Probing previously unknown neuropeptides and/or peptide hormones is essential for our understanding of the regulation of energy homeostasis in the brain. We recently performed a cDNA subtractive screening of the chicken hypothalamus, which contained one of the feeding and energy metabolic centers. We found a gene encoding a novel protein of 182 amino acid residues, including one putative small secretory protein of 80 amino acid residues. The C-terminal amino acids of the small protein were Gly-Leu-NH2, and as a result, the small protein was termed neurosecretory protein GL (NPGL). Subcutaneous and intracerebroventricular infusions of NPGL increased body mass gain in chicks, suggesting a central role for this protein in regulating growth and energy homeostasis. A database search revealed that the Npgl gene is conserved in vertebrates, including mice and rats. This review summarizes the advances in the characterization, localization, and biological action of NPGL, in birds and rodents.

Sea lampreys (Petromyzon marinus) are basal vertebrates that exhibit reproductive control via a hypothalamic-pituitary-gonadal axis. The function and evolution of the hypothalamic and pituitary peptide hormones are well studied in this species, whereas the functions of classical sex steroid hormones have not been well established due to their low or non-detectable plasma levels. Sea lamprey pheromone 3-keto petromyzonol sulfate (3kPZS) has been shown to increase while 3-keto allocholic acid (3kACA) decreases plasma 15α-hydroxyprogesterone (15αP) levels in prespermiating males (PSM) but not in preovulatory females (POF). However, spermiating male washings that contain both 3kPZS and 3kACA facilitate spawning in both sexes. Therefore, we wondered if the effects of pheromones on POF were elicited by classical steroid hormones such as progesterone, androstenedione, testosterone and estradiol. We hypothesized that waterborne 3kACA and 3kPZS differentially alter steroid hormone levels in prespawning sea lampreys. We determined the sex differences and pheromonal effects on steroid hormone levels in prespawning sea lampreys using sensitive ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) methods. Some PSM became spermiating (SM) at the time of sample collection, and those data were treated as a separate group. We found that males contained more androstenedione and progesterone in the gonad and more estradiol and testosterone in the plasma than POF, whereas POF contained more gonadal testosterone than males (no detectable gonadal testosterone). In POF, 3kPZS decreased gonadal androstenedione but increased gonadal progesterone and plasma estradiol, whereas 3kACA only increased gonadal progesterone levels. Exposure to 3kPZS for 4 h increased plasma 15αP in POF and SM, and gonadal 15αP in POF. Interestingly, 3kACA or 3kPZS depleted gonadal or plasma 15αP in PSM at various time points. On the other hand, both pheromones had no significant effect on androstenedione, progesterone or estradiol levels in males. Plasma testosterone levels did not change after pheromone exposure in both sexes. We conclude that sea lamprey pheromones 3kACA and 3kPZS induced differential steroidal responses in POF, PSM and SM.

Ghrelin has been implicated in the regulation of gastric functional development, and its physiological functions are mediated by Ghrelin-O-acyltransferase (GOAT) which is capable of generating the active form of this polypeptide hormone. However, whether and how ghrelin gene silencing may modify gastric acid secretion and GOAT-Ghrelin system is yet to be explored. Leptin has been well-established as a canonical anorexic peptide hormone in mammals, though much of its function in fish remains obscure. In this study, the cDNAs of two leptin isoforms (leptin-A and leptin-B) were cloned from the liver of a small cyprinid fish, Tanichthys albonubes. The two T. albonubes leptins, sharing low primary amino acid sequence homology with their mammalian counterparts, and between themselves, are highly conserved in three-dimensional protein structures and gene structures. Liver is a major source of leptin mRNA in T. albonubes with leptin-A being the dominant form. The expression of hepatic leptin-A but not leptin-B mRNA in female fish is significantly higher than in male fish. Transcriptional hepatic levels of leptin-A and leptin-B in both male and female fish were demonstrated to increase after long-term fasting (10-25days) but decline upon re-feeding (3days). Strikingly, estrogen (E2) administration induced only leptin-A but not leptin-B hepatic mRNA expression in both male and female fish. Our study here provides the first evidence for differential regulation of two leptins in fish, and sheds new light on the possible origin of leptin in lower vertebrates.

Hormones can mediate suites of correlated traits. Melanocortins regulate melanin synthesis and elements of the melanocortin system can directly, and indirectly, affect a number of other traits, such as stress reactivity. Trait correlations within the melanocortin system have been studied mainly in birds and mammals but less so in reptiles. We examined adult male western fence lizards (Sceloporus occidentalis) and if melanization was correlated with plasma levels of three hormones, including peptide hormone α-melanocyte stimulating hormone (α-MSH), testosterone and corticosterone, and ectoparasite loads. This lizard is darker at higher elevations in California, and we compared five high-elevation and four low-elevation populations during comparable periods of the breeding season at each site. We first validated use of an α-MSH assay kit with lizard plasma. Since Anolis carolinensis is one of the few species with published values for α-MSH plasma levels, we assayed both Anolis and Sceloporus plasma and compared hormone values to those we generated for Anolis to the publish values. We also evaluated effects of different methods of storing spiked plasma pools on resulting α-MSH concentrations. Plasma levels of α-MSH did not differ significantly, but some populations differed significantly in mean corticosterone and mean testosterone. Combining all individuals from the nine populations, we found that individual variation in α-MSH was not associated with individual variation in melanization, but levels of α-MSH were positively associated with plasma testosterone and negatively associated with corticosterone. The lack of association between individual levels of melanization and expression of most other traits differs from a growing number of within-population studies of melanization, and we discuss what differences in physiological mechanisms could produce different hypothetical patterns. Circulating levels of -MSH are only one element of the melanocortin system; in situ synthesis of α-MSH by the skin and the diversity of melanocortin receptors could also contribute to variation in traits mediated by the melanocortin system and should be examined.

Tardigrades are a key group for understanding the evolution of the Ecdysozoa, a large clade of molting animals that also includes arthropods and nematodes. However, little is known about most aspects of their basic biology. Neuropeptide and peptide hormone signaling has been extensively studied in arthropods and nematodes (particularly regarding their roles in molting in arthropods), but very little is known about neuropeptide signaling in other ecdysozoans. In this work, different strategies were used to search for neuropeptide and peptide hormone precursors in the genomes of the tardigrades Hypsibius dujardini and Ramazzottius varieornatus. In general, there is a remarkable similarity in the complement of neuropeptides and their sequences between tardigrades and arthropods. The precursors found in tardigrades included homologs of achatin, allatostatins A, B and C, allatotropin, calcitonin, CCHamide, CCRFa, corazonin, crustacean cardioactive peptide, diuretic hormone 31, diuretic hormone 44, ecdysis triggering hormone, eclosion hormone, gonadotropin-releasing hormone (GnRH), GSEFLamide, insulin-like peptides, ion transport peptide, kinin, neuropeptide F, orcokinin, pigment dispersing hormone, proctolin, pyrokinin, RYamide, short neuropeptide F, sulfakinin, tachykinin, trissin and vasopressin. In most cases, homologs of known cognate receptors for each neuropeptide family could only be identified when the precursors were also present in the genome, further supporting their identification. Some neuropeptide precursor genes have undergone several duplications in tardigrades, including allatostatin A and C, corazonin, GnRH, eclosion hormone, sulfakinin and trissin. Furthermore, four novel families of candidate neuropeptide precursors were identified (two of which could also be found in several arthropods). To the best of my knowledge, this work represents the first genome-wide search for neuropeptide precursors in any ecdysozoan species outside arthropods and nematodes, and is a necessary first step towards understanding neuropeptide function in tardigrades.

