Objective: To explore the molecular mechanism of dexmedetomidine (Dex) protecting ischemia/reperfusion (I/R) myocardium. Methods: Wild type mice were grouped into control (Control) group, sham operation (Sham) group, WT I/R group, WT Dex group, and Dectin-1 knock out mice were grouped into KO I/R group and KO Dex group in the in vivo study (n=6). TTC staining was used to determine the myocardial infarction area (%) of the above six groups of mice. HE staining and pathological analyze was used to determine the myocardial injury. Serum TNF-α, IL-6 and IL-10 levels in mice were detected by ELISA. Flow cytometry (FCM) was used to count and sort of infiltrating M2 macrophages and neutrophils in myocardium. qPCR assay was used to determine the Dectin-1 mRNA expression in the above sorted cells. Results: TTC results showed that there was no myocardial infarction in the mice of Control group and Sham group. Compared with the WT I/R group, the infarct volume was significantly lower in WT Dex group, KO I/R group and KO Dex group (P<0.05) . Compared with the KO I/R group, the infarct volume was reduced in KO Dex group (P< 0.05). The results of HE staining showed that the myocardial fibers of the WT I/R group of mice were disorderly arranged, with a large number of broken myocardial fibers, while the myocardial fibers of the WT Dex group, KO I/R group and KO Dex group of mice had a little breakage, the structural damage was not significant, and the myocardial arrangement was relatively neat. ELISA results showed that compared with Sham group, the serum TNF-α and IL-6 levels of the mice in WT I/R group were significantly increased, and the IL-10 level was significantly decreased. Compared with WT I/R group, serum TNF-α and IL-6 levels of the mice in WT Dex group and KO I/R group were significantly decreased, and IL-10 level was significantly increased. Compared with KO I/R group, the serum TNF-α and IL-6 levels of the mice in KO Dex group were significantly decreased, and the IL-10 level was significantly increased (P<0.05). FCM cell counting results showed that compared with Sham group, a large number of M2 macrophages and neutrophils were infiltrated in the myocardium of WT I/R group of mice (P<0.05) . Compared with WT I/R group, the M2 macrophages and neutrophils infiltrated in the myocardium were significantly decreased in WT Dex group, KO I/R group and KO Dex group of mice (P<0.05) . While there was no significant difference between the KO I/R group and the KO Dex group mice (P>0.05) . qPCR results showed that compared with Sham group, the expression level of Dectin-1 mRNA in the myocardial infiltrated M2 macrophages and neutrophils were significantly up-regulated in WT I/ R group of mice (P<0.05) . While compared with WT I/R group, the expression level of Dectin-1 mRNA in Dex group of mice was significantly lower (P<0.05) . Mice in KO I/R group and KO Dex group did not express Dectin-1. Conclusion: The protective mechanisms of Dex preconditioning on I/R injured myocardium involves reducing the infiltrating number of M2 macrophages and neutrophils in myocardium after I/R injury, which may be achieved by inhibiting the expression of Dectin-1. Objective: To investigate the expression of eight subunits of chaperonin containing t-complex 1 (CCT) in thyroid cancer (TC) tissues and their correlation with TC staging, patient prognosis, immune cell infiltration, immune checkpoint expression, and chemotherapy drug sensitivity. Methods: Data from The Cancer Genome Atlas (TCGA) database were used to analyze the expression of each CCT subunit in TC tissues and para-cancerous tissues as well as their relationship with the prognosis of TC patients. The biological function of each CCT subunit was analyzed by the Gene Set Enrichment Analysis (GSEA). Data from the TCGA and TIMER2.0 databases were used to analyze the correlations between the expression of each CCT subunit and the tumor microenvironment, infiltration of immune cells, chemotherapeutic drug sensitivity, and immune checkpoint expression. Results: Database analysis showed high mRNA expression of CCT3, CCT7, and CCT8 and low mRNA expression of CCT1, CCT2, CCT5, and CCT6B in TC tissues (P < 0.01 or P < 0.001). CCT3, CCT6B, and CCT8 mRNA expression were correlated with T staging (P < 0.05 or P < 0.01); CCT6B mRNA expression was associated with lymph node metastasis (P < 0.01); CCT5 mRNA expression was associated with distant metastasis (P < 0.05); and CCT6B may serve as an independent prognostic biomarker for overall survival (OS) in TC patients. CCT subunit expression was primarily enriched in signaling pathways such as graft rejection, complement, and interferongamma.Low expression groups of CCT subunits demonstrated significantly higher TC stromal score, immune infiltration score and composite score than the high expression groups (P < 0.05 or P < 0.01 or P < 0.001), displaying a negative correlation between the expression of each CCT subunit and TC stromal, immune infiltration and composite scores (all P < 0.01). The mRNA expression of each CCT subunit was positively correlated with CD8+ T cell and macrophage infiltration (all P < 0.05), the mRNA expression of most CCT subunits (except CCT6B and CCT7) was positively correlated with neutrophil infiltration (P < 0.05 or P < 0.01 or P < 0.001), whereas the mRNA expression of CCT3, 4, 7, and 8 was negatively correlated with CD4+ T-cell infiltration (all P < 0.05). Compared to the low-expression group, patients with high mRNA expression of most CCT subunit had significantly higher IC50 for chemotherapeutic agents such as sorafenib, levatinib, darafenib, trametinib, vandetanib, and cabozantinib (all P < 0.05 or P < 0.01 or P < 0.001). CCT subunit expression in TC tissues was significantly correlated with the expression of PD-1, PD-L1, PD-L2, CTLA4, CD80, and CD86 (P < 0.05 or P < 0.01 or P < 0.001). Conclusion: CCT complexes may promote TC development by affecting the tumor microenvironment thus impacting patient prognosis, potentially serving as a target for the diagnosis and immunotherapy of refractory TC. Objective: To explore the effect of Annexin A1 (ANXA1) peptidomimetic Ac2-26 on acute kidney injury (AKI) and neutrophil apoptosis in septic rats. Methods: Experimental groups included control group, Ac2-26 group, AKI group, AKI+Ac2-26 group, with 15 rats in each group. After cecal ligation and perforation were used to establish a sepsis-induced AKI model, Ac2-26 was intravenously infused for treatment, once a day for 14 days; after the end, ELISA was used to detect levels of serum creatinine (Scr), urea nitrogen( BUN), IL-1β, IL-6 and TNF-α; HE staining and periodic acid Schiff( PAS) staining were used to observe the pathological changes of rat kidney tissues in each group; immunohistochemical staining was used to detect expression of ANXA1 in kidney tissue of each group of rats; neutrophils were isolated from rat peripheral blood, Giemsa staining and trypan blue staining were used to detect cell purity and viability; Annexin V-FITC/PI double staining method and TUNEL staining were used to determine apoptosis level of neutrophils in each group. Results: Compared with control group, levels of Scr and BUN in serum of rats in AKI group were increased (P<0.05), levels of IL-1β, IL-6 and TNF-α also increased (P<0.05), renal tubules and glomeruli in kidney tissue were both significantly