Introduction: Considerable use of neonicotinoid insecticides has resulted in their detection in surface waters globally, with imidacloprid (IM) and thiamethoxam (TM) frequently found together. Neonicotinoids are selective agonists for invertebrate nicotinic acetylcholine receptors (nAChR) leading to paralysis and death. While not overtly toxic to vertebrates, growing evidence suggests that chronic exposure to individual neonicotinoids can cause adverse health effects in fish. This work examined whether chronic exposure to binary mixtures of imidacloprid (IM) and thiamethoxam (TM) would be more toxic to fathead minnow ( Pimephales promelas ) larvae than either insecticide alone. Materials and Methods: Embryos were exposed to a 1:1 mixture of IM and TM (0.2, 2, 20, 200 or 2,000 μg/L of each pesticide) or a 1:5, 1:10, or 1:20 mixture of IM and TM (0.02 μg/L of IM with 0.1, 0.2, or 0.4 μg/L of TM) for a total of 8 days. Survival, developmental toxicity, embryonic motor activity, and startle escape responses were quantified. Results: Survival and growth were reduced, and hatching induced by exposure to a 1:1 mixture containing > 2 μg/L of each insecticide, but not following exposure to mixtures containing environmentally-relevant concentrations. Acute exposure to a 1:1 mixture did not alter embryonic motor activity; however, chronic exposure to these mixtures resulted in a slight but significant decrease in embryonic movements. Only 1:1 mixtures of high concentrations of IM and TM altered the startle escape response by increasing latency of response; however, a significant proportion of fish exposed to 1:1 mixtures had altered latency and burst speed. Taken together, these behavioral indicators of nAChR activation suggest that in mixtures, neonicotinoids could interfere with nAChR signaling despite their low affinity for the nAChR. Conclusion: Our findings suggest that toxicity of binary mixtures of IM and TM is primarily driven by IM, and that mixtures of IM with TM do not appear to cause significant additive toxicity when compared with our previous studies evaluating each neonicotinoid alone. Imidacloprid (IM) has emerged as a contaminant of concern in several areas within the United States due to its frequent detection in aquatic ecosystems and its pseudo-persistence, which pose potential risks to nontarget species. We evaluated the sublethal toxicity of IM to fathead minnow larvae following chronic exposure beginning just after fertilization. Our in silico analysis and in vivo bioassays suggest that IM has a low binding affinity for the vertebrate nicotinate acetylcholine receptor (nAChR), as expected. However, chronic exposure to ≥0.16 µg IM/L reduced survival by 10%, and exposure to ≥18 µg IM/L reduced survival by approximately 20%-40%. Surviving fish exposed to ≥0.16 µg IM/L showed reduced growth, altered embryonic motor activity, and premature hatching. Furthermore, a significant proportion of fish exposed to ≥0.16 µg IM/L were slower to respond to vibrational stimuli and slower to swim away, indicating that chronic exposure to IM has the potential to impair the ability of larvae to escape predation. The adverse health effects we observed indicate that chronic exposure to environmentally relevant concentrations of IM may elicit sublethal responses that culminate in a significant increase in mortality during early life stages, ultimately translating to reduced recruitment in wild fish populations. In general, the photonic crystal (PC) is a periodical optical structure, but there are some studies considering aperiodic structures. If we insert a defect layer into a one-dimensional periodic PC to break its translational symmetry order (TSO), some peaks, called defect modes, appear in the transmittance spectrum. The defect layer thickness governs the frequencies of these defect modes but almost does not affect the other part of the spectrum. The discovery of quasi-crystals tells us that not only the TSO but also other orders can produce Bragg diffraction. It is well known that triadic Cantor set (TCS) PCs, which lack TSO but have a self-similar symmetry order (SSO), still exhibit narrow transmission peaks. In this work, we try to break the SSO in TCS PCs and find the resulting optical phenomena, where single-negative materials and dielectrics are chosen as the constituents of PCs. The study method is the transfer matrix method, and the calculation results show that the background intensity of the transmittance spectrum rather than the frequency of peaks obviously periodically changes with the break of SSO. It follows that the SSO does have physical meaning, and not only the transmission peaks but also the background should be treated as a significant optical property.

Down syndrome is the most common chromosomal disorder in the United States, occurring in about 14.14/10,000 births. It is associated with multiple medical anomalies, including cardiac, gastrointestinal, musculoskeletal, and genitourinary abnormalities, which increases the burden of morbidity for this patient population. Management is typically directed toward optimizing health and function throughout childhood and into adulthood; however, consensus regarding their management in adulthood is controversial. The burden of congenital cardiac diseases in children with trisomy 21 is well established, seen in more than 40% of cases. Although screening echocardiography is performed routinely within 1 month of birth, current consensus advocates for diagnostic echocardiography only in symptomatic adults with Down syndrome. Here, we advocate that screening echocardiography should be performed routinely in this patient population at all ages, particularly in late adolescence and early adulthood, because of a high percentage of residual cardiac defects and an increased risk of developing valvular and structural cardiac disease.

