Three new 2,4-diaminoquinazolines, the 5,6-difluoro, 6,7-difluoro and 7,8-difluoro isomers were prepared by the reaction of the requisite trifluorobenzonitrile and guanidine carbonate. Surprisingly, 2,3,6-trifluorobenzonitriles gave 2,4-diamino-5,6-difluoroquinazoline exclusively as determined by high resolution nuclear magnetic resonance spectroscopy. On the other hand, 3-amino-2,6-difluorobenzonitrile on reaction with guanidine carbonate yielded only 5-fluoro-2,4,8-triaminoquinazoline. This compound was subsequently converted to 8-chloro-2,4-diamino-5-fluoroquinazoline using the Sandmeyer procedure. The nitration of 2,4-diamino-8-fluoroquinazoline occurred exclusively at position six yielding 2,4-diamino-8-fluoro-6-nitroquinazoline, which upon reduction with stannous chloride afforded 8-fluoro-2,4,6-triaminoquinazoline. In a similar fashion 7-fluoro-2,4-diaminoquinazoline underwent nitration at position six and was then reduced to give 7-fluoro-2,4,6-triaminoquinazoline. Finally, both of these triaminoquinazolines were converted to the 6-chloro derivatives under Sandmeyer conditions to yield 6-chloro-2,4-diamino-8-fluoroquinazoline and 6-chloro-2,4-diamino-7-fluoroquinazoline, respectively.

The target folate analogue 5-trifluoromethyl-5,8-dideazafolic acid has been elaborated in a seven-step reaction sequence beginning with 2-fluoro-6-trifluoromethylbenzonitrile. The bridge-reversed isomer 5-trifluoromethyl-5,8-dideazaisofolic acid was prepared in six steps using the common intermediate 2,6-diamino-3,4-dihydro-4-oxo-5-trifluoromethylquinazoline. The key intermediate 2-amino-3,4-dihydro-4-oxo-5-trifluoromethyl-quinazoline was prepared using two distinct synthetic routes and the resulting products were found to be identical.

The new folate antagonist, 5-fluoro-5,8-dideazaisoaminopterin was synthesized in four steps beginning with 2,4-diamino-5-fluoroquinazoline. It was found to be a potent inhibitor of human dihydrofolate reductase. Against L1210 leukemia in mice, 5-fluoro-5,8-dideazaisoaminopterin was equiactive with methotrexate at approximately one half of the total dose employed.

Recently, we reported that appropriately substituted 2-fluorobenzonitriles undergo cyclization with guanidine carbonate to afford 2,4-diaminoquinazolines usually in good to excellent yield. This paper describes the preparation of a variety of new 2,4-diaminoquinazolines substituted at positions five or seven. In addition, the reactions of selected 2-fluorobenzonitriles with formamidine acetate or acetamidine acetate were examined. The results obtained demonstrate that the analogous 4-amino- and 2-methyl-4-aminoquinazolines can be prepared by this approach but that the yields are considerably lower than when guanidine carbonate is employed as the cyclization reagent.

The folate analogue, 10-thia-5-deazafolic acid, was obtained via a multistep synthetic sequence beginning with the known intermediate, 2,4-diaminopyrido[2,3-d]pyrirnidine-6-carboxaldehyde. Reduction of this aldehyde with sodium borohydride gave 2,4-diamino-6-(hydroxymethyl)pyrido[2,3-d]pyrimidine, which when heated in base gave 2-amino-3,4-dihydro-6-(hydroxymethyl)-4-oxopyrido[2,3-d]pyrimidine. Treatment of the latter compound with phosphorus tribromide in tetrahydrofuran afforded 2-amino-6-(bromomethyl)-3,4-dihydro-4-oxopyrido[2,3-d]pyrimidine, thus constituting the first successful synthesis of this elusive intermediate. The aforementioned bromomethyl compound reacted smoothly with the sodium salt of ethyl 4-mercaptobenzoate, and the resulting ester was saponified to give 10-thia-5-deazapteroic acid. Conventional peptide bond coupling to di-tert-butyl L-glutamate followed by treatment with trifluoroacetic acid afforded the target compound in respectable yield. Attempts to prepare its 5,6,7,8-tetrahydro derivative by catalytic hydrogenation were unsuccessful.

