The search for new forms to extend foodstuffs’ shelf-life through the use of natural compounds and extracts continues to grow among researchers and the industry. In this line of thought, the main objective of this work was to develop, characterize and evaluate the effectiveness of an active food packaging based on polylactic acid (PLA) and incorporated with natural extracts obtained from grape and/or pomegranate by-products. In vitro, antioxidant methods were applied to evaluate the antioxidant capacity of three extracts: wort extract, freeze-dried pomegranate peels extract (PPE-FD), and natural pomegranate peels (PPE-N). Punicalagin (A + B) and ellagic acid were determined in PPE-FD, PPE-N, and active films by UHPLC-DAD. New PLA-based active packaging with PPE-FD (PLA/3PPE) and pomegranate peels (PLA/3PP) were developed, and their chemical, mechanical, and barrier properties were evaluated. Moreover, the effectiveness of the new film was carried out through the study of lipid oxidation state and microbial contamination of two high-fat content foodstuffs, almonds and beef meat. PPE-FD presented high antioxidant capacity and high content in total phenolics and flavonoid compounds. The PLA/3PPE and PLA/3PP prevented the lipid oxidation of meat and presented antimicrobial activity against Staphylococcus aureus. The addition of the PPE-FD and the pomegranate peels to the PLA did not affect the morphology of the polymer. Although both PPE-FD and pomegranate peels presented punicalagin (A + B) and ellagic acid, only ellagic acid was identified and quantified in the active PLA. Active PLA films were not effective in delaying the lipid oxidation of almonds, but they showed to be significantly effective in delaying the lipid oxidation of beef meat and reducing the microbial growth in this food matrix over time. publishersversion published

Due to their high content of bioactive compounds, several extraction techniques can be applied to obtain extracts rich in valuable compounds and further application into food applications. Distillation and solvent extraction continues to be the most used and applied extraction techniques, followed by newer techniques such as microwave-assisted extraction and pulsed electric field extraction. Although the composition of these extracts and essential oils directly depends on the edaphoclimatic conditions to which the fruit/plant was exposed, the main active compounds are D-limonene, carotenoids, and carbohydrates. Pectin, one of the most abundant carbohydrates present in Citrus peels, can be used as a biodegradable polymer to develop new food packaging, and the extracted bioactive compounds can be easily added directly or indirectly to foods to increase their shelf-life. One of the applications is their incorporation in active food packaging for microbiological and/or oxidation inhibition, prolonging foods’ shelf-life and, consequently, contributing to reducing food spoilage. Cynara cardunculus L. (Asteraceae) is a multipurpose crop native to the Mediterranean area, commonly called cardoon. It comprises three botanical varieties: the globe artichoke (var. scolymus (L.) Fiori), the cultivated cardoon (var. altilis DC.), and the wild cardoon (var. sylvestris (Lamk) Fiori). Cardoon can develop in adverse climate conditions, low inputs, high temperature, severe drought, and infertile soils as a resistant species. Yet, it can persist for several years, re-sprouting annually. Cardoon can be used for different purposes due to its composition. For instance, the fleshy stems and the immature heads are used in Mediterranean cuisine. The flowers are used in the production of some cheeses as vegetal rennet. In traditional medicine cardoon is used due to its high content in cynarin and silymarin. Kidneys and liver malfunction, high blood cholesterol levels, diabetes, and cancer are some of the medicinal applications. On the other hand, cardoon’s by-products, leaves, stems and seeds, can be used to produce biomass for energy, biodiesel, seed oil, animal feed and paper pulp. Cardoon’s leaves bioactive properties (e.g.: antioxidant and antimicrobial activities) are due to its excellent polyphenol profile, rich in chlorogenic acid and flavonoids. As leaves represent, on average, about 60% of total cardoon waste, in the context of a circular economy it is essential to take advantage of its composition in bioactive compounds. Concerning the food industry, leaves, and