Soy protein is wildly used in food industry due to its high nutritional value and good functionalities. However, the poor storage stability of commercial soy protein products has puzzled both the producers and the users for a long time. The current study assessed the changes in protein solubility, aggregation, oxidation, and conformation of soy protein isolate (SPI) with various soluble aggregates formed at different pH values (pH 5–8) during storage. During storage, SPI samples showed a reduced protein solubility (p, SPI powder with higher pH (before spray drying) exhibited a lower storage stability. Higher pH produced more disulfide‐mediated aggregates. More soluble aggregates in fresh SPI appeared to induce faster loss of protein solubility.

The back-propagation neural network is the most commonly used neural network for designing intelligent control systems for highway bridges. To improve the stability of an intelligent controller for a bridge with magnetorheological dampers and to reduce energy consumption, optimal performance was achieved by rationalising the number of hidden-layer elements and using a genetic algorithm (GA). Simulation results showed that a GA can achieve better optimisation results than those of simply optimised hidden-layer elements and can reduce energy consumption effectively.

The inclusion complexes of oxyresveratrol (Oxy) with β-cyclodextrin (β-CD) and hydroxypropyl-β-cyclodextrin (HP-β-CD) were prepared and characterized by UV–vis spectroscopy, FT-IR spectroscopy, XRD, TG-DTG and SEM. Based on the optimal formulation, 222.23 ± 1.52 μg and 232.65 ± 2.64 μg of Oxy was included in 1.00 mg of the Oxy-β-CD and Oxy-HP-β-CD dried powder with the water solubility up to 14.44 ± 0.83 mg/mL and 47.33 ± 0.78 mg/mL, yielding about 30 and 100-fold increase in Oxy solubility (about 0.47 mg/mL), respectively. The physical stability and dissolution rate study showed that the formation of inclusion complexes significant enhanced the storage stability of Oxy and its dissolution rate. These results indicated that β-CD and HP-β-CD may be suitable delivery systems of Oxy. Furthermore, as illustrated in this study, inclusion complexes exhibited strong antibrowning effects on fresh grape juice in 24 h, showing its great potential application in the food industry.

In this study, a food grade water in oil (O/W) microemulsion to improve the solubility and stability of 7,2′,4′-trihydroxyflavanone was developed, and its ability to inhibit on the browning of fresh apple juice was also investigated. The results showed that the solubility of 7,2′,4′-trihydroxyflavanone in the aqua coconut oil microemulsion was up to 22.380 ± 0.336 mg/mL, which is approximately a 2000-fold solubility increase compared with that in water (0.011 mg/mL). The results of acceleration and long-term testing indicated thats the selected microemulsions were stable. The addition of VC into the 7,2′,4′-trihydroxyflavanone microemulsion (TME) greatly improved the solubility and stability of 7,2′,4′-trihydroxyflavanone. A 0.01% final concentration of TME performed well at inhibiting the browning of fresh apple juice during storage for 24 h at room temperature and for 7 days at 4 °C, and the antibrowning effect was further improved when a 0.05% VC concentration was added in combination with the TME. These results demonstrated that 7,2′,4′-trihydroxyflavanone microemulsion holds potential as a new antibrowning agent for fresh apple juice.

Oil-in-water microemulsions (O/W MEs) allow the preparation of insoluble compounds into liquid. In this study, we prepared O/W MEs to improve the solubility and stability of steppogenin (S) in aqueous liquid, and studied their ability to inhibit fresh apple juice browning. The ME technique greatly increased steppogenin solubility up to 3000-fold higher than that in water. All SMEs demonstrated good stability after acceleration and long-term storage. In particular, 0.01% SME was associated with dramatic inhibition of fresh apple juice browning after 24 h at room temperature and 7 days at 4 °C, and its antibrowning effects were further improved when combined with 0.05% ascorbic acid. On the other hand, simultaneous encapsulation of steppogenin with vitamin E or butylated hydroxytoluene into ME did not greatly improve SME antibrowning effects. Taken together, these results suggested that steppogenin might serve as a potential antibrowning agent to preserve fresh apple juice.

Flavonoids are an important type of natural tyrosinase inhibitor, but their inhibitory activity and mechanism against tyrosinase are very different because of their different structures. In this study, the inhibitory activity and mechanism differences between norartocarpetin and luteolin for tyrosinase were investigated by a combination of kinetic studies and computational simulations. The kinetic analysis showed that norartocarpetin reversibly inhibited tyrosinase in a competitive manner, whereas luteolin caused reversible noncompetitive inhibition. Both norartocarpetin and luteolin showed a single type of quenching and a static-type quenching mechanism. A computational simulation indicated that the hydroxyl groups of the B ring of norartocarpetin interacted with tyrosinase residues Asn81 and His85 in the active pocket, while the hydroxyl groups of the B ring of luteolin bound residues Asn81 and Cys83. HPLC and UPLC-MS/MS further confirmed that luteolin acted as a substrate or a suicide inhibitor, yet norartocarpetin acted as an inhibitor.

Artocarpus heterophyllus is an evergreen tree distributed in tropical regions, and its fruit (jackfruit) is well-known as the world's largest tree-borne fruit. Although A. heterophyllus has been widely used in folk medicines against inflammation, its potential in cancer chemoprevention remains unclear. Herein we identified artocarpin from A. heterophyllus as a promising colorectal cancer chemopreventive agent by targeting Akt kinase. Phenotypically, artocarpin exhibited selective cytotoxicity against human colon cancer cells. Artocarpin impaired the anchorage-independent growth capability, suppressed colon cancer cell growth, and induced a G1 phase cell cycle arrest which was followed by apoptotic as well as autophagic cell death. Mechanistic studies revealed that artocarpin directly targeted Akt 1 and 2 kinase activity evidenced by in vitro kinase assay, ex vivo binding assay as well as Akt downstream cellular signal transduction. Importantly, oral administration of artocarpin attenuated colitis-associated colorectal tumorigenesis in mice. Taken together, artocarpin, a bioactive component of A. heterophyllus, might merit investigation as a potential colorectal cancer chemopreventive agent.

