (90.43%). Hence, it can be concluded that these two extracts could be the suitable combinations to create desirable antibacterial properties in the manufacture of papers with sanitary purposes.

An effective method to functionalize chitosan with 4-hexyloxyphenol (HP) under homogeneous reaction conditions was developed using laccase as the catalyst. The resulting copolymer was characterized for chemical structure, grafted-HP content, surface morphology, thermal stability, antioxidant capacity, hydrophobic properties and tensile strength. Solid-state 13 C NMR spectrum confirmed the incorporation of HP onto chitosan. X-ray diffraction (XRD) showed a decrease in the degree of crystallinity for laccase/HP treated chitosan compared to pure chitosan. The grafted-HP content in laccase/HP-treated chitosan first increased and then declined with increase of the initial HP/chitosan ratio. A heterogeneous surface with spherical particles on the laccase/HP treated chitosan was observed by environmental scanning electron microscopy (ESEM) and scanning probe microscopy (SPM). The laccase/HP treatment of chitosan improved the thermal stability of copolymer. More significantly, the HP functionalized chitosan showed greatly improved ABTS + and DPPH radicals scavenging capacity, compared with pure chitosan. The hydrophobicity property of the HP functionalized chitosan also significantly increased although its tensile strength decreased. Tea polyphenols have received much attention from the pharmaceutical and food industries owing to their extraordinary antioxidant and antibacterial characteristics. However, tea polyphenols are very unstable in processing and storage, since they are sensitive to the environmental factors like temperature, light and pH. Therefore, the effective application of tea polyphenols requires a protective mechanism to maintain its activity. The utilization of compounded tea polyphenols, instead of raw materials, can potentially help to improve their stability. This review focuses on the summarization of the compounding technologies for tea polyphenols, including physical technologies, chemical-interfacial technologies and nano-scale compounding technologies. Softwood bark is an important source for producing chemicals and materials as well as bioenergy. Extraction is regarded as a key technology for obtaining chemicals in general, and valorizing bark as a source of such chemicals in particular. In this paper, properties of 237 compounds identified in various studies dealing with extraction of softwood bark were described. Esculin has many pharmacological effects, but these are difficult to observe after oral administration owing to poor lipid solubility. In our previous study, five acylated derivatives with different acyl chain lengths (EA, EP, EO, EL, and EM) were synthesized to improve the lipophilicity of esculin. In this study, the bioavailability and antioxidant activity of the five derivatives were investigated. The logP of esculin, EA, EP, EO, EL, and EM were -1.1 ± 0.1, -0.3 ± 0.14, 0.1 ± 0.17, 1.6 ± 0.09, 2.4 ± 0.11, and 2.8 ± 0.18, and their P app were 0.71 ± 0.02, 1.24 ± 0.18, 1.74 ± 0.11, 11.6 ± 3.6, 4.11 ± 1.03, and 2.64 ± 0.97 × 10 -6 cm/s, respectively. Besides, the bioavailability of EO, EL, and EM were seriously affected by carboxylesterase. The results of ABTS, ORAC, and DPPH assays indicated that the antiradical ability of the five derivatives did not exceed that of esculin. However, EA, EP, and EO showed more effective inhibition of AAPH-induced oxidative hemolysis than esculin did (p < 0.05), and EL and EM were less effective than esculin (p < 0.05). The mechanism was related to the distribution and localization of the derivatives in "oil-water interface" between the cytomembrane and the aqueous phase.

