Mulberry (Morus alba L.) plants are rich in 1-deoxynojirimycin (DNJ), which is a potential α-glucosidase inhibitor exhibiting various physiological activities. Compared to other tissues, Morus alba L. seeds contain the highest DNJ content, however, the DNJ biosynthesis mechanisms are unclear. In this study, we examined fruits of 27 mulberry varieties and found that variety MS02 had the highest DNJ levels (22.28 mg/g), whereas variety MS15 contained the lowest DNJ levels (0.37 mg/g). Through comparative transcriptomics, 1,719 differentially expressed genes (DEGs) were identified, 1,170 of which were upregulated, and 549 were downregulated in MS02 compared to MS15. DEGs were associated with cellular processes, metabolic processes, and catalytic activity. Specifically, nine DEGs were identified to be involved in alkaloid biosynthesis pathways, according to Kyoto Encyclopaedia of Genes and Genomes enrichment analysis, and four enzymes, i.e. polyphenol oxidase, tyrosine aminotransferase, aromatic-L-amino-acid decarboxylase, and tropinone reductase, are proposed to play important roles in DNJ biosynthesis. In conclusion, DNJ biosynthesis in mulberry seeds appears to be mediated by upregulation of polyphenol oxidase, tyrosine aminotransferase, aromatic-L-amino-acid decarboxylase, and tropinone reductase. Three new prenylated dihydroflavones, moralbaflavones A–C (1–3), together with four known ones (4a/4b, 5, and 6) were isolated from the root barks of Morus alba L. Their structures including the absolute configurations were determined by the analysis of HRMS, NMR, and ECD data. The neuroprotective properties of these prenylated dihydroflavones were screened at the concentration of 10 µM in the sodium nitroprusside-induced rat pheochromocytoma PC-12 cells, and the results showed moralbaflavone C (3) possessed significant neuroprotective activity, being more potent than the positive control edaravone. In the evolving field of food safety, rapid and precise detection of antibiotic residues is crucial. This study aimed to tackle this challenge by integrating advanced inkjet printing technology with sophisticated microfluidic paper‐based analytical devices (µPADs). The µPAD design utilized “green” quantum dots synthesized via an eco‐friendly hydrothermal method using green white mulberry leaves as the carbon source, serving as the key fluorescent detection material. The action mechanism involved a photoinduced electron transfer system using red carbon dots (CDs) as electron donors and blue CDs combined with two‐dimensional layered molybdenum disulfide (MoS2) nanosheets as electron acceptors. This system could quickly detect antibiotics within 10 min in pork and water samples, demonstrating high sensitivity and recovery rates: 6.5 pmol/L at 99.75%–110% for sulfadimethoxine, 3.3 pmol/L at 99%–105% for sulfamethoxazole, and 8.5 pmol/L at 98.5%–105% for tetracycline. It achieved a relative standard deviation under 5%, ensuring reliability and reproducibility. Context: The purpose of this study is to review the effectiveness of medicinal plants on memory and learning in both human and animal models. Material and Methods: Various electronic databases, including PubMed, Embase, Scopus, and Cochrane Library, were searched for randomized controlled trials published between 2010 and 2023. Results: The studies focused on thirty different herbs, including Bacopa monnieri , Polygonum odoratum , Morus alba , Ginkgo biloba , Salvia officinalis L., Rosmarinus officinalis L., Melissa officinalis , Panax ginseng , Saraca asoca , Polygala tenuifolia , Boswellia papyrifera , Boswellia serrata , and Aegle marmelos. Conclusions: The inclusion criteria were adults, participants with cognitive impairment, and healthy participants, without age or gender restrictions. Simple Summary: The presence of secondary metabolites like flavonoids, alkaloids, and phenolic compounds in white mulberry (Morus alba) make it an ornamental tree mainly in the Asian subcontinent. These secondary metabolites are synthesized through various biosynthetic pathways. Flavonoids like rutin, quercetin, and kaempferol are produced via the phenylpropanoid and flavonoid biosynthetic pathways. The key enzymes responsible for this pathway include phenylalanine ammonia-lyase, chalcone synthase, and flavonoid hydroxylases. Phenolic compounds like resveratrol and oxyresveratrol are produced via the phenylpropanoid pathway using phenylalanine as a precursor. Presences of these secondary metabolites, like in the case of flavonoids (rutin and quercetin), in the mulberry tree have potential biological activities like antioxidant, anti-inflammatory and neuroprotective effects. Also, they have been found to have great medicinal importance, for example in treating diabetes, obesity and neurodegenerative diseases. Also, phenolic compounds like reservertrol exhibit cardio-protective, anticancer, and anti-aging properties. Traditional natural products have been the focus of research to explore their medicinal properties. One such medicinally important plant is the white mulberry, Morus alba, widely distributed in the Asian subcontinent. It is one of the most cultivated species of mulberry tree and has attracted more focus from researchers because of its abundance in phytochemicals as well as multipurpose uses. The leaves, fruits and other parts of the white mulberry plant act as a source of valuable bioactive compounds like flavonoids, phenolic acids, terpenoids and alkaloids. These secondary metabolites have manifold healthy uses as they possess antioxidant, anti-inflammatory, antidiabetic, neutrotrophic, and anticancer properties. Despite the increasing scientific interest in this plant, there are very few reviews that highlight the phytochemistry and biological potential of white mulberry for biomedical research. To this end, this review elaborates the phytochemistry, biosynthetic pathways and pharmacological activities of the glycoside flavonoids of Morus alba. A comprehensive analysis of the available literature indicates that Morus alba could emerge as a promising natural agent to combat diverse conditions including diabetes, cancer, inflammation and infectious diseases. To achieve such important objectives, it is crucial to elucidate the biosynthesis and regulation mechanisms of the bioactive compounds in white mulberry as well as the multifaceted pharmacological effects attributed to this plant resource. The present review paper is intended to present a summary of existing scientific data and a guide for further research in the phytochemistry and pharmacology of white mulberry. Further, a biosynthetic pathway analysis of the glycoside flavonoid in mulberry is also given. Abiotic stress, especially drought stress, poses a significant threat to terrestrial plant growth, development, and productivity. Although mulberry has great genetic diversity and extensive stress-tolerant traits in agroforestry systems, only a few reports offer preliminary insight into the biochemical responses of mulberry leaves under drought conditions. In this study, we performed a comparative metabolomic and transcriptomic analysis on the "drooping mulberry" (Morus alba var. pendula Dippel) under PEG-6000-simulated drought stress. Our research revealed that drought stress significantly enhanced flavonoid accumulation and upregulated the expression of phenylpropanoid biosynthetic genes. Furthermore, the activities of superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) content were elevated. In vitro enzyme assays and fermentation tests indicated the involvement of flavonol synthase/flavanone 3-hydroxylase (XM_010098126.2) and anthocyanidin 3-O-glucosyltransferase 5 (XM_010101521.2) in the biosynthesis of flavonol aglycones and glycosides, respectively. The recombinant MaF3GT5 protein was found to recognize kaempferol, quercetin, and UDP-glucose as substrates but not 3-/7-O-glucosylated flavonols and UDP-rhamnose. MaF3GT5 is capable of forming 3-O- and 7-O-monoglucoside, but not di-O-glucosides, from kaempferol. This implies its role as a flavonol 3, 7-O-glucosyltransferase. Diabetes is a leading cause of death, according to statistics published by the Department of Health, Executive Yuan of Taiwan. In modern