Thiamethoxam (TM) is a neonicotinoid insecticide that acts as a nicotinic acetylcholine receptor (nAChR) agonist. While designed to specifically target invertebrate nAChRs, recent studies have reported adverse effects of neonicotinoid exposure in early life-stage fish. This study examined the health and neurobehavioral impacts of chronic exposure to various concentrations of TM or nicotine (NIC) in early life zebrafish ( Danio rerio ) in conjunction with in-silico molecular docking to compare their ligand-receptor interactions with vertebrate nAChR. Chronic exposure to both reduced survival by approximately 20% (163 µg TM/l) and 25-100% (≥0.49 µg NIC/l). Hatching and growth were impaired following exposure to ≥0.21 µg TM/l or 4.9 µg NIC/l. Both TM and NIC produced morphological and behavioral indicators of neurotoxicity, with more potent effects following NIC exposure. NIC impaired embryonic motor activity by 40% (49 µg NIC/l), while both TM and NIC significantly altered predator escape response in larvae, specifically the latency and the initial burst movement of the response were impacted. Molecular docking predicted variations in the type and strength of interactions that occur between NIC or TM and vertebrate nAChR. These findings demonstrate that chronic exposure to TM might impact general health and neurobehavior of early-stage zebrafish. Our data support hypotheses that TM presents low affinity for vertebrate nAChR but may still pose an adverse risk to larval fish growth and neurobehavior.

Introduction: The IMMray PanCan-d test combines an 8-plex biomarker signature with CA19-9 in a proprietary algorithm to detect pancreatic ductal adenocarcinoma (PDAC) in serum samples. This study aimed to validate the clinical performance of the IMMray PanCan-d test and to better understand test performance in Lewis-null (le/le) individuals who cannot express CA19-9., Methods: Serum samples from 586 individuals were analyzed with the IMMray PanCan-d biomarker signature and CA19-9 assay, including 167 PDAC samples, 203 individuals at high risk of familial/hereditary PDAC, and 216 healthy controls. Samples were collected at 11 sites in the United States and Europe. The study was performed by Immunovia, Inc (Marlborough, MA), and sample identity was blinded throughout the study. Test results were automatically generated using validated custom software with a locked algorithm and predefined decision value cutoffs for sample classification., Results: The IMMray PanCan-d test distinguished PDAC stages I and II (n = 56) vs high-risk individuals with 98% specificity and 85% sensitivity and distinguished PDAC stages I-IV vs high-risk individuals with 98% specificity and 87% sensitivity. We identified samples with a CA19-9 value of 2.5 U/mL or less as probable Lewis-null (le/le) individuals. Excluding these 55 samples from the analysis increased the IMMray PanCan-d test sensitivity to 92% for PDAC stages I-IV (n = 157) vs controls (n = 379) while maintaining specificity at 99%; test sensitivity for PDAC stages I and II increased from 85% to 89%., Discussion: These results demonstrate the IMMray PanCan-d blood test can detect PDAC with high specificity (99%) and sensitivity (92%)., (Copyright © 2022 The Author(s). Chronic lymphocytic leukemia (CLL) is the most commonly encountered leukemia in the clinical laboratory. Cytoskeletal defects in CLL lymphocytes can result in the formation of up to 75% smudge cells (SCs) during blood film preparation. Failure to account for these damaged lymphocytes in the white blood cell (WBC) differential diminishes the accuracy and reproducibility of the results. Lacking clear practice standards on handling SCs in CLL, different laboratories may employ different methods to mitigate SC-induced errors. This review explores the pathophysiology of SCs, their effect on WBC differentials in CLL, and how these results can impact clinical decisions. The pros and cons of various SC corrective methods are described to assist laboratories in developing an optimized protocol to reduce errors and inconsistencies in WBC differentials. Finally, the potential utility of SC enumeration as an indicator of CLL prognosis is discussed in terms of laboratories with differing access to technology., (© American Society for Clinical Pathology, 2021. The idea of artificial intelligence for social good (henceforth AI4SG) is gaining traction within information societies in general and the AI community in particular. It has the potential to tackle social problems through the development of AI-based solutions. Yet, to date, there is only limited understanding of what makes AI socially good in theory, what counts as AI4SG in practice, and how to reproduce its initial successes in terms of policies. This article addresses this gap by identifying seven ethical factors that are essential for future AI4SG initiatives. The analysis is supported by 27 case examples of AI4SG projects. Some of these factors are almost entirely novel to AI, while the significance of other factors is heightened by the use of AI. From each of these factors, corresponding best practices are formulated which, subject to context and balance, may serve as preliminary guidelines to ensure that well-designed AI is more likely to serve the social good.