The heterogeneity of lignin chemical structure and molecular weight results in the lignin inhomogeneous properties which also covers the antioxidant performance. In order to evaluate the effects of lignin heterogeneity on its antioxidant activity, four lignin fractions from enzymatic hydrolysis lignin were classified by sequential organic solvent extraction and further evaluated by DPPH (1,1-Diphenyl-2-Picrylhydrazyl) free radical scavenging capacity and reducing power analysis. The characterization including FTIR, 1 H NMR and GPC showed that the fractionation process could effectively separate lignin fractions with distinctly different molecular weight and weaken the heterogeneity of unfractionated lignin. The antioxidant performance comparison of lignin fractions indicated that the dichloromethane fraction (F1) with lowest molecular weight (4585g/mol) and highest total phenolics content (246.13mg GAE/g) exhibited the highest antioxidant activity whose value was close to commercial antioxidant BHT (butylated hydroxytoluene). Moreover, the relationship between the antioxidant activity and the structure of lignin was further discussed to elucidate the mechanism of antioxidant activity improvement of lignin fractionation. Extracts from the wood of Juniperus excelsa ssp. polycarpos were analysed for their antioxidant activity using the DPPH method and compared with ascorbic acid and butylated hydroxytoluene. The most active extracts were analysed for their chemical composition using gas chromatography-mass spectrometry. Acetone extract was found to be moderately active as an antioxidant agent at 58.38%, which was lower than the value of vitamin C (98.56%) at the concentration of 14.20 mg/mL. The major components identified in the acetone extract as trimethylsilyl (TMS) derivatives were pimaric acid TMS (24.56%), followed by α-d-glucopyranoside,1,3,4,6-tetrakis-O-(TMS)-β-d-fructofuranosyl 2,3,4,6-tetrakis-O-(TMS) (21.39%), triflouromethyl-bis-(TMS)methyl ketone (9.32%), and cedrol (0.72%). The dissolved water:methanol (1:1 v/v) partitioned from acetone extract afforded 12 fractions; among them, the F9 fraction was found to have good antioxidant activity (88.49%) at the concentration of 14.20 mg/mL. The major compounds identified in F9 fraction were α-d-glucopyranoside, 1,3,4,6-tetrakis-O-(TMS) (20.22%) and trifluoromethyl-bis-(TMS)methyl ketone (5.10%).

For the first time, a pale amorphous coumarin derivative, 5-methoxyl aesculetin (MOA), was isolated from the dried bark of Fraxinus rhynchophylla Hance (Oleaceae). MOA modulates cytokine expression in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages, but the precise mechanisms are still not fully understood. We determined the effects of MOA on the production of inflammatory mediators and pro-inflammatory cytokines in the LPS-induced inflammatory responses of RAW 264.7 macrophages. MOA significantly inhibited the LPS-induced production of nitric oxide (NO), prostaglandin E₂ (PGE₂), tumor necrosis factor-α (TNF-α), interleukin-6, and interleukin-1β. It also effectively attenuated inducible nitric oxide (NO) synthase, cyclooxygenase-2, and TNF-α mRNA expression and significantly decreased the levels of intracellular reactive oxygen species. It inhibited phosphorylation of the extracellular signal-regulated kinase (ERK1/2), thus blocking nuclear translocation of activation protein (AP)-1. In a molecular docking study, MOA was shown to target the binding site of ERK via the formation of three hydrogen bonds with two residues of the kinase, which is sufficient for the inhibition of ERK. These results suggest that the anti-inflammatory effects of MOA in RAW 264.7 macrophages derive from its ability to block both the activation of mitogen-activated protein kinases (MAPKs) and one of their downstream transcription factors, activator protein-1 (AP-1). Our observations support the need for further research into MOA as a promising therapeutic agent in inflammatory diseases.

Unlabelled: Enteral immunomodulatory nutrition is considered as a promising therapy for the treatment of acute lung injury and acute respiratory distress syndrome (ALI/ARDS). However, there are still some divergences, and it is unclear whether this treatment should be recommended for patients with ALI/ARDS. Therefore, we conducted this systematic review and meta-analysis to assess the efficacy and safety of an enteral immunomodulatory diet on the clinical outcomes of ALI/ARDS patients., Methods: We retrieved potentially relevant clinical trials though electronic databases. All trials of enteral immunomodulatory diet for ALI/ARDS were included. Analyses of the overall all-cause mortality, 28-day ventilator-free days and 28-day intensive care unit (ICU) free days were conducted., Results: In total six controlled trials were evaluated. The pooled results did not show a significant reduction in the risk of all-cause mortality (M-H RR (the overall Mantel-Haenszel relative risk), 0.81 (95% CI, 0.50-1.31); p = 0.38; 6 trials, n = 717) in ALI/ARDS patients treated with the immunomodulatory diet. This treatment also did not extend the ventilator-free days and ICU-free days. However, patients with high mortality might benefit from this treatment., Conclusions: The enteral immunomodulatory diet could not reduce the severity of the patients with ALI/ARDS. Whereas, for ALI/ARDS patients with high mortality, this treatment might reduce the all-cause mortality, but its use should be treated with discretion.

