Abstract: CAR-T cells represent a new anti-tumor immunotherapy which has shown its clinical efficacy in B-cell malignancies. The results of clinical trials carried out in this context have shown that certain immunological characteristics of patients before (at the time of apheresis) and after the administration of the treatment, or of the CAR-T cells themselves, are correlated with the response to the treatment or to its toxicity. However, to date, there are no recommendations on the immunological monitoring of patients treated in real life. The objectives of this workshop were to determine, based on data from the literature and the experience of the centers, the immunological analyses to be carried out in patients treated with CAR-T cells. The recommendations relate to the characterization of the patient's immune cells at the time of apheresis, the characterization of the injected CAR-T cells, as well as the monitoring of the CAR-T cells and other parameters of immune reconstitution in the patient after administration of the treatment. Harmonization of practices will allow clinical-biological correlation studies to be carried out in patients treated in real life with the aim of identifying factors predictive of response and toxicity. Such data could have a major medico-economic impact by making it possible to identify the patients who will optimally benefit from these expensive treatments.

Abstract: International audience; Second-harmonic generation (SHG) is a nonlinear optical method allowing the study of the local structure, symmetry, and ferroic order in noncentrosymmetric materials such as ferroelectrics. The combination of SHG microscopy with local polarization analysis is particularly efficient for deriving the local polarization orientation. This, however, entails the use of tedious and time-consuming modeling methods of nonlinear optical emission. Moreover, extracting the complex domain structures often observed in thin films requires a pixel-by-pixel analysis and the fitting of numerous polar plots to ascribe a polarization angle to each pixel. Here, the domain structure of GeTe films is studied using SHG polarimetry assisted by machine learning. The method is applied to two film thicknesses: A thick film containing large domains visible in SHG images, and a thin film in which the domains' size is below the SHG resolution limit. Machine learning-assisted methods show that both samples exhibit four domain variants of the same type. This result is confirmed in the case of the thick film, both by the manual pixel-by-pixel analysis and by using piezoresponse force microscopy. The proposed approach foreshows new prospects for optical studies by enabling enhanced sensitivity and high throughput analysis.

Abstract: International audience; Ancient fluvial deposits typically display repetitive patterns in their depositional architecture such as alternating intervals of laterally-stacked, high-amalgamation (HA) channels, and floodplain-dominated intervals with vertically-stacked, low-amalgamation (LA) channels. Such patterns are usually ascribed to base-level driven changes in accommodation space. However, “upstream” factors such as water discharge and sediment flux have also been recognized for their potential role in controlling stratigraphic architecture but have not been tested in ancient fluvial systems. Here, we use paleohydraulic reconstructions to document riverbed gradient evolution within three middle Eocene (~40 Ma) fluvial HA-LA sequences in the Escanilla formation in the south-Pyrenean foreland basin. We show, in an ancient fluvial system, that river slope was primarily driven by climate-controlled water discharge variations rather than base-level changes as commonly assumed. These results have fundamental implications for the interpretation of the fluvial stratigraphic record and for our ability to reconstruct ancient hydroclimates.

Abstract: International audience; Introduction: This article presents the initial recommendations of the French Rheumatology Society (Société Française de Rhumatologie - SFR) and the Osteoporosis Research and Information Group (Groupe de Recherche et d'Informations sur les Ostéoporoses - GRIO) on the role of diet in the prevention and treatment of osteoporosis.Methods: The recommendations were produced by a working group composed of rheumatologists, physician nutrition specialists and a geriatrician. Fifteen (15) questions pertaining to "daily practices" were preselected by the working group. For the literature review, the working group focussed mainly on the effects of diet on bone mineral density (BMD) and fractures, and primarily on meta-analyses of longitudinal studies and dietary intervention studies.Results: A Mediterranean-type diet and the daily consumption of 2 to 3 dairy products are recommended. Together, these provide the calcium and "high quality" protein required to maintain a normal calcium-phosphorus balance and bone metabolism, and are associated with lower fracture risk. Conversely, unbalanced Western diets, vegan diets, weight-loss diets in non-overweight individuals, alcohol consumption and daily consumption of sodas are advised against. In terms of the beneficial effects on bone mineral density and fracture risk, current scientific data are either insufficient or too divergent to recommend increasing or restricting the consumption of tea or coffee, vitamins other than vitamin D, vitamin D-enriched or phytoestrogen-rich foods, calcium-enriched plant-based beverages, oral nutritional supplements, or dietary sources of prebiotics and probiotics.Conclusions: These are the first set of recommendations addressing the role of diet in the prevention and treatment of osteoporosis. More research is necessary to direct and support guidelines.

Abstract: The genetic landscape of male infertility is highly complex. It is estimated that at least 4000 genes are involved in human spermatogenesis, but only few have so far been extensively studied. In this study, we investigated by whole exome sequencing two cases of idiopathic non-obstructive azoospermia (NOA) due to severe hypospermatogenesis. After variant filtering and prioritizing, we retained for each patient a homozygous loss-of-function (LoF) variant in a testis-specific gene, C1orf185 (c.250C>T; p.Gln84Ter) and CCT6B (c.615-2A>G), respectively. Both variants are rare according to the gnomAD database and absent from our local control cohort (n = 445). To verify the implication of these candidate genes in NOA, we used the CRISPR/Cas9 system to invalidate the mouse orthologs 4930522H14Rik and Cct6b and produced two knockout (KO) mouse lines. Sperm and testis parameters of homozygous KO adult male mice were analyzed and compared with those of wild-type animals. We showed that homozygous KO males were fertile and displayed normal sperm parameters and a functional spermatogenesis. Overall, these results demonstrate that not all genes highly and specifically expressed in the testes are essential for spermatogenesis, and in particular, we conclude that bi-allelic variants of C1orf185 and CCT6B are most likely not to be involved in NOA and male fertility.

