This work will describe a system to quantify ultra-low-level tritium concentrations in ∼100 mL water samples. Tritium decays into 3He, thus we can take advantage of high sensitivity noble gas mass spectrometry.

Efforts to model and simulate various aspects of liquid chromatography (LC) separations (e.g., retention, selectivity, peak capacity, injection breakthrough) depend on experimental retention measurements to use as the basis for the models and simulations. Often these modeling and simulation efforts are limited by datasets that are too small because of the cost (time and money) associated with making the measurements. Other groups have demonstrated improvements in throughput of LC separations by focusing on “overhead” associated with the instrument itself – for example, between-analysis software processing time, and autosampler motions. In this paper we explore the possibility of using columns with small volumes (i.e., 5 mm x 2.1 mm i.d.) compared to conventional columns (e.g., 100 mm x 2.1 mm i.d.) that are typically used for retention measurements. We find that isocratic retention factors calculated for columns with these dimensions are different by about 20%; we attribute this difference – which we interpret as an error in measurements based on data from the 5 mm column – to extra-column volume associated with inlet and outlet frits. Since retention factor is a thermodynamic property of the mobile/stationary phase system under study, it should be independent of the dimensions of the column that is used for the measurement. We propose using ratios of retention factors (i.e., selectivities) to translate retention measurements between columns of different dimensions, so that measurements made using small columns can be used to make predictions for separations that involve conventional columns. We find that this approach reduces the difference in retention factors (5 mm compared to 100 mm columns) from an average of 18% to an average absolute difference of 1.7% (all errors less than 8%). This approach will significantly increase the rate at which high quality retention data can be collected to thousands of measurements per instrument per day, which in turn will likely have a profound impact on the quality of models and simulations that can be developed for many aspects of LC separations.

RATIONALE Compound-specific stable isotope analysis (CSIA) is a powerful tool for a better understanding of trophic transfer of dietary molecules in and across ecosystems. Hydrogen isotope values (δ2 H) in consumer tissues have potential to more clearly distinguish dietary sources than 13 C or 15 N values within and among habitats, but have not been used at the fatty acid level for ecological purposes. METHODS Here we demonstrate a new online high-capacity gas chromatography-isotope ratio mass spectrometry technique (2 H-CSIA) that offers accurate and reproducible determination of δ2 H values for a range of fatty acids from organisms of aquatic food webs. RESULTS We show that lipid extracts obtained from aquatic sources, such as biofilms, leaves, invertebrates, or fish muscle tissue, have distinctive δ2 H values that can be used to assess sources and trophic interactions, as well as dietary allocation and origin of fatty acids within consumer tissue. CONCLUSIONS The new 2 H-CSIA method can be applied to evaluate sources and trophic dynamics of fatty acids in organisms ranging from food web ecology to migratory connectivity.

The electronic surroundings of phosphorus and lithium atoms in the ionic conductor lithium dihydrogen phosphate (LDP) have been studied by single-crystal nuclear magnetic resonance (NMR) spectroscopy at room temperature. From orientation-dependent NMR spectra of a large homegrown LDP single crystal, the full 31P chemical shift (CS) and 7Li quadrupole coupling (QC) tensor was determined, using a global fit over three rotation patterns. The resulting CS tensor is characterized by its three eigenvalues: &delta, 11 P A S = ( 67.0 &plusmn, 0.6 ) ppm, &delta, 22 P A S = ( 13.9 &plusmn, 1.5 ) ppm, and &delta, 33 P A S = ( &minus, 78.7 &plusmn, 0.9 ) ppm. All eigenvalues have also been verified by magic-angle spinning NMR on a polycrystalline sample, using Herzfeld&ndash, Berger analysis of the rotational side band pattern. The resulting 7Li QC tensor is characterized by its quadrupolar coupling constant &chi, = Q 33 P A S = ( &minus, 71 &plusmn, 1 ) kHz and the two eigenvalues Q 11 P A S = ( 22.3 &plusmn, 0.9 ) kHz, and Q 22 P A S = ( 48.4 &plusmn, 0.8 ) kHz. The initially unknown orientation of the mounted crystal, expressed by the orientation of the rotation axis in the orthorhombic crystal frame, was included in the global data fit as well, thus obtaining it from NMR data only.

