Abstract: International audience; Most insect herbivores are adapted to feed on a few host plants only, but the mechanisms underlying plant specialization are poorly understood. One of the dominant hypotheses is that insects inject an oral secretion into the plant that manipulates the plant's defences resulting in induced susceptibility. This hypothesis was tested on the pea aphid (Acyrthosiphon pisum), which forms multiple biotypes, each specialized on specific legume hosts. In particular, the hypothesis that a pea-adapted biotype can induce host susceptibility on pea and increase the fitness of a non-pea-adapted biotype was tested. Survival and fecundity of an alfalfa-adapted biotype on peas were enhanced by co-infestation but not by pre-infestation with a pea-adapted biotype. The electrical penetration graph (EPG) method was then used to dissect the components of the alfalfa biotype feeding behaviour on peas with and without co-infestation by the pea biotype. While probing time did not differ between these two conditions, differences were observed in most other EPG parameters. When co-infested with the pea-adapted biotype, the alfalfa-adapted biotype was more likely to establish a phloem feeding and exhibited a longer ingestion time on peas. Our results indicated that the pea-adapted biotype induced pea susceptibility by increasing the accessibility of non-pea-adapted aphids to the host phloem sap. As a result, the alfalfa biotype increased its survival and fecundity. These results contribute to understanding how aphids manipulate their specific host plants.

Abstract: Cuticle are multifunctional hydrophobic biocomposites that protect aerial organs of plants. Along plant development, plant cuticle must accommodate different mechanical constraints combining extensibility and stiffness, the corresponding structure-function relationships are unknown. Recent data showed a fine architectural tuning of the cuticle architecture and the corresponding chemical clusters along fruit development which raise the question of their impact on the mechanical properties of the cuticle.We investigated the in-depth nanomechanical properties of tomato fruit cuticle from early development to ripening, in relation to chemical and structural heterogeneities by developing a correlative multimodal imaging approach.Unprecedented sharps heterogeneities were evidenced with the highlighting of an in-depth mechanical gradient and a ‘soft’ central furrow that were maintained throughout the plant development despite the overall increase in elastic modulus. In addition, we demonstrated that these local mechanical areas are correlated to chemical and structural gradients.This study shed light on a fine tuning of mechanical properties of cuticle through the modulation of their architecture, providing new insight for our understanding of structure-function relationships of plant cuticle and for the design of biosinpired material.

Abstract: International audience; Yield reduction over the years is a phenomenon that affects many fruit tree species, including hazel (Corylus avellana). Studying plant architecture could provide useful information for improving crop management, such as pruning, to reduce the impact of yield reductions. This study aimed to compare the relationship between bud composition and shoot length in hazel shoots. The experiment was carried out in January 2020 and 2021 in Deruta, Perugia (Italy). In 2020, 120 1-year-old shoots of C. avellana 'Tonda di Giffoni' were sampled. In 2021, the same observations were performed on lateral and apical 1-year-old shoots that had burst from parental buds in spring 2020. Depending on its length, each shoot was sorted into four categories: short (Sh) when shorter than 5 cm, medium (Me) when between 5 and 20 cm, long (Lo) when between 20 and 40 cm, and very long (VLo) when longer than 40 cm. For each shoot, shoot length, axillary and apical bud fate was recorded at each node: vegetative bud, latent bud, catkin, and mixed bud (V, B, C, and M, respectively). Percentage of lateral mixed buds was higher in longer shoots than in shorter ones while that of vegetative buds remained constant whatever the length. Fruit set, computed as the ratio between the number of nuts and the number of mixed buds, was significantly higher in Lo and VLo shoots. It was observed that the length of lateral proleptic shoots, developed in 2021, was related to the length of the bearer shoot from which it was developed. Proleptic lateral Sh shoots were the most frequent among all length categories while longer proleptic shoots (Lo and VLo) were originated only from Lo and VLo shoots. Managing tree architecture, considering a good balance between long and short shoots, and mixed and vegetative buds, could help control and prevent yield decrease in hazelnut trees.

Abstract: Class II water-soluble chlorophyll proteins (WSCPs) from Brassicaceae are non-photosynthetic proteins that bind with chlorophyll (Chl) and its derivatives. The physiological function of WSCPs is still unclear, but it is assumed to be involved in stress responses, which is likely related to their Chl-binding and protease inhibition (PI) activities. Yet, the dual function and simultaneous functionality of WSCPs must still be better understood. Here, the biochemical functions of Brassica napus drought–induced 22-kDa protein (BnD22), a major WSCP expressed in B. napus leaves, were investigated using recombinant hexahistidine-tagged protein. We showed that BnD22 inhibited cysteine proteases, such as papain, but not serine proteases. BnD22 was able to bind with Chla or Chlb to form tetrameric complexes. Unexpectedly, BnD22–Chl tetramer displays higher inhibition toward cysteine proteases, indicating (i) simultaneous Chl-binding and PI activities and (ii) Chl-dependent activation of PI activity of BnD22. Moreover, the photostability of BnD22–Chl tetramer was reduced upon binding with the protease. Using three-dimensional structural modeling and molecular docking, we revealed that Chl binding favors interaction between BnD22 and proteases. Despite its Chl-binding ability, the BnD22 was not detected in chloroplasts but rather in the endoplasmic reticulum and vacuole. In addition, the C-terminal extension peptide of BnD22, which cleaved off post-translationally in vivo, was not implicated in subcellular localization. Instead, it drastically promoted the expression, solubility and stability of the recombinant protein.

Cet article de synthèse présente l’état de l’art des répliques foliaires et des phénomènes concomitants, pour une application spatiale.

Abstract: Plant ecologists and molecular biologists have long considered the hypothesis of a trade-off between plant growth and defence separately. In particular, how genes thought to control the growth–defence trade-off at the molecular level relate to trait-based frameworks in functional ecology, such as the slow–fast plant economics spectrum, is unknown. We grew 49 phenotypically diverse rice genotypes in pots under optimal conditions and measured growth-related functional traits and the constitutive expression of 11 genes involved in plant defence. We also quantified the concentration of silicon (Si) in leaves to estimate silica-based defences. Rice genotypes were aligned along a slow–fast continuum, with slow-growing, late-flowering genotypes versus fast-growing, early-flowering genotypes. Leaf dry matter content and leaf Si concentrations were not aligned with this axis and negatively correlated with each other. Live-fast genotypes exhibited greater expression of OsNPR1, a regulator of the salicylic acid pathway that promotes plant defence while suppressing plant growth. These genotypes also exhibited greater expression of SPL7 and GH3.2, which are also involved in both stress resistance and growth. Our results do not support the hypothesis of a growth–defence trade-off when leaf Si and leaf dry matter content are considered, but they do when hormonal pathway genes are considered. We demonstrate the benefits of combining ecological and molecular approaches to elucidate the growth–defence trade-off, opening new avenues for plant breeding and crop science.

Abstract: International audience; The metabolic scaling theory (MST) predicts quasi-universal trait-size relationships in plants, characterised by a unique allometric exponent within and across large taxonomic scales. However, recent studies have identified variability in allometric relationships, without a clear understanding of the modulating role played by genetic variation and environment.Here, we investigated (1) the allometric relationships for two central traits of MST, namely total leaf area and plant growth rate, in the model species Arabidopsis thaliana, (2) the variability of plant allometries between genotypes and (3) the plastic responses of plant allometries under water deficit, high temperature and their interaction.Using a population of 120 genotypes, we found that intraspecific allometries adhered on average with MST predictions. However, a broad variability but a moderate plasticity in the allometric exponents was observed across genotypes and environments. Allometric exponents were impacted significantly, yet weakly, by water deficit, but not by high temperature. Moreover, genotypes that deviated from MST predictions exhibited more plasticity in trait-size relationships than genotypes that followed MST predictions.Our study suggests that plant allometry is genetically variable and might be related to different adaptive strategies to cope with stressing conditions. Thus, our results highlights the need of assessing trait-size relationships within species to understand the mechanisms of plant adaptation to contrasted environments.

