Abstract: International audience; Après avoir été développée en traitement automatique du langage, l'architecture Transformer s'est démocratisée dans de nombreux domaines de l'apprentissage automatique. Elle a permis de surpasser l'état de l'art dans de nombreuses tâches et a conduit à la création de très grands jeux de données afin d'améliorer les performances des modèles. En multimodalité vision-langage, les résultats encourageants des Transformers favorisent la collecte de données image-texte à très grande échelle. Cependant, il est difficile d'évaluer la qualité de ces nouveaux jeux de données, ainsi que leur influence sur la performance de ces modèles, car notre compréhension des Transformers vision-langage est encore limitée. Nous explorons les études du domaine pour mieux comprendre les processus de collecte des jeux de données, les caractéristiques de ces données et leurs impacts sur les performances des modèles.

Abstract: International audience; In recent years, joint text-image embeddings have significantly improved thanks to the development of transformerbased Vision-Language models. Despite these advances, we still need to better understand the representations produced by those models. In this paper, we compare pre-trained and finetuned representations at a vision, language and multimodal level. To that end, we use a set of probing tasks to evaluate the performance of state-of-the-art Vision-Language models and introduce new datasets specifically for multimodal probing. These datasets are carefully designed to address a range of multimodal capabilities while minimizing the potential for models to rely on bias. Although the results confirm the ability of Vision-Language models to understand color at a multimodal level, the models seem to prefer relying on bias in text data for object position and size. On semantically adversarial examples, we find that those models are able to pinpoint finegrained multimodal differences. Finally, we also notice that fine-tuning a Vision-Language model on multimodal tasks does not necessarily improve its multimodal ability. We make all datasets and code available to replicate experiments.

Abstract: Sex identification from fragmentary archeozoological assemblages is particularly challenging in the Equid family, including for horses, donkeys and their hybrids. This limitation has precluded in-depth investigations of sex-ratio variation in various temporal, geographic and social contexts. Recently, shallow DNA sequencing has offered an economical solution to equine sex determination, even in environments where DNA preservation conditions is not optimal. In this study, we applied state-of-the-art methods in ancient DNA-based equine sex determination to 897 osseous remains in order to assess whether equal proportions of males and females could be found in a range of archeological contexts in France. We found Magdalenian horse hunt not focused on isolated bachelors, and Upper Paleolithic habitats and natural traps equally balancing sex ratios. In contrast, Iron Age sacrificial rituals appeared to have been preferentially oriented to male horses and this practice extended into the Roman Period. During Antiquity, the Middle Ages and the Modern Period, cities emerged as environments largely dominated by horse males. This strong sex-bias was considerably reduced, and sometimes even absent, in various rural contexts. Combined with previous archaeozoological work and textual evidence, our results portray an urban economy fueled by adult, often old, males, and rural environments where females and subadults of both sexes were maintained to sustain production demands., We thank Agnès Orsoni, Michela Leonardi, and Stefanie Wagner for lab assistance and all members of the AGES research team at CAGT for fruitful discussions. 7. Synapse-specific modulation of synaptic responses by brain states in hippocampal pathways