Different levels of changes of different nature and intensity are considered in order to appreciate the transformations that can take place from the integration of new media devices and the blurring of boundaries between social spheres characteristic of today's communicative practices.; L’évolution des formes éducatives à l’aune de l’intégration des technologies numériques au sein des pratiques pédagogiques intéresse de nombreux chercheurs, mais aussi les décideurs institutionnels. Cet article théorique propose un cadre de référence pour interroger et analyser les perturbations possibles engendrées par l’irruption, dans les pratiques pédagogiques, de pratiques médiatiques issues de la sphère personnelle. Considérée comme forme sociale de référence, la forme scolaire permet de définir et de décrire la forme universitaire sous l’angle de quatre dimensions constitutives. Plusieurs niveaux de changements de nature et d’intensité différentes sont envisagés pour apprécier les transformations qui peuvent s’opérer à partir de l’intégration de nouveaux dispositifs médiatiques et de l’effacement des frontières entre les sphères sociales caractéristiques des pratiques communicationnelles d’aujourd’hui.

Abstract: International audience; This paper presents a system that generates cinematic replays for dialogue-based 3D video games. The system exploits the narrative and geometric information present in these games and automatically computes camera framings and edits to build a coherent cinematic replay of the gaming session. We propose a novel importance-driven approach to cinematic replay. Rather than relying on actions performed by characters to drive the cinematography (as in idiom-based approaches), we rely on the importance of characters in the narrative. We first devise a mechanism to compute the varying importance of the characters. We then map importances of characters with different camera specifications, and propose a novel technique that (i) automatically computes camera positions satisfying given specifications, and (ii) provides smooth camera motions when transitioning between different specifications. We demonstrate the features of our system by implementing three camera behaviors (one for master shots, one for shots on the player character, and one for reverse shots). We present results obtained by interfacing our system with a full-fledged serious game (Nothing for Dinner) containing several hours of 3D animated content.

Abstract: Multiwavelength variability of blazars offers indirect insight into their powerful engines and on the mechanisms through which energy is propagated from the centre down the jet. The BL Lac object Mkn 421 is a TeV emitter, a bright blazar at all wavelengths, and therefore an excellent target for variability studies. Mkn 421 was observed by INTEGRAL and Fermi-LAT in an active state on 16-21 April 2013. Well sampled optical, soft, and hard X-ray light curves show the presence of two flares. The average flux in the 20-100 keV range is 9.1e-11 erg/s/cm2 (~4.5 mCrab) and the nuclear average apparent magnitude, corrected for Galactic extinction, is V ~12.2. In the time-resolved X-ray spectra (3.5-60 keV), which are described by broken power laws and, marginally better, by log-parabolic laws, we see a hardening that correlates with flux increase, as expected in refreshed energy injections in a population of electrons that later cool via synchrotron radiation. The hardness ratios between the JEM-X fluxes in two different bands and between the JEM-X and IBIS/ISGRI fluxes confirm this trend. During the observation, the variability level increases monotonically from the optical to the hard X-rays, while the large LAT errors do not allow a significant assessment of the MeV-GeV variability. The cross-correlation analysis during the onset of the most prominent flare suggests a monotonically increasing delay of the lower frequency emission with respect to that at higher frequency, with a maximum time-lag of about 70 minutes, that is however not well constrained. The spectral energy distributions from the optical to the TeV domain are satisfactorily described by homogeneous models of blazar emission based on synchrotron radiation and synchrotron self-Compton scattering, except in the state corresponding to the LAT softest spectrum and highest flux., Comment: 11 pages, 6 figures, in press in A&A

Abstract: The recovery of an intact epithelium following injury is critical for restoration of lung homeostasis, a process that may be altered in cystic fibrosis (CF). In response to injury, progenitor cells in the undamaged areas migrate, proliferate and re-differentiate to regenerate an intact airway epithelium. The mechanisms regulating this regenerative response are, however, not well understood. In a model of circular wound injury of well-differentiated human airway epithelial cell (HAEC) cultures, we identified the gap junction protein Cx26 as an important regulator of cell proliferation. We report that induction of Cx26 in repairing HAECs is associated with cell proliferation. We also show that Cx26 is expressed in a population of CK14-positive basal-like cells. Cx26 silencing in immortalized cell lines using siRNA and in primary HAECs using lentiviral-transduced shRNA enhanced Ki67-labeling index and Ki67 mRNA, indicating that Cx26 acts a negative regulator of HAEC proliferation. Cx26 silencing also markedly decreased the transcription of KLF4 in immortalized HAECs. We further show that CF HAECs exhibited deregulated expression of KLF4, Ki67 and Cx26 as well enhanced rate of wound closure in the early response to injury. These results point to an altered repair process of CF HAECs characterized by rapid but desynchronized initiation of HAEC activation and proliferation. Abstract: Chronic infection and inflammation of the airways is a hallmark of cystic fibrosis (CF), a disease caused by mutations in the CF transmembrane conductance regulator (CFTR) gene. The response of the CF airway epithelium to the opportunistic pathogen Pseudomonas aeruginosa is characterized by altered inflammation and apoptosis. In this study, we examined innate immune recognition and epithelial responses at the level of the gap junction protein connexin43 (Cx43) in polarized human airway epithelial cells upon infection by PAO1. We report that PAO1 activates cell surface receptors to elicit an intracellular signaling cascade leading to enhancement of gap junctional communication. Expression of Cx43 involved an opposite regulation exerted by JNK and p38 MAPKs. PAO1-induced apoptosis was increased in the presence of a JNK inhibitor, but latter effect was prevented by lentiviral expression of a Cx43-specific short hairpin RNA. Moreover, we found that JNK activity was upregulated by pharmacological inhibition of CFTR in Calu-3 cells, whereas correction of a CF airway cell line (CF15 cells) by adenoviral expression of CFTR reduced the activation of this MAPK. Interestingly, CFTR inhibition in Calu-3 cells was associated with decreased Cx43 expression and reduced apoptosis. These results indicate that Cx43 expression is a component of the response of airway epithelial cells to innate immune activation by regulating the survival/apoptosis balance. Defective CFTR could alter this equilibrium with deleterious consequences on the CF epithelial response to P. aeruginosa. Copyright © 2014 by The American Association of Immunologists, Inc.

