Antibiotic discs were prepared, using several several batches of papers meeting Food and Drug Administration specifications. The analysis of 1,152 zones of inhibition produced showed no performance differences among these batches. Other discs were prepared using papers of different grades. These produced large differences in performance. It is obvious, therefore, that the use of a specified disc paper is necessary for standardizing the performances of the products of various manufacturers and that reproducible results can be attained with the grade of paper specified.

Modified paper chromatography procedures for the analysis of fatty acids produced by anaerobic bacteria are described. Both ethylamine and hydroxylamine derivatives of fatty acids were prepared from inoculated anaerobic culture broth. The derivatives were spotted on chromatography paper and developed with appropriate solvents. Paper chromatography is a valuable alternative to gas liquid chromatography as an ancillary procedure in the identification of anaerobic bacteria in the clinical bacteriology laboratory.

Microbial penetration of sterile packs was studied using single-wrap (two layers) muslin, double-wrap (four layers) muslin, and two-way crepe paper (single layer) to wrap 20 gauze sponges (2 by 2 inch). These packs were stored in the central sterile supply departments of two hospitals and processed for sterility at predetermined intervals. Microorganisms penetrated single-wrap muslin as early as 3 days and double-wrap muslin and single-wrap two-way crepe paper in 21 to 28 days stored in open shelves. The time required for microbial penetration was at least twice as long when closed cabinets were used. Single-wrap muslin packs stored in sealed, impervious plastic bags remained sterile for at least 9 months. All sterile materials in pervious wrappers should be handled as little as possible and then only with extreme care and caution. Closed cabinets offer more protection than open shelves, and single wrappers are not recommended.

Ikawa, Miyoshi (University of New Hampshire, Durham), James W. Morrow, and Sheila J. Harney. Paper chromatographic system for the identification of glycerol in bacterial cell walls. J. Bacteriol. 92:812-814. 1966.-The solvent system consisting of isopropanol-5% boric acid (7:1, v/v) separates glycerol from the other carbohydrate constituents which are found in hydrolysates of bacterial cell walls. This system is useful for the identification of glycerol even when anhydroribitol and rhamnose are both present, and has been found to be applicable on cell wall hydrolysates as well as on synthetic mixtures.

Gram-nagative organisms were tested with commercially available reagentimpregnated strips (PATHO-TEC). Of the 291 strains, all were tested by using seven paper tests and their conventional counterparts. Excellent correlation was obtained with the oxidase, phenylalanine-deaminase, and Voges-Proskauer tests. Indole tests made on liquid medium cultures also gave complete correlation, but some false-negative results with indole-positive Proteus strains were obtained when growth from solid medium was tested by the strip method. Paper strip urease tests were positive within 2 hr with all Klebsiella and some Serratia, Herellea, and Citrobacter strains as well as with Proteus strains. Approximately 15% of citrate strip test results differed from those of the conventional tests, and reproducibility was poor on retest. The lysine decarboxylase strip test showed a number of discrepancies and posed problems of interpretation and readability. Paper reagent strip methods are simple and convenient and merit further development to increase the specificity of those which depend on pH change up to that achieved with the Voges-Proskauer, oxidase, phenylalanine, and indole methods.

These packs were stored on open shelves of a central sterile supply department of a hospital and processed for sterility at weekly intervals. Microorganisms penetrated the double-wrap muslin as early as 28 days, the single-wrap muslin and single-wrap two-way crepe paper combination in 77 days, and the single-wrap muslin and single-layer BAR-BAC combination in 63 days.