A series of thirty eight 2,4-diaminoquinazolines having diverse substitution patterns on the aromatic ring was evaluated for inhibitory activity against dihydrofolate reductase (DHFR) obtained from a human lymphoblast cell line. Many of these compounds were also evaluated as inhibitors of rat liver DHFR under the same experimental conditions. In most instances the results obtained with each enzyme were comparable indicating that the rodent enzyme is a suitable model for the human DHFR as far as the determination of I50 values is concerned. The results demonstrate that relatively simple 5-substituted- or 5,6-disubstituted-2,4-diaminoquinazolines can be potent DHFR inhibitors. The presence of a nonpolar substituent at position 7 or 8 was highly detrimental to inhibitory potency.

Nitration of 2-amino-4-oxo-(3H)-5-trifluoromethylquinazoline is shown to occur exclusively at C6 as determined from an analysis of long range 1H and 19F scalar couplings to ring carbons. Nitration of 2-amino-4-oxo-(3H)-5-fluoroquinazoline is found to occur both at C6 and C8 as evident from an analysis of the 19F and 1H couplings of the ring protons.

The reactions of ten ortho-fluoroketones with guanidine carbonate in N,N-dimethylacetamide were investigated as a new synthetic approach to 2-amino-4-alkyl- and 2-amino-4-arylquinazolines. The yields obtained ranged from low to moderate and were highly dependent upon the nature of the substituents on the reactant. Eight new quinazolines were elaborated in this study.

The reaction of guanidine carbonate with various ortho-fluorobenzaldehydes in N,N-diinethylacetamide was investigated as a potential route for preparing 2-aminoquinazolines. Eleven new 2-aminoquinazolines were elaborated in this manner in low to moderate yields. In general the best results were obtained with ortho-fluorobenzaldehydes possessing an electron withdrawing substituent at the other ortho position. Complex mixtures were obtained using 2-fluorobenzaldehyde, 2,5-difluorobenzaldehyde and 2-fluoro-5-methoxybenzaldehyde which were not resolved.

Five new N alpha-(5,8-dideazapteroyl)-L-ornithines have been prepared using multistep synthetic sequences. These include N alpha-[5-(trifluoromethyl)-5,8-dideazapteroyl]-L-ornithine, 3, as well as N alpha-[5-(trifluoromethyl)-5,8-dideazaisopteroyl]-L-ornithine, 4, and its 5-fluoro and 5-chloro analogues. Both of the compounds containing a 5-(trifluoromethyl) group (3 and 4) were found to be excellent inhibitors of homogeneous hog liver folylpolyglutamate synthetase, having Ki values in the same range as N alpha-(5-chloro-5,8-dideazapteroyl)-L-ornithine, 2, (approximately 10 nM). However, the bridge-reversed isomer of 2 was 60-fold less inhibitory than 2.

The new folate analogue, 2-desamino-2-methyl-5,8-dideazaisofolic acid, 2c , was synthesized and evaluated using a variety of biochemical and antitumor assays. For purposes of comparison, its 2-desamino, 2b , and 2-amino, 2a , counterparts, as well as N 10 -propargyl-5,8-dideazafolic acid, 1a , and the corresponding 2-desamino, 1b , and 2-desamino-2-methyl, 1c , modifications were included in these studies. Compound 2c was found to be a potent inhibitor of the growth of L1210 and MCF-7 cells in culture, being only 2-fold and 5-fold less effective than 1c , respectively. However, although analogue 2c was 189-fold less inhibitory toward L1210 thymidylate synthase (TS) than 1c , its cytotoxicity was reversed completely by thymidine alone which suggests that the compound behaves as a TS inhibitor in cells. Enzymatically synthesized polyglutamates of 2c were substantially more inhibitory toward human TS than the parent compound. Compound 2c was the most efficient substrate for mammalian folylpolyglutamate synthetase of the compounds studied having a V max / K m nearly 12-fold larger than 1c . Both 1c and 2c were effective inhibitors of the uptake of [ 3 methotrexate into MOLT-4 cells, implying that each is efficiently transported into tumor cells. These results suggest that a weak inhibitor of TS in vitro can be a potent cytotoxic agent if it can readily gain entry into target cells and be converted to polyglutamated metabolites.

