Bone substitution materials are widely used in dentistry to replace bone defects. Xenografts offer an alternative to autografts, allografts and alloplast by addressing their limitations. Objective: To analyze characteristics of cancellous rib freeze-dried bovine bone as a potential bone substitute material. Bovine 4th cancellous rib were cut into 10 mm x 10 mm x 10 mm. Then soak in H2O2 for 7 x 24 hours, then cleaned with 95% alcohol in ultrasonic shaker for 6 x 30 minutes, then Freeze-dried for 4x24 hours. Test method using FTIR, XRD, SEM-EDX, ICP-OES. There are functional groups -OH, PO4 3-, N-H, C-N, CO3 2- and C-O-C, amorphous crystal form, pore size diameter 50.89 µm - 530.7 µm, porosity percentage 95.94%, and bone density 0.094 g/cm3, and there are C(Carbon), N(Nitrogen), O(Oxygen), F(Fluorine), Mg(Magnesium), Al(Aluminum), Si(Silicon), P(Phosporus), Ca(Calcium) content, and heavy metals Pb and Ni below toxicity limit (Pb<30ppm, Ni<25ppm). Freeze-Dried Bovine Bone Scaffold (FDBB) and Decellularized Freeze-Dried Bovine Bone (DCFDBB) are promising new alternative of xenograft material in bone tissue engineering. However, their biocompatibility is still unknown. To investigate whether FDBB and DC-FDBB scaffolds are biocompatible, able to induce cell proliferation attachment in vitro. Human umbilical cord mesenchymal stem cells (hUC-MSCs) culture was exposed to FDBB, DC-FDBB, Deproteinized Bovine Bone Matrix (DBBM) scaffold (as positive control) conditioned medium for 24, 48 and 72 hours (n=8). MTT assay was then performed to measure the number of viable cells at each observation time. Normality (Shapiro-Wilk) and homogeneity (Levene) test were done on the results of the MTT assay, then ANOVA test was performed. To support the finding, further SEM observation was performed. hUC-MSCs were seeded on the surface of each scaffold type (n=3) and incubated for 24, 48 and 72 hours, then SEM observation was done on the scaffold surface to analyse their surface cell attachment. The mean percentage of living cells in FDBB group was the highest among all groups. There were significant differences (p<0.05) in each treatment group at each observation time, except between FDBB and DC-FDBB (48 hours), DC-FDBB and DBBM (48 hours), FDBB and DC-FDBB (72 hours) (p>0.05). SEM examination showed the same results, as the highest number of cell colonization was found on FDBB group at each observation time. Hydroxyapatite can also be obtained from synthetic and natural materials, such as bovine tooth hydroxyapatite for bone graft. Gelatin is a type-l collagen which is considered to be the main organic component in bone. Chitosan has the same structure as glycosaminoglycans (GAGs), non-collagenous organic components of bone. This study aims to investigate the compressive strength and porosity of the HAGS-Gel-Kt scaffold with a ratio of 70:15:15 (w/w/w), Sigma-Aldrich Germany synthetic HA was used as comparison. HAGS:Gel:Kt scaffold was made with a ratio of 70:15:15, made with a weight of 1.75 gr HAGS powder, 0.375 gr gelatin and 0.375 gr chitosan. The HAGS-Gel-Kt scaffold used for pore size testing is a scaffold with a diameter of 5 mm and a height of 5 mm. The procedure for measuring the size of the porosity is carried out using scanning electron microsope (SEM). The compressive strength test was carried out using diameter of 8 mm and a height of 17 mm scaffold examined with universal testing instrument. HAGS-Gel-Kt cross-scaffold has higher size pore average than HAGS-Gel-Kt surface scaffold, HASyn-Gel-Kt surface scaffold, HASyn-Gel-Kt cross scaffold with significant different. HASyn-Gel-Kt has higher compressive strength than HAGS:Gel:Kt with significantly different (p<0.05). The compressive strength of the HAGS:Gel:Kt scaffold is equal to 0.221 MPa and the compressive strength of the HASyn:Gel:Kt scaffold is 0.370 MPa. Various biomaterials have been developed to be used as bone substitutes or materials to stimulate bone growth such as beta Tricalcium phosphate (beta-TCP). In addition to the selection of biocompatible materials, any technological developments have led to the project of making substitute materials leading to the manufacture of nano-sized materials. The combination of biomaterials types and size modification aims to obtain materials with biodegradability and biocompatibility abilities. The