It is well known in the wire and cable industry that rodents will chew plastic casing materials. Squirrels in particular are notorious for chewing aerial cable causing damage that results in immediate service outages as well as long term reliability issues. Polymer masterbatch materials containing capsaicin, a known rodent aversive, are commercially available but have been reported to become ineffective over time in the field. Here we report our investigation into the concentration of capsaicin in three commercially available polymer masterbatches (A, B, and C) under simulated weather conditions. Samples were compounded at double the recommended let-down amount and extruded into strips for testing. Samples were weathered using the ASTM G154 cycle 6 standard and capsaicin concentrations were monitored using Ultra Performance Liquid Chromatography. Results from this study showed that sample B had significantly higher capsaicin concentration at the beginning of the study than samples A & C which contained similar levels. Capsaicin levels in samples A & B decreased dramatically after one week of aging and were undetectable after six weeks in the chamber. Both samples A & B also showed appreciable capsaicin depletion after four weeks in ambient lab conditions. Sample C demonstrated better capsaicin retention with 4% of the initial amount of capsaicin remaining at the end of the twelve-week study. However, the concentration still dropped to ng/ml levels, so the effectiveness of the remaining capsaicin as a deterrent is uncertain. Because capsaicin is a key aversive ingredient, our results suggest that the effectiveness of rodent aversive masterbatch materials would be relatively short-lived as compared to the predicted lifetime of a deployed aerial cable. They also demonstrate that the retention of capsaicin within the polymer matrix exposed to environmental conditions cannot be increased by simply increasing the concentration of aversive material. In this study, we evaluated the efficacy, expressed as a mean weight decrease of the whole echinococcal cyst mass, of novel benzimidazole salt formulations in a murine Echinococcus granulosus infection model. BALB/c mice were intraperitoneally infected with protoscoleces of E. granulosus (genotype G1). At 9 months post-infection, treatment with albendazole (ABZ), ricobendazole (RBZ) salt formulations, and RBZ enantiomer salts (R)-(+)-RBZ-Na and (S)-(−)-RBZ-Na formulations were initiated. Drugs were orally applied by gavage at 10 mg kg−1 body weight per day during 30 days. Experimental treatments with benzimidazole sodium salts resulted in a significant reduction of the weight of cysts compared to conventional ABZ treatment, except for the (S)-(−)-RBZ-Na enantiomer formulation. Scanning electron microscopy and histological inspection revealed that treatments impacted not only the structural integrity of the parasite tissue in the germinal layer, but also induced alterations in the laminated layer. Overall, these results demonstrate the improved efficacy of benzimidazole salt formulations compared to conventional ABZ treatment in experimental murine cystic echinococcosis.