To confer cellulose fibers antimicrobial and antioxidant activities, chitosan (CS)/lignosulfonates (LS) multilayers were constructed on fibers surfaces through layer-by-layer deposition technique. The formation of CS/LS multilayers on cellulose fibers surfaces was verified by X-ray photoelectron spectroscopy (XPS) and zeta potential measurement. The surface morphologies of CS/LS multilayers on fibers surfaces were observed by atomic force microscopy (AFM). The results showed that characteristic element (i.e. N and S element) content increased with increasing bilayers number, the surface LS content increased linearly as a function of bilayers. Zeta potential of modified fibers was inversed after deposition of each layer. AFM phase images indicated that the cellulose microfibrils on fibers surfaces were gradually covered by granular LS aggregate. The antimicrobial testing results demonstrated that CS/LS multilayers modified fibers with CS in the outermost layer exhibited higher antimicrobial activity against Escherichia coli. The antioxidant testing results showed that antioxidant activity of CS/LS multilayers modified fibers was better than that of original fibers under the same oxidation conditions., (Copyright © 2015 Elsevier Ltd. Oxidative stress-mediated cellular injury has been considered as a major cause of neurodegenerative diseases including Alzheimer and Parkinson diseases. The scavenging of reactive oxygen species (ROS) mediated by antioxidants may be a potential strategy for retarding the disease's progression. Macranthoin G (MCG), isolated from Eucommia ulmoides, is a derivative from chlorogenic acid methyl ester and caffeic acid. This study is aimed to investigate the protective role of MCG against the cytotoxicity induced by hydrogen peroxide (H2O2) and to elucidate potential protective mechanisms in rat pheochromocytoma (PC12) cells. The results showed that the treatment of PC12 cells with MCG prior to H2O2 exposure effectively increased the cell viability, and stabilized the mitochondria membrane potential (MMP); furthermore, it enhanced the antioxidant enzyme activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) and the levels of intracellular glutathione (GSH); it also decreased the malondialdehyde (MDA) content, intracellular ROS, caspase-3 activation, as well as cell apoptosis. In addition, the MCG treatment minimized the cell injury by H2O2 via down-regulation of the NF-κB pathway as well as activation of phosphorylation of IκBα, p38, and the extracellular signal-regulated kinase (ERK). The hydrolysate from duck egg white protein (DEWP) prepared by "SEEP-Alcalase" at degree of hydrolysis (DH) value of 21% (namely HSA21) exhibited high antioxidant capacity in different oxidation systems. A consecutive chromatographic method was then developed for separation and purification of HSA21, including ion-exchange chromatography, macroporous adsorption resin (MAR) and gel filter chromatography. The final peptides "P21-3-75-B" were obtained with significantly enhanced antioxidant activity (p<0.05). It was further confirmed that the product mainly consisted of five oligopeptides (Mr: 202.1, 294.1, 382.1, 426.3, and 514.4Da). Furthermore, the antioxidant activity of P21-3-75-B kept stable after in vitro digestive simulation. Antioxidant capacity of the purified peptides was closely related to the molecular mass, hydrophobic amino acid residues, acidic amino acid and some antioxidant amino acids. Biocatalytic hydrolysis reactions were designed for preparation of bioactive hydrolysate of duck egg white protein (DEWP) employing two enzymes in one pot. Firstly, the fresh DEWP was thermal treated at 95 °C, for 40 min at pH 10, to effectively deactivate enzyme inhibitors thus facilitating the following two-step enzymatic hydrolysis. Compared with single-enzyme processes, the two-step enzymatic procedures showed much higher reaction efficiency. The first enzymatic step (in the presence of Alcalase or hydrolase SEEP) allowed to hydrolyze DEWP with degree of hydrolysis (DH) of 8.8-10% and soluble peptide yield (SEP) of 60.5-70.2% in a short period (4 h). The second enzymatic step (in the presence of Trypsin or Alcalase) gave a further degradation of DEWP with DH and SEP being more than 26.2% and 90.4%, respectively. The final hydrolysates exhibited high antioxidant activity in an evident DH dependent manner. The hydrolysates achieved by sequential addition of the proteinase SEEP and Alcalase at DH value 21% gave the highest antioxidant activity, which was mainly ascribed to the changes in the amino acid compositions that the contents of some key amino acids and total hydrophobic amino acids were significantly improved by the enzymatic hydrolysis.

