Finding the right college is a challenge for all students, but Black families face additional challenges and questions when navigating the admissions process. Veteran admissions experts Timothy L. Fields and Shereem Herndon-Brown demystify this complexity by advising families on when to begin the process, where to apply, and how to be a competitive applicant. Fields and Herndon-Brown address specific concerns that are not often addressed by school counselors or other resources. They highlight how recent social justice movements and legal cases have amplified the necessity of considering both Historically Black Colleges and Universities and predominantly white institutions, while covering everything from athletic recruitment and artistic talents to financial aid and step-by-step instructions for how to search for colleges and then apply to them. The second edition includes new chapters on: (1) prioritizing students' and parents' mental health; (2) understanding the influence of artificial intelligence tools such as ChatGPT on college essay writing; (3) reviewing recent Supreme Court decisions about race-conscious admission and their likely impact on Black applicants; and (4) navigating the admission process as a transfer applicant. Housing Justice in Unequal Cities is a global research network funded by the National Science Foundation (BCS 1758774) and housed at the Institute on Inequality and Democracy at UCLA Luskin. This open-access volume, co-edited by Ananya Roy and Hilary Malson, brings together movement-based and university-based scholars to build a shared field of inquiry focused on housing justice. Based on a convening that took place in Los Angeles in January 2019, at the LA Community Action Network and at the University of California, Los Angeles, the essays and interventions situate housing justice in the long struggle for freedom on stolen land. Embedded in the stark inequalities of Los Angeles, our work is necessarily global, connecting the city’s Skid Row to the indebted and evicted in Spain and Greece, to black women’s resistance in Brazil, to the rights asserted by squatters in India and South Africa. Learning from radical social movements, we argue that housing justice also requires a commitment to research justice. With this in mind, our effort to build a field of inquiry is also necessarily an endeavor to build epistemologies and methodologies that are accountable to communities that are on the frontlines of banishment and displacement.

In 1986, the International Board of Standards for Training, Performance, and Instruction (IBSTPI) published the first edition of "Instructional Design [ID] Competencies: The Standards." It was the culmination of work that began in 1978. In this third edition, IBSTPI presents its latest view of the competencies of instructional designers. It is a greatly expanded view that reflects the complexities of current practice and technology, theoretical advancements, and the social tenor of the times. The level of proficiency described in the 1986 Competencies was taken to represent an instructional designer who would probably have at least three years of experience in the field beyond entry-level training. The current revision takes this notion considerably further in two ways. First, it discriminates between the essential and the advanced levels. Second, it discriminates between competencies which are universally recognized as required of all practitioners and those which have broad but not universal support. The current edition has added a section called "Professional Foundations." This section explicitly recognizes the importance of a knowledge base for ID and the professional responsibility practitioners have for career-long learning and update of that knowledge base. This recognition of knowledge as a foundation to practice was left implicit in the first version. The current revision has also found a way to recognize the importance of technological competence for the practitioner while continuing to recognize both the volatility and the context-specificity of expertise with any particular technology. The section now called "Implementation and Management" represents a considerable strengthening of the intent of the original. This represents both a better awareness of the role these competencies play in ID and also the increasing importance of ID in the success of knowledge-based enterprises, especially in business environments. Chapters are: (1) "Instructional Design Competence"; (2) "The 2000 IBSTPI Instructional Design Competencies"; (3) "The ID Competencies: Discussion and Analysis"; (4) "The Role and Use of ID Competencies"; (5) "The Competencies and ID Specialization"; and (6) "The Competency Validation Research." Chapter 16 of a book on school law provides a legal overview of employment discrimination on the basis of race. A litany of cases has been adjudicated or are currently in litigation. Selected cases are reviewed briefly and arguments for and against the affirmative action guidelines are presented. The main area of complaint about the guidelines is their controversial quota, or percentage, systems. A number of cases are discussed that concern quotas, among them the Regents of the University of California v. Allen Bakke and Weber v. Kaiser Aluminum and Chemical Corporation. The author concludes that the courts are sending out confusing signals and that a ruling on constitutional grounds under the Fourteenth Amendment would help to clarify the apparent conflicts. Protein-protein interactions are governed by a variety of structural features. The sequence specificities of such interactions are usually easier to establish than the "topological specificities," whereby interactions may be classified based on recognition of distinct three-dimensional structural