The aim of the present work was to develop a new scalable and cost-efficient process to isolate bovine immunoglobulin G from colostral whey with high purity and minimal loss of activity. The mixed mode material Mercapto-Ethyl-Pyridine-Hypercel™ was identified appropriate for direct capture of immunoglobulin G. The binding mechanism is primarily based on hydrophobic interactions at physiological conditions. As compared to immunoglobulin G, all other low molecular whey proteins such as α-Lactalbumin or β-Lactoglobulin, except lactoperoxidase, are more hydrophilic and were therefore found in the flow-through fraction. In order to remove lactoperoxidase as an impurity the column was combined in series with a second mixed mode material (Capto™- with N-benzoyl-homocysteine as ligand) using the same binding conditions. At pH 7.5 the carboxyl group of this ligand is negatively charged and can hence bind the positively charged lactoperoxidase, whose isoelectric point is at pH 9.6. After sample application, the columns were eluted separately. By combining the two columns it was possible to obtain immunoglobulin G with a purity of >96.1% and yield of 65-80%. The process development was carried out using 1 mL columns and upscaling was performed in three steps up to a column volume of 8800 mL for the Hypercel™ column and 3000 mL for the Capto™- column. At this scale it is possible to obtain 130-150 g pure immunoglobulin G from 3 L colostrum within five hours, including the regeneration of both columns. Additionally, the impact of freeze-drying on the isolated immunoglobulin G was studied. The nativity of the freeze dried immunoglobulin was above 95%, which was proven by reversed phase liquid chromatography and validated by differential scanning calorimetry. The activity of immunoglobulin G was preserved over the isolation process and during drying as measured by enzyme-linked immunosorbent assay. In conclusion, by applying the proposed isolation process, it becomes feasible to obtain pure, active and stable imunnunoglobulin G at large scale.

The in-process denaturation of native whey protein-based coatings, film temperature, residual humidity, and protein degree of denaturation was simulated with combined existing models. The simulation was validated experimentally through determining the degree of denaturation of the protein in the coating. Whey protein-based formulations were applied, and denaturation was induced via infrared heating during the drying process. The oxygen transmission rate was measured and correlated with the degree of denaturation. The observed influence of the denaturation degree allowed the identification of a critical denaturation degree improving oxygen barrier. The developed simulation appears as an efficient tool for optimizing the processing and properties of protein-based materials.

Lead Zirconate Titanate (PZT) ceramics Pb 1−1.5 x Bi x V Pb ′′ 0.5 x Zr 0.90 Ti 0.10 O 3 containing Bismuth ( x = 0.005–0.12) were structurally and dielectrically characterized. Results are compared to Lanthanum modified Pb 1−1.5 x La x V Pb ′′ 0.5 x Zr 0.90 Ti 0.10 O 3 , ( x = 0.01–0.08) ceramics. For Bi a solubility limit is observed at x = 0.04. The Lanthanum-doped series proved to be single phase in the investigated range. Lanthanum doping leads to a continuous reduction of cell volume causing a change from rhombohedral to orthorhombic symmetry at x = 0.03 accompanied by switching from ferroelectric to antiferroelectric behaviour. Such a change was not observed with Bismuth. This might be due to the fact that a critical cell volume necessary for structural transition could not be achieved within the solubility range. For Lanthanum doped PZT with x = 0.03 orthorhombic structure was determined already at room temperature, but with ferroelectric polarization curve, which switched to antiferroelectric polarization at 120 °C. This raises new questions on the appearance and stability of antiferroelectric ordering.

In this case study we investigate the dependence of the RF-induced power deposition (P) and the temperature rise (ΔT p ) in gel at lead electrode on the electrode geometry for helix and straight leads at 128 MHz. The results indicate that shortening the electrode length from 10 mm to 3 mm resulted in up to fourfold increase of Δ T p .

Various compositions of the solid solution system (100 − x ) Bi 0,5 Na 0,5 TiO 3 – x Bi 0,5 K 0,5 TiO 3 ( x = 0, 10, 25, 50, 75, 90, 100) were prepared by the mixed oxide route. The formation reaction was analyzed by thermogravimetry coupled with mass spectroscopy and differential scanning calorimetry. In situ high temperature X-ray diffraction up to 770 °C indicated emerging and vanishing of phases during the calcination. Intermediate phases such as alkalipolytitanate (Na/K) 2 Ti 6 O 13 and bismuth titanate Bi 2 Ti 2 O 7 were identified as forming the perovskite phase. The formation reactions were proposed based on the data obtained. Furthermore the microstructure and the dielectric behavior of the sintered samples were observed by scanning electron microscopy, impedance spectrometry and polarization measurements.

Accurate knowledge of the spatial magnetic field distribution is necessary when measuring field gradients. Therefore, a MEMS magnetic field gradiometer is reported, consisting of two identical, but independent laterally oscillating masses on a single chip. The sensor is actuated by Lorentz force and read out by modulation of the light flux passing through stationary and moving arrays of the chip. This optical readout decouples the transducer from the electronic components. Both phase and intensity are recorded which reveals information about the uniformity of the magnetic field. The magnetic flux density is measured simultaneously at two points in space and the field gradient is evaluated locally. The sensor was characterised at ambient pressure by performing frequency and magnitude response measurements with coil and various different permanent magnet arrangements, resulting in a responsivity of 35.67 V/T and detection limit of 3.07 &micro, T/ Hz (@ 83 Hz ENBW). The sensor is compact, offers a large dynamic measurement range and can be of low-cost by using conventional MEMS batch fabrication technology.

