No. of bitstreams: 0 Previous issue date: 2020-06-26 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Background:Photobiomodulation therapy (PBMT) and creatine (Cr) intake have been used in conjunction with heavy training, but little is known about their possible effects during a long-term training program. Objective:We assessed long-term use of PBMT and Cr in an exercise training program. Methods:Twenty-five male Wistar rats weighing similar to 300 g were randomly allocated to one of five groups: a nontraining control group, a training group, a training group receiving Cr, a training group receiving PBMT, and a training group receiving both PBMT and Cr. The training program consisted of 12 weeks of daily swimming training. PBMT was delivered in six points with a laser device (808 nm, 100 mW, 30 s per point of irradiation, 3 J, 75 J/cm(2)). Results:All training groups showed significantly higher peak force and longer time to 50% decay of force, and lower creatine kinase (CK) levels than the nontraining control group, thus confirming the benefit of the training program. In all outcomes related to muscle performance, the groups receiving PBMT with or without Cr supplement performed significantly better (p < 0.05) peak force and time of force decay during an electrical stimulation protocol than all the other groups. In addition, CK levels were also significantly lower for the PBMT groups than for the other groups. Conclusions:We conclude that PBMT alone or in conjunction with Cr supplement during a 12-week training program resulted in significantly better muscle performance and lower levels of CK, a biochemical marker of muscle damage. A completely randomized design which consisted of 11 treatments and three replicates each was used. The treatments included a control diet (CD) containing 55% of nonpurified natural ingredients and 45% of purified synthetic amino acids and ingredients, and other ten isonitrogenous and isoenergetic EAA limiting diets (LDs), each being deficient in 44.4 ± 0.02% of the respective EAA. Pacu juveniles with initial average body weight of 6.22 ± 0.09 g were distributed among 33 fiber glass tanks. Fish were fed with semipurified and extruded diets for 113 days two times a day until apparent satiation. The ideal ratio of each dietary EAA was calculated on the basis of the relationship between body N retention and amount of EAA deleted from the respective EAA LD. Based on the AA deletion method, the ideal ratios of digestible EAAs for pacu juveniles, relative to lysine requirement of 100% were estimated as: methionine 14.6%, threonine 35.0%, tryptophan 6.6%, arginine 62.8%, histidine 13.6%, isoleucine 26.3%, leucine 43.7%, phenylalanine 27.2%, and valine 35.8%. Thus, the aim of this study was to evaluate the changes on soil microbial biomass and enzyme activity when native vegetation (NV) was converted to cropland (CL), such as maize or sugarcane in six oxisols from Sao Paulo state, Brazil. Soil microbial biomass C (MBC) and N (MBN), and the activity of arylsulphatase, dehydrogenase and fluorescein diacetate hydrolysis (FDA) were assessed in samples collected at 0-0.20 m. In general, MBC was higher under NV than CL (about + 40%), while MBN and FDA did not show a consistent pattern between NV and CL. All soils showed higher values of arylsulfatase (increased from 101 to 717%) and dehydrogenase (increased 15 to 220%) under NV than CL. In conclusion, soil microbial biomass C is usually higher under native vegetation than cropland. Arylsulphatase and dehydrogenase were the attributes that presented better differentiation between native and cropped soils. No. of bitstreams: 0 Previous issue date: 2019-04-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Objective: The present study aimed to describe the morphology of oral apparatus and oral cavity of bullfrog tadpoles during their development and metamorphosis. Design: The oral apparatus and oropharyngeal cavity of tadpoles from hatching up to metamorphosis stage was dissected for further analysis. These structures were fixed in Karnovsky solution, afterwards in osmium tetroxide and metalized in palladium gold and electron-micrographed using the scanning electron microscope. Results: The development of oral apparatus started with the formation and keratinization of the jaw sheaths and labial teeth followed by the formation of marginal and sub-marginal papillae. Degeneration of oral apparatus and formation of mouth was observed during metamorphosis. From stage-42 (metamorphic climax) to stage-43, the jaw sheath and labial tooth rows were disappeared progressively while the size and number of labial papillae were decreased. At stage-44, mouth formation started with the development of anterior and posterior labium though the labial papillae were still present. At stage-45 and 46, mouth was already formed, being very similar to the adult and characterized by the progressive increase in