Artificial intelligence (AI) research and regulation seek to balance the benefits of innovation against any potential harms and disruption. However, one unintended consequence of the recent surge in AI research is the potential re-orientation of AI technologies to facilitate criminal acts, term in this article AI-Crime (AIC). AIC is theoretically feasible thanks to published experiments in automating fraud targeted at social media users, as well as demonstrations of AI-driven manipulation of simulated markets. However, because AIC is still a relatively young and inherently interdisciplinary area-spanning socio-legal studies to formal science-there is little certainty of what an AIC future might look like. This article offers the first systematic, interdisciplinary literature analysis of the foreseeable threats of AIC, providing ethicists, policy-makers, and law enforcement organisations with a synthesis of the current problems, and a possible solution space.

We theoretically study the one-way absorption in two 1D defective asymmetric photonic crystals, air/(DB) N A(BD) M /air and air/(DB) N A(BD) M A(DB) N A(BD) M /air, where A and B are dielectrics, D is the semiconductor, n-InSb, and N, M are stack numbers with N≠M. It is revealed that their absorption spectra exhibit one-way properties. We also find that the number of one-way absorption peaks depends on the symmetry and number of defect layers, which are similar to the defect modes in the transmittance spectra of the usual symmetry photonic crystals. Additionally, effects of the incident angles for both TE and TM waves on the one-way feature are also presented. At a large incident angle, the TE wave is almost reflected, whereas the TM wave can have a partial absorption.

The widespread use of lead (Pb) shot in shooting activities, including at former shooting ranges, continues to pose environmental risks. The La Crosse River Marsh (located in Wisconsin, USA) is a biologically diverse urban riparian wetland with a legacy of Pb-contaminated sediment resulting from its use as a trap shooting range from 1929-1963. Within the shot fall zone, shot densities exceed 43,000 pellets/m 2 and surface sediments exceed 25,000 mg/kg in some areas. This study used the Zebrafish as a model to determine the acute toxicity of these contaminated sediments. Zebrafish were exposed to sediments containing approximately 13 to 13,450 mg/kg Pb for 5 days (8-120 hr post-fertilization). The toxic responses to sediments were non-monotonic. Only exposure to sediments containing "mid-range" concentrations of Pb (4580 mg/kg) induced mild skeletal malformations and a sluggish C-start response indicating that Pb was marginally bioavailable. Expression of δ-aminolevulinic acid dehydratase (ALA-D) also indicated the potential for uptake of Pb from sediments. Our findings suggest that Pb within the La Crosse River Marsh sediments is not readily bioavailable to Zebrafish, and while this metal poses a minimal acute toxicological risk, toxicity due to chronic exposure of low concentrations of Pb is possible. Further, our data demonstrated that induction of ALA-D gene expression in Zebrafish embryos shows promise as an alternative to ALA-D enzyme activity as a biomarker for acute Pb exposure under lab conditions.

Geometric morphometrics allows for the characterization of shape using Cartesian geometric coordinates rather than linear or volumetric measurements, which are dependent upon size and are insufficient to capture geometric shape. By using landmarks on specimens, variations in position, orientation and scale between specimens can be removed to better compare variations in shape. This method has primarily been used in the fields of evolutionary biology and taxonomy. Here we describe how geometric morphometrics can be used to delineate variations in shape caused by teratogenic compounds in zebrafish.

We theoretically study wave properties for one-dimensional defective asymmetric photonic crystals, air/(AB) M G(BA) N /air, air/(AQ) M G(QA) N /air, and air/(BQ) M G(QB) N /air, where A is a lossy epsilon-negative material, B is a lossy mu-negative material, G and Q are dielectrics with different refractive indexes, and M and N are stack numbers with M≠N. Special attention has been paid to their absorption spectra. It is found that at certain frequencies the absorption can exhibit unidirectional properties. Our calculated results show two kinds of unidirectional absorption peaks. One is a single absorption peak whose frequency depends on the thickness of defect layer G. For the other peaks, its frequency does not change when the defect layer's thickness changes. In addition, in the second kind of peaks, the peak numbers for forward and backward propagation are different, that is, there are (M-1) absorption peaks for forward propagation, while there are (N-1) absorption peaks for backward propagation. When the two kinds of unidirectional absorption peaks are merged, some new peaks appear, and both forward and backward propagation will have (M+N-1) absorption peaks.

The ferroelectric material KTaO 3 (KTO) has a very high refractive index, which is advantageous to the photonic crystal (PC) design. KTO polycrystalline crystal has a high extinction coefficient. In this work, we perform a theoretical study of the transmission properties of a PC bandpass filter made of polycrystalline KTO at terahertz (THz) frequencies. Our results show that the defect modes of usual PC narrowband filters no longer exist because of the existence of the high loss. We provide a new PC structure for the high-extinction materials and show that it has defect modes in its transmittance spectra, providing a possible bandpass filter design in the THz region.