motifs. Approaches to explore topological specificities have been based primarily on assembly of mini-proteins with well defined secondary, tertiary, and/or quarternary structures. The present chapter focuses on three approaches for constructing topologically well defined mini-proteins: template-assembled synthetic proteins (TASPs), disulfide-stabilized structures, and peptide-amphiphiles (PAs). Specific examples are given for applying each approach to explore topologically-dependent protein-protein interactions. TASPs are utilized to identify a metastatic melanoma receptor that binds to the α1(IV)1263-1277 region of basement membrane (type IV) collagen. A disulfide-stabilized structure incorporating a sarafotoxin (SRT) 6b model was examined as a matrix metalloproteinase (MMP)-3 inhibitor. PAs were developed as (a) fluorogenic triple-helical or polyPro II substrates for MMPs and aggrecanase members of the a disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) family and (b) glycosylated and nonglycosylated ligands for metastatic melanoma cells. Topologically constrained mini-proteins have proved to be quite versatile, helping to define critical primary, secondary, and tertiary structural elements that modulate enzyme and receptor functions. The profiling of protein function is one of the most challenging scientific tasks in the postgenomic age. Traditional protein expression methodologies have focused only on the quantification of proteins under varying conditions or pathologies. Determining the functional differences between protein populations allows for a more accurate view of the outcomes in normal vs diseased proteomes. Because the presence or absence of a protein's function can affect its complex surroundings (consisting of multiple other proteins and substrates), the study of proteome functionality yields information on protein-protein interactions, amplification cascades, signaling pathways, and posttranslational modifications. Of significant interest are proteinases, as proteolysis is responsible for tight regulation of various cellular and tissue processes. Proteinase activities, or lack there of, alter the proteome makeup by regulating other proteins or by generating cleavage products. This chapter describes current proteolytic profiling technologies using activity or target-based formats. Effective solvation of the peptide resin is perhaps the most crucial condition for efficient chain assembly during solid-phase peptide synthesis (SPPS) (1). 1H-, 2H-, 13C-, and 19F-nuclear magnetic resonance (NMR) experiments have shown that, under proper solvation conditions, the linear polystyrene chains of copoly(styrene-l%-divinylbenzene)resin (PS) are nearly as accessible to reagents as if free in solution (2-6). When PS is well solvated, diffusion of reagents is not ratelimiting (7-9). PS swelling tests are thus recommended strongly prior to synthesis (1). The swelling capability of peptidyl-PS increases with increasing peptide length owing to a net decrease in free energy from solvation of the linear peptide chains (10). Chapter XIII of the Part Two of the book "Mama, PhD: Women Write About Motherhood and Academic Life," edited by Elrena Evans and Caroline Grant is presented. It focuses on the life of the author as a mother and her teaching profession. She took her first teaching job at a state university while pregnant with her first child, and she had two more children while working on a third master's degree. At first her life was messy, but later on she was able to manage between teaching and child rearing.

Naturally occurring cyclic depsipeptides, peptides that contain one or more ester bonds in addition to the amide bonds, have emerged as an important source of pharmacologically active compounds or promising lead structures for the development of novel synthetically derived drugs. This class of natural products has been found in many organisms, such as fungi, bacteria, and marine organisms. It is very well known that cyclic depsipeptides and their derivatives exhibit a diverse spectrum of biological activities, including insecticidal, antiviral, antimicrobial, antitumor, tumor-promotive, anti-inflammatory, and immunosuppressive actions. However, they have shown the greatest therapeutic potential as anticancer and particularly antimicrobial agents. Difficulties associated with isolation and purification of larger quantities of this class of natural products and, particularly, unlimited access to their synthetic analogs significantly hampered cyclic depsipeptides exploitation as lead compounds for development of new drugs. As an alternative, total solution or solid-phase peptide synthesis of these important natural products and combinatorial chemistry approaches can be employed to elucidate structure-activity relationships and to find new potent compounds of this class. Native peptides exhibit various biological activities from which the antimicrobial property is one of the most frequently studied. A convenient way of telling whether peptides influence the life cycle of bacteria is the broth microdilution assay. In this measure, growing bacteria are incubated with peptides and growth inhibition is detected with colorimetric methods. Highly charged and protease-sensitive peptides need special considerations in assay design and readout interpretation to reveal the true antimicrobial efficacy and potential utility as human or veterinary therapeutics. Control of gene expression using RNA interference (RNAi) technology constitutes a method of choice for investigating gene function in mammalian cells. However, like most oligonucleotide-based strategies, the major limitation of interfering RNA is their poor cellular uptake due to low permeability of the cell membrane to nucleic acids. Several strategies have been developed