Background: Syzygium cumini (locally known as Kalo Jam) seed powder is being used as folk medicine in various diseases since unknown times in Bangladesh. However, data on the systematic studies in human subjects are rare. Objective: To determine the effect of Syzygium cumini seed powder on the hepatic function-related enzymes of the alcoholic transport laborers. Methods: Here, we investigated the effects of oral administration of Syzygium cumini seed powder on hepaticfunction markers that included activities of Gamma-glutamyl transferase (GGT), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and renal-function markers such as creatinine and urea along with lipid profile, blood pressure and body mass index (BMI) upon chronic alcoholic human volunteers. Results: The levels of serum ALT, AST and GGT levels decreased, respectively, by 15.0, 8.5 and 23.0% after oral administration of the S. cumini powder in the alcoholic subjects. However, the levels of creatinine and urea were not altered significantly. In contrast, the effects of administration of S. cumini on the hepatic ALT, AST and GGT enzymes in the non-alcoholic control subjects were less prominent than those found in the alcoholic subjects. Blood pressure decreased only by 5-4 mmHg, while BMI was not altered in either the alcoholic or the non-alcoholic groups. Furthermore, there were significant reduction in the levels of TC (by 10%) and LDL-C (by >20%) and elevation of HDL-C (by 11%) in the S. cumini-prescribed alcoholic subjects. TC and LDL-C levels also decreased in the non-alcoholic subjects, but the rate was not statistically significant. Conclusions: Our results suggest that S. cumini seed extract ameliorated GGT, ALT and the atherosclerotic lipid parameters TC and LDL-C without having a significant side effect on the kidney functions, as indicated by the unaltered levels of serum creatinine levels in the alcoholic human subjects without any effect on BP and BMI of the same subjects. Chitin provides immense beneficial roles to the humanity and environment. Most of the chitin extracted worldwide are from the shell of the crustaceans. An alternative source of chitin has been observed as the number of crustaceans has been dilapidating. Here, extraction of chitin from the edible and medicinal macrofungi, mushrooms, have been described. This is a novel approach to meet the ever increasing chitin demand worldwide. Among four mushroom species (Pleurotus ostreatus, Agaricus bisporus, Lentinula edodes and Ganoderma lucidum), the reishi mushroom or ling zhi (G. lucidum) had been found containing the highest amount of chitin on percentage basis of dried mushroom powder. Chitin content trend observed in this study was: G. lucidum (44%) > L. edodes (35%) > A. bisporus (18%) > P. ostreatus (10%). Thus, mushrooms could be considered as important source of chitin and its deacetylated product, chitosan. In this way, mushrooms could aid in biomedical and environmental intervention through suppliance of chitin and chitosan.

Raoultella ornithinolytica is an emerging pathogen that causes human infections. We report the isolation and genome sequencing of R. ornithinolytica from an oral swab of a Persian pet cat in Dhaka, Bangladesh. The genome length was 5,375,160 bp, with 55.9% G+C content. It carries putative genes associated with resistance to antibiotics and metals.

The purpose of this cross sectional study was to assess the extent of depression, and the quality of life (QoL) among confirmed Covid-19 Bangladeshi patients tested positive for Covid19 from October 2020 to March 2021. This tele-interview survey included 380 patients. Eligible participants were identified from all available phone numbers of the research population obtained from various RT-PCR labs run by the government of the People's Republic of Bangladesh. The level of depression, pain and QoL was assessed with PHQ-9, Visual analogue scale (VAS), and short form-12 V2 (SF-12v2) survey instrument respectively. Depression was prevalent in 4 out of every 5 patients. More than two-third of participants had worse physical and mental health than the average citizen after being diagnosed as COVID-19 positive case. Around two-third of respondents had mild to severe pain after recovery from COVID-19. Younger participants (aged ≤50 years) had relatively better QoL and less pain intensity compared to those of older participant (aged >50 years). The respondents who had higher intensity of subjective pain, and worse than the average physical health had higher frequency of depression. Individuals having history of covid-19 are especially vulnerable to develop pain which was associated with depression and lower quality of life during the COVID-19 pandemic.