purpose of this scooping review was to get the latest description of beta tri calcium phosphate nanoparticles as scaffold combination material to increase bone formation. To obtain the data, a search was conducted using three databases, they were PubMed, Science Direct and Scopus. From all three databases, 259 articles were found. Data were then selected based on the inclusion and exclusion criteria. The title, abstract and full text of each study were carefully analyzed. Data extraction was made based on cell proliferation, cell toxicity, and new bone formation process. There were 33 selected articles and the results displayed that there were 15 other combinations of biomaterials used with beta-TCP. The in vitro studies revealed the increase in cell proliferation and the absence of toxicity from the combination of beta-TCP/hydroxyapatite and beta-TCP/Platelet Rich Plasma. While in vivo studies revealed the new bone formation from the combination of beta-TCP/hydroxyapatite and beta-TCP/ gelatin. In conclusion, the comparison of ingredients combined with beta-TCP in this scooping review were hydroxyapatite, platelet rich plasma and gelatin. Background: Composite resins have disadvantages such as low hardness level, shrinkage, and micro-leakage. In the case of deep cavity restorations, bulk-fill composite resins have begun to be used because they facilitate the restoration work and make the restoration time shorter. Purpose: This study was used to determine whether home bleaching treatment using hydrograph peroxide and carbamide peroxide can reduce the surface hardness of flowable bulk-fill composite resins. Methods: The research sample is cylindrical with a diameter of 5 mm and a height of 2 mm, this study was divided into 3 groups. Then each group was measured by Vickers Hardness Tester. Results: In the One Way Anova test the value of p = 0.020 was obtained. This indicates a significant difference in the surface hardness of bulk-fill flowable composite resins and a Post hoc test - Tukey HSD must be performed. The purpose of this to examine the effect of hydroxyapatite extracted from Indonesian limestone on bone defects healing by means of the number of osteoblasts, fibroblasts and osteoclasts. This study design is an true experimental with randomized post-test only control group design. Femurs of eighteen Cavia cobaya were drilled to make bone defects and divided into 3 groups; group 1 (G1): defects without bone graft treatment, group2 (G2): bone defects were filled with bovine hydroxyapatite and group 3 (G3): bone defects were filled with Indonesian limestone. After 14 days, the animals were sacrificed and femurs were extracted. The number of osteoblast, fibroblast and osteoclast were documented by histologically. The analysis of variance (ANOVA) continued by Least Significant Different (LSD) were conducted to analyze the data with p-value < 0.05 considered as statistically significant. The number of osteoblasts were higher in G3 than that in group G1 (p <0.05) but not with group G2 (p > 0.05). The number of osteoclasts were significantly lower in group G3 compared with group G1 and G2 (p<0.05). The number of fibroblast were higher in G3 than that in group G1 and group G2 (p<0.05). Periodontitis is an inflammatory disease of tooth supporting tissue caused by specific microorganisms, one of which is the bacteria Porphyromonas gingivalis (P. gingivalis). Based on the Basic Health Research (RISKESDAS) in 2018, dental and mouth problems reached 57.6% in Indonesia and only 10.2% had received services from medical personnel. In pregnancy there are hormonal changes that can affect periodontal tissue. Increasing the number of macrophages in pregnancies with chronic periodontitis can lead to a risk of preeclampsia. The aim of this study is toprove that pregnant mice with chronic periodontitis can increase the number of macrophages as a risk of pre-eclampsia. Thirty females Rattus novergicus, weighing 250-300 g with age 5-6 months, divided into 3 groups, consisting of 1 control group and 2 treatment groups. The control group is only pregnant mice. In treatment group 1 is the group of pregnant rats with chronic periodontitis and in treatment group 2 is the group of rats with chronic periodontitis. P. gingivalis ATCC 33277 was injected locally 0.03ml with a concentration of 1x109 CFU / ml under