leaves extracts bioactivity may represent an added value as a food additive, as an ingredient in developing a novel food, or in active food packaging to control lipid oxidation and microbial deterioration. Hence, this study aims to address the current food applications of cardoon’s leaves and their functional properties and explore new potential ones adding value to its properties. More and more, consumers are demanding natural products with longer shelf-life. Extracts from rosemary (Rosmarinus officinalis L.) and green tea (Camellia sinensis L.) have high antimicrobial and antioxidant activities. These extracts can be incorporated into polymers produced from renewable sources, to produce active biopackaging. This concept intends to positively interact with packaged food, to inhibit the natural degradation of food, maintaining the quality of the packaged food and increasing its shelf-life. The antioxidant activity of food grade extracts from rosemary and green tea was evaluated and the total content in phenolic compounds and flavonoids was determined. Atualmente, a indústria das embalagens alimentares, devido a várias solicitações resultantes da consciencialização dos consumidores sobre questões ambientais, procura soluções "verdes", mais amigas do ambiente no contexto de uma economia sustentável. Infelizmente, o uso de biopolímeros apresenta limitações, uma vez que apresentam propriedades mecânicas e barreira limitativas. Este fato pode ser mitigado adicionando componentes de tamanho nano para formar nanocompósitos. Além destes novos desenvolvimentos, a incorporação de substâncias de extratos naturais nos nanocompósitos pode substituir os aditivos sintéticos utilizados na indústria das embalagens. A dissertação de doutoramento apresentada divide-se em duas partes distintas, em que a primeira aborda a Inovação das Embalagens Alimentares e a segunda a Segurança das mesmas. Este trabalho resulta da compilação de uma série de estudos que dão resposta aos objetivos traçados. Na primeira parte do trabalho, procedeu-se a uma extensa revisão bibliográfica acerca de nanoreforços, nomeadamente acerca da Nanocelulose (CNC) e da argila Montmorillonita (MMT), no âmbito das suas aplicações em embalagens alimentares. Os restantes artigos, traduzem o desenvolvimento e o estudo de filmes bionanocompósitos, suas propriedades estruturais, mecânicas e de barreira e na sua atividade antioxidante. No decorrer destes estudos foram produzidos diferentes filmes com diferentes composições, quer no que se refere à percentagem de nanoreforço utilizado, quer no que se refere à quantidade de extrato de chá verde incorporado. Os diferentes filmes formulados resultaram num filme biopolimérico ativo de ácido polilático/extracto de chá verde (PLA/GTE) e em filmes bionanocompósitos ativos de PLA/CNC, PLA/MMT e PLA/CNC/GTE. É ainda de destacar, que nesta parte do trabalho foi elaborado um filme biodegradável ativo à base de proteína de soro de leite concentrada (WPC) incorporado com extrato de chá verde. Os diferentes filmes produzidos foram caracterizados recorrendo a uma série de técnicas/métodos laboratoriais. Face aos resultados obtidos, no caso dos compósitos PLA/CNC, as imagens de microscopia eletrónica de varrimento (SEM) indicam que as nanopartículas são dispersas homogeneamente na matriz PLA sendo que, a adição de CNC aumenta ligeiramente a percentagem cristalina da matriz polimérica, apesar da sua sinergia com o GTE desencadear uma estrutura aberta que restringe o processo de cristalização. Nos filmes PLA/CNC/GTE, observa-se que com uma maior percentagem de CNC (ou seja, 5%) ocorre a formação de aglomerados de nanocristais, dificultando o nível de dispersão. Todos os nanocompósitos de CNC e CNC/GTE baseados em PLA, quando comparados com o filme de PLA, indicam um efeito positivo nas propriedades de barreira, sendo esse efeito mais evidente para o filme de PLA/2%CNC/1%GTE. O filme de PLA/2%CNC apresenta os melhores resultados de eficácia no que se refere à inibição da oxidação lipídica do alimento modelo testado, salame fatiado. O filme ativo de proteína de soro de leite revelou-se eficaz no atraso da oxidação lipídica até ao 14º dia de armazenamento de amostras de salmão fresco embalado. A segunda parte desta dissertação, centra-se no domínio da Segurança das Embalagens alimentares. É um fato que existe migração de componentes plásticos da embalagem para o alimento, assim sendo, se