The synthesis of Bi 0.5 Na 0.375 K 0.125 TiO 3 (BNT–BKT or NBT-KBT) by the mixed oxide route was investigated using thermogravimetry, X-ray diffraction, scanning electron microscopy and electron microprobe analysis. The results show the formation of a paraelectric secondary phase with the composition (K,Na) 2 Ti 6 O 13 during sintering. Thermogravimetric measurements indicate that the main loss of volatile components mainly occurs during the calcinations step, exceeding the theoretical loss by more than 2%. These losses, most likely caused by evaporation of K 2 O induce a high number of Schottky-vacancies. These vacancies highly affect the electrical properties of the material. The formation of the secondary phase leads to a shifted composition of the ceramic to approximately Bi 0.5 Na 0.4 K 0.1 TiO 3 .

The Gram-positive, mesophilic bacterium Paenibacillus alvei CCM 2051(T) possesses a two-dimensional crystalline protein surface layer (S-layer) with oblique lattice symmetry composed of a single type of O-glycoprotein species. Herein, we describe a strategy for nanopatterned in vivo cell surface co-display of peptide and glycan epitopes based on this S-layer glycoprotein self-assembly system. The open reading frame of the corresponding structural gene spaA codes for a protein of 983 amino acids, including a signal peptide of 24 amino acids. The mature S-layer protein has a theoretical molecular mass of 105.95kDa and a calculated pI of 5.83. It contains three S-layer homology domains at the N-terminus that are involved in anchoring of the glycoprotein via a non-classical, pyruvylated secondary cell wall polymer to the peptidoglycan layer of the cell wall. For this polymer, several putative biosynthesis enzymes were identified upstream of the spaA gene. For in vivo cell surface display, the hexahistidine tag and the enhanced green fluorescent protein, respectively, were translationally fused to the C-terminus of SpaA. Immunoblot analysis, immunofluorescence staining, and fluorescence microscopy revealed that the fused epitopes were efficiently expressed and successfully displayed via the S-layer glycoprotein matrix on the surface of P. alvei CCM 2051(T) cells. In contrast, exclusively non-glycosylated chimeric SpaA proteins were displayed, when the S-layer of the glycosylation-deficient wsfP mutant was used as a display matrix.

An automated approach for rapid on-line monitoring of the solid and liquid phases present in bioprocesses based on mid-IR Fourier transform spectrometry is introduced. The principles of this new approach are presented using the example of the quantitative and qualitative analysis of poly(beta-hydroxybutyric acid) (PHB) accumulated in living bacterial cells as well as dissolved glucose during two 27-h fermentation processes. FT-IR spectra were recorded on-line using a diamond-attenuated total reflection (ATR) cell connected to the fermentation broth by means of a computer-controlled flow system. For calibration of the FT-IR method, standard reference analysis procedures for PHB (gas chromatography) and glucose (HPLC) were used. While pumping the fermentation broth through the flow cell, the recorded spectra corresponded to the fermentation solution, which allowed the determination of glucose in a range from 3.8 to -10.3 g/L. Upon stopping the flow, the cells settled on the ATR surface, and the thus recorded spectra enabled the determination of the intracellular PHB content of Escherichia coli in a range from 0.005 to 0.766 g/L. Errors of cross-validation of 0.264 g/L for glucose and 0.037 g/L for PHB were obtained. Application of one PLS calibration model to another fermentation was possible with prediction errors of 0.493 g/L for glucose and 0.105 g/L for PHB. Furthermore, from the position and shape of the PHB carbonyl band, it could be concluded that the PHB granules inside the E. coli are predominantly amorphous.

S-layer fusion protein technology was used to design four different fluorescent fusion proteins with three different GFP mutants and the red fluorescent protein mRFP1. Their absorption spectra, steady-state fluorescence, and fluorescence lifetime were investigated as a function of pH. It was found that fluorescence intensities and lifetime of the GFP mutant S-layer fusion proteins decreased about 50% between pH 6 and pH 5. The spectral properties of the red S-layer fusion protein were minimally affected by pH variations. These results were compared with His-tagged reference fluorescent proteins, demonstrating that the S-layer protein did not change the general spectral properties of the whole fusion protein. In addition, the pK(a) values of the fluorescent S-layer fusion proteins were calculated. Finally, it was shown that the S-layer fusion proteins were able to self-assemble forming 2D nanostructures of oblique p2 symmetry with lattice parameters of about a = 11 nm, b = 14 nm, and gamma = 80 degrees . The fluorescence tag did not hinder the natural self-assembly process of the S-layer protein. The combination of the fluorescence properties and the self-assembly ability of the engineered fusion proteins make them a promising tool to generate biomimetic surfaces for future applications in nanobiotechnology at a wide range of pH.