A mutant of Gluconobacter cerinus var. ammoniacus, IFO 3267, has been isolated which is deficient with respect to fructose 5-dehydrogenase, the enzyme catalyzing the oxidation of d-fructose to 5-keto-d-fructose (5 KF). Growth of this mutant on fructose as the sole carbon source was impaired unless the culture medium was supplemented with 5 KF. Significant randomization of the 1 and 6 positions of fructose has been reported previously for the wild-type organism during growth on this ketohexose. The pattern of (3)H incorporation into the C5 position of ribonucleic acid-ribose when the mutant was grown on [1-(3)H]fructose and [6-(3)H]fructose in the presence of 5 KF indicated that such randomization did not occur in this variant. The randomization observed in the wild type is, therefore, a consequence of the partial oxidation of fructose to the symmetrical 5 KF intermediate prior to its conversion to pentose. When the mutant was grown on [1-(3)H]fructose in the presence of unlabeled 5 KF, [5-(3)H]fructose appeared in the culture medium. Thus, 5 KF served as the oxidant for the nicotinamide adenine dinucleotide phosphate, reduced form, generated during growth on fructose.

Crude preparations of Desulfotomaculum nigrificans were found to reduce bisulfite to trithionate, thiosulfate, and sulfide. The bisulfite reductase of this organism was partially purified and observed to reduce bisulfite to trithionate as the major product and with thiosulfate and sulfide as minor products. The enzyme exhibited spectral properties identical to the carbon monoxide-reacting pigment (P582) isolated from this organism. It is concluded that the bisulfite reductase of D. nigrificans is P582 and that this organism utilizes a pathway which involves trithionate during the reduction of bisulfite to sulfide.

Uptake of alpha-aminoisobutyric acid (AIB) by a leucine-tyrosine auxotroph of a thermophilic microorganism starved for leucine was studied. AIB was taken up by the cells against a substantial concentration gradient (300:1) and was present there in a free and unchanged form. Various energy inhibitors and sulfhydryl reagents strongly inhibited the accumulation of AIB. AIB uptake obeyed saturation kinetics, and the Lineweaver-Burk plot is characterized by a biphasic curve. AIB most probably shares a common transport system(s) with alanine, serine, and glycine. A mutant defective in l-alanine uptake was isolated by using the suicide effect due to accumulation of the tritiated substrate. The mutant also exhibited impaired transport activity towards AIB, glycine, and l-serine, but not to phenylalanine or valine. The transport of AIB, glycine, l-alanine, and l-serine was induced by d-alanine (5 x 10(-3) M) during growth in a succinate- and ammonia-containing medium. De novo protein synthesis was required for the induction of AIB transport; the induction was inhibited when growth occurred in glucose-containing media. The apparent differential rate of synthesis of the AIB transport system was decreased considerably in glucose-grown cells as compared to succinate-grown cells. A common genetic basis of either the regulatory or structural nature for the transport of AIB, alanine, glycine, and serine in a thermophilic microorganism is suggested.

Two pathways are involved in the fermentation of glutamate to acetate, butyrate, carbon dioxide, and ammonia-the methylaspartate and the hydroxyglutarate pathways which are used by Clostridium tetanomorphum and Peptococcus aerogenes, respectively. Although these pathways give rise to the same products, they are easily distinguished by different labeling patterns of the butyrate when [4-(14)C]glutamate is used as substrate. Schmidt degradation of the radioactive butyrate from C. tetanomorphum yielded equally labeled propionate and carbon dioxide, whereas nearly all the radioactivity of the butyrate from P. aerogenes was recovered in the corresponding propionate. This procedure was used as a test for the pathway of glutamate fermentation by 15 strains (9 species) of anaerobic bacteria. The labeling patterns of the butyrate indicate that glutamate is fermented via the methylaspartate pathway by C. tetani, C. cochlearium, and C. saccarobutyricum, and via the hydroxyglutarate pathway by Acidaminococcus fermentans, C. microsporum, Fusobacterium nucleatum, and F. fusiformis. Enzymes specific for each pathway were assayed in crude extracts of the above organisms. 3-Methylaspartase was found only in clostridia which use the methylaspartate pathway, including Clostridium SB4 and C. sticklandii, which probably degrade glutamate to acetate and carbon dioxide by using a second amino acid as hydrogen acceptor. High levels of 2-hydroxyglutarate dehydrogenase were found exclusively in organisms that use the hydroxyglutarate pathway. The data indicate that only two pathways are involved in the fermentation of glutamate by the bacteria analyzed. The methylaspartate pathway appears to be used only by species of Clostridium, whereas the hydroxyglutarate pathway is used by representatives of several genera.

