Our aim was to determine the levels of arginine decarboxylase (ADC), ornithine decarboxylase (ODC) and agmatine, ornithine, nitric oxide (NO) and arginase activity in the acute and subacute phases of acute ischemic stroke (AIS), by revealing the possible role of these levels in neuroinflammation and neurogenesis that are postulated to begin after AIS. 35 patients diagnosed with AIS and 35 controls were included in the study. ADC, ODC, agmatinase and NO levels were determined by ELISA kits. Arginase activity and ornithine levels were measured spectrophotometrically. There was no statistically significant difference between the levels of agmatinase, ornithine and arginase activity on days 1 and 7 in the AIS group and the levels measured in the control group (p > 0.05). High ADC and low ODC and NO values were found in the patients compared to the controls in both acute and subacute periods (p < 0.05). It was shown that polyamine synthesis decreased in both acute and subacute periods after AIS; and this was due to the decrease in ODC levels, the rate-regulating step of the major pathway. WOS: 000461427300003, Objective: SCUBE1 [signal peptide-CUB (complement C1r/C1 s)-EGF (epidermal growth factor)-like domain-containing protein 1] is a novel biochemical marker. SCUBE1 is thought to play roles both in platelet activation and inflammation, which are important stages for the development of acute ischemic stroke (AIS). The purpose of our study was to determine the diagnostic and prognostic values and temporal change of plasma SCUBE1 levels in AIS patients. Materials and methods: Thirty-five patients diagnosed with AIS at the Cumhuriyet University Faculty of Medicine Neurology Department, between June and December 2017, and a control group of 35 healthy volunteers were included. Results: Median first day SCUBE1 value in the patient group was 97.51 ng/mL, and the median 7th day SCUBE1 value was 32.72 ng/mL. Median control group SCUBE1 value was 27.51 ng/mL. The first day SCUBE1 levels were significantly higher than the 7th day and the control group SCUBE1 levels (p = 0.001, p < 0.001, respectively). The ROC analysis showed that SCUBE1 levels above 68.8 ng/mL can be used as an indicator with high sensitivity and specificity for AIS diagnosis. Multivariate analysis revealed that the first day SCUBE1 had significant independent effects on development of AIS. In correlation analysis, plasma SCUBE1 levels showed a significantly positive correlation with lesion volume, NIHSS and MRS values (p < 0.01). Conclusion: First day plasma SCUBE1 values in AIS patients rised at significant levels compared to the control group. SCUBE1 could use both in the early diagnosis and prediction of prognosis and lesion volume of AIS patients.

OBJECTIVES: This study aims to compare the serum vascular endothelial growth factor (VEGF) and vascular endothelial growth factor receptor-1 (VEGFR-1) levels between patients with fibromyalgia syndrome (FMS) and healthy controls. PATIENTS AND METHODS: The study included 40 female patients (mean age 39.9±10.2 years; range, 22 to 52 years) diagnosed with primary FMS according to the American College of Rheumatology criteria (1990) and 40 healthy female volunteers (mean age 40.9±8.3 years; range, 25 to 53 years). The sociodemographic data of both groups were recorded. The disease duration and the number of tender points were recorded for patients with FMS, and venous blood samples were collected from the two groups for the measurement of serum VEGF and VEGFR-1 levels. RESULTS: The FMS and control groups were comparable in terms of age and body mass index (p>0.05). Background: Multiple sclerosis (MS) is a demyelinating autoimmune disease characterized by the infiltration of T cells into the central nervous system. Polyamines, which contribute to cell proliferation, hypertrophy and tissue development, have some tissue-specific roles in brain tissue. So, we aimed in this study to present the possible effects of polyamines on MS. Method: Thirty- five patients with MS and 35 sex and age-matched control were included in this study. Arginine decarboxylase (ADC), ornithine decarboxylase (ODC) and agmatinase levels were measured by ELISA kits. Results: The patient group had higher ODC and agmatinase levels than controls. The correlation analyses between ODC, ADC, and agmatinase levels and disease duration were revealed that there was a negligible positive relationship between disease duration and agmatinase, with negligible negative relationship between disease duration and ODC and ADC. Additionally, there were showed respectively moderate and weak positive correlations between EDSS (Expanded Disability Status Scale) scores and newly developed lesions and ODC and agmatinase levels. Conclusions: Elevated polyamine synthesis in MS patients was presented by detecting increased ODC, ADC and agmatinase levels compared to controls. Besides this increased polyamine synthesis in MS patients was also related with disease duration, number of newly developed lesions and disability.

