Die vorliegende Arbeit beschreibt die Entwicklung und Charakterisierung eines mikromechanischen kraftgekoppelten Schwingsystems für die resonante Detektion niederfrequenter Schwingungen. Es wird ein neuartiges Prinzip vorgestellt, das es ermöglicht, niederfrequente Vibrationen frequenzselektiv zu erfassen. Mittels Amplitudenmodulation wird das niederfrequente Signal in einen höheren Frequenzbereich umgesetzt. Durch Ausnutzung der mechanischen Resonanzüberhöhung wird aus dem breitbandigen Signal ein schmales Band herausgefiltert, die anderen Frequenzbereiche werden unterdrückt. Auf diese Weise wird direkt die spektrale Information des niederfrequenten Signals gewonnen. Eine Fourier-Transformation ist hierbei nicht notwendig. Die Abstimmung des Sensors erfolgt über eine Wechselspannung und führt dadurch zu einer einfachen Auswertung. Die Schwerpunkte der Arbeit liegen in den theoretischen Untersuchungen zum neuartigen Sensorprinzip, in der Entwicklung einer mikromechanischen Sensorstruktur zum Einsatz des neuen Prinzips sowie in der Entwicklung und Charakterisierung eines Messsystems zur Detektion niederfrequenter mechanischer Schwingungen mit dem neuen Sensor. This thesis describes the development and characterization of a micromechanical force coupled oscillator system for the resonant detection of low frequency vibrations. It presents a novel working principle that enables spectral measurements of low frequency vibrations. The low frequency spectral content is converted into a higher frequency range by means of amplitude modulation. Due to the mechanical resonance a narrow band is filtered out of the wide band vibration signal. The remaining frequency content is suppressed. Hence, the spectral information is directly obtained with the sensor system without a fast Fourier transform. The tuning is done with an AC voltage resulting in a simple analysis. The main focuses of the work are the theoretical analysis of this novel sensor principle, the development of the micromechanical sensor structure for the use of the novel principle as well as the development and characterization of a measurement system for the spectral detection of low frequency mechanical vibrations with the developed sensor system.

Most high-efficiency solar cells are fabricated from monocrystalline Czochralski silicon (Cz-Si) wafers because the material quality is higher than multicrystalline silicon (mc-Si) wafers. However, the material study presented in this paper reveals strong variations in the material quality of commercially available Cz-Si wafers, leading to a loss in solar cell efficiency of 4% absolute. The reason for this is the presence of defects, which appear as dark rings in photoluminescence (PL) images of the finished solar cells. It is shown that these efficiency-limiting defects originate from oxygen precipitation during emitter diffusion. It is demonstrated that an incoming inspection in the as-cut state is difficult, as strong ring structures in as-cut wafers turn out to originate most often from thermal donors. These are dissolved during high-temperature treatments and are therefore harmless, whereas moderate ring structures in the as-cut state may become severe. That is why critical wafers can be identified and sorted out reliably only after emitter diffusion, by using QSSPC-based lifetime measurements or PL imaging. The two-year statistics gathered from the research line at Fraunhofer ISE on the occurrence of ring defects in Cz-Si wafers indicate that ring defects are highly relevant in terms of material yield.

The spatially resolved bulk lifetimes of silicon bricks can be obtained by spectral photoluminescence intensity ratio imaging. The underlying principle of this method, relevant experimental data and studies discussing its residual artefacts and limitations were published recently. To further discuss the accuracy and drawbacks of the method, the impact of lateral carrier diffusion into grain boundaries and the influence of free carrier absorption are analysed theoretically. A criterion is given by which the impact of minority carrier diffusion on the bulk lifetime image can be rated depending on both the distance to the grain boundary and the local grain bulk lifetime. We find that the impact of free carrier absorption on the calculation of quantitative bulk lifetime can be neglected for bricks commonly used for solar cell fabrication. However, free carrier absorption needs to be accounted for bricks with lifetimes in the millisecond range such as can be found in Czochralski grown material.

Broad range injection-dependent carrier lifetime measurements in crystalline silicon are most relevant for both defect level spectroscopy, and for the investigation of recombination properties of novel solar cell technologies. The approach presented in this paper combines a determination of the effective carrier lifetime via time-dependent (quasi-steady-state) photoluminescence with a steady-state photoluminescence lifetime scan over a broad range of excess carrier densities. Thereby, the power of a virtually artifact-free time-domain approach is combined with the high sensitivity of steady-state photoluminescence at extremely low injection conditions. Time-dependent fluctuations of dark photocurrent measurements are identified as the essential source of uncertainty under such low injection. A measurement design which eliminates this source of uncertainty is presented and tested. Lifetime measurements at excess carrier densities as low as 10 8 cm −3 are shown in detail.

We are studying the thermal stability of thick hydrogenated amorphous aluminum oxide (Al2O3) layers (20-50 nm) prepared by a high-throughput plasma-enhanced chemical-vapor-deposition (PECVD) technique for the electrical passivation of crystalline silicon surfaces. These passivation layers can be applied alone or covered by a capping layer like amorphous hydrogenated silicon nitride (SiNx) or amorphous hydrogenated silicon oxide (SiOx), also prepared by PECVD. After firing at 870 degrees C for approximately 3 s, the layers show blistering for Al2O3 of 30 nm or higher, independently from the capping layer. For thinner Al2O3, no blistering can be observed even using scanning electron microscope (SEM). Very long carrier lifetimes up to 900 mu s was obtained in passivated p-Si (1 Omega cm) wafer after annealing and firing, without observing a strong influence of the layer thickness and the capping layer. All the layer stacks, including the stacks with SiNx capping layer, show high negative charge densities in the layer (1-4 x 10(12) cm(-2)). Additionally, low interface defect densities (similar to 10(11) cm(-2) eV(-1)), which could be achieved with and without a hydrogenated capping layer, were measured even after firing. To explain these phenomena, hydrogen concentration depth profiles were measured by nuclear reaction analysis. These measurements have shown that, at the Al2O3-Si interface, hydrogen atomic concentration ranging 5-7% after annealing and 4% after firing are obtained independently from the capping hydrogen concentration. We conclude that PECVD Al2O3 layers of 20 nm or thicker can provide enough hydrogen to passivate the interface defects, even after a high te! mperature step. However, the layer thickness should be limited to 30 nm in order to avoid the blistering.

The passivation mechanisms and qualities of Al2O3, SiNx, SiO2 and a-Si:H( i ) on p - and n -type silicon are investigated by quasi-steady-state photoluminescence measurements. This technique allows effective lifetime measurements in an extremely large injection range between 1010 cm-3 and 1017 cm-3. The measurements are discussed focusing on injections below 1012 cm-3 in order to determine the most effective passivation layer for solar cells arranged for indoor applications. Fixed negative charges in the passivation layer cause field-effect passivation due to band bending leading to either accumulation or inversion at the passivation layer/silicon interface. Accumulation causes a stable passivation quality at low level injection. Inversion leads to effective lifetime losses similar to the losses in the space charge region. On p -type silicon the most effective surface passivation at low injections is provided by Al2O3 or a-Si:H( i ). The n -type silicon samples passivated with a-Si:H( i ) show the best effective lifetimes. SiNx and SiO2 show lifetimes one order of magnitude below a-Si:H( i ). Al2O3 on n -type is the most effective passivation at high injections around 1015 cm-3. Due to inversion losses at low level injections the passivation quality decreases more than two orders of magnitude for injections around 1010 cm-3.

An accurate quantitative description of the Auger recombination rate in silicon as a function of the dopant density and the carrier injection level is important to understand the physics of this fundamental mechanism and to predict the physical limits to the performance of silicon based devices. Technological progress has permitted a near suppression of competing recombination mechanisms, both in the bulk of the silicon crystal and at the surfaces. This, coupled with advanced characterization techniques, has led to an improved determination of the Auger recombination rate, which is lower than previously thought. In this contribution we present a systematic study of the injection-dependent carrier recombination for a broad range of dopant concentrations of high-purity $n$-type and $p$-type silicon wafers passivated with state-of-the-art dielectric layers of aluminum oxide or silicon nitride. Based on these measurements, we develop a general parametrization for intrinsic recombination in crystalline silicon at 300 K consistent with the theory of Coulomb-enhanced Auger and radiative recombination. Based on this improved description we are able to analyze physical aspects of the Auger recombination mechanism such as the Coulomb enhancement.

Quasi-steady-state photoluminescence is a versatile technique to determine carrier lifetime in silicon. A recent approach extracts carrier lifetime without a priori information about dopant concentration [Appl. Phys. Lett. 97, 092109 (2010)]. It utilizes the phase shift between a time-modulated optical irradiation and the radiative recombination of a sample, while requiring a minimum of two measurements. The present paper is aimed at a generalization thereof that requires only one measurement. It brings about a substantial experimental simplification, significantly improved accuracy and precision, and it opens up paths to access material properties other than lifetime.

Modulated quasi-steady-state photoluminescence is used in photovoltaics in order to measure the injection-dependent effective minority carrier lifetime of silicon wafers. In spite of the very advantageous properties of this measurement technique, its wide dissemination in photovoltaics has been hampered so far because of a relatively poor sensitivity limit in terms of carrier lifetime. A systematic analysis of the sensitivity-limiting mechanisms led to a substantial upgrade of sensitivity, tackling the range of effective lifetimes of 1s. This paper discusses the requirements to reach this level of sensitivity. Most prominently, the dependence of a silicon photodiode's response time with respect to the wavelength of the detected light is addressed. As an alternative, InGaAs photodiodes are implemented. The sensitivity of this method with respect to carrier lifetime is evaluated.

This paper presents a class of methods to determine net dopant concentration of silicon wafers by means of dynamic or quasi-steady-state photoluminescence. In contrast to resistivity measurements, this approach is independent of assumptions about dopant type and majority carrier mobility. The latter fact makes it particularly interesting for the determination of net dopant concentration in compensated silicon, where conventional mobility models involving only one dopant species have been reported to fail. Our approach also allows access to majority carrier mobility via combination of net dopant concentration as inferred from photoluminescence with a resistivity measurement.

In the last years photoluminescence (PL) imaging has become a standard characterization method for conventional silicon solar cells and wafers providing spatially resolved information for material characterization and process control. In this work, the method of the c oupled determination of d ark saturation c urrent and series r esistance (C-DCR) is evaluated on conventional silicon solar cells in terms of its accuracy, reliability and informative value based on a large number of conventional multicrystalline silicon solar cells. The statistical evaluation is based on a comparison of the series resistance mean values obtained from the C-DCR method with the global values obtained from the IV-characteristics within the standard cell testing. Furthermore, examples for its application are given and demonstrate that the minimum exposure time needed for the C-DCR method is short enough to allow an inline application of the method.

We studied the precipitation of chromium in multicrystalline silicon during the crystallization process and temperature treatments typical for solar cell processing. A model which was already successfully used for simulating heterogeneous precipitation of iron is transferred to chromium, allowing two-dimensional simulations of dissolved chromium and precipitate density. The observed accordance with spatially resolved measurements demonstrates the similarity of chromium to iron precipitation and the ability of our model to predict the dissolved chromium concentration in multicrystalline silicon. After the crystallization process, a high impact of chromium on the carrier lifetime of wafers originating from an ingot intentionally contaminated with 20 ppma chromium in the melt was observed. The concentration of dissolved chromium was significantly reduced by phosphorus diffusion gettering or oxidation at 815°C. Copyright © 2012 John Wiley & Sons, Ltd.

During the production of silicon solar cells crack detection systems can help to sort out damaged wafers and reduce wafer breakage before they enter the production line. In order to be cost effective, the crack detection system needs to minimize false detections as much as possible. False detections in crack detection systems occur when bad wafers are not detected or when good wafers are falsely detected as bad. The first error leads to an increase in cell breakage, the second error raises cell costs because non-damaged wafers are sorted out prior to cell processing. In this work a model has been developed to calculate the maximum allowable error rates of crack detection systems in order to achieve a cost per wafer benefit. Therefore a breakage rate dependent throughput calculation, based on manufacturing data, has been implemented. A sensitivity analysis shows that avoiding a high sorting out rate is crucial to favor the implementation of a crack detection system.