a migração destes componentes não for controlada adequadamente e se os seus níveis excederem os valores toxicológicos ou legislativos, eles podem produzir efeitos nocivos para a Saúde e afetar as propriedades organoléticas do alimento. O bisfenol A (BPA) é um composto orgânico, com atividade estrogénica, usado para fabricar plásticos de policarbonato e revestimentos de resinas epóxi. Neste trabalho, foi desenvolvido e validado, com sucesso, um método analítico baseado em cromatografia líquida de alta eficiência com deteção de fluorescência (HPLC-FLD) para a determinação de BPA em vegetais enlatados. Estes resultados foram confirmados por cromatografia líquida acoplada a espectrometria de massa (LC-MS/MS). Currently, the food packaging industry, due to several requests resulting from consumer awareness of environmental issues, is looking for "green" solutions, more environmentally friendly in the context of a sustainable economy. Unfortunately, the use of biopolymers has limitations, since they have to limit mechanical and barrier properties. This fact can be mitigated by adding nano-sized components to form nanocomposites. In addition to these new developments, the incorporation of substances from natural extracts into nanocomposites may replace the synthetic additives used in the packaging industry. The doctoral dissertation presented is divided into two distinct parts, in which the first address the Innovation of Food Packaging and the second the Safety thereof. This work results from the compilation of a series of studies that respond to the objectives set. In the first part of the work, an extensive bibliographic review was carried out on nanoreinforces, namely on Nanocellulose (CNC) and Montmorillonite clay (MMT), within the scope of their applications in food packaging. The remaining articles reflect the development and study of bionanocomposite films, their structural, mechanical and barrier properties, and their antioxidant activity. In the course of these studies, different films were produced with different compositions, both in terms of the percentage of nanofiller used and in terms of the amount of green tea extract incorporated. The different formulated films resulted in an active biopolymeric film of polylactic acid / green tea extract (PLA/GTE) and active bionanocomposite films of PLA/CNC, PLA/MMT, and PLA/CNC/GTE. It should also be noted that in this part of the work, an active biodegradable film based on whey protein concentrate (WPC) incorporated with green tea extract was elaborated. The different films produced were characterized using a serie of laboratory techniques/methods. Given the results obtained, in the case of PLA/CNC composites, the scanning electron microscopy (SEM) images indicate that the nanoparticles are dispersed homogeneously in the PLA matrix, with the addition of CNC slightly increasing the crystalline percentage of the polymer matrix, despite synergy with the GTE trigger an open structure that restricts the crystallization process. In the PLA/CNC/GTE films, it is observed that with a higher percentage of CNC (that is, 5%), the formation of clusters of nanocrystals occurs, hindering the level of dispersion. All PLA-based CNC and CNC/GTE nanocomposites, when compared to PLA film, indicate a positive effect on barrier properties, this effect being more evident for PLA/2%CNC/1%GTE film. The PLA/2%CNC film shows the best results in terms of inhibiting the lipid oxidation of the tested model food, sliced salami. The active film of whey protein proved to be effective in delaying lipid oxidation until the 14th day of the storage of fresh packaged salmon samples. The second part of this dissertation focuses on the safety of food packaging. It is a fact that there is a migration of plastic components from the packaging to the food, so if the migration of these components is not adequately controlled and if their levels exceed toxicological or legislative values, they can produce harmful effects on health and affect health. organoleptic properties of the food. Bisphenol A (BPA) is an organic compound, with estrogenic activity, used to manufacture polycarbonate plastics and epoxy resin coatings. In this work, an analytical method based on high-performance liquid chromatography with fluorescence detection (HPLC-FLD) for the determination of BPA in canned vegetables was successfully developed and validated. UIDB/04077/2020 UIDB/00211/2020. Cardoon, Cynara