Trabalho de Projeto do Mestrado em Engenharia Biomédica apresentado à Faculdade de Ciências e Tecnologia Este projecto compreende o desenvolvimento de hidrogéis altamente elásticos e resistentes que podem ser degradados pela exposição à luz com comprimentos de onda citocompatíveis. Para isso, foram preparados hidrogéis híbridos de rede dupla de alginato e poliacrilamida. A rede dupla consiste em uma rede de alginato, reticulada ionicamente por catiões divalentes (Ca2+), misturada com uma rede de poliacrilamida, reticulada covalentemente por um reticulador sintetizado à base de ONB. O reticulante à base de ONB é o responsável pela sensibilidade do hidrogel à luz, uma vez que ONB é um grupo fotodegradável. A combinação sinérgica destes dois polímeros resulta em hidrogéis com propriedades mecânicas superiores e permite o controle da degradação por exposição à luz.O reticulante foi preparado através de substituições estruturais no anel aromático e na posição benzílica do ONB, o que resulta em um aumento do comprimento de onda máximo de absorção, aumento da cinética de fotoclivagem e produção de subprodutos da reação de degradação menos tóxicos. Além disso, cadeias de PEG foram incorporadas no reticulante, tornando-o solúvel em água. Diferentes pesos moleculares de PEG (600, 1000 e 2000 g/mol) foram usados para estudar a influência do comprimento da cadeia de PEG nas propriedades fotodegradáveis e mecânicas do hidrogel.A síntese dos reticulantes fotodegradáveis (ONB-PEG) foi acompanhada por 1H NMR e FTIR, e as suas propriedades fotodegradáveis foram analisadas por UV-Vis e, novamente, 1H NMR. Os resultados indicaram uma funcionalização bem-sucedida do reticulante e uma degradação efetiva por luz UV a 365 nm.As propriedades mecânicas dos hidrogéis com diferentes pesos moleculares de PEG (H600, H1000 e H2000) foram testadas e comparadas por testes de tração e reologia, e demonstrou-se que o H600 possui o melhor desempenho mecânico.As propriedades fotodegradáveis do hidrogel foram estudadas por testes de reologia e tração. Os resultados mostraram que ao aumentar o tempo de irradiação UV, mais rapidamente a rede de PAAm colapsa, resultando na perda de tenacidade do hidrogel. A análise por SEM da morfologia dos hidrogéis após irradiação UV mostra uma estrutura porosa interligada com inúmeros orifícios circulares, o que confirma o colapso da rede de PAAm, que compromete o desempenho mecânico do gel.O método de deslocamento de solvente foi usado para trocar as moléculas de água nos hidrogéis por moléculas de glicerol para preparar organo-hidrogéis de redes-duplas não secantes. A estratégia proporcionou estabilidade ao longo do tempo, ao mesmo tempo em que melhorou a rigidez e manteve as propriedades fotodegradáveis dos hidrogéis de redes-duplas.Assim, um hidrogel de rede-dupla fotodegradável com propriedades mecânicas aprimoradas e estabilidade ao longo do tempo foi preparado com sucesso, o que expande as aplicações do gel para áreas emergentes como a bioeletrônica. This project comprises the development of highly stretchable and tough hydrogels that can be degraded by its exposure to light with cytocompatible wavelengths. For that purpose, double-network hybrid hydrogels of alginate and polyacrylamide were prepared. The DN consists of an alginate network, ionically crosslinked by divalent cations (Ca²⁺), mixed with a polyacrylamide network, covalently crosslinked by a synthesized ONB-based crosslinker. The ONB based crosslinker is the responsible for the light-sensibility of the hydrogel, since ONB is a photodegradable group. The synergetic combination of these two polymers results in hydrogels with superior mechanical properties and enables the control over degradation upon light exposure.The crosslinker was prepared through structural substitutions on the aromatic ring and at the benzyl position of the ONB unit, which results in a redshift of the maximum absorption wavelengths, an increase of the photocleavage kinetics and the production of less toxic by-products of the degradation reaction. Furthermore, PEG backbone was incorporated on the crosslinkers, making them soluble in water. Different PEG molecular weights (600, 1000 and 2000 g/mol) were used in order to study the influence of the length of the PEG backbone on the photodegradable and mechanical properties of the hydrogel. The synthesis of the photodegradable crosslinkers (ONB-PEG) was followed by 1H NMR and FTIR, and their photodegradable properties were analysed by UV-Vis and, again, 1H NMR. The results indicated a successful functionalization of the crosslinker, and an effective degradation by UV light at 365 nm. The mechanical properties of the hydrogels with different PEG molecular weights (H600, H1000 and H2000) were tested and compared by tensile and rheology tests, demonstrating that H600 has the higher mechanical performance.The photodegradable properties of the hydrogel were studied by rheology and tensile tests. The results showed that by increasing the time of UV irradiation, more rapidly the PAAm network collapsed, resulting in a loss of toughness of the hydrogel. SEM analysis of the morphology of the hydrogels after UV irradiation shows a porous structure interconnected with numerous circular holes, which confirms the collapse of the PAAm network, which compromises the mechanical performance of the gel. The solvent displacement method was used to displace the water molecules in the hydrogels by glycerol molecules to prepare non-drying DN organohydrogels. The strategy provided stability over time, while enhancing the stiffness and maintaining the photodegradable properties of the DN hydrogels.Thus, a photodegradable DN hydrogel with enhanced mechanical properties and stability over time was successfully prepared, which expands the applications of the gel to emerging areas such as bioelectronics.