Epitaxial emitters deposited by atmospheric pressure CVD have been studied using different characterization methods such as spreading resistance profiling (SRP) or electrochemical capacitance voltage profiling (ECV). Comparing simulations with measured data, very low minority carrier lifetimes of tau(eff) = 20 mu s have been determined for FZ-material after a standard epitaxy process. LBIC measurements showed very low and inhomogeneous distributed diffusion lengths in the range of 100-250 mu m before process optimization. Therefore an emitter epitaxy process working at lower temperatures was developed [1] leading to diffusion lengths of 750 mu m. The effective minority carrier lifetimes and emitter saturation currents were determined using QSSPC measurements. For samples with an emitter, deposited at optimized temperature, a best value for the effective minority carrier lifetime of tau(eff) = 212 mu s was measured and an emitter saturation current could be determined to J(0e) = 46 fA/cm(2). In order to investigate the stress in the epitaxial layers mu Raman and mu PL measurements were performed showing no increased values at the interface. The doping density in boron doped emitters was determined with mu PL and in good agreement with measured ECV-profiles. The Shockley-Read-Hall lifetime in the emitter bulk showed no increased values at the interface between substrate and epitaxial layer.

Metallozymes are composed of a biomolecular scaffold harboring a metal complex. Such hybrid systems connect the “best” features of enzymatic (biomolecule) and homogenous (metal) catalysis. Chemogenetic optimization of metallozymes can lead to a wide range of applications in biotechnology and medicine. In this perspective, catalysis with a focus on broad reaction scopes, high activity and selectivity under mild reaction conditions as well as the development of artificial restriction enzymes for gene correction are the current topics of interest. The use of membrane proteins as proteinshell for metallozymes could broaden the range of applications due to their rigid and defined structure, structural integrity in e.g. organic solvents and at elevated temperatures as well as their easy modifiability. Still, researchers forgo the area of membrane proteins because the handling of the protein is highly challenging and requires lots of skillfulness. The constant presence of detergents mimicking the native protein environment is a drawback just as the expression and extraction in large scale with “good” quality. Crystal structures are rarely available and to set-up analytics is not easy. This thesis overcame the challenges of the development of a metallozyme based on a transmembrane ß-barrel protein (FhuA) of E. coli. FhuA, made of 22 ß-strands, was engineered and expression/extraction was optimized to make it a suitable scaffold. To gain catalytic activity, a ruthenium based cyclopentadienyl metal complex [Ru-Cp*] was coupled to the engineered FhuA. Analytical methods for FhuA metallozyme characterization were developed. In the first chapter, different FhuA variants were (re)engineered and molecular dynamic computer simulations were used to analyze the surrounding of the catalyst binding site. Following, for the developed variants, expression conditions in terms of media, temperature and time were optimized. Expression was scaled-up from 0.25 L flask expression to 10 L fermentation. Extraction was compared by protein solubilization with different detergents and harsh membrane dissolution with subsequent protein precipitation using organic solvents. The protein could be refolded by the polymer polyethylene-polyethylene glycol (PE-PEG). Regarding good yields and purity, protein solubilization with dodecylmaltoside followed by n-hexane-EtOH-SDS dissolution proved to be superior to other detergent and organic solvent based extraction methods, achieving up to 800 mg protein with a purity of 90% from one liter extracted culture broth. Stability investigations in the organic cosolvents tetrahydrofuran (THF), ethanol (EtOH), chloroform/methanol (C/M) and dioxane were performed in the second chapter. Highest stability of selected variants was obtained in up to 40 vol% THF, 25 vol% dioxane, 10 vol% EtOH or C/M. Basic pH values are preferable over acidic ones and the variant FhuA del1-159_C556 (having least mutations referring to the wildtype DNA sequence) showed higher temperature stability than FhuA del1-159_C556_tev, possessing more mutation (melting point 74°C vs. 55°C). The third chapter focused on the development and characterization of the metallozyme based on FhuA-[Ru-Cp*]. The coupling efficiency was verified by cysteine titration and analyzed by MALDI-TOF-MS. Structural integrity after coupling was confirmed by CD spectrophotometry. In summary, in this thesis, for the first time, a large, highly hydrophobic engineered membrane protein was used as a biomolecular scaffold for the development of a metallozyme. Advantages of using FhuA are its high purity and tremendous stability in organic cosolvents, elevated temperatures and basic pH values. After assembly of the protein and the metal catalyst, the system was characterized and provides fundamentals for the use of other membrane proteins in metallozyme applications. Additionally on a shortterm basis, the herein designed protein including the developed methods is used for the generation of a “platform catalytic system”.

We present a qualitative method for series resistance imaging that generates lateral current flow in solar cells without electrical contacting. The applicability of the method is demonstrated on a multicrystalline silicon solar cell, and the results are briefly discussed with respect to lifetime artifacts and injection dependence of the series resistance distribution. The method overcomes one of the major drawbacks of state-of-the-art series resistance imaging methods, which require electrical contacting for current injection or extraction. It allows for all-contactless measurements, reduces the cell breakage rate, and simplifies the measurement setup and sample handling.