This paper describes the development of planar and fiber optic oxygen sensors utilizing surface layer (S-layer) proteins as immobilization matrix for oxygen sensitive dyes. S-layer proteins have the intrinsic capability to reassemble into two-dimensional arrays in suspension and at interfaces. Due to their crystalline character the distribution of functional groups, such as carboxylic groups, is repeated with the periodicity of the lattice and thus allows the reproducible and geometrically distinct binding of functional molecules. For the development of oxygen sensors an oxygen sensitive Pt(II) porphyrin dye was covalently bound to the S-layer matrix. Measurement of the oxygen concentration was performed by phase modulation fluorimetry. Setups comprising low cost optoelectronic components like LEDs and silicon photodiodes were constructed. For both sensor setups (planar and fiber optic) variations in the oxygen concentrations resulted in distinct and reproducible changes in luminescence lifetime and intensity. The luminescence quenching efficiency of these sensors was found to be 1.5-1.9 (expressed as the ratio of signal under nitrogen and air) which compares well to other sensor systems using luminophores embedded in polymer matrices. These results demonstrated the application potential of S-layers as immobilization matrices in the development of (bio-)sensors.

A rapid analytical method for sensitive determination of 1,3-dichloro-2-propanol (1,3-DCP) in river water has been developed. 1,3-DCP is extracted from water with ethyl acetate. After filtration through sodium sulfate the ethyl acetate phase is analyzed by gas chromatography-mass spectrometry. The method uses 1,3-DCP-d5 as internal standard. Different extraction solvents, concentrations of ammonium sulfate in the water samples, and the effect of humic acid were tested and their influence on the recovery of DCP has been evaluated. The method quantification limit was 0.1 microg L(-1). For spiked water samples (0-5.2 microg L(-1), n=21) a repeatability coefficient of variation of 5.4% was obtained. The average recovery rate of 1,3-DCP was 105+/-3% (n=21). Stability tests, which were carried out with Danube river water, led to an estimated 1,3-DCP degradation rate of 0.008+/-0.0008 day(-1) at 6 degrees C.

Four different coloured pH dependent biocolloids have been designed with fluorescent S-layer fusion proteins. The intrinsic self assembling capability of the S-layer protein SgsE on silicon dioxide particles lead to crystalline p2 lattice symmetry which was combined with the fluorescent properties of fused cyan ECFP, green EGFP, yellow YFP and red mRFP1. Confocal microscopy was used to monitor the pH dependence of the fluorescent S-layer coating. Electrophoretic measurements were carried out to understand the colloidal behaviour of the S-layer protein coated particles. It was found with flow cytometry that the stable SgsE-ECFP, SgsE-EGFP, SgsE-YFP and SgsE-mRFP bioparticle suspensions lost 50% of their fluorescence emission at the pKa of the chromophores. These novel fluorescent S-layer biocolloids can be used as pH sensors and might have an important role in surface pH determination.

Eisen im Wein wurde nach Aufschlus mit Salpetersaure-Perhydrol durch Differential-Puls-Polarographie bestimmt. Vergleichswerte wurden photometrisch mito-Phenanthrolin erhalten. Die direkte photometrische Methode liefert nur bei Weisweinen exakte Ergebnisse, dagegen nicht bei Rotwein. Hier ist ein Aufschlus erforderlich. 18 Weinsorten wurden untersucht.

Zur Bestimmung von Glutaraldehyd in der Raumluft von Krankenhausern wurde ein neues Verfahren ausgearbeitet, mit dem subppm-Mengen erfasbar sind. Die Raumluft wird durch eine Schicht Aluminiumoxid geleitet, wobei Glutaraldehyd quantitativ adsorbiert wird. Nach Desorption des Aldehyds durch Wasser im Ultraschallbad wird die wasrige Losung an einer Tenax-GC-Saule chromatographiert. Die quantitative Bestimmung des Aldehyds erfolgt durch einen Flammenionisationsdetektor.

Eine Methode zur Bestimmung von Quecksilber in Oberflachen-, Quell- und Grundwassern wurde ausgearbeitet. Quecksilber wird dabei vorerst elektrolytisch an einer Goldelektrode angereichert, dann durch Erhitzen desorbiert und die Absorption des Quecksilberdampfes in einem Atomabsorptionsspektrometer gemessen. Die Nachweisgrenze der Methode liegt bei 1,5 ng/Liter. Die elektrolytische Anreicherung und die Adsorptionseffekte an Glasgefasen wurden naher untersucht.

