International audience; Gamhepathiopine (also known as M1), is a multi-stage acting antiplasmodial 2-tert-butylaminothieno[3,2-d]pyrimidin-4(3H)-one hydrochloride that was first described in 2015. The development of this compound is limited by poor microsomal stability, insufficient aqueous solubility and low intestinal permeability. In order to obtain new optimized derivatives, we conducted a scaffold hopping strategy from compound M1, resulting in the synthesis of 20 new compounds belonging to six chemical series. All the compounds were tested on the K1 multi-resistant strain of Plasmodium falciparum and the human HepG2 cell-line, to evaluate their antiplasmodial activity and their cytotoxicity. Analogues’ biological results also highlighted the mandatory presence of a heteroatom at position 5 of the thieno[3,2-d]pyrimidin-4(3H)-one moeity for the antiplasmodial activity. However, modifications at position 7 were detrimental for the antiplasmodial activity. We identified furane bioisostere 3j as a promising candidate, showing good blood stage antiplasmodial activity, better water solubility and highly improved intestinal permeability in the PAMPA assay.

International audience; Drimane sesquiterpenoid dialdehydes are natural compounds with antiproliferative properties. Nevertheless, their mode of action has not yet been discovered. In this publication, we have demonstrated that various drimanes are potent inhibitors of MCL-1 and BCL-xL, two proteins of the BCL-2 family that are overexpressed in various cancers, including lymphoid malignancies. Subtle changes in their structure significantly modified their activity on the target proteins. The two most active compounds are MCL-1 selective and bind in the BH3 binding groove of the protein. Complementary studies, by NMR and mass spectrometry analyses but also synthesis showed that they are covalent inhibitors of MCL-1, via the formation of a pyrrole adduct. In addition, cytotoxic assays revealed that these two compounds show a cytotoxic selectivity for BL2, a MCL-1/BCL-xL dependent cell line and induce apoptosis.

Surface functionalization of lipid-based nanocarriers (LBNCs) with targeting ligands has attracted huge interest in the field of nanomedicines for their ability to overcome some physiological barriers and their potential to deliver an active molecule to a specific target without causing damage to healthy tissues. The principal objective of this review is to summarize the present knowledge on LBNC decoration used for biomedical applications, with an emphasis on the ligands used, the functionalization approaches, and the purification methods after ligand corona formation. The most potent experimental techniques for the LBNC surface characterization are described. The potential of promising methods such as nuclear magnetic resonance spectroscopy and isothermal titration calorimetry to characterize ligand surface corona is also outlined.

International audience; Alzheimer's disease is the most common form of senile dementia in the world, and amyloid β peptide1-42 (Aβ1-42) is one of its two principal biological hallmarks. While interactome concept was getting forward the scientific community, we proposed that the study of the molecular interactions of amyloid β peptide with the biological membranes will allow to highlight underlying mechanisms responsive of AD. We have developed two simple liposomal formulations (phosphatidylcholine, cholesterol, phosphatidylglycerol) mimicking neuronal cell membrane (composition, charge, curvature radius). Interactions with Aβ1-42 and mutant oG37C, a stable oligomeric form of the peptide, were characterized according to a simple multiparametric procedure based on ThT fluorescence, liposome leakage assay, ATR-FTIR spectroscopy. Kinetic aggregation, membrane damage and peptide conformation provided our first methodologic bases to develop an original model to describe interactions of Aβ peptide and lipids.

International audience; Human malaria infection begins with a one-time asymptomatic liver stage followed by a cyclic symptomatic blood stage. For decades, the research for novel antimalarials focused on the high-throughput screening of molecules that only targeted the asexual blood stages. In a search for new effective compounds presenting a triple action against erythrocytic and liver stages in addition to the ability to block the transmission of the disease via the mosquito vector, 2-amino-thienopyrimidinone derivatives were synthesized and tested for their antimalarial activity. One molecule, named gamhepathiopine (denoted as “M1” herein), was active at submicromolar concentrations against both erythrocytic (50% effective concentration [EC50] = 0.045 μM) and liver (EC50 = 0.45 μM) forms of Plasmodium falciparum. Furthermore, gamhepathiopine efficiently blocked the development of the sporogonic cycle in the mosquito vector by inhibiting the exflagellation step. Moreover, M1 was active against artemisinin-resistant forms (EC50 = 0.227 μM), especially at the quiescent stage. Nevertheless, in mice, M1 showed modest activity due to its rapid metabolization by P450 cytochromes into inactive derivatives, calling for the development of new parent compounds with improved metabolic stability and longer half-lives. These results highlight the thienopyrimidinone scaffold as a novel antiplasmodial chemotype of great interest to search for new drug candidates displaying multistage activity and an original mechanism of action with the potential to be used in combination therapies for malaria elimination in the context of artemisinin resistance.IMPORTANCE This work reports a new chemical structure that (i) displays activity against the human malaria parasite Plasmodium falciparum at 3 stages of the parasitic cycle (blood stage, hepatic stage, and sexual stages), (ii) remains active against parasites that are resistant to the first-line treatment recommended by the World Health Organization (WHO) for the treatment of severe malaria (artemisinins), and (iii) reduces transmission of the parasite to the mosquito vector in a mouse model. This new molecule family could open the way to the conception of novel antimalarial drugs with an original multistage mechanism of action to fight against Plasmodium drug resistance and block interhuman transmission of malaria.