Triclosan (TCS) is an antimicrobial agent found in personal care products that has become prevalent in surface waters. TCS readily bioaccumulates within aquatic organisms, and has been found to be toxic to fish. In larval fishes, exposure to TCS disrupts a variety of developmental processes, impairs hatching success, and causes pericardial edema. In mammals, TCS exposure disrupts excitation-contraction-coupling in cardiac cells, which is associated with reductions in cardiac output. Here, we examine the impacts of TCS on heart function to better understand potential risks that TCS may pose to wild fish. Zebrafish were exposed to 0, 0.4, 40, and 400 μg TCS/L from 8 to 120 h postfertilization via static waterborne exposure with daily renewal. We examined the incidence of pericardial edema, and the impacts on heart structure and heart function. While incidence of pericardial edema increased following exposure to ≥40 μg TCS/L and the structure of the heart was altered, cardiac output was only reduced following exposure to 400 μg TCS/L. A small but significant proportion of embryos showed increased incidence of regurgitation following exposure to ≥0.4 μg TCS/L. Our findings suggest that acute exposure to TCS has the potential to cause subtle cardiac toxicity in developing fish, and further evaluation of the risks TCS pose to wild fish and human health is needed.

Triclosan (TCS) is an ionizable synthetic antimicrobial that has been found to be a persistent environmental contaminant with potential for bioaccumulation. Standard laboratory assays have shown that TCS is toxic to aquatic organisms; however, varied environmental conditions could impact this risk. For example, we would predict that sorption to dissolved organic carbon (DOC) in natural surface waters would reduce the bioavailability and, therefore, toxicity of TCS. To better understand the potential risk that TCS poses to wild fish, we evaluated the toxicity of TCS to zebrafish in the presence of DOC. Zebrafish were exposed to TCS (0-900 μg TCS/L), DOC (0-25 mg/L), or TCS (0-900 μg TCS/L) together with either 10 or 25 mg DOC/L from 8 to 120 h postfertilization through static waterborne exposure. We compared impacts of TCS alone or in conjunction with DOC on mortality, development, and hatching success. Exposure to TCS in the presence of DOC improves survival and hatching success, and reduces the incidence of developmental toxicity. However, since the presence of DOC did not completely prevent sublethal toxicity, our data suggest that given its bioaccumulation potential, developmental toxicity of TCS under environmental conditions still warrants concern.

In this work, we propose a filter structure using a one-dimensional ferroelectric-dielectric periodic multilayer, air/[(ABA) N s C] Np (ABA) N s /air, where N s and N p are the two numbers of periods. Here, B is a dielectric material of SiO 2 , C is the same as B with a different thickness, and A is taken to be a ferroelectric material Ba 55 Sr 45 TiO 3 +30%Mg 2 SiO 4 , whose dielectric constant is very high (ϵ=439 at 10 GHz). The results show that the transmittance spectra have N s -channel groups at microwave frequencies and these groups can be classified into two types. The first type has only one channel group with N p narrower channels. The other has N s -1 groups, each of which has N p +1 broader channels. In this filter structure the group number and channel number of each group can be determined simply by changing N s and N p .

An 82-year-old Japanese nonsmoking man presented with persistent dry cough and small left apical pneumothorax. High resolution CT scan of the chest demonstrated bilateral upper lobe pleuroparenchymal thickening and architectural distortion. Serial imaging revealed mild progression and development of small bilateral pneumothoraces, and pneumomediastinum. Exposure to endocrine disruptors during critical periods of development can impact the sustainability of wild fish populations. Anti-androgenic compounds have received less attention, but are capable of modulating gonad differentiation and maturation, and impairing reproduction in fish. The fungicide vinclozolin (VZ) has been shown to impair reproduction in adult fish, but less is known about its effects following exposure earlier in development. Here we show that waterborne exposure to 400μg VZ/L during critical periods of sex differentiation (21-35 days post fertilization) permanently shifts sex ratios towards females, and alters the maturation of the gonad. Both fecundity and fertility were reduced, even when oogenesis and spermatogenesis recover and sperm motility is not altered. The novel heterometallic polyoxotitanate cage [Ti18MnO30(OEt)20(MnPhen)3] (1), obtained by solvothermal reaction of Ti(OEt)4 with Mn(AcO)3·(H2O)2 and 1,10-phenanthroline (Phen) in EtOH, has a C3 symmetric core structure containing an interstitial tetrahedral Mn(II) ion and is surrounded by three Mn(II)(Phen) fragments. The molecular structure is retained in thin film electrodes of 1 deposited by solution drop-casting onto fluorinated tin oxide (FTO). Both solid state and solution phase electrochemical measurements show dual redox couples, consistent with the two distinct Mn coordination environments in the cage structure. Sintering of 1 in air at 600 °C produces a black crystalline solid which consists of Mn-doped TiO2 (mainly in the rutile phase) together with α-Mn2O3. Such a composite semiconductor has an optical band gap of ca. 1.80 eV, similar to that of α-Mn2O3.