Oxidative stress has been considered as a major cause of cell damage in various neurodegenerative disorders. One of the reasonable strategies for delaying the disease's progression is to prevent reactive oxygen species (ROS) mediated cellular injury by dietary or pharmaceutical augmentation of free radical scavengers. Isocampneoside II (ICD) is an active phenylethanoid glycoside isolated from the medicinal hardwood genus Paulownia. This study was designed to explore free radical scavenging potential of ICD in different in vitro systems and its protective role in hydrogen peroxide (H₂O₂)-induced oxidative stress and apoptotic death in cultured rat pheochromocytoma (PC12) cells. The results showed ICD eliminated approximately 80.75% superoxide radical at the concentration of 0.1mg/ml and inhibited metal chelating by 22.07% at 8 mg/ml. Additionally, ICD showed a strong ability on reducing power and provided protection against oxidative protein damage induced by hydroxyl radicals. Pretreatment of PC12 cells with ICD prior to H₂O₂ exposure elevated cell viability, enhanced activity of superoxide dismutase and catalase, and decreased levels of malondialdehyde and intracellular ROS. Furthermore, ICD inhibited cell apoptosis and Bax/Bcl-2 ratio induced by H₂O₂. In this study, an attempt was made to prepare novel D-glucosamine hydrochloride-rectorite (DGH-REC) nanocomposites with improved antioxidant and anti-ultraviolet activity via the solution-intercalation method. The structure and morphology of DGH-REC nanocomposites were characterized by XRD, TEM, (13)C CP/MAS NMR, FT-IR, XPS and SEM. The results showed that the interlayer distance of REC was enlarged after intercalation of DGH; the largest value reached 11.76 nm, and in this case exfoliation of the REC layer was observed. Moreover, most strong crystals of DGH were greatly disrupted while two other weak crystals of DGH were stronger after intercalation of DGH into REC. More importantly, it was found that DGH-REC nanocomposites showed pronounced antioxidant activity, in contrast with DGH, and the nanocomposites had anti-ultraviolet capacity, which was not observed in DGH. Furthermore, the nanocomposites did not show apparent cytotoxicity. Therefore, DGH-REC nanocomposites have great potential in health or functional food application as compared to DGH.

Microwave-assisted extraction in organic acid aqueous solution (formic acid/acetic acid/water, 3/5/2, v/v/v) was applied to isolate lignin from bamboo. Additionally, the structural features of the extracted lignins were thoroughly investigated in terms of C₉ formula, molecular weight distribution, FT-IR, (1)H NMR and HSQC spectroscopy. It was found that with an increase in the severity of microwave-assisted extraction, there was an increase of phenolic hydroxyl content in the lignin. In addition, an increase of the severity resulted in a decrease of the bound carbohydrate content as well as molecular weight of the lignin. Antioxidant activity investigation indicated that the radical scavenging index of the extracted lignins (0.35-1.15) was higher than that of BHT (0.29) but lower than that of BHA (3.85). The results suggested that microwave-assisted organic acid extraction provides a promising way to prepare lignin from bamboo with good antioxidant activity for potential application in the food industry., (Copyright © 2012 Elsevier Ltd. Study on the EtOAc soluble fraction from the bark of Populus ussuriensis Kom. resulted in the isolation of three phenolic glycosides, including populoside (1), 7-O-ρ-coumaroylsalirepin (2) and 7-O-caffeoylsalirepin (3), among which 3 is a new compound. The structures of these compounds were elucidated on the basis of spectroscopic evidence. Phenolic glycosides 1-3 exhibited excellent antioxidant activity, evaluated in the ABTS⁺ radical scavenging assay.

