BACKGROUND Fugacity, the driving force for transdermal uptake of chemicals, can be difficult to predict based only on the composition of complex, non-ideal mixtures such as personal care products. OBJECTIVE Compare the predicted transdermal uptake of benzophenone-3 (BP-3) from sunscreen lotions, based on direct measurements of BP-3 fugacity in those products, to results of human subject experiments. METHODS We measured fugacity relative to pure BP-3, for commercial sunscreens and laboratory mixtures, using a previously developed/solid-phase microextraction (SPME) method. The measured fugacity was combined with a transdermal uptake model to simulate urinary excretion rates of BP-3 resulting from sunscreen use. The model simulations were based on the reported conditions of four previously published human subject studies, accounting for area applied, time applied, showering and other factors. RESULTS The fugacities of commercial lotions containing 3-6% w/w BP-3 were ~20% of the supercooled liquid vapor pressure. Simulated dermal uptake, based on these fugacities, are within a factor of 3 of the mean results reported from two human-subject studies. However, the model significantly underpredicts total excreted mass from two other human-subject studies. This discrepancy may be due to limitations in model inputs, such as fugacity of BP-3 in lotions used in those studies. SIGNIFICANCE The results suggest that combining measured fugacity with such a model may provide order-of-magnitude accurate predictions of transdermal uptake of BP-3 from daily application of sunscreen products.

Coupling of membrane distillation with bioreactors containing enzymes such as ‘laccase’ forms an enzymatic membrane bioreactor resulting in complete retention of both trace organic contaminants and enzyme, facilitating simultaneous trace organic contaminants degradation. Integration of enzymatic membrane bioreactor and ultraviolet photolysis may result in further improved degradation of trace organic contaminants in membrane-concentrate. We studied the degradation as well as by-products formation of five selected trace organic contaminants, namely, sulfamethoxazole, diclofenac, bisphenol A, oxybenzone, and carbamazepine by ‘membrane distillation — ultraviolet photolysis’ system and ‘enzymatic membrane bioreactor — ultraviolet photolysis’ system. In the former, the membrane effectively retained the trace organic contaminants and then ultraviolet photolysis of membrane-concentrate resulted in trace organic contaminant degradation in the following order: diclofenac (88 %) > sulfamethoxazole (71 %) > oxybenzone (35 %) > bisphenol A (33 %) > carbamazepine (27 %). By contrast, the enzymatic membrane bioreactor — ultraviolet photolysis system resulted in 100 % degradation of diclofenac, sulfamethoxazole, and bisphenol A and around 70 % degradation of oxybenzone and carbamazepine. This system also resulted in more than 50 % reduction in number of degradation products with 60–70 % lower abundance. Our results indicate that laccase degradation led to products that were more amenable to the post-treatment by ultraviolet photolysis. Overall, it can be concluded that for enzymatic membrane bioreactor — ultraviolet photolysis system, enzymatic pre-treatment not only helped in better degradation of the parent trace organic contaminants but also led to the formation of fewer by-products with lower abundance (i.e., more complete degradation).

A sensitive method was developed and validated for the determination of UV filters (oxybenzone, octocrylene, 4-methyl-benzylidene camphor, 2-ethylhexyl-4-methoxycinnamate and avobenzone) in tilapia muscle using matrix solid-phase dispersion followed by on-line solid-phase extraction–liquid chromatography/UV. Laccase-catalysed degradation is a promising technology for the degradation of trace organic contaminants (TrOCs). However, depending on their physicochemical properties (e.g., chemical structures), non-phenolic TrOCs appear to be resistant to laccase-catalysed degradation, thus requiring an additional treatment process. In this study, the performance of an integrated laccase and persulfate (PS) mediated oxidation process was explored and elucidated for the first time. Compared to that achieved during treatment with laccase alone, the addition of PS at 1–10 mM concentration into batch enzymatic bioreactors showed concentration-dependent improvement in the degradation of the five TrOCs selected, namely diclofenac, sulfamethoxazole, carbamazepine, bisphenol A and oxybenzone. The results obtained from a series of batch tests also confirmed that the structural components (e.g., carbohydrate moieties in the enzyme) as well as the possibility of PS acting as a final acceptor of electrons from the type II and type III active sites of laccase may have led to PS activation. A continuous treatment system was developed by integrating a nanofiltration (NF) membrane with the laccase/PS process (NF-MBRlaccase/PS), which achieved TrOC-specific degradation. Importantly, the degradation of non-phenolic TrOCs (diclofenac, sulfamethoxazole and carbamazepine) further improved by 10 to 65% in the NF-MBRlaccase/PS system. This improvement could be attributed to: (i) the prolonged contact time between laccase/PS and TrOCs, which was possible due to their complete retention by the NF membrane; and (ii) retention of oxidative cross coupling agents or secondary radicals enhancing TrOC degradation. The NF membrane not only retained the moderately degraded carbamazepine and sulfamethoxazole effectively but also effectively removed toxic transformation products and residual estrogenicity of the final effluent as evaluated by cytotoxicity and estrogenicity assays, respectively. These observations indicate the significance of the high retention membrane in the NF-MBRlaccase/PS system for producing high quality permeate suitable for disposal and potentially for water reuse applications.

Household dust is a reservoir of various consumer product chemicals. Thus, characterizing comprehensive chemical profiles of house dust may help improve our understanding of residential chemical exposure. We have previously developed a method for detecting a broad spectrum of chemicals in dust by applying a combination of target, suspect screening, and non-target methods with mass spectrometry preceded by liquid chromatography and gas chromatography. Building upon a previous study that detected 271 compounds in 38 dust samples, we presented concentrations of 144 compounds that were confirmed and quantified by standards in the same set of samples. Ten compounds were measured with median concentrations greater than 10000ng/g of dust: cis-hexadec-6-enoic acid, squalene, cholesterol, vitamin E, bis(2-ethylhexyl) phthalate, dioctyl terephthalate, linoleic acid, tricaprylin, tris(1-chloroisopropyl) phosphate, and oxybenzone. We also reviewed in vitro toxicity screening data to identify compounds that were not previously detected in indoor dust but have potential for adverse health effects. Among 119 newly detected compounds, 13 had endocrine-disrupting potential and 7 had neurotoxic potential. Toxicity screening data were not available for eight biocides, which may adversely affect health. Our results strive to provide more comprehensive chemical profiles of house dust and identified information gaps for future health studies.