Thymoquinone is a pharmacologically active component of Nigella sativa Linn. seeds. Despite the diverse neuropharmacological attributes of TQ, limited reports related to adult neurogenesis and memory research are available. In this study, we investigated the effects of TQ on the proliferation and neural differentiation of cultured neural stem/progenitor cells (NSCs/NPCs). We also investigated the effect of TQ chronic administration on neurogenesis and memory in adult rats. Under proliferation conditions, TQ (0.05-0.3 μM) significantly increased NSCs/NPCs viability, neurosphere diameter, and cell count. TQ treatment under differentiation conditions increased the proportion of cells positive for Tuj1 (a neuronal marker). Furthermore, chronic oral administration of TQ (25 mg/kg/day for 12 weeks) to adult rats increased the number of bromodeoxyuridine (BrdU)-immunopositive cells double-stained with a mature neuronal marker, neuronal nuclei (NeuN), and a proliferation marker, doublecortin (Dcx), in the dentate gyrus of the hippocampus. TQ-administered rats showed a profound beneficial effect on avoidance-related learning ability, associated with an increase in the hippocampal mRNA and protein levels of brain-derived neurotrophic factor (BDNF), as measured by both real-time PCR and ELISA. Western blot analysis revealed that TQ stimulates the phosphorylation of cAMP-response element-binding protein (CREB), the upstream signaling molecule in the BDNF pathway. Furthermore, chronic administration of TQ decreased lipid peroxide and reactive oxygen species levels in the hippocampus. Taken together, our results suggest that TQ plays a role in memory improvement in adult rats and that the CREB/BDNF signaling pathways are involved in mediating the actions of TQ in hippocampal neurogenesis.

Ever increasing physiological and environmental abnormalities warrant natural, safe, easy to extract, less expensive and eco-friendly bio-components of versatile usage. Chitin, the second most abundant polysaccharide in the world, seems promising in fighting diseases, boosting immunity up and maintaining environment fresh. Chitin is a polysaccharide of N-acetyl-D-glucosamine forms the exoskeletons of arthropods, mollusks, and insects, cell walls of fungi and scales of fishes. Based on extraction, purification and integration enhancement, chitin could be of greater importance for the maintenance of human health, agriculture and environment. Present article compares and contrasts the methods utilized for extraction and purification of chitin from different sources and review its current fields and suggest the future aspects.

Parkinson’s disease (PD) results mostly from the degeneration of dopaminergic neurons in the substantia nigra pars compacta. It is characterized by the manifestations of anxiety, tremor, muscle rigidity, bradykinesia and cognitive impairments. Oxidative stress is attributed to the neurodegeneration of PD. Curcuma longa is a commonly used spice and has high levels of anti-oxidative phyto-constituents. Here, we evaluated whether the chronic oral administration of C. longa extract affects the degree of anxiety, neuromotor and cognitive deficits of 6-hydroxydopamine (6-OHDA)-infused PD model rats. Following the surgery, animals were orally administered with C. longa extract for twelve weeks. The degree of anxiety/depression, motor and cognitive performances were determined with a variety of neurobehavioral experimental paradigms, including elevated plus, Morris water maze task, rotarod, open field, forced swim, sucrose preference and tail suspension tests. The pathogenic mechanism of dementia is still unknown, and the fundamental treatment remains to be established. Thus, there is growing interest in preventing dementia through diet. One of the functional ingredients attracting attention is docosahexaenoic acid. We conducted a 12-month, randomized, double-blind, placebo-controlled clinical trial in healthy elderly Japanese individuals with a Mini-Mental State Examination score of 28 or higher at baseline using a docosahexaenoic acid-enriched milk beverage containing 297 mg docosahexaenoic acid and 137 mg eicosapentaenoic acid. Consumption of a docosahexaenoic acid-enriched milk beverage increased the fatty acid levels of docosahexaenoic acid and eicosapentaenoic acid in erythrocyte membranes, which was the primary outcome of this study. Moreover, intake of this beverage prevented age-related cognitive decline and decreased serum bone resorption marker levels. Our data demonstrate that, even at a low dose, long-term daily intake of docosahexaenoic acid prevents dementia and may show beneficial effect on bone health.