Abstract: Synaptic changes play a major role in memory processes. Modulation of synaptic responses by brain states remains, however, poorly understood in hippocampal networks, even in basal conditions. We recorded evoked synaptic responses at five hippocampal pathways in freely moving male rats. We showed that, at the perforant path to dentate gyrus (PP-DG) synapse, responses increase during wakefulness compared with sleep. At the Schaffer collaterals to CA1 (SC-CA1) synapse, responses increase during non-REM sleep (NREM) compared with the other states. During REM sleep (REM), responses decreased at the PP-DG and SC-CA1 synapses compared with NREM, while they increased at the fornix to nucleus accumbens synapse (Fx-NAc) during REM compared with the other states. In contrast, responses at the fornix to medial PFC synapse (Fx-PFC) and at the fornix to amygdala synapse (Fx-Amy) were weakly modulated by vigilance states. Extended sleep periods led to synaptic changes at PP-DG and Fx-Amy synapses but not at the other synapses. Synaptic responses were also linked to local oscillations and were highly correlated between Fx-PFC and Fx-NAc but not between Fx-Amy and these synapses. These results reveal synapse-specific modulations that may contribute to memory consolidation during the sleep–wake cycle.SIGNIFICANCE STATEMENTSurprisingly, the cortical network dynamics remains poorly known at the synaptic level. We tested the hypothesis that brain states would modulate synaptic changes in the same way at different cortical connections. To tackle this issue, we implemented an approach to explore the synaptic behavior of five connections upstream and downstream the rat hippocampus. Our study reveals that synaptic responses are modulated in a highly synapse-specific manner by wakefulness and sleep states as well as by local oscillations at these connections. Moreover, we found rapid synaptic changes during wake and sleep transitions as well as synaptic down and upregulations after extended periods of sleep. These synaptic changes are likely related to the mechanisms of sleep-dependent memory consolidation.

Abstract: The production of royal pheromones by reproductives (queens and kings) enables social insect colonies to allocate individuals into reproductive and non-reproductive roles. In many termite species, nestmates can develop into neotenics when the primary king or queen dies, which then inhibit the production of additional reproductives. This suggests that primary reproductives and neotenics produce royal pheromones. The cuticular hydrocarbon heneicosane was identified as a royal pheromone in Reticulitermes flavipes neotenics. Here, we investigated the presence of this and other cuticular hydrocarbons in primary reproductives and neotenics of this species, and the ontogeny of their production in primary reproductives. Our results revealed that heneicosane was produced by most neotenics, raising the question of whether reproductive status may trigger its production. Neotenics produced six additional cuticular hydrocarbons absent from workers and nymphs. Remarkably, heneicosane and four of these compounds were absent in primary reproductives, and the other two compounds were present in lower quantities. Neotenics therefore have a distinct ‘royal’ blend from primary reproductives, and potentially over-signal their reproductive status. Our results suggest that primary reproductives and neotenics may face different social pressures. Future studies of these pressures should provide a more complete understanding of the mechanisms underlying social regulation in termites.

Abstract: SummaryIntracellular calcium signaling underlies the astroglial control of synaptic transmission and plasticity. Mitochondria-endoplasmic reticulum contacts (MERCs) are key determinants of calcium dynamics, but their functional impact on astroglial regulation of brain information processing is currently unexplored. We found that the activation of astrocyte mitochondrial-associated CB1 receptors (mtCB1) determines MERCs-dependent intracellular calcium signaling and synaptic integration. The stimulation of mtCB1 receptors promotes calcium transfer from the endoplasmic reticulum to mitochondria through specific mechanisms regulating the activity of the mitochondrial calcium uniporter (MCU) channel. Physiologically, mtCB1-dependent mitochondrial calcium uptake determines the precise dynamics of cytosolic calcium events in astrocytes upon endocannabinoid mobilization. Accordingly, electrophysiological recordings in hippocampal slices showed that genetic exclusion of mtCB1 receptors or specific astroglial MCU inhibition blocks lateral synaptic potentiation, a key example of astrocyte-dependent integration of distant synapses activity. Altogether, these data reveal an unforeseen link between astroglial MERCs and the regulation of brain network functions.

