Maior planície de inundação do mundo, o Pantanal desempenha um papel importante na manutenção da diversidade biológica. A Estação Ecológica de Taiamã (EET) é uma unidade de conservação localizada no norte do Pantanal e é considerada uma área relevante para a conservação de aves. A composição de espécies dessa área deve ser estudada e conhecida para a elaboração de planos de manejo específicos. Além disso, as aves aquáticas são importantes na avaliação da qualidade ambiental e com alta sensibilidade a possíveis alterações. Assim, investigamos a composição das aves aquáticas da EET e entorno e avaliamos os níveis de riqueza e abundância entre os locais e o nível do rio durante um ano de amostragem. Foi realizado um esforço amostral total de seis campanhas, com quatro locais amostrados: dois na EET e dois nos arredores. Identificamos um total de 45 espécies de aves aquáticas, divididas em nove ordens, 20 famílias e 36 gêneros. O grande número de indivíduos observado em fevereiro de 2012 pode estar relacionado a um fenômeno natural que ocorre no Pantanal, chamado dequada, o qual está associado aos processos de decomposição da biomassa submersa das plantas no início da cheia anual do bioma. Ao analisar as espécies mais comuns neste estudo, foi verificado que as abundâncias de Nannopterum brasilianus, Ardea alba e Butorides striata aumentaram durante esse período. A maior abundância e o maior número de espécies foram observados no local de amostragem dentro da EET, de forma que a existência dessa área protegida para conservação é um fator significativo para a manutenção das comunidades de aves da região. Desta forma, fica evidenciada a importância de áreas protegidas para a conservação de aves no Pantanal. Atualmente, uma parcela muito pequena (5,37%) do bioma está protegida; portanto, aumentar essa proporção deve ajudar na manutenção dos habitat existentes. Pesquisas subsequentes devem ser conduzidas para corroborar ou refutar as tendências sazonais observadas neste estudo.

Energetic subsidies between terrestrial and aquatic ecosystems can strongly influence food webs and population dynamics. Our objective was to study how aquatic subsidies affected jaguar (Panthera onca) diet, sociality, and population density in a seasonally flooded protected area in the Brazilian Pantanal. The diet (n = 138 scats) was dominated by fish (46%) and aquatic reptiles (55%), representing the first jaguar population known to feed extensively on fish and to minimally consume mammals (11%). These aquatic subsidies supported the highest jaguar population density estimate to date (12.4 per 100 km2) derived from camera traps (8,065 trap nights) and GPS collars (n = 13). Contrary to their mostly solitary behavior elsewhere, we documented social interactions previously unobserved between same-sex adults including cooperative fishing, co-traveling, and play. Our research demonstrates that aquatic subsidies seen in omnivores can be highly influential to obligate carnivores leading to high population density and altered social structure.

O entendimento da dieta alimentar de peixes é uma importante ferramenta na obtenção de informações sobre a estrutura das comunidades aquáticas e interações biológicas, principalmente no que se refere à qualidade ambiental. Entretanto, no Pantanal, o pulso de inundação acarreta uma transformação ambiental que promove uma dinâmica na dieta de várias espécies de peixes, e aquelas espécies-base de cadeia alimentar podem sofrer uma pressão ainda maior devido a amplitude hídrica e disponibilidade dos recursos alimentares. Assim, neste trabalho, foi realizada a descrição dos recursos alimentares explorados por Tetragonopterus argenteus no Rio Paraguai, Pantanal Norte, Cáceres (MT), nos períodos de enchente (2012), cheia, vazante e estiagem (2013). Os espécimes foram coletados e levados para o laboratório de Ictiologia da UNEMAT, onde foi realizado a pesagem, evisceração, e caracterização dieta baseada na frequência de ocorrência (% Fi), proporção volumétrica (% Vi), combinados ao Índice Alimentar (IAi %), bem como através do diagrama de Costello e curva de rarefação. A alimentação de T. argenteus variou durante os períodos do ciclo hidrológico com itens como macroinvertebrados, material vegetal e resto de peixes. T. argenteus pode ser classificado como herbívoro oportunista e que ocorre uma influência do pulso de inundação na dieta, alterando a disponibilidade dos itens alimentares no decorrer do ciclo hidrológico, bem como a possível interferência das cevas no hábito alimentar de espécies usadas como iscas pela população local.