International audience; To study the antikinetoplastid 3-nitroimidazo[1,2-a]pyridine pharmacophore, a structure-activity relationship study was conducted through the synthesis of 26 original derivatives and their in vitro evaluation on both Leishmania spp and Trypanosoma brucei brucei. This SAR study showed that the antitrypanosomal pharmacophore was less restrictive than the antileishmanial one and highlighted positions 2, 6 and 8 of the imidazopyridine ring as key modulation points. None of the synthesized compounds allowed improvement in antileishmanial activity, compared to previous hit molecules in the series. Nevertheless, compound 8, the best antitrypanosomal molecule in this series (EC50 = 17 nM, SI = 2650 & E° = −0.6 V), was not only more active than all reference drugs and previous hit molecules in the series but also displayed improved aqueous solubility and better in vitro pharmacokinetic characteristics: good microsomal stability (T1/2 > 40 min), moderate albumin binding (77%) and moderate permeability across the blood brain barrier according to a PAMPA assay. Moreover, both micronucleus and comet assays showed that nitroaromatic molecule 8 was not genotoxic in vitro. It was evidenced that bioactivation of molecule 8 was operated by T. b. brucei type 1 nitroreductase, in the same manner as fexinidazole. Finally, a mouse pharmacokinetic study showed that 8 displayed good systemic exposure after both single and repeated oral administrations at 100 mg/kg (NOAEL) and satisfying plasmatic half-life (T1/2 = 7.7 h). Thus, molecule 8 appears as a good candidate for initiating a hit to lead drug discovery program.

Background Pyridoclax is an original lead, recently identified as very promising in treatment of chemoresistant ovarian cancers. To correct the unfavorable intrinsic physico-chemical properties of this BCS II drug, a formulation strategy was implied in the drug discovery step. Pyridoclax-loaded nanoemulsions (NEs) were developed to permit its preclinical evaluation. Results The resulting nanoemulsions displayed a mean size of about 100 nm and a high encapsulation efficiency (>95%) at a drug loading of 2 wt%, enabling a 1,000-fold increase of the Pyridoclax apparent solubility. NEs have enabled a sustained release of the drug as assayed by a dialysis bag method. In addition, anti-tumor effects of the Pyridoclax-loaded nanoemulsions (PNEs) showed a 2.5-fold higher activity on chemoresistant ovarian cancer cells than free Pyridoclax. This effect was confirmed by a drastic increase of caspase 3/7 activation from 10 µM PNEs, as newly objectified by real time apoptose imaging. The Pyridoclax bioavailability was kept unchanged after encapsulation in nanoemulsions as determined in a mice model after oral administration. Conclusion Thus, NEs should permit valuable Pyridoclax oral administration, and valorization of this promising anticancer drug by maintaining its original anticancer activity, and by reducing the Pyridoclax therapeutic concentration.

International audience; An antikinetoplastid pharmacomodulation study was done at position 8 of a previously identified pharmacophore in 3-nitroimidazo[1,2-a]pyridine series. Twenty original derivatives bearing an alkynyl moiety were synthesized via a Sonogashira cross-coupling reaction and tested in vitro, highlighting 3 potent (40 nM ≤ EC50 blood stream form≤ 70 nM) and selective (500 ≤ SI ≤ 1800) anti-T. brucei brucei molecules (19, 21 and 22), in comparison with four reference drugs. Among these hit molecules, compound 19 also showed the same level of activity against T. cruzi (EC50 amastigotes = 1.2 μM) as benznidazole and fexinidazole. An in vitro comet assay showed that nitroaromatic derivative 19 was not genotoxic. It displayed a low redox potential value (-0.68 V/NHE) and was shown to be bioactivated by type 1 nitroreductases both in Leishmania and Trypanosoma. The SAR study indicated that an alcohol function improved aqueous solubility while maintaining good activity and low cytotoxicity when the hydroxyl group was at position beta of the alkyne triple bond. Hit-compound 19 was also evaluated regarding in vitro pharmacokinetic data: 19 is BBB permeable (PAMPA assay), has a 16 min microsomal half-life and a high albumin binding (98.5%). Moreover, compound 19 was orally absorbed and was well tolerated in mouse after both single and repeated administrations at 100 mg/kg. Its mouse plasma half-life (10 h) is also quite encouraging, paving the way toward further efficacy evaluations in parasitized mouse models, looking for a novel antitrypanosomal lead compound.

International audience; Pyridoclax is considered a promising anticancer drug, acting as a protein-protein interaction disruptor, with potential applications in the treatment of ovarian, lung, and mesothelioma cancers. Eighteen sensibly selected structural analogues of Pyridoclax were synthesized, and their physicochemical properties were systematically assessed and analyzed. Moreover, considering that drug-membrane interactions play an essential role in understanding the mode of action of a given drug and its eventual toxic effects, membrane models were used to investigate such interactions in bulk (liposomes) and at the air-water interface. The measured experimental data on all original oligopyridines allowed the assessment of relative differences in terms of physicochemical properties, which could be determinant for their druggability, and hence for drug development.