Die quantitative Bestimmung von Bromessigsaure und deren Estern in Wein wurde bezuglich ihrer Fehlerquellen untersucht. Nach Extraktion der Bromessigsaure und Verseifung mit Lauge kann das entstandene Bromid entweder — nach Oxydation zu Bromat — titriert werden oder — nach Reaktion mit Phenolrot und Chloramin-T zu Bromphenolblau — photometrisch bei 585 nm bestimmt werden. Die Ursachen fur positive und negative Abweichungen vom Sollwert wurden ermittelt. Bei der titrimetrischen Bestimmung kann das Hypochlorit bzw. dessen Herstellung einen positiven Fehler verursachen. Ein negativer Fehler entsteht, wenn der pH-Wert bei der Titration groser als 0,9 ist. Bei der photometrischen Bestimmung mus eine bestimmte Relation zwischen Bromid/Phenolrot/Chloramin-T eingehalten werden. Positive Fehler kommen dadurch zustande, das sich chlorierte Produkte des Phenolrots bilden, die wie Bromphenolblau bei 585 nm absorbieren. Das Ausmas des Fehlers hangt von der Reaktionszeit sowie von der Chloramin-Menge ab. Die optimale molare Relation Bromid: Phenolrot ist 4 ∶ 1,2 bis 1,35. Ist die Phenolrot-Menge entweder wesentlich groser oder kleiner, entstehen negative Fehler. Die optimale Reaktionszeit betragt 60 Sek.; dann mus Chloramin-T durch Zugabe von Thiosulfat reduziert werden. In dieser Zeit ist die quantitative Bildung von Bromphenolblau aus Bromid bereits abgeschlossen. Die storenden Nebenreaktionen (Bildung chlorierter Produkte des Phenolrots sowie Oxydation des Bromphenolblaus durch Chloramin) wirken sich als langsame Reaktionen zu diesem Zeitpunkt noch wenig auf das Resultat aus.

Verschiedene Verfahren zur Bestimmung von Mikrogramm-Mengen Calcium in biologischem Material wurden getestet, um eine optimale Arbeitsvorschrift fur die Bestimmung von Calcium in Rattenherzen zu erhalten. Etwa 100 mg Probe werden trocken bei 510°C im Tiegelofen verascht. Nach Losen des Ruckstandes wird Calcium mit AGTA [Athylenglykol-bis-(2-aminoathylather)-tetraacetat] bei pH 13 mit Calcein als Indikator fluorometrisch titriert. Um die Fallung von Magnesium bei pH 13 zu verhindern, wird es mit Tartrat komplexiert. Mg2+, Zn2+ sowie Phosphat konnen bis zu einem l00fachem Uberschus vorhanden sein.

Zur Bestimmung von Ascorbinsaure in Fruchtsaften und Limonaden wurde die Differential Pulspolarographie herangezogen. Ascorbinsaure gibt zwei Peaks, deren Hohen nicht nur von der Konzentration, sondern auch vom pH und von bestimmten Gerateparametern abhangen. Auch der Einflus verschiedener Inhalts- und Zusatzstoffe in Fruchtsaften und Limonaden wurde untersucht.

Die inverse Differential-Puls-Polarographie wurde zur Bestimmung kleinster Quecksilbermengen herangezogen. Bei Verwendung einer wachsimpragnierten Graphitelektrode gelang es, 10 ng Hg/ml zu bestimmen. Mit Hilfe einer „Glassy Carbon“-Elektrode konnte noch 1 ng/ml erfast werden.

Zur Bestimmung von N,N′-Dimethylformamid in Luftproben wurde ein neues gaschromatographisches Verfahren ausgearbeitet, mit dem noch ppb-Mengen erfasbar sind. Die Luft wird durch zwei Frittenwascher geleitet, die mit Wasser beschickt sind, wobei DMF quantitativ absorbiert wird. Die wasrige Losung wird anschliesend an einer Saule mit Carbowax 20 M auf Carbopack B chromatographiert. Die quantitative Bestimmung erfolgt durch einen Flammenionisationsdetektor.

Eine Methode zur Bestimmung von Isothiocyanaten (z. B. Allyliso-thiocyanat) in Milligramm-bzw. Mikrogramm-Mengen wurde ausgearbeitet. Das durch Wasserdampfdestillation abgetrennte Isothiocyanat wird mit Ammoniak zum entsprechenden Thioharnstoff umgesetzt. Durch Reaktion mit Cadmiumacetat erhalt man Cadmiumsulfid; dieses wird entweder jodometrisch nach Zimmermann oder photometrisch (als Methylenblau) nach Roth bestimmt.