Habitat loss and fragmentation represent major threats for the conservation of apex predators, such as the jaguar (Panthera onca). Investigating species’ resource selection behavior in response to landscape alteration is critical for developing relevant conservation management plans. The jaguar is found across a variety of habitats with different gradients of human disturbance, making them a good candidate to study how apex predators respond to increasing intensity of human land use. We developed resource selection models to characterize patterns of jaguar resource selection at two different spatial scales, home range (coarse) and foraging scale (fine). This analysis was based on the largest existing GPS-location dataset for jaguars (n = 40 individuals, n = 87,376 locations), spanning the species’ geographic range in Brazil and Argentina. We found that both males and females jaguars exhibited an overall preference for forests and areas close to watercourses at both the home range and foraging scale. At the foraging scale, areas of high livestock density “attracted” male jaguars. We also performed a follow-up analysis to test for context-dependent resource selection (i.e., functional responses) by relating individual behavior to local habitat characteristics. We found that jaguars in heavily-forested landscapes showed strong avoidance of non-forest. Furthermore, we found that only the individuals in closest proximity to watercourses showed positive selection for water. Our results highlight that jaguars display different patterns of resource selection in different areas, demonstrating a considerable ability to use or tolerate a wide variety of different conditions across the species geographic range. This plasticity may allow jaguars to adjust their behavior according to land use changes but also increases human-jaguar conflict and jaguar mortality, especially in areas with high livestock density. Jaguars (Panthera onca), like other apex predators, are highly susceptible to habitat loss and fragmentation given their low demographic potential and large habitat area requirements. Across their range, the Pantanal is considered critical for the jaguar's long-term conservation. Here we provide the first multi-scale path selection function model for jaguars, and the first empirically-based movement model covering the entire Pantanal ecosystem. Out of eight investigated variables, six were related to jaguar habitat use in the Pantanal: terrain roughness, human population density, grassland, percentage of tree cover, flooded habitats and shrubland. The results of scale optimization revealed that jaguars responded primarily to landscape variables at broad scales (32 km) of habitat availability, with only one variable (grassland) influencing jaguar path selection at a finer scale (4 km). Jaguar habitat use was positively associated with flooded habitats and densely forested areas and negatively associated with grassland, terrain roughness, and human population density, with the latter having the strongest negative effect on jaguar movement. The prediction map suggested that only 9.3% of the total suitable jaguar habitat in Pantanal is protected by Conservation Units. Among the most suitable areas, the largest continuous habitats were located in the northwestern portions of the Pantanal, which corresponds to the interfluvial areas between Corixo Grande and Cuiaba rivers. Our results suggested that the implementation of already proposed North Pantanal Conservation Unit Mosaic in this area would be highly valuable for jaguar conservation. This study provides a foundation for future research to delineate and prioritize core areas and corridors for jaguars in the region.

The largest feline in the Americas and the third largest in the world, the jaguar is an apex predator in the food chain and a key species in the ecosystems where it occurs, developing important ecological functions in maintaining ecosystem balance. In Brazil, the Pantanal is considered an important refuge for the species, and protected areas such as the Taiamã Ecological Station (TES) are relevant for conservation of pristine ecosystem where the species persist. Thus, considering that this area is located in one of the regions with the highest concentration of this large cat in the Pantanal, the objective of this study was to evaluate the effectiveness of the protection given by the TES for the jaguar population. The station is a flooded island in the middle of the Paraguay river and surrounded by extensive wetlands. We monitored ten jaguars using GPS collar at the TES and its surroundings. The samples were separated into high-water season (January to June) and low-water season (July to December), and the estimated home ranges were grouped as: 1) residents only and 2) all monitored individuals. The stabilization of the home ranges of eight jaguars, considered residents, was determined through variograms. When all jaguars were analysed together there was 55% overlap between the clustered areas of the two analysed seasons. In the analysis excluding non-resident individuals there was 72% overlap between the clustered areas. The type of land cover inside these areas was very similar between these periods. The range of this protected area is not sufficient to effectively protect these jaguar populations, since the grouped home ranges of the resident animals studied are 3.5(wet)/2.5(dry) times larger than the area of the TES. However, the surroundings of the TES have low human impact and no deforestation and, in this way, the landscape context of the station may have a significant effect on the persistence of a large carnivore like the jaguar. However, it is necessary to create more protected areas in order to prevent possible future environmental changes in the areas surrounding TES.