medicine, diabetes can be controlled using various medications; however, some drugs often have undesirable side effects. It therefore became a goal to find plant‐based material that can reduce glucose concentration in the blood while reducing the incidence of complications and not causing side effects. Alpha‐glucosidase inhibitors (AGIs) are effective glucose‐lowering medicines and are enzymes essential to carbohydrate digestion. Inhibition of α‐glucosidase leads to a delayed and reduced rise in postprandial blood glucose levels. This study evaluates the inhibitory effect of mixed extracts of Psidium guajava L. and Morus alba L. leaves on α‐glucosidase activity and postprandial hyperglycemia in normal and diabetic rats. The inhibition of α‐glucosidase activity was assayed in vitro. Half maximal inhibitory concentration (IC50) values of Psidium guajava L. and Morus alba L. were 2.25 and 0.1 mg/mL, respectively. The IC50 value of a commercial anti‐hyperglycemic agent (Glucobay) is 6.41 mg/mL. The IC50 value of a mixture of extracts of Psidium guajava L. and Morus alba L. was 0.07 mg/mL. In cytotoxicity tests, survival percentages and shape did not significantly affect the murine embryonic liver cell line (BNL CL.2) when treated with varying concentrations of mixture extracts for varying periods of time. In summary, Psidium guajava L. and Morus alba L. showed positive anti‐diabetes activity and suggested promising potential for alternative functional foods for diabetes mellitus (DM) patients. In this experiment, we explored the chemical composition and community structure of Mulberry "Wuhedashi" (Morus alba L., hereinafter referred to as WHDS) in different stages and obtained data support for its resource utilisation. Five ripening stages were established: S1, deep-red stage; S2, red with black stage; S3, black with red stage; S4, black stage; S5, overripe stage. The results showed that there were significant differences in the indicators of WHDS in the different stages. Immature WHDS contains high levels of amino acids (2.14 ± 0.15 mg/g), organic acids (43.10 ± 0.75 mg/g), K (3019.52 ± 78.00 mg/L), and Ca (1617.86 ± 24.45 mg/L) and is an important source of amino acid and mineral supplementation for the body. Total phenolic compounds (26.15 ± 0.43 g/L), total flavonoids (2.46 ± 0.03 g/L), total anthocyanins (587.60 ± 7.38 mg/L), the ABTS radical scavenging rate (94.20 ± 2.18%), the DPPH radical scavenging rate (95.13 ± 1.44%), and total flavour compounds (1279.09 ± 33.50 mg/L) peaked at S4, which is the optimal stage for the consumption and processing of WHDS. High-throughput sequencing identified 10 dominant genera, including Bacillus, Brevibacterium, Pseudomonas, and Tatumella. Nesterenkonia was the most highly associated micro-organism. Bacterial genera such as Pantoea and Pseudomonas were significantly positively correlated with esters, enhancing the floral and fruity flavours of wine. The results of the study revealed the characteristics of WHDS in different stages, which can help to target the development of nutritious mulberry derivatives and provide a reference for enhancing the added value of mulberry. Establishing the leaf productivity of the species Morus alba L. and Morus nigra L. and the influence of the mulberry species on some basic vegetative traits was the aim of this paper. The study was conducted during 2018-2020 at the Training Experimental Station of the Sericulture section of the Faculty of Agriculture at Trakia University - Stara Zagora. Object of the study were representatives of the genus Morus - M. alba L. (variety "Vratsa 1") and M. nigra L. The studied mulberry trees were from a lowstemmed plantation of an intensive type. The analysis of variances shows that the type of mulberry has a highly significant influence (p>0.001) in all analyzed cases characterizing leaf productivity. The established differences between the species in terms of the studied vegetative traits are in favor of M. alba. However, for M. nigra the average values of some morphological indicators are within the limits of those generally established in practice. This gives reason to consider that the data obtained in the present study are a useful addition to the general characterization of the species and are relevant for the purposes of selection and production of mulberry cultivars and hybrids. Morus sp. (mulberry) has a long tradition of use as a medicinal treatment, including for cardiovascular disease and type 2 diabetes, being shown to have antioxidant properties and to promote wound healing. Extracellular vesicles (EVs) are sub-micron, membrane-enclosed particles that were first identified in mammalian bodily fluids. EV-like particles have been described in plants (PDVs) and shown to have similar characteristics to mammalian EVs. We hypothesised that some of the health benefits previously attributed to the fruit of Morus sp. could be due to the release of PDVs. We isolated PDVs from Morus nigra and Morus alba via ultracentrifugation and incubated THP-1 monocytes, differentiated THP-1 macrophages, or HMEC-1 endothelial cells with pro-oxidant compounds DMNQ (THP-1) and glucose oxidase (HMEC-1) or lipopolysaccharide (LPS) in the presence of different fractions of mulberry EVs. Mulberry EVs augmented ROS production with DMNQ in THP-1 and caused the downregulation of ROS in HMEC-1. Mulberry EVs increased LPS-induced IL-1β secretion but reduced CCL2 and TGF-β secretion in THP-1 macrophages. In scratch wound assays, mulberry EVs inhibited HMEC-1 migration but increased proliferation in both low and high serum conditions, suggesting that they have opposing effects in these two important aspects of wound healing. One of the limitations of plant-derived therapeutics has been overcoming the low bioavailability of isolated compounds. We propose that PDVs could provide the link between physiological dose and therapeutic benefit by protecting plant active compounds in the GIT as well as potentially delivering genetic material or proteins that contribute to previously observed health benefits. Morus alba Linn. is a popular medium-sized tree with a potent neuroprotective activity such as anti-Alzheimer. To date, it is not known which phytocompound of M. alba exerts anti-Alzheimer activity. Therefore, the present study was aimed to identify the phytochemicals of M. alba responsible for the anti-Alzheimer activity by targeting proteins such as acetylcholinesterase (AChE) and glycogen synthase kinase-3 beta (GSK-3β) that are associated in the pathogenesis of Alzheimer disease (AD). Molecular docking, molecular dynamics (MD) simulations and MM-GBSA calculations were performed for the study. Phytocompounds with code 32, 41, 67, and 70 showed the best glide gscore against AchE and GSK-3β. All these phytocompounds interacted with multiple crucial amino acids present at the active binding site of the proteins through conventional hydrogen bonds and hydrophobic interactions. MD simulations of 100 ns revealed stable conformation for all protein-ligand complexes. Based on the computational findings, we conclude that M. alba's phytoconstituents (32, 41 67, and 70) exerts anti-Alzheimer potential by inhibiting AChE and GSK-3β. [ABSTRACT FROM AUTHOR]