The mutagenicity of four organic UV filters namely oxybenzone (benzophenone-3), dioxybenzone (benzophenone-8), avobenzone, and octyl methoxycinnamate, in chlorinated bromide-rich water (artificial seawater) was investigated. Mutagenicity was evaluated using Ames test in Salmonella typhimurium TA98 without S9 mix. Chemical analysis using high-resolution mass spectrometry was carried out to elucidate the mutagenic transformation products. Among the studied UV filters, only dioxybenzone exhibited a clear mutagenic activity following chlorination in seawater at ratio 1:10 (UV filter:chlorine). In contrast, no mutagenic activity was detected when chlorine was added at higher doses (ratio 1:1000). High-resolution mass spectrometry analysis showed that mutagenic extracts contained several brominated transformation products of dioxybenzone. Time course analysis of the transformation products at increasing chlorine doses showed that they were unstable and disappeared more quickly at higher chlorine doses. This instability explained the absence of mutagenic activity of dioxybenzone when 1000-fold excess chlorine was added, as no transformation products were detected. Relevance of these findings to the context of swimming pool is discussed. Further investigations taking into consideration the mutagenicity of not only the intermediate transformation products but also the final disinfection byproducts are needed to determine the overall impact of high levels of chlorine on the overall mutagenicity. This study highlights the importance of considering the reactivity of organic UV filters and their transformation products in disinfected recreational waters when sunscreen formulations are prepared.

Rationale Exposure to UV light can induce adverse effects on human health, such as photo-aging, immunosuppression, and cancer. Sunscreens are used to prevent the absorption of UV rays, but certain UV-filtering compounds have been shown to disrupt endocrine systems or act as carcinogens. To assess the effects of the exposure to such compounds, it is important to study the pathways by which they are biotransformed in the body. Methods Liquid chromatography coupled to high-resolution tandem mass spectrometry (LC/HRMS/MS) was employed to evaluate the oxidative metabolism and, specifically, the formation of reactive metabolites of six active ingredients commonly used in sunscreen formulations: oxybenzone, avobenzone, homosalate, octisalate, octocrylene, and octinoxate. In vitro incubations were performed with human and rat liver microsomes in the presence of β-nicotinamide adenine dinucleotide phosphate and glutathione. An LC/HRMS/MS method was developed to identify metabolites employing a biphenyl reversed-phase column for separating parent molecules, metabolites, and glutathione (GSH) adducts. Results Each tested compound resulted in the formation of several metabolites, including at least one GSH adduct. Compounds containing ester groups were hydrolyzed, and some metabolites of the free acid forms were also detected. High-resolution MS/MS data was crucial for the structural elucidation of metabolites and GSH adducts. Fragmentation pathways were proposed for all parent compounds, as well as each described metabolite and adduct. Conclusions The results of this study will help better understand the metabolism and detoxification pathways of these xenobiotics.

An analytical method was developed and validated for the simultaneous determination in fish muscle of two kinds of emerging pollutants: polycyclic aromatic musks (galaxolide and tonalide) and UV filters [oxybenzone, 3-(4-methylbenzylidene) camphor, padimate-O, 2-ethylhexyl 4-methoxycinnamate, octocrylene], which are very common ingredients of personal care products, by programmable split/splitless injector gas chromatography-mass spectrometry (PSSI-GC-MS) using a matrix solid-phase dispersion (MSPD) for extraction and clean-up procedure in a single step. The method was evaluated for precision (relative standard deviation 0.9829); accuracy, reported as % of recovery; limit of quantification (0.013 to 0.040 μg g−1, referred to dry mass); and limit of detection (0.004 to 0.012 μg g−1, referred to dry mass) for each of the analytes. The average recoveries in muscle samples spiked at two levels: 0.1 μg g-1 and 1.0 μg g-1 ranged between 77–111% and 55–108%, respectively. The method was applied for the determination of the mentioned pollutants in an exposure in vivo study with rainbow trout, and the results showed a larger absorption of musks (tonalide 43.0 μg g−1) than UV filters (padimate-O, 1.0 μg g−1). The proposed procedure has demonstrated to be able for simultaneous monitoring of these two different kinds of common emerging pollutants coming from personal care products, which would make it a useful tool for quality control and safety studies.

UV filters (UV-Fs) are a group of hormonally active chemical compounds used to protect against the deleterious effects of UVA and UVB solar radiation, which are currently present in most consumer goods (personal care products, plastics, fabrics, paints, etc). Last years the concern about these emerging contaminants has been on the rise, and increasing efforts are being taken in order to properly asses the hazard that the exposure to these compounds in the early stages of life may pose. In this study, a new method for the analysis of 11 UV-Fs residues in human breast milk samples has been developed. The method is based on turbulent flow chromatography coupled to liquid chromatography-tandem mass spectrometry (TFC-HPLC-MS/MS). The validated method was successfully applied to 79 human breast milk samples from mothers in Barcelona (Spain). Twenty-four per cent of the samples contained UV-Fs, with major contributors being oxybenzone (benzophenone 3, BP3), its metabolite 4,4'-dihydroxybenzophenone (4DHB), and UV320 showing maximum concentrations of 779.9, 73.3, and 523.6ngg-1 milk, respectively. Additionally, the plastic containers of the milks were also analysed, revealing high concentrations of BP3 and 4DHB, up to 10.6µgg-1 plastic. The calculated mean ΣUV-Fs were useful to estimate the daily intake (EDI) by babies, which were 69.1µg d-1kg-1 body weight.

This study provided the first comprehensive data on the occurrence and removal of twenty-five target emerging contaminants (ECs) in a full-scale water reclamation plant (WRP) in the Southeast Asian region. Nineteen out of the twenty-five ECs were ubiquitously detected in raw influent samples. Concentrations of the detected ECs in raw influent samples ranged substantially from 44.3 to 124,966ng/L, depending upon the compound and sampling date. The elimination of ECs in full-scale conventional activated sludge (CAS) and membrane bioreactor (MBR) systems at a local WRP was evaluated and compared. Several ECs, such as acetaminophen, atenolol, fenoprofen, indomethacin, ibuprofen, and oxybenzone, exhibited excellent removal efficiencies (>90%) in biological wastewater treatment processes, while some of the investigated compounds (carbamazepine, crotamiton, diclofenac, and iopamidol) appeared to be persistent in the both CAS and MBR systems. Field-based monitoring results showed that MBR outperformed CAS in the elimination of most target ECs. The relationship between molecular characteristics of ECs (i.e. physicochemical properties and structural features) and their removal efficiencies during biological wastewater treatment was also elucidated. Excellent removal efficiencies (>90%) were often noted for ECs with the sole presence of electron donating groups (i.e. phenolic [OH], amine [NH2], methoxy [OCH3], phenoxy [OC6H5], or alkyl groups). Conversely, ECs with the absence of electron donating groups or the predominance of strong electron withdrawing groups (e.g. halogenated, carbonyl, carboxyl, and sulfonamide) tended to show poor removal efficiencies (