Abstract: International audience; The root system of a plant is composed of the primary, lateral and adventitious roots (ARs). Lateral roots always develop from roots, whereas ARs form from stem or leaf‐derived cells. AR formation is part of the normal development of the plant and occurs naturally, like in most monocotyledonous for which they constitute the main root system or in many dicotyledonous species that propagate vegetatively. Adventitious rooting is an essential step for vegetative propagation of economically important horticultural and woody species as it allows clonal propagation and rapid fixation of superior genotypes prior to their introduction into production or breeding programmes. Development of ARs is a complex process that is affected by multiple endogenous and environmental factors, including phytohormones; light; nutritional status; associated stress responses, such as wounding; and genetic characteristics.

Abstract: International audience; Emergence of secondary roots through parental tissue is a highly controlled developmental process. Although the model plant Arabidopsis has been useful to uncover the predominant role of auxin in this process, its simple root structure is not representative of how emergence takes place in most plants, which display more complex root anatomy. White lupin is a legume crop producing structures called cluster roots, where closely spaced rootlets emerge synchronously. Rootlet primordia push their way through several cortical cell layers while maintaining the parent root integrity, reflecting more generally the lateral root emergence process in most multilayered species. In this study, we showed that lupin rootlet emergence is associated with an upregulation of cell wall pectin modifying and degrading genes under the active control of auxin. Among them, we identified LaPG3, a polygalacturonase gene typically expressed in cells surrounding the rootlet primordium and we showed that its downregulation delays emergence. Immunolabeling of pectin epitopes and their quantification uncovered a gradual pectin demethylesterification in the emergence zone, which was further enhanced by auxin treatment, revealing a direct hormonal control of cell wall properties. We also report rhamnogalacturonan-I modifications affecting cortical cells that undergo separation as a consequence of primordium outgrowth. In conclusion, we describe a model of how external tissues in front of rootlet primordia display cell wall modifications to allow for the passage of newly formed rootlets.

Abstract: International audience; The coordinated distribution of inorganic phosphate (Pi) between roots and shoots is an important process that plants use to maintain Pi homeostasis. SHORT-ROOT (SHR) is well characterized for its function in root radial patterning. Here we demonstrate a role of SHR in controlling Pi allocation from root to shoot by regulating PHOSPHATE1 in the root differentiation zone. We recovered a weak mutant allele of SHR in Arabidopsis that accumulates much less Pi in the shoot and shows a constitutive Pi starvation response under Pi-sufficient conditions. In addition, Pi starvation suppresses SHR protein accumulation and releases its inhibition on the HD-ZIP III transcription factor PHB. PHB accumulates and directly binds the promoter of PHOSPHATE2 to upregulate its transcription, resulting in PHOSPHATE1 degradation in the xylem-pole pericycle cells. Our findings reveal a previously unrecognized mechanism of how plants regulate Pi translocation from roots to shoots.Plants confronted with nutrient deficiency allocate more resources to roots to maximize nutrient acquisition and growth. This study uncovers how plants repress phosphate uploading to curtail the long-distance transportation of phosphate to shoots.

Abstract: We investigated the impact of uranium (U) on Arabidopsis thaliana seed germination and on the early stages of seedling development. Using an in vitro device, we demonstrated that uranyl ion had a very low impact on Arabidopsis seed germination but had a drastic effect on the development of Arabidopsis seedlings. We showed that the two soil bacterial strains Microbacterium sp. ViU2A and Stenotrophomonas bentonitica BII-R7, which are able to tolerate high concentrations of U, strongly reduced the toxic effects of the metal on the seedling development. This protective effect is specific to these soil bacteria, as E. coli was not able to protect seedlings. The analysis of the distribution of U between Arabidopsis seedlings and soil bacteria showed that the protective effect of the bacteria was due to their ability to sequester U either by biosorption at the level of the cell surface and/or by intracellular or extracellular biomineralization.This study reveals that these bacteria are very good candidates for use in phytoremediation strategies in the case of phytostabilisation of U-polluted soils. They would be also useful to limit the contamination of the food chain by U because they would limit the entry of this toxic element in crop plants.

Abstract: International audience; As perennial plants, fruit trees must cope, individually, with the fluctuating threat of multiple pathogens over the years. In this long-lasting battle for plant immunity, disease resilience is emerging as a key mechanism for tree survival and fitness. More fundamental research is required to improve our understanding of disease resilience mechanisms, which in turn could be particularly relevant for a more sustainable fruit production. As this approach is novel for that field, we propose i) a clear definition of resilience for fruit producing trees, ii) a methodology for studying its phenotypic components which requires repeated measures of “resilience biomarkers”, iii) to decipher the genetic architecture of resilience components, and iv) an innovative strategy based on high-throughput phenotyping and genomics for identifying resilient genotypes. All in all, disease resilience appears as a meaningful breeding perspective in a context of unprecedented plant protection restrictions.

Abstract: Plant architecture plays a major role in flowering and therefore in crop yield. Attempts to visualize and analyse strawberry plant architecture have been few to date. Here, we developed open-source software combining two- and three-dimensional representations of plant development over time along with statistical methods to explore the variability in spatio-temporal development of plant architecture in cultivated strawberry. We applied this software to six seasonal strawberry varieties whose plants were exhaustively described monthly at the node scale. Results showed that the architectural pattern of the strawberry plant is characterized by a decrease of the module complexity between the zeroth-order module (primary crown) and higher-order modules (lateral branch crowns and extension crowns). Furthermore, for each variety, we could identify traits with a central role in determining yield, such as date of appearance and number of branches. By modeling the spatial organization of axillary meristem fate on the zeroth-order module using a hidden hybrid Markov/semi-Markov mathematical model, we further identified three zones with different probabilities of production of branch crowns, dormant buds, or stolons. This open-source software will be of value to the scientific community and breeders in studying the influence of environmental and genetic cues on strawberry architecture and yield.

Abstract: During meiosis, the chromosome axes and synaptonemal complex mediate chromosome pairing and homologous recombination to maintain genomic stability and accurate chromosome segregation. In plants, ASYNAPSIS 1 (ASY1) is a key component of the chromosome axis that promotes inter-homolog recombination, synapsis and crossover formation. Here, the function of ASY1 has been cytologically characterized in a series of hypomorphic wheat mutants. In tetraploid wheat, asy1 hypomorphic mutants experience a reduction in chiasmata (crossovers) in a dosage-specific manner, resulting in failure to maintain crossover (CO) assurance. In mutants with only one functional copy of ASY1, distal chiasmata are maintained at the expense of proximal and interstitial chiasmata, indicating that ASY1 is required to promote chiasma formation away from the chromosome ends. Meiotic prophase I progression is delayed in asy1 hypomorphic mutants and is arrested in asy1 null mutants. In both tetraploid and hexaploid wheat, single asy1 mutants exhibit a high degree of ectopic recombination between multiple chromosomes at metaphase I. To explore the nature of the ectopic recombination, Triticum turgidum asy1b-2 was crossed with wheat-wild relative Aegilops variabilis. Homoeologous chiasmata increased 3.75-fold in Ttasy1b-2/Ae. variabilis compared to wild type/Ae. variabilis, indicating that ASY1 suppresses chiasma formation between divergent, but related chromosomes. These data suggest that ASY1 promotes recombination along the chromosome arms of homologous chromosomes whilst suppressing recombination between non-homologous chromosomes. Therefore, asy1 mutants could be utilized to increase recombination between wheat wild relatives and elite varieties for expediting introgression of important agronomic traits.