Morus alba known as a white mulberry is a medicinal plant that has been used in food ingredients and traditional medicine. M. alba leaves contain various bioactive phenolic compounds, in particular chlorogenic acid (CGA), which is a major bioactive ingredient. Their anticancer potency of M. alba leaf extracts derived from Soxhlet extraction was evaluated based on cytotoxicity and antimigratory and antiinvasive properties. The dichloromethane extract exhibited the highest nitric oxide radical scavenging activity with a half-maximal inhibitory concentration (IC50) value of 780 μg/mL, promising cytotoxicity against HuCCA-1, MCF-7, and A-549 cells with IC50 values of 59.18, 62.20, and 103.25 μg/mL, respectively. CGA selectively inhibited the growth of MCF-7 cells with an IC50 value of 26.75 μg/mL and showed potent radical scavenging activity against DPPH radicals (IC50 = 18.85 μg/mL). An ethanolic extract derived from the gradient Soxhlet extraction suppressed A549 lung cancer cell migration and invasion more effectively than CGA with no migratory inhibition effect on noncancerous HaCaT cells. Furthermore, the ethanolic extract and CGA accelerated HaCaT wound closure at 20 µg/mL, which was the same as allantoin. Bioactive ingredients including triterpenes, steroids, phenolics, and flavonoids were mainly detected in all extracts. The highest content of CGA (52.23 g/100 g dry weight) was found in the ethanolic extract derived from the gradient Soxhlet extraction. These findings show the potency of the dichloromethane extract as a cytotoxic agent against various cancer types and the ethanolic extract as an antimetastatic agent by their antimigratory and antiinvasive activities. Diabetes mellitus (DM) is an endocrine-metabolic disorder that has limited approaches to treat effectively. Morus alba L., also known as mulberry, is a well-known medicinal plant, and its branch bark has shown hypoglycemic activity. It is rich in antioxidant and anti-inflammatory ingredients. In this study, we used metabolomics combined with network pharmacology to investigate the molecular mechanism and potential key targets of mulberry branch bark powder (MBBP) for treating DM. Serum metabolomics was performed to analyze the differences in metabolites and enrich metabolic pathways. Network pharmacology, based on systems biology tools, was applied to generate the pathway-target-compound network. Integrated analyses were then used to screen for key targets. To verify the obtained key targets, we used a molecular docking method and experimental validation. Our findings revealed that thirty-five endogenous metabolites contributed to the therapeutic impact of MBBP against DM. The analysis of 10 hub genes in the compound-target network partially supported the enrichment of metabolic pathways. Further analysis focused on two compounds (eugenol and mulberrofuran A) and three key targets (NOS2, MAOA, and CYP1A1). Mulberry leaf is a widely used protein feed and is often used as a strategy to reduce feed costs and improve meat quality in the livestock industry. However, to date, there is a lack of research on the improvement of meat quality using mulberry leaves, and the exact mechanisms are not yet known. The results showed that fermented mulberry leaves significantly reduced backfat content but had no significant effect on intramuscular fat (IMF). Lipidomic analysis showed that 98 and 303 differential lipid molecules (p < 0.05) were identified in adipose and muscle tissues, respectively, including triglycerides (TG), phosphatidylcholine, phosphatidylethanolamine, sphingolipids, and especially TG; therefore, we analysed the acyl carbon atom number of TG. The statistical results of acyl with different carbon atom numbers of TG in adipose tissue showed that the acyl group containing 13 carbon atoms (C13) in TG was significantly upregulated, whereas C15, C16, C17, and C23 were significantly downregulated, whereas in muscle tissue, the C12, C19, C23, C25, and C26 in TG were significantly downregulated. Acyl changes in TG were different for different numbers of carbon atoms in different tissues. We found that the correlations of C (14–18) in adipose tissue were higher, but in muscle tissue, the correlations of C (18–26) were higher. Through pathway enrichment analysis, we identified six and four metabolic pathways with the highest contributions of differential lipid metabolites in adipose and muscle tissues respectively. These findings suggest that fermented mulberry leaves improve meat quality mainly by inhibiting TG deposition by downregulating medium‐ and short‐chain fatty acids in backfat tissue and long‐chain fatty acids in muscle tissue. Typhlodromus (Anthoseius) bagdasarjani Wainstein & Arutunjan and T. (A.) khosrovensis Arutunjan have been frequently recorded from Iran. While investigating the population dynamics of these two predatory mite species on white mulberry in 2022 and 2023, in Tehran (Iran), our study unveiled intriguing morphological abnormalities and variations. Specifically, variations were observed in the number of dorsal shield solenostomes, insertion of ventrianal setae, shape of spermatheca and length of peritreme. Cardiovascular diseases are a broadly understood concept focusing on vascular and heart dysfunction. Lack of physical exercise, type 2 diabetes, obesity, hypertension, dyslipidemia, thromboembolism, and kidney and lung diseases all contribute to the development of heart and blood vessel dysfunction. Although effective and important, traditional treatment with diuretics, statins, beta blockers, calcium inhibitors, ACE inhibitors, and anti-platelet drugs remains a second-line treatment after dietary interventions and lifestyle changes. Scientists worldwide are still looking for an herbal product that would be effective and free from side effects, either taken together with or before the standard pharmacological intervention. Such herbal-originated medication therapy may include Morus alba L. (white mulberry), Elaeagnus rhamnoides (L.) A. Nelson (sea-buckthorn), Allium sativum L. (garlic), Convallaria majalis L. (lily of the valley), Leonurus cardiaca L. (motherwort), and Crataegus spp. (hawthorn). Valuable herbal raw materials include leaves, fruits, seeds, and even thorns. Morus alba L., a common traditional Chinese medicine (TCM) with a centuries-old medicinal history, owned various medicinal parts like Mori folium, Mori ramulus, Mori cortex and Mori fructus. Different medical parts exhibit distinct modern pharmacological effects. Mori folium exhibited analgesic, anti-inflammatory, hypoglycemic action and lipid-regulation effects. Mori ramulus owned antibacterial, anti-asthmatic and diuretic activities. Mori cortex showed counteraction action of pain, inflammatory, bacterial, and platelet aggregation. Mori fructus could decompose fat, lower blood lipids and prevent vascular sclerosis. The main chemical components in Morus alba L. covered flavonoids, phenolic compounds, alkaloids, and amino acids. This article comprehensively analyzed the recent literature related to chemical components and pharmacological actions of M. alba L., summarizing 198 of ingredients and described the modern activities of different extracts and the bioactive constituents in the four parts from M. alba L. These results fully demonstrated the medicinal value of M. alba L., provided valuable references for further comprehensive development, and layed the foundation for the utilization of M. alba L. Magnesium (Mg) deficiency is a major factor limiting the growth and development of plants. Mulberry (Morus alba L.) is an important fruit tree crop that requires Mg for optimal growth and yield, especially in acid soils. However, the molecular mechanism of Mg stress tolerance in mulberry plants remains unknown. In this study, we used next-generation sequencing technology and biochemical analysis to profile the transcriptome and physiological changes of mulberry leaves under different Mg treatments (deficiency: 0 mM, low: 1 mM, moderate low: 2 mM, sufficiency: 3 mM, toxicity: 6 mM, higher toxicity: 9 mM) as T1, T2, T3, CK, T4, T5 treatments, respectively, for 20 days. The results showed that Mg imbalance altered the antioxidant enzymatic activities, such as catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD), and non-enzymatic, including soluble protein, soluble sugar, malondialdehyde (MDA), and proline (PRO), contents of the plant. The Mg imbalances disrupted the ultrastructures of the vital components of chloroplast and mitochondria relative to the control. The transcriptome data reveal that 11,030 genes were differentially expressed (DEGs). Genes related to the photosynthetic processes (CAB40, CAB7, CAB6A, CAB-151, CAP10A) and chlorophyll degradation (PAO, CHLASE1, SGR) were altered. Antioxidant genes such as PER42, PER21, and PER47 were downregulated, but DFR was upregulated. The carbohydrate metabolism pathway was significantly altered, while those involved in energy metabolism processes were perturbed under high Mg treatment compared with control. We also identified several candidate genes associated with magnesium homeostasis via RT-qPCR validation analysis, which provided valuable information for further functional characterization studies such as promoter