Doctoral thesis in Chemical Engineering, supervised by Professor Doctor Jorge Fernando Jordão Coelho and Professor Doctor Paulo Jorge da Silva Bártolo, submitted to the Chemical Engineering Department, Faculty of Science and Technology, University of Coimbra. The goal of the present PhD work was the development of new biobased unsaturated polyesters (UPs) to be applied in additive manufacturing technologies, such as microstereo-thermal-lithography (STLG), aiming to produce 3D scaffolds for biomedical applications. UPs are widely used in industrial fields, however their excellent thermomechanical properties and tunability make them excellent candidates for tissue engineering applications. UPs were produced by step-growth polycondensation, using several glycols and diacids of sustainable resources, such as succinic acid (SuCA) and sebacic acid (SeBA). Fumaric acid (FA) was used as the source of double bonds to promote posterior crosslinking reactions with vinylic monomers. In a first approach, the synthesis of UPs using renewable monomers was performed. The properties of the polyesters were extensively studied, namely thermal and mechanical properties, and the final products compared with a commercial resin named Crystic 272, which has been successfully tested on STLG. To understand the polyesters properties, the study of the ratio between diacids and glycols as well as the type of monomers used was performed. By carefully selecting the monomers and the ratio between diacids and glycols it is possible to control the final properties of the polymers. The produced oligomers, characterized by low molecular weights and obtained from aliphatic monomers and natural based diacids, showed high thermal stability, with Ton higher than 330 ºC. Thermal stability remains constant even with the replacement of isophthalic acid by aliphatic diacids. To achieve the proper viscosity to be applied in µSTLG experiments, 2-hydroxyethyl methacrylate (HEMA) and styrene (St) were used as the crosslinking agents. The results were very promising, with the production of two layered scaffolds based on bio resins. Preliminary cell viability tests showed that the synthesized polyesters are biocompatible. The choice of the crosslinking agent also plays a determinant role in the properties of the final unsaturated polyester resins (UPRs). Although St is one the most used unsaturated monomer (UM) to promote the formation of 3D networks, a more friendly UM is desired. Therefore, the prepared biobased UPs were crosslinked with different vinylic monomers, such as HEMA and N-vinyl-2-pyrrolidinone (NVP). Since the final viscosity of the formulations is a crucial parameter, determining the applicability on the µSTLG, the viscosity of the formulations was measured and HEMA was selected as the proper monomer due to its low viscosity and biocompatible character. Photo and thermal polymerization studies were performed to evaluate the properties of the final biobased UPRs. The thermal crosslinked polyesters showed low gel content with variable water contact angles (WCA) with hydrophilic character. Morphology and roughness of the polymers were also studied since these parameters strongly influences the cell adhesion. Posteriorly, a full characterization of the UPRs, cytotoxicity and cell adhesion tests were performed. After 48 h of incubation, for the STLG UPRs, 50-60% of the cells were viable, but both thermal and STLG scaffolds presented high levels of acidity. Alongside with the synthesis and processing of the polyesters, new formulations were tested. Due to the low incorporation of the diacids observed in the previous formulations, new synthesis were performed in two distinct steps, which also resulted in a reduction of the time of the polycondensation reaction. The formulations based on SuCA, diethylene glycol (DG) and other renewable sources (UP7 to UP11s formulations) were prepared in a two-step bulk polycondensation reaction. Glutaric acid (GA) an isosorbide (IS) were some of the chosen renewable sources for the development of the new UPs formulations. Afterwards, the UPs were crosslinked with HEMA. Overall, the developed formulation showed better reaction times, and had the desired properties for the intended final application. GA also proved to be an interesting diacid to be used in the preparation of new UPs for biomedical areas, with high water solubility, high thermal stability and low Tg. Several problems arise concerning the viability of the materials regarding their use in biomedical field. The UPs showed surface acidity and to overcome this issue, several strategies were attempted to eliminate the scaffolds acidity. Reflux treatment with ethanol and oxygen plasma were some of the protocols used to change the polyesters surface. The simplest technique, reflux with ethanol, demonstrate to be adequate in the elimination of surface acidity. Results obtained from plasma treatments suggested that they are not effective to solve the acidity problem and enhance biocompatible character of the polyesters. The µSTLG scaffolds Scf6 and Scf7, based on formulations UP6 and UP7, respectively, were subjected to reflux and showed cell viabilities around 100% after 48 h. The use of renewable sources to obtain fully biobased UPs was accomplished