A Área Úmida Pantanal mato-grossense apresenta uma complexidade geomorfológica, hidrológica e biológica, que resulta na sua heterogeneidade ambiental. O conhecimento dos macrohabitats encontrados em áreas protegidas do Pantanal pode dar suporte considerável na tomada de decisão para a sua conservação. O objetivo desta pesquisa foi identificar e caracterizar unidades funcionais, subclasses e macrohabitats da Estação Ecológica de Taiamã (EET), para contribuir com a gestão e as estratégias de manejo desta Unidade de Conservação (UC). A identificação dos macrohabitats no campo foi realizada entre os anos 2015 e 2016. Com o uso de uma cena Landsat 8, os dados foram processados por meio de classificação supervisionada. As unidades funcionais encontradas foram Áreas Permanentemente Aquáticas, Áreas de Transição Aquática e Terrestre com predominância Terrestre e Áreas Pantanosas. Seis subclasses e seis macrohabitats foram identificados. Os macrohabitats mais representativos foram Campo Flutuante (Batumes), correspondendo a 48%, seguido de Campo Inundado, com 23%, e de Floresta Monoespecífica de Erythrina fusca Lour. (Abobral), com 16%. Os macrohabitats registrados na EET reproduzem o mosaico paisagístico do Pantanal, cujas funções ecológicas precisam ser protegidas por uma legislação específica, que supra as lacunas referentes à preservação das áreas úmidas brasileiras.

No. of bitstreams: 0 Previous issue date: 2021-08-09 Consejo Nacional de Investigaciones Científicas y Técnicas WWF International Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Large terrestrial carnivores have undergone some of the largest population declines and range reductions of any species, which is of concern as they can have large effects on ecosystem dynamics and function.1–4 The jaguar (Panthera onca) is the apex predator throughout the majority of the Neotropics; however, its distribution has been reduced by >50% and it survives in increasingly isolated populations.5 Consequently, the range-wide management of the jaguar depends upon maintaining core populations connected through multi-national, transboundary cooperation, which requires understanding the movement ecology and space use of jaguars throughout their range.6–8 Using GPS telemetry data for 111 jaguars from 13 ecoregions within the four biomes that constitute the majority of jaguar habitat, we examined the landscape-level environmental and anthropogenic factors related to jaguar home range size and movement parameters. Home range size decreased with increasing net productivity and forest cover and increased with increasing road density. Speed decreased with increasing forest cover with no sexual differences, while males had more directional movements, but tortuosity in movements was not related to any landscape factors. We demonstrated a synergistic relationship between landscape-scale environmental and anthropogenic factors and jaguars’ spatial needs, which has applications to the conservation strategy for the species throughout the Neotropics. Using large-scale collaboration, we overcame limitations from small sample sizes typical in large carnivore research to provide a mechanism to evaluate habitat quality for jaguars and an inferential modeling framework adaptable to the conservation of other large terrestrial carnivores. Habitat loss and fragmentation are important threats to carnivores worldwide and are especially intense for large predators. Jaguars have been extirpated from over half of their original distribution, and few regions still maintain large populations. The Pantanal is among the best examples of such regions and can be used to better understand several aspects of jaguar biology that are relevant for conservation planning throughout the species' range. Thus, in this study we used microsatellite markers and field data to: (i) assess the genetic structure and gene flow of jaguars (n = 110) from the northern and southern Pantanal; (ii) verify if females are more philopatric than males; (iii) produce a timeline pedigree to allow the identification of distances from offspring to their mothers; and (iv) estimate the generation time for jaguars. Our results are consistent with the hypothesis that Pantanal jaguars represent a panmictic population, although exhibiting some degree of local differentiation. The Paraguay River seems to be an important factor promoting gene flow among the studied populations, highlighting its relevance for regional conservation efforts. Our data provide the first genetic evidence of female philopatry and male-biased dispersal in jaguars. In addition, we report the first timeline pedigree for a wild jaguar population and the first direct estimate of the species' generation time. Our results contribute to the construction of more realistic assessments of jaguar population dynamics and long-term genetic viability, thus contributing to the design of improved conservation strategies on behalf of this species.