The major phospholipids of two strains of Bdellovibrio bacteriovorus were characterized. Both strain UKi1, which is obligately saprophytic, and strain UKi2, which is facultatively parasitic, contained phosphatidylethanolamine and phosphatidylglycerol as their major glycerophosphatides. A branched, 15-carbon fatty acid is the major component of these alkali-labile lipids. Absent from UKi1 but present in UKi2 were three alkali-stable lipids (compounds 8, 9, and 11) which appear to be phosphosphingolipids. After acid hydrolysis, both compound 8 and 9 yield the identical phosphorus-containing substance that is water soluble, dipolar ionic, and ninhydrin positive. This substance appears to contain a C-P bond since P(i) could not be released from this substance by treatment with alkaline phosphatase or by very harsh mineral acid treatment. Based on chromatographic comparisons, this phosphonate appears to be a novel lipid constituent. Upon degradation, compound 8 yields 1 mol of dihydroxy long-chain base and compound 9 yields 1 mol of a trihydroxy long-chain base. These bases appear to have a 17-carbon, possibly branched, structure based on gas-liquid chromatography retention times. Degradation of both sphingolipids yields a mixture of hydroxy fatty acids, the major component being a branched, 15-carbon hydroxy acid.

Microbes in ruminal contents incorporated (14)C into cells when they were incubated in vitro in the presence of [(14)C]carboxyl-labeled indole-3-acetic acid (IAA). Most of the cellular (14)C was found to be in tryptophan from the protein fractions of the cells. Pure cultures of several important ruminal species did not incorporate labeled IAA, but all four strains of Ruminococcus albus tested utilized IAA for tryptophan synthesis. R. albus did not incorporate (14)C into tryptophan during growth in medium containing either labeled serine or labeled shikimic acid. The mechanism of tryptophan biosynthesis from IAA is not known but appears to be different from any described biosynthetic pathway. We propose that a reductive carboxylation, perhaps involving a low-potential electron donor such as ferredoxin, is involved.

A procedure using nutritionally minimal media is presented for the plating isolation, but not enumeration, of various catalase-negative microorganisms from soil.

The rigid layer (peptidoglycan) of the wall of the chemolithotroph Ferrobacillus ferrooxidans was isolated after various chemical treatments. The removal of specific components was followed by noting in an electron microscope changes in the appearance of the cell surface. The final peptidoglycan was virtually free from proteins and was sensitive to the action of lysozyme. Results of chemical analyses of acidhydrolyzed peptidoglycan revealed three major amino acids and two amino sugars: glutamic acid, alpha,epsilon-diaminopimelic acid, alanine, glucosamine, and muramic acid in a ratio of 1:1:2.33:062:088.

A procedure for detecting the degradation of amino acids by microorganisms is described, and examples of its use in the characterization of bacteria are presented. The procedure involves inoculating a buffered solution of amino acids with a suspension of bacteria, incubating, chromatographing a sample of the suspension, and detecting degradation in terms of absence of ninhydrin-positive spots.

Rumen microorganisms rapidly metabolized glycine at rates varying from 0.014 to 0.241 mumole of glycine per ml per min. The main metabolic products were carbon dioxide, acetic acid, and ammonia; little glycine was incorporated into bacterial protein. Use of carboxyl or methylene-labeled glycine showed that the carbon dioxide came mainly from the carboxyl of glycine, whereas both carbons of acetic acid were derived partly from the methylene carbon of glycine and partly from carbon dioxide. The ratio of carbon-14 to nitrogen-15 in glycine isolated from the protein of rumen bacteria incubated in the presence of N(15)- and C(14)-labeled glycine indicated that most of the extracellular glycine incorporated into protein was incorporated without intervening deamination.
