Data useful for a characterization of the thermodynamic behaviour of a few nonionic aminoacid anhydrides (diketopiperazines) in water are reported. In particular, results of heat of dilution, heat of solution and osmotic coefficient measurements (the latter two only for sarcosine anhydride) are illustrated. As an interesting extension of our study, the likelihood that the compounds considered might exhibit a denaturant action on globular proteins has been tested using ribonuclease. Our results clearly point out that the diketopiperazines are strong denaturants indeed. A few results of spectral measurements of the monomer-dimer equilibrium of acridine orange in aqueous diketopiperazines solutions are also reported, as they appear to pertain to the mechanism of ribonuclease unfolding promoted by the diketopiperazines.