Building bridges between environmental and political agendas is essential nowadays in face of the increasing human pressure on natural environments, including wetlands. Wetlands provide critical ecosystem services for humanity and can generate a considerable direct or indirect income to the local communities. To meet many of the sustainable development goals, we need to move our trajectory from the current environmental destructive development to a wiser wetland use. The current article contain a proposed agenda for the Pantanal aiming the improvement of public policy for conservation in the Pantanal, one of the largest, most diverse, and continuous inland wetland in the world. We suggest and discuss a list of 11 essential interfaces between science, policy, and development in region linked to the proposed agenda. We believe that a functional science network can booster the collaborative capability to generate creative ideas and solutions to address the big challenges faced by the Pantanal wetland. O pantanal é uma das maiores áreas úmidas do mundo, caracterizada por apresentar alterações durante os ciclos hidrológicos, que correspondem a períodos que se alternam durante enchentes e vazantes. Este estudo teve como objetivo analisar a dieta de Brycon hilarii (Valenciennes, 1850), em diferentes períodos hidrológicos da Estação Ecológica de Taiamã em Mato Grosso Brasil, uma unidade de conservação de interesse mundial, e sua relação com a formação vegetacional. Foram capturados 353 espécimes em campanhas mensais, durante os quatro períodos hidrológicos, nos meses de agosto de 2013 a julho de 2015, em seguida, acondicionados em caixas térmicas e encaminhado para o laboratório, onde foram anotados os dados biométricos e a remoção dos estômagos para as análises de importância alimentar (IAi). Os resultados mostram que Erythrina fusca Lour. (abobreiro) é o principal item alimentar da dieta de B. hilarii. Durante os períodos de enchente, apresentou IAi de 95% e na cheia 60%. Além disso, B. hilarii mostrou ser generalista em se alimentar de E. fusca durante os períodos de cheia, vazante e estiagem, sendo especialista no período de enchente. Com relação do volume de sementes intactas e trituradas comparado ao tamanho padrão, verificou-se que as sementes são ingeridas conforme o tamanho do espécime. Este fato contribui com a importância dos períodos hidrológicos, junto a monodominancia de E. fusca no local, o que propicia o aumento de recursos alimentares para B. hilarii, através de estratégias, coincidindo com a maior disponibilidade de E. fusca neste ambiente.

No. of bitstreams: 0 Previous issue date: 2018-07-01 The field of movement ecology has rapidly grown during the last decade, with important advancements in tracking devices and analytical tools that have provided unprecedented insights into where, when, and why species move across a landscape. Although there has been an increasing emphasis on making animal movement data publicly available, there has also been a conspicuous dearth in the availability of such data on large carnivores. Globally, large predators are of conservation concern. However, due to their secretive behavior and low densities, obtaining movement data on apex predators is expensive and logistically challenging. Consequently, the relatively small sample sizes typical of large carnivore movement studies may limit insights into the ecology and behavior of these elusive predators. The aim of this initiative is to make available to the conservation-scientific community a dataset of 134,690 locations of jaguars (Panthera onca) collected from 117 individuals (54 males and 63 females) tracked by GPS technology. Individual jaguars were monitored in five different range countries representing a large portion of the species' distribution. This dataset may be used to answer a variety of ecological questions including but not limited to: improved models of connectivity from local to continental scales; the use of natural or human-modified landscapes by jaguars; movement behavior of jaguars in regions not represented in this dataset; intraspecific interactions; and predator-prey interactions. In making our dataset publicly available, we hope to motivate other research groups to do the same in the near future. Specifically, we aim to help inform a better understanding of jaguar movement ecology with applications towards effective decision making and maximizing long-term conservation efforts for this ecologically important species. The catfish family Heptapteridae (order Siluriformes) is endemic to the Neotropics and is one of the most common of the fish families in small bodies of water. Although over 200 species have been identified in this family, very few have been characterized cytogenetically. Here, we analyze the chromosome genomes of four species of Heptapteridae: Cetopsorhamdia iheringi (Schubart & Gomes, 1959), 2n = 58, comprising 28 metacentric (m) + 26 submetacentric (sm) + 4 subtelomeric (st) chromosomes; Pimelodella vittata (Lütken, 1874), 2n = 46, comprising 16m + 22sm + 8st; Rhamdia prope quelen (Quoy & Gaimard, 1824), 2n = 58 comprising 26m + 16sm + 14st + 2 acrocentric; and Rhamdiopsis prope microcephala (Lütken, 1874), 2n = 56, comprising 12m + 30sm + 14st. The nucleolus organizer regions (NORs) were located in a single chromosome pair in all species. The two species that belonged to the subclade Nemuroglanis, Cetopsorhamdia iheringi and Rhamdia prope quelen, had a diploid chromosome number of 58 and an interstitial NOR adjacent to a C+ block located on one of the larger chromosome pairs in the complement. Our results from conventional cytogenetic techniques in combination with FISH using 18S and 5S rDNA probes corroborated the taxonomical hypothesis for the formation of the Nemuroglanis subclade.