Biodiesel is generally prepared from renewable biological sources such as vegetable oils by transesterification. Jatropha curcas seed oil is a promising feedstock for biodiesel production. During biodiesel production from Jatropha oil, many co-products such as glycerol, fatty acid dis-tillate and seed cake, among others, are obtained. The efficient use of these co-products would enhance the economic viability of the Jatropha based biofuel industry. However, the possible presence of phorbol esters (PEs) in these co-products restricts their efficient utilization. During biodiesel production, Jatropha oil is subjected to many treatments (stripping, degumming and esterification) wherein PEs present in the oil undergo partial or complete destruction depending on the treatment conditions. One of aims of this study was to develop and integrate methodolo-gies for using the PEs as a value added product instead of simply allowing them to be destroyed during biodiesel production. Potential uses of the phorbol ester enriched fraction (PEEF), ob-tained from Jatropha oil in agro-pharmaceutical applications were also investigated. The reason for choosing this group of compounds (PEs) was that they are highly bioactive both in vitro and in vivo, but they are currently considered to be merely toxic, unwanted biomaterial in the Jatro-pha biodiesel production chain. The recent increase in the cultivation of Jatropha cultivation means that there are potentially huge quantities of PEs that could be used for many purposes. This study revealed that a large proportion (85.7%) of PEs was localized in the endosperm portion of the Jatropha seed. Interestingly, the kernel coat contained PEs in high concentration. The endosperm portion of the kernel also contained antinutritional factors such as phytate (96.5%) and trypsin inhibitor (95.3%). The presence of high levels of antinutritional/toxic com-ponents in the kernel was presumed to be one of the factors that protect Jatropha seeds against predatory organisms during post harvest storage. Based on the presence or absence of PEs, a qualitative method was developed to differentiate between toxic/nontoxic Jatropha genotypes. In this method the methanol extract of seeds is passed through a solid phase extraction (SPE) column and the absorption (280 nm) of the result-ing eluate is measured. After screening Jatropha seeds collected from different parts of the world for toxic and non-toxic genotypes using the pre-established HPLC method for PEs, a cut off value of the absorbance was set up to differentiate toxic and nontoxic genotypes. Raw kernels whose SPE eluates had an absorbance ≥0.056 were considered as toxic and ≤0.032 as nontoxic. Corresponding absorbances for the SPE eluates of defatted kernel meal were ≥0.059 (toxic) and ≤0.043 (nontoxic). However, confirmation of the presence of PEs especially in Jatropha products for food applications should be carried out using the pre-established and validated HPLC method. The developed qualitative method could find its applications for screening the toxicity of products and co-products obtained from the Jatropha biodiesel industry. Conditions were optimized for the extraction of PEs as a phorbol ester enriched fraction (PEEF) from Jatropha oil using methanol as a solvent and a magnetic stirrer/Ultra-turrax as ex-traction tools. The extent of PE reduction in Jatropha oil was >99.4% using methanol as the sol-vent. The PEEF obtained (48.4 mg PEs/g) was 14 fold higher in PEs than in the original oil and this fraction was highly bioactive as determined by the most sensitive snail bioassay (LC100, 1 ppm) (see below). As the removal of PEs from oil took 60 min, which might be considered a long time in an industrial process, further conditions were optimized to extract maximum PEs in the shortest possible time with minimum solvent. The tools used for PE extraction (Ultra-turrax and magnetic stirrer) were effective with a treatment time of 2 and 5 min, resulting in 80 and 78% extraction of PEs, respectively. The biodiesel prepared from both the residual oils met European (EN 14214:2008) and American biodiesel standard (ASTM D6751-09) specifications. It was evident from the study that PEs could be easily extracted by either of the two methods with a high yield and the residual oil could be processed to produce high quality biodiesel. Also the residual oil with a lower PE content is expected neither to harm the environment nor the workers who had to handle it. The extracted PEEF was evaluated for its agricultural potential as a bio control agent. The PEEF had a high biological activity in aquatic bioassays using snails (Physa fontinalis), brine shrimp (Artemeia salina) and daphnia (Daphnia magna), when compared with microorganisms. The EC50 (48 h) of the PEEF was 0.33, 26.48 and 0.95 ppm PEs for snail, brine shrimp and daphnia respectively. High MIC (minimum inhibitory concentration) values (≥215 ppm) and EC50 values (≥58 ppm) were obtained for both the bacterial and fungal species. Among the bio-assays tested, the snail bioassay was the most sensitive, producing LC100 at 1 μg of PEs/ml. The snail bioassay could be used to monitor the presence of PEs in various Jatropha derived products, contaminated soil and other matrices in the ecosystem that might be involved in the production or use of Jatropha and its products. The study also demonstrated that the PEs exhibit molus-cicidal, antifungal and antibacterial activities. The shelf life of the PEEF was investigated. The PEEF was more susceptible to degradation when stored at room temperature (50% degradation after 132 days) than when stored at 4 °C or -80 °C (8% and 4% degradation respectively). Similarly, the PEEF lost biological activity (the snail bioassay) more rapidly at room temperature becoming ineffective after 260 days; while at 4 °C and -80 °C, only 27.5% and 32.5% activity was lost after 870 days. The degradation of PEs was due to auto-oxidation. Changes in fatty acid composition, increase in peroxide value and decrease in free radical scavenging activity of the PEEF reflected the auto-oxidation. Inclusion of antioxidants as additives (butylated hydroxyanisole (BHA), Baynox and α-tocopherol) pro-tected the PEs against degradation. The study demonstrated that the PEEF was susceptible to oxidation and addition of antioxidant stabilised the PEs during storage. In soil, PEs present in both the PEEF (2.6 mg/g soil mixture) (silica was used to adsorb PEs) and Jatropha seed cake (0.37 mg/g soil mixture) were completely degraded as the temperature and moisture content of the soil increased. PEs from silica-bound PEEF were completely de-graded after 19, 12, 12 days (at 13% moisture) and after 17, 9, 9 days (at 23% moisture) at room temperature (22 −23°C), 32 °C and 42 °C respectively. Similarly, at these temperatures, PEs from seed cake were degraded after 21, 17 and 17 days (at 13% moisture) and after 23, 17, and 15 days (at 23% moisture). The toxicity of PE-amended soil extracts when tested using the snail bioassay decreased with the decrease in PE concentration. The study demonstrated that PEs pre-sent in the PEEF or Jatropha seed cake are completely biodegradable in soil and the degraded products are innocuous. In preliminary studies, the PEEF exhibited potent insecticidal activity against Spodoptera frugiperda, which is a common pest in corn fields damaging maize crop across the tropi-cal/subtropical countries such as Mexico and Brazil. The PEEF produced contact toxicity with an LC50 of 0.83 mg/ml (w/v). The PEEF at higher concentration (0.25 mg/ml, w/v) also reduced food consumption, relative growth rate and food conversion efficiency (FCE) by 33%, 42% and 38% respectively. The study demonstrated that the PEEF has a potential to be used as a bio-control agent. Further in-depth field experiments on the effects of the PEEF on S. frugiperda will pave the way for its use under field conditions. The pharmaceutical potential of Jatropha PEs was also investigated. The PEs from Jatropha oil were purified. At least six purified PEs (designated as factors C1 to C6) were present in Jatropha oil. The identities of the purified PEs (factors C1 and C2) were confirmed by NMR. Whereas, factor C3 and factors (C4 + C5) were both obtained as mixtures. However, comparison of peak areas for phorbol 12-myristate 13-acetate (PMA) and Jatropha factor C1 in the HPLC method showed a difference in sensitivity of absorption at 280 nm of 41.3 fold. All the individual purified Jatropha PEs (factors C1, C2, C3mixture and (C4+C5)) and PEs-rich extract (factors C1 to (C4 + C5)) were biologically active when tested in the snail and brine shrimp bioassays. In ad-dition, all the Jatropha PEs produced platelet aggregation in vitro with an effective order of (based on ED50 (μM)): Jatropha factor C2 < factor C3mixture < factor C1 < factor (C4+C5). The PEs-rich extract (contains factor C1 to C6) was toxic to mice upon intra gastric administration, with an LD50 of 27.34 mg/kg body mass as PMA equivalent or 0.66 mg/kg body mass as factor C1 equivalent. The prominent histopathological symptoms were observed in lung and kidney. The Jatropha purified PEs-rich extract, purified PEs (factor C1, factor C2, factor C3mixture and factors (C4+C5)) and toxic Jatropha oil produced severe cellular alterations/disintegration of the epithelium and also increased the inflammatory response (interleukin-1α and prostaglandin E2 release) when applied topically to reconstituted human epithelium (RHE) and human corneal epithelium (HCE). In RHE, the nontoxic oil (equivalent to the volume used for toxic oil) pro-duced a lower cellular and inflammatory response than the toxic oil and the response increased with an increase in concentration of the PEs. In HCE, nontoxic oil (equivalent to the volume used for toxic oil) produced marked cellular alterations. The study demonstrated that the pres-ence of PEs in Jatropha oil increased the toxicity, both towards RHE and HCE. In addition, all the purified Jatropha PEs gave positive responses in the tumour promotion assay and negative responses in the tumour initiation assay in vitro (the assay was based on foci formation in Bhas 42 cells). In the tumour promotion assay, the order of transformed foci/well formation was: PEs-rich extract > factor (C4+C5) > factor C3mixture > factor C1 > factor C2. The tumour promotion ac-tivity was mediated by the hyper activation of protein kinase C (PKC). The aforementioned studies demonstrated that Jatropha PEs are toxic when administered orally or when applied topically to the skin or eye tissues. The data obtained should help in establishing safety measures for people working with Jatropha PEs. The potential of Jatropha PEs as a feedstock intermediate for the synthesis of Prostratin, a promising adjuvant in anti HIV therapy, was evaluated. The studies demonstrated that the Jatro-pha PEs could be synthesized sequentially by converting them first to crotophorbolone and then to prostratin. As analyzed by Nano-LC-ESI-MS/MSR, the prostratin synthesized from Jatropha PEs had similar mass and peak retention time to the reference prostratin (Sigma, St. Louis), The study showed that prostratin could be synthesized from Jatropha PEs. However, further optimi-zation studies are required to ascertain the synthesis reactions and yield of prostratin synthesized from Jatropha PEs. Some of the preliminary requirements for any successful bio-control agent are that it should have a high bioactivity on the target organism, a long shelf-life and a high biodegradability in soil. In addition, the bioactive phytochemical should be available in large quantities, it should be easily extractable and continuously available. The PEEF potentially satisfies these aforesaid re-quirements. The abundance and novelty of PEs present in Jatropha species could form a new ?stock? for the agro-pharmaceutical industries. Considering the projected oil yield of 26 million tons/annum by 2015 (GEXSI, 2008), huge amount of raw materials will be available for both biodiesel and pharmaceutical industries. PEs in the form of the PEEF could be used either as in-sect controlling agents in agricultural applications or as a ?stock? biomaterial for synthesizing prostratin in pharmaceutical applications. Biodiesel wird herkömmlich durch die Transesterifikation erneuerbarer biologischen Quellen, wie beispielsweise pflanzliche Öle, hergestellt. Das aus Samen von Jatropha Curcas gewonnene Öl ist eine vielversprechende alternative für die Biodieselproduktion. In der Herstellung des Biodiesels aus Jatrophaöl fallen viele brauchbare Nebenprodukte, unter anderem Glycerin, Fettsäuredestillate und Pressskuchen, an. Der sinnvolle Einsatz dieser Nebenprodukte könnte die Wirtschaftlichkeit der auf Jatropha Biodiesel basierenden Branche steigern. Allerdings