In this work, the photonic band gap (PBG) structure in a one-dimensional ferroelectric photonic crystal (PC) is theoretically investigated. We consider a PC, air/(AB) N /air, in which layer A is a dielectric of MgO and layer B is taken to be a ferroelectric of Ba 0.55 Sr 0.45 TiO 3 (BSTO). With an extremely high value in the dielectric constant in BSTO, the calculated photonic band structure at microwave frequencies exhibits some interesting features that are significantly different from those in a usual dielectric-dielectric PC. First, the photonic transmission band consists of multiple and nearly discrete transmission peaks. Second, the calculated bandwidth of the PBG is nearly unchanged as the angle of incidence varies in the TE wave. The bandwidth will slightly reduce for the TM mode. Thus, a wide omnidirectional PBG can be obtained. Additionally, the effect of the thickness of the ferroelectric layer on the PBG is much more pronounced compared to the dielectric layer thickness. That is, the increase of ferroelectric thickness can significantly decrease the PBG bandwidth.

Rationale: Interferon-γ release assays have significant advantages over tuberculin skin testing in many clinical situations. However, recent studies have called into question their reliability in serial testing of healthcare workers because of reportedly high rates of positivity and high conversion/reversion rates on retesting., Objectives: To define the performance characteristics of the T-SPOT.TB test, an interferon-γ release assay, during serial screening programs of healthcare workers at 19 U.S. hospitals., Methods: A total of 42,155 T-SPOT.TB test results from healthcare workers at 19 geographically diverse hospitals obtained for routine tuberculosis screening programs were analyzed to determine the rates of positivity, reversion, and conversion in serial testing data., Measurements and Main Results: In 19,630 evaluable serial pairs from 16,076 healthcare workers, the mean test positivity rate was 2.3% (range, 0.0-27.4%). The mean conversion rate was 0.8% (range, 0.0-2.5%), and the mean reversion rate was 17.6%. Positivity and conversion rates correlated with known tuberculosis risk factors including age and sex. The observed specificity of the T-SPOT.TB test was at least 98.6%., Conclusions: The high concordance and test completion rates in this study suggest that the T-SPOT.TB test is a reliable tool for healthcare worker serial screening. As expected, the observed positivity rates were lower compared with the tuberculin skin test, likely reflecting the higher specificity of this test. Furthermore, the observed rates of conversion were low and significantly correlated with the geographic incidence of tuberculosis. Our findings suggest that the T-SPOT.TB test is an accurate and reliable way to screen healthcare workers.

Previous theoretical studies of C3B have suggested that boron-doped graphite is a promising H2- and Li-storage material, with large maximum capacities. These characteristics could lead to exciting applications as a lightweight H2-storage material for automotive engines and as an anode in a new generation of batteries. However, for these applications to be realized a synthetic route to bulk C3B must be developed. Here we show the thermolysis of a single-source precursor (1,3-(BBr2)2C6H4) to produce graphitic C3B, thus allowing the characteristics of this elusive material to be tested for the first time. C3B was found to be compositionally uniform but turbostratically disordered. Contrary to theoretical expectations, the H2- and Li-storage capacities are lower than anticipated, results that can partially be explained by the disordered nature of the material. Dioxin (2,3,7,8-tetrachlorodibenzo-p-dioxin; TCDD) is an aryl hydrocarbon receptor (AHR) agonist, an endocrine disruptor, and a potent global pollutant. TCDD exposure is associated with diseases of almost every organ system, and its toxicity is highly conserved across vertebrates. While the acute developmental effects of dioxin exposure have been extensively studied, the ability of early sublethal exposure to produce toxicity in adulthood or subsequent generations is poorly understood. This type of question is difficult to study because of the time frame of the effects. With human subjects, such a study could span more than a lifetime. We have chosen zebrafish (Danio rerio) as a model because they are vertebrates with short generation times and consistent genetic backgrounds. Zebrafish have very modest housing needs, facilitating single and multigenerational studies with minimal time and expense. We have used this model to identify transgenerational effects of TCDD on skeletal development, sex ratio, and male-mediated decreases in reproductive capacity. Here we compare these findings with transgenerational effects described in laboratory rodent species. We propose that the zebrafish is a cost-effective model system for evaluating the transgenerational effects of toxic chemicals and their role in the fetal basis of adult disease., (Copyright © 2014 Elsevier Ireland Ltd. Metal-doped polyoxotitanium cages (M-POTs) of the type [TixOy(OR)zMnXm] (M = a main group, transition metal or lanthanide; X = an anion such as a halide) can be regarded as molecular fragments of metal-doped TiO2. As such M-POTs can be used as structural models for the inclusion of metal ions into the TiO2 lattice and the ways in which well-defined microstructural changes affect photo-induced hole-electron separation. They are also potential organically-soluble redox-catalysts for a range of organic transformations and have been shown to be useful single-source precursors for the deposition of metal-doped TiO2. The applications of M-POTs as molecular precursors to metal-doped TiO2 offers a high degree of atomic control in the low temperature fabrication of photocatalytic thin films, which have applications in pollution control and water splitting. This perspective highlights the structural trends in M-POTs, their electronic behaviour and their applications as single-source precursors, looking at current and future trends in the development of inorganic precursors for device applications.