Ethnopharmacological relevance Morus australis, one of the major Morus species growing in East Asia, is rich in phenolic compounds. The extract of M. australis has been used as skin whitening components for a long period. The action mechanisms of its principal constituents are still unclear. This study aims to evaluate the skin lightening effects of phenolic compounds extracted from the root of M. australis in different melanocyte systems and artificial skin models. Materials and methods The depigmenting effect of resorcinol type polyphenols (RTPs) from the root extract of M. australis was evaluated in murine b16 and melan-a cell lines using a combined sulforhodamine B assay. Tyrosinase activity and the expression of melanogenesis proteins were evaluated for the mechanism study. The artificial skin model is used as a replacement of the animal test. Results Only Kuwanon O and Sanggenon T were found to have significant depigmenting effects in both murine b16 and melan-a cell lines. Their depigmenting mechanisms are slightly different in the two cell systems. In b16 cells, Kuwanon O and Sanggenon T, together with the other two RTPs, induced post-transcriptional degradations of MITF without suppressing its mRNA expression, leading to significant decreases of TRP-1 and TRP-2 production. While in melan-a cells, the levels of tyrosinase families were suppressed via MITF downregulation at both transcription and translation level by RTPs, with Kuwanon O inducing the greatest suppression. Further evaluations in artificial skin model demonstrated the outstanding depigmenting effects of Kuwanon O and Sanggenon T. Conclusions Kuwanon O and Sanggenon T from M.australis root extract are two potential skin whitening ingredients. To screen resorcinol flavonone derivatives with an isoprenyl group in the Diels-Alder substituent might be an option for the search of potent hypopigmenting agents from plants.

The purpose of this study is to prepare an oxyresveratrol (Oxy) microemulsion (ME) with improved Oxy's solubility and stability and to investigate its antibrowning effects on fresh-cut lotus root slices. The formula of OxyME consisted of ethyl butyrate, Tween 80, PEG400, and water with w/w of 4%, 10.67%, 5.33%, and 80%, respectively. Encapsulating Oxy into OxyME greatly increased its solubility and stability compared with that of in water. Strong antibrowning effects were observed on fresh-cut lotus root slices treated with OxyME, even better than 4-hexylresorcinol. The addition of ascorbic acid (VC) into OxyME greatly improved the Oxy stability in long-term storage and antibrowning effects on fresh-cut lotus root slices. However, the simultaneous addition of calcium chloride and VC did not obviously improve the antibrowning effects compared with the addition of VC alone. These results indicated that Oxy+VCME may be suitable as an antibrowning agent for fresh-cut vegetables.

In this study, pentacyclic triterpenoids (PTTs) were extracted from loquat leaves by subcritical water extraction (SWE) technique in a dynamic mode. The results revealed that PTTs yield increased up to 5.38±0.12 mg/g with the increasing temperature at 180 °C for corosolic acid (CA) and up to 7.20±0.11 mg/g at 200 °C for ursolic acid (UA) within experimental times. The optimum flow rates to obtain concentrated CA and UA extracts were found to be 33.33 and 41.67 mL/min, respectively. Furthermore, extraction temperatures strongly influenced the extraction rate as demonstrated by the rate constant of each temperature tested. Moreover, the kinetic rate constant decreased as the function of temperature indicating the yield of both CA and UA were significantly influenced by subcritical temperatures and extraction times.

Norartocarpetin, quercetin and naringenin were found to effectively inhibit 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) formation through trapping its phenylacetaldehyde and form their adducts in roast beef patties. Six adducts [8-C- or 6-C-(E-phenylethenyl) flavonoids] formed between phenylacetaldehyde and three flavonoids were detected in roast beef patties by UPLC-MS analyses and compared with their synthetic references. These flavonoid-phenylacetaldehyde adducts were synthesised and further subjected to cytotoxicity tests on three liver cancer cell lines HepG2, SMMC-7721 and QGY-7703. The adduct 8-C-(E-phenylethenyl)norartocarpetin (NARA1) was found to significantly induce cancer cell death with IC50 values about 7 μM. After pre-treating with MAPK and caspase inhibitors, alteration of the cell morphology and cleaved-PARP were detected in liver cancer cell lines administered with NARA1. These data indicated that norartocarpetin could inhibit PhIP formation in roast beef patties and form norartocarpetin-phenylacetaldehyde adducts. The adduct NARA1 has anticancer potential via intrinsic caspase-dependent and cell context-dependent MAPKs pathways.

Chalcones and their derivatives have attracted great interests in recent years for their comprehensive biological activities. In this study, 2,4,2',4'-tetrahydroxychalcone and its two derivatives, 1,3,5-tris-(2,4-dihydroxy-phenyl)pentane-1,5-dione (new compound) and 7,2',4'-trihydroxyflavanone, were synthesized through one-pot green procedure catalyzed by boric acid in polyethylene glycol 400. Their structures were identified by ESI-MS and NMR spectral. Tyrosinase inhibitory activity and antibrowning test results showed that compounds 1-3 exhibited strong tyrosinase inhibitory activities and significant antibrowning effects on the fresh-cut lotus root slices at room temperature in 48 h. Among them, 0.01% 1,3,5-tris-(2,4-dihydroxy-phenyl)pentane-1,5-dione combined with 0.5% VC showed the best antibrowning ability. In brief, this study offers a protocol for one-pot green synthesis of high efficiency tyrosinase inhibitors which may be suitable as antibrowning agents for fresh-cut vegetables. More important, this study developed a new type of 1,5-dione derivative which may serve as new lead structures for novel tyrosinase inhibitors discovery.