Abstract: International audience; Story generation (including interactive narrative) consists in creating a narrative experience on computer by generating narrative events. It requires building an abstract computational model that can generate a variety of narrative events from a limited set of authored content. These models implement a "story logic", as they formalize the occurrence of an event in the story according to various algorithms. At the same time, these stories aim to be expressed to an audience using digital media, which requires a "media logic". However, it is not quite clear how the logical constraints in a theory of story should impact the media representation. Reciprocally, how media constraints influence the story logic remains overlooked. As a result, projects which aim at providing a generative and appealing experience adopt a simple "pipeline" approach where the story is sent as a series of events to the media logic, and each event is separately translated to the digital media at hand. This has the advantage of simplicity but it raises two issues: - The media logic possesses limited information from the story logic and performs an "uninformed" flat representation of story events. - The story logic generates story events without fully exploiting the strengths of the available media. In this contribution, a theory describing the interrelation between story and media is sketched. After reviewing the contrasting positions concerning the media (in)dependance in narrative theories, core principles are exposed. They consider narrative as a dynamic generative process composed of a series of independent voices that negotiate with each other to fulfill their respective goals. The creation of story events, the introduction of a plot, the visual narration, the audio narration, etc. are example of voices and sub-voices. We illustrate our theory with examples drawn from our own computer-generated stories in different digital media.

Abstract: International audience; In this paper, we describe our generic approach for transferring part-of-speech annotations from a resourced language towards an etymologically closely related non-resourced language, without using any bilingual (i.e., parallel) data. We first induce a translation lexicon from monolingual corpora, based on cognate detection followed by cross-lingual contextual similarity. Second, POS information is transferred from the resourced language along translation pairs to the non-resourced language and used for tagging the corpus. We evaluate our methods on three language families, consisting of five Romance languages, three Germanic languages and five Slavic languages. We obtain tagging accuracies of up to 91.6%.

Abstract: International audience; BACKGROUND: Observational studies on mechanical ventilation (MV) show practice variations across ICUs. We sought to determine, with a case-vignette study, the heterogeneity of processes of care in ICUs focusing on mechanical ventilation procedures, and whether organizational patterns or physician characteristics influence practice variations. METHODS: We conducted a cross-sectional multicenter study using the case-vignette methodology. Descriptive analyses were calculated for each organizational pattern and respondent characteristics. An Index of Qualitative Variation (IQV, from 0, no heterogeneity, to a maximum of 1) was calculated. RESULTS: Forty ICUs from France (N = 33) and Switzerland (N = 7) participated; 396 physicians answered our case-vignettes. There was major heterogeneity of management processes related to MV within and across centers (mean IQV per center 0.51, SD 0.09). We observed the lowest variability (mean IQV per question 0.6) for questions related to management of endotracheal tube or suctioning, management of sedation and analgesia, and respect of autonomy. Heterogeneity was independent of respondent characteristics and of the presence of written procedures. There was a correlation between the processes associated with the highest variability (mean IQV per question > 0.6) and the annual volume of ICU admission (r = 0.32 (0.01 to 0.58)) and MV (r = 0.38 (0.07 to 0.63)). Within ICUs there was a large heterogeneity regarding knowledge of a local written procedure. CONCLUSIONS: Large clinical practice variations were found among ICUs. High volume centers were more likely to have heterogeneous practices. The presence of a local written procedure or respondent characteristics did not influence practice variation.

Abstract: International audience; Vertebrate genomes contain around 20,000 protein-encoding genes, of which a large fraction is still not associated with specific functions. A major task in future genomics will thus be to assign physiological roles to all open reading frames revealed by genome sequencing. Here we show that C2orf62, a highly conserved protein with little homology to characterized proteins, is strongly expressed in testis in zebrafish and mammals, and in various types of ciliated cells during zebrafish development. By yeast two hybrid and GST pull-down, C2orf62 was shown to interact with TTC17, another uncharacterized protein. Depletion of either C2orf62 or TTC17 in human ciliated cells interferes with actin polymerization and reduces the number of primary cilia without changing their length. Zebrafish embryos injected with morpholinos against C2orf62 or TTC17, or with mRNA coding for the C2orf62 C-terminal part containing a RII dimerization/docking (R2D2) - like domain show morphological defects consistent with imperfect ciliogenesis. We provide here the first evidence for a C2orf62-TTC17 axis that would regulate actin polymerization and ciliogenesis.

Abstract: The spatial and temporal quantification of rockfall frequency remains a major challenge in mountain environments, especially also in terms of rockfall management. Approaches that have been used traditionally to quantify rockfall frequency include historical records, remote sensing, or in situ monitoring, but have been shown repeatedly to suffer from a lack of completeness or to rely on rather short time series (of a few years) that are, in addition, limited to small areas. As such, they normally cannot meet the stringent requirements of hazard and risk analyses. Here, we propose a new procedure coupling field analysis of rockfall deposits in mitigation structures and growth disturbances in tree-ring series with three-dimensional (3D), process-based rockfall modeling to estimate rockfall frequencies for individual cliff compartments. This procedure has been tested on a slope in the Swiss Alps (La Fory, Sembrancher VS), where rockfall triggered from a 38 hm $$^{2}$$ cliff to threaten a 1-km-long section of the road to the Grand St. Bernard tunnel and the local railway line. Based on 84 rockfall deposits retrieved from 420-m-long protection barrier and growth disturbances in trees over the 1994–2017 period, we estimate rockfall hazard to between 0.043 and 0.348 events yr $$^{-1}$$ at various locations on the slope. In total, we used 68,680,000 rockfall simulations to translate hazard values into rockfall frequencies at the cliff level. Converging release frequencies between tree-ring analyses (0.99–1.35 events yr $$^{-1}$$ hm $$^{-2}$$ ) and values obtained from deposits in the protection barrier (0.82 events yr $$^{-1}$$ hm $$^{-2}$$ ) confirm the reliability of our procedure. Despite remaining limitations, our approach enables precise quantification of rockfall release frequency in a holistic and reproducible manner both in space (a few hm $$^{2}$$ ) and time (several decades), and thus yields the data needed for hazard and risk mapping. We therefore recommend, where applicable, to include this procedure in future rockfall management strategies.