damaged, accompanied by a large number of inflammatory cell infiltration, and pathological score increased (P<0.05), while proportion of ANXA1 positive staining area was decreased (P<0.05); neutrophils identified by Giemsa staining and trypan blue staining had complete morphology and high activity; compared with control group, apoptosis rate of neutrophils in AKI group was decreased( P<0.05), and the positive rate of TUNEL was decreased( P<0.05). Compared with AKI group, levels of Scr and BUN in serum of rats in AKI+Ac2-26 group were decreased (P<0.05), levels of IL-1β, IL-6 and TNF-α also decreased (P<0.05), pathological manifestations of renal tubules and glomeruli in renal tissue were significantly reduced, and pathological score was reduced (P<0.05), while the proportion of ANXA1 positive staining area was increased (P<0.05), at the same time, apoptosis rate of rat neutrophils was increased (P<0.05), positive rate of TUNEL was also increased (P<0.05). Conclusion: ANXA1 peptidomimetic Ac2-26 can increase expression of ANXA1 in kidney tissue of AKI in septic rats, promote neutrophil apoptosis, and have a protective effect on kidney tissue damage in rats caused by sepsis. Objective To analyze the correlation of interferon-induced protein with tetratricopeptide repeats 1 (IFIT1) with immune infiltration and prognosis of ovarian cancer (OC). Methods GEO database was employed to select the tumor immune related genes, and Kaplan-Meier and Prognoscan databases were used to identify the genes significantly associated with OC prognosis. The differential expression of IFIT1 between OC tissue and normal tissue were confirmed with GEPIA, Human Protein Atlas, and Timer databases. The expression level of IFIT1 in OC tissues with different grades and stages were analyzed in the UALCAN database. In addition, based on David database, GO enrichment analysis was used to analyze the interacting genes and proteins of IFIT1 in the String and Genemania databases. Timer and Tisidb databases verified the correlation between IFIT1 and immune cells mutually. Finally, after IFIT1 knockdown xenograft model was constructed based on lentiviral vector of IFIT1 shRNA, the tumor growth was observed in the transplanted nude mice, and infiltration of neutrophils was observed with immunohistochemical assay. Results FIT1, a tumor immune gene, selected from the GEO database, Kaplan-Meier and Prognoscan databases, was negatively correlated with the OC prognosis. GEPIA, Human Protein Atlas, Timer database, and UALCAN database indicated that the expression level of IFIT1 was significantly higher in the OC tissues than the normal ovarian tissues, and had no obvious correlation with tumor stage and grade. Analysis in String, Genemania, and David database found the interaction genes and proteins of IFIT1 were enriched in activation of 2'-5' oligonucleotide synthase, virus defense, and innate immunity, and other processes. The Timer database presented that IFIT1 was positively correlated with the infiltration of CD8+T cells, B cells, dendritic cells, neutrophils, and macrophage in OC, with neutrophils having the most significant correlation. Tisidb and GSCA also confirmed the positive correlation between IFIT1 and neutrophil infiltration in OC (P < 0.05). RT-qPCR and Western blotting confirmed that IFIT1 knockdown resulted in suppressed tumor growth in OC cells xenograft nude mice (P < 0.05) and reduced neutrophil infiltration in the xenograft tissues. Conclusion IFIT1 may influence the malignant progression of OC by promoting neutrophil infiltration.

Bronchial asthma (asthma) is a common clinical respiratory disease mediated by multiple cells and components, and neutrophils, as one of the classic innate immune cells, play an important role in the pathogenesis of asthma. In the inflammatory state, neutrophils release neutrophils extracellular trap (NET) into the periphery through the NETosis procedure. Although NET is a beneficial antibacterial defense structure, its excessive accumulation can trigger different response mechanisms that can also adversely affect the lungs and exacerbate asthma. Based on the basic principles of traditional Chinese medicine constitution, exploring the important role of NET in the pathogenesis of asthma, it holds that the life process theory body-spirit composition theory, endowment inheritance theory, and environmental constraint theory are related to the gradual formation of NET and different stimuli affect the heterogeneity of NET, thereby mediating different biological effects during the onset of asthma. Objective To investigate the correlation and clinical significance of peripheral blood serum amyloid A (SAA) to C-reactive protein (CRP) ratio (SAA/CRP), neutrophils lymphocytes ratio (NLR) and HBVDNA levels and disease severity in patients with chronic hepatitis B (CHB), so as to find out the correlation between SAA and CRP (SAA/CRP), neutrophils lymphocytes ratio (NLR) and hepatitis B virus-deoxyribonucleic acid (HBV-DNA). Methods From June 2020 to June 2022, 100 patients with CHB in our hospital were selected as the study subject and were divided into the mild group (simple CHB, 36 cases), the moderate group (cirrhosis in the compensated stage of hepatitis B, 33 cases) and the severe group (cirrhosis in the decompensated stage of hepatitis B, 31 cases) according to the degree of disease. And at the same time, another 50 healthy volunteers with the same age were selected as the control group. Different disease degrees, general information, serum SAA/CRP and NLR levels in the control group, and serum SAA/CRP and NLR levels in patients with different HBV-DNA loads in the study group were compared respectivly and the correlation between the serum SAA/CRP and NLR levels in patients with CHB and HBV-DNA loads and disease degrees were analyzed. The correlation between the serum SAA/CRP, NLR levels and HBV-DNA load and the degree of disease in CHB patients was analyzed. All patients were treated with the antiviral therapy for 24 weeks, and serum SAA/CRP, NLR levels and the change values of patients with different antiviral efficacies were compared before the treatment and 12 weeks and 24 weeks after the treatment and the value of the change values of serum SAA/CRP, NLR levels before and after the treatment was analyzed so as to predict the efficacy of the therapy. Results Serum SAA/CRP and NLR levels in severe patients > moderate patients > mild patients > control group ( P < 0.05); serum SAA/CRP and NLR levels in high carrier patients > moderate carrier patients > mild carrier patients (P < 0.05); serum SAA/CRP and NLR levels in CHB patients were positively correlated with HBV-DNA load (r = 0.756, 0.709), and disease extent (r = 0.776, 0.745) were positively correlated (P < 0.05);The levels of peripheral blood SAA/CRP and NLR at 12 and 24 weeks after the treatment were higher in non-responders than in responders, and the change values were lower than in responders (P < 0.05); the AUCs predicted by SAA/CRP∆1 and NLR∆1 alone were 0.752, 0.773 for SAA/CRP△1 and NLR△1 alone, and 0.861 for combined prediction of △1; the AUCs for SAA/CRP △2 and NLR△2 alone were 0.796 and 0.819, respectively, and 0.967 for combined prediction of △2, which were greater than those for combined prediction of △1 (P < 0.05). Conclusion SAA/CRP and NLR are significantly correlated with HBVDNA load