verhindert das potentielle Vorkommen von Phorbolestern (PEs) in diesen Nebenprodukten deren effektiven Einsatz. In der Biodieselproduktion wird das Jatrophaöl verschiedensten Behandlungen (Ausgasen flüchtiger Stoffe, Degummierung und Veresterung) ausgesetzt, wobei die im Öl vorkommenden PEs teilweise bis vollständig zerstört werden. Dies variiert je nach den vorherrschenden Bedingungen während der Behandlung. Eins der Ziele dieser Untersuchung war es, Methoden zu entwickeln, Verwendungen für die PEs zu finden und diese in die Produktionskette zu integrieren, anstelle zuzulassen dass sie ungenutzt bleiben bzw. sogar als Abfallstoffe übrig bleiben. Potentielle Einsatzmöglichkeiten der Phorbolesterangereicherten Fraktion (PEAF), gewonnen aus Jatrophaöl von agro-pharmazeutischen Produkte, wurden ebenfalls untersucht. Das große Interesse an den PEs rührt daher, dass sie sowohl in vitro als auch in vivo als hoch bioreaktiv einzustufen sind. Jedoch werden sie momentan nur als toxisches und unerwünschtes Nebenprodukt der Biodieselproduktion gesehen. Der starke Anstieg der Jatropha Kultivierung und deren Anbau lässt auf eine potentiell große verfügbare Menge an PEs schließen, welche vielseitig eingesetzt werden könnten. Diese Untersuchung zeigte, dass ein großer Anteil (85,7 %) der PEs in dem Endosperm der Jatropha Samen vorkommen. Interessant ist außerdem, dass die Samenhaut auch eine beachtliche Menge an PEs enthält. Das Endosperm des Kerns beinhaltet zusätzlich große Anteile verschiedenster antinutritiver Substanzen wie Phytat (96,5 %) und Trypsin Inhibitoren (95,3 %). Es wird angenommen, dass das Vorkommen dieser antinutritiven, bzw. toxischen, Substanzen einer der Eigenschaften der Jatropha Kerne ist, welche sie gegen Fraßfeinde während der Lagerung schützt. Eine qualitative Methode um zwischen toxischen und nicht toxischen Jatropha Genotypen zu unterscheiden, wurde entwickelt basierend auf dem Vorkommen bzw. dem Fehlen von PEs. Hierbei durchläuft ein Methanolextrakt der Kerne eine Festphasenextraktionssäule (SPE). Die Absorption des Eluats wird dann bei 280 nm bestimmt. Zunächst wurden Jatrophakerne aus verschiedensten Teilen der Welt auf deren Toxizität anhand der bestehenden HPLC Methode für PEs geprüft. Dann wurde ein Grenzwert auf Basis derer Absorptionen definiert um nicht toxische von toxischen Arten zu unterscheiden. Unbehandelte Kerne deren SPE Eluate eine Absorption von ≥ 0,056 aufwiesen wurden als toxisch eingestuft, wohingegen Werte ≤;0,032 als nicht toxisch angesehen wurden. Entsprechend wurden Absorptionen von ≥0,059 (toxisch) und ≤ 0,043 (nicht toxisch) für entfettetes Kernmehl festgelegt. Trotzallem sollte das Vorkommen von PEs in Jatropha Produkten, insbesondere in Lebensmittelprodukten, durch die bereits etablierte und validierte HLC Methode festgestellt werden. Die im Rahmen dieser Untersuchung entwickelte qualitative Methode könnte der toxikologischen Überprüfung von Produkten sowie Nebenprodukten der Jatropha Biodiesel Produktion dienen. Die Bedingungen der Extraktion von PEs als PEAF aus Jatrophaöl mit Hilfe von Methanol als Lösungsmittel, einem Magnetrührer und einem Ultraturrax wurden optimiert. PEs des Jatrophaöls konnten um bis zu >99.4 % reduziert werden, wenn Methanol als Lösungsmittel eingesetzt wurde. Die PEAF beinhaltete (48,4 mg PEs/g) 14 mal mehr als die unbehandelten Öle. Außerdem war diese Fraktion äußerst bioreaktiv, wie Anhand eines empfindlichen Schnecken Biotests (LC100 - 1 ppm) festgestellt wurde (siehe unten). Das Entfernen der PEs aus dem Öl dauerte 60 min, eine für die industrielle Aufreinigung sehr lange Dauer. Daher wurden die Bedingungen, unter welchen man den höchsten Anteil an PEs in der geringsten Zeit extrahieren kann, optimiert. Die zur Extraktion verwendeten Geräte (Magnetrührer und Ultraturrax) funktionierten bei 2 und 5 min am effektivsten und konnten hier entsprechend 80 und 78 % der PEs entfernen. Der aus beiden resultierenden Ölen hergestellte Biodiesel entsprach sowohl den europäischen (EN 14214:2008) als auch den amerikanischen Biodiesel Standards (ASTM D6751-09). So konnte gezeigt werden, dass PEs einfach und effizient Anhand beider Methoden zu extrahieren waren und das behandelte Öl ohne weiteres zu qualitativ hochwertigem Biodiesel weiterverarbeitet werden kann. Zudem stellt das durch die Extraktion aufgereinigte Öl keinerlei Gefahr für die Umwelt und den Menschen dar. Die PEAF wurde auf den Einsatz als potentielles Spritzmittel biologischen Ursprungs getestet. Sie zeigten eine hohe biologische Aktivität in aquatischen Biotests bei Wasserschnecken (Physa fontinalis), Salzkrebse (Artemia salina) and Wasserflöhen (Daphnia magna), anders als bei Mikroorganismen. Die EC50 (48 h) der PEAF lagen bei 0,33 (Wasserschnecken), 26,5 (Salzkrebse) und 0,95 (Wasserflöhe) ppm PEs. Für Bakterien und Pilze wurden hohe minimale inhibitorische konzentrationen (MIC) (≥215 ppm) und EC50 (≥58 ppm) Werte ermittelt. Unter allen durchgeführten Biotests reagierten die Wasserschnecken am empfindlichsten mit einem LC100 Wert von 1 µg PEs/ml. Dieser Test könnte dem Nachweis der PEs in verschiedensten Jatropha Produkten, verunreinigten Böden und weiteren Matrizes verschiedenster Ökosysteme, welche dem Anbau von Jatropha und/oder der Verarbeitung deren Produkte dienen. Hier konnte nachgewiesen werden, dass die PEs molluskizide, bakterizide und fungizide Wirkungen aufweisen. Die Lagerungsbeständigkeit der PEAF wurde auch untersucht. Sie wurden über 132 Tage bei verschiedenen Temperaturen gelagert und wiesen bei vorherrschender Raumtemperatur mit 50 % die höchste Abbaurate auf. Bei 4 °C und -80 °C konnte jedoch nur ein Abbau von 8 % bzw. 4 % nachgewiesen werden. Dementsprechend sank auch die biologische Aktivität (anhand des Wasserschneckentest überprüft) der bei Raumtemperatur gelagerten am schnellsten. Ihnen konnte bereits nach 260 Tagen kein Effekt mehr nachgewiesen werden, wohingegen die anderen nach 870 Tagen nur 27,5 % bzw. 32,5 % ihrer Aktivität einbüßten. Dieser Effekt trat aufgrund der Autooxidation ein. Veränderungen der Fettsäurenmuster, ein Anstieg der Peroxid Werte und ein Abfall der freien Radikale in der PEAF spiegeln dies wider. Die Zugabe von Antioxidantien (Butylhydroxianisol (BHA), Baynox und α-Tocopherol) konnte die PEs gegen diesen Abbau schützen. Hiermit wurde gezeigt, dass die PEAF eine begrenzte Lagerungsfähigkeit besitzt, welche jedoch mit Hilfe von Antioxidantien verbessert werden kann. In Böden wurden sowohl die PEs in der PEAF (2,6 mg/g Boden) (Silikate wurden verwendet um die PEs zu absorbieren) als auch die PEs im Jatropha Presskuchen (0,37 mg/g Boden) wurden mit steigenden Temperaturen und Feuchtigkeitsgehalten im Boden komplett abgebaut. Die Silikatgebundenen PEs der PEAF wurden nach 19, 12 und 12 Tagen (bei 13 % Feuchtigkeit) und nach 17, 9 und 9 Tagen (bei 23 % Feuchtigkeit) entsprechend bei Raumtemperatur (22−23 °C), 32 °C und 42 °C komplett abgebaut. Bei denselben Temperaturen wurden die PEs im Presskuchen nach 21, 17 und 17 Tagen (bei 13% Feuchtigkeit) und 23,17 und 15 Tagen (bei 23% Feuchtigkeit) komplett abgebaut. Die Toxizität der mit PE belasteten Bodengemische (anhand des Wasserschneckentests bestimmt) wurde mit sinkenden PE Konzentrationen geringer. Die Untersuchung konnte aufzeigen, dass sowohl in Böden als auch in der PEAF vorkommende PEs komplett biologisch abbaubar und deren Abbauprodukte ungefährlich sind. In Vorversuchen zeigte die PEAF eine insektizide Wirkung gegen Spodoptera frugiperda, ein häufig auftretender Schädling von Maispflanzen der tropischen/subtropischen Länder wie Mexiko und Brasilien. Die PEAF fungierte als Kontaktgift mit einem LC50 Wert von 0,83 mg/ml (w/v) Bei hohen Konzentrationen (0,25 mg/ml (w/v) wurden die Futteraufnahme, die relative Wachstumsrate und die Futterverwertung um 33%, 42% und 38% gesenkt. Hiermit konnte gezeigt werden, dass die PEAF potentiell als Insektizid eingesetzt werden könnte. Feldversuche wären an dieser Stelle notwendig um herauszufinden welchen Effekt sie unter Bedingungen im Freiland auf S. frugiperda hat. Das pharmazeutische Potential der PEs wurde ebenfalls untersucht. Zunächst wurden die PEs aus Jatrophaöl aufgereinigt. Es konnten mindestens sechs verschiedene PEs (als C1 bis C6 ?bezeichnet) im Jatrophaöl nachgewiesen werden. Die Identität von C1 und C2 konnte durch Kernspinresonanzspektroskopie bestätigt werden. Der Vergleich der Flächen unter den durch die HPLC gewonnenen Kurven für "Phorbol 12-myristate 13-acetate (PMA)" und Jatropha Faktor C1 zeigte eine 41,3-fach höhere Absorption bei 280 nm. Jedem der aufgereinigten PE Faktoren (C1, C2, C3mixture und (C4+C5)) und einem PE Gemisch konnte eine biologische Aktivität in den Wasserschnecken- und Salzkrebstests nachgewiesen werden. Zusätzlich haben alle Jatropha PEs eine Blutplättchenaggregation verursacht. Diese Aggregation in vitro war am stärksten (basierend auf ED50 (µM)) für Jatropha Faktor C2 < Faktor C3mixture > Faktor C1 > Faktor (C4+C5). Das PE Gemisch zeigte bei intragastrischer Anwendung toxische Wirkungen mit einem LD50 Wert von 27,3 mg/kg Körpermasse als PMA Äquivalente oder 0,66 mg/kg Körpermasse als Faktor C1 Äquivalente. Die auffälligsten histopathologischen Symptome konnten in der Lunge und den Nieren beobachtete werden. Die aufgereinigten Jatropha PEs, das PE Gemisch und das toxische Jatrophaöl riefen starke Veränderungen der Zellstrukturen, bis hin zum Auflösen der Gewebestruktur, der Epithelzellen hervor. Ferner erhöhte es die inflammatorische Reaktion (Interleukin-1α und Prostaglandin E2 Ausschüttung) wenn es topisch an nachgebildetem menschlicher Haut (RHE) und nachgebildetem Hornhautgewebe (HCE) angewandt wurde. Im Falle des RHEs rief das nicht toxische Jatrophaöl eine geringere Zell-und inflammatorische Antwort als das toxische hervor, wenn sie in gleichen Volumina eingesetzt wurden. Die Reaktionen auf das toxische Öl wurden mit steigender PE Konzentration stetig stärker. Wurden wieder die gleichen Volumina für sowohl das toxische als auch das nicht toxische Öl eingesetzt, traten im Falle des nicht toxischen Öls im HCE Zellstrukturveränderungen auf. Diese Untersuchung zeigte, dass das Vorkommen von PEs in Jatrophaöl dessen Toxizität gegenüber RHE und HCE steigert. Dazu kommt, dass alle aufgereinigten Jatropha PEs positive Reaktionen auf das Tumorwachstum und negative Reaktionen auf die Tumorbildung in vitro zeigten (der Test basierte auf der Bildung von Entzündungsherden in Bhas 42 Zellen). In dem Tumorwachstumstest war die Reihenfolge von Entzündungsbildenden Substanzen wie folgt: PE-Gemisch > Faktor (C4+C5) > Faktor C3mixture > Factor C1 > Factor C2. Die Wachstumsförderung wurde durch die Hyperaktivierung der ProteinKinase C (PKC) reguliert. Die hier dargestellten Ergebnisse zeigen, dass Jatropha PEs toxische Wirkungen haben, sofern sie oral aufgenommen oder auf die Haut oder das aufgetragen werden. Die hieraus gewonnen Daten könnten in der Etablierung von Sicherheitshinweisen für den ungefährlichen Umgang mit Jatropha PEs dienen. Jatropha PEs wurden auch auf deren potentielle Tauglichkeit als Futterzusatzstoff zur Förderung der Prostratrinsynthese, ein vielversprechendes Adjuvanz in der Therapie gegen HIV, untersucht. Die Untersuchungen zeigten, dass Jatropha PEs schrittweise synthetisiert werden können. Zunächst werden sie zu "Crotophorbolone" umgewandelt und dann zu Prostratin. Wie die Nano-LC-ESI-MS/MSR Analyse ergab, besaß das aus Jatropha PEs hergestellte Prostratin eine ähnliche Masse und auch eine ähnliche Spitzenretentionszeit wie das Referenz-Prostratin (Sigma, St. Louis). Die Untersuchung ergab, dass Prostratin aus Jatropha PEs synthetisiert werden kann. Nichtsdestotrotz müssten an dieser Stelle Folgeuntersuchungen durchgeführt werden um diesen Prozess zu optimieren und Schlüsse auf den Ertrag an Prostratin aus Jatropha PEs ziehen zu können. Einige der Anforderungen eines erfolgreichen biologischen Insekten- oder Pflanzenschutzschutzmittels sind folgende: Es sollte eine hohe Wirksamkeit gegen den Zielorganismus aufweisen, eine lange Lagerungsbeständig besitzen und im Boden biologisch Abbaubar sein. Noch dazu sollte es in hohen Mengen verfügbar, einfach zu gewinnen und nachhaltig zur Verfügung stehen. Die PEAF kann diesen Anforderungen gerecht werden. Das Vorkommen und die Neuentdeckung der PEs in Jatropha Spezies könnte eine neue Quelle für die Pharmaindustrie werden. Wenn man den geschätzten Öl Ertrag von 26 Millionen Tonnen jährlich bis zum Jahr 2015 (GEXSI, 2008) betrachtet, könnten durchaus riesige Mengen an Ausgangsmaterial für sowohl die Biodieselproduktion als auch die Pharmaindustrie zur Verfügung stehen. PEs in Form der PEAF könnten entweder als Insektenschutzmittel dem Landwirtschaftlichen Sektor oder alsAusgangsmaterial für die Prostratinsynthese der Pharmaindustrie dienen.