Abstract: La mise en place du programme de recherche pluridisciplinaire d’intérêt régional VOLAILLES, à partir de 2018, a notamment permis de mener une collecte à visée exhaustive des données archéozoologiques produites en région Centre-Val de Loire depuis près de 60 ans. Les informations rassemblées concernent 282 sites archéologiques et couvrent une période allant de la Préhistoire à nos jours. Dans le cadre de ce projet, elles ont permis d’établir une cartographie détaillée de la répartition des différents oiseaux de basse-cour au cours du temps, mais surtout de définir les dynamiques précises d’introduction et de diffusion des poules, oies, canards, pigeons, dindes et paons au sein de l’espace régional et sur le long terme. Cette étude d’ampleur inédite a également permis de synthétiser les données morphologiques, tirées des mensurations osseuses de la poule domestique, et de faire le point sur les connaissances relatives à la volaille à l’échelle française et européenne. The multidisciplinary research programme of regional interest, VOLAILLES, from 2018, has in particular made it possible to carry out an exhaustive collection of archaeozoological data produced in the Centre-Val de Loire region for almost 60 years. The information gathered concerns 282 archaeological sites and covers a period from Prehistory to the present day. Within the framework of this project, it has made it possible to establish a detailed map of the distribution of the various farmyard birds over time, but above all to define the precise dynamics of the introduction and spread of domestic fowl, goose, duck, pigeon, turkey and peacock within the region and over the long term. This unprecedented study also made it possible to synthesise morphological data, drawn from bone measurements of the domestic fowl, and to take stock of knowledge on poultry on a French and European scale.

Abstract: We previously linked TSHZ3 haploinsufficiency to autism spectrum disorder (ASD) and showed that embryonic or postnatal Tshz3 deletion in mice results in behavioral traits relevant to the two core domains of ASD, namely social interaction deficits and repetitive behaviors. Here, we provide evidence that cortical projection neurons (CPNs) and striatal cholinergic interneurons (SCINs) are two main and complementary players in the TSHZ3-linked ASD syndrome. We show that in the cerebral cortex, TSHZ3 is expressed in CPNs and in a proportion of GABA interneurons, while not in cholinergic interneurons or glial cells. TSHZ3-expressing cells, which are predominantly SCINs in the striatum, represent a low proportion of neurons in the ascending cholinergic projection system. We then characterized two new conditional knockout (cKO) models generated by crossing Tshz3flox/flox with Emx1-Cre (Emx1-cKO) or Chat-Cre (Chat-cKO) mice to decipher the respective role of CPNs and SCINs. Emx1-cKO mice show altered excitatory synaptic transmission onto CPNs and plasticity at corticostriatal synapses, with neither cortical neuron loss nor impaired layer distribution. These animals present social interaction deficits but no repetitive patterns of behavior. Chat-cKO mice exhibit no loss of SCINs but changes in the electrophysiological properties of these interneurons, associated with repetitive patterns of behavior without social interaction deficits. Therefore, dysfunction in either CPNs or SCINs segregates with a distinct ASD behavioral trait. These findings provide novel insights onto the implication of the corticostriatal circuitry in ASD by revealing an unexpected neuronal dichotomy in the biological background of the two core behavioral domains of this disorder.

Abstract: Long-term storage of information into memory is supposed to rely on long-term synaptic plasticity processes. Detection of such synaptic changes after training in long-term or reference memory (RM) tasks has yet been scarce, variable and only studied on a short time scale. On the other hand, short-term or working memory (WM) is largely known to depend on persistent neuronal activity or short-term plasticity processes. However, processing information into WM could also involve long-term synaptic changes that could be responsible for the erasure/forgetting of items previously stored in WM playing the role of proactive interference. In order to study long-term synaptic changes associated with RM or WM, we trained chronically implanted rats in three different radial maze tasks: a classical RM task and two WM tasks involving different levels of proactive interference. Synaptic responses at the perforant path to dentate gyrus synapse were recorded on a long-time scale (24h) in freely-moving rats after training in one of these three tasks. We found that consolidation of long-term information leads to a delayed synaptic potentiation, occurring 9 hours after RM training and predicting good behavioral performance on the following day. In contrast, optimal information processing into WM triggers a synaptic depression immediately observed after training and lasting 3 hours, that could act as a mechanism for interference erasure/forgetting.