Prompted by recent disclosures concerning the potent antitumor activities of 5-deaza-5,6,7,8-tetrahydrofolic acid and 5,10-dideaza-5,6,7,8-tetrahydrofolic acid (DDATHF), we have prepared 5-deazaisofolic acid (3a) and 5-deaza-5,6,7,8-tetrahydroisofolic acid (4a). Reductive condensation of 2,6-diamino-3,4-dihydro-4-oxopyridol [2,3-d] pyrimidine with di-tert-butyl N-(4-formylbenzoyl)-L-glutamate and subsequent deprotection with trifluoroacetic acid yielded 5-deazaisofolic acid in good yield. Catalytic hydrogenation of this analogue then gave 4a. The 9-CH 3 and 9-CHO modifications of 3a and the 9-CH 3 derivative of 4a were also synthesized. Each of the new analogues was evaluated with a variety of folate-requiring enzymes as well as MCF-7 cells in culture. Compound 4a had an IC 50 of ca. 1 μM against MCF-7 cells and was nearly 100-fold less potent than DDATHF in this regard. The three oxidized isofolate analogues were all poor inhibitors of tumor cell growth

GPI-linked membrane folate receptors (MFRs) have been implicated in the receptor-mediated uptake of reduced folate cofactors and folate-based chemotherapeutic drugs. We have studied the biosynthetic transport to and internalization of MFR isoform alpha in KB-cells. MFR-alpha was synthesized as a 32-kD protein and converted in a maturely glycosylated 36-38-kD protein 1 h after synthesis. 32-kD MFR-alpha was completely soluble in Triton X-100 at 0 degree C. In contrast, only 33% of the 36-38-kD species could be solubilized at these conditions whereas complete solubilization was obtained in Triton X-100 at 37 degrees C or in the presence of saponin at 0 degree C. Similar solubilization characteristics were found when MFR-alpha at the plasma membrane was labeled with a crosslinkable 125I-labeled photoaffinity-analog of folic acid as a ligand. Triton X-100-insoluble membrane domains containing MFR-alpha could be separated from soluble MFR-alpha on sucrose flotation gradients. Only Triton X-100 soluble MFR-alpha was internalized from the plasma membrane. The reduced-folate-carrier, an integral membrane protein capable of translocating (anti-)folates across membranes, was completely excluded from the Triton X-100-resistant membrane domains. Internalized MFR-alpha recycled slowly to the cell surface during which it remained soluble in Triton X-100 at 0 degree C. Using immunoelectron microscopy, we found MFR-alpha along the entire endocytic pathway: in clathrin-coated buds and vesicles, and in small and large endosomal vacuoles. In conclusion, our data indicate that a large fraction, if not all, of internalizing MFR-alpha bypasses caveolae.