activity assay or gene overexpression experiments using transient expression systems. Traditional Chinese medicine (TCM) possesses the potential of providing good curative effects with no side effects for the effective management of slow transit constipation (STC), an intestinal disease characterized by colonic dyskinesia. Mulberry leaves (Morus alba L.) and black sesame (Sesamum indicum L.), referred to as SH, are processed and conditioned as per standardized protocols. SH has applications as food and medicine. Accordingly, we investigated the therapeutic potential of SH in alleviating STC. The analysis of SH composition identified a total of 504 compounds. The intervention with SH significantly improved intestinal motility, reduced the time for the first black stool, increased antioxidant activity, and enhanced water content, thereby effectively alleviating colon damage caused by STC. Transcriptome analysis revealed the SH in the treatment of STC related to SOD1, MUC2, and AQP1. The analysis of 16S rRNA gene sequences indicated notable differences in the abundance of 10 bacteria between the SH and model. Metabolomic analysis further revealed that SH supplementation increased the levels of nine metabolites associated with STC. Integrative analysis revealed that SH modulated amino acid metabolism, balanced intestinal flora, and targeted key genes (i.e., SOD1, MUC2, AQP1) to exert its effects. Background: Obesity is a serious disease with an alarmingly high incidence that can lead to other complications in both humans and dogs. Similar to humans, obesity can cause metabolic diseases such as diabetes in dogs. Natural products may be the preferred intervention for metabolic diseases such as obesity. The compound 1-deoxynojirimycin, present in Morus leaves and other sources has antiobesity effects. The possible antiobesity effect of 1-deoxynojirimycin containing Morus alba leaf-based food was studied in healthy companion dogs (n = 46) visiting the veterinary clinic without a history of diseases. Body weight, body condition score (BCS), blood-related parameters, and other vital parameters of the dogs were studied. Whole-transcriptome of blood and gut microbiome analysis was also carried out to investigate the possible mechanisms of action and role of changes in the gut microbiome due to treatment. Results: After 90 days of treatment, a significant antiobesity effect of the treatment food was observed through the reduction of weight, BCS, and blood-related parameters. A whole-transcriptome study revealed differentially expressed target genes important in obesity and diabetes-related pathways such as MLXIPL, CREB3L1, EGR1, ACTA2, SERPINE1, NOTCH3, and CXCL8. Gut microbiome analysis also revealed a significant difference in alpha and beta-diversity parameters in the treatment group. Similarly, the microbiota known for their health-promoting effects such as Lactobacillus ruminis, and Weissella hellenica were abundant (increased) in the treatment group. The predicted functional pathways related to obesity were also differentially abundant between groups. Conclusions: 1-Deoxynojirimycin-containing treatment food have been shown to significantly improve obesity. The identified genes, pathways, and gut microbiome-related results may be pursued in further studies to develop 1-deoxynojirimycin-based products as candidates against obesity. Objective: The plant known as white mulberry, or Morus alba L., has several uses and is employed in traditional medicine in many different cultures. The objectives of this study are to determine the total phenolic component and antioxidant levels of the methanol and ethanol extracts of leaves of Morus alba L. (White mulberry). Methods: Using the techniques of DPPH (1,1-diphenyl-2-picrylhydrazyl), FRAP (Iron ion that decreases antioxidant power), and CUPRAC (Copper ion that reduces antioxidant capacity), the antioxidant activities of methanol and ethanol extracts of Morus alba L. leaves were examined. Additionally, the Folin-Ciocalteu Reagent (FCR) technique was used to evaluate the total phenolic content of the ethanol and methanol extracts. To determine the antioxidant capacity of extracts, reference samples were used to prepare various concentrations ranging from 1 to 100 µg/mL. The extract's equivalent antioxidant capacity was determined using sample concentrations of 125, 250, and 500 µg/mL. Results: The methanol and ethanol extracts of the leaves of the Morus alba L. (White mulberry) plant were found to have the greatest level of phenolic compounds at the 500 µg/mL concentration of extracts. It was determined that the Trolox Eq value of methanol extract was higher in the DPPH, FRAP and FCR methods, and the Trolox Eq value of the ethanol extract was higher in the CUPRAC method. Conclusion: The possible folk medicinal application of Morus alba L. is supported by this study. Mulberry leaves (Morus alba L.) exhibit antidiabetic properties; however, their taste is unappetizing, leading to limited practical applications. Although Fu brick tea has a favorable flavor, its hypoglycemic effects are weak. Hence, we combined the two to create mulberry leaf Fu brick tea (SF) and investigated its sensory perception, safety, microorganism content, key metabolites, and impact on hyperglycemic mice using the same raw materials and techniques to prepare Fu brick tea (F) and mulberry leaf brick tea (S) individually. Our study showed that SF, which comprises blended mulberry leaf primary dark tea and primary dark tea (1 : 3 g/g), was sweeter and smoother, with a median lethal dose of 22.3 g/kg, indicating that it is safe for consumption. Compared to F, caffeine levels were reduced in SF and 1-deoxynojirimycin and aminobutyric acid levels were increased. Phenolic substances and flavonoids showed increased levels in SF compared with S. These changes contribute to the taste and functionality of SF. The primary microorganisms in SF were Bacillus, Lactobacillus, and Aspergillus, which contributed to the improved quality. SF demonstrated its antidiabetic effects by reducing the blood sugar levels of hyperglycemic mice, restoring the function of pancreatic cells, reducing lipid levels, and improving the antioxidant capacity of the liver. BACKGROUND: With the requirements of environmental, cost and economic sustainability, new sources of alternative proteins in the livestock industry are receiving increasing attention. Mulberry (Morus alba L.) leaves are a unique feed resource because of their high protein content and large availability. Therefore, mining sustainable protein suitable for the animal husbandry industry in sericulture resources could achieve a win–win situation. RESULTS: The protein content in mulberry leaves is 232.10–386.16 g kg−1, and the mean value of crude fat content is 43.76 ± 8.48 g kg−1, which has the advantages of protein content and energy. In addition, the average content of phytic acid in mulberry leaves is only 1.88 ± 0.56 g kg−1, which means that it is not inhibited in terms of nutrient absorption. Meanwhile, the digestibility of protein was Bean pulp > Sample 8 ≈ Alfalfa ≈ Sample 13 ≈ Cottonseed meal > Fish meal, and the β‐turn and particle size of mulberry leaf protein are more conducive to digestion in vitro. Furthermore, the protein of Sample 13 had the richest essential amino acids (252.00 g kg−1) and the highest essential amino acid index (EAAI), which was superior to conventional feed protein. In addition, the partial substitution of mulberry leaf protein (15%) significantly increased the EAAI value of conventional feed protein. However, to balance nutrition, it is necessary to combine mulberry leaf protein with other proteins to further broaden its application field. Mulberry (Morus alba L.), a significant fruit tree crop, requires magnesium (Mg) for its optimal growth and productivity. Nonetheless, our understanding of the molecular basis underlying magnesium stress tolerance in mulberry plants remains unexplored. In our previous study, we identified several differential candidate genes associated with Mg homeostasis via transcriptome analysis, including the xyloglucan endotransglucosylase/hydrolase (XTH) gene family. The XTH gene family is crucial for plant cell wall reconstruction and