and disease extent in CHB, and the changes in their levels can be used clinically to assess the disease and predict the prognosis, so as to target the follow-up treatment and improve the prognosis. Objective To explore the factors influencing the severity of acute phase and short-term prognosis of anti-N-methyl-D-aspartate receptor (NMDAR) encephalitis, and search for potential indicators for assessing disease severity and predicting prognosis. Methods A total of 60 patients with anti-NMDAR encephalitis admitted to West China Hospital, Sichuan University from January 2020 to July 2023 were included. According to the modified Rankin Scale (mRS) during the acute phase, they were divided into mild group (mRS ≤ 3 score, n = 25) and severe group (mRS > 3 score, n = 35), as well as good outcome group (mRS ≤ 3 score, n = 32) and poor outcome group (mRS > 3 score, n = 28) based on the mRS score after one month of treatment at our hospital. Univariate and multivariate Logistic regression analyses were used to determine the risk factors of anti-NMDAR encephalitis with severe condition during the acute phase and poor short-term prognosis. Results Logistic regression analysis showed consciousness disorder (OR = 8.975, 95%CI: 2.048-39.327; P = 0.004) and high level of neutrophil percentage-to-albumin ratio (NPAR; OR = 5.004, 95%CI: 1.138-22.011, P = 0.033) were the risk factors of anti-NMDAR encephalitis with severe condition during the acute phase, while high level of serum IgE was the protective factor (OR = 0.994, 95%CI: 0.989-0.999; P = 0.026). Female (OR = 4.380, 95%CI: 1.205-15.929; P = 0.025), consciousness disorder (OR = 5.493, 95%CI: 1.535-19.657; P = 0.009) and high level of NPAR (OR = 2.949, 95%CI: 1.010-8.612; P = 0.048) were the risk factors of anti-NMDAR encephalitis with poor short-term prognosis. Conclusions Consciousness disorder, low serum IgE level and high NPAR are risk factors of anti-NMDAR encephalitis with severe condition during the acute phase, while female, consciousness disorder and high NPAR are risk factors of anti-NMDAR encephalitis with poor short-term prognosis. The purpose of this experiment was to study the effects of adding Modified Zhizhu Powder to diet on the growth performance, blood routine examination, serum biochemical indexes, and intestinal digestive enzymes of weaned rabbits. Ninety 35-day-old healthy New Zealand white rabbits with similar weight were randomly divided into five groups with six replicates in each group and three rabbits in each replicate. The rabbits in the control group were fed with basal diet, while the experimental group was fed with 2‰ enrofloxacin powder and 1‰, 3‰, and 5‰ Modified Zhizhu Powder, respectively. The pre-test period was seven days, and the formal test period was 14 days. The results showed that compared with the control group, the average daily gain of weaned rabbits in the Modified Zhizhu Powder group increased significantly (P<0.05), the diarrhea rate decreased significantly, and there was no death of rabbits. Compared with the control group, the immune organ index of weaned rabbits in the Modified Zhizhu Powder group increased significantly (P<0.05), and it was dose-dependent. Compared with the control group, the number of white blood cells, lymphocytes, and neutrophils in the Modified Zhizhu Powder group decreased significantly (P<0.05). The content of immunoglobulin in 5‰ Modified Zhizhu Powder group increased significantly (P<0.05). Modified Zhizhu Powder can enhance the secretion of lipase, α-amylase, cellulase, and trypsin in small intestine, and promote the absorption of energy in intestinal tract of young rabbits by up-regulating the gene expression level of glucose transporter in small intestine. The study indicates that the addition of Modified Zhizhu Powder in the diet of weaned rabbits can improve the growth performance, enhance the immune function, reduce the inflammatory level of the body, and improve the digestion and absorption capacity of intestinal tract. Objective: To explore the effect of neutrophil elastase (NE) on neutrophil inflammatory recruitment. Methods: Mice bone marrow-derived neutrophils were pretreated with an exogenous elastase inhibitor-sivelestat sodium. The effects of elastase inhibition on the in vivo inflammatory recruitment, chemotaxis, adhesion, cell polarization and spreading of NE were examined by peritonitis adoptive transfer assay, dunn chamber, flow chamber, immunofluorescence staining and spreading assay, respectively. The effects of elastase inhibition on NE phagocytosis and reactive oxygen species (ROS) release capacity were detected by flow cytometry and the luminol chemiluminescence system. Results: Sivelestat sodium pretreatment significantly attenuated neutrophil in vivo inflammatory recruitment (P<0.001); impaired neutrophil perception of chemotaxis in vitro (P<0.05), slowed chemotactic velocity (P< 0.05), and decreased the chemotactic distance (P<0.05); reduced the adhesion of neutrophils to inflamed endothelial cells (P<0.0001) and inhibited the phagocytosis of bacteria by neutrophils (P<0.01); however, there was no significant effect on neutrophil spreading, polarization and ROS. Conclusion: NE inhibition significantly impaired the inflammatory recruitment cascade response and phagocytosis of neutrophils in vitro and in vivo but had no significant effect on the spreading, polarization and ROS release of neutrophils. Objective: To explore the clinical application value of neutrophil extracellular traps (NETs) in the evaluation of psoriasis patients. Methods: Peripheral blood (2 ml) of 63 patients with psoriasis and 27 healthy controls were collected, neutrophils were isolated by density gradient method, and the formation of NETs was observed by immunofluorescence staining. Calculated the proportion and fluorescence intensity of NETs, and analyzed their correlation with clinical classification, severity and serological indica-tors. Skin lesions of 5 patients with psoriasis were collected, and the NETs were observed by immunofluorescence method. Results: Proportion and fluorescence intensity of spontaneous NETs in patients with psoriasis were significantly higher than those in healthy controls, and were positively correlated with the PASI scores. Production of NETs in psoriasis patients with metabolic abnormalities or TNF-α abnormalities was higher than that in psoriasis patients without metabolic abnormalities or TNF-α normalcy. Expressions of NETs in psoriatic lesions were increased. Conclusion: Levels of NETs are correlated with disease severity of psoriasis and patients with metabolic abnormalities, and may be used as a clinical indicator to reflect the inflammatory state of psoriasis and metabolic comorbidity of psoriasis for disease evaluation. Neutrophil extracellular traps play an important role in the body's defense against pathogens, but over-activated neutrophil extracellular traps may damage normal tissues and participate in the occurrence and development of idiopathic inflammatory myopathy associated interstitial lung disease. This review summarized the relationship between neutrophil extracellular traps and idiopathic inflammatory myopathy associated interstitial lung disease as well as the possible pathogenesis found so far, and potential therapeutic targets and directions for future research was discussed based on these findings.