Ein Beitrag zur voltametrischen Titration von Metallionen mit ADTA (nach Kraft) wurde mitgeteilt. Als Kathode dient blankes Platin, als Anode wird ein Metalloxid verwendet. Dieses wird vor der Titration aus einer Metallsalzlosung auf einer Platinelektrode abgeschieden. Folgende Oxide eignen sich: Tl2O3, Co2O3, MnO2, PbO2 und PtOx. Die Elektroden werden mit konstanter Stromstarke polarisiert, die je nach dem Oxid 2 bis 10 Mikroampere betragt. Die Oxidanode indiziert einen Uberschus an ADTA durch eine plotzliche Spannungsanderung. Deren Grose hangt sehr stark vom pH-Wert der Losung sowie vom Oxid ab. Fur jede Oxidanode wird die optimale Polarisationsstromstarke und ihr Verwendungszweck angegeben. Weiters wird die praktische Verwendung der Oxidelektroden bei der Bestimmung verschiedener Metallionen beschrieben.

Eine Methode zur direkten Bestimmung von Chinin in Tonic-Wassern und Limonaden wurde ausgearbeitet. Hierzu wurde die Differential-Pulspolarographie verwendet. Chinin gibt bei dieser Methode — je nach pH-Wert-ein oder zwei Peaks. Die Hohe dieser Peaks hangt bei vorgegebenem pH von der Konzentration ab. Auch das Potential im Peakmaximum ist eine Funktion des pH. Eine maximale Detektion erhalt man zwischen pH 6,5 und 9. Aus den gewonnenen Daten war auch ein Einblick in die Elektrodenreaktionen moglich.

Die Wechselstrompolarographie eignet sich gut, um den Kupfergehalt von Bodenproben aus Fischteichen zu bestimmen. Gleichzeitig ist es moglich, auch Blei, Cadmium und Zink bei Anwesenheit von Eisen zu erfassen. Das Leitsalz Triathanolamin/Natronlauge (pH 12) ermoglicht die Bestimmung von Kupfer neben Eisen. Fur die Bestimmung von Pb, Cd und Zn in Anwesenheit von Eisen bewahrt sich Ammoniumacetat/Hydroxylamin-Hydrochlorid. Zur Charakterisierung der Bodenprobe wird der Gesamt-Kupfergehalt, das extrahierbare und okkludierte Kupfer sowie die Kupfer-Sorption (Sattigungswert) durch AC-Polarographie bestimmt.

Eine Methode wurde entwickelt, um Calcium und Magnesium in naturlichen Wassern bei pH 10 an einer Tl2O3-Anode unter Verwendung von ADTA und AGTA als Titrationsmittel zu bestimmen. Die dabei verwendete Apparatur eignet sich gut fur Serienbestimmungen, da die Zugabe des Titrationsmittels und die Aufzeichnung des Kurvenverlaufs automatisch erfolgen. Die Auswertung wird graphisch durchgefuhrt.

Eine Methode zur quantitativen Bestimmung von Isosorbiddinitrat und Diazepam in pharmazeutischen Praparaten wurde ausgearbeitet. Diese Verbindungen werden durch HPLC an Nucleosil C18 getrennt (mobile Phase: Methanol-Ethylacetat-Acetatpuffer). Die Detektion erfolgt amperometrisch an einer stationaren Quecksilber-Tropfelektrode mit Pulstechnik bei −1,15 V (Basispotential: −0,4 V, Pulsamplitude: −0,75 V). Die Nachweisgrenze liegt bei 8 ng Isosorbiddinitrat bzw. 20 ng Diazepam (S/N=3). Bei Praparaten, die zusatzlich Lecithine oder Triglyceride enthalten, ist eine vorherige Aufarbeitung erforderlich. Lecithine werden durch Adsorption an einer Silicagel-Kartusche abgetrennt, Triglyceride durch Extraktion mit Petrolether. Die amperometrische Detektion wird auch mit der UV-Detektion verglichen. Isosorbiddinitrat wird amperometrisch empfindlicher detektiert. Bei Diazepam hingegen ist die UV-Absorption gunstiger.

Dimethylamin (DMA) in Luft wird in 0,1N Schwefelsaure absorbiert und angereichert. Die saure Absorptionslosung wird in die Septumflasche eines Head-Space-Samplers ubergefuhrt und mit festem Natriumhydroxid alkalisch gemacht. Nach Einstellung des Gleichgewichts wird aus dem Dampfraum eine gemessene Menge Dampf entnommen und gaschromatographisch analysiert. Zur Detektion wird ein FID verwendet. Bei einer Probemenge von 250 l Luft last sich noch ein Zehntel des MIK-Wertes (=2μg DMA/m3) bestimmen.