The preparation of ultra-thin oxide layers on mono-crystalline silicon substrate surfaces with ozone dissolved in ultra pure water at ambient temperature was investigated as a low cost alternative to current wet-chemical cleaning and passivation processes in solar cell manufacturing. Surface photovoltage technique was applied as fast, nondestructive, and surface sensitive method, to provide detailed information about the influence of oxidation rate and substrate surface morphology on electronic properties of the oxidised silicon interfaces and subsequently prepared hydrogen terminated surfaces. Sequences of wet-chemical oxidation in ozone containing ultra pure water and subsequent oxide removal in diluted hydrofluoric acid solution could be utilised to prepare hydrophobic substrates, which are predominantly required as starting point for layer deposition and contact formation. On so prepared hydrogen-terminated substrates values of interface state densities D it,min ≈ 5 × 10 11 eV −1 cm −2 could be achieved, comparable to values obtained on the same substrates by the standard RCA process followed by HF dip.

Charakteristisch für Perlcellulose als Regenerat vom Typ Cellulose II sind sphärisch geformte, poröse Partikel mit einer hohen spezifischen Oberfläche und einer guten Bioverträglichkeit. Aufgrund ihrer Eigenschaften sind diese Cellulosemikropartikel besonders gut für medizinische Anwendungen geeignet. Im Mittelpunkt der Arbeit standen Herstellung, Charakterisierung und Modifizierung von Perlcellulosen mit Partikelgrößen von etwa 1 bis10 µm. Im Rahmen der Arbeit wurden zunächst sieben technische Cellulose-2,5-acetate mit vergleichbaren molekularen Eigenschaften auf ihre Eignung zur Herstellung von Perlcellulose nach dem in EP0750007 beschriebenen Acetatverfahren untersucht. Dabei erfolgte der Vergleich verschiedener Eigenschaften. Aus allen untersuchten Celluloseacetaten können Perlcellulosen synthetisiert werden. Als besonders geeignet erwies sich ein Produkt mit einer Molmasse von über 100.000, einem Verhältnis der Molmasse zur numerischen Molmasse von etwa 1,5 und einer guten Löslichkeit in Ethylacetat / Methanol (100:17,5). Die hergestellte Perlcellulose hat eine geringe Partikelgröße und eine relativ enge Größenverteilung. Damit erfüllt dieses Cellulose-2,5-acetat alle Anforderungen für die Synthese von Perlcellulose. Der entscheidende Verfahrensschritt zur Herstellung von Perlcellulose ist das Dispergieren der Emulsion mittels Inline-Ultraturrax. Die Partikelgrößenverteilung hängt im Wesentlichen von den Bedingungen während des Dispergierens ab. Im Rahmen der Arbeit gelang die reproduzierbare Herstellung von Cellulosemicrospheres mit einer Partikelgröße unter 5 µm. Für die Herstellung von Cellulosemikropartikeln mit definierten Eigenschaften ist neben den Synthesebedingungen auch die Charakterisierung der Perlcellulosen von entscheidender Bedeutung. Dafür wurden zunächst etablierte Verfahren verwendet (Partikelgrößenmessung, REM und Quecksilberporosimetrie). Parallel dazu erfolgte die Entwicklung bzw. Einführung neuer Methoden. Im Vordergrund stand die Untersuchung des Sedimentationsverhaltens der Perlcellulosen durch analytisches Zentrifugieren. Davon ausgehend konnte ein Verfahren zur Berechnung der Porosität aus dem Sedimentationsvolumen entwickelt werden. Zum Nachweis der kompletten Deacetylierung der Proben wurde die Ramanspektroskopie genutzt. Durch die Anwendung vorhandener und die Entwicklung neuer Methoden wird die genaue Einstellung von Eigenschaften der Perlcellulosen (z. B. Partikelgröße und deren Verteilung, Morphologie sowie Porosität) ermöglicht und deren Reproduzierbarkeit gewährleistet. Weitere Versuche hatten die Entwicklung von Endotoxinadsorbermaterial auf Basis von Perlcellulose und Polymyxin-B-sulfat (PMB) zum Ziel. Die Kopplung des PMB erfolgte meist nach Aktivierung der Proben mit Epichlorhydrin. Zunächst wurde die eingesetzte Epichlorhydrinmenge variiert, um das Optimum für die Aktivierung der Perlcellulosen zu finden. Weiterhin wurden unterschiedliche Mengen PMB angebunden und die Anbindung an nicht aktivierte Proben untersucht. Die Planung aller Versuche erfolgte jeweils nach Auswertung der an der Donau-Universität Krems durchgeführten Limulus- Amöbocyten- Lysat (LAL)-Tests. Mittels dieser Batchtests wurde die Wirksamkeit des Endotoxinadsorbermaterials sowohl im Vergleich zu unbehandeltem Blutplasma und als auch zu kommerziell erhältlichen Adsorbern auf Polystyrenbasis getestet. Endotoxinadsorber, die bei diesen Tests besonders gut bewertet wurden, konnten in einem up-scale- Versuch erstmals in größeren Mengen synthetisiert werden. Auch die direkte Herstellung von Endotoxinadsorbermaterial aus Perlcelluloseacetat konnte realisiert werden. Bei diesem neu entwickelten Verfahren erfolgen Deacetylierung und Aktivierung in einem Schritt. Damit kann die Herstellung vereinfacht werden. Zur Gewährleistung der Erstfehlersicherheit in extrakorporalen Blutreinigungssystemen sollen magnetisierte Perlcellulosepartikel als bioverträgliche Marker eingesetzt werden. Versuche zur Magnetisierung von Cellulosemikropartikeln während des Herstellungsprozesses zeigten, dass die Einbindung von Magnetit bei Erhalt der sphärischen Partikelstruktur prinzipiell auch auf diesem homogenen Syntheseweg möglich ist.:1. Because of their properties these cellulose microspheres are especially suited for medical applications. The focus of this work was the synthesis, characterization and modification of bead cellulose with particle sizes between 1 to 10 µm. In the frame of this work seven technical cellulose-2.5-acetates were investigated with regard to their suitability for making bead cellulose according to the process described in EP0750007. These cellulose acetates have comparable molecular characteristics. Different properties were compared. Bead celluloses can be synthesized from all investigated cellulose acetates. A product with a molecular weight of more than 100,000 and with a ratio between molecular weight and numeric molecular weight of about 1.5 is special suited. This cellulose-2.5-acetate has a good solubility in ethyl acetate / methanol (100:17.5). The bead cellulose made from it has a low particle size and a relative narrow size distribution. Thus this cellulose acetate complies with the requirements for making bead cellulose. The most important process step for making bead cellulose is the dispersing of the emulsion using an inline-ultraturrax. The distribution of particle size depends mainly on the conditions during dispersing. A reproducible synthesis of cellulose microspheres with a particle size range below 5 µm was successfully achieved. In addition to determining conditions for manufacturing bead cellulose the characterization of the microspheres is essential to obtain bead cellulose with well defined properties. At first well-established methods of characterization were used (particle size measurement, SEM and mercury porosimetry). In parallel new methods were developed and implemented. The main focus was the investigation of sedimentation behaviour of bead cellulose using analytical centrifugation. Based on this knowledge of the sedimentation volume a new method to calculate the porosity was designed. Raman spectroscopy was used for detecting the complete deacetylation of the samples. By using well-established and newly developed methods properties of bead cellulose such as particle size and distribution, morphology and porosity can be accurately adjusted. In this way the reproducible synthesis of cellulose microspheres can be ensured. The aim of further experiments was to develop an endotoxin adsorber material based on a coupling of bead cellulose with Polymyxin B sulfate (PMB). The coupling with PMB was carried out after activation of the samples by using epichlorohydrin. At first the added epichlorohydrin amount was diversified in order to find the optimum for the activation of bead cellulose. Later the coupling of different amounts of PMB took place and the linking of PMB to non activated samples was investigated too. The planning of all experiments occurred after evaluation of Limulus amebocyte lysate (LAL) tests at Danube-University Krems. Using these batch tests the effectiveness of the endotoxin adsorber material was tested compared to untreated blood plasma as well as commercial available adsorbers based on polystyrene. Endotoxin adsorbers showing the best adsorption rate were then synthesized for the first time in larger quantities. Also the direct synthesis of endotoxin adsorber material based on bead cellulose acetate could be realized. Using this newly developed method, deacetylation and activation occur during the same step. This means manufacturing process can be simplified. Using magnetized bead cellulose as biocompatible marker particles is planned to achieve first fault safety in case of a membrane rupture during extracorporeal blood purification. Initial tests have shown that the magnetization of cellulose microspheres is possible during the manufacturing process. Today’s raising demand for energy relies to a degree of 85% on the consumption of fossil fuels. A change to regenerative forms of energy is an important and inevitable step in order to face the challenges of climate change and fading natural resources. Photovoltaic’s (PV) plays a special role within the various forms of renewable energy since it converts sunlight, our most important and virtually endless energy source, directly into electricity. However, currently available PV-systems are still very expensive and, in combination with their relatively low performance, can hardly or cannot compete with conventional sources of energy from an economical point of view. One possibility to overcome this problem is the combination of highly efficient multi junction solar cells with cost-efficient concentrator optics that focus the incident sunlight to a small spot. The material system (GaIn)(NAs) is envisioned to play an important role in a future generation of multi junction solar cells for concentrator applications being a further development of existing device concepts. However, especially the carrier diffusion lengths in (GaIn)(NAs)-based solar cell layers are currently to low for the fabrication of highly efficient PV-structures. In this work, two novel techniques for the characterization of solar cells are developed and evaluated by experiments on test structures and numerical simulations. Both are based on the measurement of laser-induced currents. Spatially-resolved photocurrent spectroscopy (SRPS) allows a spatially-resolved determination of locally induced photocurrents at a fixed bias voltage while spatially-resolved IV-characteristics (SRIV) are measurements of local I-V-characteristics at a certain position. It is found that SRPS and SRIV allow for a reliable and meaningful characterization of solar cell prototypes with a high spatial resolution. Especially the local p-n-parameters of the sample become accessible. These are the short circuit current, the saturation current, the ideality factor and the excitation-induced shunt resistance. Subsequently, SRPS and SRIV served as characterization techniques for the evaluation of annealing experiments on processed (GaIn)(NAs) solar cell layers for concentrator applications. The goal of these experiments was to improve the material quality of such structures by treating them with high electrical currents and strong lasers pulses. Such enhanced devices could be used in a future generation of PV-systems leading to a significantly increased efficiency. Concerning this some first achievements were made. The use of high electrical currents led to a significant increase of the short circuit current, especially if applied as reverse bias. However, this also caused a reduction of the shunt resistance which eventually dropped down to a few ohms. The application of intensive laser radiation led under certain conditions to a considerable increase of the carrier lifetime and photoluminescence but no improvement of the electrical properties of the devices could be found. A second project dealt with the effect of carrier-depletion on the evaluation of data obtained by the variable stripe length method which is a technique for measuring the optical gain of potential laser materials. Numerical simulations and accompanying experiments on a GaAs-quantum well structure showed that this approach is only valid within a small parameter range. Outside this range an unavoidable carrier depletion caused by stimulated emission processes leads to significant errors in the obtained results. The determination of the corresponding borders is very complicated and requires a detailed knowledge of the sample properties. As the variable stripe length method is typically used to get a first insight into the properties of a novel sample such knowledge is generally missing. Therefore the reliability of experimental results obtained by this technique is doubtful., Der stetig steigende Weltenergieverbrauch wird derzeit zu 85% durch fossile Energieträger gedeckt. Im Hinblick auf den Klimawandel und schwindende Ressourcen ist hier ein Wandel hin zu regenerativen Formen der Energieerzeugung dringend geboten und auf lange Sicht alternativlos. Hier kommt speziell der Photovoltaik (PV) eine besondere Rolle zu, stellt sie doch den direktesten Weg dar um elektrischen Strom aus unserer wichtigsten Energiequelle, der Sonne, zu gewinnen. Aktuelle Generationen von PV-Anlagen besitzen aber nach wie vor eine zu geringe Leistungsfähigkeit bei zu hohen Kosten, um mit konventionellen Formen der Energieerzeugung wirtschaftlich konkurrieren zu können. Diesbezüglich bieten hocheffiziente Mehrschichtsolarzellen in Verbindung mit kostengünstigen Konzentratoroptiken zur Fokussierung einfallender Sonnenstrahlung einen möglichen Ausweg. Große Hoffnungen im Rahmen der Weiterentwicklung solcher Systeme ruhen auf dem Materialsystem (GaIn)(NAs), mit dem sich bereits bestehende Mehrschichtsolarzellenkonzepte nahezu ideal ergänzen ließen. Allerdings sind speziell die Ladungsträgerdiffusionslängen, welche sich derzeit in (GaIn)(NAs)-basierten Solarzellenschichten erzielen lassen, nicht hoch genug, um zufriedenstellende Wirkungsgrade erzielen zu können. Im Rahmen dieser Arbeit wurden zwei neuartige Verfahren zur Charakterisierung von Solarzellen, basierend auf der Messung laserinduzierter Photoströme, entwickelt und deren Zuverlässigkeit und Aussagekraft anhand eingehender Testmessungen sowie numerischer Simulationen untersucht. Hierbei handelt es sich um Methoden zur ortsaufgelösten Messung lokaler Photoströme bei fester Vorspannung (engl.: spatially-resolved photocurrent spectroscopy, SRPS) beziehungsweise zur Aufnahme lokaler Strom-Spannungs-Kennlinien an einer bestimmten Probenposition (engl.: spatially-resolved IV-characteristics, SRIV). Es zeigt sich, dass mit Hilfe von SRPS und SRIV eine zuverlässige und aussagekräftige Charakterisierung von Solarzellenprototypen möglich ist, die insbesondere eine Bestimmung der lokalen p-n-Parameter wie Kurzschlussstrom, Sättigungsstrom, Idealitätsfaktor und anregungsinduzierter Nebenwiderstand mit sehr hoher Ortsauflösung erlaubt. Aufbauend auf diesen Erkenntnissen dienten SRPS und SRIV als Bewertungsgrundlage in Versuchen zur Verbesserung prozessierter (GaIn)(NAs) Solarzellenschichten für Konzentratoranwendungen. Diese hatten zum Ziel durch den Einsatz starker elektrischer Ströme und intensiver Laserpulse die Materialqualität solcher Strukturen derart zu verbessern, dass deren Verwendung in einer zukünftigen Generation von PV-Systemen mit deutlich erhöhtem Wirkungsgrad möglich wird. Hier konnten erste Erfolge verbucht werden. So ließ sich zeigen, dass starke elektrische Ströme insbesondere in Sperrrichtung des p-n-Übergangs zu einer deutlichen Erhöhung des Kurzschlussstroms führten, wobei allerdings der Nebenwiderstand der untersuchten Probe bis auf wenige Ohm abfiel. Ferner führte intensive Laserbestrahlung unter bestimmten Voraussetzungen zu einer signifikanten Erhöhung der Ladungsträgerlebensdauer und Photolumineszenz, was sich allerdings nicht in einer Verbesserung der elektrischen Eigenschaften manifestierte. In Rahmen eines zweiten Projekts wurde der Einfluss einer möglichen Ladungsträgerverarmung auf die Auswertung von Strichlängenexperimenten, welche zur Bestimmung der optischen Verstärkung potentieller Lasermaterialien dienen, untersucht. Hier konnte anhand numerischer Simulationen und begleitender Messungen an einem GaAs-Quantenfilm klar gezeigt werden, dass die Methode der variablen Strichlänge in ihrer klassischen Form lediglich innerhalb eines sehr schmalen Parameterbereichs ihre Gültigkeit hat. Außerhalb dieser Grenzen, welche sich nur mit großem Aufwand und bei genauer Kenntnis der Probeneigenschaften bestimmen lassen, führt eine unvermeidliche Ladungsträgerverarmung infolge stimulierter Emissionsprozesse zu einer deutlichen Verfälschung der Auswertungsergebnisse. Da in aller Regel die erwähnten Kenntnisse im Vorfeld einer Messung naturgemäß nicht vorliegen, stellt dies die Zuverlässigkeit und Aussagekraft dieses Verfahrens generell in Frage.