Abstract: The elevation of atmospheric CO2 leads to a decline in the plant mineral content, which poses a major threat to food security in the coming decades. To date, very few genes have been identified as having a role in the negative effect of elevated CO2 on plant mineral composition. Yet, several studies have shown a certain degree of diversity in the ionome's response to elevated CO2, associated with genotypic variation. This suggests the existence of genetic factors controlling the effect of CO2 on ionome composition. However, no large-scale studies have been carried out to date to explore the genetic diversity of the ionome responses to elevated CO2. Here, we used six hundred Arabidopsis thaliana accessions, representing geographical distributions ranging from worldwide to regional and local environments, to analyze the natural genetic variation underlying the negative effect of elevated CO2 on the ionome composition in plants. We show that the growth under elevated CO2 leads to a global and important decrease of the ionome content whatever the geographic distribution of the population. We also observed a high range of genetic diversity in the response of the ionome composition to elevated CO2, and we identified sub-populations, showing effects on their ionome ranging from the most pronounced to resilience or even to a benefit in response to elevated CO2. Using genome-wide association mapping on the response of each mineral element to elevated CO2 or on integrative traits, we identified a large set of QTLs and genes associated with the ionome response to elevated CO2. Finally, we demonstrate that manipulating the function of one of these genes can mitigate the negative effect of elevated CO2 on the plant mineral composition. Therefore, this resource will contribute to understand the genetic mechanisms underlying the negative effect of elevated CO2 on the mineral composition of plants, and to the development of biofortified crops adapted to a high-CO2 world.

Abstract: International audience; Tsetse flies (Glossina spp.) are major vectors of African trypanosomes, causing either Human or Animal African Trypanosomiasis (HAT or AAT). Several approaches have been developed to control the disease, among which is the anti-vector Sterile Insect Technique. Another approach to anti-vector strategies could consist of controlling the fly's vector competence through hitherto unidentified regulatory factors (genes, proteins, biological pathways, etc.). The present work aims to evaluate the protein abundance in the midgut of wild tsetse flies (Glossina palpalis palpalis) naturally infected by Trypanosoma congolense s.l. Infected and non-infected flies were sampled in two HAT/AAT foci in Southern Cameroon. After dissection, the proteomes from the guts of parasite-infected flies were compared to that of uninfected flies to identify quantitative and/or qualitative changes associated with infection. Among the proteins with increased abundance were fructose-1,6-biphosphatase, membrane trafficking proteins, death proteins (or apoptosis proteins) and SERPINs (inhibitor of serine proteases, enzymes considered as trypanosome virulence factors) that displayed the highest increased abundance. Abstract: Phenotypic integration is a concept related to the cascade of trait relationships from the lowest organizational levels, i.e. genes, to the highest, i.e. whole-organism traits. However, the cause-and-effect linkages between traits are notoriously difficult to determine. In particular, we still lack a mathematical framework to model the relationships involved in the integration of phenotypic traits. Here, we argue that allometric models developed in ecology offer testable mathematical equations of trait relationships across scales. We first show that allometric relationships are pervasive in biology at different organizational scales and in different taxa. We then present mechanistic models that explain the origin of allometric relationships. In addition, we emphasized that recent studies showed that natural variation does exist for allometric parameters, suggesting a role for genetic variability, selection and evolution. Consequently, we advocate that it is time to examine the genetic determinism of allometries, as well as to question in more detail the role of genome size in subsequent scaling relationships. More broadly, a possible—but so far neglected—solution to understand phenotypic integration is to examine allometric relationships at different organizational levels (cell, tissue, organ, organism) and in contrasted species.

Abstract: The loss of large frugivores leads to seed dispersal loss and regeneration failure of numerous large-seeded trees near mother trees. Although Janzen–Connell effects are considered as the primary underlying cause, other factors remain understudied. Here, we used a field experiment to test the impact of flesh persistence on the recruitment of two large-seeded Sapotaceae species that lost their dispersers. In the rainforest of Mare Longue (Réunion), we sowed 3840 seeds in a four-factor design: seed treatment (seed cleaning; flesh persistence), canopy closure (understory; gap), year of sowing (01/2018; 11/2019) and species (Labourdonnaisia calophylloides, Mimusops balata). We also used camera traps to evaluate the impact of extant vertebrates. Seed treatment was by far the most influential factor: flesh persistence led to seedling recruitment divided by 3,2 on average, mainly due to failure of germination or seedling emergence. There were also significant variations in recruitment between species, years and canopy closure levels, notably due to the behaviour of the invasive fauna, especially giant snails that could unexpectedly restore recruitment by feeding on fruit flesh. Together, our results demonstrate strongly depleted recruitment due to flesh persistence and the importance of field experiments to understand the processes at work in complex ecosystems with novel plant–animal interactions.

Abstract: Background and Aims Condensed tannins, responsible for berry and wine astringency, may have been selected during grapevine domestication. This work examines the phylogenetic distribution of condensed tannins throughout the Vitaceae phylogenetic tree. Methods Green berries and mature leaves of representative true-to-type members of the Vitaceae were collected before ‘véraison’, freeze-dried and pulverized, and condensed tannins were measured following depolymerization by nucleophilic addition of 2-mercaptoethanol to the C4 of the flavan-3-ol units in an organic acidic medium. Reaction products were separated and quantified by ultrahigh pressure liquid chromatography/diode array detection/mass spectrometry. Key Results and Conclusions The original ability to incorporate epigallocatechin (EGC) into grapevine condensed tannins was lost independently in both the American and Eurasian/Asian branches of the Vitaceae, with exceptional cases of reversion to the ancestral EGC phenotype. This is particularly true in the genus Vitis, where we now find two radically distinct groups differing with respect to EGC content. While Vitis species from Asia are void of EGC, 50 % of the New World Vitis harbour EGC. Interestingly, the presence of EGC is tightly coupled with the degree of leaf margin serration. Noticeably, the rare Asian EGC-forming species are phylogenetically close to Vitis vinifera, the only remnant representative of Vitis in Eurasia. Both the wild ancestral V. vinifera subsp. sylvestris as well as the domesticated V. vinifera subsp. sativa can accumulate EGC and activate galloylation biosynthesis that compete for photoassimilates and reductive power.

Abstract: Data from functional trait databases have been increasingly used to address questions related to plant diversity and trait-environment relationships. However, such databases provide intraspecific data that combine individual records obtained from distinct populations at different sites and, hence, environmental conditions. This prevents distinguishing sources of variation (e.g., genetic-based variation vs. phenotypic plasticity), a necessary condition to test for adaptive processes and other determinants of plant phenotypic diversity. Consequently, individual traits measured under common growing conditions and encompassing within-species variation across the occupied geographic range have the potential to leverage trait databases with valuable data for functional and evolutionary ecology. Here, we recorded 16 functional traits and leaf hyperspectral reflectance (NIRS) data for 721 widely distributed Arabidopsis thaliana natural accessions grown in a common garden experiment. These data records, together with meteorological variables obtained during the experiment, were assembled to create the AraDiv dataset. AraDiv is a comprehensive dataset of A. thaliana’s intraspecific variability that can be explored to address questions at the interface of genetics and ecology.