size. Conclusion: The sequence of events that happen during the development of oral apparatus of Lithobates catesbeianus Shaw, 1802 tadpoles follows the same pattern as occur in other anuran species but metamorphic atrophy of the oral apparatus follows the sequence of morphogenesis. Methods: Forty-four female Wistar rats were randomly divided into four experimental groups: Lesion control (L), Lesion and FS (LS), Lesion and AE (LE) and Lesion and FS associated to AE (LSE). The edema volume (EV), collagen ratio, and histopathological analysis were evaluated after 7, 14, and 21 days of partial tendon transection. Results: The EV was statistically reduced for all treatment groups after 7 and 21 days when compared to L group. The LS and LSE had the highest EV reduction after 21 days of treatment. The FS group didn't induce tissue necrosis or infections on the histopathological analysis. It was observed tenocytes proliferation, granulation tissue and collagen formation in the tendon partial transection area in the FS group. The LSE demonstrated higher amount of granulation tissue and increased the collagen deposition at the injury site. Conclusions: Our data suggests that the therapeutic potential of the association of heterologous fibrin biopolymer sealant with aquatic exercise program should be further explored as it may stimulate the regeneration phase and optimize calcaneal tendon recovery. No. of bitstreams: 0 Previous issue date: 2021-03-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) National Council for Scientific and Technological Development Electrospun ultrathin fibrous scaffold filed with synthetic nanohydroxyapatite (nHAp) and graphene nanoribbons (GNR) has bioactive and osteoconductive properties and is a plausible strategy to improve bone regeneration. Poly(butylene-adipate-co-terephthalate) (PBAT) has been studied as fibrous scaffolds due to its low crystallinity, faster biodegradability, and good mechanical properties; however, its potential for in vivo applications remains underexplored. We proposed the application of electrospun PBAT with high contents of incorporated nHAp and nHAp/GNR nanoparticles as bone grafts. Ultrathin PBAT, PBAT/nHAp, and PBAT/nHAp/GNR fibers were produced using an electrospinning apparatus. The produced fibers were characterized morphologically and structurally using scanning electron (SEM) and high-resolution transmission electron (TEM) microscopies, respectively. Mechanical properties were analyzed using a texturometer. All scaffolds were implanted into critical tibia defects in rats and analyzed after two weeks using radiography, microcomputed tomography, histological, histomorphometric, and biomechanical analyses. The results showed through SEM and high-resolution TEM characterized the average diameters of the fibers (ranged from 0.208 mu m +/- 0.035 to 0.388 mu m +/- 0.087) and nHAp (crystallite around 0.28, 0.34, and 0.69 nm) and nHAp/GNR (200-300 nm) nanoparticles distribution into PBAT matrices. Ultrathin fibers were obtained, and the incorporated nHAp and nHAp/GNR nanoparticles were well distributed into PBAT matrices. The addition of nHAp and nHAp/GNR nanoparticles improved the elastic modulus of the ultrathin fibers compared to neat PBAT. High loads of nHAp/GNR (PBATnH5G group) improved the in vivo lamellar bone formation promoting greater radiographic density, trabecular number and stiffness in the defect area 2 weeks after implantation than control and PBAT groups. Methods: A 1.5 cm ostectomy was performed in the radial diaphysis of 60 New Zealand white rabbits. The rabbits were divided into three groups according to surgical treatment of the bone defect (group I - control, group II - bone graft, group III - 3D PLLA). Each group was divided into four subgroups with different radiographic and histopathologic evaluation times (T1 - 15 days, T2 - 30 days, T3 - 60 days, T4 - 90 days). Results: The implant group had greater clinically lameness (p = 0.02), edema (p = 0.007), pain (p = 0.04) and more complications at the surgical site (p = 0.03). Histologically, this group showed greater congestion (p = 0.04), hemorrhage (p = 0.04) and inflammation. Osteogenesis was microscopically similar between days (p = 0.54) and treatments (p = 0.17), even though radiographically, more effective bone healing occurred in the graft group (II), with more callus and bone bridge formation. Conclusions: The customization of a 3D PLLA/HA scaffold was successful. However, in animals receiving the polymer-ceramic composite less bone callus and bone bridge was formed compared to the graft group. No. of bitstreams: 0 Previous issue date: 2020-12-14 Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) The fibrous scaffolds for bone tissue engineering that mimic the extracellular matrix with bioactive and bactericidal properties could provide adequate conditions for regeneration of damaged bone. Electrospun ultrathin fiber covered with