stress responses. These genes have been identified and thoroughly investigated in various plant species. However, there is no research pertaining to XTH genes within the M. alba plant. This research systematically examined the M. alba XTH (MaXTH) gene family at the genomic level using a bioinformatic approach. In total, 22 MaXTH genes were discovered and contained the Glyco_hydro_16 and XET_C conserved domains. The MaXTHs were categorized into five distinct groups by their phylogenetic relationships. The gene structure possesses four exons and three introns. Furthermore, the MaXTH gene promoter analysis reveals a plethora of cis-regulatory elements, mainly stress responsiveness, phytohormone responsiveness, and growth and development. GO analysis indicated that MaXTHs encode proteins that exhibit xyloglucan xyloglucosyl transferase and hydrolase activities in addition to cell wall biogenesis as well as xyloglucan and carbohydrate metabolic processes. Moreover, a synteny analysis unveiled an evolutionary relationship between the XTH genes in M. alba and those in three other species: A. thaliana, P. trichocarpa, and Zea mays. Expression profiles from RNA-Seq data displayed distinct expression patterns of XTH genes in M. alba leaf tissue during Mg treatments. Real-time quantitative PCR analysis confirmed the expression of the MaXTH genes in Mg stress response. Background: Studies have reported that the phytochemical content of Mulberry (Morus alba Linn.) is influenced by the area where it grows. On the other hand, the study of the bioactivity and toxicity of mulberry leaves from Brunei Darussalam still needs to be completed. In particular, the investigation regarding the safe dose for Mulberry's application from Brunei Darussalam has yet to be studied. Hence, toxicity information must be considered even though the community has used it for generations. Aim: This study investigated Morus alba ethanolic leaf extract (MAE) to observe the acute toxicity in mice. Methods: In particular, this study utilized 12 female Institute of Cancer Research mice, 8 weeks old, divided into 2 groups: the control group and the MAE group (2,000 mg/kg single dose). Physiology, hematology, biochemistry, and histology were analyzed during the study. Results: The examination result indicated no mortality and behavioral changes throughout the testing period. However, the mice developed mild anemia and leukopenia, followed by decreased numbers of neutrophils, lymphocytes, and monocytes. In addition, the mice developed a mild hepatocellular injury, indicated by significant (p < 0.05) elevations of both alanine aminotransferase (ALT) and aspartate transaminase (AST). The histopathological findings of the liver were also consistent with the increment of ALT and AST, indicating mild hepatocellular necrosis through the eosinophilic cytoplasm and pyknosis (p > 0.05). Conclusion: It was evident that a single oral administration of MAE was not lethal for mice (LD50, which was higher than 2,000 mg/kg). Caeoma mori (≡ Aecidium mori), known as the mulberry rust which is an anamorphic rust fungus forming only aecidioid uredinia, were found on Morus alba in Ibaraki and Saitama Prefectures, Japan. Molecular phylogenetic analyses using the combined dataset of sequences from 28S and 18S of the nuclear ribosomal RNA gene and Cytochrome-c-oxidase subunit 3 of the mitochondrial DNA revealed that this anamorphic rust fungus was a member of the clade composed of the genus Gymnosporangium. Therefore, a new combination, Gymnosporangium mori is proposed for this species. Additionally, a new combination, G. brucense for Roestelia brucensis is proposed by phylogenetic evidence. Both morphological and molecular markers have been extensively used to evaluate genetic diversity; however, molecular markers are considered more reliable and can lead to improved reproductive efficiency. This study utilized inter-primer binding site (iPBS) markers to examine the genetic diversity and population structure of thirty mulberry accessions from the districts of Sahiwal and Faisalabad, Pakistan. These mulberry accessions belonged to three species: Morus nigra (n = 13), Morus alba (n = 12), and Morus rubra (n = 5). The use of nine iPBS primers in this study provided a comprehensive understanding of genetic diversity among the selected mulberry accessions. Nine iPBS primers were used in the study and generated 431 bands with allelic frequencies ranging from 21 to 75 and band sizes from 200 to 1500 base pairs. The primer 2230 showed the highest polymorphic information content (PIC) value of 0.47 and the highest Shannon's information index (I = 0.53). The Morus nigra accessions had the highest levels of expected heterozygosity (He = 0.30), unbiased expected heterozygosity (µHe = 0.33), and Shannon's information index (I = 0.45). The molecular variance analysis (AMOVA) revealed a high degree of genetic variation, as estimated by the pairwise PhiPT value of 0.21, which was significant at the p < 0.001 *** level. The neighbor joining tree, principal coordinate analysis, and structure analysis grouped the 30 mulberry accessions into four main clusters. The distinct grouping of accessions SWLS14, SWLS6, FSDS30, and SWLS7 validated their notable genetic distinctiveness. The iminosugar 1-deoxynojirimicyn (DNJ) contained in mulberry leaves has displayed systemic beneficial effects against disorders of carbohydrate metabolism. Nevertheless, its effect is impaired by the short half-life. Alginate-based carriers were developed to encapsulate a DNJ-rich mulberry extract: Ca-alginate beads, obtained by external gelation, and spray-dried alginate microparticles (SDMs). Mean size and distribution, morphology, drug loading, encapsulation efficiency, experimental yield, and release characteristics were determined for the two formulations. Ca-alginate beads and SDMs exhibited an encapsulation efficiency of about 54% and 98%, respectively, and a DNJ loading in the range of 0.43–0.63 μg/mg. The in vitro release study demonstrated the carriers' capability in controlling the DNJ release in acid and basic conditions (<50% in 5 h), due to electrostatic interactions, which were demonstrated by 1H-NMR relaxometry studies. Medicinal preparations made from plant materials have been widely used for many years due to their high pharmacological efficacy and safety of use. Therefore, a study of white mulberry fruits (Morus alba L.) for the content of substances is very important for the pharmaceutical industry such as flavonoids, alkaloids, polysaccharides, minerals, vitamins, and amino acids. White mulberry has a wide distribution area around the world, including in Kazakhstan, especially in the southern regions of the country (Almaty, Zhambyl, and Turkestan regions). The composition of the fruits of this plant is significantly influenced by the area where the trees grow, and therefore, the establishment of a specific composition of biologically active substances is very important. In the course of this study, such methods as gas chromatography were used—mass spectrometry of an extract obtained using carbon dioxide under subcritical conditions, atomic absorption, gravimetric, and spectrophotometric methods. As a result, for the first time in Kazakhstan, the composition of white mulberry fruits (Morus alba L.), namely, biologically active substances, has been identified, such as alkaloids, flavonoids, vitamins, macro- and microelements, and amino acids and fatty acids; in addition, the percentage composition of the above compounds has been determined. The results of the study show a comparative analysis of the component composition of white mulberry fruits (Morus alba L.) in various areas of tree growth, including outside of Kazakhstan. Taxilli Herba (TH) is famous for semi-parasitic feature, with hundreds of host species. The content of bioactive constituents varies in TH from different hosts. To gain insight into the molecular mechanisms of the synthesis and accumulation of bioactive constituents in TH from different hosts, we performed proteomics combined with metabolomics to investigate TH from two common hosts of Morus alba L. and Liquidambar formosana Hance. Tandem mass tag (TMT)-based quantitative proteomics were used for analysis. A total of 533 differentially expressed proteins were detected, of which 259 were significantly up-regulated and 274 significantly down-regulated in expression levels. DEPs were shown to be mostly engaged in cellular process and metabolic process by GO analysis. DEPs were considerably abundant in amino acid metabolic pathways, according to KEGG pathway analysis. 