the incisor gingival sulcus in the right and left mesials. Data on the number of macrophages were analyzed using Kolmogorov-Smirnov and One-Way Anova. There were significant differences in the mean number of macrophages between groups. Limestone, as a natural source has the main component of calcium carbonate (CaCO3) and is abundantly existed in Indonesia. Calcium carbonate is one of the compounds that could be a precursor in the fabrication of hydroxyapatite (HA) that has been commonly used in clinical application. The center of ceramics in Indonesia (BBK: Balai Besar Keramik) has synthesize HA from limestone's calcium carbonate derived from mountains in Padalang and Cirebon, West Java, Indonesia. This study aims to evaluate the characteristics and cytotoxity of hydroxyapatite made from Padalarang-Cirebon limestone by BBK as a bone graft candidate. Characterizations of HA were Fourier Transform Infra-Red (FTIR), X-Ray Diffraction (XRD) and Energy Dispersive X-ray (EDX). Cytotoxicity of the samples were evaluated with MTT assay on umbilical cord mesenchymal stem cells (UC-MSCs) with the concentrations of 50µg/ml, 25µg/ml, 12.5µg/ml, 6.25µg/ml, 3.12µg/ml, 1.56µg/ml, 0.78µg/ml, 0.39µg/ml, and 0.19µg/ml. HA from limestone has hydroxyl (OH-) dan phosphate (PO42-) functional groups, the particle was in the crystal form and has the composition of O, Ca, and P with the Ca/P ratio was 1.64. Toxicity was not found in all concentrations (p > 5). HA from Padalarang-Cirebon limestone by BBK has hydroxyl and phosphate groups, crystal particles, the composition of O, Ca, and P, Ca/P ratio of 1.64 and no toxicity, which satisfied the requirements of a bone graft candidate. The most common bacteria that cause dental caries are Streptococcus mutans. Prevention of dental caries can be done by brushing teeth using toothpaste that contains antibacterial ingredients. Chitosan is a derivative product of chitin, which is extracted from crab shell. Chitosan has been proven to be non-toxic, has good biocompatibility and antibacterial effect. To determine the inhibition of toothpaste containing 1 percent chitosan extracted from crab shell on Streptococcus mutans. This study is an experimental study with only post-control design. A diffusion method was used to determine the effect of the material on bacteria. Agar media was used to culture Streptococcus mutans and divided into four parts. Toothpaste containing 1% chitosan from crab shell with low weight molecule, medium weight molecule, high weight molecule and basic toothpaste without any chitosan, are put into each part. The average inhibition zone diameter against Streptococcus mutans in toothpaste containing chitosan are various among its molecule weight. At low molecular weight has a diameter of 16.4914 mm, medium molecular weight has a diameter of 17.500 mm, high molecular weight has a diameter of 19.0643 mm, and in basic toothpaste without chitosan has a diameter of 14.9214 mm. The jawbone is an important component because it functions as a supporting structure of the teeth, regulates mineral homeostasis and protects various organs in the oral cavity.The number of tooth extraction cases in the Oral and Maxillofacial Surgery Clinic of Dental and Oral Hospital, Faculty of Dental Medicine, UniversitasAirlangga average in 2019 every month is 800 teeth. This shows that there is potential to use wasted crowns, such as HA, which will be used as scaffold candidates. Cytotoxicity of HA dental crowns as scaffold candidates will be investigated.Using hidroxyapatite (HA) Powder Preparation and Cytotoxicity Test. The absorbance results for all concentration groups are above 50%, which means HA of dental crowns is not toxic according to CD50, which states the material is said to be toxic if the number of living cells is below 50%. Background: Induction of the proliferation and differentiation of stem cells could represent a viable alternative therapeutic method for treating bone diseases. Stem cells are essential to bone tissue regeneration; although, their availability is limited. One possible method of increasing the number of stem cells and promote osteogenic differentiation is the application of red flesh dragon fruit extract supplement. The present study was performed to identify and analyze proliferation and osteogenic differentiation of bone marrow-derived