Learning and memory impairment may result from age-related decline in synaptic plasticity-related proteins in the hippocampus. Therefore, exploration of functional foods capable of ameliorating memory and cognition decline is an interesting endeavor in neuroscience research. We report the effects of Anredera cordifolia (AC) extract on learning and memory deficits in a senescence-accelerated mouse-prone 8 (SAMP8) mouse model, which demonstrate age-related memory deficits and related pathological changes in the brain. After 8 weeks of oral administration of AC extract, the mice were trained in the Novel Object Recognition (NOR) task, and after 7 more weeks, in the Morris Water Maze (MWM) task. Following the completion of behavioral testing, the blood biochemistry parameters, the hippocampal levels of brain-derived neurotropic factor (BDNF), PSD95, and NR2A, and the p-cAMP-response element binding (p-CREB)/CREB ratio were measured. The AC-treated group spent more time exploring the novel objects in the NOR task, and showed faster acquisition and better retention in the MWM task than the negative control (CN) group. In addition, AC enhanced the levels of the aforementioned neuronal plasticity-related proteins, and did not affect the blood biochemistry parameters. Therefore, our data suggest that the AC extract may improve learning and memory without causing any noticeable side effects in the body.

Although brown rice (BR) contains significantly higher levels of nutrients than the traditionally used polished white rice (WR), its consumption among the population is still not noteworthy. WR and BR are essentially same grain. The only difference between the two is the application of an exhaustive milling procedure during the processing of WR that removes all other layers of the grain except the portion of its white endosperm. BR, on the other hand, is prepared by removing only the outer hull of the rice seed. Thus, in addition to its inner endosperm, the bran and germ are also left on the BR. Hence, BR retains all its nutrients, including proteins, lipids, carbohydrates, fibers, vitamins, minerals, tocopherols, tocotrienols, γ-oryzanol, and γ-aminobutyric acid (GABA) packed into the bran and germ of the seed. Since BR tastes nutty and takes longer to cook than WR, it is not appreciated by the consumers. However, these problems have been circumvented using non-thermal ultra-high hydrostatic pressure (UHHP)-processing for the treatment of BR. A superior modification in the physicochemical and functional qualities of UHHPBR, along with its ability to curb human diseases may make it a more palatable and nutritious choice of rice over WR or the untreated-BR. Here, we have reviewed the mechanism by which UHHP treatment leads to the modification of nutrients such as proteins, lipids, carbohydrates, and fibers. We have focused on the effects of rice on cell and animal models of different conditions such as hyperlipidemia, diabetes, and hypertension and the possible mechanisms. Finally, we have emphasized the effects of UHHPBR in human cases with rare conditions such as osteoporosis and brain cognition - two age-related degenerative diseases of the elderly population.

Lablab purpureus (L) bean is an undervalued or underutilized orphan crop in many tropical countries, where different forms of malnutrition are associated with stunting growth and cognitive deficiencies. We previously reported that L. purpureus contains ω‐3 α‐linolenic acid (ALA, C18:3, ω‐3), which can act as the precursor of ω‐3 docosahexaenoic acid (DHA, C22:6, ω‐3). Inadequate level of DHA impairs growth, development, and cognitive performance. Therefore, we evaluated if supplementation of L. purpureus seed flour (LPS) affects the nutritional status, in terms of body weight gain, plasma proteins, and DHA levels of malnourished model rats. Three groups of rats, namely, controls, malnourished alone (MN), and 15% LPS‐supplemented malnourished (MN + LPS) rats were fed with LPS for 12 weeks. Afterward, body weight, liver weight, brain weight, plasma proteins, micronutrients, lipid profile, and fatty acid profile of plasma, liver, and brains were determined by standard methods. The levels of liver lipid peroxide (LPO) and proinflammatory TNFα were also measured. The body weight, liver weight, serum total proteins and micronutrients (iron/potassium), and the levels of docosahexaenoic acid significantly increased in the plasma, liver, and brain of MN + LPS rats. Moreover, LPO and TNFα levels reduced significantly in MN + LPS rats. in vitro analysis revealed a significant free radical scavenging and antioxidative potential of L. purpureus seed extract. Thus, L. purpureus not only replenishes protein‐energy malnutrition, but also increases the levels of DHA, an indispensable polyunsaturated fatty acid for brain cognition. Finally, our results suggest that L. purpureus might benefit human malnutrition and related cognitive deficits.