Abstract: Mitochondrial ATP-synthesis is catalyzed by a F1Fo-ATP synthase, an enzyme of dual genetic origin enriched at the edge of cristae where it plays a key role in their structure/stability. The enzyme’s biogenesis remains poorly understood, both from a mechanistic and a compartmentalization point of view. The present study provides novel molecular insights into this process through investigations on a human protein called TMEM70 with an unclear role in the assembly of ATP synthase. A recent study has revealed the existence of physical interactions between TMEM70 and the subunit c (Su.c), a protein present in 8 identical copies forming a transmembrane oligomeric ring (c-ring) within the ATP synthase proton translocating domain (FO). Herein we analyzed the ATP-synthase assembly in cells lacking TMEM70, mitochondrial DNA or F1 subunits and observe a reciprocal dependence of TMEM70 and Su.c levels, regardless of the status of other ATP synthase subunits or of mitochondrial bioenergetics. Immunoprecipitation and two-dimensional blue-native/SDS-PAGE reveal that TMEM70 forms large oligomers composed of 8 TMEM70 dimers and that TMEM70 oligomers interact with Su.c not yet incorporated into ATP synthase complexes. Moreover, discrete TMEM70-Su.c complexes with increasing Su.c contents can be detected, suggesting a role for TMEM70 oligomers in the gradual assembly of the c-ring. Furthermore, we demonstrate using expansion super-resolution microscopy the specific localization of TMEM70 at the inner cristae membrane, distinct from the MICOS component MIC60. Taken together, our results show that TMEM70 oligomers provide a scaffold for c-ring assembly and that mammalian ATP synthase is assembled within inner cristae membranes.

Abstract: Finite element analysis (FEA) has been widely used to study foot biomechanics and pathological functions or effects of therapeutic solutions. However, development and analysis of such foot modeling is complex and time-consuming. The purpose of this study was therefore to propose a method coupling a FE foot model with a model order reduction (MOR) technique to provide real-time analysis of the dynamic foot function. A generic and parametric FE foot model was developed and dynamically validated during stance phase of gait. Based on a design of experiment of 30 FE simulations including four parameters related to foot function, the MOR method was employed to create a prediction model of the center of pressure (COP) path that was validated with four more random simulations. The four predicted COP paths were obtained with a 3% root-mean-square-error (RMSE) in less than 1 s. The time-dependent analysis demonstrated that the subtalar joint position and the midtarsal joint laxity are the most influential factors on the foot functions. These results provide additionally insight into the use of MOR technique to significantly improve speed and power of the FE analysis of the foot function and may support the development of real-time decision support tools based on this method.

Abstract: International audience; Intracellular calcium signaling underlies the astroglial control of synaptic transmission and plasticity. Mitochondria-endoplasmic reticulum contacts (MERCs) are key determinants of calcium dynamics, but their functional impact on astroglial regulation of brain information processing is unexplored. We found that the activation of astrocyte mitochondrial-associated type-1 cannabinoid (mtCB1) receptors determines MERC-dependent intracellular calcium signaling and synaptic integration. The stimulation of mtCB1 receptors promotes calcium transfer from the endoplasmic reticulum to mitochondria through a specific molecular cascade, involving the mitochondrial calcium uniporter (MCU). Physiologically, mtCB1-dependent mitochondrial calcium uptake determines the dynamics of cytosolic calcium events in astrocytes upon endocannabinoid mobilization. Accordingly, electrophysiological recordings in hippocampal slices showed that conditional genetic exclusion of mtCB1 receptors or dominant-negative MCU expression in astrocytes blocks lateral synaptic potentiation, through which astrocytes integrate the activity of distant synapses. Altogether, these data reveal an endocannabinoid link between astroglial MERCs and the regulation of brain network functions.