The aim of the present study was the proposal of a strategy to permit the safe use of the powerful antitumor agent Cisplatin. Considering the possibility of reducing the oxidative stress responsible for the Cisplatin induced nephrotoxicity by using antioxidants, such as flavonoids, we proposed the concomitant therapeutic association of Cisplatin with Quercetin. Considering the poor solubility of Quercetin, nanoemulsions have been proposed to encapsulate and facilitate the use of the drug. Originally, the concomitant encapsulation of Quercetin with Cisplatin has been also assessed. The obtained formulations have been characterized and tested against two human cell models, namely the human breast cancer MDA-MB-231 and the normal HEK-293 renal cells. We demonstrated that the new formulated nanoemulsions have improved the anticancer properties of both the molecules and, most importantly, the synergistic effect of the Cisplatin/Quercetin against the MDA-MB-231. Moreover, the dramatic cytotoxic effects of Cisplatin against the human renal HEK-293 cells have been significantly diminished by the use of nanoemulsions containing both the molecules, whereas the antioxidant properties of encapsulated Quercetin have been improved. Finally, the simple process used to obtain the nanoemulsions and the physico-chemical properties compatible with parenteral administration, the stability and the improved pharmaceutical effects contribute to the high potential of this strategy to be studied in future in vivo studies.

The present study aimed to determine the chemical composition, antioxidant effects and antitumor properties of a methanol extract of Anchusa azurea Mill. (Boraginaceae) aerial parts against four tumour cell lines (MCF-7, MDA-MB-231, RKO, and R2C). The antioxidant effects were assessed by using β-carotene bleaching, 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and ferric reducing ability power (FRAP) tests. HPLC analyses revealed chlorogenic acid, catechin, caffeic acid, and astragalin as the most abundant compounds. Interesting results were obtained in the β-carotene bleaching test with IC50 values of 7.6 and 27.5 μg mL−1 after 30 and 60 min of incubation, respectively. Furthermore, the A. azurea extract protects 3T3-L1 mouse cells from oxidative stress induced by menadione and exhibits good antitumor activity, with very low toxicity. Our data indicate that the antitumor properties are due to the ability to induce programmed cancer cell death through caspase 3/7 and 9 activation and interference with the cytoskeleton dynamics.

International audience; Lipophilicity must be necessarily determined in drug discovery since this physicochemical property will directly influence the pharmacokinetics of a drug as its pharmacodynamics profile. Pyridoclax is an original lead, recently identified as very promising in treatment of chemoresistant cancers. The partition coefficient (Kp) of this anticancer drug was determined by microplate assays, well adapted in drug discovery, since being rapid, and requiring only poor drug amounts. The analytical approach was performed either by UV derivative spectrophotometry after validation thanks to a set of basic, neutral and acid reference substances, or originally, by raw fluorescence spectrophotometry by taking advantage of the pyridoclax intrinsic fluorescence. Large unilamellar vesicles (LUVs) were formulated from soybean-, egg-, or dipalmitoyl-phosphatidylcholine, characterized in terms of granulometric properties, ζ potential (determined by DLS), and of phospholipid content (quantified by 1H NMR, also in presence of cholesterol). Whatever the detection method used, log Kp of pyridoclax were in the same magnitude order, and pyridoclax appeared as a lipophilic compound. It was also established that interactions between this lead and biomimetic membranes were influenced by the relative fluidity of the membranes, as confirmed by results of a liposome leakage assay

International audience; Herein we describe the drug design steps developed to increase the radical scavenging and β-amyloid aggregation inhibitory activities of a previously described series of benzylidenephenylpyrrolizinones. Among the newly synthesized derivatives, some benzylphenylpyrrolizinones exhibited interesting results in regard to those activities. Initial druggability parameters were measured, and suggest these compounds as a suitable starting point for potential alternatives in treating Alzheimer's disease

International audience; Pyridoclax is an original oligopyridine lead, very promising in treatment of chemoresistant cancers. However, from solubility measurement and permeability evaluation, it appeared that this compound can be considered as a BCS II drug, with a poor water solubility. To overcome this unfavorable property, two strategies were proposed and compared: pyridoclax di-hydrochloride salt synthesis and formulation of pyridoclax-loaded nanoemulsions (PNEs) efficiently performed by transposing the spontaneous emulsification process previously developed by our team. Whereas the salt improved the thermodynamic solubility of the drug by a factor 4, the apparent solubility of the encapsulated pyridoclax was 1000-fold higher. Their stability was assessed upon dilution in various complex biomimetic media relevant for oral administration (SGF, FaSSIF-V2, FeSSIF-V2) or for the intravenous route (PBS). The solubility of the salt was affected by the nature of the medium, indicating that it could precipitate after administration, negatively impacting its bioavailability and its efficiency in vivo. On the contrary, in all media, PNEs remained stable in terms of granulometric properties (determined by DLS), ζ-potential and encapsulation efficiency (measured by HPLC). Thus, such nanomedicines appear as a valuable option to perform preclinical studies on the promising pyridoclax

International audience; This work describes the synthesis and the biological evaluation of novel benzylidenephenylpyrrolizinones as potential antioxidant, metal chelating or amyloid β (βA) aggregation inhibitors. Some derivatives exhibited interesting results in regard to several of the performed evaluations and appear as valuable Multi-Target Directed Ligands with potential therapeutic interest in Alzheimer's disease. Among them, compound 29 particularly appears as a valuable radical and NO scavenger, a Cu(II) and Fe(II) chelating agent and exhibits moderate βA aggregation inhibition properties. These activities, associated to a good predictive bioavailability and a lack of cytotoxicity, design it as a promising hit for further in vivo investigation