mesenchymal stem cells (BMMSCs) after exposure to red flesh dragon fruit extract. Materials and Methods: This in vitro study was posttest only control group design. Red flesh dragon fruit extract was produced by means of water extraction method and subsequent dilution with different amounts of water to produce a range of concentrations. BMMSCs were obtained from the femurs of three White New Zealand rabbits. BMMSCs were then treated with 50, 100, 200, 300, and 400 µg/ml red flesh dragon fruit extract concentrations. The in vitro proliferation assay was determined by means of an 3-(4.5 dimethylthiazol 2-yl) 2,5- diphenyltetrazolium bromide assay. Osteogenic differentiation was determined by means of the degree of nodule mineralization. There were two groups as follows: group I with the addition of 50 µg/ml of red flesh dragon fruit extract and Group II without the addition of red flesh dragon fruit. Data were analyzed using analysis of variance and the Student's t test (P = 0.05). Results: 50, 100, 200, 300, and 400 µg/ml of red flesh dragon fruit extract demonstrated the capacity to significantly increase the proliferation of BMMSCs (P ≤ 0.05). Red flesh dragon fruit extract could significantly increase osteogenic differentiation (P ≤ 0.05). Background: The incision in the oral cavity is also taken for gingivectomy. Fig leaves (Ficus carica Linn) extract contains compounds of flavonoid, terpenoids and tannins and has anti-inflammatory and antioxidant activities. The important markers of wound healing were fibroblasts, macrophages and collagen density. Aim and Objectives: To investigate the topical application of fig leaves extracts on Wistar rat wounds on the number of macrophages, fibroblasts and collagen density after treatment for three and seven days. Material and Methods: This research was performed on 24 rats by making incision wounds on the backs of Wistar rats and divided into control and treatment groups. The control groups were left untreated and the treatment groups were given fig leaves extract gel on the incision wound once every day during thee and seven days, and then the animals were sacrificed. Wound tissue was removed and fixed in 10% formalin solution for histopathological test. Then it was embedded in paraffin, and stained with Hematoxylin-Eosin to observe fibroblasts and macrophages. The collagen density was observed by Masson's Trichome staining. Statistical analyses of fibroblasts and macrophages were using One-way Anova and Tukeys HSD. Collagen density was analyzed by using Kruskall-Wallis and Mann-Whitney test. Results: There were significant differences among the groups (p<0.005) on the number of fibroblasts, macrophages and collagen density after treatment for three and seven days. Conclusion: Application of fig leaves extract on Wistar rat wounds could increase the number of fibroblasts and macrophages but not collagen density in the wound healing process. Periodontitis, congenital disorder, degenerative disease, and bone tumor are the major problems in oral cavity that could damage bone continuity. The damage is irreversible and in most cases the bone cannot heal properly, so a biomimetic scaffold is needed to mediate the healing process. Scaffold made of chitosan and collagen derived-chicken shank (CDCS) is on a development in recent researches. Scaffold combined with 0.25% glutaraldehyde as cross linking agent shows a good improvement in biologic and mechanical properties. The aim of this study is to discover the profile of cross-linked chitosan-CDCS scaffold as biomaterial in tissue engineering. Methods: Scaffolds’ profiles were examined with degradation test, swelling test, WCP test and porosity size by using SEM. These scaffolds have degradation rate 20.909%, swelling ratio 1.625, WCP 59.851%, and porosity size36.06μm. Biomaterials that can be used to accelerate wound healing is the amniotic membrane that contains growth hormone. In its use, amniotic membrane tends to be difficult to use because of their sheets form. To make it easier to use, the amniotic membrane is mixed with gelatin as an adhesive material to produce sponge amnion. Sponge amnion which is used as a biomaterial needs to meet several requirements, one of them is biocompatibility. To complete these requirements cytotoxicity testing is necessary to determine the toxic potential that a