This study aimed to assess the exposure of free-living jaguars (Panthera onca) to Leptospira spp. and Brucella abortus in two conservation units in the Pantanal of Mato Grosso, Brazil. The presence of antibodies in blood samples of eleven jaguars was investigated using autochthonous antigens isolated in Brazil added to reference antigen collection applied to diagnosis of leptospirosis by Microscopic Agglutination Test (MAT). The Rose Bengal test was applied for B. abortus antibodies. Two (18.2%) jaguars were seroreactive for the Leptospira spp. antigen and the serovar considered as most infective in both animals was a Brazilian isolate of serovar Canicola (L01). All jaguars were seronegative for B. abortus. These data indicate that the inclusion of autochthonous antigens in serological studies can significantly increase the number of reactive animals, as well as modify the epidemiological profile of Leptospira spp. infection. Este estudo teve como objetivo avaliar a exposição de onças-pintadas de vida livre (Panthera onca) para Leptospira spp. e Brucella abortus em duas unidades de conservação no Pantanal de Mato Grosso, Brasil. A presença de anticorpos em amostras de sangue de onze onças foi investigada utilizando antígenos autóctones isoladas no Brasil adicionais a coleção de antígenos de referência aplicada usualmente ao diagnóstico da leptospirose pelo teste de soroaglutinação microscópica (MAT). Para os anticorpos de B. abortus, foi utilizado o teste de Rosa Bengala. Duas onças-pintadas (18,2%) foram reagentes para Leptospira spp. e o sorotipo considerado como o mais provável pela infecção em ambos os animais foi um isolado brasileiro do sorovar Canicola (L01). Todas as onças-pintadas foram soronegativas para B. abortus. Estes dados indicam que a inclusão de antígenos autóctones em estudos sorológicos pode aumentar significativamente o número de animais reativos, assim como modificar a caracterização do sorotipo mais prevalente.

Big cat genomes reveal a history of interspecies admixture and adaptive evolution of genes underlying development and sensory perception., The great cats of the genus Panthera comprise a recent radiation whose evolutionary history is poorly understood. Their rapid diversification poses challenges to resolving their phylogeny while offering opportunities to investigate the historical dynamics of adaptive divergence. We report the sequence, de novo assembly, and annotation of the jaguar (Panthera onca) genome, a novel genome sequence for the leopard (Panthera pardus), and comparative analyses encompassing all living Panthera species. Demographic reconstructions indicated that all of these species have experienced variable episodes of population decline during the Pleistocene, ultimately leading to small effective sizes in present-day genomes. We observed pervasive genealogical discordance across Panthera genomes, caused by both incomplete lineage sorting and complex patterns of historical interspecific hybridization. We identified multiple signatures of species-specific positive selection, affecting genes involved in craniofacial and limb development, protein metabolism, hypoxia, reproduction, pigmentation, and sensory perception. There was remarkable concordance in pathways enriched in genomic segments implicated in interspecies introgression and in positive selection, suggesting that these processes were connected. We tested this hypothesis by developing exome capture probes targeting ~19,000 Panthera genes and applying them to 30 wild-caught jaguars. We found at least two genes (DOCK3 and COL4A5, both related to optic nerve development) bearing significant signatures of interspecies introgression and within-species positive selection. These findings indicate that post-speciation admixture has contributed genetic material that facilitated the adaptive evolution of big cat lineages.

Accurately estimating home range and understanding movement behavior can provide important information on ecological processes. Advances in data collection and analysis have improved our ability to estimate home range and movement parameters, both of which have the potential to impact species conservation. Fitting continuous-time movement model to data and incorporating the autocorrelated kernel density estimator (AKDE), we investigated range residency of forty-four jaguars fit with GPS collars across five biomes in Brazil and Argentina. We assessed home range and movement parameters of range resident animals and compared AKDE estimates with kernel density estimates (KDE). We accounted for differential space use and movement among individuals, sex, region, and habitat quality. Thirty-three (80%) of collared jaguars were range resident. Home range estimates using AKDE were 1.02 to 4.80 times larger than KDE estimates that did not consider autocorrelation. Males exhibited larger home ranges, more directional movement paths, and a trend towards larger distances traveled per day. Jaguars with the largest home ranges occupied the Atlantic Forest, a biome with high levels of deforestation and high human population density. Our results fill a gap in the knowledge of the species´ ecology with an aim towards better conservation of this endangered/critically endangered carnivore-the top predator in the Neotropics. Este estudo investigou a exposição de onças-pintadas de vida livre a agentes virais selecionados em duas unidades de conservação federais no Pantanal de Mato Grosso, Brasil. Para a análise desses agentes virais, a maioria de caráter zoonótico, foram utilizados métodos sorológicos e moleculares. Nenhuma das onze onças-pintadas examinadas foi positiva na técnica de real-time RT-PCR para a detecção molecular dos agentes da Influenza aviária e Febre do Nilo Ocidental (WNF). Somente um animal foi positivo sorologicamente para a o vírus da Encefalite Equina do Leste (EEE) pela Microtécnica de vírus neutralização em culturas de células VERO, sendo este o primeiro relato da exposição de onças-pintadas. Todos os animais examinados s foram negativos para o vírus da Encefalite Equina do Oeste (WEE) e Venezuelana (VEE). Amostras de soro colhidas de 11 onças-pintadas foram submetidas a adois testes distintos para a detecção de anticorpos contra o vírus da raiva (Teste Rápido de Inibição de Foco de Fluorescência – RFFIT e Microteste Simplificado de Inibição da Fluorescência - SFIMT), resultando em cinco animais positivos, dos quase dois positivos para cada teste e um positivo quando submetido aos dois testes sorológicos. Além disso, três das 14 amostras submetidas a técnica de soroneutralização foram positivas para a pesquisa de anticorpos contra o vírus da cinomose (CDV) e 15 amostras positivas das 17 analisadas para a pesquisa de anticorpos contra o parvovírus canino (CPV) foram identificadas pela técnica de Inibição da Hemaglutinação (HI). De acordo com os resultados deste estudo, é pouco provável que os agentes virais aqui analisados representem ameaça à população de onças-pintadas nesta região.