International audience; The formulate-ability of six model active pharmaceutical ingredients (API), with different physico-chemical profiles, in a nanoemulsion designed to be intraveinously administrable was explored. Nanoemulsions were spontaneously generated at room temperature by pouring a phosphate buffer in an anhydrous mixture containing pharmaceutically acceptable triglycerides and non-ionic surfactants. After determination of the apparent solubility of each API in excipients and characterization of mixtures by DSC, API-loaded nanoemulsions were formulated and characterized in terms of granulometric properties, surface potential, drug recovery efficiency, pH, osmolarity, in vitro drug release, and stability. Except ciprofloxacin, a BCS class IV drug, all studied APIs were soluble in at least one excipient used, i.e. Labrasol. At 2 wt% API, all drug-loaded nanoemulsions present properties compatible with i.v. administration. The formulation should permit to increase apparent solubility of poorly water-soluble APIs, and also to prolong delivery of hydrophobic as well of more hydrophilic compounds. Herein, the relative affinity of the API for nanodroplets and the release medium would directly influence drug release profiles. Nanoemulsions were stable for 7 days. They could also been extemporaneously reconstituted before use. Such a versatile nanoemulsion would provide a valuable option as formulation strategy for improvement of drug properties

International audience; Natural or synthetic carbazole derivatives have recently attracted the attention of the scientific world because of their multiple biological activity, leading to an increase of designed, synthesized and studied analogues. In this paper, four 1,4-dimethylcarbazole derivatives, analogues of Ellipticine, have been investigated for their ability to block cancer cells growth, with low effects on the proliferation of normal cells. DNA topoisomerases inhibition assays, docking simulations, stability studies and effects on a membrane model are reported. Particularly, compounds 2 and 3 have been found thermally stable and able to inhibit, strongly and selectively, the human DNA topoisomerase II. These properties confer a good and broad antitumoral activity in vitro, with very low cytotoxic effect on the proliferation of normal cell lines and without damaging, in contrast with Ellipticine, the cell membrane model. The presented outcomes set the most active compounds as good candidates for pre-clinical studies useful in cancer treatment

International audience; From microscopic observations, it was established that an oil-in-water emulsion with droplets of a size in the micrometer range can spontaneously form at room temperature without additional external stirring as soon as a solvent that is only partly miscible to water-like dichloromethane (DCM) is put in contact with an aqueous mixture of polyethylene glycol (PEG) and a protein. Experimental results show that emulsification only occurs if the system simultaneously includes PEG with middle chain, an organic solvent partly miscible to water and for which PEG affinity is sufficiently high, and a protein. From adsorption kinetics, it appears that this spontaneous emulsification process is related to the rapid diffusion of DCM towards water through the formation of interfacial turbulences, once the accumulation of PEG close to the DCM/water interface occurs. The oil droplets formed would be then stabilized by adsorbed protein molecules. Since the presence of polylactic acid in the organic phase did not prevent the emulsion formation, we studied the feasibility of formulating microparticles using this polymer. From results, it appears that microcapsules with a polymeric shell, with a homogeneous size of about 50 microm and able to encapsulate a model hydrophobic drug, such as amiodarone, can be obtained by using this spontaneous emulsification method.

International audience; In this work, we describe the synthesis and in vitro evaluation of a novel series of multitarget-directed ligands (MTDL) displaying both nanomolar dual-binding site (DBS) acetylcholinesterase inhibitory effects and partial 5-HT4R agonist activity, among which donecopride was selected for further in vivo evaluations in mice. The latter displayed procognitive and antiamnesic effects and enhanced sAPPα release, accounting for a potential symptomatic and disease-modifying therapeutic benefit in the treatment of Alzheimer’s disease.

The purpose of the present work is to develop nanoparticles of a new antitubulin agent of the family of tripentones by means of a phase inversion process. Dynamic light scattering, transmission electron microscopy and zeta-potential measurements were used to characterize tripentone loaded nanoparticles. From interfacial tension measurements and from the study of the rheological interfacial properties of the tripentone at the Labrafac-Solutol interface, the fraction of tripentone initially present in Labrafac would stay in the oily core of nanocapsules. Moreover, the interpenetration of some tripentone molecules within the surfactant units helps to the stabilization of the formulated nanoparticles. The encapsulation efficiency was determined by high performance liquid chromatography (HPLC) and was found to be above 95%. In vitro release studies were carried out in blank nanoparticles containing phosphate buffer, pH 7.4, at 37 degrees C. The drug release kinetics was measured by HPLC. Antiproliferative activity studies on L1210 cells showed that the cytotoxic activity of tripentone was totally recovered after encapsulation of the antitubulin agent in lipid nanoparticles. This study shows that lipid nanocapsules could be a promising and effective carrier for tripentone delivery in the treatment of cancers.

In this paper, we compared the efficiency of polymer films, made of a poly(ethylene glycol) (PEG2,000)/poly( d , l -lactide) (PLA50) mixture, or a PEG2,000-PLA50 copolymer, to prevent adsorption of a model protein, the hen egg-white lysozyme (HEWL), at the air–water interface. This was achieved by analyzing the surface pressure/surface area curves, and the X-ray reflectivity data of the polymer films spread on a Langmuir trough, obtained in absence or in presence of the protein. For both the mixture and the copolymer, the amount of protein adsorbed at the air–water interface decreases when the density of the polymer surface coverage increases. It was shown that even in a condensed state, the polymer film made by the mixture can not totally prevent HEWL molecules to adsorb and penetrate the polymer mixed film, but however, protein molecules would not be directly exposed to the more hydrophobic phase, i.e. the air phase. It was also shown that the configuration adopted by the copolymer at the interface in its condensed state would prevent adsorption of HEWL molecules for several hours; this would be due in particular to the presence of PEG segments in the interfacial film.