Alzheimer's disease (AD) is the leading neurodegenerative disorder affecting memory and learning of aged people. Hypercholesterolemia had been implicated as one of the stark hallmarks of AD. Recent AD control guidelines have suggested lifestyle modification to slow down the progression of AD. In this regard, medicinal mushroom Ganoderma lucidum seems apt. In the present study, hot water extract of G. lucidum (200 mg/kg body weight) was fed to the hypercholesterolemic and AD model rats for 8 weeks. Nonspatial memory and learning abilities of the model animals was assessed using novel object recognition (NOR) test, rotarod test, and locomotor/open-field test. Then, the animals were sacrificed and transmission electron micrograph (TEM) view of the hippocampal neurons was assessed. In all the nonspatial memory and learning tests, the G. lucidum HWE fed rats performed better indicating improved memory and learning abilities. TEM view showed regular arrangement of the neurons in the G. lucidum HWE fed rats compared with those of the deranged arrangement of the AD rats. G. lucidum might have aided in restoring the memory and learning abilities of the AD model animals through maintaining neuronal structure and function. Thus, G. lucidum could be suggested as a medicotherapeutic agent against AD.

The skin mucosa of fish serves as a primary barrier against pathogens. In lesion sites in diseased fish, the mucosal barrier is expected to be compromised, with a substantial presence of potential pathogens. An understanding of the skin microbiome and its functional repertoire would provide important insights into host-microbe interactions, which has important implications for prophylactic measures in aquaculture. This study revealed the skin microbiomes and their functional annotations from healthy and diseased stinging catfish (Heteropneustes fossilis) based on 16S rRNA metagenomics. The OTUs consisted of four major phyla, Proteobacteria, Bacteroidota, Actinobacteriota and Firmicutes. Among members of the predominant phyla, Proteobacteria were rich in healthy fishes, but Bacteroidota and Firmicutes were significantly differentiated in healthy and diseased fish. The diversified microbiome was high in the skin of healthy fishes and did not significantly differ from that of the diseased groups. At the genus level, Pseudomonas showed the highest abundance in healthy fish but was nearly absent in diseased fish, whereas Flavobacterium showed the highest abundance in diseased fish. Linear discriminant analysis identified two phyla (Bacteroidota, Firmicutes) and two genera (Flavobacterium, Allorhizobium) that were consistently identified in diseased fishes. Functional prediction analysis specified that the genes related to physiological functions such as metabolism, immune and digestive systems and environmental adaptations could be highly expressed in diseased fishes. The present study indicates that the compositions, richness and functions of the bacterial community could influence the health status of cultured stinging catfish. Aquaculture-associated pathogenic bacteria may be identified, and preventive measures can be taken for the surveillance of fish health.

Plasmalogens are alkenyl-acyl glycerophospholipids and decreased in post-mortem Alzheimer’s disease (AD) brains. The aim of this study is to investigate the time-dependent changes of plasmalogens in the hippocampus of an AD model mouse (J20). Plasmalogen levels at 3, 6, 9, 12 and 15 months were analyzed by liquid-chromatography-targeted-multiplexed-selected-reaction-monitoring-tandem-mass-spectrometry (LC-SRM/MS). Reactive oxygen species (ROS) levels were evaluated using dichlorofluorescein diacetate (DCF-DA). Plasmalogen synthesizing enzyme glycerone-phosphate O-acyltransferase (GNPAT) and late endosome marker Rab7 levels were quantified by Western blotting. GNPAT localization, changes of neuronal and glial cell numbers were evaluated by immunostaining. Compared to wild-type mice (WT), total plasmalogen-ethanolamine, but not plasmalogen-choline levels, were increased at 9 months and subsequently decreased at 15 months in J20 mice. A principal component analysis of plasmalogen-ethanolamine species could separate WT and J20 mice both at 9 and 15 months. Both GNPAT and Rab7 protein were increased in J20 mice at 9 months, whereas GNPAT was decreased at 15 months. ROS levels were increased in J20 mice except for 9 months. Our results suggest that increased plasmalogen-ethanolamine could counteract ROS levels and contribute to the phagocytosis process in J20 mice at 9 months. Such results might indicate a transient protective response of plasmalogen-ethanolamine in AD conditions.