within this PhD work, and the final materials reveal to be strong candidates to be used in the fabrication of 3D scaffolds by µSTLG. The materials exhibit low viscosity, good thermo-mechanical properties, similar to commercial products, high hydrophilicity and cell viability. Ultimately, these polymers can bring new advances and opportunities for the use of these unsaturated polyesters in the preparation of tailored scaffolds for specific uses. RESUMO: O objetivo do presente trabalho de doutoramento consistiu no desenvolvimento de poliésteres insaturados para aplicação em técnicas de prototipagem rápida, nomeadamente em microestereo-termo-litografia (μSTLG). Os materiais desenvolvidos são deste modo processados tendo em vista o desenvolvimento de scaffolds para aplicações biomédicas. Os poliésteres insaturados, regra geral, são extensivamente utilizados em aplicações industriais, contudo as suas propriedades revelam-se de extrema utilidade em áreas como a engenharia de tecidos. A possibilidade de manipular as propriedades destes materiais consoante a aplicação final pretendida torna-os bastante atrativos para o desenvolvimento de scaffolds. Os poliésteres insaturados foram obtidos por policondensação de massa, tendo sido utilizados vários glicóis e diácidos de base natural, tais como o ácido succínico (SuCA) e sebácico (SeBA). O ácido fumárico (FA) foi usado em todas as reações desenvolvidas como a fonte de ligações duplas, de modo a promover a posterior reticulação com monómeros vinílicos. Numa primeira abordagem, a síntese de poliésteres insaturados tendo como materiais de partida monómeros de base natural foi executada. O estudo das propriedades destes poliésteres foi realizado, nomeadamente no que diz respeito às suas propriedades térmicas e mecânicas, e os produtos comparados com uma resina comercial, Crystic 272, previamente testada, com sucesso, no equipamento de STLG. O estudo das propriedades dos poliésteres, a razão entre diácidos e glicóis bem como o tipo de monómeros usados foi fundamental para a compreensão das características dos poliésteres. Uma seleção cuidada dos monómeros e da razão entre diácidos e dióis permite controlar a estrutura final dos polímeros. Os oligómeros obtidos, de baixos pesos moleculares, com base em monómeros alifáticos e diácidos de base natural, mostraram elevada estabilidade térmica, com Ton superiores a 330 C. A substituição do ácido isoftálico, presente na resina comercial, por ácidos alifáticos, não resultou na perda de estabilidade térmica. Os materiais obtidos foram processados por μSTLG, usando 2-hidroxietil metacrilato (HEMA) e estireno (St) como agentes reticulantes, conferindo assim uma viscosidade adequada ao processamento dos scaffolds. Os resultados foram bastante promissores, tendo-se fabricado scaffolds constituídos por duas camadas de resina de caráter bio. Resultados preliminares de viabilidade celular revelaram que os poliésteres sintetizados não apresentaram citotoxicidade. A escolha dos agentes de reticulação determina em grande parte as propriedades finais dos poliéstres insaturados reticulados (UPRs). Embora o St seja dos monómeros insaturados (UMs) mais utilizados para a formação de redes reticuladas 3D, a sua substituição por um composto de caráter bio é desejável. Por conseguinte, os poliésteres insaturados foram posteriormente reticulados com diferentes monómeros vinílicos, tais como o HEMA e N-Vinil-2-pirrolidona (NVP). Uma vez que a viscosidade final das formulações é um parâmetro crucial para a sua aplicação na μSTLG, as viscosidades das formulações desenvolvidas foram determinadas, tendo-se selecionado o HEMA devido à baixa viscosidade conferida ao poliéster, bem como ao seu caráter biocompatível. Estudos de foto reticulação e reticulação térmica foram desenvolvidos de modo a avaliar as propriedades finais das resinas de caráter bio. Os poliésteres reticulados termicamente revelaram baixo teor gel, com ângulos de contacto variáveis, apresentando na sua maioria um caráter hidrofílico. A morfologia das resinas, bem como a determinação da rugosidade, foram analisadas, uma vez que estes parâmetros influenciam a adesão das células na superfície polimérica. Após uma completa caracterização das resinas de poliéster insaturado, testes de citotoxicidade e de adesão celular foram realizados. As amostras preparadas por μSTLG demonstraram viabilidades na ordem dos 50-60% após 48 horas, tendo-se, no entanto, comprovado a acidez dos materiais, tanto dos scaffolds processados por μSTLG como dos UPs preparados por reticulação térmica. Em paralelo à síntese e processamento dos poliésteres, a otimização do processo de policondensação de massa foi efetuada. Tendo-se registado anteriormente a baixa incorporação dos ácidos, as sínteses foram realizadas em dois passos distintos, verificandose a diminuição do tempo de reação de policondensação. As formulações desenvolvidas à base de SuCA, dietilenoglicol (DEG) e outras fontes de origem renovável (formulações UP7 a UP11s) foram preparadas por policondensação em massa. O ácido glutárico (GA) e o isosorbide (IS) foram algumas das matérias-primas de fontes renováveis selecionadas para a síntese das novas formulações que, posteriormente, foram reticulados com HEMA. De um modo geral, verificou-se que