International audience; Organic ultraviolet (UV) filters are used in sunscreens and other personal-care products to protect against harmful effects of exposure to UV solar radiation. Little is known about the fate of UV filters in seawater swimming pools disinfected with chlorine. The present study investigated the occurrence and fate of five commonly used organic UV filters, namely dioxybenzone, oxybenzone, avobenzone, 2-ethylhexyl-4-methoxycinnamate, and octocrylene, in chlorinated seawater swimming pools. Pool samples were collected to monitor the variation of UV filter concentrations during pool opening hours. Furthermore, laboratory-controlled chlorination experiments were conducted in seawater spiked with UV filters to investigate the reactivity of UV filters. Extracts of chlorination reaction samples were analyzed using high-resolution mass spectrometry and electron-capture detection to identify the potentially formed byproducts. In the collected pool samples, all the UV filters except dioxybenzone were detected. Chlorination reactions showed that only octocrylene was stable in chlorinated seawater. The four reactive UV filters generated brominated transformation products and disinfection byproducts. This formation of brominated products resulted from reactions between the reactive UV filters and bromine, which is formed rapidly when chlorine is added to seawater. Based on the identified byproducts, the transformation pathways of the reactive UV filters were proposed for the first time. Bromoform was generated by all the reactive UV filters at different yields. Bromal hydrate was also detected as one of the byproducts generated by oxybenzone and dioxybenzone.

A novel membrane distillation – enzymatic membrane bioreactor (MD-EMBR) system was developed for efficient degradation of trace organic contaminants (TrOCs). Degradation of five TrOCs, namely carbamazepine, oxybenzone, diclofenac, atrazine and sulfamethoxazole was examined using two commercially available laccases (from Trametes versicolor and Aspergillus oryzae ). The MD system ensured complete retention (>99%) of both enzyme and TrOCs. Of particular interest was that the complete retention of the TrOCs resulted in high TrOC degradation by both laccases. Oxybenzone and diclofenac degradation in the MD-EMBR ranged between 80 and 99%. Compared to previously developed EMBRs, as much as 40% improvement in the removal of resistant non-phenolic TrOCs ( e.g., carbamazepine) was observed. Laccase from A. oryzae demonstrated better TrOC degradation and enzymatic stability. With the addition of redox mediators, namely 1-hydroxybenzotriazole (HBT) or violuric acid (VA), TrOC degradation was improved by 10–20%. This is the first demonstration of a laccase-based high retention membrane bioreactor for enhanced biodegradation of TrOCs.

There is a need to develop viable techniques for removal and/or recovery of organic and inorganic compounds from environmental matrices, due to their eco-toxicity, regulatory obligations or potential supplies as secondary materials. Simultaneous phosphorus (P) recovery and removal of emerging organic contaminants (caffeine, bisphenol A, 17α-ethinylestradiol and oxybenzone) from sewage sludge (SS) was assessed using the electrodialytic (ED) process. The SS samples were collected at the secondary settling tank between May and July, 2015. Different current intensities were tested continuously or sequentially (between 50, 75 and 100 mA), in a 2 compartment cell, placing the SS in the cathode end, separated from the anode end through an anion exchange membrane. After 24 h, between 45 and 55% of P electromigrated to the anode end in all experiments and the application of higher current intensities did not promote significant improvements in its recovery. After 72 h, the best results were obtained when 100 mA (0.21 mA/cm 2 ) were applied: 70% of P were recovered in the anolyte and between 79 and 96% of organic compounds were degraded. Still, the obtained anolyte enriched in P was not completely free of organic contaminants.

A new voltammetric methodology is proposed for the simultaneous determination of the organic UV filters oxybenzone (benzophenone 3, BP3) and octocrylene (OC) in personal care products and in wastewater extracts. It is based on previous adsorptive stripping voltammetric (AdSV) methods developed for hanging mercury drop electrodes, adapted to the special characteristics of carbon-based screen-printed electrodes. Among the carbon substrates tested, regular carbon screen-printed devices exhibited the best performance, with detection limits of 4.8 and 6.6 μmol L−1 and linearity ranges of 16–400 and 22–400 μmol L−1 for BP3 and OC, respectively, and, as compared to mercury electrodes, with the advantages of environmental safety, easy of use, low cost and compatibility with automation and flow measurements. Application of sunscreen is one of many ways to protect skin from the harmful effects of UV radiation. Sunscreen products are widely used and regulated as over-the-counter drug products in the United States. The U.S. Food and Drug Administration recommends an assessment of human systemic absorption of sunscreen active ingredients with a Maximal Usage Trial. The FDA conducted a clinical study to determine the systemic exposure of sunscreen active ingredients present in 4 commercially available sunscreen products of different formulation types under maximal usage conditions. To support this clinical study, a sensitive and specific LC-MS/MS method for the simultaneous determination of the two sunscreens avobenzone and oxybenzone in human plasma was developed. Phospholipid removal 96-well protein precipitation plates were used for sample clean-up and the extracted samples were chromatographed on an Ethylene-Bridged Hybrid (BEH) C18 column in isocratic flow using 10 mM ammonium formate in 0.1% formic acid and methanol (24:76, v/v) as a mobile phase. A triple quadrupole mass spectrometer in multiple reaction monitoring (MRM) mode was used to acquire data. The method was validated as per current FDA bioanalytical method validation guidance, in the ranges 0.20–12.00 ng/mL for avobenzone and 0.40–300.00 ng/mL for oxybenzone. The validated method was used to analyze these active ingredients in human clinical study samples.

A low saline backwater canal, mainly utilized for domestic and agricultural purposes, has been analyzed for the possible presence of organic micropollutants (OMP) and their potential removal was explored by multilayered microfiltration membranes. The qualitative as well as quantitative analysis were carried out for a span of one year using the technique of liquid chromatography connected with high resolution mass spectrometry (LC-Q-TOF-MS). The identification of the formally unknown compounds was initially done using non-target analysis based on the mass accuracy, isotopic pattern and MS/MS spectral interpretation. Results of the non target screening revealed the presence of 11 OMPs. Five of these OMPs were confirmed using standards; these include chlorophene (CHP), oxybenzone (OXY), N, N-diethyl-meta-toluamide (DEET), N, N-diethyl-benzamide (DEB) and dibutyl phthalate (DBP). Among the confirmed OMPs, the highest concentration was observed for DBP (244.61 ng l−1). The most frequently observed OMP in the study area was DBP while the least was DEB which is an insect repellent as well as a degradation product of DEET. The ecological risk associated with the target compounds has also been analyzed by calculating the risk quotient (RQ) and the results revealed that at the detected levels, these compounds are capable of causing low to medium risk. A series of undergraduate laboratory experiments that utilize reversed-phase HPLC separation, inductively coupled plasma spectroscopy (ICP), and scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS) are described for the analysis of commercial sunscreens. The active ingredients of many sunscreen brands include zinc or titanium oxide in addition to organic acids. Students determine the zinc content using ICP, and the chemical composition as well as particle sizes using SEM-EDS. The organic UV absorbers octocrylene and oxybenzone are quantified using HPLC. With the incorporation of these interesting characterization techniques in second or fourth-year chemistry courses, and by having students analyze sunscreen samples that are medically relevant in terms of health effects, students engage in timely research and at the same time gain exposure to a variety of instruments in the analysis of a familiar household product.