Neutrophils have traditionally been considered to have characteristics of short half-life, specific nuclear morphology and clear particle content. In recent years, it has been found that neutrophils are characterized by heterogeneity and plasticity. Under different physiological and pathological states, neutrophils have phenotypes and functional diversity. Among them, low-density neutrophils (LDNs) are important population of neutrophils, which are composed of a mixed population of activated mature neutrophils and immature neutrophils. LDNs have pro-inflammatory and immunosuppressive functions, which can provide novel treatment strategies for diseases. BACKGROUND: Intervertebral disc degeneration is clinically considered to be the main cause of low back pain, but due to the unclear pathogenesis of intervertebral disc degeneration, there is still a lack of effective means to delay the progression of the disease. Single-cell RNA sequencing technology can amplify and sequence mRNA at the single-cell level, reveal the gene expression intensity of a single cell, discover different cell subsets in tissues according to the heterogeneity of cells, study the pathogenesis of intervertebral disc degeneration at the molecular level, and provide a new theoretical basis for its early diagnosis and treatment. OBJECTIVE: To introduce the basic principles of single-cell RNA sequencing technology and review the research progress of single-cell RNA sequencing technology in intervertebral disc degeneration in recent years. METHODS: A computer was used to search PubMed, Web of Science, CNKI and WanFang databases for the literature published from 2012 to 2022. Key words were “single-cell RNA sequencing, intervertebral disc degeneration, sequencing Technology” in Chinese and English. Duplicate, poor-quality and irrelevant articles were excluded; a total of 70 articles were eventually included. RESULTS AND CONCLUSION: (1) We identified new cell subsets such as homeostatic chondrocytes, hypertrophy chondrocyte-like nucleus pulposus cells and fibrous nucleus pulposus cells, identified the marker genes and transcription factors of these cell subsets, and described the functions, differentiation paths and cell fate of these cell subsets during the development and progression of intervertebral disc degeneration, and proposed the concept of progenitor nucleus pulposus cells. A cell subpopulation with progenitor nucleus pulposus cells properties was identified and its effectiveness in treating intervertebral disc degeneration was verified in mice. (2) Fibro chondrocyte-like annulus fibrosus cells and annulus fibrosus stem cells with both cartilage and fiber properties were identified, and a new type of composite hydrogel was prepared by combining fibrous cartilage inducers silk fibroin and hyaluronic acid in vitro. Experiments in mice demonstrated that this hydrogel could repair both annulus fibrosus tissue and cartilage matrix, and was remarkably effective in the treatment of intervertebral disc degeneration. (3) Regulatory chondrocytes were found in endplate cartilage. Two distinct fates in the progression of intervertebral disc degeneration were analyzed and the differential genes in the two fates were identified. Intercellular communication analysis indicated that regulatory chondrocytes interact with endothelial cells to promote angiogenesis. (4) Immune cells such as macrophages, T cells, myeloid progenitor cells and neutrophils were identified in the degenerated intervertebral disc tissues, demonstrating the existence of immune response during intervertebral disc degeneration. It was found that apolipoprotein induced the polarization of macrophages M1 and M2 subtypes, and this polarization process affected the activity of progenitor nucleus pulposus cells by amplifying the inflammatory response through the MIF signaling pathway. The aim of the experiment was to investigate the effect of VC and bacterial enzyme complex combined with glucose on serum biochemical indexes, immune and antioxidant function of cows with antibiotic poisoning. Twenty cows with antibiotic poisoning, weighing about (250.00±10.23) kg were randomly divided into a control group and an experimental group, with five replicates in each group and two cows in each replicate. The cows in the control group received an intravenous injection of glucose and VC, and the cows in the experimental group received an oral administration of glucose and bacterial enzyme complex for seven days. The results showed that compared with the control group, on the 8th day, the leukocytes, neutrophils, erythrocytes and hemoglobin of cows in the experimental group were significantly decreased ( P<0.05), and the platelets were significantly increased ( P<0.05). The activitiy of ALT and CK, and the concentrations of TBIL, Na, P and BUN in serum significantly decreased ( P<0.05), while the concentrations of ALB and Ca significantly increased ( P<0.05). The concentrations of IgM, IgG, IL-6, IFN-γ, SOD activity and T-AOC in the serum of the cows in the experimental group significantly increased ( P<0.05), while the concentrations of MDA significantly decreased ( P<0.05). On the 8th day of the experiment, there were no significant differences in blood routine, serum biochemistry, antioxidant indexes and immune between the experimental group and healthy cows ( P>0.05). Aim To explore the association between serum neutrophil percentage to albumin ratio (NPAR) level and coronary artery layered plaque in patients with acute coronary syndrome (ACS). Methods 222 patients who underwent coronary angiography (CAG) and optical coherence tomography (OCT) examination of culprit vessels immediately after angiography at the Heart Center of the First Affiliated Hospital of Xinjiang Medical University from January 2018 to January 2023 and diagnosed with ACS were selected as the study subjects, and they were classified into 111 cases of layered plaque and 111 cases of non-layered plaque according to the OCT image results. Serum NPAR level was compared between the layered plaque group and the non-layered plaque group, and the influencing factors of coronary artery layered plaque were analyzed by multivariate Logistic regression analysis. Results Comparing the clinical and biochemical index data between the layered plaque group and the non-layered plaque group, the history of alcohol consumption, lumen stenosis rate, neutrophil percentage level, NPAR level was higher in the layered plaque group than that in the non-layered plaque group, and the albumin level was lower than that in the non-layered plaque group, the differences were statistically significant (P<0. 05). Comparison of the OCT characteristics showed that lipid arcs were larger and fibrous cap thickness was thinner, macrophage infiltration rate was higher, vulnerable plaques, plaque rupture, cholesterol crystals, microvessels, thrombus, calcified nodules, calcified plaques were detected at a higher rate, and fibrous plaques were detected at a lower rate in the layered plaque group, and the differences were all statistically significant (P<0. 05). Spearman correlation analysis showed that the serum NPAR a positive correlation with the rupture of plaques, and the TCFA (r =0. 436 and r =0. 622, respectively, both P<0. 05). Multivariate Logistic regression analysis showed that high level of NPAR was an independent risk factor for the development of layered plaque in culprit vessels in ACS patients (OR=10. 186, 95% CI: 3. 242 ~31. 999, P<0. 001). Conclusion Layered plaque is associated with plaque vulnerability characteristics, and serum NPAR level is elevated in patients with layered plaque in culprit vessels with ACS, which is an independent risk factor for coronary layered plaque in ACS patients. Objective: To investigate the relationship between preoperative trefoil factor 3 (TFF3）, heat shock protein 40 (HSP40）,neutrophil/lymphocyte ratio (NLR) and recurrence after tumor cell reduction surgery (CRS) in patients with epithelial ovarian cancer (EOC）. Methods: 140 EOC patients who received CRS treatment in Department of Gynecology of Nanjing Second Hospital from January2016 to May 2017 were selected, and they were divided into recurrent group and non recurrent group based on whether they recurred after CRS. Serum TFF3, HSP40, and NLR were detected and calculated. Single factor and multivariate logistic regression were used to analyze the influencing factors of recurrence after CRS in EOC patients, and the predictive value of recurrence after CRS in EOC patients were analyzed by Subjects’ Job Characteristics (ROC) Curve. Results: Following up for 5 years, 2 cases were lost and 138 EOC patients had a recurrence rate of 52.90% (73/138) after CRS. Compared with the non recurrence group, the recurrence group had a higher proportion of International Federation of Obstetrics and Gynecology (FIGO) stage IV, chemotherapy courses ≥6, and postoperative residual lesions with a maximum residual tumor diameter of ≤1 cm (R1）. The preoperative serum carbohydrate antigen (CA) 125, TFF3,HSP40, and NLR were increased (P＜0.05）. Multivariate logistic regression analysis showed that tumor diameter ≥3 cm, low differentiation, FIGO stage IV, residual lesions of R1 and TFF3 after surgery, HSP40, and increased NLR were independent risk factors for recurrence after CRS in EOC patients (P＜0.05）. ROC curve analysis showed that the area under the curve (AUC) of preoperative serum TFF3, HSP40, and NLR alone and in combination predicted recurrence after CRS in EOC patients were 0.766, 0.763, 0.765, and0.911, respectively. The AUC of preoperative serum TFF3, HSP40, and NLR combined predicted recurrence after CRS in EOC patients was the highest. Conclusion: Elevated levels of preoperative serum TFF3, HSP40, and NLR are independently associated with postoperative recurrence in EOC patients with CRS, and may serve as auxiliary predictive indicators for postoperative recurrence in EOC patients. Objective To investigate the association of plasma neutrophil-to-lymphocyte ratio (NLR) and lymphocyte-to-monocyte ratio (LMR) with carotid atherosclerosis (CAS) in type 2 diabetes mellitus(T2DM). Methods A total of 211 T2DM patients who attended Department of Endocrinology and Department of Health Care in our hospital from August 2021 to October 2022 were enrolled as experimental group, and according to the presence or absence of CAS, the experimental group was further divided into CAS group with 136 patients and non-CAS group with 75 patients. A total of 47 individuals who underwent physical examination in Physical Examination Center of our hospital during the same period of time were enrolled as control group. The three groups were compared in terms of general information and laboratory examination results including routine blood test and blood biochemistry. The Pearson correlation analysis was used to investigate the correlation of NLR and LMR with the clinical indices of T2DM patients with CAS, and the multivariate logistic regression analysis was used to investigate the influence factors for CAS in T2DM patients. Results There were significant differences between the three groups in age, body mass index, course of disease, systolic pressure, plasma neutrophil count, lymphocyte count, monocyte count, NLR, LMR, fasting blood glucose (FBG), glycosylated hemoglobin (HbA1c), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (χ2=33.651,14.038,F=4.714-68.693,H=6.192-112.809,P0.70, had a relatively high value in evaluating the risk of CAS in T2DM patients Conclusion The level of plasma NLR and LMR are significantly associated with the development of CAS in T2DM patients and can predict the development of T2DM with CAS to a certain extent.