This study investigated the effects of irradiation on Ne-carboxymethyl-lysine (CML) and Ne-carboxyethyl-lysine (CEL) formation in cooked red and white meats during storage. The results showed that irradiation did not affect CML/CEL formation (0 weeks). After 6 weeks, CML/CEL contents in the irradiated samples exhibited a higher growth rate than the non-irradiated samples, especially the red meat. The results of electron spin resonance spectrometry and 2-Thiobarbituric acid-reactive substances suggested irradiation had induced free-radical reactions and accelerated lipid oxidation during storage. A new method was developed for one-pot green synthesis 1,3,5-triarylpentane-1,5-dione, triarylmethane, and flavonoid derivatives from the reaction between 2,4-dihydroxybenzaldehyde and hydroxyacetophenones via Aldol, Michael, and Friedel-Crafts additions using boric acid as catalyst in polyethylene glycol 400. The synthetic compounds demonstrated significant tyrosinase inhibitory activities much stronger than that of kojic acid. More important, 1,3,5-triarylpentane-1,5-dione and triarylmethane derivatives were found to be a new class of tyrosinase inhibitors.

In this study, the inhibitory effects of eight kinds of dietary flavonoids on the formation of heterocyclic amines (HAs) were investigated in roast beef patties. The results showed that most of them exhibited significant inhibition on both total HAs and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), one of the most abundant HAs. Among the studied flavonoids, phlorizin, epigallocatechin gallate (EGCG), and quercetin were found to be the most effective in both the reductions of total HAs (55-70%) and PhIP (60-80%). The reaction activity between the flavonoid and phenylacetaldehyde, a key intermediate in PhIP formation, showed a good correlation with the inhibition of PhIP formation in an aqueous model system (R(2) = 0.8904) and a di(ethylene) glycol reaction system (R(2) = 0.6514). However, no significant correlation was found between the flavonoid antioxidant capacity and PhIP formation (R(2) = 0.2359). The postulated adducts of flavonoids-phenylacetaldehyde were further confirmed by LC-MS analysis in the chemical models. Since phenylacetaldehyde is the chief intermediate in PhIP formation, these results suggest that the inhibitory effects of flavonoids on PhIP formation are mainly dependent on their abilities to trap phenylacetaldehyde as opposed to their antioxidant capacities.

Summary This study investigated the effects of oxidised linoleic acid (18:2) on Ne-carboxymethyl-lysine (CML) and Ne-carboxyethyl-lysine (CEL) formation in Maillard reaction systems. Model systems of lysine/glucose (L/G), lysine/18:2 (L/18:2), lysine/18:2/glucose (L/18:2/G), myofibrillar protein/glucose (MFP/G), MFP/18:2 and MFP/18:2/G were maintained at 37 °C for 6 weeks. The results showed that CML/CEL contents in L/G (6.99 and 0.96 mmol mol−1 lysine, respectively) were significantly higher than those in L/18:2/G (1.43 and 0.41 mmol mol−1 lysine, respectively), and there is a small amount of CML/CEL generation in L/18:2. However, the CML/CEL levels in MFP/G (197.2 and 83.8 ng mg−1 protein, respectively) were markedly lower than those in MFP/18:2/G (283.2 and 118.5 ng mg−1 protein, respectively). 18:2 favours the formation of CML/CEL in MFP/18:2/G, not in L/18:2/G. All these findings indicated that the role of 18:2 on CML/CEL formation in Maillard reaction system was complex, and depended on CML/CEL formation rate and substrate types (lysine or lysine residue in protein).

The depigmenting effects of oxyresveratrol and trans- dihydromorin were investigated in b16F10 cell line and its synergetic melan-a cells, as well as in 3-dimensional skin models. But their inhibitory effects were different in two cell lines. In b16 cells, oxyresveratrol showed stronger depigmenting effect than trans- dihydromorin, with greater suppression on tyrosinase activity. Both of them induced post-transcriptional degradations of microphthalmia-associated transcription factor (MITF), leading to significant decreases of tyrosinase related protein 1 (TRP-1) and tyrosinase related protein 2 (TRP-2) production. In melan-a cells, trans- dihydromorin exhibited stronger inhibition on melanin synthesis. It also induced greater suppression on the levels of tyrosinase, TRP-1 and TRP-2 via MITF down-regulation at both transcription and translation level. Evaluations in artificial skin model also demonstrated the depigmenting effects of trans- dihydromorin. In conclusion, we found that trans -dihydromorin is an effective hypopigmenting agent in normal skin cells. Furthermore we demonstrated that hypopigmenting agents effective in melanoma system may not be effective on normal melanocytes, indicating that a non-tumor melanocyte system is more suitable for the screening of hypopigmenting agents.