Abstract: International audience; The Chromosome-centric Human Proteome Project (C-HPP) aims at identifying the proteins as gene products encoded by the human genome, characterizing their isoforms and functions. The existence of products has now been confirmed for 93.2% of the genes at the protein level. The remaining mostly correspond to proteins of low abundance or difficult to access. Over the past years, we have significantly contributed to the identification of missing proteins in the human spermatozoa. We pursue our search in the reproductive sphere with a focus on early human embryonic development. Pluripotent cells, developing into the fetus, and trophoblast cells, giving rise to the placenta, emerge during the first weeks. This emergence is a focus of scientists working in the field of reproduction, placentation and regenerative medicine. Most knowledge has been harnessed by transcriptomic analysis. Interestingly, some genes are uniquely expressed in those cells, giving the opportunity to uncover new proteins that might play a crucial role in setting up the molecular events underlying early embryonic development. Here, we analyzed naive pluripotent and trophoblastic stem cells and discovered 4 new missing proteins, thus contributing to the C-HPP. The mass spectrometry proteomics data was deposited on ProteomeXchange under the data set identifier PXD035768.

Abstract: Reactive oxygen species (ROS) such as hydrogen peroxide (H2O2) and the hydroxyl radical (•OH) have specific functions in biological processes, while their uncontrolled production and reactivity are known to be determining factors in pathophysiology. Methionine (Met) residues act as endogenous antioxidants, when they are oxidized into methionine sulfoxide (MetSO), thus depleting ROS and protecting the protein. We employed tandem mass spectrometry combined with IR multiple photon dissociation spectroscopy to study the oxidation induced by OH radicals produced by γradiolysis on model cyclic dipeptides c(LMetLMet), c(LMetDMet), and c(GlyMet). Our aim was to characterize the geometries of the oxidized peptides in the gas phase and to understand the relationship between the structure of the 2-center 3-electron (2c-3e) free radical formed in the first step of the oxidation process and the final compound. Density functional theory calculations were performed to characterize the lowest energy structures of the final product of oxidation and to interpret the IR spectra. Collision-induced dissociation tandem mass spectrometry (CID-MS2) experiments of oxidized c(LMetLMet)H+ and c(LMetDMet)H+ led to the loss of one or two oxidized sulfenic acid molecules, indicating that the addition of one or two oxygen atoms occurs on the sulfur atom of both methionine side chains and no sulfone formation was observed. The CID-MS2 fragmentation mass spectrum of oxidized c(GlyMet)H+ showed only the loss of one oxidized sulfenic acid molecule. Thus, the final products of oxidation are the same regardless of the structure of the precursor sulfur-centered free radical.

Abstract: Euclid will observe 15 000 deg$^2$ of the darkest sky, in regions free of contamination by light from our Galaxy and our Solar System. Three "Euclid Deep Fields" surveys covering around 40 deg$^2$ in total will extend the scientific scope of the mission to the high-redshift Universe. The complete survey will be constituted by hundreds of thousands of images and several tens of petabytes of data. About 10 billion sources will be observed. With these images Euclid will probe the expansion history of the Universe and the evolution of cosmic structures. This will be achieved by measuring the effect on galaxy shapes due to dark matter gravitational lensing, and by reconstructing the three-dimensional distribution of cosmic structures from the measured spectroscopic redshifts of galaxies and clusters of galaxies. These proceedings present the implications for cosmology and cosmological constraints of this unprecedented data set. Abstract: The early Cenozoic marine sedimentary record is punctuated by several brief episodes ( kyr) of abrupt global warming, called hyperthermals, that have disturbed ocean life and water physicochemistry. Moreover, recent studies of fluvial–deltaic systems, for instance at the Palaeocene–Eocene Thermal Maximum, revealed that these hyperthermals also impacted the hydrologic cycle, triggering an increase in erosion and sediment transport at the Earth's surface. Contrary to the early Cenozoic hyperthermals, the Middle Eocene Climatic Optimum (MECO), lasting from 40.5 to 40.0 Ma, constitutes an event of gradual warming that left a highly variable carbon isotope signature and for which little data exist about its impact on Earth surface systems. In the South Pyrenean foreland basin (SPFB), an episode of prominent deltaic progradation (Belsué–Atarés and Escanilla formations) in the middle Bartonian has been usually associated with increased Pyrenean tectonic activity, but recent magnetostratigraphic data suggest a possible coincidence between the progradation and the MECO warming period. To test this hypothesis, we measured the stable-isotope composition of carbonates (δ13Ccarb and δ18Ocarb) and organic matter (δ13Corg) of 257 samples in two sections of SPFB fluvial–deltaic successions covering the different phases of the MECO and already dated with magnetostratigraphy. We find a negative shift in δ18Ocarb and an unclear signal in δ13Ccarb around the transition from magnetic chron C18r to chron C17r (middle Bartonian). These results allow, by correlation with reference sections in the Atlantic and Tethys, the MECO to be identified and its coincident relationship with the Belsué–Atarès fluvial–deltaic progradation to be documented. Despite its long duration and a more gradual temperature rise, the MECO in the South Pyrenean foreland basin may have led, like lower Cenozoic hyperthermals, to an increase in erosion and sediment transport that is manifested in the sedimentary record. The new data support the hypothesis of a more important hydrological response to the MECO than previously thought in mid-latitude environments, including those around the Tethys.