Es wurde das elektrochemische Verhalten von Oxalsaure, Kupferron und Diathyldithiocarbamat an Oxidelektroden ermittelt. Es ergab sich ein Zusammenhang zwischen den oxydierbaren Gruppen dieser Verbindungen und der Stromanderung, die an den Oxidelektroden hervorgerufen wird. Die Stromanderung an den Oxidelektroden ist proportional dem Ausmas der Reaktion zwischen Chelatbildner und Oxid und dadurch stark pH-abhangig. Eine titrimetrische Anwendung der erwahnten Fallungsreagentien ist dann moglich, wenn sich der pH-Bereich der Fallung mit dem pH-Bereich der Indikation uberlappt. Dadurch konnen eine Reihe von Metallionen amperometrisch titriert werden. Mit Oxalsaure werden vor allem die Seltenen Erden erfast. Mit Kupferron kann man 3wertige und einen Teil der 4wertigen Ionen titrieren. Mit Diathyldithiocarbamat sind die Schwefelwasserstoffgruppe sowie die meisten Ionen der Ammonsulfid-Gruppe bestimmbar.

Zur amperometrischen Titration von Metallionen mit ADTA wird eine Oxidelektrode verwendet (Platinelektrode, die mit einer Oxidschicht, z.B. mit Tl2O3 oder Ni2O3, Co2O3, MnO2, PbO2, PtOx, Bi2O5, uberzogen ist). Diese Elektroden sind uber einen relativ grosen pH-Bereich aktiv und stabil. Sie sprechen auf uberschussiges ADTA (nach dem Aquivalenzpunkt) an. Es werden die genauen Arbeitsbedingungen angegeben: a) zur Einzelbestimmung von Metallionen; b) zur Bestimmung von zwei Ionen nebeneinander. Im Falle b) wird von der Tatsache ausgegangen, das die Stabilitat von Metall-ADTA-Komplexen stark vom pH-Wert der Losung abhangt. Zunachst wird ein pH-Wert eingestellt, bei dem nur ein Ion einen stabilen Komplex (Stabilitatskonstante groser als 107) bildet. Dieses Ion kann dann ohne Storung durch das andere Ion mit ADTA titriert werden. Sodann wird ein pH-Wert eingestellt, wo auch das zweite Ion einen hinreichend stabilen Komplex bildet.

Es wurde ein Verfahren ausgearbeitet, um die Absorptionsleistung von MnO2-Praparaten zahlenmasig bestimmen zu konnen. Hierbei zeigte sich, das die Absorptionsleistung von der angebotenen NO2-Menge, von der Schichtlange und von der Stromungsgeschwindigkeit des Spulgases abhangt. Diese Faktoren wurden auch rechnerisch erfast. Es gelang weiters, die analytisch auswertbare Kapazitat (Durchbruchskapazitat) von MnO2-Praparaten aus der empirisch leicht bestimmbaren Gesamtkapazitat zu berechnen.

Die fruher beschriebene Apparatur zur Bestimmung der primaren Aminogruppe wurde verbessert. Auch die Handhabung wurde vereinfacht. Die einzelnen Teile der Apparatur (Desaminierungsteil, Absorber) konnen nun unabhangig voneinander ausgewaschen und luftfrei gemacht werden. Die Absorber sind fur etwa 30 bis 50 Analysen benutzbar, bevor sie erneuert werden mussen.

Die bereits fruher beschriebene Apparatur zur serienmasigen Mikrobestimmung des Kohlenstoffs und Wasserstoffs, bei der Kupfer zur Reduktion der Stickoxyde verwendet wurde, konnte weiter vereinfacht werden. Im einzelnen werden die Bedingungen fur eine quantitative Oxydation organischer Substanzen imLuftstrom, sowie die Herstellung einer besonders aktiven Kupferschicht, die fur eine Tagesserie von 14 Bestimmungen ausreicht, beschrieben. Ferner werden interessante Einzelheiten uber den Verbrennungsvorgang mitgeteilt.

The reaction of different cracked products (H2, C2H2, C2H4, COS and CS2) with iodine pentoxide and anhydroiodic acid was investigated, because the cracked products develop iodine and thereby cause a positive error in the determination of oxygen. The development of iodine depends on the following factors: 1. on the chemical composition of the oxidation zone; much more iodine is developed on iodine pentoxide than on anhydroiodic acid; 2. on the cracked products, the development of iodine increases in the sequence H2 < C2H4 < C2H2 < COS < CS2; 3. on the temperature of the oxidation zone. The oxidation curves of COS and CS2 are ascending steeply already between 40 and 50° C and coincide with the oxidation curve of carbon monoxide. The oxidation curves of C2H2, C2H4 and H2 are shifted to higher temperatures.