as novas formulações possuíam as propriedades desejadas tendo em vista a aplicação final pretendida. O GA mostrou ser um diácido interessante para ser usado na preparação de novos UPs para aplicação biomédica, apresentando elevada solubilidade, estabilidade térmica e baixo Tg. Vários problemas surgiram no que diz respeito à viabilidade dos materiais para aplicação biomédica. A maioria dos UPs sintetizados apresentava uma acidez elevada, que em última instância daria origem à morte celular. De modo a contornar esta limitação, uma série de tratamentos foram efetuados, com o intuito de remover a acidez da superfície dos scaffolds. Tratamento por refluxo com etanol e tratamento por plasma com oxigénio, foram algumas das estratégias usadas para modificação da superfície dos poliésteres. O tratamento mais simples, por refluxo, revelou-se extremamente eficaz na eliminação da acidez dos UPs. Ao passo que o tratamento por plasma não foi eficiente para resolver o problema da acidez. Os materiais processados por μSTLG, Scf6 e Scf7, tratados com refluxo de etanol por 3 h, demonstraram viabilidades superiores a 100 % após 48 h. O desenvolvimento de UPs e UPRs à base de matérias-primas renováveis foi realizado com sucesso, tendo sido demonstrado que são excelentes candidatos para a fabricação de scaffolds tridimensionais por μSTLG. Propriedades tais como baixa viscosidade, performance térmica e mecânica semelhantes às exibidas pelos produtos comerciais, elevada hidrofilicidade, viabilidade e adesão celular caracterizam os polímeros obtidos, reforçando a sua potencialidade em áreas como a engenharia de tecidos.

Trabalho de Projeto do Mestrado Integrado em Engenharia Biomédica apresentado à Faculdade de Ciências e Tecnologia A engenharia de tecidos visa criar modelos in vitro de tecidos e estruturas biológicas funcionais recorrendo a diferentes tecnologias, conforme o objectivo final. Uma estratégia importante para adquirir um bom conhecimento do funcionamento dos órgãos humanos e da sua interações enquanto sistema biológico passa por devenvolver modelos in vitro que representem, da melhor forma possível, a sua anatomia e fisiologia.Desenvolvimento de um modelo 3D in vitro de alvéolo humano por microengenharia. Cavidades com semelhança de forma e dimensional à do órgão a ser modelado foram formadas em policarbonato por microthermoforming assistido por gás. Porosidade destas membranas foi introduzida por ion track etching. e forma a alcançar uma estrutura para a cavidade o mais semelhante possível ao alvéolo humano, diferentes parâmetros para optimização do microthermoforming assistido por gás e do ion track etching foram testados. A avaliação do impacto que a curvatura poderá ter em células epiteliais de pulmão baseou-se na análise de resultados obtidos para proliferação celular, apoptose, densidade celular, adesão célula-substrato, interacção célula-célula e teste de actividade metabólica. A caracterização destes resultados foi efectuada por marcação de estruturas específicas com anticorpos, microscópio electrónico de varrimento (MEV) e por microscopia laser de varrimento (3D profilómetro). Após desenvolvida a plataforma porosa com curvatura, células epiteliais de alvéolo pulmonar humano foram cultivadas. Avaliação de actividades celulares como proliferação, apoptose, densidade celular, adesão celular, actividade metabólica e interacção célula-célula, as quais foram quantificadas. Desenvolvimento e estudo preliminar de uma interface ar-líquido com a curvatura referida, aproximando mais o modelo à barreira ar-sangue responsável pela troca gasosa a nível alveolar. Tissue engineering aims to create functional tissues and in vitro biological structures using different technologies, approaches and to address different goals. An important strategy to have a better understanding of human organs and their interactions is by developing in vitro models that represent their anatomy and physiology. Development of a microengineered 3D in vitro model of human alveoli. Cavities similar to the shape and dimensions of human alveoli were created in polycarbonate by gas-assisted microthermoforming. Porosity in these membranes was introduced by ion track etching. Different parameters were tested to optimise gas-assisted microthermoforming and ion track etching in order to achieve a structure that was as close as possible to the human alveoli' dimensions and shape. To assess the impact of substrate’s curvature on epithelial cells, proliferation, apoptosis, cell density, cell-substrate adhesion, cell-cell interaction and metabolic activity assays were performed. Samples were analysed and characterised by specific immunostainings by respective fluorescent imaging, by scanning electron microscope (SEM) and by laser scanning microscope profilometer. After achieving the desired design for the cavities, lung epithelial cells were cultured and their basic activities were analysed and quantified such as proliferation, apoptosis, cell density, metabolic activity, cell-cell interaction and cell-substrate adhesion. Trial of an Air-Liquid Interface (ALI) and preliminary study of cell barrier intefrity and epithelial cells differentiation when exposed to air, mimicking the air-blood barrier responsible for gas exchange in the human alveoli.