Abstract: International audience; Tree stem respiration (R S) is a substantial component of the forest carbon balance. The mass balance approach uses stem CO 2 efflux and internal xylem fluxes to sum up R S , while the oxygen-based method assumes O 2 influx as a proxy of R S. So far, both approaches have yielded inconsistent results regarding the fate of respired CO 2 in tree stems, a major challenge for quantifying forest carbon dynamics. We collected a data set of CO 2 efflux, O 2 influx, xylem CO 2 concentration, sap flow, sap pH, stem temperature, nonstructural carbohydrates concentration and potential phosphoenolpyruvate carboxylase (PEPC) capacity on mature beech trees to identify the sources of differences between approaches. The ratio of CO 2 efflux to O 2 influx was consistently below unity (0.7) along a 3-m vertical gradient, but internal fluxes did not bridge the gap between influx and efflux, nor did we find evidence for changes in respiratory substrate use. PEPC capacity was comparable with that previously reported in green current-year twigs. Although we could not reconcile differences between approaches, results shed light on the uncertain fate of CO 2 respired by parenchyma cells across the sapwood. Unexpected high values of PEPC

Abstract: International audience; Analysis of plant metabolite 13C-enrichments with gas-chromatography mass spectrometry (GC/MS) has gain interest over the last years, due to the ease of its implementation and the ability to combine information from multiple fragments of a same metabolite. However, measurement of carbon isotopologue distribution (CID) by GC-MS can be prone to analytical and matrix biases, that could be further enhanced with positional approaches (combination of multiples errors). Here, we took advantage of tailor-made 13C-Pascal Triangle standards, harbouring known CIDs and positional 13C-enrichments, to evaluate the reliability of GC-MS measurements and positional approaches for TMS-derivatives of organic and amino acids. Overall, we successfully validated the accuracy of the method, but some important biases were identified for proline, valine and threonine. Conversely, positional approaches were essentially validated for glycine and serine and questioned the wide application of such methodology without a preliminary validation step. Finally, we applied this method to explore the regulation of plant central carbon metabolism under dark and light conditions, by performing incorporation kinetics of U-13C-pyruvate and U-13C-glucose into Brassica napus leaf discs for up to 6 hours. The results showed interesting isotopic patterns related to the regulation of glycolysis, the tricarboxylic acid cycle and its phosphoenolpyruvate carboxylase anaplerotic pathway, and the photorespiratory-dependent biosynthesis of serine.

Abstract: International audience; A large part of the soil protist diversity is missed in metabarcoding studies based on 0.25 g of soil environmental DNA (eDNA) and universal primers due to ca. 80% co-amplification of non-target plants, animals and fungi. To overcome this problem, enrichment of the substrate used for eDNA extraction is an easily implemented option but its effect has not yet been tested. In this study, we evaluated the effect of a 150 μm mesh size filtration and sedimentation method to improve the recovery of protist eDNA, while reducing the co-extraction of plant, animal and fungal eDNA, using a set of contrasted forest and alpine soils from La Réunion, Japan, Spain and Switzerland. Total eukaryotic diversity was estimated by V4 18S rRNA metabarcoding and classical amplicon sequence variant calling. A 2- to 3- fold enrichment in shelled protists (Euglyphida, Arcellinida and Chrysophyceae) was observed at the sample level with the proposed method, with, at the same time, a 2-fold depletion of Fungi and a 3- fold depletion of Embryophyceae. Protist alpha diversity was slightly lower in filtered samples due to reduced coverage in Variosea and Sarcomonadea, but significant differences were observed in only one region. Beta diversity varied mostly between regions and habitats, which explained the same proportion of variance in bulk soil and filtered samples. The increased resolution in soil protist diversity estimates provided by the filtration-sedimentation method is a strong argument in favour of including it in the standard protocol for soil protist eDNA metabarcoding studies

Abstract: Chloroplasts are the powerhouse of the plant cell, yet they are resource-intensive and will cause photooxidative damage if their activity overshoots the demands of growth. The adjustment of chloroplast activity to match growth is therefore vital for stress acclimation. Here we identify a novel post-translational mechanism linking the conserved eukaryotic TOR kinase that promotes growth and the guanosine tetraphosphate (ppGpp) signaling pathway of prokaryotic origin that regulates chloroplast activity, and photosynthesis in particular. We show that RelA SpoT Homologue 3 (RSH3), a nuclear-encoded chloroplastic enzyme responsible for ppGpp biosynthesis, interacts directly with the TOR complex via a plant-specific N-terminal region (NTR) which is hyper-phosphorylated in a TOR-dependent manner. Downregulation of TOR activity reduces NTR phosphorylation, enhances ppGpp synthesis by RSH3, and causes a ppGpp-dependent decrease in photosynthetic capacity. Altogether we demonstrate that the TOR-RSH3 signaling axis is a novel and direct post-translational mechanism that allows chloroplast activity to be matched with plant growth, setting a new precedent for the regulation of organellar function by TOR.One sentence summaryThe TOR kinase post-translationally controls guanosine tetraphosphate signaling to regulate plant photosynthetic activity.

Abstract: International audience; Numerous studies report different types of responses following exposure of plants to high frequency electromagnetic fields (HF-EMF). While this phenomenon is related to tissue heating in animals, the situation is much less straightforward in plants where metabolic changes seem to occur without tissue temperature increase. We have set up an exposure system allowing reliable measurements of tissue heating (using a reflectometric probe and thermal imaging) after a long exposure (30 min) to an electromagnetic field of 2.45 GHz transmitted through a horn antenna (about 100 V m−1 at the plant level). We did not observe any heating of the tissues, but we detected rapid increases (60 min) in the accumulation of transcripts of stress-related genes (TCH1 and ZAT12 transcription factor) or involved in ROS metabolism (RBOHF and APX1). At the same time, the amounts of hydrogen peroxide and dehydroascorbic acid increased while glutathione (reduced and oxidized forms), ascorbic acid, and lipid peroxidation remained stable. Therefore, our results unambiguously show that molecular and biochemical responses occur rapidly (within 60min) in plants after exposure to an electromagnetic field, in absence of tissue heating.

Abstract: Grafting has been widely used in horticulture to induce dwarfing and avoid stress-derived limitations on plant growth and yield by affecting plant architecture and leaf functions. However, the respective effects on plant photosynthesis and water use efficiency (WUE) of leaf distribution and functions that depend on both rootstock and scion have not been fully elucidated. This study aimed to (i) clarify the scion × interstock impacts on the variability of leaf photosynthetic traits and WUE, and (ii) decipher the respective effects of leaf distribution and functions on canopy photosynthesis and WUE (WUEc). Leaf gas exchange over light gradients and responses to light, CO2, temperature, and vapor pressure deficit were measured in two apple cultivars, ‘Liquan Fuji’ (‘Fuji’) and ‘Regal Gala’ (‘Gala’), grafted onto rootstocks combined with interstocks: a vigorous (VV, ‘Qinguan’), or a dwarf one (VD, M26). The 3D architecture-based RATP model was parameterized to estimate the canopy photosynthesis rate (Ac), transpiration rate (Ec), and WUEc. Then, virtual scenarios were used to compare the relative contributions of cultivar and interstock to canopy Ac, Ec, and WUEc. These scenarios changed the leaf distribution and functions of either cultivar or interstock. At the leaf scale, VD trees had significantly higher leaf nitrogen per area but a lower maximum carboxylation rate and dark respiration in both cultivars. In parallel with higher leaf stomatal conductance (gs) and transpiration in VD ‘Fuji’ and similar gs in VD ‘Gala’, VD trees showed significantly lower leaf photosynthesis rate and WUE than VV trees. However, lower leaf photosynthetic capacities in VD trees were compensated at the canopy scale, with Ac and WUEc for ‘Fuji’ significantly improved in VD trees under both sunny and cloudy conditions, and for ‘Gala’ significantly improved in VD trees under cloudy conditions compared with VV trees. Switching scenarios highlighted that ‘Gala’ leaf functions and distribution and VD leaf distributions enhanced Ac and WUEc simultaneously, irrespective of weather conditions. Up-scaling leaf gas exchange to the canopy scale by utilizing 3D architecture-based modeling and reliable measurements of tree architecture and leaf functional traits provides insights to explore the influence of genetic materials and tree management practices.