Oxybenzone (OBZ), known as Benzophenone-3, is a commonly used UV filter in sun tans and skin protectants, entering aquatic systems either directly during recreational activities or indirectly through wastewater treatment plants discharge. To study the potential degradation capacity of plants for OBZ in phytotreatment, a well-established hairy root culture (Armoracia rusticana) was treated with OBZ. More than 20% of spiked OBZ (100μM) was eliminated from the medium by hairy roots after 3h of exposure. Two metabolites were identified as oxybenzone-glucoside (OBZ-Glu) and oxybenzone-(6-O-malonyl)-glucoside (OBZ-Mal-Glu) by LC-MS/MS and TOF-MS. Formation of these metabolites was confirmed by enzymatic synthesis, as well as enzymatic and alkaline hydrolysis. Incubation with O-glucosyltransferase (O-GT) extracted from roots formed OBZ-Glu; whereas β-d-Glucosidase hydrolyzed OBZ-Glu. However, alkaline hydrolysis led to cleavage of OBZ-Mal-Glu and yielded OBZ-Glu. In the hairy root culture, an excretion of OBZ-Glu into the growth medium was observed while the corresponding OBZ-Mal-Glu remained stored in root cells over the incubation time. We propose that metabolism of oxybenzone in plants involves initial conjugation with glucose to form OBZ-Glu followed by malonylation to yield OBZ-Mal-Glu. To our best knowledge this first finding presenting the potential of plants to degrade benzophenone type UV filters by phytoremediation.

This study systematically compares the performance of ultrafiltration (UF) and nanofiltration (NF) based enzymatic membrane bioreactors (EMBRs) for the degradation of five micropollutants, namely atrazine, carbamazepine, sulfamethoxazole, diclofenac and oxybenzone to elucidate the impact of effective membrane retention of micropollutants on their degradation. Based on the permeate quality, NF-EMBR achieved 92–99.9% micropollutant removal (i.e., biodegradation + membrane retention), while the removal of these micropollutants by UF-EMBR varied from 20 to 85%. Mass balance analysis revealed that micropollutant degradation was improved by 15–30% in NF-EMBR as compared to UF-EMBR, which could be attributed to the prolonged contact time between laccase and micropollutants following their effective retention by the NF membrane. A small decline in permeate flux was observed during EMBR operation. However, the flux could be recovered by flushing the membrane with permeate.

Ultraviolet radiation (UV-R) causes genotoxic and aging effects on skin, and sunscreens are used to alleviate the damage. However, sunscreens contain synthetic shielding agents that can cause harmful effects in the environment. Nature-derived substances may have potential as replacement materials for the harmful sunscreen chemicals. However, screening of a broad range of samples is tedious, and often requires a separate genotoxicity assessment. We describe a simple microplate technique for the screening of UV protective substances using a recombinant Escherichia coli biosensor. Both absorbance-based and bioactivity-based shields can be detected with simultaneous information about the sample genotoxicity. With this technique, a controversial sunscreen compound, oxybenzone offers physical or absorbance-based shield but appears genotoxic at higher concentrations (3.3 mg/mL). We also demonstrate that pine needle extract (PiNe ) shields the biosensor from UV-R in a dose-dependent manner without showing genotoxicity. The physical shield of 5 mg/mL PiNe was similar to that of one of the most common UV-shielding compound TiO2 concentration 0.80 mg/mL. The bioactivity-based shield of PiNe also reaches the extent of the physical shield with the highest concentration (3.3 mg/mL). We conclude that our technique is suitable in detecting the UV-shielding potential of natural substances, and gives simultaneous information on genotoxicity.

The UVA absorbers such as avobenzone are widely used for sunlight protection; however, they show a significant skin penetration. The aim of the present study was to formulate UVA absorbers into mesoporous silicas (MSs) for enhanced UVA protection with reduced percutaneous absorption. Two MSs prepared with different structure-directing agents (Pluronic P123 as single MS and combined Pluronic P123-Pluronic F68 as hybrid MS) were synthesized in this study. The hybrid MS exhibited higher specific surface area (853 m2/g) than the single MS (764 m2/g). The particle sizes of single MS and hybrid MS were about 1 and 1.5 μm, respectively. The adsorbed avobenzone had greatly decreased crystallinity compared with free avobenzone. The in vitro photoprotection determined by UVA/UVB ratio showed that the MS-loaded avobenzone in hydrogel endowed a synergistic effect on UVA protection compared to the free avobenzone. The skin absorption test using Franz diffusion cell indicated that the skin permeation of avobenzone and oxybenzone from MSs in semisolid preparations was one-third to one-half of those from free control. This effect was observed by using both pig skin and UVA-damaged nude mouse skin as the penetration barriers. Topical application of hybrid MS on nude mouse skin before UVA irradiation had prevented the increased transepidermal water loss (TEWL), furrow formation, keratinocyte apoptosis, and neutrophil infiltration. Our findings conclude that MSs containing avobenzone or oxybenzone effectively ameliorated UVA-induced skin disruption and reduced the possible toxicity elicited by percutaneous penetration.

Accurate, reliable, and sensitive spectrophotometric and chemometric methods were developed for simultaneous determination of octinoxate (OMC), oxybenzone (OXY), and octocrylene (OCR) in a sunscreen formulation without prior separation steps, including derivative ratio spectra zero crossing (DRSZ), double divisor ratio spectra derivative (DDRD), mean centering ratio spectra (MCR), and partial least squares (PLS-2). With the DRSZ technique, the UV filters could be determined in the ranges of 0.5–13.0, 0.3–9.0, and 0.5–9.0 μg/mL at 265.2, 246.6, and 261.8 nm, respectively. By utilizing the DDRD technique, UV filters could be determined in the above ranges at 237.8, 241.0, and 254.2 nm, respectively. With the MCR technique, the UV filters could be determined in the above ranges at 381.7, 383.2, and 355.6 nm, respectively. The PLS-2 technique successfully quantified the examined UV filters in the ranges of 0.5–9.3, 0.3–7.1, and 0.5–6.9 μg/mL, respectively. All the methods were validated according to the International Conference on Harmonization guidelines and successfully applied to determine the UV filters in pure form, laboratory-prepared mixtures, and a sunscreen formulation. The obtained results were statistically compared with reference and reported methods of analysis for OXY, OMC, and OCR, and there were no significant differences with respect to accuracy and precision of the adopted techniques.