Objective To investigate the predictive value of preoperative hematological inflammatory markers in gastric cancer (GC) with lymphovascular invasion(LVI). Methods A retrospective study was conducted on 341 GC patients who undergoing surgical resection and confirmed by pathological examination in Jiangsu Cancer Hospital from October 2020 to July 2022. The patients were divided into a positive group (n=163) and a negative group (n=178) based on the presence or absence of vascular cancer thrombi in postoperative pathology. Before operation, the following clinical data were collected, including neutrophil to lymphocyte ratio (NLR), platelet-lymphocyte ratio (PLR), lymphocyte-monocyte ratio(LMR), fibrinogen-lymphocyte ratio (FLR) and common tumor markers. The receiver operating characteristic (ROC) curve was used to evaluate the predictive ability of NLR, PLR and FLR for LVI to find the optimal cut-off values. Univariate logistic regression was used to analyze the influencing factors of LVI in GC patients, and a logistic regression model for predicting LVI in GC patients was constructed. Results Compared with those in the patients with LVI negative, the levels of NLR, PLR, FLR and carbohydrate antigen (CA)19-9, the tumor infiltration depth and the rate of lymph node metastasis rate significantly increased in LVI positive patients(P＜0.05). ROC curve analysis showed that the best cutoff values of NLR, PLR and FLR were 1.68, 115.29 and 1.96 respectively. The logistic regression model of LVI in GC patients showed that Logit(P)=-2.117+0.890×X1+0.286×X2+0.917×X3(X1=PLR, X2=cT, X3=lymph node metastasis) had a good model fit(χ=58.405, P＜0.01), with the prediction accuracy of 67.26% and an AUC of 0.733 (95%CI: 0.680-0.786, P＜0.01), and this model analysis suggested that PLR(＞115.29, OR=2.436, 95%CI: 1.386-4.282, P=0.002), cT stage (OR=1.331, 95%CI: 1.031-1.749, P=0.030) and lymph node metastasis(OR=2.503, 95%CI: 1.507-4.156, P＜0.001) were the independent risk factors for LVI in patients with GC. Conclusion Preoperative monitoring of PLR level has a certain predictive value in evaluating the occurrence of LVI in GC patients, and can be used as a supplementary indicator to judge the formation of LVI.

Objective To investigate the value of plasma neutrophil-to-lymphocyte ratio (NLR) combined with global registry of acute coronary events (GRACE) score in evaluating the long-term prognosis of patients with non-ST-elevation myocar-dial infarction (NSTEMI). Methods An analysis was performed for 278 patients with NSTEMI who were hospitalized in The Affiliated Hospital of Qingdao University in 2018 and received long-term follow-up after discharge, and related clinical data were collected to calculate plasma NLR and GRACE score. With the onset of major adverse cardiovascular events (MACE) during follow-up as the endpoint, the patients were divided into MACE group and non-MACE group, and clinical data were compared between the two groups. A multivariate logistic regression analysis was used to investigate the risk factors for MACE, and a receiver operating characteristic (ROC) curve analysis was performed for plasma NLR and GRACE score to investigate their value in eva-luating patient prognosis. The multivariate Cox regression model was used to explore the impact of plasma NLR and GRACE score on the specific survival time of patients. Objective To investigate the association of neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and systemic immune-inflammation index (SII) with the degree of adenoid hypertrophy in children. Methods This study was conducted for 181 children who attended Department of Otorhinolaryngology & Head and Neck Surgery in our hospital and were diagnosed with adenoid hypertrophy by fibrolaryngoscopy from February 2020 to February 2021. According to the type and degree of adenoid hypertrophy, the children were divided into pathological adenoid hypertrophy (degree Ⅲ-Ⅳ hypertrophy) group and physiological adenoid hypertrophy (degree Ⅰ-Ⅱhypertrophy) group (group A), and the pathological adenoid hypertrophy group was further divided into degree Ⅲ hypertrophy group (group B) and degree Ⅳ hypertrophy group (group C). Objective: To explore the relationship between neutrophil/lymphocyte ratio (NLR), platelet/lymphocyte ratio (PLR) and the treatment effect of iodine 131 (131Ⅰ) thyroidectomy after operation for differentiated thyroid cancer (DTC) and the predictive value of the effect of 131I thyroidectomy after DTC operation. Methods: 150 DTC patients who underwent thyroidectomy and underwent 131Ⅰ thyroidectomy after operation in Xuzhou Hospital Affiliated to Jiangsu University from March 2019 to December 2021 were selected as the study subjects, and the patients were divided into successful thyroidectomy group (107 cases) and unsuccessful thyroidectomy group (43 cases) according to the effect of 131Ⅰ thyroidectomy, neutrophil count, platelet count, lymphocyte count were obtained by blood routine examination and NLR and PLR were calculated, and the NLR and PLR were compared between the two groups. The influencing factors of 131Ⅰ thyroidectomy efficacy was analyzed by single factor and multiple factor logistic regression model, and the predictive value of NLR and PLR on the efficacy of 131Ⅰ thyroidectomy was analyzed by receiver operating characteristic curve (ROC). Results: The NLR and PLR in the successful thyroidectomy group were lower than those in the unsuccessful thyroidectomy group (P＜0.05), single factor analysis showed that the levels of thyroid stimulating hormone (TSH) was higher than that in the unsuccessful thyroidectomy group, thyroglobulin (Tg) in the successful thyroidectomy group was lower than those in the unsuccessful thyroidectomy group, and the maximum diameter of the lesion in the successful thyroidectomy was smaller than that in the unsuccessful thyroidectomy group (P＜0.05) .Multivariate logistics regression analysis showed that, high NLR, PLR and Tg were independent risk factors for failure of 131Ⅰ thyroidectomy (P＜0.05) .The area under ROC curve (AUC) of NLR, PLR and combined detection predicting the efficacy of 131Ⅰ thyroidectomy was 0.760, 0.732, 0.829respectively, and the AUC of NLR and PLR combined detection was higher than that of the two alone. Conclusion: High NLR and PLR are independent risk factors for unsuccessful 131Ⅰ thyroidectomy after DTC operation, and early detection of NLR and PLR levels has good predictive value for the efficacy of 131Ⅰ thyroidectomy after DTC operation. Objective: To investigate the predictive value of quantitative parameters of contrast-enhanced ultrasound combined with carcinoembryonic antigen (CEA) and neutrophil/lymphocyte ratio (NLR) for recurrence and metastasis of breast cancer after modified radical mastectomy. Methods: 120 patients with breast cancer who underwent modified radical mastectomy in our hospital were selected from December 2020 to January 2022. All patients underwent breast CEUS to obtain contrast-enhanced images and quantitative CEUS parameters. Serum CEA levels and NLR were measured, and the incidence of recurrence and metastasis after surgery were statistically analyzed. The