Hintergrund und Ziele Herpesviren und andere Viren, die im Zellkern replizieren, greifen für den Export ihrer mRNAs auf zelluläre und/oder eigene Transportfaktoren zurück. Als solche herpesviralen RNA-Exportfaktoren konnten die Regulatorproteine der ICP27-Familie identifiziert werden, zu denen auch das UL69-Protein des humanen Cytomegalovirus (HCMV) gehört. Die Proteine dieser Familie besitzen als virale RNA-Exportfaktoren charakteristische Eigenschaften: sie wandern zwischen Zellkern und Zytoplasma (Shuttle-Aktivität), sie binden direkt an RNA und exportieren diese durch Interaktion mit zellulären mRNA-Exportfaktoren aus dem Nukleus. Für das UL69-Protein konnte durch in vitro Analysen gezeigt werden, daß die Interaktion mit dem zellulären mRNA-Exportfaktor UAP56 essenziell ist für den nukleären Export intronloser RNA. Obwohl für UL69 auch eine RNA-Bindungsaktivität detektiert wurde, zeigten RNA-bindungsdefiziente Mutanten keinen Verlust der RNA-Exportaktivität. Das Ziel dieser Arbeit bestand darin, rekombinante Cytomegaloviren zu konstruieren, die Mutationen in der UAP56- sowie der RNA-Bindungsdomäne des UAP56 aufwiesen. Die Charakterisierung dieser Viren sollte Aufschluss darüber geben, inwieweit die bislang durch in vitro-Analysen untersuchten Wechselwirkungen mit UAP56 und RNA auch für die virale Replikation von Bedeutung sind. Methoden Zur Konstruktion rekombinanter Cytomegaloviren wurde ein zweistufiges Verfahren verwendet, das auf homologer Rekombination BAC (bacterial artificial chromosome)-klonierter viraler DNA in E. coli beruht. In einem ersten Schritt wurde das UL69-Gen durch den Selektionsmarker Galaktosekinase ersetzt, der sowohl Positiv- als auch Negativ-Selektion ermöglicht. In einem zweiten Schritt erfolgte die Insertion mutierter UL69-Sequenzen. Zur Verifizierung der strukturellen Integrität der rekombinanten BACs wurden diese durch PCR, Restriktionsenzym-Verdau und Southern Blot-Analysen charakterisiert. Die Rekonstitution rekombinanter Cytomegaloviren erfolgte durch Transfektion von primären humanen Fibroblasten mit gereinigter BAC-DNA. Replikationskinetiken wurden sowohl durch Quantifizierung der viralen DNA durch Taqman-PCR als auch der infektiösen Viren im Zellkulturüberstand erstellt. Durch Western Blot-Analysen wurde die Proteinexpressionskinetik rekombinanter Viren charakterisiert. Ergebnisse Durch homologe Rekombination in E.coli konnten die Genome rekombinanter Cytomegaloviren mit Deletion bzw. Punktmutationen des Leserahmens UL69 konstruiert werden (UL69WT, UL69-galK, UL69mUAP, UL69∆R1∆RS, UL69∆R2∆RS, bzw. UL69Rev). Die Integrität der viralen Genome sowie die korrekte Insertion der gewünschten Mutationen wurde durch PCR, Restriktionsenzym- und Nukleotidsequenzanalysen sowie durch Southern Blot bestätigt. Nach Transfektion gereinigter BAC-DNA gelang es in allen Fällen rekombinante Viren zu rekonstituieren. Dies weist darauf hin, daß sowohl UL69 insgesamt als auch die mutierten Domänen (UAP56-Bindungsstelle, RNA-Bindedomäne) nicht essenziell sind für die virale Replikation. Wachstumskurven zeigten jedoch, dass die Mutanten AD169-UL69mUAP und AD169-UL69∆R1∆RS, welche eine punktmutierte bzw. deletierte UAP56-Bindestelle des UL69 enthielten, eine deutlich verzögerte Replikation im Vergleich zu Wildtypviren aufwiesen. Interessanterweise konnte auch für die Mutante AD169-UL69∆R2∆RS (Mutation der RNA-Bindungsdomäne des UL69) eine verzögerte Replikation beobachtet werden. Dies zeigt, daß die UL69 RNA-Bindung in vivo relevant ist für effiziente virale Vermehrung. Eine vergleichende Analyse der Freisetzung infektiöser Viruspartikel und viraler DNA im Zellkulturüberstand ergab eine gute Korrelation, so daß die Deletion bzw. Mutation von UL69 keinen Einfluß auf die Stabilität infektiöser Viruspartikel hat. Zur weiteren Charakterisierung der rekombinanten Viren wurde durch Western Blot-Analysen die Kinetik der viralen Proteinexpression untersucht. Dabei wurde deutlich, dass die Deletion von UL69 bzw. die Mutation der UAP56-Bindungsstelle insbesondere die Expression der späten viralen Proteine beeinflusst. Praktische Schlußfolgerungen Zusammenfassend gelang es mit dieser Arbeit mittels BAC-Rekombination humane Cytomegaloviren mit Mutation in der UAP56- und/oder RNA-Interaktionsdomäne des RNA-Exportfaktors pUL69 zu konstruieren. Diese Viren zeigten im Vergleich zu Wildtypviren ein verzögertes Replikationsverhalten, so daß beide Proteindomänen funktionelle Relevanz während der viralen Replikation aufweisen. Durch die Konstruktion dieser Viren konnte eine wichtige Voraussetzung geschaffen werden, um durch weitere detaillierte Charakterisierung zu klären, welche viralen Gene auf Ebene der Transkription oder auf mRNA-Exportebene durch pUL69 reguliert werden. Setting and targets Herpesviruses as well as other viruses that replicate within the nucleus have to make use of cellular and/or viral regulatory factors in order to facilitate the export of their mRNAs. The ICP27 protein family which includes the protein UL69 of human cytomegalovirus (HCMV), has been shown to encode herpesviral RNA export factors. All characterized members of this protein family exhibit characteristic features of RNA export factors: they shuttle between the nucleus and the cytoplasm, they directly interact with RNA and mediate its nuclear export via interaction with components of the cellular mRNA export machinery. In vitro analyses could demonstrate that a direct interaction of pUL69 with the cellular mRNA export factor UAP56 is essential for its stimulating effects on the nuclear export of intronless RNAs. Although RNA-binding could be detected for pUL69, an RNA binding deficient mutant did not lack its RNA export activity. Thus, the major goal of this work was to generate recombinant HCMVs carrying a mutated UAP56-binding domain and/or a deleted RNA-binding domain of UL69. The characterization of these recombinant viruses should clarify the role of UAP56 and RNA interaction of pUL69 for viral replication. Methods To construct rekombinant HCMVs a two-step selection procedure was applied that was based on homologous recombination of BAC (bacterial artificial chromosome)-cloned viral DNA in E. coli. In a first step, the UL69 gene was replaced by the selection marker galaktokinase which could be used for both positive and negative selection. In a second step the mutated/recombinant UL69 sequences were inserted. To verify the structural integrity of the recombinant BACs, PCR, endonuclease restriction digestion and Southern Blot analyses were performed. The reconstitution of recombinant HCMVs could be obtained by transfection of primary human fibroblasts with purified BAC DNA. Replication kinetics were analyzed by the quantification of viral DNA via Taqman-PCR as well as by titration of infectious virus in cell culture supernatants. Protein expression kinetics of recombinant HCMVs were characterized by Western Blot analyses. Results Recombinant HCMVs with deletions and point mutations of the open reading frame UL69 (UL69WT, UL69-galK, UL69mUAP, UL69∆R1∆RS, UL69∆R2∆RS, UL69Rev) were constructed via homologous recombination in E. coli. The integrity of the viral genome as well as the correct insertion of the intended mutations were verified by PCR, restriction enzyme digestion, nucleotide sequencing and Southern Blot analyses. After transfection of purified BAC-DNA recombinant HCMVs could be reconstituted for each UL69 BAC. This suggested that UL69 itself as well its mutated domains (UAP56-binding, RNA-binding domain) are not essential for viral replication. However, growth curve analyses demonstrated that the mutants AD169-UL69mUAP and AD169-UL69∆R1∆RS comprising a point-mutated or deleted UAP56-binding domain of UL69 showed a significantly reduced replication in contrast to wildtype virus. Surprisingly, a reduced replication could also be observed for the mutant AD169-UL69∆R2∆RS (mutated RNA-binding domain of UL69). This demonstrates that UL69 RNA-binding is a prerequisite for efficient viral replication in vivo. A comparative analysis revealed a good correlation of viral particle release and of viral DNA in supernatants of infected cells. This excludes a major effect of mutated forms of pUL69 on the stability of viral particles. Western blot analyses of protein expression after infection with genuine or mutated HCMVs revealed that a deletion of UL69 or a mutated UAP56-binding had a major effect on the expression of late viral proteins. Conclusion In summary, recombinant HCMVs harbouring mutations of the UAP56- and/or RNA interaction domains of the RNA export factor pUL69 could be constructed via BAC-recombination. Compared to wildtype HCMV, these recombinant viruses showed a reduced replication indicating a functional relevance of the respective protein domains for effective viral replication. The construction of these recombinant HCMVs is an important prerequisite to further characterize which viral genes are regulated by pUL69 on the level of transcription or mRNA export.

In the last few years, literary studies have experienced what we could call the rise of quantitative evidence. This had happened before of course, without producing lasting effects, but this time it’s probably going to be different, because this time we have digital databases, and automated data retrieval. As Michel’s and Lieberman’s recent article on "Culturomics" made clear, the width of the corpus and the speed of the search have increased beyond all expectations: today, we can replicate in a few minutes investigations that took a giant like Leo Spitzer months and years of work. When it comes to phenomena of language and style, we can do things that previous generations could only dream of. When it comes to language and style. But if you work on novels or plays, style is only part of the picture. What about plot – how can that be quantified? This paper is the beginning of an answer, and the beginning of the beginning is network theory. This is a theory that studies connections within large groups of objects: the objects can be just about anything – banks, neurons, film actors, research papers, friends... – and are usually called nodes or vertices; their connections are usually called edges; and the analysis of how vertices are linked by edges has revealed many unexpected features of large systems, the most famous one being the so-called "small-world" property, or "six degrees of separation": the uncanny rapidity with which one can reach any vertex in the network from any other vertex. The theory proper requires a level of mathematical intelligence which I unfortunately lack; and it typically uses vast quantities of data which will also be missing from my paper. But this is only the first in a series of studies we’re doing at the Stanford Literary Lab; and then, even at this early stage, a few things emerge.