Cardiovascular diseases, which are mostly related to high levels of low density lipoproteins (LDL), are currently one of the major causes of death. Currently, there are various polymer based drugs (hydrogels) capable of lowering the cholesterol levels in the blood, through bile acid complexation. However, all of the commercially available bile acid sequestrants have problems in terms of efficiency. Thus, in order to try to create a more effective drug, hydrogels of 2-hydroxyethyl acrylate (HEA) and ((3- acrylamidopropyl)trimethylammonium chloride) (AMPTMA) were prepared using two different polymerization methods: free radical polymerization (FRP) and supplemental activator and reducing agent atom transfer radical polymerization (SARA ATRP). It was found that the new hydrogels were able to bind the sodium cholate (NaCA) (bile acid model molecule), but showing lower binding capacity than the products of the same category that are already present on the market. The effect of the amount of HEA present on the hydrogels on the binding parameters was also investigated. Generally, the results showed that an increase on the amount of HEA lead to an decrease of the binding capacity of the new BAS. The results also showed that BAS produced using SARA ATRP techniques present a higher binding capacity than the ones produced using FRP techniques. Finally, it was observed that copolymers of PAMPTMA-co-PHEA showed no sign of degradation in solutions that mimic the stomach and intestine environment. Keywords: Hydrogels; Bile acid sequestrants; FRP; SARA ATRP; 2-hydroxyethyl acrylate; ((3- acrylamidopropyl)trimethylammonium chloride) As doenças cardiovasculares, que na sua maioria, estão relacionadas com elevados níveis de lipoproteínas de baixa densidade (colesterol LDL), constituem atualmente uma das principais causas de morte. De momento, já existem vários fármacos baseados em polímeros (hidrogéis) capazes de diminuir os níveis de colesterol no sangue, através da complexação de ácidos biliares. Porém, todos os sequestradores de ácidos biliares(BAS) presentes no mercado apresentam problemas ao nível da eficiência. Assim, de modo a tentar criar um fármaco mais eficaz, recorreu-se à preparação de hidrogéis de acrilato de 2-hidroxietil(HEA) e cloreto de (3-acrilamidopropil)trimetil amónio (AMPTMA), usando dois métodos de polimerização distintos: polimerização radicalar livre (FRP) e polimerização radicalar por transferência de átomo com ativador suplementar e agente de redução (SARA ATRP). Verificou-se que os novos hidrogéis são capazes de se ligar ao sal de sódio do ácido cólico (NaCA) (ácido biliar modelo), mas apresentam uma capacidade de ligação inferior aos produtos da mesma categoria que já se encontram no mercado. Foi ainda investigado o efeito que a quantidade de HEA presente no hidrogel tem nos parâmetros de ligação. De um modo geral, verificou-se que a capacidade de ligação diminui com o aumento da quantidade de HEA nos BAS. Os resultados mostraram ainda que os BAS produzidos usando técnicas de SARA ATRP apresentam uma maior capacidade de ligação que os produzidos por técnicas de FRP. Finalmente, foi observado que o copolímero PAMPTMA-co-PHEA não mostrou sinais de degradação pela solução que simulou o ambiente estomacal e intestinal.

Dissertação de Mestrado Integrado em Engenharia Biomédica apresentada à Faculdade de Ciências e Tecnologia da Universidade de Coimbra. Estima-se que, atualmente, cerca 67 milh˜oes de pessoas em todo o mundo sofram de glaucoma, doen¸ca esta que pode levar `a cegueira quando n˜ao tratada. Os tratamentos dispon´ıveis no mercado possuem efeitos secund´arios adversos e/ou necessitam de destreza manual por parte do paciente para serem administrados corretamente. Nesta perspectiva, e de forma a melhorar a biodisponibilidade do f´armaco no olho atrav´es da administra¸c˜ao de uma dose mais baixa de medicamento, diversos tipos de sistemas de liberta¸c˜ao tˆem vindo a ser desenvolvidos. Devido essencialmente `a sua elevada biocompatibilidade e interesse para aplica¸c˜oes biom´edicas, o poli(´acido l´actico) (PLA) foi o pol´ımero escolhido para o desenvolvimento deste trabalho. Contudo, sendo um material duro e quebradi¸co, a sua utiliza¸c˜ao em diversas aplica¸c˜oes tem sido limitada. Por forma a viabilizar a sua utiliza¸c˜ao na aplica¸c˜ao final, o uso de plastificantes tem-se mostrado como uma promissora alternativa. Assim, o presente trabalho tem como finalidade o estudo do PLA como material pass´ıvel de ser utilizado na entrega de f´armacos anti glaucoma. Para tal, numa primeira etapa, procedeu-se `a s´ıntese e caracteriza¸c˜ao de poli´esteres saturados (SP) para plastifica¸c˜ao do PLA e, utilizando os filmes de PLA desenvolvidos, estudou-se numa segunda fase a capacidade de liberta¸c˜ao controlada de f´armaco