In this study, the encapsulation mechanism of oxyresveratrol and β-cyclodextrin (β-CD) and hydroxypropyl-β-cyclodextrin (HP-β-CD) was studied. As this research shows, oxyresveratrol and two cyclodextrins (CDs) were able to form inclusion complexes in a 1:1 stoichiometry. However, the interaction with HP-β-CD was more efficient, showing up as higher encapsulation constant (KF) (35,864.72 ± 3415.89 M−1). The KF values exhibited a strong dependence on temperature and pH, which decreased as they increased. From the thermodynamic parameters (ΔH0, ΔS0, and ΔG0) of the oxyresveratrol loaded β-CD (oxyresveratrol-β-CD) and HP-β-CD (oxyresveratrol-HP-β-CD), it could be seen that the complexation process was spontaneous and exothermic, and the main driving forces between oxyrsveratrol and CDs were hydrogen bonding and van der waals force. Besides, molecular docking combined with 1H-NMR were used to explain the most possible mode of interactions between oxyresveratrol and CDs.

In this study, a convenient approach and green procedure for the synthesis of 4-phenacylideneflavenes has been developed from the reaction between 2,4-dihydroxybenzaldehyde and substituted acetophenones using boric acid as a catalyst in polyethylene glycol 400. Seven 4-phenacylideneflavenes were synthetized and their structures were confirmed by NMR and mass spectral analyses. Meanwhile, their possible mechanism of formation was also discussed. These products were found to have potential cytotoxic effect on HepG2 cell line with IC50 values from 12.5 to 50 µM.

A new preparation procedure, allowing the analysis of 11 polar heterocyclic aromatic amines (HAAs) in meat samples, is proposed and applied to commercial meat products. It is based on ultrahigh-performance liquid chromatography with tandem mass spectrometry (UHPLC-MS/MS). The procedure involves a one-step extraction using hydrochloric acid coupled with a solid-phase extraction clean-up using Oasis MCX cartridges. Significantly higher recoveries were obtained (ranging from 58.2% to 107.6%) for every target polar HAAs from roasted pork samples prepared in a laboratory compared with a commonly used reference method based on multiple extractions. The UHPLC-MS/MS parameters were optimized to simultaneously analyze all the 11 HAAs within 17 min with the limits of quantification (LOQs) < 4.3 μg L−1 using standard solutions. The LOQs for the spiked meat extract (0.026–0.659 ng g−1) and adequate day-to-day precision ranging from 6.17% to 12.03% (N = 15) were obtained. Five commercial meat products (smoked fish, roasted chicken wing, roasted pork, grilled beef, and sausage) were spiked with three concentrations (low, medium and high) of the HAAs standards. Satisfactory recoveries (about 50–110%) were obtained, which verify the applicability and advantage of the proposed method.

The twigs of Cudrania tricuspidata were found to show strong tyrosinase inhibitory activity, and further detailed component analysis resulted in the isolation of a new flavanol glucoside, (2S,3S)-2,3-trans-dihydromorin-7-O-β-d-glucoside (1), plus twenty-seven known compounds (2-28). Their structures were elucidated on the basis of ESI-MS and NMR spectral data. Among the isolated compounds, trans-dihydromorin (8), oxyresveratrol (9), and steppogenin (12) were found to exhibit significant tyrosinase inhibition activities. Moreover, the structure-activity relationship of these isolated compounds was also discussed.

The twigs of Morus alba L. were found to show strong tyrosinase inhibition activity, and the responsible active components in the extract were further investigated in this study. A flavone, named morusone (1), and sixteen known compounds 2–17 were isolated from M. alba twigs and their structures were identified by interpretation of the corresponding ESI-MS and NMR spectral data. In the tyrosinase inhibitory test, the compounds steppogenin (IC50 0.98 ± 0.01 µM), 2,4,2′,4′-tetrahydroxychalcone (IC50 0.07 ± 0.02 µM), morachalcone A (IC50 0.08 ± 0.02 µM), oxyresveratrol (IC50 0.10 ± 0.01 µM), and moracin M (8.00 ± 0.22 µM) exhibited significant tyrosinase inhibition activities, much stronger than that of the positive control kojic acid. These results suggest that M. alba twig extract should served as a good source of natural tyrosinase inhibitors for use in foods as antibrowning agents or in cosmetics as skin-whitening agents.

Medicinal properties of loquat leaf extracts (LLEs) are associated with their constituents of phenolic compounds and triterpenes. In this study, the efficacy of subcritical water extraction (SWE) technique was assessed by comparing with conventional solid-liquid extraction (CE) and Soxhlet extraction (SE). Results showed that the highest yields of total polyphenols were 82.7 ± 1.5 mgGAE/g leaf weight (LW), total flavonoids (54.1 ± 4.1 mgQE/g LW) and total triterpenoids (37.5 ± 3.2 mgUAE/g LW) were obtained by SWE compared to total polyphenols (61.8 ± 3.3 mgGAE/g LW), total flavonoids (43.2 ± 0.6 mgQE/g LW) and total triterpenoids (28.7 ± 2.3 mgUAE/g LW) extracted by SE and total polyphenols (50.3 ± 1.8 mgGAE/g LW), total flavonoids (40.4 ± 2.1 mgQE/g LW) and total triterpenoids (22.9 ± 3.2 mgUAE/g LW) obtained by CE. The extraction efficiency of triterpenes using SWE was about 1.7 times higher than those obtained using traditional extraction methods, and their main structural pattern of the cured extracts was comparable to the extracts obtained using traditional extraction methods.The infrared spectra obtained from the three extraction techniques appeared identical, but the variation in the intensity of the peak of absorption was visible among the three extraction techniques. The similarity of the infrared spectral pattern (peak coincided peak by peak) implies that the triterpenes in the extract obtained by the three techniques were identical by LC/MS. The findings of this study have demonstrated that SWE can be employed as an alternative green extraction technology to get important phytochemicals from plant sources. cope Quercetin, a flavonoid, widely distributed in edible fruits and vegetables, was reported to effectively inhibit 2-amino-1-methyl-6-phenylimidazo[4, 5-b]pyridine (PhIP) formation in a food model (roast beef patties) with itself being converted into a novel compound 8-C-(E-phenylethenyl)quercetin (8-CEPQ). Here we investigated whether 8-CEPQ could be formed in a real food system, and tested its anticancer activity in human colon cancer cell lines. Methods and results LC-MS was applied for the determination of 8-CEPQ formation in onion/beef soup. Anticancer activity of 8-CEPQ was evaluated by using cell viability assay and flow cytometry. Results showed that 8-CEPQ suppressed proliferation and caused G2 phase arrest in colon cancer cells. Based on immunofluorescent staining assay, western blot assay, and RNA knockdown data, we found that 8-CEPQ did not cause apoptotic cell death. Instead, it induced autophagic cell death. Moreover, treatment with 8-CEPQ induced phosphorylation of extracellular signal-regulated kinase (ERK). Inhibition of ERK phosphorylation by the mitogen-activated protein kinase kinase (MEK)/ERK inhibitor U0126 attenuated 8-CEPQ-induced autophagy and reversed 8-CEPQ-mediated cell growth inhibition. Conclusion Our results demonstrate that 8-CEPQ, a novel quercetin derivative, could be formed in onion/beef soup. 8-CEPQ inhibited colon cancer cell growth by inducing autophagic cell death through ERK activation.