The Astyanax species of the bimaculatus group include morphologically similar representatives that share the same color pattern, corresponding to at least 20 species. Specimens from the Sao Francisco River basin and from the Grande River (Upper Parana basin) are included in this group. In the early 1960’s, the Piumhi River, an original tributary of the Grande River, was transposed to the Sao Francisco River basin due to the construction of the Furnas Hydroelectric Power, in the state of Minas Gerais, Brazil. In this context, we characterized representatives of the bimaculatus group from both basins and from the transposition region of the Piumhi River, using chromosome characteristics obtained by conventional analyzes and fluorescence in situ hybridization with rDNA probes. All the analyzed specimens presented 2n = 50 chromosomes and similar localization of the nucleolus organizer regions (Ag-NORs), 18S rDNA and 5S rDNA sites. However, diagnostic chromosome differences were detected concerning the number of submetacentric and acrocentric chromosomes in the karyotypes, making possible the characterization of five distinct karyotypes (Cytotypes A–E), where the specimens from the Sao Francisco and from the Grande River correspond to Cytotypes A and B, respectively. In addition, these two Cytotypes were sympatrically found in the Piumhi River with specimens carrying a clear intermediate karyotype (Cytotype C), as well as with others harboring different karyotypes (Cytotypes D and E). Our data indicate that Cytotypes A and B probably diverged from each other during the prior geographic isolation. However, despite their chromosomal differentiations, they still maintain a close relationship which allows interbreeding and the formation of a secondary hybrid zone, due to the breakdown of the geographic isolation.

Constitutive heterochromatin makes up a substantial portion of the genome of eukaryotes and is composed mainly of satellite DNA repeating sequences in tandem. Some satellite DNAs may have been derived from transposable elements. These repetitive sequences represent a highly dynamic component of rapid evolution in genomes. Among the genus Astyanax, the As51 satellite DNA is found in species that have large distal heterochromatic blocks, which may be considered as derived from a transposable DNA element. In the present study, As51 satellite DNA was mapped through in situ fluorescent hybridization in the chromosomes of five species of the genus. The possible roles of this type of saltatory DNA type in the genome of the species are discussed, along with its use for the phylogenetic grouping of the genus Astyanax, together with other shared chromosomal characters. However, the number of As51 clusters is presented as a homoplastic characteristic, thereby indicating evident genomic diversification of species with this type of DNA.

In the 1960s, as a part of the construction of the Furnas Hydroelectric Power Dam, Minas Gerais, Brazil, the Rio Piumhi was diverted from the Rio Grande drainage to the São Francisco River basin, with the resulting introduction of species from one basin to the other. Chromosomal characteristics of various populations of Astyanax fasciatus sensu Eigenmann from the transect region were studied using cytogenetic techniques, with the goal to identify and map the dispersal of invasive species in the Rio São Francisco. Populations of the Rio Grande and Rio Piumhi are distinct from those of the São Francisco basin based on chromosome markers associated to the heterochromatin, Ag-NORs, 18S rDNA, 5S rDNA, and As-51 satellite DNA sites. Individuals with karyomorph originally present in the Piumhi drainage are now present in the São Francisco basin, downstream from the transposition channel. This expansion of the distribution corroborates a hypothesis of progressive substitution of native populations by the invasive Piumhi form.

Chromosomal characterization of constitutive heterochromatin was performed in Astyanax sp. D, a species found in the Iguaçu river, using C-banding, fluorescent in situ hybridization (FISH) of the repetitive DNA family As51, localization of the 18S rDNA sites and base-specific fluorochromes. The sites of the satellite DNA coincide with large heterochromatic blocks present in this species, namely CMA3- and DAPI-. The many 18S rDNA sites are CMA3+. Similarity between the large heterochromatic blocks is suggested, as well as the identification of two types of heterochromatin, one related to the 18S rDNA domains and the other to the As51 regions.