16 bioactive constituents were quantitatively determined by ultrafast liquid chromatography simultaneously coupled with triple quadrupole/linear ion trap tandem mass spectrometry (UFLC-QTRAP-MS/MS). Meanwhile, the changes in the contents of 16 bioactive constituents were mostly compatible with the expression patterns of relevant structural proteins. The results of this study will provide basic information about the role of protein in the secondary metabolism biosynthesis pathway and the molecular mechanism of the quality formation of TH. Background: Leaf coloration in plants, attributed to anthocyanin compounds, plays a crucial role in various physiological functions, and also for pharmaceutical and horticultural uses. However, the molecular mechanisms governing leaf coloration and the physiological significance of anthocyanins in leaves remain poorly understood. Results: In this study, we investigated leaf color variation in two closely related mulberry genotypes, one with purplish-red young leaves (EP) and another with normal leaf color (EW). We integrated transcriptomic and metabolomic approaches to gain insights into the metabolic and genetic basis of purplish-red leaf development in mulberry. Our results revealed that flavonoid biosynthesis, particularly the accumulation of delphinidin-3-O-glucoside, is a key determinant of leaf color. Additionally, the up-regulation of CHS genes and transcription factors, including MYB family members, likely contributes to the increased flavonoid content in purplish-red leaves. Conclusion: These findings enhance our understanding of the molecular mechanisms responsible for the purplish coloration observed in mulberry leaves and also offer supporting evidence for the hypothesis that anthocyanins serve a protective function in plant tissues until the processes of light absorption and carbon fixation reach maturity, thereby ensuring a balanced equilibrium between energy capture and utilization. Gastrointestinal cancers are a major global health challenge, with high mortality rates. This study investigated the anti-cancer activities of 30 monomers extracted from Morus alba L. (mulberry) against gastrointestinal cancers. Toxicological assessments revealed that most of the compounds, particularly immunotoxicity, exhibit some level of toxicity, but it is generally not life-threatening under normal conditions. Among these components, Sanggenol L, Sanggenon C, Kuwanon H, 3′-Geranyl-3-prenyl-5,7,2′,4′-tetrahydroxyflavone, Morusinol, Mulberrin, Moracin P, Kuwanon E, and Kuwanon A demonstrate significant anti-cancer properties against various gastrointestinal cancers, including colon, pancreatic, and gastric cancers. The anti-cancer mechanism of these chemical components was explored in gastric cancer cells, revealing that they inhibit cell cycle and DNA replication-related gene expression, leading to the effective suppression of tumor cell growth. Additionally, they induced unfolded protein response (UPR) and endoplasmic reticulum (ER) stress, potentially resulting in DNA damage, autophagy, and cell death. Moracin P, an active monomer characterized as a 2-arylbenzofuran, was found to induce ER stress and promote apoptosis in gastric cancer cells, confirming its potential to inhibit tumor cell growth in vitro and in vivo. These findings highlight the therapeutic potential of Morus alba L. monomers in gastrointestinal cancers, especially focusing on Moracin P as a potent inducer of ER stress and apoptosis. This paper presents for the first time the results of studying the total content of antioxidants in the seeds of Morus L. The composition of fatty and steric acids of the seeds of mulberry fruits Morus L. was also studied: polymorphic species Morus alba (white-fruited, pink-fruited, black-fruited) and Morus nigra (Khartut variety). As a result of the studies, 12 fatty acids were identified. The main ones were linoleic acid, palmitic oleic acid, and stearic acid. The predominant fatty acid in all samples was linoleic, which varied from 67.5 to 79.0%. 15 sterols were found, among which β-Sitosterol accounts for about 90%. Determination of the total antioxidant content of Morus alba seeds (white-fruited, pink-fruited, black-fruited forms) and Morus nigra (Khartut variety) revealed the accumulation of antioxidants in all samples. Increased oxidative stress and acetylcholinesterase (AChE) activity are key pathological characters contributing to the memory disorders. Thus, drugs targeting both oxidative stress and AChE are being explored for the management of cognitive dysfunction. Morus alba fruits (commonly consumed for its high nutritious value) are known to have antioxidant and AChE inhibitory effects. However, the role of Morus alba fruits in the management of memory disorders has not reported yet. This investigation was conducted to assess the antioxidant and AChE inhibitory potential of Morus alba fruit extracts in-vitro and to identify the components responsible for such effects. Further, the obtained bioactive component was studied for possible memory improvement effects against streptozotocin (STZ) induced dementia. To isolate the bioactive component in-vitro DPPH and AChE assays guided fractionation was performed. Memory functions in mice were determined using Morris Water Maze test while brain biochemical parameters were measured to understand the mechanism of action. In-vitro assays revealed strong AChE and DPPH inhibitory potential of methanol extract (ME), therefore, it was further fractionated. Among various fractions obtained, ethyl-acetate fraction (EAF) was found to possess marked AChE and DPPH inhibitory activities. On subsequent fractionation of EAF, bioactivity of obtained sub-fractions was found to be inferior to EAF. Further, both ME and EAF improved STZ (intracerebroventricular) induced cognitive dysfunction in animals by restoring endogenous antioxidant status (superoxide dismutase and reduced glutathione) and reducing thiobarbituric acid reactive species and nitric oxide levels along with brain AChE and myeloperoxidase activity. TLC densitometric studies showed appreciable levels of phenolic acids and quercetin in both EAF and ME. It can be concluded that Morus alba fruit extract has the ability to modulate cholinergic and oxidative system due to presence of phenolic and flavonoid compounds and hence, could aid in the management of memory disorders. In this work, Pollen analysis was carried out on two honey samples from two different localities (Dabhad village and Vaghavaadi village) of Kheralu taluka, Mehsana district, Gujarat. Melissopalynological examinations were made to isolate and identify pollen types in the honey. The aim of the study is to determine whether the honey samples are unifloral or multifloral. Moreover, to determine pollen types to understand the forage patterns of bee species. Pollen analysis was performed using the acetolysis method. A total of seventeen morphotypes of pollen were identified. Brassica juncea (L.) Czern., Morus alba L., Sonchus oleraceus L., Solanum virginianum L., Foeniculum vulgare Mill., Ricinus communis L., Achyranthes aspera L., Tridax procumbens (L.), Amaranthus viridis L., Parthenium hysterophorus L., Cyanthillium cinereum (L.) H.Rob., Digera muricata (L.) Mart., Tagetes erecta L., Peristrophe bicalyculata (Retz) Nees, Eclipta prostrata (L.), Setaria glauca (L.) Beauv. & unknown pollen of plant. Among these plants, the pollen of Brassica juncea (L.) Czern. was recorded with 53.03% and 41.71% in the Dabhad locality as well as the Vaghavaadi locality with the highest frequency percentage. Metabolic product accumulation exhibited variations among mulberry (Morus alba L.) leaves (MLs) at distinct growth stages, and this assessment was conducted using a combination of analytical techniques including high-performance liquid chromatography (HPLC), gas chromatography–mass