Six new analogues of 5,8-dideazaisofolic acid and 5,8-dideazaisoaminopterin were synthesized in an effort to obtain enhanced antitumor activity. The modifications included the replacement of the 2-amino group by hydrogen or methyl as well as the inclusion of a methyl substituent at position 9. Based upon activity against L1210 leukemia cells in culture, three of the new analogues together with one compound described previously were evaluated for cytotoxicity in vitro using three human tumor cell lines (Colo 320 DM, Hep G2 and HL-60). The most effective compound was 2-desamino-N9-methyl-5,8-dideazaisoaminopterin (2c) with the HL-60 cells being the most sensitive to its cytotoxic effects. These analogues were evaluated in vitro as inhibitors of dihydrofolate reductase (DHFR) and thymidylate synthase (TS) from human as well as bacterial (Lactobacillus casei) sources. All four of the 4-amino analogues were most effective toward L. casei DHFR compared with human DHFR, with 2-desamino-2-methyl-5,8-dideazaisoaminopterin (2d) and its 9-methyl derivative (2e) having 818- and 430-fold greater selectivity (L. casei/human). Most of the compounds studied were found to be only modest inhibitors of human TS (I50 values = 1.5 to 20 μM) and were therefore at least 40-fold less inhibitory than 10-propargyl-5,8-dideazafolic acid. Nevertheless, reversal of cytotoxicity studies with thymidine, hypoxanthine and folinic acid using the HL-60 cell line suggested that TS is the primary target for these analogues.

The compound 5,8-dideazaisofolic acid (IAHQ), 1a, was found to posses modest antitumor activity against a variety of human and murine tumor cells both in vitro and in vivo. 1–5 Its lack of potency was attributed to slow influx into target cells as demonstrated with [3H] IAHQ in the presence of HCT-8 human colon adenocarcinoma cells in culture.6 The analogues 2-desamino-5,8-dideazaisofolic acid, 1b, and 2-desamino-2-methyl-5,8-dideazaisofolic acid, 1c, were prepared subsequently and found to be 6-and 44-fold more cytotoxic toward L1210 cells than 1a 7,8 This activity enhancement correlated well with increased affinity for the reduced folate transporter as measured by competition for the uptake of [3H] MTX into MOLT-4 cells.8

The folate analogue 5,8-dideazaisofolic acid (IAHQ), 1a, has demonstrated antitumor activity against a variety of mammalian tumor cells both in vitro and in vivo.1–5 Its derivative, 2-desamino-2-methyl-5,8-dideazaisofolic acid, 1b, was found to be 20-and 44-fold more cytotoxic toward MCF-7 and L1210 cells in culture, respectively.6 In order to evaluate the effects of other substituents located at position five of the quinazoline nucleus upon antitumor activity, we have prepared 5-fluoro-5,8-dideazaisofolic acid, 2a, and desamino-2-methyl-5-fluoro-5,8-dideazaisofolic acid, 2c, together with their 9-methyl derivatives, 2b and 2d. This report describes the chemistry and preliminary biological evaluation of these new compounds.

A novel folic acid analogue, N alpha-(5-deaza-5,6,7,8-tetrahydropteroyl)-L-ornithine, 3, was prepared via a multistep synthetic sequence. The key steps involved the conversion of 5-deazapteroic acid to its N10-formyl derivative followed by catalytic hydrogenation of the pyridine ring and subsequent heating in dilute sodium hydroxide to afford the new 5-deaza-5,6,7,8-tetrahydropteroic acid. After trifluoroacetylation, this compound was coupled to N delta-(tert-butyl-oxycarbonyl)-L-ornithine using conventional peptide bond forming conditions. Deprotection first in base and then in acid gave the title compound. Compound 3 was an effective inhibitor of hog liver folylpolyglutamate synthetase (Kis, estimated = 64 nM), and was shown to retard the formation of polyglutamates of a structurally related folic acid analogue in HCT-8 cells in vitro.

The in vitro metabolism of a variety of 5,8-dideazafolate and 5,8-dideazaisofolate analogues by pig liver folylpolyglutamate synthetase and the specificity of the enzyme for some polyglutamate derivatives of these analogues have been investigated. All 4-oxo-quinazoline analogues were metabolized to long chain polyglutamate derivatives, primarily the pentaglutamate, whereas 4-amino-quinazolines were metabolized to a lesser extent, with the accumulation of di- and triglutamate derivatives. This pattern of metabolism was consistent with the large drop in V max K m and V max values for folylpolyglutamate synthetase observed with diglutamate derivatives of 4-aminofolate analogues. The extent of metabolism of the various analogues did not correlate with the relative substrate effectiveness of their parent monoglutamate derivatives. The 5-chloro and 5-methyl substitutions of quinazolines enhanced the addition of glutamate residues to 4-amino derivatives but markedly impaired the metabolism of 4-oxo derivatives.