Ghrelin and leptin are key peripherally secreted appetite-regulating hormones in vertebrates. Here we consider the ghrelin gene (GHRL) of birds (class Aves), where it has been reported that ghrelin inhibits rather than augments feeding. Thirty-one bird species were compared, revealing that most species harbour a functional copy of GHRL and the coding region for its derived peptides ghrelin and obestatin. We provide evidence for loss of GHRL in saker and peregrine falcons, and this is likely to result from the insertion of an ERVK retrotransposon in intron 0. We hypothesise that the loss of anorexigenic ghrelin is a predatory adaptation that results in increased food-seeking behaviour and feeding in falcons.

In teleosts, as their names suggest, the main target cells of melanocyte-stimulating hormone (MSH) and melanin-concentrating hormone (MCH) are the chromatophores in the skin, where these peptide hormones play opposing roles in regulating pigment migration. These effects are obvious especially when their activities are examined in vitro. On the contrary, while MCH also exhibits activity in vivo, MSH does not always stimulate pigment dispersion in vivo because of predominant sympathetic nervous system. A series of our investigations indicates that this is also the case in barfin flounder, Verasper moseri. Interestingly, we observed that mch expression and the tissue contents of MCH can be easily influenced by changes in environmental color conditions, while gene expression and tissue contents related to MSH scarcely respond to color changes. Transcripts of MSH and MCH receptor genes have been identified in a variety of tissues of this fish species, suggesting that these are multifunctional peptide hormones. Nevertheless, chromatophores in the skin still offer important clues in the efforts to elucidate the functions of melanotropic peptides. Herein, we review the most recent advancements of our studies on MSH and MCH and their receptors in the barfin flounder and discuss the interrelations between these peptides, focusing on their roles in influencing pigment migration in the skin.

Knowledge about the endocrine regulation of energy balance in fish is of interest for basic as well as aquaculture research. Ghrelin is a peptide hormone that was first identified in fish 10 years ago and has important roles in the control of food intake and metabolism. Both ghrelin and its receptor, the growth hormone secretagogue receptor (GHS-R), have been found in numerous fish species. Their tissue distributions support the idea that ghrelin has an integrative role in the regulation of energy balance at both the central nervous system level and systemic level. In tilapia and goldfish, ghrelin treatment appears to increase food intake and to stimulate lipogenesis and tissue fat deposition to promote a more positive energy status. In rainbow trout, on the other hand, ghrelin decreases food intake. Goldfish and rainbow trout are the fish species in which the mode of action of ghrelin on food intake has been most thoroughly investigated. The results from these studies indicate that ghrelin alters food intake by acting on well-known appetite signals, such as CRH, NPY and orexin, in the hypothalamus in a species-specific manner. In goldfish, sensory fibres of the vagus nerve convey the signal from gut-derived ghrelin to modulate appetite. The data also indicate that ghrelin may modulate foraging/swimming activity and the perception of food in fish. Results related to the effects of energy status, temperature, and stressors on plasma ghrelin/tissue ghrelin mRNA levels are occasionally inconsistent between short- and long-term studies, between the protein and mRNA, and between species. Recent data also imply a role of ghrelin in carbohydrate metabolism. More functional studies are required to understand the role of ghrelin and its mechanisms of action in the regulation of energy balance among fish.

Apart from providing an up-to-date review of the literature, considerable emphasis was placed in this article on the historical development of the field of "crustacean eyestalk hormones". A role of the neurosecretory eyestalk structures of crustaceans in endocrine regulation was recognized about 80 years ago, but it took another half a century until the first peptide hormones were identified. Following the identification of crustacean hyperglycaemic hormone (CHH) and moult-inhibiting hormone (MIH), a large number of homologous peptides have been identified to this date. They comprise a family of multifunctional peptides which can be divided, according to sequences and precursor structure, into two subfamilies, type-I and -II. Recent results on peptide sequences, structure of genes and precursors are described here. The best studied biological activities include metabolic control, moulting, gonad maturation, ionic and osmotic regulation and methyl farnesoate synthesis in mandibular glands. Accordingly, the names CHH, MIH, and GIH/VIH (gonad/vitellogenesis-inhibiting hormone), MOIH (mandibular organ-inhibiting hormone) were coined. The identification of ITP (ion transport peptide) in insects showed, for the first time, that CHH-family peptides are not restricted to crustaceans, and data mining has recently inferred their occurrence in other ecdysozoan clades as well. The long-held tenet of exclusive association with the eyestalk X-organ-sinus gland tract has been challenged by the finding of several extra nervous system sites of expression of CHH-family peptides. Concerning mode of action and the question of target tissues, second messenger mechanisms are discussed, as well as binding sites and receptors. Future challenges are highlighted.

Arginine vasotocin (AVT) and isotocin (IT) are two neurohypophysial peptide hormones for which a role in adaptation to environmental changes has been suggested in fish. In teleosts, there are only a few available studies about circadian changes of AVT and IT levels, and a role of those peptides in the circadian system has been mainly suggested on the basis of the role of the homologous hormone AVP in mammals. Herein, we evaluated the diurnal rhythms in plasma AVT, pituitary AVT and IT content and the hypothalamic pro-vasotocin (pro-VT) expression in rainbow trout kept under a natural photoperiod, as well as their persistence in constant darkness as a tool for defining circadian dependence. Trout kept under a natural light cycle showed clear diurnal rhythms in both circulating and pituitary AVT levels with peak values around the last hours of the light phase. Hypothalamic pro-VT mRNA was also rhythmically expressed with similar peak characteristics. These rhythms persisted in fish kept under constant darkness for nearly two consecutive days, although peaks were progressively attenuated and phase-advanced. An IT rhythm was also found in pituitary of the trout maintained under a natural photoperiod, but not in those kept under continuous darkness. These results suggest that rhythms of hypothalamic AVT synthesis might be regulated by endogenous circadian mechanisms, and these rhythms contribute to maintain a similar fluctuation in pituitary AVT secretion into the blood. A potential role for AVT in the circadian and seasonal time-keeping system of teleost fish, either as a component of the neural machinery that participates in the adaptation to cyclic environmental changes, or as a circadian/seasonal output signal, is also discussed.

A decade has passed since the peptide hormone ghrelin was first discovered in rat stomach. During this period, ghrelin has been identified not only in other mammals but also in fish, amphibians, reptiles and birds, and its physiological functions have been widely investigated. Avian ghrelin was first identified in chickens in 2002 and to date, the amino acid sequences of six different avian ghrelin peptides have been reported. In mammals, ghrelin is the only known gut-derived hormone to stimulate food intake when administered centrally or peripherally. In studies on chickens and quail, however, ghrelin inhibits food intake when injected centrally, while the effects on feeding behavior elicited by ghrelin injected peripherally are equivocal. This review summarizes what is currently known about the regulation of food intake and energy balance by ghrelin in birds.

The present study was conducted to establish a homologous radioimmunoassay (RIA) for quantifying plasma leptin (Lep) levels in salmonid species, and to study Lep levels in relation to nutritional status. A part of the Lep peptide, a 14 amino acid long sequence, identical between a Salmo and an Oncorhynchus species was synthesised. Polyclonal antibodies were raised in rabbit against this antigen and both were subsequently used in the development of a RIA protocol for assessing plasma Lep levels. The limit of detection of the assay was 0.3 nM, and intra- and interassay coefficient of variation (CV) were 8.4% and 13%, respectively. Apart from Atlantic salmon and rainbow trout, the assay exhibits measuring parallelism for a range of fish species, including arctic char, Atlantic cod and turbot, suggesting that the established RIA is useful for quantifying Lep levels in several fish species. The RIA indicates that Lep is found in salmonid plasma at levels of 0.5-5 nM, which is comparable with other peptide hormones, and well within the measuring range of the RIA. A study of fed and fasted rainbow trout showed elevated plasma Lep levels during fasting. In addition there was no correlation between Lep levels and condition factor. These data suggest that the relation between circulating Lep levels and energy status differs from that in mammals. While Lep is linked to energy balance, it may not act as an adiposity signal in salmonids, possibly pointing to functional divergence among ectothermic and endothermic vertebrates.