ROC curve of subjects was used to analyze the value of quantitative parameters of contrast-enhanced ultrasound combined with CEA and NLR in predicting the recurrence and metastasis of breast cancer after modified radical mastectomy. Results:Compared the preoperative sonographic features of the two groups, the recurrent metastasis group showed slightly higher enhancement intensity, most of which were hyperenhanced or globally enhanced, and the contrast agent distribution was not yet uniform. The results showed that 18 patients experienced postoperative recurrence and metastasis (in the recurrence and metastasis group）. The peak intensity (PI) of the recurrence and metastasis group was higher than that of the non recurrence and metastasis group, and the serum CEA level and NLR were higher than those of the non recurrence and metastasis group. The time to peak (TTP) was lower than that of the non recurrence and metastasis group (P＜0.05）. The area under the curve (AUC) predicted by PI, TTP, CEA and NLR for recurrence and metastasis of breast cancer after modified radical mastectomy were 0.693, 0.764, 0.763 and 0.781, and the AUC predicted by PI, TTP, CEA and NLR for recurrence and metastasis of breast cancer after modified radical mastectomy was 0.909, which was greater than that predicted by each index alone. Conclusion: In patients with recurrence and metastasis of breast cancer after modified radical mastectomy, contrast-enhanced ultrasound parameters PI increased and TTP decreased, serum CEA and NLR increased. Objective To investigate the relationship between blood eosinophil (EOS), neutrophil (NEU), procalcitonin (PCT) and hypersensitive C-reactive protein (hs-CRP) levels and severity and outcome in neonatal acute respiratory distress syndrome (ARDS). Methods From January 2018 to June 2022, a total of 55 patients who were transferred to Department of Neonatal in our hospital within 24 hours after birth and met the inclusion criteria of the study were selected. According to oxygenation index, they were divided into the mild ARDS group (26 cases), the moderate ARDS group(13 cases) and the severe ARDS group(16 cases). According to the outcome, they were divided into the good outcome group (34 cases) and the comprehensive adverse outcome group(21 cases). The clinical data, EOS, NEU, PCT and hs-CRP levels of patients with different degrees and outcomes were compared separately; ROC curves were used to analyze the value of EOS, NEU and PCT in the assessment of comprehensive adverse outcomes in neonates with ARDS. Results The levels of NEU, and PCT in the severe group were higher than in the mild and moderate groups (P<0.05). The levels of hs-CRP in the severe group was higher than in the mild group (P<0.05), but the difference in hs-CRP was not significant in the moderate and severe groups (P>0.05); the levels of NEU, NLR, and PCT were higher in the comprehensive adverse outcome group than those in the good outcome group, and the levels of EOS were lower than those in the good outcome group (P<0.05); EOS, NEU, and PCT were independent risk factors for poor outcome in neonatal ARDS (P<0.05); the area under the curves for NEU, PCT levels and combined diagnosis to predict poor neonatal ARDS outcome were 0.877.0. 835 and 0.912. respectively. Conclusion The levels of serum EOS, NEU and PCT in neonatal ARDS are closely related to the severity of disease and outcome, and can be used as indicators for the assessment the outcome of neonatal ARDS. Objective To explore the risk factors of intestinal necrosis in patients with incarcerated inguinal hernia and the diagnostic value of predictors. Methods A total of 107 patients with incarcerated hernia underwent emergency surgery in Jinshan Hospital, Fudan University from Jan 2017 to Jun 2021 were analyzed retrospectively. Cases of intestinal necrosis were the observation group (n=24)and no intestinal necrosis was the control group (n=83). The cutoff value of continuous variables was determined by receiver operating characteristic curve (ROC) to evaluate the diagnostic value of intestinal necrosis predictors. Univariate and multivariate analysis were applied to analyze the potential risk factors for intestinal necrosis in incarcerated hernia. Results Among 107 patients who met the inclusion criteria，24 (22.4%) underwent intestinal resection for intestinal necrosis. Univariate analysis showed that intestinal necrosis was associated with female (P=0.028), elderly (P=0.004), femoral hernia (P=0.013), incarceration time (P=0.023), leukocyte count (P=0.002), neutrophil count (P<0.001), lymphocyte count (P=0.016), neutrophil/lymphocyte count (P<0.001), C-reactive protein (P=0.031), plasma fibrin (P=0.018), increased D-Dimer (P<0.001), intestinal obstruction (P<0.001) and peritonitis signs (P=0.002). Multivariate regression analysis showed that the number of neutrophils >7×109 /L (OR=6.188, 95%CI：1.350-41.326，P=0.023) and D-dimer >0.3 mg/L (OR=24.961，95%CI：3.328-186.659，P= 0.002) were the risk factors of intestinal necrosis in incarcerated hernia. The AUC area of ROC curve predicted by neutrophil count (>7×109 /L) and D-dimer (>0.3 mg/L) was 0.894. The sensitivity, specificity, positive predictive value and negative predictive value for the diagnosis of intestinal necrosis were 95.8%，91.6%，76.7% and 98.7%, respectively. Conclusion Elevated neutrophils and D-dimers may be associated with the risk of intestinal necrosis in patients with incarcerated hernia, which can help predict and diagnose intestinal necrosis before surgery. Eutrophils are the first innate immune cells to reach the site of inflammation. Neutrophils produce neutrophil extracellular traps (NETs) that can quickly capture and limit the spread of pathogens, facilitating the removal of pathogens and their debris. Neutrophils in the oral cavity are specifically transformed from circulating neutrophils in the blood, and the number of NETs released by oral neutrophils is much higher than that of circulating neutrophils, thus better maintaining the balance of the oral microenvironment. As a bimorphic fungus, only the mycelium phase of Candida albicans can induce NETs, which is related to the neutrophils' ability to sense the size of pathogenic microorganisms through neutrophil elastase. However, spherical Staphylococcus aureus are much smaller than Candida albicans, and they can still induce NETs. Porphyromonas gingivalis, as one of the microorganisms in the periodontitis complex, induces fewer NETs than Streptococcus oralis and Actinomycetes, which are two common oral microorganisms, and there may be a mechanism allowing them to escape neutrophilic immunity in the early stage of periodontitis. Although the two main pathways of NET production have been studied in detail, the mechanisms involved in the induction of NETs by different microorganisms, especially from oral neutrophils, are not well understood. This review describes the mechanism of the immune effects of pathogenic microorganisms on neutrophil NETs in the oral cavity, providing a reference for the search for therapeutic targets and the development of key drugs for treating oral infectious diseases.