Abstract: TP53INP1 is a stress-induced protein, which acts as a dual positive regulator of transcription and of autophagy and whose deficiency has been linked with cancer and metabolic syndrome. Here, we addressed the unexplored role of TP53INP1 and of its Drosophila homolog dDOR in the maintenance of neuronal homeostasis under chronic stress, focusing on dopamine (DA) neurons under normal ageing- and Parkinson’s disease (PD)-related context. Trp53inp1−/− mice displayed additional loss of DA neurons in the substantia nigra compared to wild-type (WT) mice, both with ageing and in a PD model based on targeted overexpression of α-synuclein. Nigral Trp53inp1 expression of WT mice was not significantly modified with ageing but was markedly increased in the PD model. Trp53inp2 expression showed similar evolution and did not differ between WT and Trp53inp1−/− mice. In Drosophila, pan-neuronal dDOR overexpression improved survival under paraquat exposure and mitigated the progressive locomotor decline and the loss of DA neurons caused by the human α-synuclein A30P variant. dDOR overexpression in DA neurons also rescued the locomotor deficit in flies with RNAi-induced downregulation of dPINK1 or dParkin. Live imaging, confocal and electron microscopy in fat bodies, neurons, and indirect flight muscles showed that dDOR acts as a positive regulator of basal autophagy and mitophagy independently of the PINK1-mediated pathway. Analyses in a mammalian cell model confirmed that modulating TP53INP1 levels does not impact mitochondrial stress-induced PINK1/Parkin-dependent mitophagy. These data provide the first evidence for a neuroprotective role of TP53INP1/dDOR and highlight its involvement in the regulation of autophagy and mitophagy in neurons.

Abstract: The chronic relapsing nature of cocaine addiction suggests that chronic cocaine exposure produces persistent neuroadaptations that may be temporally and regionally dynamic in brain areas such as the dopaminergic (DA) system. We have previously shown altered metabolism of DA-target structures, the ventral and dorsal striatum, between early and late abstinence. However, specific changes within the midbrain DA system were not investigated. Here, we investigated potential time- and region-specific changes of activity in the ventral tegmental area (VTA) and the substantia nigra pars compacta (SNc) in rats that had extended or limited access to cocaine and later underwent a period of abstinence. We found that DA activity is decreased only in the VTA in rats with extended access to cocaine, with no changes in SNc DA activity. These changes in VTA DA activity may participate in the negative emotional state and the incubation of drug seeking that occur during abstinence from cocaine.

Abstract: International audience; In filamentous fungi, amyloid signaling sequences allow Nod-like receptors (NLRs) to activate downstream cell-death inducing proteins with HeLo and HeLo-like (HELL) domains and amyloid RHIM and RHIM-related motifs control immune defense pathways in mammals and flies. Herein, we show bioinformatically that analogous amyloid signaling motifs exist in bacteria. These short motifs are found at the N-terminus of NLRs and at the C-terminus of proteins with a domain we term BELL. The corresponding NLR and BELL proteins are encoded by adjacent genes. We identify 10 families of such bacterial amyloid signaling sequences (BASS), one of which (BASS3) is homologous to RHIM and a fungal amyloid motif termed PP. BASS motifs occur nearly exclusively in bacteria forming multicellular structures (mainly in Actinobacteria and Cyanobacteria). We analyze experimentally a subset of seven of these motifs (from the most common BASS1 family and the RHIM-related BASS3 family) and find that these sequences form fibrils in vitro. Using a fungal in vivo model, we show that all tested BASS-motifs form prions and that the NLR-side motifs seed prion-formation of the corresponding BELL-side motif. We find that BASS3 motifs show partial prion cross-seeding with mammalian RHIM and fungal PP-motifs and that proline mutations on key positions of the BASS3 core motif, conserved in RHIM and PP-motifs, abolish prion formation. This work expands the paradigm of prion amyloid signaling to multicellular prokaryotes and suggests a long-term evolutionary conservation of these motifs from bacteria, to fungi and animals.