A fast gas chromatography/mass spectrometry method was developed and validated for the analysis of the potential endocrine disrupters octinoxate and oxybenzone in swimming pool water samples based on the solvent-free solid-phase microextraction technique. The low-pressure gas chromatography/mass spectrometry method used for the fast identification of UV filter substances was compared to a conventional method in terms of sensitivity and speed. The fast method proposed resulted in 2 min runs, leading to an eightfold decrease in the total analysis time and a sevenfold improvement in detection limits. The main parameters affecting the solid-phase microextraction process were also studied in detail and the optimized conditions were as follows: fiber coating, polyacrylate; extraction mode, direct immersion; extraction temperature, 25°C; sample volume, 5 mL; extraction time 45 min; pH 6.5. Under the optimized conditions, a linear response was obtained in the concentration range of 0.5-25 μg/L with correlation coefficients in the range 0.990-0.999. The limits of detection were 0.17-0.29 μg/L, and the recoveries were 80-83%. Combined method uncertainty was assessed and found to be less than 7% for both analytes for concentrations equal to or higher than 5 μg/L. Pool water samples were analyzed to demonstrate the applicability of the proposed method. Neither octinoxate nor oxybenzone were detected in the swimming pool water samples at concentrations above the respective limits of detection.

Given that previous studies have highlighted the anti-inflammatory nature, which is influenced by UV radiation, of a certain number of ultraviolet filters currently used in Europe, it seemed interesting to evaluate the persistence of this type of effect. The persistence of the anti-inflammatory effect of fourteen preparations, each containing one of the ultraviolet filters authorized by Regulation (CE) No. 1223/2009, and of 10 commercially-available sun products was studied using the phorbol-myristate-acetate test on mice, up to 6 and a half hours after application. We can observe that a benzophenone, oxybenzone, a PABA derivative, octyldimethylPABA and a derivative of cinnamic acid, OMC as well as 3 commercially-available products, display a very marked anti-inflammatory effect at the end of our experimentation phase. The type of effect observed could encourage users to prolong the time that they are exposed to the sun, due to there being no warning signs, namely sunburn.

A simple method for the simultaneous determination of personal care product ingredients: galaxolide, tonalide, oxybenzone, 4-methylbenzyliden camphor, padimate-o, 2-ethylhexyl methoxycinnamate, octocrylene, triclosan, and methyl triclosan in lettuce by ultrasound-assisted extraction combined with solid-phase microextraction followed by gas chromatography with mass spectrometry was developed. Insect repellent DEET and sunscreen ingredient oxybenzone play an essential role in minimizing vector-borne diseases and skin cancers. The purpose of this study was to investigate the effects of emulsion type, addition of thickening agent and droplet size in three emulsion-based lotions on percutaneous permeation of DEET and oxybenzone using in vitro diffusion experiments, in order to minimize overall systemic permeation of the substances. Formulation C (water-in-oil emulsion) significantly increased overall permeation of DEET through human skin (56%) compared to Formulation A (oil-in-water emulsion). Formulation B (oil-in-water emulsion with thickening agent xanthan gum) significantly decreased the size of oil droplet containing DEET (16%), but no effect on oil droplets containing oxybenzone. Adding xanthan gum also increased overall permeation of DEET and oxybenzone (21% and 150%) when compared to Formulation A; presence of both ingredients in Formulation B further increased their permeation (36% and 23%) in comparison to its single counterparts. This study used three kinds of wax and three kinds of oil, with fixed mixture ratio including UV-blocking materials of ethylhexyl methoxycinnamate, oxybenzone, and avobenzone, and applied hot high-pressure homogenization process to prepare nanolipid sunscreen formulations. The measured particle size of the sunscreen formulations was 100~300 nm around PDI of 0.2 having a moderate polydisperse system. The distribution of zeta potential was −50 mV to −35 mV, showing a stable system. The UV light-absorbing range of 9 groups of sunscreen formulations was 275 nm~380 nm ranging within UVA and UVB. The rheological analysis found that the viscosity change is shear, thinning exhibiting colloid behavior. Taguchi analysis found that the optimum combinations are the carnauba wax and the blackcurrant oil combination for crystallinity and the beeswax and CPG oil for UV absorption. In addition, UV-blocking ability shows that the SPF was 51.5 and PFA was three stars for SU9 formulation. Finally, the effect of temperature on the properties of sunscreen formulations was also explored.

This study examined effects of Fenton oxidation on trace level pharmaceuticals and personal care products (PPCPs) commonly occurring in wastewater. The tested PPCPs included acetaminophen, atenolol, atrazine, carbamazepine, metoprolol, dilantin, DEET, diclofenac, pentoxifylline, oxybenzone, caffeine, fluoxetine, gemfibrozil, ibuprofen, iopromide, naproxen, propranolol, sulfamethoxazole, bisphenol-A and trimethoprim. Transformations of effluent organic matter (EfOM) caused by Fenton oxidation were also quantified. All tested PPCPs, except atrazine and iopromide, were completely removed by Fenton treatment carried out using a 20mg/L Fe (II) concentration and a 2.5 H(2)O(2)/Fe (II) molar ratio. Up to 30% on the total carbon concentration was removed during Fenton treatment which was accompanied by the oxidation of EfOM molecules and formation of oxidation products such as oxalic, formic and acetic acids and, less prominently, formaldehyde, acetaldehyde, propionaldehyde and glycolaldehyde. The absorbance of EfOM treated with Fenton reagent at varying Fe (II) concentration and contact time underwent a consistent decrease. The relative decrease of EfOM absorbance was strongly and unambiguously correlated with the removal of all tested PPCPs.

Removal of a large suite of pharmaceuticals and endocrine disrupting compounds (EDCs) was measured through both pilot- and full-scale riverbank filtration (RBF) facilities. The pilot-scale RBF effectively reduced low ng/L concentrations of most compounds by greater than 90% following a 36-day experiment. Breakthrough of the conservative tracer occurred after 10 days and reached 90% recovery after 28 days. Only four compounds (diazepam, meprobamate, phenytoin and tris(2-chloroethyl) phosphate (TCEP)) were between 20 and 80% removed, and three compounds (atrazine, carbamazepine, and sulfamethoxazole) were less than 20% removed by the pilot. Removal of compounds through the pilot-scale RBF is consistent with results from two full-scale RBF systems: atrazine, carbamazepine, meprobamate, phenytoin and sulfamethoxazole persisted through infiltration into drinking water, whereas caffeine, DEET, gemfibrozil, ibuprofen, iopromide, TCEP, triclosan and trimethoprim were removed. Removal during RBF was attributed to (1) microbial degradation for compounds which were either negatively charged or were neutrally charged and had low log KOW values (such as acetaminophen, caffeine, and trimethoprim), or (2) adsorption for neutrally-charged compounds with high log KOW values (including estradiol, estrone, ethynylestradiol, fluoxetine, oxybenzone, progesterone, testosterone, and triclosan).