Key peptide hormones involved in the control of appetite in vertebrates were identified, their genes characterized and their regulation studied in Atlantic salmon: two agouti-related proteins (AgRP), cocaine- and amphetamine-regulated transcript (CART) and neuropeptide Y (NPY). The AgRP-1 and AgRP-2 genes encode prepro-proteins of 142- and 117-amino acids, respectively. The deduced AgRP-2 protein has 10 cysteine residues in the C-terminal polycysteine domain, while the AgRP-1 lacks the 6th and 7th cysteine residues observed in other species. AgRP-1 was principally expressed in the pituitary and skin, while AgRP-2 was highly expressed in the mid-gut, red muscle and gonads. The CART gene, encoding 118-amino acids, was strongly expressed in the brain and eye. In addition to salmon CART, we identified three to six variants of the CART gene in lower vertebrates by mining available databases. The salmon NPY gene, encoding 100-amino acids, was mainly expressed in the brain and eye. AgRP-1 and CART mRNA levels in the brain decreased after 6 days of fasting while AgRP-2 and NPY showed no significant change, suggesting that AgRP-1 and CART are involved in feeding regulation in Atlantic salmon. The identification of multiple variants of these appetite-regulating genes emphasizes the importance to further investigate the complex regulation of these genes.

In fish, pituitary growth hormone family peptide hormones (growth hormone, GH; prolactin, PRL; somatolactin, SL) regulate essential physiological functions including osmoregulation, growth, and metabolism. Teleost GH family hormones have both differential and overlapping effects, which are mediated by plasma membrane receptors. A PRL receptor (PRLR) and two putative GH receptors (GHR1 and GHR2) have been identified in several teleost species. Recent phylogenetic analyses and binding studies suggest that GHR1 is a receptor for SL. However, no studies have compared the tissue distribution and physiological regulation of all three receptors. We sequenced GHR2 from the liver of the Mozambique tilapia (Oreochromis mossambicus), developed quantitative real-time PCR assays for the three receptors, and assessed their tissue distribution and regulation by salinity and fasting. PRLR was highly expressed in the gill, kidney, and intestine, consistent with the osmoregulatory functions of PRL. PRLR expression was very low in the liver. GHR2 was most highly expressed in the muscle, followed by heart, testis, and liver, consistent with this being a GH receptor with functions in growth and metabolism. GHR1 was most highly expressed in fat, liver, and muscle, suggesting a metabolic function. GHR1 expression was also high in skin, consistent with a function of SL in chromatophore regulation. These findings support the hypothesis that GHR1 is a receptor for SL. In a comparison of freshwater (FW)- and seawater (SW)-adapted tilapia, plasma PRL was strongly elevated in FW, whereas plasma GH was slightly elevated in SW. PRLR expression was reduced in the gill in SW, consistent with PRL's function in freshwater adaptation. GHR2 was elevated in the kidney in FW, and correlated negatively with plasma GH, whereas GHR1 was elevated in the gill in SW. Plasma IGF-I, but not GH, was reduced by 4 weeks of fasting. Transcript levels of GHR1 and GHR2 were elevated by fasting in the muscle. However, liver levels of GHR1 and GHR2 transcripts, and liver and muscle levels of IGF-I transcripts were unaffected by fasting. These results clearly indicate tissue specific expression and differential physiological regulation of GH family receptors in the tilapia.

Insect ecdysis sequence is composed of pre-ecdysis, ecdysis and post-ecdysis behaviors controlled by a complex cascade of peptide hormones from endocrine Inka cells and neuropeptides in the central nervous system (CNS). Inka cells produce pre-ecdysis and ecdysis triggering hormones (ETH) which activate the ecdysis sequence through receptor-mediated actions on specific neurons in the CNS. Multiple experimental approaches have been used to determine mechanisms of ETH expression and release from Inka cells and its action on the CNS of moths and flies. During the preparatory phase 1-2 days prior to ecdysis, high ecdysteroid levels induce expression of ETH receptors in the CNS and increased ETH production in Inka cells, which coincides with expression of nuclear ecdysone receptor (EcR) and transcription factor cryptocephal (CRC). However, high ecdysteroid levels prevent ETH release from Inka cells. Acquisition of Inka cell competence to release ETH requires decline of ecdysteroid levels and beta-FTZ-F1 expression few hours prior to ecdysis. The behavioral phase is initiated by ETH secretion into the hemolymph, which is controlled by two brain neuropeptides-corazonin and eclosion hormone (EH). Corazonin acts on its receptor in Inka cells to elicit low level ETH secretion and initiation of pre-ecdysis, while EH induces cGMP-mediated ETH depletion and consequent activation of ecdysis. The activation of both behaviors is accomplished by ETH action on central neurons expressing ETH receptors A and B (ETHR-A and B). These neurons produce numerous excitatory or inhibitory neuropeptides which initiate or terminate different phases of the ecdysis sequence. Our data indicate that insect ecdysis is a very complex process characterized by two principal steps: (1) ecdysteroid-induced expression of receptors and transcription factors in the CNS and Inka cells. (2) Release and interaction of Inka cell peptide hormones and multiple central neuropeptides to control consecutive phases of the ecdysis sequence.

Insulin is an extensively studied peptide hormone in mammals. However, insulin is not restricted to vertebrates, but has also been identified in invertebrates, among whom several insect species. These insulin-like peptides (ILPs) show structural and-at least some-functional homology with mammalian insulin and act through a conserved pathway. Yet many aspects of insulin function in insects remain to be unveiled. We analyzed the presence of ILPs in the cotton leafworm, Spodoptera littoralis, at two levels: (1) cellular localization of ILPs in whole tissues of the central nervous system from S. littoralis, and (2) detection and identification of ILPs at nucleotide level. To our knowledge, nothing about the presence of ILPs in S. littoralis has been described so far. By whole mount in situ immunolocalization, we localized bombyxin-like material in S. littoralis in four pairs of pars intercerebralis cells and in the corpus cardiacum-corpus allatum complexes. In addition, we have cloned two different S. littoralis ILP precursor cDNAs by a combination of PCR and RAcE. The corresponding precursor polypeptides ('Sl-ILPP1' and 'Sl-ILPP2') show significant sequence homology with precursors for bombyxin and other bombyxin-related peptides. Chicken growth hormone secretagogue receptor (GHSR) is a receptor for ghrelin (GHRL), a peptide hormone produced by chicken proventriculus, which stimulates growth hormone (GH) release and food intake. The purpose of this study was to search for single nucleotide polymorphisms (SNPs) in exon 2 of GHSR gene and to analyze their effect on the appetite, growth traits and expression levels of GHSR, GHRL, and GH genes as well as serum levels of GH and GHRL in Mandara chicken. Two adjacent SNPs, A239G and G244A, were detected in exon 2 of GHSR gene. G244A SNP was non-synonymous mutation and led to replacement of lysine amino acid (aa) by arginine aa, while A239G SNP was synonymous mutation. The combined genotypes of A239G and G244A SNPs produced three haplotypes; GG/GG, GG/AG, AG/AG, which associated significantly (P0.05) with growth traits (body weight, average daily gain, shank length, keel length, chest circumference) at age from4 to 16w. Chickens with the homozygous GG/GG haplotype showed higher growth performance than other chickens. The two SNPs were also correlated with mRNA levels of GHSR and GH (in pituitary gland), and GHRL (in proventriculus and hypothalamus) as well as with serum level of GH and GHRL. Also, chickens with GG/GG haplotype showed higher mRNA and serum levels. This is the first study to demonstrate that SNPs in GHSR can increase appetite, growth traits, expression and level of GHRL, suggesting a hunger signal role for endogenous GHRL.