Abstract: International audience; BACKGROUND: The introduction of viral transneuronal tracers in the toolbox of neural tract-tracing methods has been an important addition in the field of connectomics for deciphering circuit-level architecture of the nervous system. One of the added values of viral compared to conventional retrograde tracers, in particular of rabies virus, is to provide a Golgi staining-like view of the infected neurons, revealing the thin dendritic arborizations and the spines that are major post-synaptic seats of neuronal connections. NEWMETHOD: Here, we comparatively illustrate the characteristics of the labeling obtained in the same model system, the basal ganglia circuitry, by different retrograde viral tracing approaches, using the Bartha strain of pseudorabies virus, the SAD and CVS strains of rabies virus and by the conventional retrograde tracer cholera toxin B. To best contrast the differences in the capacity of these tracers to reveal the dendritic morphology in details, we focused on one population of first-order infected neurons in the striatum, which exhibit high spine density, after tracer injection in the substantia nigra. RESULTS AND CONCLUSION: None of the viruses tested allowed to detect as many neurons as with cholera toxin B, but the SAD and CVS strains of rabies virus had the advantage of enabling detailed Golgi-like visualisation of the dendritic trees, the best numerical detection being offered by the transneuronal rCVS-N2c-P-mCherry while poor labeling was provided by rCVS-N2c-M-GFP. Results also suggest that, besides different viral properties, technical issues about constructs and detection methods contribute to apparently different efficiencies among the viral approaches.

Abstract: International audience; Edited by Ruma Banerjee Heme (iron protoporphyrin IX) is a well-known prosthetic group for enzymes involved in metabolic pathways such as oxygen transport and electron transfer through the mitochondrial respiratory chain. However, heme has also been shown to be an important regulatory molecule (as "labile" heme) for diverse processes such as translation, kinase activity, and transcription in mammals, yeast, and bacteria. Taking advantage of a yeast strain deficient for heme production that enabled controlled modulation and monitoring of labile heme levels, here we investigated the role of labile heme in the regulation of mitochondrial biogenesis. This process is regulated by the HAP complex in yeast. Using several biochemical assays along with EM and epifluorescence microscopy, to the best of our knowledge, we show for the first time that cellular labile heme is critical for the post-translational regulation of HAP complex activity, most likely through the stability of the transcriptional co-activator Hap4p. Consequently, we found that labile heme regulates mitochondrial biogenesis and cell growth. The findings of our work highlight a new mechanism in the regulation of mitochondrial biogenesis by cellular metabolites.

Abstract: International audience; Glial cells have a major role in protecting neurons against various forms of stress. Especially, astrocytes mediate the bulk of glutamate clearance in the brain via specific membrane transporters (GLAST and GLT1), thereby preventing the occurrence of excitotoxic events. Although glutamate-mediated mechanisms are thought to contribute to nigral dopaminergic neuron degeneration in Parkinson's disease, detailed information on the organization of glia in the substantia nigra is still lacking. The present study was performed to provide quantitative information on the organization of astroglia and on the relationships between astrocytes and excitatory synapses in the rat substantia nigra. Using immunolabeling of GLT1 and confocal imaging, we found that the substantia nigra was filled with a dense meshwork of immunoreactive astrocyte processes. Stereological analysis performed on electron microscope images revealed that the density of immunoreactive astrocyte plasma membranes was substantial, close to 1 μm2 /μm3 , in the substantia nigra neuropil, both in the pars compacta and the pars reticulata. Excitatory synapses had on average two thirds of their perimeters free from glia, a disposition that may favor transmitter spillover. The density of glutamatergic synapses, as quantified on confocal images by the simultaneous detection of bassoon and of vesicular glutamate transporter 1 or 2, was very low (0.01 and 0.025 per μm3 in the reticulata and compacta subdivisions, respectively). Thus the ratio of GLT1-expressing glial membrane surface to glutamatergic synapses was very high (40-100 μm2 ), suggesting an efficient regulation of extracellular glutamate concentrations.