A novel non-targeted isoflavone profiling method was developed using the diagnostic fragment-ion-based extension strategy, based on ultra-high performance liquid chromatography coupled with photo-diode array detector and electrospray ionization-mass spectrometry (UPLC-PDA-ESI-MS). 16 types of isoflavones were obtained in positive mode, but only 12 were obtained in negative mode due to the absence of precursor ions. Malonyldaidzin and malonylgenistin glycosylated at the 4'-O position or malonylated at the 4″-O position of glucose were indicated by their retention behavior and fragmentation pattern. Three possible quantification methods in one run based on UPLC-PDA and UPLC-ESI-MS were validated and compared, suggesting that methods based on UPLC-ESI-MS possess remarkable selectivity and sensitivity. Impermissible quantitative deviations induced by the linearity calibration with 400-fold dynamic range was observed for the first time and was recalibrated with a 20-fold dynamic range. These results suggest that isoflavones and their stereoisomers can be simultaneously determined by positive-ion UPLC-ESI-MS in soymilk.

The phytochemical profiles of Morus australis roots, stems and twigs were firstly compared by HPLC analysis. It was found that Morus australis stem extract mainly contained one known tyrosinase inhibitor, oxyresveratrol, while its root and twig extract might contain some unknown potential tyrosinase inhibitors. The root extract of Morus australis was further investigated in this study. One new compound, austraone A, together with 21 known compounds, was isolated and their structures were identified by interpretation of MS and NMR data. In the tyrosinase inhibitory testing, some of them, such as oxyresveratrol, moracenin D, sanggenon T, and kuwanon O, exhibited stronger tyrosinase inhibitory activities than that of kojic acid. These results suggested the Morus australis root extract as a good source of natural tyrosinase inhibitors with a great potential to be used in foods as anti-browning agents and in cosmetics as skin-whitening agents.

Recently correlation studies between dietary advanced glycation end products (AGEs) and the progression of chronic diseases have attracted much attention. To explore the impact of dietary AGEs on the health risk of people consuming a high-fat diet (HFD), male Sprague-Dawley rats were used as the research subject. Under HFD, free N(e)-(carboxymethyl)lysine (CML, a major AGE, 60 mg/kg body weight/day) was administered by gavage for 12 consecutive weeks. The results indicated that protein-bound CML accumulation in the kidney, heart, lung, pancreas, and muscle significantly increased to 178 ± 36, 161 ± 2, 106 ± 11, 39 ± 8, and 141 ± 20 μg/g dry matter, respectively, compared to HFD control levels of 86 ± 9, 127 ± 10, 89 ± 6, 23 ± 2, and 95 ± 3 μg/g dry matter, whereas no statistical increase was found in the liver and spleen. An increase of the protein-bound CML might be due to free CML binding to proteins in tissues by covalent bonds. Moreover, the rats' serum low-density lipoprotein cholesterol concentration, fasting blood glucose levels, and energy expenditure also increased obviously. These findings indicated that long-term intake of high-dose free CML might be a health risk factor for rats with HFD. This could provide valuable information for further study on the possible effects of long-term consumption of CML-rich fatty foods on human health, involving the progression of chronic disease.

A thinner-THGEM (Thick Gas Electron Multiplier) with a thickness of 200 μm and small rim has been produced by high speed drilling and global etching. Some performances have been studied and the working conditions have been tested and selected, using various Ar and Ne based gas mixtures. Gain values above 3×10 3 under relatively low working voltage, energy resolution of 15.9% with 5.9 keV X-rays and good gain stability are obtained. Using Monte Carlo simulation, the dependence of the energy resolution on drift electric field was analyzed and compared with experimental results.

Resveratrol exhibits anti-tumor properties against different types of cancer. In this study, several polyhydroxylated resveratrol derivatives were prepared with the aim of discovering new leading compounds with clinical potential for human colon cancer chemotherapy. Among these compounds, 3,3′,4,5,5′-pentahydroxy- trans- stilbene (PHS) displayed the most potent cytotoxicity and triggered apoptosis in HT-29 cells as evidenced by increased poly(ADP-ribose) polymerase (PARP) cleavage, elevated levels of cytoplasmic nucleosomes and DNA fragmentation. Further mechanistic analysis revealed that PHS-induced apoptosis was caspase-dependent and mediated by its pro-oxidative action through up-regulation of reactive oxidative species generation and depletion of intracellular glutathione.