Taxonomic studies of the genus Astyanax from the Iguac xu River (Brazil) indicate that they may be differentiated into 11 distinct species, some of which have not been formally described and named so for. This study focuses on three of these species, Astyanax sp. B, Astyanax sp. C and Astyanax sp. D from the Upper Iguac xu River Basin. Comparative cytogenetic analyses of Cbanding, Ag-NORs (silver nitrate stained nucleolar organizer region) and 18S and 5S rDNA corroborate that they are distinct species. A diploid number of 50 chromosomes and similar karyotypic formulae were observed in the three taxa, with the exception of Astyanax sp. D that differs in the number of submetacentric and subtelocentric chromosomes. However, the NOR silver-staining pattern, the heterochromatic bands (C-bands) and the mapping of the 18S and 5S rDNA sites in the chromosomes showed divergences between all three species under study, supporting the occurrence of distinct evolutionary units.

a State, Brazil). All specimens had a diploid number of 48 chromosomes, with a karyotype constituted by 4mþ14smþ18stþ12a and fundamental number of 84. The C-positive heterochromatin was distributed over the nucleolar organizer regions (NORs) in the centromeric regions and on short arms of metacentric and submetacentric chromosomes. Most of this heterochromatin was AT-rich, except in the NORs, which were rich in GC, as detected by double staining with chromomycin A3/49-6-diamin-2-phenylindole. Cytogenetic analysis with Astyanax sp. D revealed a karyotype of 2n=50 with 2M+26SM+6ST+16A, besides a triploid specimen showing 2n=75 chromosomes (3M+39SM+9ST+24A). C-banding strongly stained the terminal regions of several SM-ST-A chromossomes. Two pairs of acrocentric chromosomes presented interstitial heterochromatin, this state being polymorphic and occuring due to possible paracentric inversions. The results obtained with the AluI restriction enzyme and A3 chromomycin were similar to the C-banding. Relationships were proposed between Astyanax sp. D and A. scabripinnis, as well as considerations for a possible origin of the triploid specimen (2n=3x=75). When comparing the present results with cytogenetic features of other endemic Astyanax species in the Iguaçu river (A. sp. B and C), a clear differentiation was observed between them, indicating cytogenetics as an important cytotaxonomic tool.Análises citogenéticas em Astyanax sp. D evidenciaram 2n=50 cromossomos e um cariótipo com 2M+26SM+6ST+16A. O bandamento C destacou as regiões teloméricas de diversos cromossomos SM-ST-A. Dois pares de cromossomos acrocêntricos possuem heterocromatina intersticial, sendo este estado polimórfico decorrente de prováveis inversões paracêntricas. O resultados obtidos com a enzima de restrição AluI e a Cromomicina A3 foram semelhantes aos do bandamento C. São propostas relações de parentesco entre Astyanax sp. D e Astyanax scabripinnis, bem como considerações sobre a possível origem do exemplar triplóide (2n=3x=75). Ao comparar os resultados deste trabalho com outras espécies de Astyanax do Rio Iguaçu, estas espécies se tornam claramente distinguíveis, evidenciando a citogenética como uma importante ferramenta taxonômica.

Cytogenetic studies have assisted in the taxonomic classification of organisms, especially those involving species with highly similar morphologic characteristics, or so-called cryptic species. Strongylura marina and Strongylura timucu collected from Paranagua Bay, Parana Coast in Southern Brazil are considered cryptic species, and the identification of interspecific variations based on the number and/or morphology of its chromosomes may serve as differentiating cytotaxonomic markers. Chromosomes of the two species were subjected to different banding and staining methods (C-, Ag-, and DAPI-CMA3), as well as chromosomal mapping of major rDNA (45S), revealed with an 18S probe by fluorescence in situ hybridization (FISH). The pattern of distribution of constitutive heterochromatin showed distinct features involving the pericentromeric and telomeric bands in both species. In S. marina, chromosome 1 represents the main species-specific marker, appearing almost entirely heterochromatic. In both species, the 45S rDNA is located at terminal region of the short arm of the chromosome 6, as detected by silver nitrate staining and FISH. Despite the apparent conserved diploid number of 48 chromosomes, data on the karyotype microstructure characterize the cytogenetic profile of the genus and may allow the establishment of cytotaxonomic and evolutionary inferences for these fishes.

The characids belong to the greater freshwater fish family of the world with around 700 recognized species. Whithin this group the genus Astyanax includes several species with unclear phylogenetic relationships, showing a wide karyotypic diversity. In the highly endemic Iguacu river basin these problems are also observed being distinguished 6 unnamed Astyanax species (A to F). In the present work were cytogenetically studied 2 populations of A. sp. C from the first plateau of the Iguacu river (Parana State, Brazil). Fishes of both populations are similar in chromosome number, having 2n=50, however were evidenced clear differences in C-banding and nucleolar organizing regions (NORs) patterns. With the use of the restriction enzyme Alu I were obtained the same results as C-bands. These results indicate the complexity of Astyanax and the importance of further taxonomic revisions joining morphological, cytogenetic and molecular approachings.