Ovarian cancer is the leading cause of death from gynecological malignancies worldwide. Although the majority of tumors initially respond to standard treatments combining surgery and chemotherapy with platinum based chemotherapy, frequent recurrence and subsequent acquired chemoresistance are responsible for the therapeutic failure, leading to an overall 5 years survival rate of 30%. Considering the usual initial sensitivity of the ovarian tumors to chemotherapy, over the past decade efforts have been focused over the past decade to cure ovarian cancer using the currently available chemotherapeutic agents in various combinations, dosages, schedules (durations and/or routes of administration). However, with such a systemic chemotherapeutic approach, considerable limitations exist including toxicities to healthy tissues and low achievable drug concentrations at tumor sites. Considerable efforts are implemented to engineer systems capable of ferrying large doses of cytotoxic agents specifically into targeted malignant cells while sparing healthy cells. The purpose of the present review is to index the main targeted colloidal systems used for drug delivery to ovarian tumors. These nanocarriers will be analyzed by citing examples of their use in preclinical development.

We report the development of folate-functionalized nanoparticles able to target folate receptors, and to deliver a poorly water soluble cytotoxic agent, a tripentone, in ovarian carcinoma. The stability under incubation of lipid nanoparticles formulated by a low-energy phase inversion temperature method was investigated. Thanks to the presence of Labrasol ® , a macrogolglyceride into the composition of the nanocarriers, the conjugation of different quantities of a folate derivate (folic acid-polyethylene glycol 2000 -distearylphosphatidylethanolamine) to nanoparticles was possible by a rapid, soft, very simple post-insertion process. As determined by dynamic light scattering, nanoparticles present a monodisperse diameter of about 100 nm, a spherical shape as attested by transmission electron micrographs, a weakly negative surface zeta potential, and are able to encapsulate the tripentone MR22388. The presence of folate receptors on SKOV3 human ovarian cancer cells was identified by fluorescent immunocytochemistry. Cellular uptake studies assessed by flow cytometry indicated that these nanoparticles reached the SKOV3 cells rapidly, and were internalized by a folate-receptor mediated endocytosis pathway. Moreover, nanoparticles allowed the rapid delivery of the antitumor agent tripentone into cells as shown in vitro by real-time cellular activity assay. Such folate-lipid nanoparticles are a potential carrier for targeted delivery of poorly water soluble compounds into ovarian carcinoma.

A series of nitrated 2-substituted-quinolines was synthesized and evaluated in vitro toward Leishmania donovani promastigotes. In parallel, the in vitro cytotoxicity of these molecules was assessed on the murine J774 and human HepG2 cell lines. Thus, a very promising antileishmanial hit molecule was identified (compound 21), displaying an IC(50) value of 6.6 μM and CC(50) values ≥ 100 μM, conferring quite good selectivity index to this molecule, in comparison with 3 drug-compounds of reference (amphotericin B, miltefosine and pentamidine). Compound 21 also appears as an efficient in vitro antileishmanial molecule against both Leishmania infantum promastigotes and the intracellular L. donovani amastigotes (respective IC(50) = 7.6 and 6.5 μM). Moreover, hit quinoline 21 does not show neither significant antiplasmodial nor antitoxoplasmic in vitro activity and though, presents a selective antileishmanial activity. Finally, a structure-activity relationships study enabled to define precisely the antileishmanial pharmacophore based on this nitroquinoline scaffold: 2-hydroxy-8-nitroquinoline.

Lipidic nanoparticles (NP), formulated from a phase inversion temperature process, have been studied with chemometric techniques to emphasize the influence of the four major components (Solutol®, Labrasol®, Labrafac®, water) on their average diameter and their distribution in size. Typically, these NP present a monodisperse size lower than 200 nm, as determined by dynamic light scattering measurements. From the application of the partial least squares (PLS) regression technique to the experimental data collected during definition of the feasibility zone, it was established that NP present a core-shell structure where Labrasol® is well encapsulated and contributes to the structuring of the NP. Even if this solubility enhancer is regarded as a pure surfactant in the literature, it appears that the oil moieties of this macrogolglyceride mixture significantly influence its properties. Furthermore, results have shown that PLS technique can be also used for predictions of sizes for given relative proportions of components and it was established that from a mixture design, the quantitative mixture composition to use in order to reach a targeted size and a targeted polydispersity index (PDI) can be easily predicted. The objective of the present paper is to develop lipidic nanoparticles (NP) able to encapsulate drugs presenting limited solubility in both water and lipids, with high loading rates, and without using organic solvents. In this goal, a solubility enhancer, a macrogolglyceride (Labrasol®), was incorporated in a formulation process based on a low-energy phase inversion temperature method. From electrical conductivity through the temperature scans, it appears that presence of Labrasol® does not prevent the phase inversion, and it takes part in the microemulsion structuring, probably of bicontinuous type. After screening pseudo-ternary diagrams, the feasibility of NP was established. From results of a partial least square analysis, it appears that these NP present a core–shell structure where Labrasol® is well encapsulated and contributes to the formation of the oily liquid core of the NP. The diameter of the NP, assessed by dynamic light scattering, remains kinetically stable. These NP, smaller than 200 nm, spherical in shape as attested by cryo-transmission electron micrographs, are able to encapsulate a tripentone, a new anticancer agent, with drug loading rates up to 6.5% (w/w). So highly drug-loaded lipidic nanocarriers were developed without using the slightest organic solvent trace, and making it easily possible dose adjustment.