nano-hydroxyapatite is a favorable fibrous scaffold design. We developed a fast and reproducible strategy to produce polyvinylidene fluoride (PVDF)/nano-hydroxyapatite (nHAp) nanofibrous scaffolds with bactericidal and bioactive properties. Fibrous PVDF scaffolds were obtained first by the electrospinning method. Then, their surfaces were modified using oxygen plasma treatment followed by electrodeposition of nHAp. This process formed nanofibrous and superhydrophilic PVDF fibers (133.6 nm, fiber average diameter) covered with homogeneous nHAp (202.6 nm, average particle diameter) crystals. Energy-dispersive X-ray spectrometry demonstrated the presence of calcium phosphate, indicating a Ca/P molar ratio of approximately 1.64. X-ray diffraction, Fourier transform infrared spectroscopy, and Raman spectroscopy spectra identified beta-phase of nHAp. Thermal analysis indicated a slight reduction in stability after nHAp electrodeposition. Bactericidal assays showed that nHAp exhibited 99.8% efficiency against Pseudomonas aeruginosa bacteria. The PVDF/Plasma and PVDF/nHAp groups had the highest cell viability, total protein, and alkaline phosphatase activity by 7 days after exposure of the scaffolds to MG63 cell culture. Therefore, the developed scaffolds are an exciting alternative for application in bone regeneration. No. of bitstreams: 0 Previous issue date: 2020-11-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Due to increasing demand of ingredients of animal origin, studies on the availability of nutrients in ingredients of plant origin are worthy in carnivorous species including bullfrog. This study aimed to determine apparent digestibility coefficients (ADCs) of amino acids (AAs) in three ingredients as plant protein sources (soybean protein concentrate (SPC), corn gluten meal 60 % (CGM), soybean meal (SM)) and two ingredients as plant energy sources (corn grain (CG) and wheat bran (WB)) which commonly use in bullfrog diets. A reference diet and five test diets containing 69.5 % of the reference diet, 30 % of each test ingredient and 0.5 % of chromium (III) oxide as an inert marker were prepared. Among plant protein ingredients, CGM showed better digestibility of essential amino acids (EAAs) (> 80 %) at the three post-metamorphic (initial, growing and finishing) phases of bullfrog development, except tryptophan which presented a low digestibility at growing and finishing phases. Among plant energy ingredients, CG presented better digestibility of AAs at both initial and growing phases than WB which showed better digestibility only at finishing phase. It was observed that methionine and lysine are the most limiting EAAs in tested ingredients, indicating that additional amounts of these AAs should be supplemented when these ingredients are used in bullfrog diets. No. of bitstreams: 0 Previous issue date: 2020-09-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) This 45-day work aimed to determine the response of juvenile pacu (Piaractus mesopotamicus) to balanced digestible protein (BDP) and to use these responses to determine whether the optimum economic levels of BDP would differ depending on the form in which the fish is sold. Six isoenergetic diets containing 163, 201, 238, 272, 315 and 348 g/kg BDP (dry-matter basis) were prepared through the serial dilutions of a high-protein diet with the low-protein diet. Fish (initial average body weight, 10.82 ± 0.14 g) were fed with respective experimental diets three times a day until apparent satiation. The optimum biological level of BDP was calculated as 326 g/kg (dry-matter basis) by the quadratic regression model for maximum body weight gain of juvenile pacu. To maximize economic returns (US$/kg) for different end products, the optimum economic levels of BDP were calculated as 311, 317 and 319 g/kg (dry-matter basis) by an economic model for whole-body, eviscerated and sliced juvenile pacu respectively. This finding revealed that obtaining maximum biological performance of fish in the case of high feed costs or low prices of the end product in consumer market would substantially reduce the economic returns. Here, we propose the combination of PBAT [poly(butylene-adipate-co-terephthalate)] and synthetic nHAp (at 3 and 5 wt%). PBAT is a relatively a new polymer with low crystallinity and attractive biodegradability and mechanical properties for orthopedic applications, however, with a still underexplored potential for in vivo applications. Then, we performed a careful biological in vitro and in vivo set of experiments to evaluate the influence of PBAT containing two different nHAp loads. For in vitro assays, osteoblast-like MG63 cells were used and the bioactivity and gene expression related to osteogenesis were evaluated by qRT-PCR. For in vivo experiments, twenty-four male rats were used and a tibial defect model was applied to insert the scaffolds. Micro-computed tomography (Micro-CT) and histological