A fractal multilayer structure made of two dielectric materials can exhibit photonic bandgap (PBG). In this work, with the use of this PBG, we study the transmission properties of periodic triadic Cantor set structures. The results indicate that the structure can be used to design multichannel filters with channel number equal to N-1 for a given number of periods, N. In addition, the channel frequencies can be designed at will. The considered structure provides another new type of design for a tunable multichannel filter.

To what extent the presence of transition metal ions can affect the optical properties of structurally well-defined, metal-doped polyoxotitanium (POT) cages is a key question in respect to how closely these species model technologically important metal-doped TiO2. This also has direct implications to the potential applications of these organically-soluble inorganic cages as photocatalytic redox systems in chemical transformations. Measurement of the band gaps of the series of closely related polyoxotitanium cages [MnTi14(OEt)28O14(OH)2] (1), [FeTi14(OEt)28O14(OH)2] (2) and [GaTi14(OEt)28O15(OH)] (3), containing interstitial Mn(II), Fe(II) and Ga(III) dopant ions, shows that transition metal doping alone does not lower the band gaps below that of TiO2 or the corresponding metal-doped TiO2. Instead, the band gaps of these cages are within the range of values found previously for transition metal-doped TiO2 nanoparticles. The low band gaps previously reported for 1 and for a recently reported related Mn-doped POT cage appear to be the result of low band gap impurities (most likely amorphous Mn-doped TiO2).

As a ubiquitous, persistent environmental contaminant, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has the potential to cause lethal deformities in larval fishes. Few studies have examined its impacts on larval growth and craniofacial development in conjunction with feeding capability. The authors used morphological and behavioral assessments to demonstrate that feeding capability of larvae is impaired even when craniofacial structures are not grossly malformed. Zebrafish embryos were exposed to 25 pg TCDD/mL, 50 pg TCDD/mL, or 100 pg TCDD/mL or <0.1% dimethyl sulfoxide for 1 h at 4 h postfertilization and then raised in clean water for 21 d or 90 d to assess craniofacial morphology, feeding capability, and long-term survival. The lower jaw was 5% smaller in 21-d larvae exposed to ≥ 50 pg TCDD/mL, and those larvae caught 10% fewer prey items; survival was reduced by 13% to 23%. The direct cause of TCDD's impacts on feeding capability is not known, but feeding success was correlated with growth, length of lower jaw, and survival. Since low larval mortality rates are key for recruitment, this suggests that exposure to concentrations of TCDD during embryonic development that do not initially cause mortality still has the potential to impact the recruitment success of feral fish. Furthermore, the present work provides additional evidence that behavioral end points are often more sensitive than morphological ones and should be included when assessing the sublethal toxicity of environmental contaminants.

Environmental contaminants are known to exert endocrine-disrupting effects on the reproductive axis of animals. Many of these molecules can affect steroid biosynthesis or estrogen-receptor signaling by behaving as estrogen-like molecules ("xenoestrogens"), or by exerting estrogenmodulatory effects. Exposure to some compounds has been correlated with the skewing of sex ratios in aquatic species, feminization and demasculinization of male animals, declines in human sperm counts, and overall diminution in fertility of birds, fish, and mammals. We herein devote space to several classes of endocrine-disrupting compounds (EDCs), including estrogenic substances such as bisphenol A (BPA), molecules that can behave at times anti-estrogenically while activating the aromatic hydrocarbon receptor (AHR), such as dioxins (a known human carcinogen), and novel, ubiquitous molecules such as nanoparticles, particularly gold nanoparticles (GNPs), that appear to alter the sexsteroid biosynthetic pathway.

Photoactive in one step! Once released into the environment, engineered nanoparticles (eNPs) are subjected to processes that may alter their physical or chemical properties, potentially altering their toxicity vis-à-vis the as-synthesized materials. We examined the toxicity to zebrafish ( Danio rerio ) embryos of CdSecore/ZnSshell quantum dots (QDs) before and after exposure to an in vitro chemical model designed to simulate oxidative weathering in soil environments based on a reductant-driven Fenton's reaction. Exposure to these oxidative conditions resulted in severe degradation of the QDs: the Zn shell eroded, Cd(2+) and selenium were released, and amorphous Se-containing aggregates were formed. Products of QD weathering exhibited higher potency than did as-synthesized QDs. Morphological endpoints of toxicity included pericardial, ocular and yolk sac edema, nondepleted yolk, spinal curvature, tail malformations, and craniofacial malformations. To better understand the selenium-like toxicity observed in QD exposures, we examined the toxicity of selenite, selenate, and amorphous selenium nanoparticles (SeNPs). Selenite exposures resulted in high mortality to embryos/larvae while selenate and SeNPs were nontoxic. Co-exposures to SeNPs + CdCl2 resulted in dramatic increase in mortality and recapitulated the morphological endpoints of toxicity observed with exposure to products of QD weathering. Cadmium body burden was increased in larvae exposed to weathered QDs or SeNP + CdCl2 suggesting the increased potency of products of QD weathering was due to selenium modulation of cadmium toxicity. Our findings highlight the need to examine the toxicity of eNPs after they have undergone environmental weathering processes.