Objective To investigate the effect and mechanism of tetramethylpyrazine (TMP) on the formation of neutrophil extracellular traps (NETs) induced by high glucose. Methods The viability of neutrophils treated with TMP of 0, 5, 10, 20, 40, and 80 μmol/L for 4 h was measured by CCK-8 assay. Neutrophils were divided into Control group (1.5 g/L glucose), TMP group (40 μmol/L TMP), HG group (4.5 g/L glucose), HG+TMP group (40 μmol/L TMP+4.5 g/L glucose), HG+TMP+ML385 group (4.5 g/L glucose+40 μmol/L TMP+10 μmol/L ML385), HG+ML385 group (4.5 g/L glucose+10 μmol/L ML385) and HG+DPI group (4.5 g/L glucose+10 μmol/L DPI). The structure of NETs was observed with immunofluorescence assay. The contents of reactive oxygen species (ROS) and superoxide anions were detected using dichloro-dihydro-fluorescein diacetate (DCFH-DA) assay and dihydroethidine (DHE) method. MPO-DNA and dsDNA in the culture medium was measured with ELISA and Pico-Green kit, respectively. Western blotting was used to detect the intracellular expression of H3-cit, NRF2, HO-1, and NOX2 proteins. Results TMP of 80 μmol/L had significant effect on neutrophil (P < 0.01), but no such effects were seen in TMP at other doses viability. Compared with the Control group, the expression levels of NRF2 and HO-1 were increased significantly in TMP group (P < 0.01), while that of H3-cit was obviously decreased after TMP treatment (P < 0.05), and the contents of dsDNA and MPO-DNA in the medium were decreased notably (P < 0.01). NETs formation was not observed by immunofluorescence assay in the HG+TMP group. The productions of ROS and superoxide anions in the HG+TMP group were significantly lower than those produced in the HG group (P < 0.01). The effects of TMP on the expression of H3-cit, NRF2, and HO-1 were reversed after treatment with NRF2 inhibitor ML385. Compared with the HG+TMP group, the contents of dsDNA and MPO-DNA were increased significantly (P < 0.01), and the contents of intracellular ROS and superoxide anions were elevated remarkably in HG+TMP+ML385 group (P < 0.01). Conclusion TMP can activate the NRF2/HO-1 pathway and then suppress the production of ROS, and thus inhibit the NETs formation induced by high glucose.

Pericyte, a kind of mesenchymal cell, surrounds the wall of microvessels. Inflammation is the body’s defense response to tissue damage, which is helpful to the proliferation and repair of damaged tissues and the removal of harmful substances. However, excessive inflammatory reaction will also cause damage to the body and cause diseases. Studies in recent years have paid more and more attention to the role of pericytes in inflammation. Pericytes can quickly respond to inflammatory stimulation, regulate neutrophilic infiltration in process, and exert the interaction of endothelial cells through direct contact, paracrine and a series of signal transduction pathways, to maintain stable neurovascular unit and physiological barrier. In addition, pericytes themselves have the immune function of phagocytosis and antigen presentation, and have the dual role of anti ⁃ inflammatory and proinflammatory in the regulation of inflammation. These effects make pericytes play an important role in inflammatory diseases, and provide a possible research direction for the treatment of pericytes. Cholestatic liver injury (CLI) is a liver disease with high mordity. Accumulation of bile acids in the liver is thought to be a major driver of liver injury during cholestasis. The disease can develop into be liver fibrosis, cirrhosis, liver failure and liver cancer. This review focuses on the molecular mechanism of CLI in the perspective of inflammatory response. The roles of neutrophils, Kupffer cells, bile duct cells, and the signaling pathways involved in CLI are reviewed to provide clues for research of new therapeutic targets and treatment strategies.

Objective To explore the characteristics and clinical significance of airway microbiota in the patients with neutrophilic asthma. Methods Induced sputum samples were collected from 44 patients with clinically stable neutrophilic asthma attending the second Affiliated Hospital of Army Medical University from August 2019 to January 2020. After DNA extraction, 16S rRNA and internal transcribed space (ITS) sequencing were performed by PacBio Sequel Systems. Two subgroups were identified by hierarchical cluster analysis on sequencing data, and the differences in their bacterial and fungal microbiota and clinical characteristics were explored. Results Hierarchical clustering analysis of bacterial sequencing data from 44 subjects identified 2 clusters. Compared to the larger cluster (group A1, n=32), the smaller cluster (group A2, n=12) was characterized by a predominance of Haemophilus in their bacterial microbiota, and the patients in this group demonstrated a higher percentage of sputum neutrophils (Z=-4.14, P < 0.001) and poorer FEV1% pred (t=2.15, P=0.037). In bacterial microbiota, there were significant differences in both α-diversity (Chao1 and Simpson indexes, P < 0.001) and β-diversity (Adonis, R2=0.290, P=0.001) between the 2 groups. At the species level, LEfSe analysis revealed that the group A1 was significantly enriched in Streptococcus pneumonia, Haemophilus parainfluenzae, Fusobacterium nucleatum, Lautropia mirabilis, Neisseria meningitides, Porphyromonas gingivalis, Prevotella melaninogenica, Prevotella pallens and Ralstonia solanacearum, while the group A2 was enriched in Haemophilus influenzae. In fungal microbiota, no significant differences were observed between the 2 groups in α-diversity (Chao1 index, P=0.230; Simpson index, P=0.097), whereas significant differences were observed between their β-diversity (Adonis, R2=0.068, P=0.002). At the species level, LEfSe analysis revealed that the group A1 was enriched in Schizophyllum commune, and group A2 was enriched in Trametes versicolor and Irpex lacteus. Interaction network analysis indicated that group A2 showed more correlations between genus than group A1, and the strength of association between genus was more intense. Conclusion Among the patients with stable neutrophilic asthma, there is a subgroup characterized by the presence of Haemophilus influenzae as its airway predominant colonizing bacteria, which differs significantly from the non-Haemophilus influenzae dominant group in terms of bacterial and fungal microbiota, and presents clinical characteristics with more severe airway inflammation and poorer lung function.