Pregerminated brown rice (GBR) is assumed to be more beneficial than polished white rice (WR), with regard to nutrition and cardiovascular health. To support this with scientific evidence, cholesterol-lowering and antioxidative effects of GBR were studied in the present investigation. The most popular rice variety in Bangladesh BIRI-29 was used to prepare GBR and WR. Initially, we analyzed the proximate composition, antioxidative phytochemicals, in vitro 2, 2-diphenyl-1-picrylhydrazyl (DPPH)-free radical scavenging ability and anti-hemolytic effects of GBR. To examine the dietary impact and possible benefits of the GBR, experimentally-induced hypercholesterolemic (HC, 1% cholesterol) rats were fed with GBR against WR for 12 wk. At the end, plasma total cholesterol (TC), low- and high-density lipoprotein-cholesterol (LDL-C and HDL-C), triglyceride (TG), fecal TC, and hepatic TC, lipid peroxide (LPO) and proinflammatory TNFα levels were determined. Relative to WR, GBR contained higher amounts of total polyphenols, total flavonoids, β-carotene and lycopene, and exhibited a stronger in vitro DPPH-free radical scavenging ability and antihemolytic potentials. Levels of plasma TC, LDL-C, TG, and hepatic TC and TG significantly decreased, while plasma HDL-C and fecal TC levels significantly increased in the GBR-fed HC-rats, indicating dietary GBR demonstrates a stronger antilipidemic effect than WR. An experiment was conducted to evaluate the effects of salinity and variety on yield and yield contributing characters of Red amaranth at Horticulture Farm of Bangladesh Agricultural University, Mymensingh. The experiment consisted of three varieties viz; Altapety, RM, Deshi and four levels of salinity viz; 0mM, 25mM, 50mM and 75mM. The experiment was laid out following RCBD with three replications. Variety had significant effect on yield characters and yield. The highest average weight of plant (9.69 g/pot), highest average root weight (2.13 g/pot), highest average root length (10.28 g/pot) and highest yield per pot (237.08g) were found from the variety Altapety and the lowest average weight of plant (7.67 g/pot), lowest average root weight (2.03 g/pot), lowest average root length (8.83 g/pot) and yield per pot (198.58g) were found from the variety RM. Salinity had significant effect on yield characters and yield. The highest average weight of plant (10.22 g/pot) was recorded from the variety Altapety at the application of 25mM NaCl, highest average of root weight (2.27 g/pot) was recorded from the variety Altapety at the application of 25mM NaCl, highest average of root length (10.29 g/pot) was recorded from the variety Altapety at the application of 25mM NaCl and yield (259.56g/pot) were found from the application of 25mM NaCl. Oxidative stress plays an important role in age-associated cognitive decline. We recently reported that dietary intake of perilla seed oil (PO), a rich source of α-linolenic acid (LNA, C18:3, ω-3), helps in maintaining good mental health in adults. This study aimed to investigate the impacts of dietary PO intake on cognitive functions and mental health in healthy, elderly Japanese individuals. Seventy-five healthy volunteers aged 64–84 years were randomly divided into two groups: a control group and a PO-administered group. At baseline and at 12 months of intervention, cognitive function, mental health condition, fatty acid profile of the red blood cell plasma membranes (RBC-PM), and serum biochemical parameters were evaluated. Results showed that serum biological antioxidant potential and LNA levels in the RBC-PM at 12 months after the trial were significantly higher in the PO group compared to the control group. Further, both the cognitive function measures, as evaluated by the Frontal Assessment Battery test and the apathy scores, tended to be improved after 12 months in the PO group. Our results demonstrate that dietary PO intake enhances the antioxidant potential and prevents the age-related cognitive and mental decline in healthy elderly individuals by enhancing the blood LNA levels.