Abstract: International audience; Cells maintain a constant dialog between the extracellular matrix and their plasma membrane to fine tune signal transduction processes. We found that the receptor kinase FERONIA (FER), which is a proposed cell wall sensor, modulates phosphatidylserine plasma membrane accumulation and nano-organization, a key regulator of Rho GTPase signaling in Arabidopsis. We demonstrate that FER is required for both Rho-of-Plant 6 (ROP6) nano-partitioning at the membrane and downstream production of reactive oxygen species upon hyperosmotic stimulus. Genetic and pharmacological rescue experiments indicate that phosphatidylserine is required for a subset of, but not all, FER functions. Furthermore, application of FER ligand shows that its signaling controls both phosphatidylserine membrane localization and nanodomains formation, which, in turn, tunes ROP6 signaling. Together, we propose that a cell wall–sensing pathway controls via the regulation of membrane phospholipid content, the nano-organization of the plasma membrane, which is an essential cell acclimation to environmental perturbations.

Abstract: International audience; Plant-herbivore interactions mediated by plant-plant signalling have been documented in different species but its within-species variability has hardly been quantified. Here, we tested if herbivore foraging activity on plants was influenced by a prior contact with a damaged plant and if the effect of such plant-plant signalling was variable across 113 natural genotypes of Arabidopsis thaliana. We filmed the activity of the generalist herbivore Cornu aspersum during 1 h on two plants differing only in a prior contact with a damaged plant or not. We recorded each snails' first choice, and measured its first duration on a plant, the proportion of time spent on both plants and leaf consumption. Overall, plant-plant signalling modified the foraging activity of herbivores in A. thaliana. On average, snails spent more time and consumed more of plants that experienced a prior contact with a damaged plant. However, the effects of plant-plant signalling on snail behaviour was variable: depending on genotype identity, plant-plant signalling made undamaged plants more repellant or attractive to snails. Genome-wide associations revealed that genes related to stress coping ability and jasmonate pathway were associated to this variation. Together, our findings highlight the adaptive significance of plant-plant signalling for plant-herbivore interactions.

Abstract: International audience; Introduction - Tree architecture is a fundamental part of a functional structural plant model. This is particularly evident in perennial tree crops where the structure of the plant influences the position of source and sink organs, as well as light interception inside the canopy, and serves as storage pool for exceeding carbohydrates. Therefore, a precise description of tree 3D structure is essential to realistically allocate carbohydrates in a FSPM (Costes et al., 2006). Hazelnut tree architecture has different characteristics compared to other fruit tree crops that have been modelled (i.e., peach, mango, apple, and kiwi). It is a monoecious species. Female flowers are grouped into inflorescences located into mixed buds, while male flowers are arranged into inflorescences in the apical position of sylleptic shoots. Each node could bear more than one bud and, the following year, the new shoots are much shorter than the shoots from which they were born. The present study aimed to analyse hazelnut one-year-old shoots 'architecture in order to build the structural part of L-HAZELNUT: a functional structural plant model of hazelnut (Corylus avellana). Material and Methods - Own-rooted tree clones of C.avellana, cultivar “Tonda di Giffoni”, were chosen, during winter, in 2020 and 2021 in Deruta (Italy). Plants were normally irrigated and fertilized. No pruning occurred during the two years. 104 one-year-old proleptic shoots were sampled according to four length categories: short (Sh) when shorter than 5 cm, medium (Me) when they were between 5 and 20 cm, long (Lo) when they were between 20 and 40 cm, and very long (VLo) when longer than 40 cm. For each shoot, diameter, length, and number of nodes were recorded. From the base of the shoot to its tip, the fate of each node was noted. Four fates are distinguished in C.avellana: blind nodes, vegetative buds, sylleptic shoots and mixed buds. In the year subsequent to sampling, the same measurements were repeated on the lateral shoots that were born from the buds. Statistical analyses were done in 2022, using packages from RStudio. First, exploratory analyses were performed to investigate the distribution of the bud types or sylleptic shoots depending on the rank node along the one-year-old proleptic shoots and thus, to address if there were some homogeneous zone (i.e., zones characterized by a stable distribution of the possible bud fates (Guédon et al., 2001)). This led us to draw a flux diagram (Figure 1). Each part of the diagram corresponds to specific logical connections, between FSPM components, which were addressed using specific probabilistic models, inferred from the collected data set. Results and Discussion - During this study new knowledge regarding the architecture of C.avellana was acquired. Even though male flowers were stated to be located on proleptic shoots 'nodes (Botta & Valentini, 2018), we instead highlighted the presence of male flowers in the apical position of sylleptic shoots. It was also observed that hazelnut has a sympodial growth. Each statistical model, associated with the FSPM components, was used to define functions to answer each of the questions addressed in the flow diagram. All the functions were coded in L-py (Boudon et al., 2012) to build a L-system program able to generate the architecture of hazelnut (Figure 1). Conclusion - This led to a first coarse FSPM named L-HAZELNUT, in reference to some previous FSPM for fruit tree crops (Lopez et al., 2010). It simulates the growth of a hazelnut tree over two successive years. The conception of an FSPM is a long and meticulous process that requires having in mind a clear scheme that describes the growing and branching processes. Such a scheme can be acquired by appropriate observations and analyses of how the plant grows and develops. However, as many aspects of the plant have to be considered, the accuracy of the presented 3D structure has still to be evaluated and further studies will be necessary to validate them. Moreover, L-HAZELNUT model should be complemented in the next future with a functional part describing carbon acquisition and partitioning within the tree.

Abstract: International audience; Aged plants are more difficult to infect than young plantlets. This modification of susceptibility is described as mature plant resistance (MPR). For potato virus Y (PVY), MPR is known to lead to low infection rates of plants inoculated at the postflowering stage and a decrease in the number of infected daughter tubers. However, the impact of inoculation date on the capacity of PVY to accumulate in daughter tubers has not been studied so far. Field and greenhouse experiments were carried out to better understand PVY epidemiology and to help potato growers to evaluate consequences of early/late infections on the quality of their crops. In field trials, potato plants (cv. Bintje) were covered by insectproof nets from planting to harvest except for a 14-day period to expose plants to natural PVY infections. Under controlled conditions, potato plants were mechanically inoculated with PVY at different dates from preflowering stages (early inoculations) to postflowering stage (late inoculations). At harvest, daughter tubers were individually collected and analysed to define proportions and viral load of infected tubers according to the time between virus inoculation and harvest. Our results showed that although the age of plants at the time of inoculation can modify their susceptibility to PVY infection, in return, early and late PVY inoculations lead to similar rates of infected tubers at the plant scale and equivalent viral accumulation in infected tubers. All together, these data revealed that both early/late infections are high risks for the sanitary quality of potato tubers.