Ascidians are the closest phylogenetic neighbors to vertebrates and are believed to conserve the evolutionary origin in chordates of the endocrine, neuroendocrine, and nervous systems involving neuropeptides and peptide hormones. Ciona intestinalis harbors various homologs or prototypes of vertebrate neuropeptides and peptide hormones including gonadotropin-releasing hormones (GnRHs), tachykinins (TKs), and calcitonin, as well as Ciona-specific neuropeptides such as Ciona vasopressin, LF, and YFV/L peptides. Moreover, molecular and functional studies on Ciona tachykinin (Ci-TK) have revealed the novel molecular mechanism of inducing oocyte growth via up-regulation of vitellogenesis-associated protease activity, which is expected to be conserved in vertebrates. Furthermore, a series of studies on Ciona GnRH receptor paralogs have verified the species-specific regulation of GnRHergic signaling including unique signaling control via heterodimerization among multiple GnRH receptors. These findings confirm the remarkable significance of ascidians in investigations of the evolutionary processes of the peptidergic systems in chordates, leading to the promising advance in the research on Ciona peptides in the next stage based on the recent development of emerging technologies including genome-editing techniques, peptidomics-based multi-color staining, machine-learning prediction, and next-generation sequencing. These technologies and bioinformatic integration of the resultant "multi-omics" data will provide unprecedented insights into the comprehensive understanding of molecular and functional regulatory mechanisms of the Ciona peptides, and will eventually enable the exploration of both conserved and diversified endocrine, neuroendocrine, and nervous systems in the evolutionary lineage of chordates.

Mastotermes darwiniensis is the most basal living member of the Isoptera (termites), yet it exhibits an extremely advanced level of eusocial organization. Given the interest in, and the high levels of differential developmental and behavioral control needed for, eusociality, it is surprising that essentially nothing is known about the native peptides of M. darwiniensis , which undoubtedly represent the largest and most diverse class of hormones present in this species. The recent public deposition of a 100,000 + -sequence transcriptome for M. darwiniensis provides a means for peptide discovery in this termite. Here, this resource was mined for putative peptide-encoding transcripts via the BLAST algorithm tblastn and known arthropod neuropeptide precursor queries; mature peptide structures were predicted from the deduced pre/preprohormones using a well-vetted bioinformatics workflow. Thirty-four M. darwiniensis peptide-encoding transcripts were identified, with 163 distinct mature peptides predicted from these sequences. These peptides included members of the adipokinetic hormone, adipokinetic hormone-corazonin-like peptide, allatostatin A, allatostatin C, allatotropin, bursicon β, CCHamide, corazonin, crustacean cardioactive peptide, crustacean hyperglycemic hormone/ion transport peptide, diuretic hormone 31, diuretic hormone 44, FMRFamide-like peptide, insulin-like peptide, leucokinin, myosuppressin, neuroparsin, neuropeptide F, orcokinin, pigment dispersing hormone, pyrokinin, RYamide, short neuropeptide F, SIFamide, sulfakinin and tachykinin-related peptide families. This peptidome is the largest thus far predicted for any member of the Isoptera, and provides a foundation for initiating studies of peptidergic signaling in this and other termites, including ones directed at understanding the roles peptide hormones play in the developmental and behavioral control required for eusociality.

In this study, we established a radioimmunoassay (RIA) specific for ghrelin from the bullfrog Rana catesbeiana using a novel antibody raised against the C-terminal amino acid sequence of bullfrog ghrelin [13-28]. We also examined the distribution of ghrelin-producing cells in the stomachs of bullfrogs using this antibody and a cRNA probe specific for the bullfrog ghrelin gene. Ghrelin levels in plasma and stomach extracts were approximately 150 fmol/ml and 83-135 fmol/mg wet tissue, respectively. Reverse-phase high performance liquid chromatographic analysis, combined with bullfrog ghrelin RIA, revealed that ghrelin immunoreactivity in the stomach was composed of non-acylated ghrelin (des-acyl ghrelin) and several acylated forms of ghrelin bearing different fatty acid modifications, which could induce increases in intracellular Ca2+ in cells expressing the rat GH secretagogue receptor. In the stomach, the major storage form was acylated ghrelin. In bullfrog plasma, however, the majority of ghrelin immunoreactivity was des-acyl ghrelin and C-terminal fragments of frog ghrelin. Acylated ghrelin forms comprised only minor peaks. Ghrelin-immunopositive and ghrelin mRNA-expressing cells were observed within the mucosal layer of the stomach. Following starvation, significant increases in plasma ghrelin levels and stomach ghrelin mRNA levels were observed as early as 10 days after starvation. These results indicate that ghrelin is present in the stomach and plasma of the bullfrog, which can be detected with our novel antibody. Interestingly, the primary storage form of ghrelin in the stomach differed from the circulating form dominating in the plasma. Furthermore, increases in ghrelin levels in plasma and mRNA levels in the stomach after starvation suggest the possible involvement of ghrelin in energy homeostasis in the bullfrog.

In fish, the peptide hormones cholecystokinin (CCK) and peptide Y (PY) may be involved in pancreatic exocrine secretion, as found with mammalian CCK and peptide YY (PYY); CCK stimulates, whereas PYY inhibits, pancreatic exocrine secretion in mammals. However, there is very little information on these hormones in fish; in particular, the function of PY is still unknown. Therefore, as a first step for understanding the role of CCK and PY in regulating pancreatic exocrine in fish, the cDNAs of CCK and PY were cloned from the digestive tract of yellowtail (Seriola quinqueradiata). The peptide sequence of yellowtail CCK-8, DYLGWMDF, is identical to sequences found in several teleosts. The mature form of yellowtail PY consists of 36 amino acids and has high identity to other fish PYs (88.9-97.2%). Real-time quantitative RT-PCR assays were developed to measure yellowtail CCK and PY mRNA levels. CCK mRNA levels were extremely high in the brain and, among the digestive organs, high concentrations were found in the pyloric caeca and anterior intestine. PY mRNA levels were low in the brain and highest in the anterior intestine. In fasting experiments, mRNA levels of CCK and PY in the anterior intestine showed an antagonistic change after fasting; CCK decreased whereas PY increased. These data suggest that CCK and PY in yellowtail may relate to digestion including, enzyme secretion.