L';amélioration des techniques de caractérisation (EBSD, MET, AFM) permet actuellement une meilleure compréhension des mécanismes plastiques intra-granulaires pour des poly-cristallin déformé. Les observations expérimentales montrent que les processus de plastification sont fortement hétérogènes et intermittent à l';intérieur des grains. Les modèles micromécaniques à champs moyens développés ces dernières décennies ne considèrent pas proprement les hétérogénéités intra-granulaires du glissement plastique. Or il est aujourd';hui démontré (simulations de Dynamique des Dislocations Discrètes par exemple) que la prise en compte de l';auto-organisation des dislocations à l';intérieur des grains est fondamentale pour mieux comprendre et expliquer les effets de taille de grains sur le comportement mécanique des polycristaux. Dans cette étude, deux approches complémentaires ont été développées : Une approche théorique qui consiste aux calculs des champs élastiques (contrainte interne et rotation de réseau) dus à une distribution discrète de boucles de dislocations contraintes par le joint de grains, et une approche expérimentale dont le but est de caractériser quantitativement les longueurs caractéristiques (espacements inter-bandes, et niveau de plasticification dans les bandes) pour des polycristaux à plusieurs tailles de grains se déformant plastiquement et de mesurer les rotations de réseau locales associées (mesure EBSD de désorientation de réseau cristallin) en vue de les comparer au champs de rotations élastiques calculés par le modèle. Le modèle a également été étendu pour permettre l';étude à des microstructures plus complexes, comme par exemple, les cellules de dislocations. The improvement of the materials characterization techniques in the last years has given access to new important information about the microstructure of polycrystalline metals. From experimental studies of deformed polycrystals, plastic strain within grains is known to be strongly heterogeneous and intermittent. As a consequence of the collective motion of dislocations, sample surfaces are indeed characterized by the presence of slip lines and slip bands (as slip traces). In the present study, a new micromechanical approach is developed to derive the mechanical fields (stresses, distortion, lattice curvature, elastic energy) arising from the presence of an inelastic strain field representing a typical internal "microstructure" as the one observed during the plastification of metallic polycrystals. This "microstructure" is due to the formation of discrete (spatial-temporal) intra-granular plastic slip heterogeneities which are modelled using discrete distributions of circular glide dislocation loops for a grain embedded in an infinite elastic matrix. Then, field equations have been solved using the method of Fourier Transforms. In contrast with the mean field approach based on the Eshelby formalism, it is then found that stress and lattice curvature fields are not more uniform inside the grain. A grain boundary layer actually appears where strong gradients occur and whose thickness depends on the introduced internal lengths. These results are compared with experimental measurements of local lattice rotation fields obtained by orientation imaging mapping (OIM). The model is able to capture different behaviours between near grain boundary regions and grain interior. The model was also develop to allow the study of more complex microstructures like the dislocation cells.

Micro-Raman (μRS) and micro-photoluminescence spectroscopy (μPLS) are demonstrated as valuable characterization techniques for fundamental research on silicon as well as for technological issues in the photovoltaic production. We measure the quantitative carrier recombination lifetime and the doping density with submicron resolution by μPLS and μRS. μPLS utilizes the carrier diffusion from a point excitation source and μRS the hole density-dependent Fano resonances of the first order Raman peak. This is demonstrated on micro defects in multicrystalline silicon. In comparison with the stress measurement by μRS, these measurements reveal the influence of stress on the recombination activity of metal precipitates. This can be attributed to the strong stress dependence of the carrier mobility (piezoresistance) of silicon. With the aim of evaluating technological process steps, Fano resonances in μRS measurements are analyzed for the determination of the doping density and the carrier lifetime in selective emitters, laser fired doping structures, and back surface fields, while μPLS can show the micron-sized damage induced by the respective processes.

Summary The impact on solar cell performance of transition metals like iron, chromium, nickel, titanium and co pper is the topic of this extended abstract. Each impurity has been intentionally added to silicon feedstock used to gr ow p-type directionally solidified multicrystalline silicon i ngots. A state of the art screen print solar cell process ha s been applied to wafers cut from the bottom to the top of these ingots. Adding 50 ppmwt of iron or 40 ppmwt of nickel or chromium to silicon feedstock, results in comparabl e solar cell performances to reference uncontaminated material in the range 40% to 70% of the ingot height. Addition of 10 ppmwt of titanium dramatically reduces the efficien cy along the entire ingot. Impurities like iron, chromium an d titanium cause a reduction in the diffusion length. Nickel d oes not reduce the diffusion length. On the other hand affe cts strongly the emitter recombination reducing the sol ar cell performance significantly. Copper has the peculiari ty to impact both bulk recombination as well as emitter recombination. A model based on Scheil distribution of impurity ha s been derived to fit the degradation along the ingot. So lar cell performance has been modeled as function of base bulk recombination and emitter recombination. The model fits very well the experimental data and has been also successfully validated. Unexpectedly, the Scheil di stribution of impurity along the ingot leaves its finger-print also at the end of the solar cell process. A measure of impurit y impact has been defined as the level of impurity which cau ses a degradation of less than 2% up to 90% of the ingot height. The advantage of this parameter is that comprises t he different impurities physical characters in one sin gle parameter, easy to compare.

Minority carrier lifetime is the most crucial material parameter for the performance of a silicon solar cell. While numerous methods exist to determine carrier lifetime on solar cell precursors prior to metallization, only very few techniques are capable of implicitly extracting effective minority carrier lifetimes from metallized silicon samples. In this paper, a measurement technique for effective minority carrier lifetime on silicon solar cells and metallized cell precursors via quasi-steady-state photoluminescence is presented. The setup requirements for this measurement technique are elaborated, experimental evidence of the reliability of such measurements down to carrier lifetimes in the range of a microsecond is provided, and a lifetime calibration of spatially resolved photoluminescence images of solar cells via the presented measurement technique is sketched. Finally, the very good agreement between the obtained effective carrier lifetime and the corresponding open circuit voltage of a solar cell is demonstrated.

We studied the behavior of iron in multicrystalline silicon during phosphorus diffusion by spatially resolved measurements and physical modeling. We present improvements to the previously used models for internal gettering in multicrystalline silicon and phosphorus diffusion gettering. 2-dimensional simulations are used for optimization of the phosphorus diffusion processes for intentionally contaminated wafers regarding the iron distribution, without changing the emitter characteristics. Simulations and experimental results show a reduced interstitial iron concentration after an additional low temperature step at the end of the phosphorus diffusion. The concentration of iron precipitates was reduced by a short annealing at 900 degrees C before the phosphorus diffusion, leading to a carrier lifetime three times higher than compared to the standard process.

Surface-passivated and surface-unpassivated aluminum-alloyed p +-layers are characterized. By varying the firing conditions and the thickness of the screen-printed aluminum paste, different sheet resistances Rsh of the p+-layer were fabricated. The emitter saturation current density J0eplotted versus Rsh follows distinctly different trends for the passivated and unpassivated samples. An aluminum paste with a boron additive achieves a much higher doping concentration and a lower sheet resistance but nevertheless follows the same J 0ecurves as the pure Al paste. The aluminum p+-layer was quantitatively analyzed with microphotoluminescence and FanoRaman measurements. The latter shows an increased defect recombination at the interface between the p+-layer and the moderately doped Si bulk. The lower ShockleyReadHall lifetime in this region can be attributed to a high defect concentration in the most highly doped layer, represents an impediment to the reduction of J 0efor Al- doped emitter regions, and needs to be optimized in future investigations.

Valuation of photovoltaic devices depends strongly on the measured power output of the device. This quantity is usually determined under artificial sunlight in production line measurement systems or industrial or research test labs. A practical calibration chain is realized essentially with measurements at solar simulators. The measurement conditions are defined in the IEC 60904 series of standards. An important part of the standard testing conditions is the definition of a specific spectral distribution of the sunlight (AM1.5 global). The inevitable deviations of the spectrum of artificial light sources from the standard spectrum have to be taken into account by a spectral mismatch factor. The uncertainty of this crucial correction is spectrally dependent, in most cases unknown and complex and inconvenient to evaluate. In this article a randomizing method is proposed which allows one to calculate the uncertainty of the mismatch factor from the uncertainties of the inpu t parameters determined with high spectral resolution. Based on a range of different spectral responses of solar cells on the one hand and variations of the solar simulator spectral distribution on the other, we are able to generalize the results to a broad set of measurement configurations. A sensitivity analysis reveals the crucial wavelength regions and thus allows the systematic optimization of simulator spectra and selection of reference cells.

Ziel ist die Entwicklung eines alternativen Elektrolytmaterials (YSZ/ScSZ-Komposits) für SOFCs im abgesenkten Temperaturbereich unter Berücksichti-gung der Perkolationstheorie. Der neue Komposit-Elektrolyt soll den Vorteil des reinen ScSZ-Materials, eine hohe ionische Leitfähigkeit im Temperaturbereich zwischen 600-800 °C, gepaart mit den Kostenvorteilen des Standardmaterials YSZ besitzen.

Only recently, methods for quality control of multicrystalline silicon wafers have been published, which allow the efficiency of solar cells to be predicted precisely from photoluminescence (PL) images taken in the as-cut state. In this letter it is shown that oxygen precipitates, present in standard Czochralski silicon wafers, can cause efficiency losses of more than 4% (absolute) within an industrial solar cell proc- ess. These efficiency losses correlate with ring-like defect structures of reduced intensity in the PL image. In comparison with QSSPC-based lifetime measurements, we introduce a PL-based method of quality control which allows the critical wafers to be identified and sorted out reliably at an early state of production and thus increases yield and average efficiency of production lines.

Photoluminescence Imaging has been proven to be very useful for the characterization of silicon material characterization. By detecting emitted photons from the band-to-band recombination with a CCD camera information about the excess carrier density is gained. Besides carrier lifetime spatially resolved concentration measurements of the most prominent metallic impurities, namely interstitial iron and chromium, can be obtained in boron doped silicon. These measurements are based on a careful preparation of different metastable chemical states of the species under test by appropriate temperature and illumination conditions. Interstitial chromium as well as iron may reversibly bind to boron which changes their electrical defect parameters and thus the injection dependent carrier lifetime. From PL-images in both states the specific quantitative point defect concentration can be extracted. An analysis of the depth dependent carrier profiles gives an estimation on systematic errors, ideal measurement conditions, and a correction procedure for non-ideal measurement conditions. This contribution summarizes the state-of-the-art of impurity imaging methods with PL and introduces new aspects on quantitative PL-analysis of silicon material. A possible impact of lateral carrier diffusion on measurement errors is discussed by simulations of the carrier density distribution around extended defects. We present spatially resolved PL-measurements of the BO-defect concentration which expand the analysis of specific impurities. Being the most prominent factor which limits the carrier lifetime in Czochralski silicon this defect proves to significantly reduce material quality in multicrystalline silicon, too. For the separation of the impact of different metastable species present in the same sample a sequence of conditioning steps and measurements is proposed. Combining all techniques on the detection of metastable defects the quantitative impact of the specific detected impurity concentrations on the electrical material quality is deduced and can be tracked during solar cell processing resulting in a comprehensive analysis of efficiency-limiting defects in silicon.

Luminescence imaging is a very rapid technique for the characterisation of silicon samples. Megapixel luminescence images on silicon bricks, unprocessed wafers, partially processed wafers and fully processed cells can be captured with acquisition times of typically a few seconds or less than one second. A number of specific luminescence imaging applications have been developed over the last three years, allowing quantitative spatial information to be gained about a variety of crucial material and device parameters. This paper reviews some of the intriguing possibilities for inline monitoring in PV production at an early stage of production that result from the above mentioned short measurement times.

Consolidated tables showing an extensive listing of the highest independently confirmed efficiencies for solar cells and modules are presented. Guidelines for inclusion of results into these tables are outlined and new entries since June 2010 are reviewed.

Minority carrier recombination is studied in multicrystalline ingot cast silicon solar cells. The normalized recombination strength Gamma of dislocations is obtained by correlating topograms of the internal quantum efficiency (IQE) with those of the dislocation density rho. Gamma is obtained by fitting an extended theory of Donolato to the experimental data. The measured Gamma-values vary significantly between adjacent dislocation clusters and correlate with the spatial pattern of the dislocations. All Gamma-values are strongly dependent on the parameters of the solar cell process. The influence of phosphorus diffusion and hydrogenation is shown. After solidification of the silicon, impurities from the crucible enter the ingot and deteriorate its border regions during cooling to room temperature. These deteriorated border regions can be significantly improved by an additional low temperature anneal that is applied after phosphorus diffusion. The experiments indicate that the mechanism of the anneal is external phosphorus gettering into the emitter.

The evolution during silicon solar cell processing of performance-limiting iron impurities is investigated with synchrotron-based x-ray fluorescence microscopy. We find that during industrial phosphorus diffusion, bulk precipitate dissolution is incomplete in wafers with high metal content, specifically ingot border material. Postdiffusion low-temperature annealing is not found to alter appreciably the size or spatial distribution of FeSi2 precipitates, although cell efficiency improves due to a decrease in iron interstitial concentration. Gettering simulations successfully model experiment results and suggest the efficacy of high- and low-temperature processing to reduce both precipitated and interstitial iron concentrations, respectively.