Objective: To investigate the differentially expressed proteins in lesions of patients with psoriasis vulgaris (PV) by proteomics technology. Methods: A total of six patients with PV and six patients with pigmented nevus presented to our hospital between June 2020 and September 2020 were enrolled. The lesions of PV and pigmented nevus-adjacent normal skin tissues were obtained by surgical removation. These differentially expressed proteins between the lesions of patients with PV (n=4) and normal skin tissues (n=4) was explored by proteomics technique. These differentially expressed proteins were further analyzed by GO and KEGG enrichment analysis. The protein protein interaction (PPI) analysis was used to screen out key proteins in the lesions of PV. Key proteins were independently verified in the lesions with PV (n=2) and normal skin tissues (n=2). Results: When compared with normal skin tissues, 291 proteins were significantly up-regulated and 163 proteins were significantly down-regulated in lesions of PV. These involved biological processes were mainly associated with neutrophil activation, neutrophil-mediated immunity, TNF-α-mediated signaling pathway, B cell-mediated immunity, epithelial cell differentiation and keratinization. The involved pathogenesis was mainly associated with IL-17 signaling pathway, oxidative phosphorylation, PPAR signal pathways and others. The PPI results showed that S100A7, S100A9, neutrophil gelatinase-associated lipocalin, cytochrome c oxidase subunit NDUFA4, fatty acid-binding protein 5, peroxisomal acyl-coenzyme A oxidase 1, platelet glycoprotein 4, cathepsin B, eosinophil cationic protein, haptoglobin, haptog-1lobin, apoptosis-associated speck-like protein containing a CARD may paly the key role in the pathogenesis of PV. Objective: To analyze the significance of peripheral blood inflammatory cells in the patients with serum prostate specific antigen（PSA）4-20 ng/ml in differentiating prostate cancer from prostatic hyperplasia, and establish a nomogram model to predict the risk of prostate cancer based on meaningful parameters. Methods: Clinical data of the patients who underwent prostate biopsy by transrectal of ultrasound in our hospital from June 2013 to December 2020 were retrospectively analyzed. Variables including neutrophil, lymphocyte, monocyte, PSA, prostate volume（PV）by ultrasound, and pathological examination were collected. Of these patients, 80% were randomly selected as the modeling group, and the other 20% as the validation group. The independent risk factors of prostate cancer were screened in the modeling group using logistic regression analysis, and then a prediction model based on regression equation was established to predict the result of prostate cancer. A receiver operating characteristic (ROC) curve was used to determine the diagnostic value of the nomogram model. Results: Total of 351 patients (147 patients with prostate cancer and 204 patients with benign prostatic hyperplasia ) with integrity data, which PSA was between 4 ng/mL to 20 ng/mL, were enrolled in this study. There were significant differences in free PSA(fPSA), f/tPSA (free to total PSA), PV (prostate volume), PSAD (PSA density) and NLR between two groups. And there were no differences in LMR and NMR between two groups. Logistic regression analysis showed that f/tPSA, PV and NLR were the independent risk factors in the modeling group. The nomogram model with these variables was established. And the area under the ROC curve (AUC) of the model (0.915) was greater than that of f/tPSA(0.765), PSAD(0.880) and NLR(0.550) (P＜0.05). Conclusion: The NLR-based nomogram model established in study has better diagnostic value for prostate cancer, which is a simple and convenient method without additional medical spending. Objective: To analyze the expression and clinical significance of E3 ubiquitin ligase (HECW2) in gastric adenocarcinoma using a bioinformatics approach to provide new clues for finding diagnostic and prognostic biomarkers for gastric adenocarcinoma. Methods: The expression of HECW2 in pan-cancer and its relationship with the prognosis of pan-cancer were analyzed by R language. The expression of HECW2 in gastric adenocarcinoma tissues and its relationship with clinicopathological features were analyzed with the help of UCSC Xena, HPA, and Kaplan-Meier Plotter databases. The WB method was used to detect HECW2 protein levels in Chinese gastric adenocarcinoma tissues and their paraneoplastic tissues to verify the analysis results in the database. The relationship between HECW2 and immune infiltration of gastric adenocarcinoma was analyzed with the help of TIMER and Cibersort databases. GO functional analysis and KEGG signaling pathway enrichment analysis and correlation gene analysis were performed for HECW2 in gastric adenocarcinoma by LinkedOmics database. Results: The results of raw signal analysis showed that HECW2 was significantly highly expressed in 12 tumors including gastric adenocarcinoma among 33 different types of tumors (all P<0.05), and the results of WB method showed that HECW2 was also significantly highly expressed in Chinese gastric adenocarcinoma tissues (P<0.05). Higher HECW2 expression level was associated with shorter OS and higher abundance of infiltration of immune cells including CD4+ T cells, macrophages, neutrophils and dendritic cells in gastric adenocarcinoma patients (all P<0.01). In addition, HECW2-associated signaling pathways were mainly enriched in extracellular matrix receptor interactions, adherent spots, cell adhesion and oxidative phosphorylation (all P<0.01). Conclusion: HECW2 is significantly overexpressed in gastric adenocarcinoma tissues and is closely related to the poor prognosis and immune invasion of gastric adenocarcinoma, and has great potential as a prognostic biomarker and target therapy for gastric adenocarcinoma. Objectives: To explore the risk factors of major adverse cardiovascular events (MACEs) in immune checkpoint inhibitor (ICI) related myocarditis and establish a predictive model. Methods: This was a retrospective case-control study. Tumor patients diagnosed with ICI related myocarditis in the First Affiliated Hospital of Guangzhou Medical University from May 2019 to August 2023 were selected and divided into non-MACE group and MACE group based on whether MACE occurred. Clinical and imaging data of the two groups were collected. Univariate and multivariate logistic regression models were used to analyze the risk factors for MACE in patients with ICI related myocarditis. According to the results of multivariate logistic regression analysis, R 4.1.0 software was used to construct the MACE risk prediction model for these patients and draw a nomogram. The receiver operating characteristic curve was used to evaluate the prediction ability of the prediction model. Results: A total of 35 patients with ICI related myocarditis, aged (63.9±8.2) years, were included, including 28 males (80%). There were 18 patients in the non-MACE group and 17 patients in the MACE group. Multivariate logistic regression analysis showed that elevated neutrophil to lymphocyte ratio ( OR =1.115, 95% CI 1.007-1.235, P =0.036) and ST-T segment changes ( OR =24.942, 95% CI 1.239-502.194, P =0.036) were risk factors for MACE in patients with ICI related myocarditis. The receiver operating characteristic curve indicated that the area under the curve of the prediction model was 0.967 (95% CI 0.916-1.000, P <0.001), with a sensitivity of 88.2% and specificity of 100%, demonstrating good predictive ability. Conclusion: Elevated neutrophil to lymphocyte ratio and ST-T segment change are independent risk factors for MACE in patients with ICI related myocarditis. Risk prediction model based on the above two indicators can assist in the early identification and individualized intervention of ICI related myocarditis patients.