International audience; From adsorption kinetics and interfacial rheological studies performed by using a pendant-drop method, i.e. in conditions close to those of the primary emulsion of the water-in-oil-in-water emulsion-encapsulation technique, it was shown that adsorption of the hen egg-white lysozyme (HEWL) at the water/dichloromethane (DCM) interface can be efficiently slowed down by modulating some parameters. It was shown that a decrease of the ionic strength of the aqueous phase, and the optimization of the density of the poly(ethylene glycol) (PEG) adsorbed film by increasing the PEG concentration or by modulating the polymer chain length, can significantly decrease the rate of adsorption of HEWL at the water/DCM interface. Moreover, it was shown that the choice of the dissolution phase of PEG (DCM or water) clearly influences the results.

Background Our work has been carried out in the context of the therapeutic failure in ovarian carcinoma, which remains the leading cause of death by gynecologic malignancy. In these tumours, recurrence and subsequent acquired chemoresistance constitute major hurdles to successful therapy. Here we studied the interest of a member of the tripentone chemical family, MR22388, for the treatment of chemoresistant ovarian cancer cells. Findings MR22388 activity has been assessed in vitro on cisplatin-resistant (SKOV3 and IGROV1-R10) ovarian cancer cell lines by conventional analysis, alone or combined to a BH3-mimetic molecule, ABT-737. MR22388 exerts its activity on cisplatin resistant cells, and we showed that it induces a decrease of the Mcl-1 anti-apoptotic protein expression. Considering our previous work demonstrating that the efficiency of Bcl-xL targeting strategies is conditioned to the concomitant inhibition of Mcl-1 we studied the interest of the association of this MR22388 with ABT-737, and showed that this combination was highly cytotoxic in chemoresistant cells. Conclusions This work thus opens new perspectives for the use of this promising molecule for the treatment of highly chemoresistant ovarian cancer cells and for sensitization of emerging Bcl-xL targeting strategies such as the use of BH3-mimetic molecules.

Nanomedicines, which are hopeful vectors for pathologies such as cancer, are the subject of more and more studies. The enthusiasm for these active substance delivery systems raises new questions: what characteristics should be taken into account in the development of a nanodrug to ensure that the active substance will reach its therapeutic target? Among the most important elements, such as size, morphology, surface charge, etc., what is the role of the mechanical properties of nanovectors? Do these properties have an impact on the biological machinery? In this manuscript, we demonstrate that the mechanical properties of nanoparticles have an undeniable influence on biological mechanisms, and that it seems essential to integrate a systematic and standardized characterization of these properties, in the development of any new nanomedicine.; Les nanomédicaments, vecteurs d’espoir dans les pathologies telles que le cancer notamment, font l’objet d’études de plus en plus nombreuses. L’engouement envers ces systèmes de délivrance de substances actives soulève de nouvelles interrogations : quelles caractéristiques sont à prendre en compte dans le développement d’un nanomédicament, pour s’assurer que la substance active atteindra sa cible thérapeutique ? Parmi les éléments les plus déterminants, que sont la taille, la morphologie, la charge de surface etc…, quelle est la place des propriétés mécaniques des nanovecteurs ? Ces dernières ont-elles un impact sur les rouages de la machinerie biologique ? Dans ce manuscrit, nous démontrons que les propriétés mécaniques des nanoparticules ont une influence indéniable sur les mécanismes biologiques, et qu’il semble primordial d’intégrer une caractérisation systématique et standardisée de celles-ci, dans le développement de tout nouveau nanomédicament.

Burns are still a major public health problem in France today. The dispensary pharmacist is regularly called to assess the severity, as a local health actor. He can treat first degree and superficial second degree burns in the dispensary, in the absence of a gravity factor. For those of the deep second degree and third degree, he redirects the patient to an emergency service. Dressings are used at all steps of severity, except for first degree burns. The pharmacist recommends the most suitable dressing according to the size and depth of the lesion, and above all according to its level of exudation and the risk of infection of the burn to allow rapid and quality healing. He also has the role of advising other health professionals on the dressing to use according to the described characteristics of the patient's wound. Some medical devices are also used to limit scar sequelae. Finally, the dispensary pharmacist discusses nutritional advices with his patient to allow good healing of the wound and must recall the prevention advice to limit the risk of burns.; Les brûlures sont encore, à l’heure actuelle, un problème majeur de santé publique en France. Le pharmacien d’officine est régulièrement appelé à en évaluer la gravité, en tant qu’acteur de santé de proximité. Il peut prendre en charge à l’officine, les brûlures du premier degré et du deuxième degré superficiel, en l’absence de facteurs de gravité. Pour celles du deuxième degré profond et du troisième degré, il réoriente le patient vers une structure d’urgence. Les pansements sont utilisés à tous les stades de gravité, sauf pour les brûlures du premier degré. Après avoir rappelé des règles d’hygiène, le pharmacien conseille le pansement le plus adapté selon la taille, la profondeur de la lésion, et surtout selon son niveau d’exsudation et le risque infectieux de la brûlure pour permettre une cicatrisation rapide, et de qualité. Il a aussi pour rôle de conseiller les autres professionnels de santé sur le pansement à utiliser selon les caractéristiques décrites de la plaie du patient. Certains dispositifs médicaux sont également utilisés pour limiter les séquelles cicatricielles. Enfin, le pharmacien d’officine évoque avec son patient les conseils nutritionnels pour permettre une bonne cicatrisation de la plaie et doit rappeler les conseils de prévention pour limiter les risques de brûlures.