Recent approaches to the management of dementia have focused mainly on continual preventive treatment. In this study, we investigated whether intake of 297 mg docosahexaenoic acid (DHA) and 137 mg eicosapentaenoic acid (EPA) per day, which is a smaller amount than in previous reports, would maintain higher cognitive function in healthy elderly people. Eighty-seven participants were enrolled in this 12-month randomized, double-blind, placebo-controlled trial. Cognitive function test, blood biochemical analysis and anthropometry were conducted at baseline, 6 and 12 months of DHA beverage intake. Intake of DHA-enriched milk beverage for 6 and 12 months significantly increased erythrocyte plasma membrane DHA and EPA levels. Furthermore, intake of DHA-enriched milk beverage for 12 months protected against age-related cognitive decline. The preventive mechanism of this small amount of DHA is not known. Thus, further investigation is needed to reveal the mechanism underlying its role in cognitive function. Trial registration: UMIN Clinical Trial Registry, UMIN000031699.

Ganoderma lucidum has been hailed as medicinal mushroom. Its effect on memory and learning related behavioral performance along with related protein markers has been evaluated using Alzheimer’s disease (AD) and hypercholesterolemic model rats in the present study. AD model rats were prepared infusing amyloid beta peptide into the right ventricles of the rats. Hypercholesterolemia was evoked feeding 1% cholesterol and 1% cholic acid with basal diet of the rats for 8 weeks. Hot water extract of G. lucidum was ingested orally (200 mg/kg bw) to the HC and AD model rats. Memory and learning related behavioral tests were performed using Barnes maze while protein markers (BDNF, SNAP2, PSD-95, VAchT) were detected using ELISA. Observed findings suggest hypocholesterolemic, lipid profile improving and enhanced cognitive performance of the G. lucidum fed rats. Memory and learning related protein markers also substantiate this fruition. Thus, therapeutic potentiality of Ganoderma lucidum in AD amelioration seems promising. In this study, a series of bacteria capable of degrading starch and cellulose were isolated from the aging flue-cured tobacco leaves. Remarkably, there was a thermophilic bacterium, Bacillus subtilis ZIM3, that can simultaneously degrade both starch and cellulose at a wide range of temperature and pH values. Genome sequencing, comparative genomics analyses, and enzymatic activity assays showed that the ZIM3 strain expressed a variety of highly active plant biomass-degrading enzymes, such as the amylase AmyE1 and cellulase CelE1. The in vitro and PhoA-fusion assays indicated that these enzymes degrading complex plant biomass into fermentable sugars were secreted into ambient environment to function. Besides, the amylase and cellulase activities were further increased by three- to five-folds by using overexpression. Furthermore, a fermentation strategy was developed and the biodegradation efficiency of the starch and cellulose in the tobacco leaves were improved by 30–48%. These results reveal that B. subtilis ZIM3 and the recombinant strain exhibited high amylase and cellulase activities for efficient biodegradation of starch and cellulose in tobacco and could potentially be applied for industrial tobacco fermentation.

Hypercholesterolemia has been implicated as one of the pathomechanistic factors of Alzheimer's disease (AD), the most common neurodegenerative disorder affecting memory and learning abilities. In the present study, ameliorative effect of hot water extract (HWE) of mushroom Ganoderma lucidum to the memory and learning related behavioral performance of hypercholesterolemic and AD rats was investigated using Morris water maze (MWM). Male Wistar rats were randomly grouped into control, extract fed control, hypercholesterolemic, extract fed hypercholesterolemic, AD, and extract fed AD groups, each group containing 8 animals. Hypercholesterolemia was induced in rats by adding 1% cholesterol and 1% cholic acid with the basal diet of the respective group. Alzheimer's disease model rats were prepared through infusion of amyloid β(1-42) to the right ventricle. Memory and learning related performance of all the rats was tested for 6 consecutive days that included time taken to reach the submerged platform (sec) and distance traveled (m). G. lucidum HWE fed rats took less time and traveled less distance to find the submerged platform, which indicates the spatial learning and memory related behavioral amelioration of the extract fed rats compared with their non-fed counterparts. Thus, usage of G. lucidum seems promising in withstanding hypercholesterolemia-induced Alzheimer's disease pathogenesis.