A new furanoflavone, 7-(2,4-dihydroxyphenyl)-4-hydroxy-2-(2-hydroxy propan-2-yl)-2, 3-dihydrofuro(3, 2-g)chromen-5-one (artocarpfuranol, 1), together with 14 known compounds, dihydromorin (2), steppogenin (3), norartocarpetin (4), artocarpanone (5), artocarpesin (6), artocarpin (7), cycloartocarpin (8), cycloartocarpesin (9), artocarpetin (10), brosimone I (11), cudraflavone B (12), carpachromene (13), isoartocarpesin (14), and cyanomaclurin (15) were isolated from the wood of Artocarpus heterophyllus. Their structures were identified by interpretation of MS,( 1)H-NMR,( 13)C-NMR, HMQC, and HMBC spectroscopic data. Among them, compounds 1-6 and 14 showed strong mushroom tyrosinase inhibitory activity with IC(50) values lower than 50 microM, more potent than kojic acid (IC(50) = 71.6 microM), a well-known tyrosinase inhibitor. In addition, extract of A. heterophyllus was evaluated for its antibrowning effect on fresh-cut apple slices. It was discovered that fresh-cut apple slices treated by dipping in solution of 0.03 or 0.05% of A. heterophyllus extract with 0.5% ascorbic acid did not undergo any substantial browning reaction after storage at room temperature for 24 h. The antibrowning effect was significantly better than samples treated with the extract (0.03 or 0.05%) or ascorbic acid (0.5%) alone. The results provide preliminary evidence supporting the potential of this natural extract as antibrowning agent in food systems.

Aim of the study Fissistigma oldhamii (Hemsl.) Merr, a traditional Chinese herb medicine, is used for treating rheumatoid arthritis in China. In our previous study, an effective compound, 7′-(3′,4′-dihydroxyphenyl)-N-[(4-methoxyphenyl) ethyl] propenamide (Z23), from this herb has showed potent immunosuppressive effects both in vitro and in vivo. However, its anti-inflammatory effect and mechanism is still need to explore. Materials and methods We examined the in vitro effects of Z23 on the production of nitric oxide (NO), prostaglandin E2 (PGE2) and cytokines by lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Results Z23 significantly decreased the production of PGE2, NO, tumour necrosis factor alpha (TNFalpha) and IL6 production. Inducible nitric oxide synthase (iNOS) and cyclooxygenase2 (COX2) gene expression were also significantly reduced. Conclusions These results demonstrated that Z23 exerted an anti-inflammatory effect through modulating the synthesis of several mediators and cytokines involved in the inflammatory process. This study provided evidence to understand the therapeutic effects of Fissistigma oldhamii (Hemsl.) Merr and indicated that Z23 has the potential for treatment of various inflammatory diseases where the overproduction of NO, PGE2 and inflammatory cytokines has been shown to play a role, e.g. rheumatoid arthritis.

The anti-glycation activity of four kinds of beans including mung bean, black bean, soybean and cowpea were evaluated. Aqueous alcohol extract of mung bean exhibited the strongest inhibitory activity against the formation of fluorescent advanced glycation endproducts (AGEs) in a bovine serum albumin (BSA)-glucose model, and the inhibitory activities of extracts of the four beans were found to be highly correlated with their total phenolic contents ( R 2 = 0.95). Subsequent HPLC analysis of mung bean extract revealed two major phenolics which were purified and identified as vitexin and isovitexin by spectral methods. In the anti-glycation assays, both vitexin and isovitexin showed significant inhibitory activities against the formation of AGEs induced by glucose or methylglyoxal with efficacies of over 85% at 100 μM. In another assay, vitexin and isovitexin failed to directly trap reactive carbonyl species, such as methylglyoxal, suggesting that their anti-glycation activities may mainly be due to their free radical scavenging capacity.

Fractionation of a chloroform-soluble extract from twigs of Broussonetia papyrifera , led to the isolation of one new compound, 3,5,7,4′-tetrahydroxy-3′-(2-hydroxy-3-methylbut-3-enyl)flavone ( 1 ), and 10 known compounds, uralenol ( 2 ), quercetin ( 3 ), isolicoflavonol ( 4 ), papyriflavonol A ( 5 ), broussoflavonol F ( 6 ), 5,7,3′,5′-tetrahydroxyflavanone ( 7 ), luteolin ( 8 ), isoliquiritigenin ( 9 ), broussochalcone A ( 10 ) and 5,7,3′,4′-tetrahydroxy-3-methoxyflavone ( 11 ). Their structures were identified by interpretation of MS, 1 H NMR, 13 C NMR, HMQC and HMBC data. Their inhibitory activities on mushroom tyrosinase using l -tyrosine as substrate were investigated and the IC 50 values of 3,5,7,4′-tetrahydroxy-3′-(2-hydroxy-3-methylbut-3-enyl)flavone, uralenol, quercetin and broussoflavonol F were found to be 96.6, 49.5, 57.8, and 82.3 μM, respectively, better than arbutin, a well-known tyrosinase inhibitor.