An audit of 28 patients treated with acupuncture for musculo-skeletal pain, of whom visual analogue scores were analysed for 20, showed a marked decrease in pain following treatment. Needles used in treatment cost less than 50p per session, but consultations were doubled in length to 20min.

The quinazoline couterpart of folic acid (5,8-deazafolic acid) as well as its 10-methyl analogue has been shown to be an effective inhibitor of thymidylate synthetase from several different sources. This paper describes the synthesis of modifications in which the nitrogen atom at position 10 is replaced by sulfur, oxygen, or methylene affording 10-thia-5,8-deazafolic acid, 10-oxa-5,8-deazafolic acid, and 5,8,10-deazafolic acid, respectively. In preliminary testing, each of the target compounds displayed marginal activity against L1210 leukemia in mice.

A series of six 2,4-diaminoquinazoline analogues of folic acid which bear close structural resemblance to methotrexate, 1a, were synthesized by unequivocal routes. Three of these have not been described previously, while complete structural characterization of the remaining compounds is presented for the first time. Each of the compounds was a potent inhibitor of dihydrofolate reductase (DHFR) from rat liver or L1210 leukemia cells having I50 values in a range similar to that of 1a. However, a wide divergence in inhibitory activity toward the growth of human gastrointestinal adenocarcinoma or L1210 leukemia cells in vitro was observed. Compounds having a normal folate configuration at positions 9 and 10 were more inhibitory than their isomeric reversed-bridge counterparts. The N-formyl modifications were the least active of the compounds studied. Unsubstituted or N-methyl modifications competed effectively with tritiated 1a for uptake into L1210 leukemia cells, while N-formyl modifications did not. Against an L1210 cell line resistant to 1a by virtue of altered transport and overproduction of DHFR, partial but not complete cross-resistance was observed for certain analogues. Of the three compounds selected for in vivo evaluation against L1210 leukemia in mice, two had a similar level of antitumor activity to that of 1a. The compound 5,8-dideazamethopterin, 2b, however, was slightly more active than 1a but at substantially reduced dose levels.

Infrared and Raman spectra of 2-fluoro-4,6-dichloro-s-triazine and 2,4-difluoro-6-chloro-s-triazine have been recorded and an assignment of the normal vibrations in the series C3Cl3−nFnN3 is proposed. Infrared spectra of the series C3Br3−nFnN3 are evaluated in somewhat less detail.

An investigation was made of certain pharmacological and chemical characteristics of the antimycobacterial agents glycyl hydroxamic acid (GHA) and β-alanyl hydroxamic acid (BAHA). Inhibition by GHA of the growth of Mycobacterium smegmatis was antagonized by D-, L-, and DL-alanine, pyridoxal phosphate (PyP), and ascorbic acid. Inhibition by BAHA was reversed by D-alanine. Oxidation of L-alanine by the organism was inhibited by GHA, and this inhibition was partially antagonized by PyP and by L-alanine. BAHA had virtually no effect on oxidation of L-alanine. GHA inhibited the dihydroxyphenylalanine (DOPA) decarboxylase from guinea pig liver, and this inhibition was antagonized by PyP. BAHA was without effect on DOPA decarboxylase. Ultraviolet absorption spectroscopy suggested an alteration of PyP by GHA and BAHA. The test for hydroxylamine was found unsuitable as a means for determining stability of each compound due to falsely high values. Synthesis of each compound is described in an appendix.