Cobalt-containing polyoxotitanates (TiCo) are excellent precursors for the simple and scalable preparation of Nocera-type CoOx water-oxidation electrocatalysts. The TiCo cages serve as a reservoir for cobalt ions in a titania matrix on fluoride-doped tin oxide electrodes, and form, in situ, the active CoOx catalyst for O2 evolution with high stability in phosphate buffer in pH neutral water.

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD or dioxin) is a global environmental contaminant and the prototypical ligand for investigating aryl hydrocarbon receptor (AHR)-mediated toxicity. Environmental exposure to TCDD results in developmental and reproductive toxicity in fish, birds and mammals. To resolve the ecotoxicological relevance and human health risks posed by exposure to dioxin-like AHR agonists, a vertebrate model is needed that allows for toxicity studies at various levels of biological organization, assesses adverse reproductive and developmental effects and establishes appropriate integrative correlations between different levels of effects. Here we describe the reproductive and developmental toxicity of TCDD in feral fish species and summarize how using the zebrafish model to investigate TCDD toxicity has enabled us to characterize the AHR signaling in fish and to better understand how dioxin-like chemicals induce toxicity. We propose that such studies can be used to predict the risks that AHR ligands pose to feral fish populations and provide a platform for integrating risk assessments for both ecologically relevant organisms and humans., (Copyright © 2011 Elsevier Ireland Ltd. Background: Sexual risk-taking behaviors have a negative effect on the heath and future of American adolescents., Aims and Objectives: To gain insight into these behaviors and preventative efforts, this study explored the experiences and perceptions of 15 pregnant adolescents using a qualitative feminist approach and in-depth interviews., Methods: As participants discussed sex education, each identified influences on sexual decision-making that often overpowered the information received., Results: Content analysis and constant comparison of this data led to the overarching theme "Drivers for Sexual Risk-taking." Drivers were categorized as internal and external and each demonstrated a significant influence on adolescent sexual decision-making., Conclusion: Results of this study can inform the development of educational efforts, reform of social policy, and the focus of future research.

Over-expression of the multifunctional zinc-finger transcription factor Yin Yang 1 (YY1) has been associated with cellular proliferation and resistance to apoptotic stimuli. In this study, we report that YY1 was uniformly highly over-expressed in a wide range of human cancer cell lines and in human colon cancer tissue samples. The examination of YY1-specific mRNA expression demonstrated at least six mRNA isoforms ubiquitously expressed in normal human adult and fetal tissues. Substantial over-expression of two specific mRNA isoforms of 7.5 and 2.9 kb size, respectively, was detected in gastrointestinal and other cancer cells in vitro, whereby mRNA stability differed significantly between various cell lines. YY1 protein expression levels were similar in different colon cancer cell lines. Using FISH analysis of several colorectal cancer cell lines, the human YY1 locus was expectedly identified on chromosome 14q32 and no evidence of gene amplification and chromosomal translocation was observed. However, varying degree of aneuploidy was noted in vitro. YY1 immunoreactivity in human colon tumor samples was found more intense in poorly differentiated tumors than in moderately and well differentiated colon cancers and lower expression levels tended to be associated with shorter survival. In conclusion, YY1 was over-expressed in colon cancer in the absence of gene amplification and chromosomal translocation. YY1 mRNA and protein stability are important regulatory mechanisms of YY1 expression in colon cancer.

Little is understood regarding the impacts of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) exposure during early development on the health, survival, and reproductive capability of adults. Here we use zebrafish to determine whether early life stage exposure to TCDD induces toxicity in adult zebrafish and their offspring. Zebrafish were exposed to graded concentrations of TCDD (0-400 pg/ml) via waterborne exposure for 1 h/week from 0 to 7 weeks of age. The heart and swim bladder were identified as being most sensitive to TCDD exposure during early development. Subtle developmental toxic responses collectively impaired survival, and only zebrafish in the 0, 25, and 50 pg TCDD/ml groups survived to adulthood. Surviving fish exhibited TCDD toxicity in craniofacial structures (i.e., operculum and jaw), heart, swim bladder, and ovary. Exposure to 25 pg TCDD/ml impaired egg production (40% of control), fertility (90% of control), and gamete quality. TCDD-treated males contributed more than females to impaired reproductive capacity. Transgenerational effects were also discovered in that offspring from parents exposed to TCDD during early life stages showed a 25% increase in mortality compared with the F1 of dimethyl sulfoxide fish, reduced egg production (30-50% of control) and fertility (96% of control). Thus, adverse effects resulting from TCDD exposure during early life stages for one generation of zebrafish were sufficient to cause adverse health and reproductive effects on a second generation of zebrafish. In the environment, transgenerational effects such as these may contribute to population declines for the most TCDD sensitive fish species.