anti glaucoma dos mesmos. Diferentes poli´esteres saturados foram sintetizados por policondensa¸c˜ao utilizando, para tal, diferentes glic´ois e ´acidos saturados. Ap´os sintetizados, os diversos SPs foram caracterizados qu´ımica, t´ermica e morfologicamente. Atrav´es das referidas an´alises verificou-se, com sucesso, a obten¸c˜ao de quatro poli´esteres: dois alif´aticos (SP1 e SP3) e dois arom´aticos (SP2 e SP4). De seguida, por solvent casting, procedeu-se `a forma¸c˜ao de filmes de PLA com poli´esteres, utilizando o clorof´ormio como solvente. Para a obten¸c˜ao do filme final, o solvente utilizado na sua prepara¸c˜ao foi evaporado numa estufa a 50 oC. Atrav´es deste m´etodo todos os poli´esteres mostraram ser capazes de formar filmes com o PLA. Contudo, apenas os poli´esteres SP1 e SP3 mostraram ser capazes de formar filmes com o PLA e cujas propriedades mecˆanicas os tornam pass´ıveis de ser estudados para liberta¸c˜ao controlada de f´armaco. Ap´os caracteriza¸c˜ao, os filmes com melhores propriedades - SP1 PLA50 (formula¸c˜ao SP1/PLA na propor¸c˜ao de 50/50), SP3 PLA70 (formula¸c˜ao SP3/PLA na propor¸c˜ao de 30/70) e SP3 PLA50 (formula¸c˜ao SP3/PLA na propor¸c˜ao de 50/50) - foram revestidos com a solu¸c˜ao polim´erica de origem (PLA com SP) ou com um silicone reativo. Posteriormente foram realizados estudos de liberta¸c˜ao de acetazolamida, um f´armaco anti glaucoma. O filme SP3 PLA70, formula¸c˜ao de PLA com poli´ester de ´acido seb´acico e 1,6-hexanodiol, provou ser o mais promissor dos trˆes no que se refere aos perfis de liberta¸c˜ao do f´armaco. No que respeita aos revestimentos estudados, ambos mostraram ser eficazes na diminui¸c˜ao da liberta¸c˜ao do f´armaco. Paralelamente, e numa an´alise comparativa, o mesmo processo de secagem foi realizado `a temperatura ambiente, avaliando assim o impacto da evapora¸c˜ao lenta do clorof´ormio sendo obtidos os mesmos resultados quando em compara¸c˜ao com a evapora¸c˜ao na estufa. Currently, about 67 million people worldwide are affected by glaucoma. Without an appropriate treatment this disease can lead to irreversible blindness. The treatments available in the market present adverse side effects and/or require manual dexterity of the patient to be administered properly. In this regard, and in order to improve the bioavailability of the drug in the eye, a variety of drug delivery systems have been developed. Poly(lactic acid) (PLA) was chosen for the development of this work due to its high biocompatibility and interest for biomedical applications. However, this polymer is a brittle and stiff material, which restricts its use in a variety of applications, namely ocular drug delivery systems. The use of plasticizers aims to improve the mechanical properties in order to meet the specifications of the final application. Thus, the present work aims to study the PLA as a material that can be used in the delivery of anti glaucoma drugs. To this end, in a first stage, different saturated polyesters (SP) were synthesized and characterized in order to plasticize PLA. In a second stage, the developed SP/PLA films were studied by their ability for controlled release of acetazolamide, an anti-glaucoma drug. Different saturated polyesters were synthesized by polycondensation using different saturated glycols and acids. Once synthesized, the different SPs have been characterized chemically, thermally and morphologically. Through these analyzes, it was verified that four polyesters, two aliphatic (SP1 and SP3) and two aromatic (SP2 and SP4), were successfully obtained. Then, using chloroform as solvent, the formation of SP/PLA films was performed by solvent casting. To obtain the final films, the solvent used in their preparation was evaporated in an oven at 50 oC. All the polyesters showed to be able to form films with PLA. However, only SP1 and SP3 polyesters showed to be able to form films with the PLA with mechanical properties of interest for drug controlled release. After characterization, the films with improved properties - SP1 PLA50 (films containing 50/50 wt.% of SP1/PLA), SP3 PLA70 (films containing 30/70 wt.% of SP3/PLA) and SP3 PLA50 (films containing 50/50 wt.% of SP3/PLA) - were coated with the film main solution (SP PLA) or with a silicone. Afterwards, acetazolamide release studies were performed. The SP3 PLA70 films proved to be the most promising of the three films used in this study, in what concerns the drug release profiles. As regards the studied coatings, both showed to be effective in lowering drug release. At the same time, the same drying process was performed at room temperature in order to evaluate the impact of slow evaporation of the chloroform in the film properties. The obtained results were similar to those obtained for the film prepared in the oven at 50 oC.