Abstract: International audience; Brettanomyces bruxellensis is a unicellular fungus of increasing industrial and scientific interest over the past 15 years. Previous studies revealed high genotypic diversity amongst B. bruxellensis strains as well as strain-dependent phenotypic characteristics. Genomic assemblies revealed that some strains harbour triploid genomes and based upon prior genotyping it was inferred that a triploid population was widely dispersed across Australian wine regions. We performed an intraspecific diversity genotypic survey of 1488 B. bruxellensis isolates from 29 countries, 5 continents and 9 different fermentation niches. Using microsatellite analysis in combination with different statistical approaches, we demonstrate that the studied population is structured according to ploidy level, substrate of isolation and geographical origin of the strains, underlying the relative importance of each factor. We found that geographical origin has a different contribution to the population structure according to the substrate of origin, suggesting an anthropic influence on the spatial biodiversity of this microorganism of industrial interest. The observed clustering was correlated to variable stress response, as strains from different groups displayed variation in tolerance to the wine preservative sulfur dioxide (SO2). The potential contribution of the triploid state for adaptation to industrial fermentations and dissemination of the species B. bruxellensis is discussed.

Abstract: Winemakers are increasingly keen to limit the use of commercial yeasts in order to reduce oenological inputs. The preparation of an indigenous winery-made fermentation starter from grapes called 'pied de cuve' (PdC) is becoming popular, especially in organic farming systems. However, the implementation of the PdC method is still empirical and knowledge is lacking regarding the impact of PdC on S. cerevisiae diversity during alcoholic fermentation. In this study, the impact of PdC on S. cerevisiae genetic diversity and wine composition was evaluated at an industrial scale. Despite very low initial population level of S. cerevisiae before inoculation, the use of PdC was as efficient as Active Dry Yeast in terms of fermentation kinetics and chemical analyses on the resulting wines, except for one modality. At mid-fermentation, the diversity of S. cerevisiae strains was different depending on the PdC used, and was also different from that in the spontaneous fermentation with, in some cases, clonal expansion. Abstract: Pathogenic enterobacteria face various oxygen (O2) levels during intestinal colonization from the O2-deprived lumen to oxygenated tissues. Using Shigella flexneri as a model, we have previously demonstrated that epithelium invasion is promoted by O2 in a type III secretion system-dependent manner. However, subsequent pathogen adaptation to tissue oxygenation modulation remained unknown. Assessing single-cell distribution, together with tissue oxygenation, we demonstrate here that the colonic mucosa O2 is actively depleted by S. flexneri aerobic respiration—and not host neutrophils—during infection, leading to the formation of hypoxic foci of infection. This process is promoted by type III secretion system inactivation in infected tissues, favouring colonizers over explorers. We identify the molecular mechanisms supporting infectious hypoxia induction, and demonstrate here how enteropathogens optimize their colonization capacity in relation to their ability to manipulate tissue oxygenation during infection. Shigella flexneri aerobic respiration depletes mucosal oxygen in the colon to promote infection.

Abstract: Local field potential (LFP) recording is a very useful electrophysiological method to study brain processes. However, this method is criticized for recording low frequency activity in a large area of extracellular space potentially contaminated by distal activity. Here, we theoretically and experimentally compare ground-referenced (RR) with differential recordings (DR). We analyze electrical activity in the rat cortex with these two methods. Compared with RR, DR reveals the importance of local phasic oscillatory activities and their coherence between cortical areas. Finally, we show that DR provides a more faithful assessment of functional connectivity caused by an increase in the signal to noise ratio, and of the delay in the propagation of information between two cortical structures.

Abstract: Study Objectives Paradoxical sleep (PS) has been shown to play an important role in memory, in particular in emotional memory processes. However, the involvement of this particular sleep stage in the systemic consolidation of remote (30 days old) memory has never been tested. We examined whether post-learning PS could play a role in the consolidation of remote fearful memory and in the brain network reorganization that depends on it. Methods Mice were PS-deprived during 6 hours after contextual fear conditioning using an automated method, and their memory was tested either 1 day or 30 days after learning. Brain activity during retrieval was assessed using the immediate early gene Egr1 (Zif 268) as a neuronal marker of activity. Results We found that PS deprivation impaired the recall of remote (30 days)-but not recent (1 day)-memory. We also showed that the superficial layers of the anterior cingulate cortex were significantly less activated during the retrieval of remote memory after PS deprivation. In contrast, after such deprivation, retrieval of remote memory significantly activated several areas involved in emotional processing such as the CA1 area of the ventral hippocampus, the basolateral amygdala and the superficial layers of the ventral orbitofrontal cortex. By performing graph-theoretical analyses, our result also suggests that post-learning PS deprivation could impact the reorganization of the functional connections between limbic areas in order to reduce the level of global activity in this network. Conclusions These findings suggest an important role for PS in the systemic consolidation of remote memory.