Pressure chamber measurements of psi(leaf) (psi(leafPC)) are most typical, yet, the practical complexity of the technique and of the underlying theory has led to ambiguous understanding of the conditions to optimize measurements. Consequently, specific techniques and precautions diversified across the global research community, raising questions of reliability and repeatability. Here, we surveyed specific methods of psi(leafPC) from multiple laboratories, and synthesized experiments testing common assumptions and practices in psi(leafPC) for diverse species: (i) the need for equilibration of previously transpiring leaves; (ii) leaf storage before measurement; (iii) the equilibration of psi(leaf) for leaves on bagged branches of a range of dehydration; (iv) the equilibration of psi(leaf) across the lamina for bagged leaves, and the accuracy of measuring leaves with artificially 'elongated petioles'; (v) the need in psi(leaf) measurements for bagging leaves and high humidity within the chamber; (vi) the need to avoid liquid water on leaf surfaces; (vii) the use of 'pulse' pressurization versus gradual pressurization; and (viii) variation among experimenters in psi(leafPC) determination. Abstract: 1. Diatoms of the Arctic Ocean annually experience extreme changes of light environment linked to photoperiodic cycles and seasonal variations of the snow and sea-ice cover extent and thickness which attenuate light penetration in the water column. Arctic diatom communities exploit this complex seasonal dynamic through a well-documented species succession during spring, beginning in sea-ice and culminating in massive phytoplankton blooms underneath sea-ice and in the marginal ice zone. The pattern of diatom taxa sequentially dominating this succession is relatively well conserved interannually, and taxonomic shifts seem to align with habitat transitions. 2. To understand whether differential photoadaptation strategies among diatom taxa explain these recurring succession sequences, we coupled lab experiments with field work in Baffin Bay at 67.5°N. Based on field data, we selected five diatom species typical of different ecological niches and measured their growth rates under light intensity ranges representative of their natural habitats. To characterize their photoacclimative responses, we sampled pigments and total particulate carbon, and conducted 14C-uptake photosynthesis response curves and variable fluorescence measurements. 3. We documented a gradient in species respective light intensity for maximal growth suggesting divergent light response plasticity, which for the most part align with species sequential dominance. Other photophysiological parameters supported this ecophysiological framing, although contrasts were always clear only between succession endmembers, Nitzschia frigida and Chaetoceros neogracilis. To validate that these photoacclimative responses are representative of in situ dynamics, we compared them to the chlorophyll a-specific light-limited slope (α*) and saturated rate of photosynthesis (P*M), monitored in Baffin Bay on sea-ice and planktonic communities. This complementary approach confirmed that unusual responses in α* and P*M as a function of light history intensity are similar between sentinel sympagic species N. frigida and natural ice-core communities. While no light-history-dependent trends were observed in planktonic communities, their α* and P*M values were in the range of measurements from our monospecific cultures. 4. Synthesis. Our results suggest that Arctic diatoms species photoadaptation strategy is tuned to the light environment of the habitats in which they dominate and indeed drives the seasonal taxonomic succession.

Abstract: International audience; Karrikins, of which KAR1 and KAR2 are representatives, are a class of abiotic plant growth regulators produced from the partial combustion of plant material (Flematti et al., 2004; Flematti et al., 2009). Released into the soil following wildfires, karrikins can promote seed germination and seedling photomorphogenesis, and thereby facilitate the revegetation process (Nelson et al., 2009; Nelson et al., 2010; Stevens et al., 2007). The butenolide moiety of karrikins is also an essential structural feature of strigolactones (SLs), which are carotenoid-derived plant hormones that regulate a range of developmental processes in plants, and also serve as a rhizosphere signal for arbuscular mycorrhizal fungi and root parasitic weeds (Waters et al., 2016; Bürger and Chory, 2020). Collectively, these butenolides possess wide-ranging biological activities with considerable agricultural and environmental significance.

Abstract: Acoustic emission analysis is a promising technique to investigate the physiological events leading to drought-induced injuries and mortality. However, the nature and the source of the acoustic emissions are not fully understood and make the use of this technique difficult as a direct measure of the loss of xylem hydraulic conductance. In this study, acoustic emissions were recorded during severe dehydration in lavender plants and compared to the dynamics of embolism development and cell lysis. The timing and characteristics of acoustic signals from two independent recording systems were compared by principal component analysis. In parallel, changes in water potential, branch diameter, loss of hydraulic conductance and electrolyte leakage were measured to quantify drought-induced damages. Two distinct phases of acoustic emissions were observed during dehydration: the first one associated with a rapid loss of diameter and a significant increase in loss of xylem conductance (90%) and the second one with a significant increase in electrolyte leakage and slower diameter changes. This second phase corresponds to a complete loss of recovery capacity. The acoustic signals of both phases were discriminated by the third and fourth principal components. The loss of hydraulic conductance during the first acoustic phase suggests the hydraulic origin of these signals (i.e. cavitation events). For the second phase, the signals showed much higher variability between plants and acoustic systems suggesting that the sources of these signals may be plural, although likely including cellular damage. A simple algorithm was developed to discriminate hydraulic-related acoustic signals from other sources, allowing the reconstruction of dynamic hydraulic vulnerability curves. Abstract: International audience; Aquatic plants are crucial for the proper functioning of aquatic environments. So far, mixotrophy, widespread in aquatic microbial communities and in certain terrestrial plants, has been studied rarely in aquatic plants other than carnivorous ones. New evidence, particularly in Lemna and other close genera, suggests that mixotrophy is much more widespread than previously thought. Three processes could be involved, namely biotrophic associations, capture of prey (e.g., in carnivorous plants), and absorbotrophy (uptake of dissolved organic carbon). The latter process seems to be the most probable hypothesis in regard to the physiology of aquatic plants, but further research at both organism and cellular levels is needed to understand and clarify the processes involved and their importance. This opinion opens a new window of opportunity in aquatic ecology, with possible unsuspected applications in water management.

Abstract: KAI2 are plant α/β hydrolase receptors, which perceive smoke-derived butenolide signals (karrikins) and putative endogenous, yet unidentified phytohormones (KAI2-ligands, KLs). The number of functional KAI2 receptors varies among plant species. It has been suggested that KAI2 gene duplication and sub-functionalization plays an adaptative role for diverse environments or ligand diversification by altering the receptor responsiveness to specific KLs. Legumes represent one of the largest families of flowering plants and contain many essential agronomic crops. Prior to legume diversification, KAI2 underwent duplication, resulting in KAI2A and KAI2B. Integrating plant genetics, ligand perception and enzymatic assays, and protein crystallography, we demonstrate that Pisum sativum KAI2A and KAI2B act as receptors and enzymes with divergent ligand stereoselectivity. KAI2B has a stronger affinity than KAI2A towards the KAI2-ligand (-)-GR24 and remarkably hydrolyses a broader range of substrates including the strigolactone-like isomer (+)-GR24. We determine the crystal structures of PsKAI2B in apo and butenolide-bound states. The biochemical and structural analyses as well as recorded mass spectra of KAI2s reveal a transient intermediate on the catalytic serine and a stable adduct on the catalytic histidine, further illuminating the role of KAI2 not only as receptors but also as bona fide enzymes. Our work uncovers the stereoselectivity of ligand perception and catalysis by evolutionarily diverged KAI2 receptors in KAR/KL signaling pathways and proposes adaptive sensitivity to KAR/KL and strigolactone phytohormones by KAI2B.