The Raman spectrum of liquid tris(trifluoromethyl)- s -triazine has been obtained at room temperature. The infrared spectrum of the gaseous compound has been recorded at room temperature from 650–4000 cm −1 , that of the liquid from 250–4000 cm −1 . The analysis of these data, particularly by correlation with the spectrum of trifluoro- s -triazine, permits a satisfactory assignment of the fundamentals. Based on this information, the infrared spectra in the 650–4000 cm −1 region of liquid tris(perfluoroethyl)- s -triazine, tris(perfluoropropyl)- s -triazine, and tris(chlorodifluoromethyl)- s -triazine are discussed and characteristic frequencies are correlated.

Dihydrofolate reductase (DHFR) inhibitors, which differ from the classical folate antagonists in physicochemical and pharmacologic parameters such as lipid solubility and mechanisms of cellular transport, were screened for DHFR inhibitory activity and biologic activity in newborn rat skin. The most effective drugs from this screen were tested for their effects on de novo DNA synthesis in psoriatic epidermis in vitro. Of the 24 compounds studied, methotrexate (MTX) was the most potent inhibitor of rat skin DHFR (I 50 = 8.6 × 10 -8 M). Methotrexate-dimethylester, methasquin-diethylester, DDEP (2,4-diamino-5-(3',4'-dichlorophenyl)-6-ethylpyrimidine), and Baker's triazine antifolate (NSC 139105), while less effective than MTX as DHFR inhibitors, were more effective than MTX as inhibitors of de novo DNA synthesis in rat skin in vitro. Baker's antifolate was the only compound tested which was considerably more effective than MTX as an inhibitor of de novo DNA synthesis in psoriatic epidermis in vitro.

A series of 2,4-diaminoquinazolines bearing an aryl function attached to the 6 position through an acetamido or related linkage was synthesized. Each compound was evaluated as an inhibitor of rat liver dihydrofolate reductase as well as for suppressive antimalarial effects against Plasmodium berghei in mice. Significant in vivo activity was found to reside primarily with 5-chloro-6-arylacetamido derivatives. Most of these compounds were also tested for prophylactic activity against sporozoite-induced Plasmodium gallinaceum in chicks. Thirteen compounds, each of which possesses a 5-Cl or 5-CH3 group, displayed curative activity in this test system. Since several of these showed markedly greater potency against the avian infection, selective inhibitory action upon preerythrocytic forms of the malaria parasite is thus implied.

SummaryA series of nine 2,4-diamino-quinazolines bearing a variety of substitu-ents at positions 5 and 6 was evaluated against three human isolates of Candida albicans in vitro. An established drug, 5-fluo-rocytosine, was chosen as a standard of comparison and each isolate was found to be sensitive to this agent. The two most active compounds, 6-(3,4-dichlorophenylacetam-ido)-2,4-diaminoquinazoline and 6-(3,4-dichlorobenzamido)-2,4-diaminoquinazo-line, were selected for acute toxicity studies in mice. At the highest dose tested (250 mg/ kg) the latter compound showed evidence of toxicity while both compounds were substantially free of untoward effects at lower doses.

A series of fourteen 5,8-dideaza analogues of folic and pteroic acids was evaluated for inhibition of 5-aminoimidazole-4-carboxamide ribonucleotide transformylase (AICAR TFase) from chicken liver. Of the 5,8-dideaza folate derivatives studies, 10-oxa-5,8-dideazafolic acid was the most potent inhibitor. The addition of one L-glutamate moiety to the gamma-carboxyl group caused a 6- to 7-fold reduction in Ki in three instances. Two compounds devoid of an L-glutamate were 4- to 6-fold less inhibitory than their parent counterparts possessing one L-glutamate residue.

The infrared spectra in the sodium chloride region of the series trifluoro- s-triazine tris (trifluoromethyl)- s-triazine and trifluoro- s-triazinetris(perfluoroethyl)- s-triazine have been recorded and normal vibrations were assigned. The transitions of characteristic frequencies are discussed.