cardunculus L., is a perennial plant whose flowers are used as vegetal rennet in cheese making. Cardoon is native from the Mediterranean area and is commonly used in the preparation of salads and soup dishes. Nowadays, cardoon is also being exploited for the production of energy, generating large amount of wastes, mainly leaves. These wastes are rich in bioactive compounds with important health benefits. The aim of this review is to highlight the main properties of cardoon leaves according to the current research and to explore its potential uses in different sectors, namely the food industry. Cardoon leaves are recognized to have potential health benefits. In fact, some studies indicated that cardoon leaves could have diuretic, hepato-protective, choleretic, hypocholesterolemic, anti-carcinogenic, and antibacterial properties. Most of these properties are due to excellent polyphenol profiles, with interesting antioxidant and antimicrobial activities. These findings indicate that cardoon leaves can have new potential uses in different sectors, such as cosmetics and the food industry; in particular, they can be used for the preparation of extracts to incorporate into active food packaging. Introdução: De acordo com o estudo Global Burden of Disease (GBD) realizado em 2017, os hábitos alimentares inadequados dos portugueses foram o terceiro fator de risco que mais contribuiu para a perda de anos de vida saudável. Na última década, diversos estudos epidemiológicos têm evidenciado uma relação entre a saúde e o consumo de certos tipos de gordura, nomeadamente pela ingestão de alguns ácidos gordos específicos. Objetivo: Caracterizar o perfil em ácidos gordos de três grupos de alimentos representativos dos hábitos/consumos alimentares da população portuguesa. Metodologia: Os grupos de alimentos estudados foram: Grupo do Peixe, produtos da pesca, anfíbios, répteis e invertebrados; Grupo dos Cereais e produtos à base de cereais e Grupo dos Pratos compostos. A recolha de amostras e sua preparação foram efetuadas no âmbito do Projeto TDS piloto (estudos de dieta total (TDS)) enquadrado no Projeto Europeu TDSExposure. Para a determinação do perfil de ácidos gordos, realizou-se a extração e metilação dos ácidos gordos em ésteres metílicos, seguida de análise por cromatografia gasosa acoplada a detetor de ionização de chama. Resultados: De acordo com os resultados obtidos, os valores de gordura total variaram entre 0,8 g/100g (Massa cozida) e 23,4 g/100g (Bolachas de chocolate). O valor mais elevado de ácidos gordos saturados (AGS) por 100g de alimento, foi observado no grupo dos Cereais e produtos à base de cereais (Bolachas de Chocolate (12,5 g/100 g)), enquanto que, o valor mais baixo de ácidos gordos saturados quantificado foi encontrado na amostra de Salmão (3,1 g /100 g). Os valores de ácidos gordos monoinsaturados (AGMI) variaram entre 0,1 g/100g (Caldo verde) e 9,9 g/100 g (Queque). O teor mais elevado de ácidos gordos polinsaturados (AGPI) foi encontrado no grupo do Pescado. Todos os alimentos analisados apresentaram valores de ácidos gordos trans (AGT) inferiores a 0,05 g/100 g (limite de quantificação do método), com exceção das amostras de Croissant e de Almôndegas, que apresentaram o valor 0,3 g/100 g. Conclusões: De acordo com os resultados obtidos, os teores mais elevados de ácidos gordos saturados (AGS), monoinsaturados (AGMI) e polinsaturados (AGPI) foram encontrados nos alimentos do grupo dos Cereais e produtos à base de cereais, tendo sido também os que apresentaram maiores teores de gordura total. O valor mais baixo de AGS foi encontrado nos alimentos do grupo do Pescado. N/A

As embalagens alimentares têm como principal função a proteção dos alimentos de fatores que alterem o seu valor nutricional, as suas propriedades organoléticas e coloquem em risco a sua segurança. Contudo, os materiais utilizados nas embalagens podem interagir com os alimentos, podendo ocorrer a migração de determinados compostos para os alimentos. A maioria das embalagens metálicas usadas nas conservas de alimentos são revestidas internamente por um material epóxi-fenólico, onde se inclui o éter diglicidílico do bisfenol A (BADGE). Nas últimas décadas, tem havido um grande desenvolvimento das tecnologias usadas na conservação dos alimentos, com consequente aumento do tempo de vida útil destes e da segurança alimentar. Neste sentido, surgiram as embalagens ativas e as