Abstract

Results reveal five palindromic sequences in the telomere terminal 116 nucleotide segment that forms the hairpin loop structure in the linear chromosome of Streptomyces griseus. The terminal inverted repeat, involved in terminal replication, is a 24 kilobase in length and contain two almost identical open reading frames.

The authors discuss cloning and sequencing of circularized chromosomes in Streptomyces griseus 404-23 and N2 to analyze their fusion junctions. Results indicate that chromosomal circulation prevents lethal deletions.

Abstract

A-factor (2-isocapryloyl-3R-hydroxymethyl-gamma-butyrolactone) is needed for the initiation of aerial mycelium formation in Streptomyces griseus. The amfR gene is one of the genes which, when cloned on a low-copy-number plasmid, suppresses the aerial mycelium-negative phenotype of an A-factor-deficient mutant of S. griseus. Findings showed that amfR is needed for aerial growth. A protein, AdpB, which binds the promoter of the amfR operon only in the absence of A-factor and repressor-like behavior of AdpB, was identified.

Abstract

A linear chromosome physical map of Streptomyces griseus 2247 was presented and its terminal construction was examined. The chromosome DNA was digested with various restriction endonucleases and studied by pulse-field gel electrophoresis (PFGE). Fifteen fragments from AseI and nine fragments from DraI were obtained. A 24-kb terminal inverted repeat was found on each end. PFGE analysis indicated the presence of a protein molecule at each end of the chromosome.

Abstract

The article presents a study on the fabrication of a biofertilizer which contains high-nitrogen (N2) fixation capacity, and phosphate and potassium solubilizing bacteria. The study involves a collection of microorganisms that produce high amount of auxin in several agricultural lands in Thailand. It employs the acetylene reduction method in measuring the N2 fixation activity. Results of the study showed the improvement of vegetable growth applied with the new biofertilizer.

Abstract

The article presents a study on the use of coconut water and sugar-cane juice to carried out extracellular polysaccharide (EPS) production by Lactobacillus confusus in liquid and solid state fermentation. The study showed that high concentrations of EPS of sugarcane juice and coconut water were produced in solid state fermentation by reducing nitrogen sources. Moreover, it mentions that coconut water and sugar-cane juice were subjected to solid state fermentation in enhancing EPS production.

Extracellular polysaccharide (EPS) production by Lactobacillus confususin liquid and solid state fermentation was carried out using coconut water and sugarcane juice as renewable wastes. High concentrations of EPS of 62 (sugarcane juice) and 18 g/l of coconut water were produced in solid state fermentation when nitrogen sources were reduced 5-fold from the original medium.

Abstract

Saccharomyces cerevisiae Rrs1p is a nuclear protein that is essential for the maturation of 25 S rRNA and the 60 S ribosomal subunit assembly. In two-hybrid screening, using RRS1 as bait, we have cloned YKR081c/RPF2. Rpf2p is essential for growth and is mainly localized in the nucleolus. The amino acid sequence of Rpf2p is highly conserved in eukaryotes from yeast to human. Similar to Rrs1p, Rpf2p shows physical interaction with ribosomal protein L11 and appears to associate with preribosomal subunits fairly tightly. Northern, methionine pulse-chase, and sucrose density gradient ultracentrifugation analyses reveal that the depletion of Rpf2p results in a delayed processing of pre-rRNA, a decrease of mature 25 S rRNA, and a shortage of 60 S subunits. An analysis of processing intermediates by primer extension shows that the Rpf2p depletion leads to an accumulation of 27 SB pre-rRNA, suggesting that Rpf2p is required for the processing of 27 SB into 25 S rRNA.

Abstract

The article presents a study on the capacity for oil and chemical oxygen demand (COD) removal of a thermotolerant photosynthetic bacterium, Rhodobacter sphaeroides Strain, at high temperatures. After isolating Rhodobacter sphaeroides, the alginate-immobilized cells of strain NAT were used in high temperature treatment of artificial sewage wastewater containing oil. It was observed that the COD was decreased by 80% and 76% of the oil was removed after 96 hours of treatment at 55 degrees Celsius.

Abstract

The gene encoding streptomycin 6-kinase involved in the self-resistance of the streptomycin-producing Streptomyces griseusHUT 6037 was cloned in the plasmid vector pIJ703. The resulting plasmid, pSP6, contained 2.5 kb inserts of S. griseusDNA. When streptomycin-susceptible S. lividans1326 was retransformed with pSP6, all transformants produced streptomycin 6-kinase. Addition of streptomycin to the culture medium of S. lividanscarrying pSP6 plasmid brought about a remarkable increase in streptomycin 6-kinase activity in the cell extracts. It is suggested from the results that the production of streptomycin 6-kinase in streptomycin producer was induced by streptomycin accumulated during cultivation.

A thermotolerant photosynthetic bacterium NAT identified as Rhodobacter sphaeroideswas isolated. When alginate-immobilized cells of strain NAT were used in high-temperature treatment of artificial sewage wastewater containing oil, the chemical oxygen demand (COD) decreased by 80% and 76% of the oil was removed after 96 h of treatment at 55 °C. Lipase activity was observed in the culture.