to improve delivery of oligonucleotides both in cultured cells and in vivo. So far, there is no universal method for their delivery, as they all present several limitations. Peptide-based strategies have been demonstrated to improve the cellular uptake of nucleic acids both in cultured cell and in vivo. This chapter describes a new peptide-based gene delivery system, MPG, which forms stable noncovalent complexes with oligonucleotides and promotes their delivery into a large panel of cell lines without the need for prior chemical covalent coupling. One of the biggest obstacles for efficient drug delivery is specific cellular targeting. Liposomes have long been used for drug delivery, but do not possess targeting capabilities. This limitation may be circumvented by surface coating of colloidal delivery systems with peptides, proteins, carbohydrates, vitamins, or antibodies that target cell surface receptors or other biomolecules. Each of these coatings has significant drawbacks. One idealized system for drug delivery combines stabilized "protein module" ligands with a colloidal delivery vehicle. Prior studies have shown that peptide-amphiphiles, whereby both a peptide "head group" and a lipid-like "tail" are present in the same molecule, can be used to engineer collagen-like triple-helical or α-helical miniproteins. The tails serve to stabilize the head group structural elements. These peptide-amphiphiles can be designed to bind to specific cell surface receptors with high affinity. Structural stabilization of the integrated targeting ligand in the peptide-amphiphile system equates to prolonged in vivo stability through resistance to proteolytic degradation. Liposomes have been prepared incorporating a melanoma targeting peptide-amphiphile ligand, and shown to be stable with retention of peptide-amphiphile triple-helical structure. Encapsulated fluorescent dyes are selectively delivered to cells. In this chapter we describe the methods and techniques employed in the preparation and characterization of peptide-amphiphiles and peptide-amphiphile-targeted large and small unilamellar vesicles (LUVs and SUVs). Fluorescence microscopy is subsequently utilized to examine the targeting capabilities of peptide-amphiphile LUVs, which should allow for improved drug selectivity towards melanoma vs normal cells based on differences in the relative abundance of the targeted cell surface receptors. Cell-penetrating peptides (CPPs) offer potential as delivery agents for the cellular administration of drugs. However, the pharmacological utility of CPPs that are derived from natural amino acids is limited by their rapid metabolic degradation, low membrane permeability, and toxicity. Various peptidomimetics able to overcome these problems have been described, including peptides formed by d-amino acids and β -peptides. This chapter summarizes the synthesis of γ-proline-derived peptides and polyproline dendrimers for drug delivery applications, and includes descriptions of several modifications in the γ-peptides (mimicking the side chains of the α -amino acids) or modulating the dendrimer surface. 5(6)-Carboxyfluorescein labeling of the aforementioned peptidomimetics for use in cell translocation studies is also described. Furthermore, different protocols for the study of the drug delivery capabilities of these compounds are reviewed, including enzymatic stability studies, cellular uptake measurements by plate fluorimetry and flow cytometry, confocal laser scanning microscopy, and cytotoxicity assays. The β-amyloid peptide aggregates via a nucleation pathway where micellar aggregates propagate to form oligomers (protofibrils), which then polymerize into insoluble fibrils. This fibrillogenic process has been linked to the pathogenesis associated with Alzheimer's disease. One purpose of this chapter is to provide a protocol for reliably producing monomeric Aβas a starting point for physical and biological studies. Many research groups have used organic solvents to disaggregate pre-seeded Aβ in an attempt to acquire monomeric starting materials. Others have used instrumental techniques such as size exclusion chromatography to isolate monomer, structural intermediates, and fibrils and study their affects on A β nucleation. This chapter discusses a modified method of A βpreparation using organic solvents followed by dissolution into aqueous phosphate buffer systems that renders monomeric A β starting solutions for kinetic experiments. Additionally, this chapter details a number of physical techniques such as scanning force microscopy, circular dichroism spectroscopy, transmission electron microscopy, fluorescence spectroscopy, fluorescence photobleaching recovery, and dynamic light scattering, together with physiological techniques such as cell viability assays to characterize Aβ nucleation, aggregation, and fibrillization and the potential biological activity of the various A βparticles. The use of radiolabeled peptides for the diagnosis and therapy of cancer has increased greatly over the last few decades. Skillfully crafted peptide systems, which have high affinity for receptors that are overexpressed in human tumors, offer the potential to improve the characterization, grading, and eventual therapy of human cancer. Robust peptide systems can be labeled with radioactive atoms for imaging purposes using single-photon emission computed tomography and positron emission tomography technologies, or can be labeled with therapeutic nuclides for the efficient killing of tumor cells. This method-based review discusses one such class of receptor-targeted peptides and their radiolabeling with radioactive metals. The somatostatin receptor is upregulated in many types of cancer, and when labeled with a radiometal atom via a bifunctional chelate, can be employed