Quantum dots (QDs) hold promise for several biomedical, life sciences, and photovoltaic applications. Substantial production volumes and environmental release are anticipated. QD toxicity may be intrinsic to their physicochemical properties, or result from the release of toxic components during breakdown. We hypothesized that developing zebrafish could be used to identify and distinguish these different types of toxicity. Embryos were exposed to aqueous suspensions of CdSe(core)/ZnS(shell) QDs functionalized with either poly-L-lysine or poly(ethylene glycol) terminated with methoxy, carboxylate, or amine groups. Toxicity was influenced by the QD coating, which also contributed to the QD suspension stability. At sublethal concentrations, many QD preparations produced characteristic signs of Cd toxicity that weakly correlated with metallothionein expression, indicating that QDs are only slightly degraded in vivo. QDs also produced distinctly different toxicity that could not be explained by Cd release. Using the zebrafish model, we were able to distinguish toxicity intrinsic to QDs from that caused by released metal ions. We conclude that developing zebrafish provide a rapid, low-cost approach for assessing structure-toxicity relationships of nanoparticles.

Introduction: We investigated clinical and pathologic features of breast cancers (BC) in an unselected series of patients diagnosed in a tertiary care hospital serving a diverse population. We focused on triple-negative (Tneg) tumours (oestrogen receptor (ER), progesterone receptor (PR) and HER2 negative), which are associated with poor prognosis., Methods: We identified female patients with invasive BC diagnosed between 1998 and 2006, with data available on tumor grade, stage, ER, PR and HER2 status, and patient age, body mass index (BMI) and self-identified racial/ethnic group. We determined associations between patient and tumour characteristics using contingency tables and multivariate logistic regression., Results: 415 cases were identified. Patients were racially and ethnically diverse (born in 44 countries, 36% white, 43% black, 10% Hispanic and 11% other). 47% were obese (BMI > 30 kg/m2). 72% of tumours were ER+ and/or PR+, 20% were Tneg and 13% were HER2+. The odds of having a Tneg tumour were 3-fold higher (95% CI 1.6, 5.5; p = 0.0001) in black compared with white women. Tneg tumours were equally common in black women diagnosed before and after age 50 (31% vs 29%; p = NS), and who were obese and non-obese (29% vs 31%; p = NS). Considering all patients, as BMI increased, the proportion of Tneg tumours decreased (p = 0.08)., Conclusions: Black women of diverse background have 3-fold more Tneg tumours than non-black women, regardless of age and BMI. Other factors must determine tumour subtype. The higher prevalence of Tneg tumours in black women in all age and weight categories likely contributes to black women's unfavorable breast cancer prognosis.

It has been reported that the phase transitions found in the quasi-one-dimensional sulfide KCu(7-x)S4 are most likely due to vacancy ordering involving Cu+-ion diffusion along the Cu(2)-Cu(2) zigzag chains. Our previous studies with both a self-consistent method and Monte Carlo simulations confirmed that phase transitions indeed exist in a one-dimensional (1D) lattice gas system in which vacancy ordering is involved. In this paper, we calculate the more nearly real case of KCu6.88S4 and further investigate the angular dependence of the phase properties in a partially occupied 1D zigzag chain with various particle occupancies. The calculated results suggest that the phase transitions that occur in the quasi-one-dimensional material KCu(7-x)S4 are presumably due to both intrachain and interchain interactions between the partially occupied Cu+ zigzag chains. Most interestingly, we found that the average particle distribution of the lowest free energy state is a linear superposition of two other solutions with different particle distributions for occupancy n(av)=1/2.

Novel systemic treatments are needed in pancreatic cancer. The authors sought to establish the frequency of overexpression of the HER-2/neu oncogene in patients with pancreatic adenocarcinoma to determine the potential role of trastuzumab (Herceptin) as a therapeutic agent in this disease. Tumor specimens from patients with pancreatic adenocarcinoma were analyzed by staining for p185HER2 protein using the DAKO immunohistochemical assay. Patients with and without HER-2/neu overexpression by immunohistochemistry were compared with respect to clinical and pathologic characteristics. HER-2/neu gene amplification was also evaluated by fluorescence in situ hybridization (FISH). Thirty-two of 154 patients (21%) had pancreatic adenocarcinoma that demonstrated HER-2/neu overexpression by immunohistochemistry. At initial diagnosis, 16% of resectable cancers, 17% of locally advanced cancers, and 26% of metastatic cancers were determined to have HER-2/neu overexpression. Three of 11 (27%) patients with HER-2/neu overexpression by immunohistochemistry had gene amplification by FISH. HER-2/neu overexpression occurs in a subset of pancreatic cancer. Evaluation of the efficacy of trastuzumab for patients with pancreatic cancer who overexpress HER-2/neu appears indicated.