Synopsis In this study, high-temperature liquid chromatographic (HTLC) and subcritical water chromatographic (SBWC) separations of sunscreens contained in skincare creams were achieved at temperatures ranging from 90 to 250°C. The columns employed in this work include a ZirChrom-DiamondBond-C18, a XTerra MS C18 and a XBridge C18 column. The quantity of methanol consumed by the greener HTLC sunscreen methods developed in this project is significantly reduced although the HTLC separation at this stage is not as efficient as that achieved by traditional HPLC. SBWC separation of sunscreens was also achieved on the XTerra MS C18 and the XBridge C18 columns using pure water at 230–250°C. Methanol was eliminated in the SBWC methods developed in this study.

High throughput analysis of sunscreen agents present in cream cosmetic has been demonstrated, typically 2 samples per minute, using neutral desorption extractive electrospray ionization mass spectrometry (ND-EESI-MS) without sample pretreatment. For the targeted compounds such as 4-Aminobenzoic acid and oxybenzone, ND-EESI-MS method provided linear signal responses in the range of 1-100 ppb. Limits of detection (LOD) of the method were estimated at sub-ppb levels for the analytes tested. Reasonable relative standard deviation (RSD=8.4-16.0%) was obtained as a result of 10 independent measurements for commercial cosmetics samples spiked with each individual sunscreen agents at 1-10 ppb. Acceptable recoveries were achieved in the range of 87-116% for direct analysis of commercial cream cosmetic samples. The experimental data demonstrate that ND-EESI-MS is a useful tool for high throughput screening of sunscreen agents in highly viscous cream cosmetic products, with the capability to obtain quantitative information of the analytes.

Nanostructured lipid carriers (NLCs) are potential active delivery systems based on mixtures of solid lipids and liquid oil. In this paper, aqueous dispersions of NLCs were prepared by a hot high-pressure homogenization technique using carnauba wax as the solid lipid and isodecyl oleate as the liquid oil. The preparation and stability parameters of benzophenone-3-loaded NLCs have been investigated concerning particle size, zeta potential and loading capacity to encapsulate benzophenone-3, a molecular sunscreen. The current investigation illustrates the effect of the composition of the lipid mixture on the entrapment efficiency, in vitro release and stability of benzophenone-3-loaded in these NLCs. A loading capacity of approximately 5% of benzophenone-3 (m(BZ-3) /m(lipids) ) was characteristic of these systems.

Picaridin and oxybenzone are two active ingredients found in repellent and sunscreen preparations, respectively. We performed a series of in vitro diffusion studies to evaluate the transmembrane permeation of picaridin and oxybenzone across human epidermis and poly(dimethylsiloxane) (PDMS) membrane. Permeation of picaridin (PCR) and oxybenzone (OBZ) across human epidermis was suppressed when both active ingredients were used concurrently; increasing concentration of the test compounds further reduced the permeation percentage of picaridin and oxybenzone. While permeation characteristics were correlative between human epidermis and PDMS membrane, permeability of PDMS membrane was significantly larger than that of human epidermis. The findings were different from concurrent use of repellent DEET and sunscreen oxybenzone in which a synergistic permeation enhancement was observed. Further comparative studies are therefore needed to understand permeation mechanisms and interactions between picaridin and oxybenzone.

Simultaneous determination of 2-ethylhexyl-4-methoxycinnamate (OMC) and oxybenzone (OB) in commercial sunscreen formulations has been achieved by using second order derivative spectrophotometry. The analytical methodology described does not require elaborate pretreatment of samples. The results obtained could be validated by reverse phase HPLC assay by using isocratic methanol: water (88: 12) solvent system. The sensitivity of the developed derivative spectrophotometry method was determined to be 9.0 × 10−3 L/mg for OB and 9.56 × 10−3 L/mg for OMC. The limit of instrumental detection (LOD) was determined to be 0.6 mg/L for OB and 1.38 mg/L for OMC with relative standard deviation of 0.001 and 0.004 for OB and OMC respectively. The method developed can be used for quick assay of commercial sunscreens.

The impact of natural organic matter (NOM) and cations on the rejection of five endocrine disrupting compounds (EDCs) and pharmaceutically active compounds (PhACs) (acetaminophen, carbamazepine, estrone, gemfibrozil, oxybenzone) by nanofiltration (NF) was examined. The water matrices included membrane bioreactor (MBR) effluent, Lake Ontario water and laboratory-prepared waters modelled to represent the characteristics of the Lake Ontario water. The impact of cations in natural waters on compound rejection was also examined by doubling the natural cation concentration (calcium, magnesium, sodium) in both the Lake Ontario water and the MBR effluent. The presence of Suwannee River NOM spiked into laboratory-grade water was found to cause an increase in compound NF rejection. In addition, the presence of cations alone in laboratory-grade water did not have a significant impact on rejection with the exception of the polar compound gemfibrozil. However, when cation concentration in natural waters was increased, a significant decrease in the rejection of EDCs and PhACs was observed. This suggests that the presence of cations may result in a reduction in the association of EDCs and PhACs with NOM.

A method using liquid chromatography/tandem mass spectrometry (LC/MS/MS) was developed for the determination of 17-ethynylestradiol in fish liver; a second method using LC/MS was developed for the determination of carbamazepine, diazepam, simvastatin, and oxybenzone in fish liver. The fish liver samples were extracted and cleaned up by using liquid-liquid extraction and solid-phase extraction before the extracts were analyzed by LC/MS or LC/MS/MS with electrospray negative and positive ionization. Recoveries of the 5 target compounds from spiked catfish liver ranged from 72 2 to 100 3. Limits of quantification for the 5 compounds were between 4.2 and 12.3 ng/g (wet weight). Ten turbot (Pleuronichthys verticalis) liver samples were analyzed; levels of 17-ethynylestradiol, carbamazepine, simvastatin, and oxybenzone were below the detection limits. Diazepam was detected in all 10 fish liver samples at concentrations ranging from 23 to 110 ng/g (wet weight).

Ibuprofen, one of the mostly prescribed nonsteroidal anti-inflammatory drugs (NSAIDs), has been proposed as a topical medication for secondary prevention against skin damage induced by sunburn. The objective of this study was to characterize transmembrane permeation of ibuprofen and sunscreen oxybenzone across poly(dimethyl siloxane) (PDMS) membrane. In vitro diffusion studies were carried out at 37 degrees and 45 degrees C, using a series of ibuprofen and oxybenzone samples, either individually or in combination. Concentrations of ibuprofen and oxybenzone in the receptor compartment for up to 6 h were measured using a high-performance liquid chromatography (HPLC) assay. Ibuprofen and oxybenzone permeated across the PDMS membrane in all diffusion studies. When applied individually, permeation percentages of ibuprofen and oxybenzone ranged from 1.0 to 4.1% and from 13.2 to 25.8%, respectively. When applied in combination, permeation percentages of ibuprofen and oxybenzone were 0.3-1.4% and 7.8-24.3%, respectively. Transmembrane permeation was significantly suppressed when both compounds were present concurrently. High temperature promoted the diffusion process of oxybenzone; a linear correlation was also observed between oxybenzone concentration and its permeation. The proposed permeation enhancement between ibuprofen and oxybenzone was not observed from this study. The potential transdermal interaction and systemic absorption from concurrent application of topical analgesics and sunscreens thus requires further systematic evaluation.