Abstract: International audience; Automated thermal-to-visible face recognition has received increased attention due to benefits related to lowlight applications. Towards improvement of related matching accuracy, we hereby present TFLD, a detector of face and landmarks operating in the thermal spectrum. Our proposed TFLD is based on the architecture of YOLOv5, integrating sequential modules for face and landmark detection. We introduce a thermal face restoration scheme, in order to enhance thermal image quality and hence detection accuracy. We address data scarcity by transferring landmarks in paired visible and thermal images. Our experimental results showcase that our proposed detector accurately detects faces, as well as landmarks in a wide range of adversarial conditions. Further, TFLD achieves promising results on three benchmark multi-spectral face and landmark datasets, namely ARL-VTF, SF-TL54 and RWTH-Aachen; thereby improving the matching accuracy in cross-spectral face recognition by providing robust face alignment based on estimated facial landmarks.

Abstract: International audience; Thermal to visible face image translation aims at synthesizing high-fidelity visible face images from thermal counterparts, placing emphasis on preserving the identity of the faces. While remarkable progress has been achieved related to the quality of synthetic images, as well as related to associated face matching accuracy, interpreting the generation process from thermal to visible face images remains an open challenge. Towards tackling this challenge, we present a novel generic attention-guided generative adversarial network (AG-GAN) for thermal to visible image translation. The AG-GAN framework is based on an encoder network that directly generates attention feature maps from an input thermal image in either, supervised or unsupervised fashion. A decoder network takes the attention maps and applies adaptive layer-instance normalization, in order to reconstruct the corresponding visible image. We show that solving thermal to visible image translation tasks through AG-GAN significantly improves the cross-spectral face matching accuracy, as well as inherently supports model explanation.

Abstract: The nitric oxide (NO) signaling pathway in hypothalamic neurons plays a key role in the regulation of the secretion of gonadotropin-releasing hormone (GnRH), which is crucial for reproduction. We hypothesized that a disruption of neuronal NO synthase (NOS1) activity underlies some forms of hypogonadotropic hypogonadism. Whole-exome sequencing was performed on a cohort of 341 probands with congenital hypogonadotropic hypogonadism to identify ultrarare variants in NOS1 . The activity of the identified NOS1 mutant proteins was assessed by their ability to promote nitrite and cGMP production in vitro. In addition, physiological and pharmacological characterization was carried out in a Nos1 -deficient mouse model. We identified five heterozygous NOS1 loss-of-function mutations in six probands with congenital hypogonadotropic hypogonadism (2%), who displayed additional phenotypes including anosmia, hearing loss, and intellectual disability. NOS1 was found to be transiently expressed by GnRH neurons in the nose of both humans and mice, and Nos1 deficiency in mice resulted in dose-dependent defects in sexual maturation as well as in olfaction, hearing, and cognition. The pharmacological inhibition of NO production in postnatal mice revealed a critical time window during which Nos1 activity shaped minipuberty and sexual maturation. Inhaled NO treatment at minipuberty rescued both reproductive and behavioral phenotypes in Nos1 -deficient mice. In summary, lack of NOS1 activity led to GnRH deficiency associated with sensory and intellectual comorbidities in humans and mice. NO treatment during minipuberty reversed deficits in sexual maturation, olfaction, and cognition in Nos1 mutant mice, suggesting a potential therapy for humans with NO deficiency.

Abstract: This paper contributes to the ongoing discussions on the scholarly access to social media data, discussing a case where this access is barred despite its value for understanding and countering online disinformation and despite the absence of privacy or copyright issues. Our study concerns YouTube's engagement metrics and, more specifically, the way in which the platform removes "fake views" (i.e., views considered as artificial or illegitimate by the platform). Working with one and a half year of data extracted from a thousand French YouTube channels, we show the massive extent of this phenomenon, which concerns the large majority of the channels and more than half the videos in our corpus. Our analysis indicates that most fakes news are corrected relatively late in the life of the videos and that the final view counts of the videos are not independent from the fake views they received. We discuss the potential harm that delays in corrections could produce in content diffusion: by inflating views counts, illegitimate views could make a video appear more popular than it is and unwarrantedly encourage its human and algorithmic recommendation. Unfortunately, we cannot offer a definitive assessment of this phenomenon, because YouTube provides no information on fake views in its API or interface. This paper is, therefore, also a call for greater transparency by YouTube and other online platforms about information that can have crucial implications for the quality of online public debate.