In der vorliegenden Arbeit wurde versucht, einen Einblick in die bisher ungeklarte anomale Reaktion verschiedener Aminosauren mit salpetriger Saure zu gewinnen. Es wird wahrscheinlich gemacht, das bei der Desaminierung der Aminosauren nicht nur eine Hydroxyverbindung entsteht, sondern auch die entsprechende Nitrolsaure. Bei den α-Aminosauren wird dabei Kohlendioxyd in Freiheit gesetzt. Das Ausmas der Bildung von Nitrolsaure hangt von der Konzentration der Nitritlosung ab; unter den Bedingungen der Aminostickstoffbestimmung entstehen etwa 30%. Die Nitrolsaure ist der Ausgangspunkt fur die anomale Gasentwicklung. Bei Temperaturerhohung reagiert namlich die Isonitrosogruppe der Nitrolsaure mit salpetriger Saure, wobei unter Entwicklung von N2 und N2O die entsprechende Carbonylverbindung entsteht. Bei Tyrosin bilden sich noch zusatzliche Gasmengen infolge der anomalen Reaktion der Phenolgruppierung. Bei Cystin ruhrt das Mehrvolumen von der Reaktion der Disulfidgruppierung mit salpetriger Saure her.

Es wird eine Vorrichtung beschrieben, die die Vergasung der organischen Substanz nach dem Prinzip der manuellen Verbrennungstechnik von F. Pregl automatisch durchfuhrt. Hierzu wird ein Stromungsmesser verwendet, der mit Elektroden versehen ist, die uber ein Relais den Vorschub des beweglichen Brenners regeln. Durch Kombination mit einem Schreibgerat ist es erstmals moglich, den Vorgang bei der individuellen Verbrennung graphisch darzustellen.

Die Verbrennung an einer Duse kann man zur Spurenbestimmung von Schwefel, Halogenen und Phosphor in organischen Stoffen einsetzen. Ein zusatzliches Zundgas (wie Wasserstoff) ist nicht notwendig. Um eine moglichst grose Probemenge durchsetzen zu konnen, mus die Probezufuhr gleichmasig und der Sauerstoffzufuhr angepast sein. Verschiedene Methoden der Probezufuhr wurden erprobt: pro Minute konnen 150 bis 300 mg Probe oxydiert werden.

Bisher konnten Mikro-Chlor- und Brombestimmungen mittels Peroxydaufschlus, abgesehen von potentiometrischen Verfahren, nur gravimetrisch vorgenommen werden, wodurch fur eine Analyse verhaltnismasig viel Zeit benotigt wurde. Die herkommlichen volumetrischen Methoden sind infolge der schwierigen Titrationsbedingungen nicht anwendbar. In dieser Arbeit wird nun eine argentometrische Titration mit Jodstarke-Indikation angegeben, die auch bei hoher Salzkonzentration und grosem Volumen einen scharfen Endpunkt liefert. Bei der Anwendung auf den Peroxydaufschlus ergaben sich zunachst gewisse Storungen, die durch eine geeignete Aufarbeitung des Aufschlusses beseitigt wurden. Nach diesem Verfahren konnen nunmehr Mikrobestimmungen von Halogen in organischen Substanzen mittels Peroxydaufschlus bei gleicher Genauigkeit wesentlich schneller als bisher ausgefuhrt werden. Das Verfahren eignet sich auch zur Bestimmung geringster Halogenmengen in organischem Material.

Die anomale Reaktion von Phenolen mit salpetriger Saure konnte an Hand von Di- und Triphenolen weitgehend geklart werden. Demnach kann das anomale Gasvolumen (N2 und N2O) durch drei Reaktionsfolgen zustande kommen. Bei derersten Reaktionsfolge tritt in p-Stellung zur phenolischen Hydroxylgruppe eine Nitrosogruppe ein. Durch Umlagerung entsteht daraus ein tautomeres Chinonoxim. Dieses geht durch Reaktion mit uberschussiger salpetriger Saure unter Entwicklung eines Mols (N2 + N2O) in das entsprechende p-Chinon uber. Diezweite Reaktionsfolge tritt ein, wenn die o-Stellung zum phenolischen Hydroxyl nitrosiert wird. Die entstandene Nitrosoverbindung wird durch uberschussige salpetrige Saure in die Diazoniumverbindung ubergefuhrt, die dann 1 Mol Stickstoff abgibt. Mit derdritten Reaktionsfolge ist dann zu rechnen, wenn sich intermediar ein p-Chinon bildet. Dies ist der Fall bei der Reaktionsfolge 1 oder, wie bei Hydrochinon, durch unmittelbare Oxydation. p-Chinone werden von uberschussiger salpetriger Saure unter Entwicklung von Kohlendioxyd und (N2 + N2O) zu Oxalsaure abgebaut. Aus den gefundenen Werten wurde die Bruttogleichung fur den Abbau der Chinone aufgestellt. Es konnten auch die Gasmengen, die bei jeder dieser Reaktionsfolgen zu erwarten sind, berechnet werden.