Residual monomers, additives and degradation products from resin-based dental restorative materials eluted into the oral cavity may influence the biocompatibility of these materials. Emphasis has been placed on studies addressing cytotoxic, genotoxic and estrogenic potential of these substances. A prerequisite for analyzing the potential of exposure to eluted compounds from dental materials is reliable quantification methods, both real time and accelerated measurements. The purpose of the present study was to quantify nine eluates; 2-hydroxyethyl methacrylate (HEMA), hydroquinone monomethyl ether (MEHQ), camphorquinone (CQ), butylated hydroxytoluene (BHT), ethyl 4-(dimethylamino)benzoate (DMABEE), triethylene glycoldimethacrylate (TEGDMA), trimethylolpropane trimethacrylate (TMPTMA), oxybenzone (HMBP) and drometrizole (TIN P) leaching from specimens of four commonly used resin-based dental materials in ethanol and an aqueous solution. All analyses were performed by use of GC/MS, each component was quantified separately and the results presented in microg mm(-2). This study has shown that elution from various materials differs significantly, not only in the types of eluates, but also regarding amounts of total and of single components. A high amount of HMBP, a UV stabilizer with potential estrogenic activity, was detected from one material in both solutions.

Benzophenone-type UV filters (BPs) are ubiquitous in the environment. Transformation products (TPs) of BPs with suspected toxicity are likely to be produced during disinfection of water by chlorination. To quickly predict the toxicity of TPs, in this study, a novel two-dimensional liquid-chromatography (2D-LC) method was established in which the objective of the first dimension was to separate the multiple components of the BPs sample after chlorination, using a reversed-phase liquid-chromatography mode. A biochromatographic system, i.e. bio-partitioning micellar chromatography with the polyoxyethylene (23) lauryl ether aqueous solution as the mobile phase, served as the second dimension to predict the toxicity of the fraction from the first dimension on the basis of the quantitative retention-activity relationships (QRARs) model. Six BPs, namely 2,4-dihydroxybenzophenone, oxybenzone, 4-hydroxybenzophenone, 2-hydroxy-4-methoxybenzophenone-5-sulfonic acid, 2,2'-dihydroxy-4,4'-dimethoxybenzophenone and 2,2'-dihydroxy-4-methoxybenzophenone, were the target analytes subjected to chlorination. The products of these BPs after chlorination were directly injected to the 2D-LC system for analysis. The results indicated that most TPs may be less toxic than their parent chemicals, but some may be more toxic, and that intestinal toxicity of TPs may be more obvious than blood toxicity. The proposed method is time-saving, high-throughput, and reliable, and has great potential for predicting toxicity or bioactivity of unknown and/or known components in a complex sample. Background. Oxybenzone, a broad spectrum sunscreen agent widely used in the form of lotion and cream, has been reported to cause skin irritation, dermatitis, and systemic absorption. Aim. The objective of the present study was to formulate oxybenzone loaded microsponge gel for enhanced sun protection factor with reduced toxicity. Material and Method. Microsponge for topical delivery of oxybenzone was successfully prepared by quasiemulsion solvent diffusion method. The effects of ethyl cellulose and dichloromethane were optimized by the 32 factorial design. The optimized microsponges were dispersed into the hydrogel and further evaluated. Results. The microsponges were spherical with pore size in the range of 0.10–0.22 µm. The optimized formulation possesses the particle size and entrapment efficiency of 72 ± 0.77 µm and 96.9 ± 0.52%, respectively. The microsponge gel showed the controlled release and was nonirritant to the rat skin. In creep recovery test it had shown highest recovery indicating elasticity. The controlled release of oxybenzone from microsponge and barrier effect of gel result in prolonged retention of oxybenzone with reduced permeation activity. Conclusion. Evaluation study revealed remarkable and enhanced topical retention of oxybenzone for prolonged period of time. It also showed the enhanced sun protection factor compared to the marketed preparation with reduced irritation and toxicity.

The objective of our present study was to prepare and evaluate gelatin microspheres of oxybenzone to enhance its sunscreening efficacy. The gelatin microspheres of oxybenzone were prepared by emulsion method. Process parameters were analyzed to optimize the formulation. The in vitro drug release study was performed in pH 7.4 using cellulose acetate membrane. Microspheres prepared using oxybenzone:gelatin ratio of 1:6 showed slowest drug release and those prepared with oxybenzone:gelatin ratio of 1:2 showed fastest drug release. The gelatin microspheres of oxybenzone were incorporated in aloe vera gel. Sun exposure method using sodium nitroprusside solution was used for in vitro sunscreen efficacy testing. The formulation C5 containing oxybenzone-bearing gelatin microspheres in aloe vera gel showed best sunscreen efficacy. The formulations were evaluated for skin irritation test in human volunteers, sun protection factor, and minimum erythema dose in albino rats. These studies revealed that the incorporation of sunscreening agent-loaded microspheres into aloe vera gel greatly increased the efficacy of sunscreen formulation more than four times.

Summary Background Benzophenone-3 (BZ-3; 2-hydroxy-4-methoxybenzophenone, oxybenzone) is commonly used to absorb ultraviolet (UV) radiation. BZ-3 penetrates the skin and can be found in the urine. The amount varies between 0·4% and 2%. This seems to be the main metabolic pathway in rats. Objectives To investigate the total amount of BZ-3 excreted in the urine after repeated topical whole-body applications of a sunscreen and to see if UV radiation has any effect on the amount excreted. Methods Twenty-five volunteers applied a commercially available sunscreen containing 4% BZ-3 morning and night for 5 days. Their urine was measured during those 5 days and during a further 5 days after the last application. They were divided into groups A (unirradiated) and B. Group B received UV radiation according to skin type: UVA between 400 and 707 J cm−2, and UVB between 0·46 and 2·0 J cm−2. Among the available cyclodextrins, hydroxypropyl-β-cyclodextrin (HP-β-CD) and especially sulfobutylether-β-cyclodextrin (SBE-β-CD) had the greatest solubilizing activity. Ethanol–water solutions containing oxybenzone free or complexed with HP-β-CD or SBE-β-CD were applied to human skin in Franz diffusion cells and the amount of sunscreen permeated into the different cutaneous compartments was determined by HPLC. As much as 20.5% of the oxybenzone applied dose diffused within the skin tissue after 6 h application. Between 39.4% and 54.9% of the penetrated UV filter was localized in the stratum corneum, with no significant difference between uncomplexed oxybenzone or its complex with HP-β-CD. Conversely, the amount retained in the stratum corneum was markedly decreased (ca. 50%) by complexation with SBE-β-CD. Considerable quantities of oxybenzone accumulated into the viable epidermis (5.7% of the applied dose) and dermis (6.2% of the applied dose) from the preparation containing the free UV filter. The sunscreen penetration to the deeper living layers of the skin was remarkably lower (1.0% and 2.0% of applied dose for epidermis and dermis, respectively) upon application of the sunscreen complexed with SBE-β-CD, whereas HP-β-CD had no effect. In addition, photostability experiments demonstrated that SBE-β-CD complexation did not alter the sunscreen photochemical properties.