Ghrelin, a peptide hormone produced by the stomach in mammals, stimulates growth hormone release and food intake. Recently, ghrelin was identified and characterized in chicken proventriculus and shown to stimulate growth hormone release but inhibit feed intake. The purpose of this work was to identify and further characterize the ghrelin gene in chickens and in turkeys. Using molecular cloning techniques we have sequenced cDNAs corresponding to chicken (White Leghorn) and turkey ghrelin mRNAs. A total of 844 (chicken) or 869 (turkey) bases including the complete coding regions (CDS), and the 5'- and 3'-untranslated regions (UTRs) were determined. Nucleotide sequence (CDS) predicted a 116 amino acid precursor protein (preproghrelin) for both the chicken and the turkey that demonstrated complete conservation of an N-terminal 'active core' (GSSF) including a serine (position 3 of the mature hormone) known to be a modification (acylation) site important for ghrelin bioactivity. Additional nucleotide sequence was found in the 5'-UTRs of both Leghorn and turkey cDNAs that was not present in broilers or the red jungle fowl. The turkey ghrelin gene, sequenced from genomic DNA templates, contained five exons and four introns, a structure similar to mammalian and chicken ghrelin genes. Ghrelin was highly expressed in proventriculus with much lower levels of expression in other tissues such as pancreas, brain, and intestine. RT-PCR was used to quantify ghrelin mRNA levels relative to 18S rRNA in 3-week-old male broiler chickens. The level of ghrelin mRNA increased in proventriculus in response to fasting but did not decline with subsequent refeeding. Plasma ghrelin levels did not change significantly in response to fasting or refeeding and did not appear to reflect changes in proventriculus ghrelin mRNA levels. Ghrelin mRNA levels declined in broiler pancreas after a 48 h fast and increased upon refeeding. Expression of the gene encoding the receptor for ghrelin (growth hormone secretagogue receptor, GHS-R) and a variant form was detected in a variety of tissues collected from 3-week-old male broiler chickens possibly suggesting autocrine/paracrine effects. These results offer new information about the avian ghrelin and ghrelin receptor genes and the potential role that this system might play in regulating feed intake and energy balance in poultry.

A cDNA that codes for the polypeptide hormone precursor proopiomelanocortin (POMC) was cloned and sequenced from a gar (Lepisosteus osseus) pituitary cDNA library. The gar POMC cDNA is 1237 bp and contains a 780-bp open reading frame. The deduced amino acid sequence for gar POMC is 259 amino acids in length. The general organization of gar POMC is very similar to that of other gnathostome POMC sequences. The beta-endorphin sequence had 91% sequence identity with sockeye A beta-endorphin and 71% sequence identity with Xenopus laevis beta-endorphin. Three melanocyte-stimulating hormone (MSH) core sequences [HFR(W)] were detected. The gar alpha-MSH sequence was identical to the alpha-MSH sequence in rat POMC. The gar beta-MSH sequence had 77% sequence identity with salmonid forms of beta-MSH and 53% sequence identity with tetrapod forms of beta-MSH. The gamma-MSH region of gar POMC only had 26% primary sequence identity with tetrapod gamma-MSH sequences. Gar gamma-MSH had an incomplete MSH core sequence (HRF), an apparent internal deletion of five amino acids, and lacked flanking paired basic amino acids essential for proteolytic cleavage. The apparent degenerate nature of gar gamma-MSH is discussed in light of the absence of this sequence in salmonid fish.

The copepod Calanus finmarchicus is the most abundant zooplankton species in the North Atlantic. While the life history of this crustacean is well studied, little is known about its peptidergic signaling systems despite the fact that these pathways are undoubtedly important components of its physiological/behavioral control systems. Here we have generated and used a de novo assembled transcriptome for C. finmarchicus (206,041 sequences in total) to identify peptide precursor proteins and receptors. Using known protein queries, 34 transcripts encoding peptide preprohormones and 18 encoding peptide receptors were identified. Using a combination of online software programs and homology to known arthropod isoforms, 148 mature peptides were predicted from the deduced precursors, including members of the allatostatin-A, allatostatin-B, allatostatin-C, bursicon, crustacean cardioactive peptide (CCAP), crustacean hyperglycemic hormone, diuretic hormone 31 (DH31), diuretic hormone 44 (DH44), FMRFamide-like peptide (myosuppressin, neuropeptide F [NPF] and extended FL/IRFamide subfamilies), leucokinin, neuroparsin, orcokinin, orcomyotropin, periviscerokinin, RYamide and tachykinin-related peptide (TRP) families. The identified receptors included ones for allatostatin-A, allatostatin-C, bursicon, CCAP, DH31, DH44, ecdysis-triggering hormone, NPF, short NPF, FMRFamide, insulin-like peptide, leucokinin, periviscerokinin, pigment dispersing hormone, and TRP. Developmental profiling of the identified transcripts in embryos, early nauplii, late nauplii, early copepodites, late copepodites, and adult females was also undertaken, with all showing the highest expression levels in the naupliar and copepodite stages. Collectively, these data radically expand the catalog of known C. finmarchicus peptidergic signaling proteins and provide a foundation for experiments directed at understanding the physiological roles served by them in this species.

Because of the high degree of primary sequence conservation in neuropeptides and low molecular weight polypeptide hormones, these polypeptides are not useful for constructing phylogenetic trees based on maximum parsimony analysis. This review focuses on the organization of neuropeptide and polypeptide hormone precursors and discusses strategies for aligning polypeptide precursors for phylogenetic analysis. Examples are provided to support the hypothesis that some neuropeptide and polypeptide precursors and some high molecular weight polypeptide hormones can be used as data sets for resolving deep divergences among vertebrate taxa.

Profiles of testosterone, 11-ketotestosterone, androstenedione, and estradiol were determined by RIA, and immunocytochemical techniques were employed to identify gonadotropin (GTH) I and II and gonadotropin releasing hormone (GnRH) in monosex and mixed sex populations of rainbow trout from 1 to 126 days postfertilization (dpf). Steroid levels were relatively high at 1 dpf and declined until 25 dpf. At 30 and 48 dpf (hatching) steroid levels increased slightly before they fell by 78 dpf and remained relatively constant thereafter. Trends toward differences in steroid content between males and females became evident around the time gonadal differentiation was histologically discernible (78 and 90 dpf). GTH I was present in the proximal pars distalis at all dates (48–126 dpf), whereas GTH II was not detectable. GnRH was found at all dates (48–126 dpf) and was distributed in several areas of the brain including the nucleus preopticus periventricularis, nucleus lateralis tuberis, and the pituitary in the region where GTH I was found. No differences were seen between males and females in the timing of appearance, localization, or intensity of staining of these peptide hormones. Given that the brain–pituitary–gonadal axis seems to be intact during the process of sexual differentiation and the fluctuations of steroid levels during this process, sex steroids may play the driving role for sexual differentiation of rainbow trout.

Leptin is a peptide hormone that plays an important role in the regulation of energy homeostasis. Studies in mammals have shown that circulating leptin levels reflect adiposity and that this adipocyte-derived cytokine acts as an afferent satiety signal to the brain, decreasing food intake and increasing energy expenditure. Since leptin has been found in the liver and adipose tissue of migratory birds that are able to accumulate fat reserves as endogenous fuel for flight, we hypothesized that individuals with higher fat score would have higher plasma leptin levels, as it had been found previously in mammals. The aim of this study was to determine if circulating leptin levels correlate with the amount of body fat in a migratory bird, the dunlin Calidris alpina. Adult dunlins were caught during autumn migration on the Baltic coast, and their fat score was determined. Blood samples from 150 birds were used to assess the levels of circulating leptin. We did not find any statistical differences between dunlins with various fat scores. In fact, plasma leptin levels tended to be lower in fat birds than in lean individuals. Our data indicate that in wild birds in migration mode leptin does not reflect the amount of accumulated fat. It suggests that leptin in birds during migration is neither involved in the regulation of energy homeostasis nor acts as a signal to control the amount of body fat.

Adrenocorticotrophic hormone (ACTH), angiotensin II (AII), and the urophysial peptides, urotensins I and II (UI and UII), stimulate cortisol secretion by interrenal preparations of seawater (SW) and freshwater (FW) adapted trout. Steroid secretion was not disturbed in sham-treated control groups. The increased cortisol secretion following perifusion of tissue with 10(-7) M ACTH in combination with 10(-7) M AII, 10(-7) M UI, or 10(-7) M UII was greater than after separate administration of ACTH, AII, UI, or UII. These responses were no greater than the summation of the separate effects of ACTH, AII, or UII in SW and FW derived tissue or of ACTH and UI in FW derived tissue. However, the increased cortisol secretion (600-700%) after UI and ACTH in combination in SW adapted fish was significantly higher than the summated responses (100-200%) to UI and ACTH when administered separately. These results suggest that in SW fish interrenal UI enhances the steroidogenic action of ACTH, a potentially important response in SW teleost fish.