Abstract: International audience; L’ensemble des données et connaissances acquises sur la chaine Pyrénéenne et l’évolution de ces bassins d’avant-pays permettent à présent d’appréhender ce système orogénique et l’évolution des bassins dans une perspective source to sink intégrée. Cette approche met en relation l’ensemble du routage sédimentaire depuis la Source (relief orogénique, craton, recyclage de bassin) en passant par la zone de transfert (périphériques ou internes au bassin) jusqu’au réservoir final (bassin flexural, bassin turbiditique profond). Nous proposons d’intégrer l’ensemble de ces données sous la forme d’une nouvelle cartographie, sur l’ensemble du domaine péri-pyrénéen du Golfe du Lion à l’Est jusqu’au Golfe de Gascogne à l’Ouest en incluant le Massif Armoricain au Nord jusqu’au bassin de Madrid au Sud. Cette représentation en cartes est complétée par des diagrammes de Wheeler « augmentés » intégrant l’évolution des reliefs. La réalisation de ces cartes s’appuie sur l’utilisation de nombreuses méthodes et outils, tels que les reconstructions cinématiques du système Ibérie-Europe-Méditerranée, les coupes restaurées, l’histoire d’exhumation par thermochronologie, le traçage des sources, la caractérisation des surfaces d’altération et d’érosion, la synthèse de l’activité des accidents structuraux majeurs, les reconstructions paléogéographiques, l’analyse des géométries sédimentaires et des directions de transport ainsi que la quantification des volumes préservés dans les bassins. Nous présenterons ici des cartes représentatives de l’évolution du routage sédimentaire au cours des premières phases de l’orogenèse du Danien au début du Bartonien. Les pas de temps choisis pour ces cartes permettent de rendre compte de différents stades d’évolution tectono-sédimentaire du système péri-pyrénéen au stade de quiescence de l’orogenèse (1) à 65 Ma, Danien, et aux stades précoces de l’orogenèse (2) Thanétien terminal (56Ma) et (3) Yprésien (52 Ma), jusqu’au début de la collision Bartonien (40 Ma). Les compilations effectuées mettent en regard domaines exhumés et zone de sédimentation en termes de flux et de volumes, et permettent de cartographier les systèmes de routage et leur évolution au cours d’un cycle de construction d’un orogène. Elles mettent en évidence l’influence du diachronisme Est Ouest de la compression pyrénéenne et de la déformation de l’avant pays sur le routage sédimentaire, l’importance de la contribution des reliefs périphériques aux budget sédimentaires au cours de l’évolution du système source to sink et de discuter de l’influence des variations climatiques sur ce système. Abstract: Effective human movement requires the coordinated participation of the whole musculoskeletal system. Here we propose to represent the human body movements as a network (that we named “kinectome”), where nodes are body parts, and edges are defined as the correlations of the accelerations between each pair of body parts during gait. We apply this framework in healthy individuals and patients with Parkinson’s disease (PD). The network dynamics in Parkinson’s display high variability, as conveyed by the high variance and the modular structure in the patients’ kinectomes. Furthermore, our analysis identified a set of anatomical elements that are specifically related to the balance impairment in PD. Furthermore, each participant could be identified based on its kinectome patterns, akin to a “fingerprint” of movement, confirming that our approach captures relevant features of gait. We hope that applying network approaches to human kinematics yields new insights to characterize human movement.

Abstract: National audience; Mountain ecosystems are particularly sensitive to climate change, which in part causes encroachment of woody plants at the treeline ecotone, with repercussions on treeline advance and ecosystem carbon balance. Yet, studies investigating the long-term trends in radial growth as well as year-to-year response of several tree and shrub species to climate change are scarce, especially in the Pyrenees where dendroecological studies are hitherto critically lacking. Here, we estimate and comparethe long-term growth trends of two shrub (Rhododendron ferrugineum and Juniperus communis) and one tree (Pinus uncinata) species, and investigate their year-toyear growth response to changing climatic conditions and advancing snow melt-out timings. Overall, our results show that all species at our site, especially shrubs, have grown increasingly well over at least the last century, probably in response to increasing temperatures during the growing season and earlier snow melt-out dates. Nevertheless, the two shrub species differ quite strongly in their response to climate. Whereas the climate signal of J. communis has been relatively stable in recentdecades despite the persistent and significant warming trend, R. ferrugineum shows a strong shift in climate sensitivity and is increasingly affected negatively by climate change and by late-frost events. Altogether, our results address the different climate sensitivity of the two most common shrubs in the Pyrenees. They also contribute to a better understanding of vegetation dynamics in the Pyrenean treeline ecotone in the context of global change.

Abstract: International audience; Aims: The aim of this study was to map the diverse factors impacting change implementation in nursing practices and investigate different implementation strategies.Design: Scoping literature review following PRISMA-ScR extension.Methods: Data were collected from PubMed, Ebsco, Scopus and ScienceDirect databases from 1990 onwards. Only English peer-reviewed studies reporting an implementation of change in nursing practice were included. Of 9,954 studies, 425 abstracts were scanned and 98 full-text articles were screened. Finally, 28 studies were selected.Results: A multifaceted approach, with a tailored intervention, was the most effective implementation strategy. Most identified factors were considered systematic, for example resource availability, leadership and knowledge. However, others related to local social and material context were identified in fewer number of studies. These seem to be operational elements for implementation processes. Both types of factors are essential and must be considered for successful implementation.Conclusion: We advocate the development of framework including systematic factors and which capture the local context flexibility.

Abstract: Photoreduction of perylenediimide (PDI) derivatives has been widely studied for use in photocatalysis, hydrogen evolution, photo-responsive gels, and organic semiconductors. Upon light irradiation, the radical anion (PDI⋅- ) can readily be obtained, whereas further reduction to the dianion (PDI2- ) is rare. Here we show that full 2-electron photoreduction can be achieved using UVC light: 1) in anaerobic conditions by 'direct photoreduction' of PDI aggregates, or 2) by 'indirect photoreduction' in aerobic conditions due to acetone ketyl radicals. The latter strategy is also efficient for other dyes, such as naphthalenediimide (NDI) and methylviologen (MV2+ ). Efficient photoreduction on the minute time-scale using simple LED light in aerobic conditions is attractive for use in dissipative light-driven systems and materials.

Abstract: International audience; On forested paths, dendrogeomorphology has been demonstrated to represent a powerful tool to reconstruct past activity of avalanches with annual resolution and for periods covering past decades to centuries. Here, we present a new semi-quantitative approach for the identification of past snow avalanche events, which relies on the assessment of the number and position of disturbed trees within avalanche path as well as on the intensity of reactions in trees. Based on a statistical evaluation of the approach, we point to the consistency and replicability of the procedure. In a subsequent step, we demonstrate how dendrogeomorpic records can contribute to the specification of expected runout distances and related return periods of extreme events, an indispensable step in avalanche hazard assessment. Based on the reconstructed distribution of runout distances of 25 events and mean event frequencies at two paths in the French Alps, we successfully derive runout values for events with return periods of ≤300 yr. Furthermore, comparison of relations between runout distance and return periods between dendrogeomorphic data and predictions of a locally calibrated statistical-dynamical model show very reasonable agreement. Within the classical intervals used in hazard zoning (i.e. 10-300 yr), mean and mean square errors amounted to 19.7 m and 28.2 m, respectively, in the first path, and to 23.5 m and 45.8 m, respectively, in the second path. Despite some level of uncertainty related to the limits of both approaches, results suggest that dendrogeomorphic time series can yield valuable information to anticipate future extreme events.