It is desirable to minimize skin penetration of some drugs, such as sunscreens and insect repellents. Available in vivo and in vitro data suggest appreciable absorption of some sunscreen agents. The purpose of this study was to investigate the effect of hydroxypropyl-β-cyclodextrin (HPβCD) on the release and permeation of the sunscreen agent 2-hydroxy-4-methoxy benzophenone (oxybenzone). The interaction between oxybenzone and HPβCD was studied in water by phase solubility analysis. The inclusion complex was characterized by thermal analysis and nuclear magnetic resonance spectroscopy. UV transmittance and percent UVA transmittance, as a preliminary measure of sun protection factor (SPF), were determined. In vitro permeation experiments were conducted in Franz-type diffusion cells at 37 °C, using the model membrane poly(dimethyl siloxane) (PDMS) and 4% bovine serum albumin in phosphate buffer solution (pH 7.4) as the receptor phase. HPβCD caused a marked increase in the aqueous solubility of oxybenzone. Data from the phase solubility experiment indicated the formation of 1:1 oxybenzone-cyclodextrin complex. UV transmittance studies indicated that the presence of HPβCD did not suppress the UV absorbing properties of oxybenzone. The release and membrane permeation of oxybenzone was significantly reduced in the presence of equimolar, 2 times molar and 1, 2 and 4% of HPβCD. It is concluded that HPβCD can reduce the release/membrane diffusion of oxybenzone whilst retaining its efficacy as a sunscreen agent. This formulation strategy may be useful in controlling skin penetration of topically applied sunscreens and other chemicals.

The objective of the current study was to investigate the influence of cyclodextrin complexation on the in vivo photoprotective effects of a model ultraviolet (UV) absorber, oxybenzone, and to compare these novel sunscreens to a commercial SPF 30 sunscreen product. Aqueous-based solutions and suspensions containing 2.7 mg/mL oxybenzone and up to 20% (w/w) hydroxypropyl-beta-cyclodextrin (HPCD) were prepared. The sunscreens were applied to the dorsal skin of SKH-1 hairless mice and the animals were exposed to up to two minimal erythemal doses (MEDs) of UV radiation. Control animals received no sunscreen treatment. Lipid damage, as quantified by decreases in the lipid melting temperature of the epidermis, was determined using differential scanning calorimetry immediately after UV exposure. The number of sunburn cells (SBCs) and the extent of edema were measured 24 hours postexposure. Results showed that all oxybenzone-containing formulations decreased the number of SBCs formed, diminished swelling, and reduced the physical damage to the skin structure, in comparison to control. Thus, complexation did not prevent oxybenzone from reacting with light. The 20% HPCD formulation exhibited more substantial photoprotection at UV exposures of one or two MEDs, as evidenced by the formation of fewer SBCs. The 5% HPCD formulation also provided substantial protection against epidermal lipid damage. These studies demonstrate that inclusion of HPCD in sunscreen formulations may enhance the in vivo photoprotective effects of the UV absorbers. No single HPCD-containing sunscreen, however, was found to be equivalent to a commercially available sunscreen product for all biomarkers investigated.

A method has been developed for the trace determination of two sunscreen constituents (2-hydroxy-4-methoxybenzophenone and octyldimethyl-p-aminobenzoic acid) in water samples, which are commonly used in commercial formulations. The method employs solid-phase microextraction (SPME) and gas chromatography with flame ionization and mass spectrometric detection. The technique was developed with headspace and direct sampling in order to demonstrate the applicability of these SPME extraction modes for the identification of these two UV absorbing compounds in waters. The main parameters affecting the SPME process, such as desorption time, extraction time profile, salt additives, pH, and temperature, were investigated. The poly(dimethylsiloxane) 100-microm and polyacrylate 85-microm fiber coatings were found to be the most efficient for the extraction of these compounds from aqueous matrices. Linear calibration curves in the wide range of 10-500 microg/l were obtained for both compounds yielding typical RSD values of 5-9% for both extraction modes. The recoveries were relatively high, 82-98%, with quantitation limits below 1 microg/l. A comparison between the proposed methods and the conventional multiresidue solid-phase extraction revealed that the proposed technique(s) can be reliably used for sunscreen residue measurement in water samples with satisfactory results.

The aim of this research was the development and validation of a high performance liquid chromatography (HPLC) method for the simultaneous and quantitative determination of benzophenone-3, octyl methoxycinnamate and octyl salicylate contained in sunscreen emulsions. The separation and quantitative determination was achieved using a LiChrospher((R)) 100 RP-18 (5 microm) Merck column, a mobile phase constituted of methanol/water (85/15, v/v), a flow-rate of 1.0 mL min(-1) and UV detection at 310 nm. The correlation coefficients and percentage of recovery for benzophenone-3, octyl methoxycinnamate and octyl salicylate were 0.9999 and 99.46%; 0.9995 and 98.85%; 0.9998 and 98.84%, respectively. The relative standard deviations (RSD) for commercial samples were between 0.50 and 0.70%.

The objective of the present study was to determine the effects of hydroxypropyl-beta-cyclodextrin (HPCD) concentration on the transdermal permeation and skin accumulation of a model ultraviolet (UV) absorber, oxybenzone. The concentration of oxybenzone was held constant at 2.67 mg/mL for all formulations, while the HPCD concentrations varied from 0 to 20% (w/w). Complexation of oxybenzone by HPCD was demonstrated by differential scanning calorimetry. A modified Franz cell apparatus was used in the transdermal experiments, with aliquots of the receptor fluid assayed for oxybenzone by high-performance liquid chromatography. From the permeation data, flux of the drug was calculated. Skins were removed from the diffusion cells at specified time points over a 24-hr period and the oxybenzone content in the skin determined. The aqueous solubility of oxybenzone increased linearly with increasing HPCD concentration, following a Higuchi AL-type complexation. The stability constant of the reaction was calculated from the phase-solubility diagram and found to be 2047 M-1. As the concentration of HPCD was increased from 0 to 10%, transdermal permeation and skin accumulation of oxybenzone increased. Maximum flux occurred at 10% HPCD, where sufficient cyclodextrin was added to completely solubilize all oxybenzone. When the concentration of HPCD was increased to 20%, both transdermal permeation and skin accumulation decreased. These data suggest the formation of a drug reservoir on the surface of the skin.