Renin-like activity (RLA) and angiotensin I-converting enzyme-like activity (ACELA), two key enzymes of the renin-angiotensin cascade (RAS), were sought in the dogfish rectal gland. RLA was 1.1 +/- 0.2 ng Ang I/mg protein/hr after incubation with porcine angiotensinogen and 0.8 +/- 0.1 ng Ang I/mg protein/hr after incubation with homologous plasma. ACELA was 7.22 +/- 1.08 and 8.87 +/- 1.9 nmol hippurate generated/min/mg protein respectively, at 0 and 37 degrees. The presence of these enzymes may indicate the presence of an endogenous RAS-like system in the rectal gland. Angiotensin II (Ang II) and atrial natriuretic peptide (ANP) binding sites were demonstrated autoradiographically in the subcapsular region of the gland, suggesting a possible interaction of the two hormones in the blind outer ends of the rectal gland tubules. Immunoreactivities toward Ang II, ANP, bombesin, vasoactive intestinal polypeptide (VIP), glucagon, and somatostatin were differentially localized in the rectal gland within three concentric zones with potentially different functional activities. In the capsule, there was a strong positive ir-glucagon reaction and a slightly weaker reaction for ir-somatostatin and VIP. In the blind outer ends of the tubules (in the subcapsular zone), strong immunoreactivity was present toward all the tested peptides except glucagon and somatostatin. In the inner zone and in the central canal, only a weak immunoreactivity toward Ang II and glucagon was observed.

The co-evolution of peptides and early cells some 3.7 billion years ago provided bioactive peptides with a long history for the proliferation and refinement of peptide hormones. Central to the adaptation and evolution of cell types in metazoans is the development of peptide signaling systems that regulate stress mechanisms. The corticotropin-releasing factor (CRF) family of peptides represents the canonical family of peptides that are pivotal to the regulation of stress in vertebrates. However, these peptides appear to have evolved at least 2 billion years after the formation of the first postulated bioactive peptides, suggesting that before this, other peptide systems played a role in stress and energy metabolism. The teneurin C-terminal associated peptides (TCAPs) are a recently discovered family of highly conserved peptides that are processed from the teneurin transmembrane proteins. This peptide/protein system is ubiquitous in multicellular organisms and evolved before the CRF family. TCAP-1 is a potent regulator of CRF-associated physiology and behavior and may play a significant role in the regulation of cell-to-cell communication and neuroplasticity in neurons.

The neurohypophysial hormones contained in the neurointermediate lobe of the pituitary of the Australian lungfish Neoceratodus forsteri have been investigated. Mesotocin was identified by coelution with the synthetic peptide in high-pressure liquid chromatography, by Edman amino acid sequencing, by mass spectrometry, by C-terminal sequencing through carboxypeptidase Y, and cleavage with prolyl endopeptidase. Vasotocin, if present, would be in very small amounts and hydrins were not detected. Oxytocin appears absent. Although Neoceratodus and Protopterus have different habitats, the former being permanently aquatic, the latter burrowing during estivation, the proportions of neurohypophysial hormones stored in neurohypophysis were roughly similar in the two species and not apparently affected by environmental conditions.

Region-specific antisera raised against different amino acid sequences of pancreastatin (Pst) (Pst-1-6, Pst-1-17, Pst-14-49 and Pst-33-49) and two antisera towards chromogranin (Cg) A and CgA/B were applied in immunofluorescence to examine the occurrence and distribution of Pst-immunoreactive (-IR) and Cg-IR cells in adrenal organs of several mammals, birds, reptiles, amphibia, and bony fish. The catecholamine-containing cells were identified using antisera against enzymes of catecholamine synthesis (tyrosine-hydroxylase, dopamine-beta-hydroxylase, and phenylethanolamine-N-methyl-transferase). No animal showed any Pst-IR or Cg-IR cells in the adrenal cortex or in its homolog, the interrenal. All antisera reacted with chromaffin cells in porcine adrenal medulla. Both adrenaline (A)- and noradrenaline (NA)-containing cells displayed Pst- and Cg-immunoreactivity. Pst- and CgA-immunoreactivities were observed in coexistence using double immunofluorescence. However, strongly reacting Pst-IR cells showed only low CgA immunoreactivity and vice versa. This inverse relationship between Pst- and CgA-immunoreactivities might reflect different levels of processing of the likely Pst-precursor CgA. In all nonmammalian vertebrates studied, Pst- and Cg-immunoreactivities were also found in both A- and NA-containing adrenal cells. However, the chromaffin cells reacted only with the antisera Pst-1-6, Pst-1-17, Pst-33-49, and CgAB. The adrenal chromaffin cells of nonmammalian vertebrates appear to contain Pst-/Cg-like peptides akin to those of the enteroendocrine cells but different from those of their endocrine pancreas. Since no immunoreactions were obtained with antiserum CgA, nonmammalian Pst may be derived from a precursor different from mammalian CgA.

Renal actions of arginine vasotocin (AVT) and atrial natriuretic peptide (ANP) in teleosts are poorly defined. Administration of these hormones changes renal function, largely explained by systemic vascular effects. The present experiments examine the potential direct action of these hormones on the telcost nephron. This has been assessed from the hormones' abilities to stimulate their second messenger systems within isolated nephroi of the rainbow trout Onchorhynchus mykiss. Following initial experiments to define the experimental conditions, AVT was found to induce a dose-dependent increase in cAMP content of a suspension of nephroi incubated for 10 min at 10°; 10 -5 M AVT provoking an increase of 846% (SW) and 829% (FW) above basal cAMP accumulation of 88 and 125 pmol/mg protein, respectively. This V 2 -type receptor response to AVT (or AVP) has previously been considered to be present only in the tetrapod kidney. ANP evoked a dose-dependent stimulation of cGMP accumulation from 1.3 (SW) or 1.9 (FW) pmol/mg protein basal levels to 5806 and 4405% of these levels, respectively, at the highest concentration used of 3.3 × 10 -7 M . Further experiments using isolated glomeruli from the eel Anguilla anguilla localized at least a part of the response to ANP to a glomerular site.

A short overview on the regional distribution of the gastro-intestinal peptide hormone cholecystokin (CCK) in fish is presented. In particular, the results of molecular and immunological studies on seabreams, Diplodus puntazzo and Diplodus sargus, are reported, which, by demonstrating CCK in the hindgut, open new questions regarding the functional role of this hormone in that part of the intestine. The putative involvement of hindgut CCK in the feedback control of digestive processes was tested by measuring CCK gene and protein expression in fed and fasted fish. The results of this study led to hypothesize different roles for the two CCK isoforms in D. sargus, one of which related to regulation of digestive processes from pyloric caeca through hindgut. On the other hand, a functional role alternative to regulation of digestive processes may be inferred for the other isoform.

Hypocalcemic potency of calcitonin isolated from the stingray (cartilaginous fish), Dasyatis akajei , was examined using the rat bioassay and compared with the activities of other calcitonins (human, pig, salmon, eel, and fowl). The potency of Dasyatis calcitonin (dCT) was estimated to be 1500–3800 IU/mg. However, when the duration of the hypocalcemic effect of dCT was taken into consideration, it was judged that dCT was approximately 2.4–6.2 times more potent than mammalian calcitonins, but about 2.3–3.5 times less active than other nonmammalian calcitonins.