Specimens of Imparfinis schubarti (Gomes, 1956) collected in the Piumhi river drainage, state of Minas Gerais, Brazil, were studied cytogenetically. The river was diverted from the Rio Grande Basin into the São Francisco basin in the early 1960s. All individuals presented 2n = 58 chromosomes, including 18 metacentric, 34 submetacentric and six subtelocentric chromosomes. A secondary constriction was observed in the interstitial region of the long arm of the largest chromosome pair, coinciding with the NOR. A single conspicuous heterochromatic block located in the largest pair of metacentric chromosomes was observed, adjacent to the secondary constriction. A detectable 18S rDNA probe hybridization region occurs in only one chromosome pair and is synthenic with the marking obtained with 5S rDNA probe. These results fit the cytogenetic pattern previously described for the genus Imparfinis Eigenmann & Norris, 1900.

In this study, five species of marine fishes from the Paranaguá Bay in the Brazilian coast were evaluated. Eucinostomus argenteus and Diapterus rhombeus (Gerreidae) presented 48 chromosomes, all of which more acrocentric (FN = 48); Strongylura timucu and S. marina (Belonidae) also presented 48 chromosomes, but with a higher karyotypic complexity than the Gerreidae, 10M+2SM+36A (FN = 60) and 4M+44A (FN = 52), respectively. The fifth species, Mugil curema (Mugilidae), different than the others, presented only 28 chromosomes 20M+4ST+4A (FN = 48). The species presented diversity in the karyotypic macro-structure, which should be relevant for the cytotaxonomy and the evolution of this group of the vertebrate.Nas últimas décadas tem ocorrido no Brasil um incremento de estudos cariotípicos em peixes marinhos. Atualmente são conhecidos os cariótipos de 118 espécies, distribuídas em 43 famílias e 80 gêneros. Foram estudadas cinco espécies de peixes marinhos do complexo estuarino da Baía de Paranaguá na costa brasileira. Eucinostomus argenteus e Diapterus rhombeus (Gerreidae), apresentaram 48 cromossomos todos acrocêntricos (NF = 48); Strongylura timucu e S. marina (Belonidae) apresentaram 48 cromossomos, porém com complexidade cariotípica maior do que apresentada pelos gerreídeos, 10M+2SM+36A (NF = 60) e 4M+44A (NF = 52), respectivamente. A quinta espécie, Mugil curema (Mugilidae), ao contrário das outras quatro espécies aqui analisadas, apresentou apenas 28 cromossomos 20M+4ST+4A (NF = 48). Apesar da tendência em se verificar um cariótipo constituído por 48 cromossomos em teleósteos marinhos, as espécies aqui analisadas apresentam uma diversidade para a macroestrutura cariotípica a ser considerada para a citotaxonomia e evolução desse grupo de vertebrados.

Chromosomal analyses were performed in the fish Astyanax sp.D collected from three different points: two streams from the right bank and one from the left bank of the Upper Iguaçu River, Paraná State, Brazil. The individuals from all localities possess 2n = 50 chromosomes and a FN = 84 (4m+24sm+6st+16a). The C-banding pattern was similar in all populations. However, within each population, an interindividual variation concerning the number and localization of heterochromatic bands was observed. Some of these variations were quantified in each population, and the results indicate that the samples were not different when studying the variable frequencies. Considering that Astyanax sp.D is typical in the headwaters of the Iguaçu River, these results were not expected. The data indicate that gene flow is occurring and that the Iguaçu River is not an ecological barrier among the Astyanax sp. D populations.

We present the karyotypic characterization of 26 specimens of the side-necked turtle Hydromedusa tectifera collected in the upper Iguaçu River, Paraná state, Brazil. The turtles were cytogenetically analyzed using Giemsa staining and other banding techniques (C, G, Ag-NOR and CMA3) as well as fluorescence in situ hybridization (FISH) with a rDNA 18S probe. All the specimens showed a diploid number of 58 composed of 22 macro and 36 microchromosomes. The Ag-NOR, CMA3 and FISH techniques permitted the identification and characterization of the chromosome pairs bearing nucleolus organizer regions (NORs), while G-banding facilitated a better recognition and pairing of macrochromosomes. These data agree with some information available in the literature and should be very useful for further cytotaxonomic and cytosystematic studies.