Over the last decade, people have been questioning the composition of medicines and cosmetic products before purchasing them. This interest increased with the arrival of new smartphone applications which can mark products and give information about their composition. A number of excipients are known to be controversial, i.e. are subject of a discussion which emerges from the population’s different opinions following the publication of scientific studies. The controversy may result from public concern about an excipient which could potentially be hazardous to human health. Such excipients include titanium dioxide and parabens. Titanium dioxide has many functions: colourant, opacifier, sun filter... It is now suspected of being a carcinogenic agent. Parabens are used as preservatives and are accused of being endocrine disruptors. The controversy over excipients can also be related to their environmental impact such as palm oil and silicones. Palm oil is used as an emollient, moisturizer or surfactant and has many derivatives. However, this substance is contentious as it is believed to be responsible for massive deforestation. Finally, silicones, used for their sensory qualities, are now being questioned as they appear to be non-biodegradable and would pollute the aquatic environment. Because of all these controversies and in order to meet the demands of the population, pharmaceutical and cosmetic industries are seeking for solutions to replace these contentious excipients and alternatives have now been found.; Depuis une dizaine d’année, la population se questionne sur la composition des médicaments et des produits cosmétiques avant de les acheter. Cet intérêt s’est intensifié avec l’arrivée de nouvelles applications pour téléphone qui permettent de noter les produits et leurs compositions. Un certain nombre d’excipients font l’objet d’une controverse, c’est-à-dire d’une discussion sur les différentes opinions de la population suite à la publication d’études scientifiques. La controverse peut résulter d’une inquiétude de la population face à un excipient potentiellement dangereux pour la santé humaine. Parmi ces excipients figurent le dioxyde de titane et les parabènes. Le dioxyde de titane possède de nombreuses fonctions : colorant, opacifiant, filtre solaire... Il est aujourd’hui suspecté d’être un agent cancérogène. Les parabènes sont utilisés comme agents conservateurs et sont accusés d’être des perturbateurs endocriniens. La controverse vis-à-vis des excipients peut aussi porter sur leur impact environnemental comme l’huile de palme et les silicones. L’huile de palme est utilisée comme émollient, hydratant ou tensioactif et possède de nombreux dérivés. Mais cette substance est controversée car elle serait responsable de déforestations massives. Enfin, les silicones, utilisés pour leurs qualités sensorielles, sont aujourd’hui remis en question car ils ne seraient pas biodégradables et pollueraient l’environnement aquatique. En raison de toutes ces controverses et afin de répondre aux exigences de la population, les industries pharmaceutiques et cosmétiques cherchent des solutions afin de remplacer ces excipients controversés et des alternatives ont été trouvées.

For several years now, cosmetics have been undergoing a real revolution. The desire for reasoned consumption has given rise to many brands of natural and organic cosmetics and at the same time new requirements, combined in the form of labels. However, natural consumption is not safe. Despite regulations requiring the labelling of allergens, some, and mainly natural are still too little known to consumers.; Depuis plusieurs années, la cosmétique connaît une véritable révolution. L’envie d’une consommation raisonnée a fait naître de nombreuses marques de cosmétiques naturels et biologiques et par la même occasion de nouvelles exigences, réunies sous forme de labels. Néanmoins, consommer naturel n’est pas sans danger. Malgré une règlementation obligeant l’étiquetage des allergènes, certains, et principalement naturels, sont encore trop peu connus des consommateurs.