139. A New Tree-Ring-Based, Semi-Quantitative Approach for the Determination of Snow Avalanche Events: use of Classification Trees for Validation

Abstract: International audience; On forested paths, dendrogeomorphology has been demonstrated to represent a powerful Vincent Jomelli* tool to reconstruct past activity of avalanches, an indispensable step in avalanche hazard Delphine Grancher* assessment. Several quantitative and qualitative approaches have been shown to yield Markus Stoffel † ‡ reasonable event chronologies but the question of the completeness of tree-ring records remains debatable. Here, we present an alternative semi-quantitative approach for the Christophe Corona † determination of past snow avalanche events. The approach relies on the assessment of Daniel Brunstein* the number and position of disturbed trees within avalanche paths as well as on the Nicolas Eckert § and intensity of reactions in trees. In order to demonstrate that no bias was induced by the dendrogeomorphic expert, we carry out a statistical evaluation (Classification and Regres-Michael Deschatres § sion Trees, or CART) of the approach. Abstract: International audience; This paper presents the specification of a context-aware system dedicated to assist home occupants in their everyday life while reducing their energy consumption. The system behavior and the interaction are built upon the definition of "situation spaces" based on a prior definition of the contexts of activity from the point of view of each actor in the home, i.e. taking into account actors' concerns. The interaction specification appears to be a way to manage the discrepancy between users' concerns and the system context, which can reduce errors. To develop context-aware systems that can easily be appropriated and thus potentially "invisible," we believe it is essential to articulate choices about architecture and interaction with models of individual-collective activities built upon real-life observations.

Abstract: Several regulators are involved in the control of cell cycle progression in the bacterial model system Caulobacter crescentus, which divides asymmetrically into a vegetative G1-phase (swarmer) cell and a replicative S-phase (stalked) cell. Here we report a novel functional interaction between the enigmatic cell cycle regulator GcrA and the N6-adenosine methyltransferase CcrM, both highly conserved proteins among Alphaproteobacteria, that are activated early and at the end of S-phase, respectively. As no direct biochemical and regulatory relationship between GcrA and CcrM were known, we used a combination of ChIP (chromatin-immunoprecipitation), biochemical and biophysical experimentation, and genetics to show that GcrA is a dimeric DNA–binding protein that preferentially targets promoters harbouring CcrM methylation sites. After tracing CcrM-dependent N6-methyl-adenosine promoter marks at a genome-wide scale, we show that these marks recruit GcrA in vitro and in vivo. Moreover, we found that, in the presence of a methylated target, GcrA recruits the RNA polymerase to the promoter, consistent with its role in transcriptional activation. Since methylation-dependent DNA binding is also observed with GcrA orthologs from other Alphaproteobacteria, we conclude that GcrA is the founding member of a new and conserved class of transcriptional regulators that function as molecular effectors of a methylation-dependent (non-heritable) epigenetic switch that regulates gene expression during the cell cycle., Author Summary Methylation of genomic DNA at a specific regulatory site can impact a myriad of processes in eukaryotic cells. In bacteria, methylation at the N6 position of adenosine (m6A) is known to mediate a non-adaptive immunity response to protect cells from foreign DNA. While m6A marks are not known to govern expression of cell cycle genes in Gammaproteobacteria, cell cycle transcription in the model alphaproteobacterium Caulobacter crescentus requires the m6A methyltransferase CcrM that introduces m6A marks at GAnTC sequences and the enigmatic factor GcrA. Investigating if a functional and biochemical relationship exists between CcrM and GcrA, we found that CcrM-dependent m6A marks recruit GcrA to the promoters of cell cycle genes in vitro and in vivo and is required for efficient transcription. GcrA interacts with RNA polymerase, explaining how cell cycle transcription is affected. Importantly, m6A-dependent binding is also seen in GcrA orthologs, indicating that this transcriptional regulatory mechanism by CcrM and GcrA is conserved in Alphaproteobacteria.

Abstract: Active Galactic Nuclei (hereafter AGN) produce powerful outflows which offer excellent conditions for efficient particle acceleration in internal and external shocks, turbulence, and magnetic reconnection events. The jets as well as particle accelerating regions close to the supermassive black holes (hereafter SMBH) at the intersection of plasma inflows and outflows, can produce readily detectable very high energy gamma-ray emission. As of now, more than 45 AGN including 41 blazars and 4 radiogalaxies have been detected by the present ground-based gamma-ray telescopes, which represents more than one third of the cosmic sources detected so far in the VHE gamma-ray regime. The future Cherenkov Telescope Array (CTA) should boost the sample of AGN detected in the VHE range by about one order of magnitude, shedding new light on AGN population studies, and AGN classification and unification schemes. CTA will be a unique tool to scrutinize the extreme high-energy tail of accelerated particles in SMBH environments, to revisit the central engines and their associated relativistic jets, and to study the particle acceleration and emission mechanisms, particularly exploring the missing link between accretion physics, SMBH magnetospheres and jet formation. Monitoring of distant AGN will be an extremely rewarding observing program which will inform us about the inner workings and evolution of AGN. Furthermore these AGN are bright beacons of gamma-rays which will allow us to constrain the extragalactic infrared and optical backgrounds as well as the intergalactic magnetic field, and will enable tests of quantum gravity and other "exotic" phenomena., 28 pages, 23 figures

Abstract: International audience; An exploratory study has been conducted in which 12 students in Bachelor of Science in Psychology were asked to use the Visu reflection tool during Computer-Supported Collaborative Learning (CSCL) situations. Visu is a web videoconferencing platform that allows participants to put reflective markers during their collaborative learning activity, and to review the traces of their synchronous collaboration later. In this study, co-learners used two types of markers: (1) free markers and (2) socio-emotional markers to express either negative or positive feelings about the way they collaborate together. Our contribution is related to the first focus of the workshop: emotion awareness in CSCL. In line with this focus, our main questions are as follows: (1) How can the Visu tool help learners express and share their feelings about collaboration? How does this affect the way they interact and learn together? (2) How do they use the Visu markers after the collaboration to self-reflect on their group processes? In this paper, we first describe the Visu platform. We then present a study we have carried out to answer the questions presented above. We finally conclude with the contributions of our research to the workshop topic.