spectrometry (GC-MS), and liquid chromatography–mass spectrometry (LC-MS). Multivariate analysis was applied to the data, and the findings were correlated with antioxidant activity and α-glucosidase inhibitory effects in vitro. Statistical analyses divided the 27 batches of MLs at different growth stages into three distinct groups. Three new compounds (1, 11, and 12), together with 32 known ones, were isolated from the root bark of Morus alba L. using various chromatographic methods. The structures of the undescribed compounds were elucidated based on 1D, 2D NMR, and HRESIMS dataanalysis, while the known ones were identified by comparison of their spectroscopic data with those reported in the literature. All the isolates were evaluated for their cytotoxic activities against human gastric cancer HGC27 cells by CCK-8 assay. Among them, compounds 5, 8, 10, and 30 exhibited cytotoxic activities on HGC27 cells with IC50 values of 33.76 ± 2.64 μM, 28.94 ± 0.72 μM, 6.08 ± 0.34 μM, and 10.24 ± 0.89 μM, respectively. We have successfully synthesized ZnFe2O4/SiO2—Morus alba L. nanoparticles as a drug delivery agent. The nanostructural and optical properties of ZnFe2O4/SiO2—Morus alba L. were evaluated using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy–energy dispersive X-ray (SEM–EDX) spectroscopy, transmission electron microscopy (TEM), and ultraviolet–visible spectroscopy. Antibacterial and drug delivery tests were conducted to examine the antibacterial activity and drug delivery performance of the nanoparticles, respectively. The peaks in the XRD patterns indicated the existence of ZnFe2O4 and SiO2 phases at 2θ = 35.4° and 22°–28°, respectively, in the sample. Meanwhile, the FTIR spectrum showed the functional group of ZnFe2O4—Morus alba L. at wavenumbers of 400–600 cm−1 and Si–O–Si at 1086 and 950 cm−1. The Si–O–Fe peak was also detected at 541–575 cm−1. The calculated bandgap energy was in the range of 3.003–3.218 eV. The TEM and SEM images revealed that the particle size varied in the range of 28.7–47.3 nm and confirmed the formation of a composite. The samples exhibited excellent antibacterial activity and inhibited the growth of S. aureus and E. coli by up to 72% and 78%, respectively. The samples also possessed a desirable doxorubicin (DOX) loading, indicated by the appearance of DOX absorption peaks at wavelengths of 200–250 nm and 450–550 nm. The objective of this study was to analyze the content of vitamin C, total phenols, total anthocyanins, flavan-3-ols and antioxidant activity in fruits of five berry fruit species: blackcurrant variety "Rosenthal", white mulberry local ecotype, black chokeberry variety "Viking", blackberry variety "Thornfree" and pomegranate autochthonous variety "Karamustafa". Determination of vitamin C was performed by classical analytical method. Total phenols, total anthocyanins, flavan-3-ols and antioxidant activity were analysed by spectrophotometric methods. The obtained results from the analysis for berry fruits showed high antioxidant activity, over 70% and high content of polyphenols. According to the genotype, black chokeberry had the highest content of total phenols (23.9 mg g-1 FW (fresh weight), anthocyanins (6.1 mg g-1 FW) and flavan-3-ols (2.8 mg g-1 FW). Blackcurrant had highest content of vitamin C (216.0 mg 100 g-1 FW) and antioxidant activity (86.2% inhibition). A positive correlation was found among all the studied fruit species. A strong positive correlation of vitamin C with antioxidant activity was determined, as well as between total phenols and total anthocyanins, i.e. flavan-3-ols. A weak negative correlation was only observed for catechin in relation to vitamin C, inhibitory antioxidant activity and vitamin C antioxidant activity. [ABSTRACT FROM AUTHOR]

The application of Morus alba L. in traditional oriental medicine and cuisine has resulted in numerous studies on its health-promoting effects. However, if the process is not monitored by the manufacturers, the processing of the leaves alters the obtained health-promoting properties and results in different health qualities in the final composition of dietary supplements. This article aims to analyze changes (using the HPLC/DAD method) in the proposed conditioned mulberry leaves in terms of key compounds (phenolic acids and flavonols) responsible for antioxidant activity after being digested in in vitro conditions. The analyzed material was leaves of white mulberry (Morus alba L.) cv. Żółwińska wielkolistna, conditioned (1–4 h) and non-conditioned. The conditioning process of mulberry proposed here, e.g., for industry production, resulted in variable transformations of polyphenols during in vitro digestion. For many polyphenols, especially those shown in the highest amounts, significant correlations were found between their content and conditioning, as well as the stage of digestion. In the case of mulberry infusions, the amounts of individual polyphenols were several times lower than in the preparations, which was due to the degree of dilution. Their amounts tended to decrease in the course of digestion. Taking this into account, it seems justified to continue research on the in vivo bioavailability of bioactive components from conditioned Morus alba L. leaves. [ABSTRACT FROM AUTHOR]

Introduction: In traditional Chinese medicine, the root bark of Morus alba L. is used to treat respiratory infections. Recently, anti-inflammatory and multiple antiinfective activities (against influenza viruses, corona virus 2, S. aureus, and S. pneumoniae) were shown in vitro for a standardized root bark extract from M. alba (MA60). Sanggenons C and D were identified as major active constituents of MA60. The aim of the present preclinical study was to evaluate, whether these findings are transferable to an in vivo setting. Methods: MA60 was orally administered to female BALB/c mice to determine 1) the maximum tolerated dose (MTD) in an acute toxicity study and 2) its antiinfluenza virus and anti-inflammatory effects in an efficacy study. A further aim was to evaluate whether there is a correlation between the obtained results and the amount of sanggenons C and D in serum and tissues. For the quantitation of the marker compounds sanggenons C and D in serum and tissue samples an UPLC-ESI-MS method was developed and validated. Results: In our study setting, the MTD was reached at 100 mg/kg. In the efficacy study, the treatment effects were moderate. Dose-dependent quantities of sanggenon C in serum and sanggenon D in liver samples were detected. Only very low concentrations of sanggenons C and D were determined in lung samples and none of these compounds was found in spleen samples. There was no compound accumulation when MA60 was administered repeatedly. Discussion: The herein determined low serum concentration after oral application once daily encourages the use of an alternative application route like intravenous, inhalation or intranasal administration and/or multiple dosing in further trials. The established method for the quantitation of the marker sanggenon compounds in tissue samples serves as a basis to determine pharmacokinetic parameters such as their bioavailability in future studies. Less common tropical fruits have been the subject of little research, leaving a vast field to be explored. In this context, a comprehensive study was carried out on the bioactive compounds and antioxidant capacity of 51 non-traditional fruits consumed in Ecuador. Vitamin C, organic acids, carotenoids, and phenolic compounds were evaluated using microextraction and rapid resolution liquid chromatography (RRLC) techniques, while antioxidant activity was measured using microplate readings. The results showed high levels of vitamin C (768.2 mg/100 g DW) in Dovyalis hebecarpa, total organic acids (37.2 g/100 g DW) in Passiflora tripartita, carotenoids (487.0 mg/100 g DW) in Momordica charantia, phenolic compounds (535.4 mg/g DW) in Nephelium lappaceum, Pourouma cecropiifolia (161.4 µmol TE/g DW) and Morus alba (80.5 µmol AAE/g DW) in antioxidant activity. Effective extraction of carotenoids was also observed using a mixture of methanol: acetone: dichloromethane (1:1:2) with an extraction time of 2 min, while an 80% solution of 