analysis were used to assess e bone neoformation after 6 weeks of implantation. Three point flexural tests measured the mechanical properties of the neoformed bone. All scaffolds showed promising in vitro properties, since they were not cytotoxic against MG-63 cells and promoted high cell proliferation and formation of mineralized nodules. From a mechanistic point-of-view, nHAp loading increased hydrophilicity, which in turn allowed for a better adsorption of proteins and consequent changes in the phenotypic expression of osteoblasts. nHAp induced better cellular responses on/in the scaffolds, which was mainly attributed to its osteoconductive and osteoinductive properties. Micro-CT images showed that nHAp at 3% and 5 wt% led to more effective bone formation, presenting the highest bone volume after 6 weeks of implantation. Considering the three point flexural tests, 5 wt% of nHAp positively influenced the flexural mode of the neoformed bone, but the stiffness was similar between the 3% and 5 wt% groups. In summary, this investigation demonstrated great potential for the application of these novel scaffolds towards bone regeneration and, thus, should be further studied. (C) 2017 Elsevier B.V. No. of bitstreams: 0 Previous issue date: 2020-07-27 Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Polycaprolactone (PCL) is a biocompatible, biodegradable synthetic polymer which in combination with nanohydroxyapatite (nHAp) can give rise to a low cost, nontoxic bioactive product with excellent mechanical properties and slow degradation. Here we produced, characterized and evaluated in vivo the bone formation of PCL/nHAp scaffolds produced by the rotary jet spinning technique. The scaffolds produced were firstly soaked into simulated body fluid for 21 days to also obtain nHAp onto PCL/nHAp scaffolds. Afterwards, the scaffolds were characterized by scanning electron microscopy (SEM), energy dispersive spectroscopy and Raman spectroscopy. For in vivo experiments, 20 male Wistar rats were used and randomly divided in 4 experimental groups (n = 5). A critical defect of 3 mm in diameter was made in the tibia of the animals, which were filled with G1 control (clot); G2-PCL scaffold; G3-PCL/nHAp (5%) scaffold; G4-PCL/nHAp (20%) scaffold. All animals were euthanized 60 days after surgery, and the bone repair in the right tibiae were evaluated by radiographic analysis, histological analysis and histomorphometric analysis. While in the left tibias, the areas of bone repair were submitted to the flexural strength test. Radiographic and histomorphometric analyses no showed statistical difference in new bone formation between the groups, but in the three-point flexural tests, the PCL/nHAp (20%) scaffold positively influenced the flexural mode of the neoformed bone. These findings indicate that PCL/nHAp (20%) scaffold improve biomechanical properties of neoformed bone and could be used for bone medicine regenerative. Poly(butylene adipate-co-terephthalate) (PBAT) solutions containing different concentrations of nano-hydroxyapatite (nHAp) (1, 2, 3, 4, 5 and 6% wt) were evaluated to assess their potential to produce scaffolds via electrospinning. The characteristics of these solutions were evaluated using surface tension analysis. Different solutions were electrospun and characterized using scanning electron microscopy, X-ray diffractometry (XRD), infrared spectroscopy (FTIR), and differential scanning calorimetry analysis. In vitro and in vivo experiments were performed using MG-63 osteoblast cells and male Wistar rats. The MTT assay was used to evaluate the cytotoxicity of scaffolds. Microtomography and a three-point bend test were used to analyze parameters of bone neoformation 4 weeks after implantation. We observed a requirement for solutions containing nHAp to reach chemical stability to produce ultrathin fibers. FTIR and XRD data demonstrated the presence of carbonate and phosphate groups, and thermal analysis showed a reduction in crystallinity of the nanocomposites when the concentration of nHAp was increased. None of the scaffolds analyzed demonstrated cytotoxicity when compared to controls. All of the PBAT/nHAp scaffolds analyzed promoted bone repair; however, a solution of PBAT with 3% nHAp improved bone volume, force, and stiffness when compared to controls. No. of bitstreams: 0 Previous issue date: 2020-06-30 The present study aimed to determine the optimal ratios of essential amino acids (EAAs) for Nile tilapia (Oreochromis niloticus) juveniles (150–200 g) using the deletion method. A total of 880 Nile tilapia juveniles (158.78 ± 0.9 g) were randomly distributed among 44 tanks (20 fish per tank/replicate) of 1500-L capacity each. During the 53 days feeding trial, fish were fed with the respective diets and daily feed intake was regularly recorded. In order to prevent any feed wastage, the fish were hand-fed three times a day until apparent satiation. A balanced