The aim of this study is to improve artocarpanone solubility by developing an O/W microemulsion with the evaluation of its antibrowning effects. The chemical and physical stabilities as well as antibrowning effects in apple juice were also evaluated. The formulation of artocarpanone microemulsion consisted of 4% w/w of ethyl butyrate, 10.67% w/w of Tween 80, 5.33% w/w of polyethylene glycol 400, and 80% w/w of water, with a maximum solubility of artocarpanone up to 10.54 ± 0.01 mg/mL, at least 3000-folds increase in solubility compared that in water. Encapsulating artocarpanone and ascorbic acid (VC) into microemulsion simultaneously decreased modest artocarpanone solubility whereas improving its stability in long-term storage. Blank, artocarpanone and artocarpanone-Vc-loaded microemulsions demonstrated steadily during accelerated and long-term storage. Artocarpanone-Vc-loaded microemulsion showed strong antibrowning effects in apple juice at room temperature in 24h, suggesting that artocarpanone-Vc-loaded microemulsion is a good antibrowning agent for apple juices.

Norartocarpetin is a flavone widely distributed in Moraceae plants with strong tyrosinase inhibitory activity. However, its poor solubility in aqueous systems and in food grade solvents (oils) limits its extensive applications. The aim of this study was to improve the solubility of norartocarpetin by developing an oil-in-water (o/w) microemulsion with food grade components. A microemulsion was developed and characterized, and its chemical and physical stabilities were also evaluated within 8 weeks. Using the construction of pseudoternary phase diagrams, the optimized formulation of 2% w/w of ethyl oleate, 12% w/w of Tween 80, 6% w/w of polyethylene glycol 400, and 80% w/w of water was obtained, with a maximum solubility of norartocarpetin up to 1.73 ± 0.21 mg/mL, at least a 1000-fold increase in solubility. The microemulsion base and norartocarpetin-loaded microemulsion were demonstrated to be stable after accelerated and long-term conditions (8 weeks). The norartocarpetin microemulsion still showed strong tyrosinase inhibitory activity and antibrowning effect on fresh-cut apple slices. These combined results indicated that norartocarpetin microemulsion may be suitable as an antibrowning agent for fresh-cut fruits.

In order to find new structural and biologically active compounds, the constituents of the bark of Cudrania tricuspidata (Carr.) Bur. were investigated and a new 6-p-hydroxybenzyltaxifolin glucoside, named tricusposide (compound 1), together with 16 known compounds, was isolated by solvent partition, macroporous adsorption resin AB-8, silica gel, Sephadex LH-20 chromatography. Using spectroscopic methods, the structures of the compounds were elucidated as 6-p-hydroxybenzyl taxifolin-7-O-β-D-glucoside (compound 1), dihydroquerctin-7-O-β-D-glucoside (compound 2), dihydrokaempferol-3-O-β-D-glucoside (compound 3), dihydroquercetin (compound 4), peonoside (compound 5), sphaerobioside (compound 6), quercimeritrin (compound 7), genistein (compound 8), aromadendrin (compound 9), kaempferol (compound 10), genistin (compound 11), 3,4-dihydroxystyryl alcohol (compound 12), sucrose (compound 13), 1,3,5,6-tetrahydroxyxanthone (compound 14), gericudranin E (compound 15), gericudranin C (compound 16), and orobol (compound 17). A novel method allowing simultaneous analysis of PhIP, 4'-OH-PhIP, and their precursors (phenylalanine, tyrosine, creatine, creatinine, glucose) has been developed as a robust kinetic study tool by using ultra high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). A direct hydrochloric acid (HCl) extraction was applied to achieve the simultaneous extraction of all seven analytes, with the mean recoveries ranging from 60% to 120% at two concentration levels. Then, an Atlantis dC18 column selected from four different chromatographic columns was ultimately used to separate these compounds within 15 min. The limits of detection range of allseven analytes were calculated as 0.14-325.00 μg L(-1). The intra- and interday precision of the proposed method were less than 15.4 and 19.9%, respectively. The proposed method was successfully applied to depict the kinetic profiles of PhIP, 4'-OH-PhIP, and their precursors in pork model, reducing the analysis time and cost in the kinetic study.

The effects of Chinese spices on the profiles of 17 heterocyclic amines (HAs) from seven HA categories were investigated in roast beef patties using Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry (UPLC-MS/MS) and principal component analysis. Three groups of HAs, imidazopyridines (PhIP, DMIP, and 1,5,6-TMIP), imidazoquinoxalines (MeIQx and 4,8-DiMeIQx), and β-carbolines (harman and norharman), were detected and quantified in all of the samples. The results demonstrated that the total HA and imidazopyridine profiles could clearly be affected by 1% pricklyash peel (14.1 ± 0.76 and 6.06 ± 0.32 ng/g), chilli (41.0 ± 0.01 and 23.0 ± 0.52 ng/g), and cumin (59.9 ± 2.44 and 31.1 ± 3.06 ng/g), in comparison with control values of 21.8 ± 2.40 and 14.3 ± 2.04 ng/g, respectively. The difference was only significant (p0.05) for cumin. The imidazoquinoxaline profile was significantly (p0.05) affected by 1% pricklyash peel (0.57 ± 0.05 ng/g) and cumin (2.36 ± 0.20 ng/g) compared to the control (1.16 ± 0.11 ng/g). The β-carboline profile was only markedly (p0.05) affected by 1% cumin (26.4 ± 0.82 ng/g) compared to the control (6.26 ± 0.26 ng/g). In general, pricklyash peel inhibited HA formation, whereas star anise, fennel, cumin, chilli, and black pepper promoted HA formation. The findings could facilitate the selection of spices in meat processing to minimize HA formation.