0.1% acidified methanol with hydrochloric acid with an extraction time of 3 min was highly effective for phenolics in fruit. These results provide a valuable basis for optimising future extraction processes of bioactive compounds from non-traditional fruits, with significant implications for their potential use in various nutritional and pharmaceutical contexts. The silkworm industry plays a crucial role in providing employment opportunities, particularly in regions like Karnataka, Andhra Pradesh, Assam, West Bengal, Jharkhand, and Tamil Nadu. This industry heavily relies on mulberry plants (Morus alba L.) as the primary food source for silkworms (Bombyx mori). The quality of mulberry leaves directly influences cocoon quality and colour, making Moriculture pivotal in sericulture. Mulberry plants face various challenges, including diseases and pest infestations, affecting leaf growth and silk quality. This study, conducted at Holy Cross College, Tamil Nadu, aims to identify and analyze arthropods damaging mulberry leaves. Arthropods from two classes, Insecta and Arachnida, were identified and classified into orders, families, and genera. Hemiptera emerged as the most diverse order among Insects. Notably, sap-sucking insects like mealy bugs, hoppers, and jassids were observed, consistent with previous research. The Shannon Weinner index, Species richness and Species evenness were found to be more in the mulberry garden as per this study. Mulberry is a multipurpose tree in horticulture. Besides nutritional importance, it is used for raising silkworms, fodder and landscape. These is quite a wide genetic diversity among different mulberry genotypes. The present study was undertaken to evaluate certain morphological and biochemical properties of 20 genotypes of Morus alba, M. rubra, and M. nigra species distributed in Golestan Province, northern Iran. The highest leaf length (15.51 cm), petiole length (5.37 cm), vitamin C (4.34 mg/100 g) and phenol (31.89 mg/g) were recorded in M12 genotype. The highest fruit length (5.52 cm), pedicle length (17 mm), soluble solids (18.26%) and sucrose (31.64 mg/g) were recorded in M10 genotype. The amount of glucose (288.82 mg/g) and fructose (121.97 mg/g) were also dominant in M17 genotype. The utmost fruit fresh and dry weights (0.3-67.80 g), fruit diameter (1.99 cm), leaf width (11.94 cm) and flavonoids (24.67 mg/g) were also observed in the M20 genotype. The M1 and M2 genotypes had higher anthocyanin (1.13 µmol/g) and total antioxidant activity (66.71%) compared to other genotypes. Furthermore, fruit fresh and dry weights were positively correlated with fruit length and diameter, pedicle length, and leaf and petiole length. The highest positive correlation was observed among anthocyanin, vitamin C and phenols. The morphological and biochemical traits explained 99 % of the total variance of four and three main components. The results of cluster analysis grouped these genotypes into 3 main groups. Therefore, the highest heritability and genetic advance were found for fruit weight, petiole length, leaf width, antioxidant activity, fructose and flavonoids. Simple Summary: Edible trees and shrubs have been evaluated to improve animal nutrition and mitigate the emission of greenhouse gasses from ruminants. In the present study, researchers visited some farmers and identified eight potentially edible Brazilian trees. Nutritional value and gas (and methane) production from these edible trees were evaluated. The leaves of Aroeira, Candeia, and Jatobá had limited nutritional value. Samples of Santa Bárbara trees, Mulberry, and Tithonia showed an improved nutritional value. Furthermore, Tithonia decreased methane emissions and is a potentially edible tree for multifunctional redesigned ruminant production systems. The present study aims to evaluate the nutritional value of different tree and shrub leaves in Brazilian ruminant production systems. Eight potentially edible trees and shrubs were identified from interviews with 30 ruminant producers: Aroeira (ARO; Lithraea molleoides), Black Mulberry (BMU; Morus nigra), Candeia (CAN; Eremanthus erythropappus), Jatobá (JAT; Hymenaea courbaril), Gliricídia (GLI; Gliricidia sepium), Santa Bárbara tree (SBT; Mélia azedarach), Tithonia (TIT; Tithonia diversifolia), and White Mulberry (WMU; Morus alba). Four leaf samples of each edible tree were sampled, and chemical analyses and in vitro assays were performed. Edible trees (except CAN and JAT) had lower neutral detergent fiber content than Mombasa grass. In addition, SBT, BMU, WMU, and TIT had lower fiber content than the other evaluated edible trees. Consequently, SBT, TIT, BMU, and WMU had improved dry matter degradation. Among the edible trees and shrubs, SMW and WMU increased the potential for gas production (a parameter). On the other hand, CAN decreased the estimated gas production 48 h after incubation. Furthermore, TIT decreased methane production up to 24 h after in vitro fermentation. Thus, except ARO, CAN, and JAT, the edible trees evaluated in the present study are potential feeds in moderate- to high-producing animals. Mulberry plants (Morus alba) have leaf shapes, ranging from unlobed to lobed, which are crucial for yield, growth, and adaptability, indicating their ability to adapt to their environment. Competing endogenous RNAs (ceRNAs) constitute a web of RNAs within the organism's transcriptional regulatory system, including protein-coding genes (mRNAs), microRNAs (miRNAs), long non-coding RNAs (lncRNAs), circular RNAs (circRNAs), and others. In this study, samples for ceRNA sequencing were categorized into two groups: whole leaves and lobed leaves, each group with three replicates. In addition, we isolated, cloned, and characterized the precursor miRNA (miR156x) from the leaves of M. alba. miR156x precursor had a length of 107 base pairs and a minimum folding free energy of 50.27 kcal/mol. We constructed a pCAMBIA-35S-GUS-miR156x dual overexpression vector and established a transient transformation system for mulberry. At an optimal transformation solution (OD600 = 0.7), the GUS gene showed a higher expression in the leaves of transiently transformed mulberry with miR156x overexpression, four days after transformation, while the target genes of miR156x had decreased expression in the same leaves. Investigations into the transgenic mulberry plants uncovered various modifications to physio-chemical parameters including POD, SOD, PRO, MDA, soluble proteins and sugars, and chlorophyl content. miRNAs in the plants were found to act as negative regulators of gene expression in response to changes in leaf shape regulation, which was confirmed in vitro using dual-luciferase reporter assays. Subsequently, we cloned Maspl3 in vitro and conducted GST-Pull down assays, obtaining multiple proteins that interacted with the Maspl3 gene. Genetic resources of mulberry trees are commonly preserved as trophosomes, which are vulnerable to environmental factors, such as natural disasters, diseases, and pests. This study establishes a basic protocol for ultra-low temperature cryopreservation of mulberry trees using a two-step freezing process. The procedure was established using the ‘‘Daeshim’’ variety and then tested on genetic resources from 24 other mulberry varieties. Samples were first dried to a moisture content of 33–43% in a low-temperature forced-air chamber at −5 °C, then slowly frozen from −5 °C to −20 °C, and preserved in liquid nitrogen (−196 °C). To determine the regeneration rate, isolated dormant buds were inoculated into MS basal medium, and grown shoots were grafted onto 1-year-old rootstock via chip budding and then cultured. After freezing in liquid nitrogen, the ‘‘Daeshim’’ variety exhibited a survival and regeneration rate of more than 70% and 50%, respectively. Applying the two-step freezing process to genetic resources from 24 mulberry species yielded average survival and regeneration rates of 85.3% and 75.5%, respectively. Morus alba showed survival and regeneration rates of 100%, confirming the efficacy of the two-step freezing method. These results indicate the high feasibility of ultra-low-temperature cryopreservation through two-step freezing of dormant buds from mulberry genetic resources.