Tese de doutoramento em Engenharia Química, apresentada ao Departamento de Engenharia Química da Faculdade de Ciências e Tecnologia da Universidade de Coimbra The aim of this work was the development of well-defined co(polymers) based on stimuli-responsive segments to further use as building blocks in the development of nanocarriers for biomedical applications. The synthesis strategy involved the use of reversible deactivation radical polymerization (RDRP) methods, as well as “click” coupling reactions that led to the preparation of well-defined polymers, with controlled molecular weight, narrow molecular weight distributions, and well-defined chain-end functionalities. The homopolymerization studies of the 2-(diisopropylamino)ethyl methacrylate (DPA) by atom transfer radical polymerization (ATRP) involving an eco-friendly catalytic system that uses sodium dithionite (Na2S2O4) as a reducing agent and supplemental activator originated well-defined pH-responsive polymers with controlled properties. The detailed mechanism of such reaction system was investigated and the structure of the initiator, solvent, concentration of the catalyst, and the ratios of Na2S2O4 were adjusted to optimize the polymerization and afford polymers with narrow molecular weight distribution (Đ < 1.15) even at high monomer conversion (~ 90%). The slow and continuous feed of Na2S2O4 solution to the reaction mixture allowed the polymerization to be carried out in the presence of only 100 ppm of CuBr2 when the ligand tris(2-pyridylmethyl)-amine (TPMA) was used. This system was successfully extended to the polymerization of oligo(ethylene oxide) methyl ether methacrylate (OEOMA). The residual metal catalyst concentrations used and non-toxic nature of the Na2S2O4 make this SARA ATRP method very attractive for the well-controlled synthesis of water soluble polymers for biomedical applications. The high conversion and preservation of the chain-end functionality allowed the direct synthesis of POEOMA-b-PDPA block copolymers through one-pot polymerization approach. The pH-dependent self-assembly behavior of these brush-like copolymers in aqueous solutions was studied and the preliminary results suggested that the preparation method plays an important role on the final morphology of the nanoaggregates. Due to the pH critical value of the DPA block, these block copolymers form stable nanostructures at physiological pH, but disassemble at pH < 6.2. Copolymers composed by longer PDPA segments were found to originate larger self-assembled particles with critical micelle concentration (CMC) in the range of 1.0 x 10-3 mg.mL-1. Despite the high versatility of the ATRP method, it is not very efficient in the polymerization of the so called non-activated monomers, namely vinyl acetate (VAc) and N-vinyl caprolactam (NVCL). The reversible addition fragmentation chain transfer (RAFT) was proposed as an alternative RDRP method for the polymerization of such monomers and two new xanthates with alkyne functionalities were designed and synthesized. The kinetic studies revealed that the protected alkyne-terminated RAFT agent (PAT-X1) was able to conduct the RAFT polymerization of both VAc and NVCL in 1,4-dioxane at 60 °C, with a good control over the molecular weight and relatively narrow MW distributions (Đ < 1.4) up to high monomer conversions. The linear evolution of Mn,GPC with conversion as well as the close agreement between Mn,th and Mn,GPC values confirmed the controlled feature of the RAFT system. The poly(N-vinyl caprolactam) (PNVCL) is a temperature-responsive polymer and its solution behaviour was fully investigated under different conditions. The stringent control over the polymer molecular weight allows the development of PNVCL with tunable phase transition temperatures around 37 °C. The deprotection of the alkyne functionality of the polymers synthesized by RAFT, allowed a further copper catalyzed azide–alkyne [3+2] dipolar cycloaddition (CuAAC) to obtain new linear block copolymers. This “click” coupling reaction allowed the conjugation of the alkyne-terminated PNVCL synthesized by RAFT with and azide-terminated POEOMA synthesized by ATRP, originating POEOMA-b-PNVCL copolymers. Such block copolymers are hydrophilic but, due to the temperature responsive nature of the PNVCL segment, they become amphiphilic at temperatures above its low critical solution temperature (LCST) and self-assemble into spherical vesicular aggregates with narrow size distributions. A small drop in the solution temperature caused the disruption of the nanostructures and induced the fast release of nile red (NR), an hydrophobic small molecule used as a model drug. Moreover, the sharp and reversible solution properties of the PNVCL block turn those copolymers interesting candidates for the development of temperature-triggered drug delivery systems (DDS). The CuAAC coupling reaction was extended for the development of responsive polymers with linear-dendritic architectures. The synthesized linear pH-responsive and temperature-responsive polymers were conjugated to polyester dendritic structures based on the monomer 2,2-bis(hydroxymethyl) propionic acid (bis-MPA), functionalized with poly(ethylene glycol) (PEG) segments, to obtain linear dendritic block copolymers (LDBC). This thesis contributed to the development of methods that allow the synthesis