Comparison of the open-circuit voltage (external voltage V(oc,ext)) determined by Suns-V(oc) measurements with the implied voltage V(oc, impl) determined by transient photoconductance decay lifetime measurements can yield a quick and easy analysis of silicon heterojunction (SHJ) solar cells, especially in regard to finding the optimum doping concentration of the emitter layer [or back surface field (BSF)]. A sufficiently high doping concentration of the emitter and BSF is mandatory to extract the internal Fermi-level splitting and thus the internal voltage, at the solar cell contacts. However increasing the concentration of doping gases during the deposition of doped amorphous silicon layers results in a reduction of the interface passivation quality and Voc, impl. The best trade off is realized when the ratio of V(oc,ext) to V(oc,impl) (external/internal V(oc)-ratio zeta) reaches a saturation value near 1 upon increasing the doping concentration. AFORS-HET (Automat FOR Simulation of HETerostructures) simulations resulted in the conclusion that the characteristics of the external/internal Voc-ratio are mainly determined by the active doping concentration (doping minus defect concentration).

The purpose of this paper is a detailed comparison of selected luminescence and lock-in thermography (LIT) results on one exemplary sample and the drawing of corresponding conclusions. Our focus is on solar cells, but some investigations on wafers will be discussed as well. The comparison will help to decide which characterization tools are needed to solve technological problems. It will be demonstrated that luminescence imaging may widely replace LIT with respect to the analysis of recombination-active bulk defects, cracks, series resistance, and junction breakdown sites. However, some important investigations can be done only by LIT. LIT allows for a quantitative analysis of different kinds of leakage currents both under forward and under reverse bias, enabling a reliable analysis of local I-V characteristics. It is shown that LIT and luminescence imaging are complementary to each other and should be used in combination.

Mittels chemischer Bildgebung auf Basis spektralanalytischer Verfahren können biomolekulare Wechselwirkungen schneller, preiswerter, genauer und zuverlässiger als bisher nachgewiesen werden. Im folgenden Beitrag illustrieren wir dies mit Schwerpunkt auf der optischen Spektroskopie.

Optical Fluorescence Tomography (OFT) of live small animals can yield optimum 3-dimensional imaging performance when large amounts of tomographic boundary information are used for reconstruction. Commonly, multiple source-detector projection measurements distributed over the tissue surface of the imaging object are used to generate raw data for tomography. Recent advances in multispectral optical tomography, however, provide an attractive alternative method to harness tomographic boundary data based on excitation-resolved fluorescence measurements. The aim of this thesis is to report on the development of a novel versatile non-contact Optical Fluorescence Tomography system, which is capable of both multi-projection and multispectral excitation-resolved fluorescence measurements in order to collect large tomographic boundary data sets of small animals in vivo. The scanning concept of the OFT system capitalizes on a rotatable complete-angle projection mechanism that is combined with an independent source-detector positioning scheme using a CCD detector for photon collection and a continuous single beam illumination to excite fluorescence in tissue. This method allows the imaging object to be scanned in free-space over 360° around its longitudinal axis supporting a large number of spatial projections, while the animal remains calm in a prone position. The use of an incoherent white light source in conjunction with exchangeable optical filters for excitation light generation provides the basis for excitation-resolved fluorescence measurements. For imaging and experimental verification of the imaging system, tissue-simulating phantoms were used that were made from epoxy resin using black ink and TiO2 particles to adjust the optical properties. As contrast agent a NIR fluorophore (DY-752, Dyomics GmbH, Germany) was used. In order to investigate the potential of multispectral fluorescence tomography as supporting modality for multi-projection-based tomography in small animal imaging, simulations were carried out on an inhomogeneous digital mouse model that contained various anatomically correct mapped organs assigned with tissue-like optical properties as a function of wavelength. Excitation-resolved simulations were performed for nine excitation wavelengths (580-660 nm / 10 nm steps) using up to four perspective views. A rescaling method was applied to reduce the influence of the tissue-heterogeneities for reconstruction. Based on preliminary studies of the basic imaging equipment, an automated, selfcontained optical tomography system was developed. Measurements for characterization of the positioning performance of the tomography system reveal a precise and mechanically stable operation. Characterization of the optical performance suggests that best projectional data collection can be obtained when source and detector are operated within a dedicated optimized projection range of 160 degrees. It is shown that accurate fluorescence projection measurements can be obtained from deep tissue-like media using the selected imaging equipment. An experimental verification example of multi-projection-based tomography reveals good reconstructed results in homogeneous phantoms with a single fluorophore inclusion. In simulations it is shown that excitation-resolved multispectral fluorescence tomography can be used to retrieve accurate 3-dimensional reconstructions of single and multiple fluorophores in inhomogeneous small animal tissue. Best results were observed when at least two perspective views are used for multispectral reconstruction emphasizing the use of combined multi-projectional and multispectral optical fluorescence tomography. In conclusion, the developed tomographic system shows great promise to be used in in vivo small animal studies imaging the 3-dimensional biodistribution of fluorescent probes in tissue. The system offers enhanced projection flexibility that capitalizes on independent source-detector positioning within a 360° complete-angle projection geometry. Additionally, excitation-resolved fluorescence tomography appears as a reasonable extension of multi-projection tomography with a high potential in functional small animal imaging. The synergistic exploitation of multi-projection and multispectral tomographic information for reconstruction is therefore expected to improve the general ill-conditioning of the OFT inverse problem, and holds out the prospect for an overall improvement of optical tomography in small animal imaging. In der Optischen Fluoreszenztomographie (OFT) an Kleintieren können optimale 3-dimensionale Rekonstruktionsergebnisse erzielt werden, wenn umfangreiche Datensätze mit hohem räumlichen Informationsgehalt zur Rekonstruktion heran gezogen werden. Üblicherweise werden dazu viele Quellen-Detektor-Projektionsmessungen von einem begrenzten Oberflächenbereich oder über den gesamten Umfang des Messobjektes verteilt. Eine weitere Möglichkeit besteht jedoch darin, tomographische Rohdaten anhand multispektraler anregungsaufgelöster Fluoreszenzmessung zu gewinnen. Das Ziel dieser Arbeit ist die Entwicklung eines neuartigen universellen Fluoreszenztomographiesystems, das die Gewinnung umfangreicher Rohdatensätze sowohl über multi-projektionsbasierte als auch über multispektrale Fluoreszenzmessungen ermöglicht. Das Konzept des OFT-Systems beruht auf einer zirkulären Projektionsanordnung mit unabhängiger Quellen-Detektor-Positionierung. Als Detektor wird eine CCD-Kamera eingesetzt und für die Fluoreszenzanregung sorgt eine kontinuierliche, gefilterte Weißlichtquelle mit breitem Ausgangsstrahl. Diese Methode ermöglicht, das Messobjekt vollständig über 360° zu erfassen und begünstigt damit die Gewinnung umfangreicher Projektionsdaten. Während der Messung befindet sich das Messobjekt dabei in Bauchlage. Die Anwendung einer Weißlichtquelle in Verbindung mit austauschbaren optischen Filtern bildet gleichzeitig die Grundlage für anregungsaufgelöste Fluoreszenzaufnahmen. Für die experimentellen Untersuchungen wurden gewebeähnliche Phantome eingesetzt, welche aus Epoxidharz hergestellt wurden, deren optischen Gewebeparameter über die Beimengung von schwarzer Tinte als Absorber und TiO2-Partikel als Streumedium bestimmt werden. Als Fluoreszenzmarker wurde der NIR-Farbstoff DY-752 verwendet. Zur Evaluierung der multispektralen Tomographie als unterstützende Modalität zur multi-projektionalen Tomographie wurden Simulationen an einem inhomogenen digitalen Mausmodell durchgeführt, welches diverse, anatomisch richtig positionierte Organe beinhaltet. Allen Organen wurden realistische optische Parameter in Abhängigkeit der Wellenlänge zugeordnet. Die anregungsaufgelöste Fluoreszenztomographie wurde für neun Wellenlängen zwischen 580 und 660 nm und mit bis zu vier Projektionsperspektiven durchgeführt. Zur Reduktion des Einflusses durch Gewebeinhomogenitäten wurde eine Reskalierungsmethode eingesetzt. Aufbauend auf grundlegende Untersuchungen der für die Bildgebung ausschlaggebenden optischen Komponenten wurde ein eigenständiges, automatisiertes optisches Fluoreszenztomographiegerät entwickelt. Die technische Bewertung der Systemmechanik weist eine präzise, robuste und wiederholbare Funktion auf und gewährleistet damit eine verlässliche Positionierung von Quelle und Detektor. Die Charakterisierung des verfügbaren Projektionsraumes weist darauf hin, dass beste Ergebnisse erzielt werden, wenn Quelle und Detektor einen bestimmten relativen Mindestwinkel zueinander nicht unterschreiten (Optimaler Projektionsraum). Es konnte gezeigt werden, dass damit die Gewinnung representativer Projektionsdaten von fluoreszierenden Objekten in gewebeähnlichen Medien erhalten werden können. Erste 3-dimensionale Rekonstruktionen experimentell gewonnener Projektionsdaten von einem homogenen Phantom zeigten ebenfalls gute Ergebnisse und bestätigen damit die Funktion des optischen Tomographiegerätes. Zusätzlich konnte gezeigt werden, dass die multispektrale anregungsaufgelöste Fluoreszenztomographie für korrekte 3-D Rekonstruktionen von ein- und mehrfachen Fluorophoreinschlüssen im inhomogenen Kleintiermodell angewendet werden kann, wenn mindestens zwei Projektionsansichten für die multispektrale Rohdatengewinnung benutzt werden. Dies belegt damit gleichzeitig den Nutzen einer kombinierten multispektralen und multi-projektionalen Fluoreszenztomographie. Das in dieser Arbeit vorgestellte tomographische System stellt einen vielversprechenden Ansatz in der funktionellen Kleintierbildgebung dar. Das System profitiert von einer vollständigen 360° Multi-Projektionsumgebung mit unabhängiger Quellen-Detektor-Positionierung. Zusätzlich erweist sich die multispektrale anregungsaufgelöste Tomographie als eine gewinnbringende Erweiterung der multi-projektionsbasierten Fluoreszenztomographie. Die gemeinsame, synergetische Nutzung multi-projektionaler und multispektraler tomographischer Informationen verspricht daher eine Optimierung des generell schlecht gestellten inversen Problems der optischen Tomographie, und lässt damit eine allgemeine Verbesserung der Fluoreszenztomographie in der Kleintierbildgebung erwarten.

The knowledge of the loss mechanisms in industrial back-contact back-junction (BC BJ) silicon solar cells and their dependence on geometrical and substrate parameters provides the opportunity to further increase the cell efficiency of this cell type. In the presented paper the influences of the different loss mechanisms on the cell parameters of BC BJ solar cells were analyzed. The basis of the simulations was an advanced 1-d model that regards the detrimental influences of intrinsic losses, optical losses, series resistance losses and recombination losses on the cell efficiency. In this context the main influence of electrical shading losses will be discussed in particular, due to the restrictions of the minimum base width as a result of industrial structuring processes. The predictions of the theoretical calculations will be compared with the measured cell parameters of BC BJ solar cells for various cell designs. In order to further improve the cell design the influences of geometrical parameters (pitch, base width) and substrate parameters (base resistivity) on the cell efficiency will be analyzed. The modeling data show that the optimum cell geometry defines a balance between series resistance and electrical shading losses.

After completion of the solar cell manufacturing process the current-density versus voltage curves (J(U) curves) are measured to determine the solar cell's efficiency and the mechanisms limiting the efficiency. An accurate and robust analysis of the measured curves is essential. In this work it is shown that fitting the two-diode model is inappropriate to quantify recombination in the space charge region and ohmic losses due to series resistance. Three fill factors, namely the fill factor of the illuminated J(U) curve, the pseudo fill factor of the sunsVoc curve and the ideal fill factor of the single diode model, are the base of a quick loss analysis that is evaluated in the present paper. It is shown that for an accurate analysis the distributed character of the series resistance and the network character of the solar cell cannot be neglected. An advanced current-voltage curve analysis including fill factors and fit is presented.