Abstract: Vineyards and wineries are ecological habitats that house a diverse community of yeast and bacteria. Grapes and cellars are two sources of S. cerevisiae strains involved in the winemaking process, but the relationship between both S. cerevisiae populations is still poorly understood. Indeed strains in vineyards samples are rarely the same as those isolated from vats. In order to better apprehend this issue, 1374 S. cerevisiae isolates were collected from 193 samples of Merlot grapes obtained across 5 Bordeaux regions and from 11 spontaneously fermenting must of 7 cellars. We obtained 402 different genotypes using 17 microsatellite markers. A first analysis of genotypes indicated that approximately ¼thof isolates presented more than 75% of similarity with commercial yeast starters, suggesting that they escaped from the cellar environment, but presented variations higher that could be detected from the analysis of their industrial batch production. The resulting S. cerevisiae populations of 302 grapes-associated and 225 cellar-associated unique profiles revealed a global low differentiation (Fst=0.036), but with differences from sites to sites. To limit potential geographic or sampling effects, a subset of 72 individuals were selected among the vineyard and cellar isolates, representing 5 cellars and the vines located in their immediate environment. Again a low differentiation was noticed (Fst= 0.03 +/- 0.001). The geneflow between vines and cellars was inferred, using a likelihood approach implemented in the software MIGRATE. Inferences showed that cellar and grape metapopulations present similar theoretical sizes and are connected by asymmetric geneflow: almost 4 times higher in the direction "grapes-to-cellar" than for "cellar-to-grapes" (number of migrants per generation 191 [166 -226] versus 55 [25 - 83], respectively). This reveals that vines and cellars are two compartments of the same ecosystem, which has deep ecological significance.

The different actors from the economic and the tourism fields as well as the cultural heritage one seek to re-anchor tango in Buenos Aires while tango keeps expanding and is globally practiced by musicians and dancers. Is Buenos Aires, as the public actors claim, the world’s capital of tango?; El tango, nacido en la cuenca del Río de la Plata entre Buenos Aires y Montevideo a fines del siglo XIX es una música, una danza que fue difundida muy temprano en los salones de la Belle Epoque. Hoy el tango se baila en todo el mundo y se convierte en uno de los motivos del turismo en Buenos Aires. La inscripción del tango como PCI (patrimonio cultural inmaterial) en 2009 marca la voluntad de legitimar los orígenes del tango y apoyar una actividad turística en pleno apogeo desde la década del 2000. Abstract: International audience; Cultural landscapes, conceived by UNESCO as the combined works of man and nature, became an operating category of World Heritage in 1992. This article analyzes, through a comparative study, the processes of heritage, narrative and tourism in three landscapes in Latin-American cultures : Quebrada de Humahuaca (Argentina), agave landscape and ancient industrial facilities of Tequila (Mexico) and the cultural landscapes of Colombia’s coffee.; Los paisajes culturales, concebidos por la UNESCO como obras combinadas del hombre y de la naturaleza, se convirtieron en una categoría operativa del Patrimonio Mundial en 1992. Este artículo analiza, a través de un estudio comparativo, los procesos de patrimonialización, de storytelling y de turismo en tres paisajes culturales latinoamericanos : la Quebrada de Humahuaca (Argentina), el paisaje de agave e instalaciones industriales antiguas de tequila (México) y los paisajes culturales del café de Colombia.; Les paysages culturels, conçus par l’UNESCO comme des œuvres combinées de l’homme et de la nature, deviennent une catégorie opérante du Patrimoine Mondial en 1992.