material may produce. The cytotoxicity test was performed on BHK-21 fibroblast cells. The aim of this study is to determine the citotoxicity effect of amnion in sponge form on BHK-21 fibroblast cells. This study was done towards the culture cell line BHK-21 using MTT assay method. In this study used four sample groups. Two groups are the control group (media control and cell control), and the other two groups are samples consisted of amniotic membrane and sponge amnion. Living cells were quantified after treatment by ELISA reader on 620 nm. The fibroblast cell life percentage after exposure with amniotic membrane and sponge amnion is 98,16% and 98.13%. Based on the ANOVA test result was obtained P-value of 0.000 (P-value <0.05) so that it can be said that the treatment result gives a significant difference. Surgical procedure using bone replacement materials are still needed to accelerate new bone formation. Tissue engineering concept develop the research in scaffold biomaterial. Chitosan, gelatin and bovine hydroxyapatite combination can be synthesized as an ideal scaffold biomaterial that has a biomimetic properties of bone tissue. To determine pore size and hydrophobicity properties of chitosan-gelatin/bovine hydroxyapatite scaffold at various eligible ratios in bone tissue engineering. Scaffold chitosan-gelatin/bovine hydroxyapatite with a ratio of 20:80, 30:70 and 40:60 synthesized using freeze dry method. Scaffold on each ratio was tested by pore size examination using Scanning Electron Microscope. The ratio of swelling and water content percentage was done by measuring the initial weight and final weight after being soaked in distillate water for 1, 3 and 7 days. The smallest pore size was obtained at a 20:80 ratio scaffold with a mean value of 254.44 ± 37.96 µm and the largest on a 40:60 ratio scaffold with a mean value of 423.04 ± 68.72 µm. Swelling ratios and water content percentage were highest on the chitosan-gelatin/bovine hydroxyapatite ratio of 40:60 at day 7 (2,904 ± 0.531 and 75.84 ± 2.6%). The pore size and hydrophobicity properties corresponding to bone tissue regeneration biomaterials were obtained on the 20:80 and 30:70 ratios chitosan-gelatin/bovine hydroxyapatite scaffold. Background: In Indonesia, the incidence rate of tooth extraction as the main form of dental treatment reached as high as 79.6% in 2014. Teeth extraction leads to periodontal tissue defect whose treatment, incorporating the use of graft material Hydroxyapatite (HA) and Platelet Rich Fibrin (PRF) with stem cells, has become increasingly widespread. The stem cell potentially put to therapeutic use is the Hematopoetic Stem Cell (HSC). Aim and Objectives: to examine the effect of hematopoetic stem cell addition to HA graft and PRF post-tooth extraction on the number of osteoblasts and amount of Osteoprotegerin (OPG) in periodontal defect treatment. Material and Methods: This study constituted laboratory-based experimental research with a post test-only control group. Twenty four rats (Rattus novergicus) represented the animal study model in this research. Alveolar bone defect in the animal study model was induced by extracting the first molar of the mandible using sterile needle holder clamps. The sample population was divided into four groups: K0: the untreated control group; K1: socket filled with HA Graft and PRF. K2: socket filled with HSC 105 cells. K3: the socket filled with HAGraft and PRF with HSC 106 cells. Results: The K3 group had both the highest number of osteoblasts when compared with the control group (350.17 ± 33.42; P <0.001) and the strongest OPG expression of all treatment groups (11,36 ± 0,54; P <0.01). Conclusion:Acombination of HA graft, PRF and HSC significantly increases the expression of OPG and the number of osteoblasts rendering it a potential treatment for tissue engineering-based periodontal defect. Vascular endothelial growth factor (VEGF) is an essential mediator during the process of angiogenesis that is an initial process of bone regeneration. Engineered bone tissue commonly encompass the use of scaffold. Collagen plays an important role in bone regeneration. Unfortunately, collagen can be extracted from chicken shank. This research was aimed to determine the effect of chicken shank collagen scaffold on the expresion of VEGF. Chicken shank was mixed by trypsin powder