WOS: 000373106100002, This study aims to investigate the relationship between sulfurtransferase (STS) activities [rhodanese (TST), mercaptopyruvate sulfurtransferase (MST)] involved in the catalysis of several biochemical reactions including detoxification of cyanide (CN-), restructuring of Fe-S cluster in proteins, and detoxification of oxygen radicals. Rats with type 1 diabetes mellitus induced by streptozotocin (STZ) were anesthetized at 14th day, and liver, lung, kidney, and heart tissues were extracted. All samples were homogenized, and mitochondrial parts were separated. Same processes were performed also in the control group, and TST and MST activities were measured in each part. The homogenate MST (MST (Homo) .) activities of the type 1 diabetes mellitus group were compared with the control group, and a decrease was observed in the lung, liver, and kidney, respectively; at the same time, an increase was seen in the heart tissue. The mitochondrial MST (MST (Mito) .) activities of rats with type 1 diabetes mellitus group were compared with the control group, and a decrease was found in all tissues. The highest decrease in the TST (Mito) . level of rats with type 1 diabetes mellitus was observed in kidney tissue. The TST activities of the type 1 diabetes mellitus group were compared with the control group, and a decrease was observed in the liver, lung, and kidney, respectively; at the same time, an increase was seen in the heart tissue. It is demonstrated in the present study that decreases occur both in enzyme levels of tissue homogenates and in mitochondria, of rats with induced type 1 diabetes mellitus. However, these results were not statistically significant. In the presence of these findings, we think that kidney, liver, lung, and heart tissue can be affected by type 1 diabetes in the long term.

Bu calismada grafen oksitin (GO) purin katabolizmasinda rol alan ksantin oksidaz (KO) enzim aktivitesi uzerine in vitro etkisinin incelenmesi amaclanmistir. GO' nun KO aktivitesi uzerine etkisini incelemek amaciyla 0.50, 1.00, 5.00 ve 10.00 ppm olmak uzere 4 farkli derisimde GO cozeltisi KO tepkime ortamina ilave edilmis ve 5 farkli substrat (hipoksantin) derisiminde aktivite degerleri spektrofotometrik olarak belirlenmistir. K m ve V max degerleri saptanmistir. KO' nun V max degeri 0.83 µM Urat/mgProtein/dakika, K m degeri 138.95 µM olarak hesaplanmistir. Tepkime ortamina 0.50, 1.00, 5.00, 10.00 ppm GO eklendiginde KO enzimin sirasiyla %36.1, 72.3, 77.1 ve 84.3 oraninda inhibe oldugu tespit edilmistir. Sonuc olarak, GO derisimi arttikca enzimin hem K m hem de V max degerlerinde dikkate deger bir azalma oldugu ve substrat derisimi artirsak da enzim inhibisyonun devam ettigi tespit edilmistir. Bu da inhibisyon tipinin unkompetetif oldugunu gostermektedir.

WOS: 000337256000005, PubMed: 24337212, Aim: Oxidative stress, which is widely recognised as an important feature of many diseases, can be defined as an increased formation of reactive oxygen species or decreased antioxidant defense. In this study we measured plasma vitamin E levels and total antioxidant activity (AOA) in patients with slow coronary flow (SCF). Methods: The plasma vitamin E levels and AOA were measured in 40 patients with angiographically diagnosed SCF. Forty subjects with normal coronary flow (NCF) served as the control group. SCF and NCF were analysed, and blood samples were taken for plasma vitamin E levels and AOA. Plasma vitamin E levels and AOA in patients with SCF were evaluated and compared to those of patients with NCF. Results: There was no significant difference between the two groups in terms of plasma AOA, lipid profile and C-reactive protein (CRP) levels but there was a significant difference in vitamin E levels between the two groups (p = 0.001). Conclusion: Vitamin E levels were found to be lowered in patients with SCF compared to the NCF group. The association between smoking and vitamin E levels is worth further investigating in larger samples.