embalagens inteligentes. Este estudo teve como objetivo avaliar os hábitos de consumo de latas de conserva numa amostra da população portuguesa e aferir o seu conhecimento sobre embalagens ativas e inteligentes. Perante os resultados obtidos, a população portuguesa consome um elevado número de produtos em latas de conserva, em especial atum (género Thunnus), e poucos inquiridos (26,4%) possuem conhecimento sobre a potencial migração de bisfenóis para os alimentos. Em relação às embalagens alimentares inovadoras, apenas uma pequena fração dos inquiridos (11%) tem conhecimento da sua existência e das funções que desempenham. The main purpose of food packaging is to protect food from factors that alter its nutritional value, its organoleptic proper ties and put its safety at risk. However, the materials used in the packaging may interact with food, and migration of cer tain compounds to food may occur. Most metal containers used in food preser ves are internally coated with an epoxyphenolic material, which includes bisphenol A diglycidyl ether (BADGE). In the last decades, there has been a great development of the technologies used in food preser vation, with a consequent increase in shelf life and food safety. In this sense, active packaging and smar t packaging have emerged. The purpose of this study was to evaluate the consumption habits of canned tins, in a sample of the Por tuguese population, and to gauge their knowledge about active and intelligent packaging. Given the results obtained, the Por tuguese population consumes a large number of canned products, especially tuna fish (Thunnus genus), and few respondents (26.4%) are aware of the potential migration of bisphenols into food. O bisfenol A (BPA) é um composto orgânico, de origem industrial, com atividade estrogénica. A sua principal utilização encontra-se na produção de plásticos policarbonatos e resinas epóxi, aplicados a embalagens alimentares e produtos médicos. Devido aos perigos associados à ingestão repetida e a longo prazo deste xenoestrogénio, é importante avaliar a perceção e o conhecimento da população portuguesa acerca do BPA e dos seus efeitos para a saúde. Deste modo, foi elaborado um questionário sobre este composto e aplicado a uma amostra da população portuguesa. A amostra foi constituída por 249 indivíduos de diversas zonas do país, pertencentes a diversas faixas etárias e com vários níveis de escolaridade. O questionário compilou questões que permitem reunir dados socioeconómicos e obter informação acerca do conhecimento dos inquiridos sobre este composto químico. Os resultados obtidos mostram que 64% dos inquiridos nunca ouviram falar do BPA. Apesar de 92% dos inquiridos (dos que conheciam o BPA) terem conhecimento que a exposição a este químico produz efeitos negativos para a sua saúde, e que estes, estão maioritariamente associados à utilização de materiais plásticos em contacto com alimentos, 50% mostram desconhecimento de quais os tipos de riscos a que estão sujeitos. Concluiu-se que nesta amostra da população portuguesa o conhecimento sobre o bisfenol A (BPA) é escasso, e que a maioria dos inquiridos não conhece o composto. Bisphenol A (BPA) is an organic compound, of industrial origin, with estrogenic activity. Its main use is in the production of plastic polycarbonates and epoxy resins, applied to food packaging and medical products. Due to the dangers associated with repeated and long-term ingestion of this xenoestrogen, it is impor tant to assess the perception and knowledge of the Por tuguese population about BPA and its health ef fects. Thus, a questionnaire was elaborated on this compound and applied to a sample of the Por tuguese population. The sample was made up of 249 individuals from dif ferent par ts of the countr y, belonging to dif ferent age groups and with dif ferent education levels. The questionnaire compiled questions that allow us to gather socioeconomic data and obtain information about the knowledge of the par ticipants about this chemical. The results show that 64% of the par ticipants never heard of BPA. Although 92% of the respondents (from those who knew BPA) knew that exposure to this chemical agent had negative ef fects on their health and that these were mostly associated with the use of plastic materials in contact with food, 50% showed lack of knowledge of the types of risks to which they are subjected. It was concluded that among this sample of the Por