and dissolved in CH3COOH until forming like fiber then was sentifuged. Supernatant was dissolved NaCl 5wt% until forming fiber then was dialyzed for three days. Chicken shank collagen was printed in mould to freeze dry for 24 hours. Rats with defected femur were divided into 2 groups. Group 1 was the control group, and group 2 was treatment group which the defected femur were administrated with chicken shank collagen scaffold. Those rats were sacrificed on 2nd week. Tissue preparation was made then immunohistochemical staining was conducted counting VEGF expression. A statistical analysis was conducted using Mann Whitney test. There was a significant difference in the expression of VEGF (p<0,05). Amniotic membrane is one of the natural biomaterial that is widely used for wound healing which has Secretory Leukocyte Protease Inhibitor (SLPI) as an active component. One of rSLPI functions is suppressing several pro-inflammatory factors, such as serine protease. Supression of serine proteinase in wound healing process may increase the amount of vascular endothelial growth factor (VEGF) and collagen. The aim of this study is to evaluate the effect of rSLP amniotic membrane fluid application on incision wound of Rattus norvegicus in expression of VEGF and collagen. The rats divided into 4 groups: control group and treated group with rSLPI (P1 0.03 cc, P2 0,045 cc, and P3 0.06 cc) (n=6). Insicion was made on rat's cervical back. On the day 4, tissues around insicion wound are collected for Masson's Trychrome (MT) and Immunohistochemistry (IHC) analysis. Expression of VEGF and collagen are calculated using microscope with 400x magnification. Kruskal-Wallis test of VEGF showed significant value with p=0,006 (p<0.05). Mann-Whitney test of VEGF showed a significant difference between control and treatment group. Oneway ANOVA test result for collagen has p value 0.000 (p<0.05). Honestly Significant Dif ference (HSD) test showed a significant differences between all group. Bone defect is still a major problem in dentistry. In oral cavity, trauma, congenital disorder, periodontitis and tumor can damage bones. A biomimetic scaffold made of bovine hydroxyapatite (BHA), gelatin (GEL), and chitosan (K) (BHA-GEL-K) is on a development in recent researches. This research tends to develop BHA-GEL-K scaffolds combined with 0.25% glutaraldehyde as cross linking agent. Scaffold's profiles were examined with FTIR, porosity size by using SEM and compressive strength by using autograph. The adhesion of bone narrow mesenchymal stem cell (BM-MSC) to crosslinked BHA-GEL-K scaffolds with 0.25% glutaraldehyde were examined by using SEM. BHA, GEL, K and glutaraldehyde components are bonded to each other. These scaffolds have 121.21 μm of porosity and 912.86 + 83.81 kPa of compressive strength value. These scaffolds have good adhesion to BM-MSC. The adhesion of scaffolds increases since day 1 to 7. The aim of this study was to examine the TLR2, NF-κB, TNFα expression as an inflammation marker to evaluate the anti-inflammation properties of combination zinc oxide and turmeric rhizome liquid extract as wound dressing. Twenty three male Wistar rats, aged 3 months, weighing 175-275 grams, adapted one week before, were devided into 3 groups. Group A without any intervention as reference for normal data (n=3), Group B (excision at 3rd day), Group C (excision at 7th day). Group B and C contains 10 animals and has two subgroups: control (excision, n=5) and experimental group (excision with dressing application, n=5).The tissues around wound excision were removed and used to analyze histopathologically and immunohistochemistry. ANOVA was used for the statistic and regression was used for path analysis. The results showed that combination of zinc oxide and turmeric rhizome liquid extract as a dressing, were found to inhibit the inflammation, as it indicates by lowering the TLR2 at the 7th day, blocking the NF-κB expression, lowering the TNFα expression. Path analysis found a strong relationship between dressing combination zinc oxide and turmeric rhizome liquid extract to TLR2 expression (B= -0,772), NF-κB (B=0,712), TNFα (B=0,722). In conclusion, these data indicate that zinc oxide with turmeric rhizome liquid extract have beneficial effects to accelerating the inflammation through lowering TLR2, NF-κB and TNFα expressions.