Abstract: International audience; This paper describes an exploratory study on the use of reflective markers set during synchronous collaborative learning sessions (reflection in action) for later construction of self-reflection reports upon the collaboration that occurred (reflection on action). During 2 sessions, students used the Visu tool for interaction and marker setting (positive, negative, free) and then report building on the interaction (using markers or not). A quantitative descriptive analysis has been conducted on the markers used and on the reflective categories of the sentences in the reports. Results show that students (1) paid more attention in repairing their relationship than reflecting on learning and task goals; (2) used mainly positive markers to both reflect in and on action; (3) used more their partner's markers in the second reports; (4) reflected more on themselves in the second reports to justify successes and failures, and to express satisfaction.

Abstract: Schweizerische Zeitschrift für Bildungswissenschaften 34 (2012) 3, S. 501-514, En référence aux enjeux de ce numéro thématique, cette contribution propose de rendre compte d’expériences de formation d’enseignants débutants au cours desquelles sont repérables des transformations de leur engagement. L’approche privilégiée accorde une importance particulière à la dimension dynamique des phénomènes dès lors décrits sur la durée à partir d’études longitudinales. Trois modalités de transformation ou reconfiguration de l’engagement des enseignants débutants ont été caractérisées sous la forme de "sorties d’impasses", de "transitions" et de "reconstructions massives". Une meilleure compréhension des processus identitaires en jeu lors de ces reconfigurations nous semble a même d’ouvrir des pistes prometteuses pour la formation. (DIPF/Orig.), Mit Bezug auf die Fragestellungen dieses Themenheftes schlägt dieser Beitrag vor, Bildungserfahrungen von Junglehrpersonen zu betrachten, bei denen im Verlauf ihrer Berufseinstiegsphase Veränderungen ihres Engagements identifiziert werden können. Das gewählte Vorgehen legt besonderen Wert auf die dynamische Dimension der betrachteten Phänomene, weshalb diese durch Längsschnittstudien beschrieben werden. Drei Arten der Transformation resp. der Rekonfiguration des Engagements von Junglehrpersonen werden charakterisiert: "Auswege aus (den) Sackgassen", "Übergänge" und "umfangreiche Rekonstruktion". Abstract: Delrez, L., et al., Exoplanets transiting bright nearby stars are key objects for advancing our knowledge of planetary formation and evolution. The wealth of photons from the host star gives detailed access to the atmospheric, interior and orbital properties of the planetary companions. ν Lupi (HD 136352) is a naked-eye (V = 5.78) Sun-like star that was discovered to host three low-mass planets with orbital periods of 11.6, 27.6 and 107.6 d via radial-velocity monitoring. The two inner planets (b and c) were recently found to transit, prompting a photometric follow-up by the brand new Characterising Exoplanets Satellite (CHEOPS). Here, we report that the outer planet d is also transiting, and measure its radius and mass to be 2.56 ± 0.09 R and 8.82 ± 0.94 M, respectively. With its bright Sun-like star, long period and mild irradiation (~5.7 times the irradiation of Earth), ν Lupi d unlocks a completely new region in the parameter space of exoplanets amenable to detailed characterization. We refine the properties of all three planets: planet b probably has a rocky mostly dry composition, while planets c and d seem to have retained small hydrogen–helium envelopes and a possibly large water fraction. This diversity of planetary compositions makes the ν Lupi system an excellent laboratory for testing formation and evolution models of low-mass planets., The MOC activities have been supported by ESA contract 4000124370. Abstract: International audience; Two millets, Panicum miliaceum and Setaria italica, were domesticated in northern China, around 6000 BC. Although its oldest evidence is in Asia, possible independent domestication of these species in the Caucasus has often been proposed. To verify this hypothesis, a multiproxy research program (Orimil) was designed to detect the first evidence of millet in this region. It included a critical review of the occurrence of archaeological millet in the Caucasus, up to Antiquity; isotopic analyses of human and animal bones and charred grains; and radiocarbon dating of millet grains from archaeological contexts dated from the Early Bronze Age (3500-2500 BC) to the 1 st Century BC. The results show that these two cereals were cultivated during the Middle Bronze Age (MBA), around 2000-1800 BC, especially Setaria italica which is the most ancient millet found in Georgia. Isotopic analyses also show a significant enrichment in 13 C in human and animal tissues, indicating an increasing C 4 plants consumption at the same period. More broadly, our results assert that millet was not present in the Caucasus in the Neolithic period. Its arrival in the region, based on existing data in Eurasia, was from the south, without excluding a possible local domestication of Setaria italica. Today, there are many genetic, archaeobotanical or biochemical resources to track the domestication processes and diffusion of plants and animals. Among them, the millets, Panicum miliaceum and Setaria italica, are two species that have their origins in China, but their path to Europe has been the subject of numerous studies for several years. Across Asia, researchers have looked at millets to describe the dispersal of peoples and languages 1 , to address the concept of food globalization 2 , to understand how these cereals have adapted to different environments 3,4 and to investigate the sedentary, semi-nomadic, or nomadic lifestyles of the early agropastoral communities 5,6. This article focuses on the Caucasus, a region that has hitherto not yet been explored