of new block copolymers having stimuli-responsive segments, and intended to extend the application of these structures to the development of tailor made nanocarriers to be used as DDS for cancer therapy. Na última década, as resinas de poliéster insaturado (UPRs) têm vindo a tornar-se um material importante na área Biomédica. Estas resinas apresentam excelentes propriedades mecânicas e uma capacidade intrínseca de biodegradação, contudo são constituídas normalmente por monómeros ou agentes de reticulação/monómeros reativos que não são biocompatíveis, o que pode comprometer a sua aplicabilidade em produtos da área da Saúde, como implantes e adesivos, scaffolds e sistemas de libertação controlada de fármacos. Esta tese pretende desenvolver UPRs mais adequados para aplicações biomédicas. Para isso, são usados monómeros de origem renovável na produção dos poliésteres insaturados (UPs), na tentativa de aumentar a sua biocompatibilidade sem comprometer as suas propriedades mecânicas. Para isso, os UPs foram reticulados com 2-hidroxietil metacrilato (HEMA), pois é bastante usado em áreas médicas, como a Oftalmologia e a Engenharia de Tecidos. Os UPs sintetizados foram caracterizados pelas técnicas de FTIR, 1H NMR (composição química), SEC (peso molecular), SDT e DMTA (propriedades termo-mecânicas). As UPRs obtidas através de reticulação térmica e fotoreticulação foram primeiramente analisadas relativamente ao seu teor de gel. Seguidamente, as amostras reticuladas termicamente foram analisadas pelas técnicas SDT e DMTA (propriedades termo-mecânicas), e as suas capacidades de absorção e degradação hidrolítica in vitro também foram avaliadas. Os resultados revelaram sucesso na preparação dos UPs e UPRs compostos por monómeros de fonte renovável, assim como a observação de efeitos interessantes nas suas propriedades, constituindo pontos de partida importantes para a futura otimização dos sistemas e respectiva aplicação no campo biomédico.

Este projecto teve como principal objectivo o desenvolvimento de uma estrutura dendrítica de quarta geração, de base poliéster, funcionalizada com um grupo alcino. O dendron de quarta geração (baseada no composto bis-MPA) foi sintetizado segundo o método double-stage convergent growth, tendo os resultados obtidos sido confirmados por ressonância magnética nuclear e espectrometria de massa. A estrutura sintetizada, com um grupo carboxílico no seu `núcleo', foi sujeita a diversas reacções de funcionalização com terminais alcino, revelando-se, no entanto, infrutíferas. Deste modo, um novo método, através do anidrido carboxílico, foi testado e um dendron de segunda geração, funcionalizado com o grupo alcino, foi sintetizado com sucesso. Com o mesmo intuito de construir uma estrutura de quarta geração, reacções de esterificação e acilação foram executadas, mas os resultados obtidos sugerem que o terminal alcino, presente inicialmente, foi afectado. Concomitantemente, reacções de funcionalização do polietilenoglicol com um terminal alcino foram também realizadas, tendo sido sintetizado com sucesso (após várias tentativas). Embora o objectivo inicial não tenho sido totalmente alcançado, as metodologias abordadas e os resultados obtidos neste projecto, permitem retirar algumas ilações acerca do processo de síntese de estruturas dendríticas. Dissertação de mestrado em Engenharia Química apresentada à Faculdade de Ciências e Tecnologia da Universidade de Coimbra The main goal of this work is to study the thermal dynamics of a lab-scale vessel that can be used for polymerization experiments. It involves a study of the thermal dynamics of a 250 mL jacketed lab-scale glass reactor intended to be used in the future for batch polymerization reaction experiments. A mathematical model is developed and is used to simulate the thermal dynamics of the system. The mathematical model available in the literature for methyl methacrylate (MMA) solution free-radical polymerization is also considered in this simulation study. The numerical implementation of the models has been made in the MATLAB programming language. The simulation results of the lab-scale reactor show the heating and cooling performance, and a feedback control scheme using a split-range control strategy is tested as well to simulate the temperature setpoint tracking of the MMA polymerization mixture in the lab-scale reactor. Neste trabalho é efectuado um estudo térmico de um reactor laboratorial em vidro, dotado de uma camisa de arrefecimento/aquecimento e com uma capacidade de 250 mL. Prentende-se no futuro utilizar este reactor para realizar ensaios experimentais de polimerização. É formulado um modelo matemático do reactor e é simulado numericamente o seu comportamento dinâmico. O estudo desenvolvido inclui também o modelo matemático para a polimerização de metacrilato de metilo (MMA) em solução em modeo descontínuo, disponibilizado na literatura cientÃfica. A implementação numérica dos modelos matemáticos é efectuada com recurso à linguagem de programação MATLAB. Os resultados de simulação demonstram o desempenho térmico do reactor, nomeadamente a sua capacidade de resposta quando é aquecido ou arrefecido. É também apresentado um estudo do reactor sob controlo por realimentação da temperatura da mistura do sistema de polimerização de MMA, em que é adoptada uma estratégia de controlo do tipo split-range.