The study of molecular interactions at the level of biological membranes is a key issue for the screening and the development of new drugs. Alzheimer's disease (AD) is the most common form of senile dementia in the world and is the leading socio-economic problem in health care. The appearance and progression of this neurodegenerative disease are associated with the aggregation of the amyloid-β peptide (Aβ). A therapeutic strategy against AD consists in the development of molecules able to interfere with specific steps of Aβ aggregation. To identify such compounds, experimental methods are required to monitor and characterize the Aβ peptide during its fibrillation process. These methods must be simple enough to remain compatible with drug discovery. In this PhD project, we have proposed to combine experimental methods to allow a multiparametric characterization of potential Aβ1-42 fibrillation modulators, by integrating liposomes of defined composition as biomimetic neuronal membranes. It is indeed established that neuronal lipids are an important factor in the formation of amyloid fibers and their toxicity. The liposomes were formulated by the lipid film rehydration method, and their physicochemical properties characterized by NMR, DLS, ζ potential. The experimental determination of the compounds partition coefficient could be carried out by spectrophotometry, including in an original way, by fluorescence, these liposomes, in miniaturized tests. Kinetic studies of Aβ1-42 peptide aggregation were performed in the presence of liposomes.The ThT fluorescence was monitored to follow the Aβ peptide fibrillation pathway, used in its wild form or with an oligomeric mutant, oG37C. A fluorophore leakage analysis from liposomes, supported by DLS measurements, was performed to evaluate the impact of peptide/membranes interactions to predict any destabilization effects. The toxic fibers formed by Aβ being mainly organized in β-sheets, the data were correlated with the analysis of the peptide secondary structure by ATR-FTIR spectroscopy. After the implementation of this approach on different model molecules and a hit of potential interest in the AD treatment, all of this work has resulted in a multiparametric test allowing the molecules/Aβ/membranes interactome characterization and the discrimination of Aβ1-42 peptide aggregation modulators. This approach may be advantageously transposed to other amyloid diseases.; L’étude des interactions moléculaires au niveau des membranes biologiques est un enjeu capital pour le développement et le screening de nouvelles molécules médicamenteuses. This study aimed at preparing and characterizing thermosensitive gel formulations based on a mixture of two hydrophilic polymers (Poloxamer 407 and 188) for a sustained delivery via liposomes of an active pharmaceutical ingredient (API) of potential interest for Alzheimer’s disease through nasal route. API was characterized as being a selective inhibitor of butyrylcholinesterase but presents a low permeability through brain blood barrier model. The osmolarity and the gelation temperature (T°sol-gel) of formulations, defined in a ternary diagram, were investigated by rheometry and visual determination. At the issue of assays, a mixture composed of p407/p188 (15/1 %, w/w) was selected, being compatible with intranasal administration in terms of (T°sol-gel = 34,5°c ± 0,3) and with the olfactory mucosal (280 ± 20 mOsmol, pH 6). Liposomes of API were prepared by the thin film hydration method. various parameters such as lipid ratio, concentration and lipid to drug ratio were investigated to obtain acceptable entrapment efficiency (EE). Formulations were evaluated for vesicle size granulometry, zeta potential, and EE determined after purification. Mucoadhesion studies were performed, and showed that in situ gel formulations present good natural mucoadhesive characteristics, that could be increased with liposomes. Drug release studies showed the API was successfully and completely released with a controlled rate, and slower than with API-loaded solutions or gels. The study demonstrated successful formulation of a promising API-loaded liposomes in a thermosensitive hydrogel for nasal delivery.; Cette étude visait à préparer et caractériser des formulations de gels thermosensibles à base de poloxamères (p407 et p188) en vue d’une délivrance prolongée, par le biais de liposomes, d’un principe actif (PA) pharmaceutique présentant un intérêt potentiel pour la maladie d’Alzheimer par voie nasale. Le PA a été caractérisé comme étant un inhibiteur sélectif de la butyrylcholinestérase, mais présente une faible perméabilité à travers le modèle de barrière hémato-encéphalique. L'osmolarité et la température de gélification (T°sol-gel) des formulations, définies dans un diagramme ternaire, ont été étudiées par analyse rhéologique et détermination visuelle. Des liposomes charges en PA ont été préparés par la méthode d'hydratation du film lipidique. Leurs propriétés ont été évaluées en termes de granulométrie, de potentiel zêta et de taux d’encapsulation. À l’issue de ces essais, un mélange composé de P407/188 15/1 (%, masse/masse) a été choisi, compatible avec l’administration intranasale et la muqueuse olfactive en termes de T°sol-gel (34,5°c ± 0,3), d’osmolarité (280 ± 20 mosmol) et de pH (6). Des études de mucoadhésion ont établi que ce gel présentait de bonnes caractéristiques mucoadhésives naturelles, et augmentées en présence de liposomes. Les cinétiques de libération ont montré que la formulation composite permettait une libération prolongée et complète du PA. Ce travail de thèse a permis le développement d’une formulation innovante, administrable par voie intranasale, assurant un temps de résidence prolongé dans la cavité nasale et libérant de façon maitrisée un PA d’intérêt thérapeutique pour le traitement d’une maladie neurodégénérative incurable à ce jour.

Psoriasis is a chronic autoimmune disease which currently affects 2 to 3% of the world's population and it’s characterized by erythematous and scaly inflammatory lesions. Many drugs can improve the symptoms without being able to cure them. This dermatosis affects seriously the patient's psychology. Also, a vicious circle exists because the stress maintains this disease. Used alone or in addition to drug treatments, dermocosmetics can improve the patient's life quality by providing him a "pleasure" for healing himself. The pharmacist can also contribute to an improvement of the patient’s life quality by accompanying him through therapeutic education and giving him advice about dermocosmetics and medical make-up. The patient becomes the "actor of his illness".; Le psoriasis est une maladie auto-immune chronique qui touche actuellement 2 à 3% de la population mondiale et qui se manifeste par des lésions inflammatoires érythémateuses et squameuses. De nombreux médicaments permettent d’améliorer les symptômes sans pouvoir les guérir. Cette dermatose affecte grandement la psychologie du patient. Il existe d’ailleurs un cercle vicieux car le stress entretient la maladie. Les dermocosmétiques, utilisés seuls ou en complément des traitements médicamenteux permettent d’améliorer la qualité de vie du patient en leur procurant un « plaisir » à se soigner. Le pharmacien peut également contribuer à une amélioration de la qualité de vie du patient, en l’accompagnant au travers de l’éducation thérapeutique et des conseils sur la dermocosmétique ainsi que sur le maquillage médical. Le patient devient alors « acteur de sa maladie ».

L'hyperpigmentation résulte d’une production excessive de mélanine au niveau de certaines zones cutanées. Elle peut avoir différentes causes et se traduit par l'apparition de taches foncées sur la peau, d'étendue et de forme variables. Ces dernières années, les femmes sont de plus en plus promptes à agir pour tenter de lutter efficacement contre ces taches parfois préoccupantes ou jugées disgracieuses. Différents moyens peuvent être envisagés qu’ils soient préventifs, mécaniques, médicamenteux ou cosmétiques. Les laboratoires cosmétiques proposent d’ailleurs de plus en plus de gammes ayant pour objectif d’améliorer « l’éclat du teint » et de faire disparaître ces taches.Après avoir présenté les causes de l’hyperpigmentation cutanée et les approches envisageables actuellement ou en devenir pour sa prise en charge, cette thèse proposera plus particulièrement une revue des approches cosmétiques pouvant être proposées en 2017.