Artocarpus heterophyllus is an evergreen fruit tree cultivated in many tropical regions. Previous studies have shown that some of its compositions exhibited potential tyrosinase inhibition activities. This study indentified 8 new phenolic compounds, artoheterophyllins E-J (1-6), 4-geranyl-2',3,4',5-tetrahydroxy-cis-stilbene (7), and 5-methoxymorican M (8) and 2 new natural compounds (9 and 10), 2,3-dihydro-5,7-dihydroxy-2-(2-hydroxy-4-methoxyphenyl)-4H-benzopyran-4-one and 6-[(1S,2S)-1,2-dihydroxy-3-methylbutyl]-2-(2,4-dihydroxyphenyl)-5-hydroxy-7-methoxy-3-(3-methyl-2-buten-1-yl)-4H-1-benzopyran-4-one, together with 23 known compounds (11-33), from the ethanol extract of the wood of A. heterophyllus. The structures of the eight new compounds (1-8) and two new natural compounds were established by extensive 1D- and 2D-NMR experiments. The anticancer effects of the isolated compounds were examined in MCF-7, H460, and SMMC-7721 human cancer cell lines by MTT assay. Compounds 5, 11, 12, and 30 significantly reduced the cell viabilities of these cell lines. Especially, compounds 11 and 30 resulted in more potent cytotoxicity than the positive control, 5-fluorouracil (5-Fu), in SMMC-7721 cell line, with IC50 values of 15.85 and 12.06 μM, whereas compound 30 exhibited more potent cytotoxicity than 5-Fu in NCI-H460 cell line, with an IC50 value of 5.19 μM. In addition, this study suggests that compounds 11 and 30 from the wood of A. heterophyllus have anticancer potential via MAPK pathways.

Based on 106 x 10(6)psi(3686) events collected with the BESIII detector at the BEPCII facility, a partial wave analysis of psi(3686) -> p (p) over bar pi(0) is performed. The branching fraction of this channel has been determined to be B psi(3686) -> p (p) over bar pi(0) = (1.65 +/- 0.03 +/- 0.15) x 10(-4). In this decay, 7 N* intermediate resonances are observed. Among these, two new resonances, N(2300) and N(2570) are significant, one 1/2(+) resonance with a mass of 2300(-30-0)(+40+109) MeV/c(2) and width of 340(-30-58)(+30+110) MeV/c(2), and one 5/2(-) resonance with a mass of 2570(-10-10)(+19+34) MeV/c(2) and width of 250(-24-21)(+14+69) MeV/c(.)(2) For the remaining 5 N* intermediate resonances [N(1440), N(1520), N(1535), N(1650) and N(1720)], the analysis yields mass and width values that are consistent with those from established resonances. Using a sample of 106×10(6) ψ(3686) events collected with the BESIII detector at the BEPCII storage ring, we have made the first measurement of the M1 transition between the radially excited charmonium S-wave spin-triplet and the radially excited S-wave spin-singlet states: ψ(3686)→γη(c)(2S). Analyses of the processes ψ(3686)→γη(c)(2S) with η(c)(2S)→K(S)(0)K(±)π(∓) and K(+)K(-)π(0) give an η(c)(2S) signal with a statistical significance of greater than 10 standard deviations under a wide range of assumptions about the signal and background properties. The data are used to obtain measurements of the η(c)(2S) mass (M(η(c)(2S))=3637.6±2.9(stat)±1.6(syst) MeV/c(2)), width (Γ(η(c)(2S))=16.9±6.4(stat)±4.8(syst) MeV), and the product branching-fraction (B(ψ(3686)→γη(c)(2S))×B(η(c)(2S)→KKπ)=(1.30±0.20(stat)±0.30(syst))×10(-5)). Combining our result with a BABAR measurement of B(η(c)(2S)→KKπ), we find the branching fraction of the M1 transition to be B(ψ(3686)→γη(c)(2S))=(6.8±1.1(stat)±4.5(syst))×10(-4).

The mass and width of the lowest-lying S-wave spin singlet charmonium state, the η(c), are measured using a data sample of 1.06×10(8) ψ(3686) decays collected with the BESIII detector at the BEPCII storage ring. We use a model that incorporates full interference between the signal reaction, ψ(3686)→γη(c), and a nonresonant radiative background to describe the line shape of the η(c) successfully. We measure the η(c) mass to be 2984.3±0.6±0.6 MeV/c(2) and the total width to be 32.0±1.2±1.0 MeV, where the first errors are statistical and the second are systematic.

With a sample of (225.2 +/- 2.8) x 10(6) J/psi events registered in the BESIII detector, J/psi -> gamma pi(+)pi(-)eta' is studied using two eta' decay modes: eta' -> pi(+)pi(-)eta and eta' -> gamma rho(0). The X(1835), which was previously observed by BESII, is confirmed with a statistical significance that is larger than 20 sigma. In addition, in the pi(+)pi(-)eta' invariant-mass spectrum, the X(2120) and the X(2370), are observed with statistical significances larger than 7.2 sigma and 6.4 sigma, respectively. For the X(1835), the angular distribution of the radiative photon is consistent with expectations for a pseudoscalar.

The Dalitz plot of eta' -> eta pi(+)pi(-) decay is studied using (225.2 +/- 2.8) x 10(6) J/psi events collected with the BESIII detector at the BEPCII e(+)e(-) collider. With the largest sample of eta' decays to date, the parameters of the Dalitz plot are determined in a generalized and a linear representation. Also, the branching fraction of J/psi -> gamma eta' is determined to be (4.84 +/- 0.03 +/- 0.24) x 10(-3), where the first error is statistical and the second systematic.

Jatrophalactam (1), a novel diterpenoid lactam possessing an unprecedented 5/13/3 tricyclic skeleton, was isolated from the roots of Jatropha curcas. The structure and relative configuration of jatrophalactam (1) were elucidated by extensive spectroscopic analysis and further determined by a single-crystal X-ray diffraction.