The effects of somatostatin-14 (SS-14) and salmon somatostatin-25 (sSS-25) on hepatic glycogenolysis were studied by incubating rainbow trout liver pieces in vitro. Glycogen content in untreated liver pieces cultured for 3 and 5 hr was 28.6 +/- 7.6 mg/g fresh wt. and 21.5 +/- 6.6 U, respectively. Treatment of liver pieces with either SS-14 or sSS-25 resulted in significant glycogen depletion; sSS-25 appeared more potent in this regard. Equimolar concentrations (10(-8) M) of SS-14 or sSS-25 reduced glycogen content to 10.6 +/- 1.6 and 2.6 +/- 2.2 U, respectively, in liver pieces incubated for 3 hr. Alterations in liver glycogen content were reflected in glucose release into medium. Basal release of glucose into culture medium over the course of a 3-hr incubation was 20 +/- 5.6 mumol/g dry wt. Both SS-14 and sSS-25 stimulated a rapid increase (500 and 600%, respectively) in glucose release during the first 0.5 hr of incubation. After 3 hr, SS-14 and sSS-25 stimulated glucose release over basal levels to 116 +/- 9.3 and 153 +/- 16 U, respectively. Both SS-14 and sSS-25 stimulated glucose release in a dose-dependent manner; ED50 for both peptides was ca. 5 X 10(-8) M. These results indicate that both SS-14 and sSS-25 directly mediate hepatic glycogenolysis in rainbow trout.

A specific and sensitive radioimmunoassay (RIA) for the measurement of plasma levels of somatostatin-25 (SS-25) in salmon was developed using antisera raised against coho salmon (Oncorhynchus kisutch) SS-25. Somatostatin-25 was iodinated by the chloramine-T method and repurified on Sephadex G-25. The RIA was performed using a double antibody (goat anti-rabbit gammaglobulin as second antibody) method under disequilibrium conditions. Plasma from several salmonids (coho, chinook, rainbow trout, brook trout, arctic char, lake trout, and whitefish) as well as plasma from some nonsalmonids (sucker, bluegill) crossreacted with the antisera; serial dilutions of plasma from rainbow trout, brook trout, chinook salmon, and coho salmon were parallel to the SS-25 standard curve. Plasma from catfish showed negligible cross-reactivity. None of the mammalian somatostatins (somatostatin-14, somatostatin-28), U II, or other pancreatic hormones (insulin, glucagon) tested showed significant cross-reactivity with the antibody in the assay system. The lowest detectable level of SS-25 was 5 pg/tube; especially reproducible results were obtained in the range of 0.15–1.20 ng/ml, which appears to be the normal range of SS-25 circulating in the plasma of salmonids. Intra- and interassay coefficients of variation were 5.7 and 12.6%, respectively. Injection of glucose into chinook salmon resulted in an elevation of plasma SS-25 titers within 30 min and was coincident with hyperglycemia.

The specific binding of 125I-labeled eel growth hormone (eGH) to liver membranes of the eel was examined. The specific binding to the 10,000g pellet was greater than that to the 600g pellet. The specific binding was linear up to about 100 mg fresh tissue, and was saturable with increasing amounts of membrane. The specific binding was pH-, temperature-, and time-dependent, with the optimum pH at 7.4, and greater specific binding was obtained at 15 and 25 degrees than at 35 degrees. Scatchard analysis of liver binding gave an association constant of 1.1 x 10(9) M-1 and a capacity of 105 fmol/mg protein. The receptor preparation was highly specific for GHs. Natural and recombinant eel GHs as well as recombinant salmon GH competed equally with 125I-eGH for the receptor sites of the 10,000g liver membrane. Ovine GH was more potent in displacing the labeled eGH than the homologous eel hormone. Tilapia GH and ovine prolactin (PRL) were needed in greater amounts (40 times) than eGH to displace the labeled eGH. Salmon and tilapia PRLs were still less potent (500 times) than eGH. There was no displacement with eel PRL. No significant change in the specific binding was seen 1 week after hypophysectomy, whereas injection of eGH into the hypophysectomized eel caused a significant reduction after 24 hr. The binding to the membrane fractions from gills, kidney, muscle, intestine, and brain was low and exclusively nonspecific, indicating the presence of specific GH receptors predominantly in the liver.

To observe a direct effect of 17beta-estradiol (E2) on thyroid activity, thyroid follicles were isolated from hypobranchial muscles of a freshwater murrel, Channa gachua. Thyroid follicles were incubated (5 X 10(6) follicles/well) in vitro at 30 degrees for 2 hr without hormone and then 3 hr with E2 or bovine thyroid-stimulating hormone (bTSH). Addition of 10 and 100 ng of E2 to thyroid follicles resulted in 2- and 3-fold increases in thyroxine (T4) release. When 100 ng of bTSH was added to check the isolated follicular function, it stimulated T4 release to more than 3-fold. Increasing doses of E2 from 1 to 100 ng caused a dose-dependent stimulation of T4 release, while a 1000-ng dose reduced T4 release compared to 100 ng. When thyroid follicles of this experiment were lysed by sonication and T4 content was determined, it corroborated the profile of T4 release in response to varied E2 doses. E2 was ineffective in increasing 125I uptake by the follicles, while bTSH elevated it by 45% over the control. Incubation of varied concentrations of [3H]estradiol with cytosol and nuclear fractions of thyroid follicles in the presence of a 100-fold excess of diethylstilbestrol showed saturable specific binding of E2.

Gastrointestinal (GI) peptide hormones, ghrelin (GHRL), cholecystokinin (CCK), and peptide YY (PYY) genes were identified in Atlantic salmon, Salmo salar. Full-length cDNAs encoding two isoforms of GHRL (GHRL-1 and GHRL-2), two isoforms of CCK (CCK-L and CCK-N) and peptide YY (PYY) cDNA were obtained. The GHRL-1 and GHRL-2 genes encoded proteins of 111- and 108-amino acids, respectively. Both types of GHRL were mainly expressed in the stomach, but also weakly expressed in the pyloric caeca, mid-gut, adipose tissue, and testis. The CCK-L and CCK-N genes encoded preproproteins of 132- and 140-amino acids, respectively. Both types of CCK were strongly expressed in the brain and comparatively weakly expressed in other tissues, including the digestive tract. In the digestive tract, CCK-L was mainly expressed in the pyloric caeca and hind-gut, while CCK-N was only expressed in the pyloric caeca. The PYY gene encoded for 97-amino acid residues and was mainly expressed in the brain and anterior part of the intestine, including the pyloric caeca. In an experiment, we demonstrated that 6 days starvation led to, increased GHRL-1 mRNA levels in the GI tract (stomach), while there no significant changes in expression levels for the other hormones in the GI tract. This suggests an orexigenic role for GHRL-1 in Atlantic salmon. These data contribute to elucidate the functional relationships among teleost gastrointestinal peptide hormones.

The effects of relaxin (RLX), a hormone that has previously been demonstrated to have mammotrophic properties, were studied in the pigeon crop sac, a well-known target organ for mammotrophic and lactogenic hormones, and compared with the effects produced by prolactin (PRL). The two hormones were injected directly over the crop at different doses and the response was evaluated after differing times of exposure. RLX causes a dose-related increase in wet and dry weights and [3H]thymidine and [3H]uridine uptake by the crop mucosa, as well as morphological changes indicating growth and differentiation of the epithelial cells similar to those occurring during physiological activation in incubation and hatching. At the doses assayed, the effects of RLX were nearly identical to those obtained following PRL in the short-term experiments, but differences in functional responses were found in the long-term experiment.