diet (BD) and other ten diets were formulated by the deletion method, where the BD diet was adjusted to result in a reduction of 45% of each tested EAA. A group of fish at the beginning and at the end of the experiment was euthanized for the determination of the carcass nitrogen (N) composition. Based on the results of the present study, the optimal balance of essential amino acids for Nile tilapia juveniles weighing 150 and 200 g (g 16 g−1 N) was estimated as, arginine 4.88, lysine 5.87, threonine 6.02, histidine 1.73, isoleucine 3.26, leucine 4.82, methionine 2.36, phenylalanine 3.66, valine 3.46 and tryptophan 0.87. Recently, some biomaterials, based on carbon nanostructures and nanohydroxyapatite (nHAp), have been investigated as bone substitutes in order to improve regeneration. The aim of this study was to access the expression of some RNA transcripts (involved in the process of osteoblast differentiation) by mesenchymal stem cells cultured over different nanocomposite surfaces. A multi-walled carbon nanotube (MWCNT) was firstly grown using chemical vapor deposition and then exfoliated using chemical and oxygen plasma treatments to obtain graphene nanoribbons (GNR). The hybrid composites nHAp/GNR were prepared using the wet method assisted by ultrasound irradiation with different amounts of GNR (1.0, 2.0 and 3.0wt %). Five groups were tested in cell cultures. Group 1: synthesized nHAp; Group 2: synthesized GNR; Group 3: nHAp and 1.0% of GNR; Group 4: nHAp and 2.0% of GNR and group 5: nHAp and 3.0% of GNR. Real time reverse transcription polymerase chain reactions were performed, and all data was submitted to Kruskal Wallis and Dunn tests, at a significance level of 5%. As a result, three nanocomposites with different proportions of GNR were successfully produced. After cell culture, the expression of osteogenic genes demonstrated no significant differences among the groups and periods. However, bone morphogenetic protein II (BMP II), integrin binding sialoprotein (IBSP), and Osterix highest expressions were observed in the group containing 3.0% of GNR. In conclusion, our hybrid composites may be useful in bone interventions requiring mesenchymal stem cell differentiation into osteoblasts for healing. [GRAPHICS] . Hatchability was not affected by addition of glycosaminoglycans and vitamin C in ovo. However, chicks that received 4 ug additive showed 2.86% reduction in total mortality when compared with chicks from non-injected eggs. Moreover, tibia area and femur bone mineral density increased in chicks from eggs injected with 2.16 and 6.00 mu g of additive, respectively. In ovo feeding with glycosaminoglycans and vitamin C can benefit bone development in embryos and reduce total mortality during the incubation period. The trial was conducted using 108 quail distributed in a completely randomized experimental design, with three treatments (0, 2.5, and 5.0 g chamomile/kg of feed), six replicates, and six birds per treatment, evaluated in six measures repeated in time (14, 28, 42, 56, 70, and 84 days of trial). The inclusion of chamomile presented a quadratic effect on sitting behavior, estimating the inclusion of 1.8 g chamomile/kg to maximize this behavior. There was a decreasing linear effect for aggressive pecking, that is, the higher the inclusion of chamomile in the diet, the lower the expression of this behavior. The inclusion of 1.8-5.0 g chamomile/kg in a Japanese quail diet reduces the behavior of aggressive pecking, in addition to keeping the birds seated longer. These results are innovative because they show in the literature for the first time that chamomile supplemented in Japanese quail diets has the capacity to modulate the behavior of the quail, leading to an improvement in the welfare of quail raised in cages. Thus, this study aimed to understand the processes influencing the physical, chemical, and biological properties of the water due to land uses and occupation in the Ipe stream basin, in Ilha Solteira - SP (Brazil), regarding the dependence of the analyzed variables. Monitoring of chemical, physical, and biological water quality was performed from 2006 to 2011, and the records of land use and occupation in 2011. A factorial multivariate analysis allowed us to understand that the processes in densely populated areas increased degradation of water by the input of organic loads, yet those of farming areas enhanced the degradation of the chemical and physical quality of spring waters. Therefore, installing irrigation systems near populated areas, for crops consumed in natura, may compromise this agricultural activity, as well as the absence of water filtration systems downstream agricultural areas. Furthermore, we concluded that multivariate statistics is a powerful tool to detect these influencing processes in water quality. In this study, this type of analysis played an important role isolating the processes acting on water quality.