The most accepted view to explaining the pathogenesis of Crimean–Congo hemorrhagic fever (CCHF) is endothelial damage. This study was conducted in a University hospital to investigate the serum levels and prognostic significance of the vascular endothelial growth factor (VEGF) and its receptor, soluble fms-like tyrosine kinase-1 receptor (sVEGFR-1) in CCHF. Forty-eight consecutive hospitalized CCHF patients (grouped into severe illness and non-severe illness) and 40 healthy adults, as controls were enrolled. There was statistically significant difference for each of VEGF (P = 0.003), and sVEGFR1 (P = 0.0001) between the patients and controls. VEGF and sVEGFR1 levels in patients with severe CCHF were found to be higher than in the control group (P = 0.0001 and P = 0.0001, respectively). A significant difference was found in VEGF (P = 0.003) and sVEGFR1 (P = 0.0001) levels when compared to patients with CCHF who died and who recovered. In patients in the group with severe illness, the sensitivity, specificity, and the area underneath the ROC curve (AUROC) belonging to those cut-off points of VEGF and sVEGFR1 were 66.7%, 76.2%, 0.747, and 77.8%, 81%, 0.849, respectively. In non-survivors, the sensitivity, specificity, and the AUROC belonging to those cut-off points of VEGF and sVEGFR1 defined as 77.8%, 76.9%, 0.813, and 88.9%, 97.4%, 0.912, respectively. In conclusion, high sensitivity, specificity, and the AUROC values were found in sVEGFR1 levels especially in the severely ill and non-survivors. Therefore, sVEGFR1 may be an important biomarker for determining the risk of severity and death as result of infection with CCHF virus. J Med. Oxidative stress is considered to be the main factor in the development of diabetic complications and tissue injury. our objective was to investigate and compare the oxidant/antioxidant conditions and detoxification mechanisms of the liver, lung, kidney, cardiac tissues, and mitochondria of rats with diabetes induced by streptozocin (STZ). Methods. Rats with diabetes induced by streptozocin were anesthetized by administering 90 mg/kg ketamine hydrochloride and 3mg/kg xylazine hydrochloride. Thoracic cavities were incised open; liver, lung, kidney, and cardiac tissues were removed and stored at -70 degrees C. All samples were homogenized and mitochondrial fractions were separated. Total Antioxidant Status (TAS), Total Oxidant Status (TOS), Oxidative Stress Index (OSI), Paraoxonase (PON), Arylesterase, Catalase (Cat), Malondialdehyde (MDA), and Glutathion-S-transferase were measured in each fraction. Results. MDA and TOS levels were significantly increased in liver tissues, and TOS and OSI were increased in the mitochondrial fractions of diabetic rats. These increases were not statistically significant compared to the control group. No significant differences were determined in the antioxidant and GST activities. Conclusion. According to our results, oxidative stress has not developed in rats with diabetes induced by streptozocin. The detoxification system was induced; however, this induction did not differ significantly from the controls.

Background: Crimean-Congo Hemorrhagic Fever (CCHF) is a disease characterized by serious course, including acute viral fever, ecchymosis, thrombocytopenia, liver dysfunction and high rate of mortality. Hypoxia Inducible Factor-1α (HIF-1α) and Vascular Endothelial Growth Factor-A (VEGF-A) play an important role both in the inflamma­tory process and plasma leakage. The aim of this study was to define HIF-1α and VEGF-A serum levels obtained from CCHF patients and control group and to investigate whether these factors were correlated with the pathogenesis of this disease. Methods: Thirty cases younger than 17 yr confirmed by RT-PCR and/or ELISA for CCHF were included in this study. Thirty age and sex matched healthy peoples were enrolled as controls. Blood samples collected from the pa­tient and control groups. Serum levels of HIF-1α and VEGF-A were measured with ELISA. Results: Levels of HIF-1α and VEGF-A were statistically significantly increased in CCHF patients compared to the control group (P< 0.05). A significant positive correlation was found between the levels of HIF-1α and VEGF-A in the patient group (P< 0.01). The levels of ALT, AST, CK, aPTT, WBC and Thrombocyte count were significantly higher in the patients than in the control group (P< 0.001). A positive correlation was found among the levels of AST and CK from biochemical parame­ters and VEGF and HIF-1α in the patient group (P< 0.05) Conclusion: HIF-1α and VEGF-A might play an important role in CCHF pathogenesis.