tuguese population the knowledge about BPA is scarce, and the majority do not know the compound. As embalagens alimentares têm como principal função a proteção dos alimentos de fatores que alterem as suas propriedades organoléticas, valor nutricional e segurança microbiológica. Contudo, os materiais utilizados no revestimento das embalagens podem interagir com os alimentos, havendo a migração de determinados compostos para os alimentos. A maioria das latas metálicas usadas para a conserva de alimentos é revestida internamente por um material epóxi-fenólico que tem como base o bisfenol A (BPA) e epicloridrina. Estes dois compostos reagem e produzem uma resina de baixo peso molecular, onde se inclui o éter diglicidílico do bisfenol A (BADGE). Com o avanço da tecnologia, surgiram as embalagens ativas e as embalagens inteligentes. As embalagens ativas podem ser de dois tipos: de emissão ou absorção. Estas têm como função a interação entre a embalagem e o alimento, através da emissão de compostos ou da absorção de gases ou substâncias provenientes do alimento. Em relação às embalagens inteligentes, a sua função principal é monitorizar as condições do alimento e do meio envolvente do alimento e informar o consumidor das mesmas. Este estudo teve como objetivo o perceber os hábitos de consumo de latas de conserva numa amostra da população portuguesa e aferir o seu conhecimento sobre embalagens ativas e inteligentes. Foi elaborado um questionário online com 26 questões. Dos inquiridos, a maioria era do sexo feminino (66 %), entre os 18 e os 24 anos de idade (62 %), e com formação superior (licenciatura, mestrado ou doutoramento) (76 %). De acordo com os resultados obtidos, verificou-se que 70 % dos inquiridos consome alimentos enlatados, dos quais aproximadamente 72 % tem preferência por atum. Em relação aos compostos químicos presentes no revestimento das embalagens de conserva, aproximadamente 74 % respondeu não ter conhecimento sobre o bisfenol A e BADGE. No tema das embalagens alimentares, apenas 15 % dos inquiridos tinha conhecimento da função principal das embalagens inteligentes, enquanto 13 % declarou conhecer o objetivo das embalagens ativas. Com os resultados obtidos, pode afirmar-se que a população portuguesa consome um elevado número de produtos em latas de conserva, nomeadamente o atum, apesar de poucos inquiridos possuírem conhecimento sobre a migração de bisfenóis para os alimentos. The goal of this work was to produce a biodegradable active film based on polylactic acid (PLA) in which green tea extract (GTE) has been immobilized by extrusion, Two GTE concentrations were tested, 1% (w/w) and 2% (w/w). Four different GTEs were characterized regarding its antioxidant capacity, total phenolics content (TPC) and total flavonoids content (TFC). The commercial GTE chosen presented with highest antioxidant capacity by the DPPH radical assay (EC50= 0.12 ± 0.00 mg/mL) and highest TPC (416 ± 9.95 mg gallic acid equivalents (GAE)/g extract) being selected to be incorporated in the active film. The new film was to evaluated regarding its mechanical properties (e.g. tensile strength decreased 12% with the incorporation of GTE in the PLA matrix and strain at break increased 9.6% and 36% with the addition of 1% and 2% of GTE, respectively), water vapour transmission rate (water-vapour barrier properties improved with the addition of higher amounts of GTE) and effectiveness to inhibit lipid oxidation of model food. Smoked salmon was selected due to be a much appreciated high-fat content food with considerable high value in the market. The methods applied were: peroxide value, p-anisidine value, thiobarbituric acid reactive substances (TBARS) assay and hexanal monitoring, after packaging smoked salmon slices during different storage times (0, 7, 15, 30, 45 and 60 days). According to the results, the incorporation of GTE in the PLA films protects the smoked salmon from lipid oxidation in the different storage times tested. However, further studies should be performed to better understand the GTE mechanism of action as the results indicate a pro-oxidant effect of PLA/GTE 2% after 60 days of storage and to evaluate the potential antimicrobial activity of active films. To our information, this is the first time that an active PLA film containing GTE is evaluated regarding mechanical and barrier properties as well as its effectiveness in inhibiting lipid oxidation of smoked salmon.