The antioxidant activity of the phenolic compounds present in industrial black liquors obtained from the two cooking processes (kraft and sulphite) used in Portugal to produce Eucalyptus globulus pulp was evaluated. The black liquors treated at several pH values were extracted with ethyl acetate. Phenolic fractions were further separated by liquid chromatography of the crude extracts of kraft liquor at pH = 6 and sulphite liquor at the original pH. Total phenolic content was determined in terms of gallic acid equivalents (Folin-Ciocalteu colorimetric method), and the antioxidant activity in the crude extracts at several pH values and in the separated fractions was measured using the DPPH test for radical scavenging capacity. The total phenolic content of crude extracts and separated fractions ranged from 92.7 to 181.6 and from 91.6 to 1,099.6 mg GAE/g, respectively, while the antioxidant activity index (AAI) ranged from 2.20 to 3.41 and from 2.21 to 11.47 respectively, showing very strong antioxidant activity in all studied cases. The fractions separated by column chromatography were submitted to mass spectrometry analysis and the results were compared to others in the literature of natural products, mainly from Eucalyptus, and the characteristic bands of functional groups were identified by ¹H-NMR and FTIR. These methods allowed the identification of 17 phenolic compounds.

Recently we demonstrated that in inflammatory bowel disease (IBD) macrophage-oxidative burst activity is increased and NADPH oxidase mRNA is induced. The herbal phenylethanoid acteoside isolated from Plantago lanceolata L. was shown to exhibit anti-oxidative potential. Using the dextran sulphate sodium (DSS)-induced colitis model, in this study we have assessed whether systemic application of acteoside affects colitis. Colitis was induced by DSS in Balb/c mice. Treatment with acteoside (120, 600 microg/mouse/day) was performed intraperitoneally. The colon lengths were determined. Colonic tissue was scored histologically (max. score 8) by a blinded investigator. T cells isolated from mesenteric lymph nodes (MLN) were stimulated with anti-CD3 antibody in the presence of interleukin (IL)-2 (final concentration 10 U/ml). After incubation for 24 h, IL-1beta, IL-6, IL-12 tumour necrosis factor (TNF)-alpha and interferon (IFN)-gamma levels in supernatants were analysed by the beadlyte cytokine detection system. Histological scoring of colonic tissue revealed that application of acteoside was followed by a significantly improved histological score. In acute colitis the histological score was 3.2 with acteoside versus 5.2 with phosphate-buffered saline (PBS) (P < 0.02). In chronic colitis both 120 microg (3.3 versus 5.2) or 600 microg acteoside (3.0 versus 5.2) significantly ameliorated colitis (both P < 0.02). Stimulated MLN from mice with chronic DSS-induced colitis treated with acteoside showed a significant down-regulation of IFN-gamma secretion (195 pg/ml with 600 microg acteoside versus 612 pg/ml with PBS, P < 0.02). Inhibition of oxidative burst activity with acteoside reduced mucosal tissue damage in DSS colitis and could be a therapeutic alternative for IBD treatment. Further studies of this agent are warranted.

A sulfated emodin glucoside, emodin 8-O-beta-D-glucopyranosyl-6-O-sulfate (1), was isolated from the roots of Rheum emodi in an investigation of the active constituents of this Nepalese medicinal plant, and its structure was determined by spectroscopic and chemical methods. Additionally, two rare auronols, carpusin (2) and maesopsin (3), besides other anthraquinones and phenolics, were isolated and identified. Compounds 2 and 3 showed significant antioxidant activity in the DPPH assay, while chrysophanol, physcion, and emodin and their 8-O-glucosides were found to be inactive.