Oxidized low-density lipoprotein (Ox-LDL) is known to be highly atherogenic. Thus, decreasing the blood levels of Ox-LDL through dietary means is an important approach to reduce cardiovascular events in high-risk individuals. In this randomized placebo-controlled human interventional trial, we aimed to evaluate whether Perilla frutescens leaf powder (PLP) ameliorates Ox-LDL and home blood pressure, along with its biological antioxidant potential. Healthy Japanese volunteers aged 30&ndash, 60 years (n = 60) were randomized to PLP and placebo groups. The PLP group consumed PLP dried using a microwave under reduced pressure, and the placebo group consumed pectin fiber daily for 6 months. Home blood pressure, serum biochemical parameters, and fatty acid profiles of erythrocyte plasma membranes were analyzed. Plasma Ox-LDL levels significantly decreased in the PLP group but not in the placebo group. Mean changes in the biological antioxidant potential and alpha-linolenic acid levels in the erythrocyte plasma membrane were significantly increased in the PLP group than in the placebo group. In subjects with prehypertension (systolic blood pressure [SBP] &sup3, 120 mmHg), the mean reduction in morning or nocturnal SBP was significantly greater in the PLP group than in the placebo group. Thus, PLP intake may be an effective intervention to prevent cardiovascular diseases.

Perilla oil (PO), rich in &alpha, linolenic acid (LNA, C18:3, &omega, 3), is increasingly alleged to have numerous health benefits in humans. However, the current reports detailing the effects of PO on human mental health are not adequate. Therefore, in the current investigation we compared the effects of PO or placebo treatment on the mental condition of healthy adult Japanese volunteers. At baseline and after 12 months of treatment, mental health condition was assessed using the Zung Self-Rating Depression Scale (SDS) and Apathy Scale, and serum biochemical parameters were determined. From baseline to 12 months of intervention, both SDS depression and apathy scores improved significantly in the PO-administered group. Compared to those of control group, serum norepinephrine and serotonin levels after 12 months decreased in the PO-administered group. The enhanced mental state observed in PO-subjects was accompanied by LNA level increases in erythrocyte plasma membranes. Our data demonstrate that PO intake enhances blood LNA levels and may maintain healthy mental conditions in adult subjects.

The effects of cholesterol-lowering statins, which substantially benefit future cardiovascular events, on fatty acid metabolism have remained largely obscured. In this study, we investigated the effects of atorvastatin on fatty acid metabolism together with the effects of TAK-085 containing highly purified eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) ethyl ester on atorvastatin-induced n-3 polyunsaturated fatty acid lowering in SHR.Cg-Leprcp/NDmcr (SHRcp) rats, as a metabolic syndrome model. Supplementation with 10mg/kg body weight/day of atorvastatin for 17 weeks significantly decreased plasma total cholesterol and very low density lipoprotein cholesterol. Atorvastatin alone caused a subtle change in fatty acid composition particularly of EPA and DHA in the plasma, liver or erythrocyte membranes. However, the TAK-085 consistently increased both the levels of EPA and DHA in the plasma, liver and erythrocyte membranes. After confirming the reduction of plasma total cholesterol, 300mg/kg body weight/day of TAK-085 was continuously administered for another 6 weeks. Supplementation with TAK-085 did not decrease plasma total cholesterol but significantly increased the EPA and DHA levels in both the plasma and liver compared with rats administered atorvastatin only. Supplementation with atorvastatin alone significantly decreased sterol regulatory element-binding protein-1c, Δ5- and Δ6-desaturases, elongase-5, and stearoyl-coenzyme A (CoA) desaturase-2 levels and increased 3-hydroxy-3-methylglutaryl-CoA reductase mRNA expression in the liver compared with control rats. TAK-085 supplementation significantly increased stearoyl-CoA desaturase-2 mRNA expression. These results suggest that long-term supplementation with atorvastatin decreases the EPA and DHA levels by inhibiting the desaturation and elongation of n-3 fatty acid metabolism, while TAK-085 supplementation effectively replenishes this effect in SHRcp rat liver.