We have developed sensitive and specific radioimmunoassays (RIA) for salmon gonadotropin-releasing hormone (sGnRH) and chicken GnRH-II (cGnRH-II). Synthetic sGnRH and cGnRH-II(2-10) were conjugated to bovine serum albumin and injected into rabbits to raise specific antisera. The antiserum against sGnRH showed cross-reactivities of 1.58 and 0.08% for cGnRH-II and lamprey GnRH, respectively. The antiserum against cGnRH-II showed cross-reactivities of 0.05 and 0.01% for sGnRH and lamprey GnRH, respectively. Both antisera were observed not to cross-react with mammalian GnRH and cGnRH-I or other peptide hormones. Synthetic sGnRH and cGnRH-II were iodinated using the chloramine-T method. The iodinated GnRH was purified by HPLC using a reverse-phase C18 column. The RIA system was developed as a double antibody method. Brain extracts of rainbow trout showed displacement curves which were parallel to the sGnRH and cGnRH-II standards in each RIA. HPLC analysis followed by RIA has revealed that rainbow trout brain contains two types of GnRH: sGnRH and cGnRH-II. Total sGnRH content in the brain was about three-fold higher than that of cGnRH-II. In the olfactory bulbs, telencephalon, optic tectum-thalamus, hypothalamus, and pituitary, sGnRH content (per region) was higher than cGnRH-II content, whereas cerebellum and medulla oblongata contained much more cGnRH-II than sGnRH. sGnRH content in the optic tectum-thalamus and pituitary was the highest in 1-year-old immature fish and 3-year-old mature fish, respectively. Medulla oblongata showed the highest cGnRH-II content in all groups. sGnRH concentrations (per milligram of protein) were high in the pituitary and intermediate in the olfactory bulbs, hypothalamus, and telencephalon. In all groups, the cGnRH-II concentration was high in the medulla oblongata, whereas the concentration in the olfactory bulbs and pituitary gland was below the detectable limit in most individuals.

Insulin-like growth factor-I (IGF-I) has been purified from chicken serum and sequenced. The peptide has eight amino acid substitutions when compared with human IGF-I: serine26, leucine38, histidine39, histidine40, lysine41, glutamine50, isoleucine64, and proline67. Chicken IGF-I (cIGF-I) has been measured using a radioimmunoassay with a human IGF-I (hIGF-I) standard and an antibody raised against hIGF-I. In this assay the cross-reactivity of cIGF-I was approximately 50% with respect to hIGF-I and the cross-reactivity of chicken IGF-II was 1.7% with respect to chicken IGF-I. To determine whether binding proteins in chicken plasma can artifactually interfere with IGF-I measurements as they do in mammals, chicken plasma was fractionated by molecular sieve chromatography at acid pH. When the fractions corresponding to the binding protein region were included in the IGF-I radioimmunoassay, essentially no apparent IGF-I was detected, indicating that the binding proteins did not interfere. This result, together with the finding that IGF-I in acid-ethanol extracts of chicken plasma produced parallel dose-response curves to pure cIGF-I and hIGF-I, allows the reliable measurement of cIGF-I in such extracts. The concentrations of IGF-I in plasma from male birds increased two- to threefold between 1 and 7 weeks after hatching to achieve 30-45 ng/ml. Smaller increases were found in female chickens from a higher value at 1 week. No diurnal pattern of IGF-I levels could be detected. In 4-week-old birds, the plasma concentration of the peptide fell from nearly 40 to 15 ng/ml after 24 hr of starvation and to 9 ng/ml 20 hr later. These effects are very similar to those described for mammals and strongly suggest that the regulation of IGF-I is conserved during evolution, notwithstanding the lower plasma concentrations of the growth factor in chickens.

Effects of growth hormone (GH) on the synthesis of mucopolysaccharide by ceratobranchial cartilages of the Japanese eel, Anguilla japonica, were examined by monitoring the in vitro uptake of ({sup 35}S)sulfate. The ({sup 35}S)sulfate uptake decreased rapidly to one-third of the initial level during the first 3 days after hypophysectomy, and decreased gradually thereafter. When hypophysectomized eels were injected intramuscularly with recombinant eel GH (2 micrograms/g), the plasma GH concentrations increased maximally after 6 hr, and declined rapidly thereafter. On the other hand, the sulfate uptake increased significantly after 12 hr, and high levels were maintained until 48 hr. The stimulating effect of GH was dose dependent (0.02-2 micrograms/g). However, the addition of eel GH (0.05-5 micrograms/ml) to the culture medium did not affect the sulfate uptake by hypophysectomized eel cartilages, suggesting that the stimulative action of GH on the sulfate uptake by the cartilages is indirect.

Acid extracts of individual lungfish pituitaries were fractionated by gel filtration on a Sephadex G-75 column and aliquots of column fractions were screened with a heterologous ACTH(1–39) radioimmunoassay (RIA). Two major, incompletely separated, peaks of ACTH-related immunoreactivity were detected. These peaks of ACTH-related material were resolved by reverse-phase high-performance liquid chromatography and were designated Peak A and Peak B. Peak A had a retention time more hydrophilic than synthetic human CLIP [ACTH(18–39)], whereas Peak B had a retention time similar to, but not identical with, human ACTH(1–39). Further analysis indicated that Peak A had an apparent molecular weight of 2.5K and an isoelectric point of 4.3. Based on these characteristics, Peak A would appear to be lungfish CLIP. Peak B had an apparent molecular weight of 4.5K. Based on chromatographic and immunological properties, Peak B would appear to be lungfish ACTH. The detection of these lungfish peptides by heterologous RIA indicates a high degree of primary sequence homology between lungfish and tetrapod ACTH-related polypeptides.

Neurohypophysial hormones in mammals are synthesized by the ovary and may influence reproductive function. Although arginine vasotocin (AVT) can induce premature oviposition in chickens, the gonadal content of AVT or mesotocin (MT) has not been examined in this species. Ovarian follicles were removed 2 hr before oviposition (-2 hr), immediately after oviposition (0 hr), and 5 hr after oviposition (+5 hr). The theca and granulosa layers of the three largest preovulatory and postovulatory follicles were isolated, extracted, and assayed for immunoreactive AVT and MT. AVT content in the theca and granulosa layers was lowest at -2 hr and increased to maximum levels at +5 hr. Pre- and postovulatory follices contained similar amounts of AVT. Maximum AVT levels in the theca and granulosa layers were 2.5 +/- 0.6 and 1.3 +/- 0.2 ng/tissue, respectively. In contrast, theca content of MT was maximal at 0 hr and the highest peptide levels were observed in the largest preovulatory follicle (0.24 +/- 0.02 ng/tissue). The results demonstrate that, compared with circulating levels of the hormones, the ovary of the chicken contains high concentrations of AVT and MT. The ovarian content of AVT and MT varies in a different manner in relation to the oviposition cycle.

The vasorelaxant effect of atrial natriuretic peptide (ANP) in two species of frogs, Rana catesbeiana and R. tigrina, were studied in vitro. Isolated arterial strips were prepared from the aortic loop, dorsal aorta, iliac, and femoral arteries in the frog. These were stimulated to contract with KCl, norepinephrine, or arginine vasotocin. When maximal contraction was attained, ANP was added to assess if it could relax the strips. Data show that ANP is vasorelaxant in these preparations. The sensitivity of these tissues depends on the contractile agents, e.g., KCl-stimulated preparations from R. catesbeiana did not respond to ANP. Data also suggest that ANP action elicited in the frog vascular tissue is due to an inhibitory effect on the mobilization of the intracellular calcium store and/or calcium influx from extracellular space to initiate contraction.