as an agent for the imaging and radiotherapy of cancer. This review will discuss the methods used in the synthesis of the somatostatin peptides, conjugation with bifunctional chelators, and radiolabeling with metal radionuclides. In the analytical approach called chemically targeted identification (CTID), peptides containing phosphorylated or glycosylated serine and threonine underwent β-elimination to produce an unsaturated double bond. Nucleophilic addition of 2-aminoethanethiol to this bond occurred, yielding aminoethylcysteine. Thus, sites containing posttranslational modifications were made susceptible to lysine endopeptidase. Structural information could then be obtained by mass analysis of the proteolytic products. The method was demonstrated by the analysis of β-casein tryptic digest peptides and an O-glycosylated peptide. Contrary to an earlier report, the glycopeptide was found to react with essentially the same kinetics as phosphopeptides. Conversion of all five phosphoserines in residues 15, 17, 18, 19, and 35 in N-terminal tryptic phosphopeptides from bovine β-casein were followed by monitoring the time course of the addition reaction. The chemistry proceeded rapidly at room temperature with a half-reaction time of 15 min. No side reaction products were observed. However, care had to be taken to minimize all counterions, which either precipitate barium or neutralize the base. In the case of 2-aminoethanethiol, excess Ba(OH)2 was needed to offset the effect of the hydrochloride. Alternatively, pre-incubation with base followed by nucleophilic addition was found to work satisfactorily. The use of water-soluble thiol allowed the procedure to be carried out in the solid phase, with a micro pipet greatly facilitating sample cleanup. [ABSTRACT FROM AUTHOR]

Peptide fragments have been widely used in biophysical studies on specific regions of integral membrane proteins. Because of their inherent insoluble nature and tendency to aggregate the preparation of such model peptides is challenging. We have developed synthetic and biosynthetic approaches to prepare peptides containing single and multiple domains of a G protein-coupled receptor. Both the synthetic and biosynthetic products can be isolated by reversed-phase highperformance liquid chromatography to near homogeneity. The biosynthetic product, a fusion protein, is processed by CNBr cleavage to yield the target peptide in various isotopic forms. The final peptides are studied by circular dichroism spectroscopy to determine their secondary structure under a variety of conditions. [ABSTRACT FROM AUTHOR]

Comparative protein profiling is a key approach to understanding the human and other proteomes. Systems-level profiling technologies, such as differential fluorescence twodimensional gel electrophoresis (DIGE), often require the identification of the proteins that are contained within 50 or more spots per gel. A major focus of this chapter therefore is devoted to a general approach for high throughput protein identification that is based on liquid chromatography (LC)/tandem mass spectrometry (MS/MS) analysis of tryptic digests of individual proteins or mixtures of only a few proteins (i.e., as are usually obtained from individual DIGE spots), and that is also applicable to the analysis of complex protein extracts. Additionally, multiple techniques will be described for identifying sites of protein posttranslational modification, with emphasis on phosphorylation and Arg methylation. [ABSTRACT FROM AUTHOR]

High-performance liquid chromatography (HPLC) has proved extremely versatile over the past 25 yr for the isolation and purification of peptides varying widely in their sources, quantity and complexity. This article covers the major modes of HPLC utilized for peptides (size-exclusion, ion-exchange, and reversed-phase), as well as demonstrating the potential of a novel mixed-mode hydrophilic interaction/cation-exchange approach developed in this laboratory. In addition to the value of these HPLC modes for peptide separations, the value of various HPLC techniques for structural characterization of peptides and proteins will be addressed, e.g., assessment of oligomerization state of peptides/proteins by size-exclusion chromatography and monitoring the hydrophilicity/hydrophobicity of amphipathic α-helical peptides, a vital precursor for the development of novel antimicrobial peptides. The value of capillary electrophoresis for peptide separations is also demonstrated. Preparative reversed-phase chromatography purification protocols for sample loads of up to 200 mg on analytical columns and instrumentation are introduced for both peptides and recombinant proteins. A future large-volume liquid scintillator detector such as the proposed 50 kton LENA (Low Energy Neutrino Astronomy) detector would provide a high-statistics measurement of terrestrial antineutrinos originating from β-decays of the uranium and thorium chains. Additionally, the neutron is scattered in the forward direction in the detection reaction $$ \bar v_e + p \to n + e^ + $$. Henceforth, we investigate to what extent LENA can distinguish between certain geophysical models on the basis of the angular dependence of the geoneutrino flux. Our analysis is based on a Monte-Carlo simulation with different levels of light yield, considering an unloaded PXE scintillator. We find that LENA is able to detect deviations from isotropy of the geoneutrino flux with high significance. However, if only the directional information is used, the time required to distinguish between different geophysical models is of the order of severals decades. Nonetheless, a high-statistics measurement of the total geoneutrino flux and its spectrum still provides an extremely useful glance at the Earth's interior. [ABSTRACT FROM AUTHOR]