Abstract: International audience; Phenomic selection is a promising alternative or complement to genomic selection in wheat breeding. Models combining spectra from different environments maximise the predictive ability of grain yield and heading date of wheat breeding lines. Phenomic selection (PS) is a recent breeding approach similar to genomic selection (GS) except that genotyping is replaced by near-infrared (NIR) spectroscopy. PS can potentially account for non-additive effects and has the major advantage of being low cost and high throughput. Factors influencing GS predictive abilities have been intensively studied, but little is known about PS. We tested and compared the abilities of PS and GS to predict grain yield and heading date from several datasets of bread wheat lines corresponding to the first or second years of trial evaluation from two breeding companies and one research institute in France. We evaluated several factors affecting PS predictive abilities including the possibility of combining spectra collected in different environments. A simple H-BLUP model predicted both traits with prediction ability from 0.26 to 0.62 and with an efficient computation time. Our results showed that the environments in which lines are grown had a crucial impact on predictive ability based on the spectra acquired and was specific to the trait considered. Models combining NIR spectra from different environments were the best PS models and were at least as accurate as GS in most of the datasets. Furthermore, a GH-BLUP model combining genotyping and NIR spectra was the best model of all (prediction ability from 0.31 to 0.73). We demonstrated also that as for GS, the size and the composition of the training set have a crucial impact on predictive ability. PS could therefore replace or complement GS for efficient wheat breeding programs.

Abstract: International audience; The growing world population and global increases in the standard of living both result in an increasing demand for food, feed and other plant- derived products. In the coming years, plant- based research will be among the major drivers ensuring food security and the expansion of the bio- based economy. Crop productivity is determined by several factors, including the available physical and agricultural resources, crop management, and the resource use efficiency, quality and intrinsic yield potential of the chosen crop. This review focuses on intrinsic yield potential, since understanding its determinants and their biological basis will allow to maximize the plant's potential in food and energy production. Yield potential is determined by a variety of complex traits that integrate strictly regulated processes and their underlying gene regulatory networks. Due to this inherent complexity, numerous potential targets have been identified that could be exploited to increase crop yield. These encompass diverse metabolic and physical processes at the cellular, organ and canopy level. We present an overview of some of the distinct biological processes considered to be crucial for yield determination that could further be exploited to improve future crop productivity.

Abstract: Flower development is crucial for the reproductive success of plants. This developmental step requires a deep genetic reprogramming, with the activation of flower-specific genes. In angiosperms, the LEAFY (LFY) Transcription Factor (TF) is a key regulator of flower development. Previous studies showed that LFY acts with cofactors that spatially specify its activity. The best-described LFY cofactor is the F-box protein UNUSUAL FLORAL ORGANS (UFO). However, how UFO regulates LFY’s activity at the molecular level remained unclear.I first showed that the role of UFO as a component of an E3 ligase complex was not essential for its functions in planta. Then, I found that LFY and UFO act together by forming a complex that recognizes specific cis-elements. The structural characterization of this complex revealed that UFO binds both LFY and a precise DNA sequence, explaining its role as a transcriptional cofactor. Finally, preliminary data suggest that the formation of the LFY-UFO complex is conserved within angiosperms and non-flowering plants. Altogether, this study presents an original case in plant where an F-box protein acts mostly as a transcriptional cofactor through its ability to bind DNA.A second part of my PhD was dedicated to the study of Arabidopsis LSH Oryza G1 (ALOG) genes, a plant-specific class of TFs regulated by LFY. Few data were available in the literature about their biochemical properties, and I focused on the interaction of the ALOG domain with DNA. I found that several ALOG proteins bind the same DNA motif and I characterized the DNA-binding mechanism.; Le développement des fleurs est crucial pour le succès reproductif des plantes. Cette étape du développement nécessite une reprogrammation génétique profonde, avec l'activation de gènes spécifiques à la fleur. Chez les angiospermes, le facteur de transcription (FT) LEAFY (LFY) est un régulateur clé de ce processus. Des études précédentes ont montré que LFY agit avec des cofacteurs qui spécifient spatialement son activité. Le cofacteur de LFY le mieux décrit est la protéine à F-box UNUSUAL FLORAL ORGANS (UFO). Cependant, comment UFO régule l'activité de LFY au niveau moléculaire restait incompris.J'ai d'abord montré que le rôle d'UFO en tant que composant d'un complexe E3 ligase n’était pas essentiel à sa fonction in planta. Puis j'ai montré que LFY et UFO agissent en formant un complexe qui reconnaît des éléments cis spécifiques. La caractérisation structurale de ce complexe a révélé qu’UFO se lie à la fois à LFY et à une séquence précise d'ADN, expliquant son rôle de cofacteur transcriptionnel. Enfin, des données préliminaires suggèrent que la formation du complexe LFY-UFO est conservée au sein des angiospermes et des plantes sans fleurs. Ainsi, cette étude présente un cas original chez les plantes où une protéine à F-box agit principalement comme cofacteur de transcription par sa capacité à lier l'ADN.Une deuxième partie de ma thèse a été consacrée à l'étude des gènes Arabidopsis LSH Oryza G1 (ALOG), une classe de FTs spécifique aux plantes et régulée par LFY. Peu de données étaient disponibles dans la littérature sur leurs propriétés biochimiques, et je me suis concentré sur l'interaction du domaine ALOG avec l'ADN. J'ai montré que plusieurs protéines ALOG lient toutes le même motif d'ADN et j’ai caractérisé le mécanisme de liaison à l’ADN.

Abstract: The complex and dynamic three-dimensional organization of chromatin within the nucleus makes understanding the control of gene expression challenging, but also opens up possible ways to epigenetically modulate gene expression. Because plants are sessile, they evolved sophisticated ways to rapidly modulate gene expression in response to environmental stress, that are thought to be coordinated by changes in chromatin conformation to mediate specific cellular and physiological responses. However, to what extent and how stress induces dynamic changes in chromatin reorganization remains poorly understood. Here, we comprehensively investigated genome-wide chromatin changes associated with transcriptional reprogramming response to heat stress in tomato. Our data show that heat stress induces rapid changes in chromatin architecture, leading to the transient formation of promoter-enhancer contacts, likely driving the expression of heat-stress responsive genes. Furthermore, we demonstrate that chromatin spatial reorganization requires HSFA1a, a transcription factor (TF) essential for heat stress tolerance in tomato. In light of our findings, we propose that TFs play a key role in controlling dynamic transcriptional responses through 3D reconfiguration of promoter-enhancer contacts.

Abstract: The regulation of intracellular pyrophosphate (PPi) level is crucial for proper morphogenesis across all taxonomic kingdoms. PPi is released as a byproduct from ~200 metabolic reactions, then hydrolyzed by either membrane-bound (H+-PPase) or soluble pyrophosphatases (PPases). In Arabidopsis, the loss of the vacuolar H+-PPase/FUGU5, a key enzyme in PPi homeostasis, results in delayed growth and a number of developmental defects, pointing to the importance of PPi homeostasis in plant morphogenesis. The Arabidopsis genome encodes several PPases in addition to FUGU5, such as PPsPase1/PECP2, VHP2;1 and VHP2;2, although their significance regarding PPi homeostasis remains elusive. Here, to assess their contribution, phenotypic analyses of cotyledon aspect ratio, palisade tissue cellular phenotypes, adaxial side pavement cell complexity, stomatal distribution, and etiolated seedling length were performed, provided that they were altered due to excess PPi in a fugu5 mutant background. Overall, our analyses revealed that the above five traits were unaffected in ppspase1/pecp2, vhp2;1 and vhp2;2 loss-of-function mutants, as well as in fugu5 mutant lines constitutively overexpressing PPsPase1/PECP2. Furthermore, metabolomics revealed that ppspase1/pecp2, vhp2;1 and vhp2;2 etiolated seedlings exhibited metabolic profiles comparable to the wild type. Together, these results indicate that the contribution of PPsPase1/PECP2, VHP2;1 and VHP2;2 to PPi levels is negligible in comparison to FUGU5 in the early stages of seedling development.