Alzheimer’s disease (AD) is the most prominent dementia-related disease and characterized by the presence of insoluble amyloid beta peptide (Aβ) fibers in or around the brain neurons of the affected person. Therefore, agent(s) capable of inhibiting brain amyloid deposition might delay the occurrence or retard the progress forwards of AD and related neurobehavioral symptoms. Here, we report whether, chronic oral administration of Syzygium cumini (locally known as Jam)-seed extract exerts protection against the progressive cognitive decline in the Aβ1-40-infused AD model rats. After 12 weeks of feeding with S. cumini seed extract (at 300 mg/kg BW), we evaluated the learning-related memory of the rats by 8-arm radial maze task, where we determined two types of memory errors, namely reference memory errors (RMEs) and working memory errors (WMEs). After completion of memory tests, rats were sacrificed and the levels of lipid peroxide (LPO), the Aβ1-40-burden, Aβ1-40-oligomers, proinflammatory TNFα, brain derived neurotrophic factor (BDNF), Tyrosine-kinase B (TrkB), postsynaptic-density protein 95 (PSD-95) and Synapse-associated protein (SNAP-25) were determined in the corticohippocampal tissues of the brain. In addition, in vitro antioxidative effects of S. cumini seed extract were evaluated. The oral administration of S. cumini extract significantly increased the memory-related learning ability of the AD model rats, concomitantly with reductions in the levels of corticohippocampal Aβ1-40-burden and Aβ1-40-oligomers. Furthermore, the extract suppressed the levels of TNFα and LPO in the corticohippocampal tissues of the AD rats and also the later in the plasma, suggesting an anti-oxidative and anti-inflammatory activities of the S. cumini extract in the brains of AD model rats. S. cumini extract also increased the levels of brain cognition and memory-related proteins, including BDNF, TrKB, PSD-95 and SNAP-25. We thus suggest that S. cumini-seed extract could be used in neurobehavioral deficits and associated pathogenesis of Alzheimer’s disease.

We examined the effects of the administration of docosahexaenoic acid (DHA)-enriched meals on cognitive function in the oldest elderly with cognitive impairment, such as dementia, living in nursing homes, and on the improvement in caregiver burden at aging agencies.Participants in elderly care facilities and nursing homes (n = 75; 88.5 ± 0.6 years) were randomized in active and placebo groups. The active group had family-style meals containing an additional 1720 mg of docosahexaenoic acid per day for 12 months. At baseline, and after 6 and 12 months of intervention, cognitive function was assessed using Hasegawa's Dementia Scale-Revised and the Mini-Mental State Examination; mental health condition was assessed with the Apathy scale and the Zung Self-Rating Depression Scale; caregiver burden was evaluated using Zarit Burden Interview scores; and participants' serum biochemical factors were measured.The participants were suggested to have dementia. After 12 months, the mean change in Mini-Mental State Examination subitem "Registration" score from baseline to month 12 showed a tendency to be greater in the active group than that in the placebo group. Mean changes in the Apathy scale from baseline to month 12 were less, and the changes in the Zung Self-Rating Depression Scale and the total Zarit Burden Interview scores showed a tendency to be lower in the active group than in the placebo group, respectively.These results suggest that docosahexaenoic acid-enriched meals protect against age-related cognitive decline, and also improve apathy and caregiver burden for the oldest-elderly Japanese with cognitive impairment, such as dementia. Docosahexaenoic acid (DHA, C22:6, ω-3), an ω-3 polyunsaturated fatty acid (PUFA), is critical for brain growth, development, and cognitive ability. It is consumed by offspring via milk during lactation. However, the toxic heavy metal lead (Pb) readily passes into the mammary glands of mother animals and then to offspring through milk. Here, we investigated whether DHA composition of milk and mammary gland tissues is affected by Pb exposure. Mother rats were exposed to Pb via drinking water (0.1%). The fatty acid profile and levels of reduced glutathione (GSH), lipid peroxide (LPO), and pro-inflammatory TNF-α in milk and mammary tissues were measured. Levels of DHA and antioxidant GSH decreased (P < 0.05), while LPO and TNF-α levels increased (P < 0.05) both in milk and mammary tissues. Our results suggest that toxic Pb exposure can upset the level of milk DHA, which may affect brain growth and development, and hence cognitive ability in adulthood and later life.