Abstract: Main conclusion Severe N stress allows an accumulation of C-based compounds but impedes that of N-based compounds required to lower the susceptibility of tomato stem toBotrytis cinerea. Abstract Botrytis cinerea, a necrotrophic filamentous fungus, forms potentially lethal lesions on the stems of infected plants. Contrasted levels of susceptibility to B. cinerea were obtained in a tomato cultivar grown on a range of nitrate concentration: low N supply resulted in high susceptibility while high N supply conferred a strong resistance. Metabolic deviations and physiological traits resulting from both infection and nitrogen limitation were investigated in the symptomless stem tissue surrounding the necrotic lesion. Prior to infection, nitrogen-deficient plants showed reduced levels of nitrogen-based compounds such as amino acids, proteins, and glutathione and elevated levels of carbon-based and defence compounds such as α-tomatine and chlorogenic acid. After B. cinerea inoculation, all plants displayed a few common responses, mainly alanine accumulation and galactinol depletion. The metabolome of resistant plants grown under high N supply showed no significant change after inoculation. On the contrary, the metabolome of susceptible plants grown under low N supply showed massive metabolic adjustments, including changes in central metabolism around glutamate and respiratory pathways, suggesting active resource mobilization and production of energy and reducing power. Redox and defence metabolisms were also stimulated by the infection in plants grown under low N supply; glutathione and chlorogenic acid accumulated, as well as metabolites with more controversial defensive roles, such as polyamines, GABA, branched-chain amino acids and phytosterols. Taken together, the results showed that nitrogen deficiency, although leading to an increase in secondary metabolites even before the pathogen attack, must have compromised the constitutive levels of defence proteins and delayed or attenuated the induced responses. The involvement of galactinol, alanine, cycloartenol and citramalate in the tomato stem response to B. cinerea is reported here for the first time.

Abstract: International audience; The hop plant (Humulus lupulus L.) has been exploited for a long time for both its brewing and medicinal uses, due in particular to its specific chemical composition. These last years, hop cultivation that was in decline has been experiencing a renewal for several reasons, such as a craze for strongly hopped aromatic beers. In this context, the present work aims at investigating the genetic and chemical diversity of fifty wild hops collected from different locations in Northern France. These wild hops were compared to ten commercial varieties and three heirloom varieties cultivated in the same sampled geographical area. Genetic analysis relying on genome fingerprinting using 11 microsatellite markers showed a high level of diversity. A total of 56 alleles were determined with an average of 10.9 alleles per locus and assessed a significant population structure (mean pairwise FST = 0.29). Phytochemical characterization of hops was based on volatile compound analysis by HS-SPME GC-MS, quantification of the main prenylated phenolic compounds by UHPLC-UV as well as untargeted metabolomics by UHPLC-HRMS and revealed a high level of chemical diversity among the assessed wild ac-cessions. In particular, analysis of volatile compounds revealed the presence of some minor but original com-pounds, such as aromadendrene, allo-aromadendrene, isoledene, fl-guaiene, alpha-ylangene and fl-pinene in some wild accessions; while analysis of phenolic compounds showed high content of fl-acids in these wild accessions, up to 2.37% of colupulone. Genetic diversity of wild hops previously observed was hence supported by their chemical diversity. Sample soil analysis was also performed to get a pedological classification of these different collection sites. Results of the multivariate statistical analysis suggest that wild hops constitute a huge pool of chemical and genetic diversity of this species.

Abstract: IntroductionDifferentiation of spore mother cells marks the somatic-to-reproductive transition in higher plants. Spore mother cells are critical for fitness because they differentiate into gametes, leading to fertilization and seed formation. The female spore mother cell is called the megaspore mother cell (MMC) and is specified in the ovule primordium. The number of MMCs varies by species and genetic background, but in most cases, only a single mature MMC enters meiosis to form the embryo sac. Multiple candidate MMC precursor cells have been identified in both rice and Arabidopsis, so variability in MMC number is likely due to conserved early morphogenetic events. In Arabidopsis, the restriction of a single MMC per ovule, or MMC singleness, is determined by ovule geometry. To look for potential conservation of MMC ontogeny and specification mechanisms, we undertook a morphogenetic description of ovule primordium growth at cellular resolution in the model crop maize.MethodsWe generated a collection of 48 three-dimensional (3D) ovule primordium images for five developmental stages, annotated for 11 cell types. Quantitative analysis of ovule and cell morphological descriptors allowed the reconstruction of a plausible developmental trajectory of the MMC and its neighbors.ResultsThe MMC is specified within a niche of enlarged, homogenous L2 cells, forming a pool of candidate archesporial (MMC progenitor) cells. A prevalent periclinal division of the uppermost central archesporial cell formed the apical MMC and the underlying cell, a presumptive stack cell. The MMC stopped dividing and expanded, acquiring an anisotropic, trapezoidal shape. By contrast, periclinal divisions continued in L2 neighbor cells, resulting in a single central MMC.DiscussionWe propose a model where anisotropic ovule growth in maize drives L2 divisions and MMC elongation, coupling ovule geometry with MMC fate.

Abstract: International audience; Blumeria graminis f.sp. tritici (Bgt) is an obligate biotrophic fungal pathogen responsible for powdery mildew in bread wheat (Triticum aestivum L.). Upon Bgt infection, the wheat plant activates basal defense mechanisms namely PAMP-triggered immunity (PTI) in the leaves during the first few days. Understanding this early stage of quantitative resistance is crucial for developing new breeding tools and evaluating plant resistance inducers for sustainable agricultural practices. In this sense, we used a combination of transcriptomic and metabolomic approaches to analyze the early steps of the interaction between Bgt and the moderately susceptible wheat cultivar Pakito. Bgt infection resulted in an increasing expression of genes encoding pathogenesis-related proteins (PR-proteins, PR1, PR4, PR5 and PR8), known to target the pathogen, during the first 48 hours post-inoculation. Moreover, RT-qPCR and metabolomic analyses pointed out the importance of the phenylpropanoid pathway in quantitative resistance against Bgt. Among metabolites linked to this pathway, hydroxycinnamic acid amides containing agmatine and putrescine as amine component accumulated from the second to the fourth day after inoculation. This suggests their involvement in quantitative resistance via cross-linking processes in cell wall for reinforcement, what is supported by the up-regulation of PAL (phenylalanine ammonia-lyase), PR15 (encoding an oxalate oxidase) and POX (peroxidase) after inoculation. Finally, pipecolic acid, which is considered as a signal involved in systemic acquired resistance (SAR), accumulated after inoculation. These new insights lead to a better understanding of basal defense in wheat leaves after Bgt infection.

Abstract: International audience; The Optimal Defense Theory (ODT) predicts that the distribution of defenses within a plant should mirror the value and vulnerability of each tissue. Although the ODT has received much experimental support, very few studies have examined defense allocation among reproductive tissues and none assessed simultaneously how these defenses evolve with age. We quantified glucosinolates in perianths, anthers and pistils at different bud maturity stages (i.e., intermediate flower buds, old flower buds and flowers) of undamaged and mechanically damaged plants of an annual brassicaceous species. The youngest leaf was used as a reference for vegetative organs, since it is predicted to be one of the most defended. In line with ODT predictions, reproductive tissues were more defended than vegetative tissues constitutively, and within the former, pistils and anthers more defended than perianths. No change in the overall defense level was found between bud maturity stages, but a significant temporal shift was observed between pistils and anthers. Contrary to ODT predictions, mechanical damage did not induce systemic defenses in leaves but only in pistils. Our results show that defense allocation in plant reproductive tissues occurs at fine spatial and temporal scales, extending the application framework of the ODT. They also demonstrate interactions between space and time in fine-scale defense allocation.
