Imidacloprid (IMI) is used extensively as an insecticide and poses a significant risk to both the ecological environment and human health. Biological methods are currently gaining recognition among the different strategies tested for wastewater treatment. This study focused on evaluating a recently discovered green alga, Scenedesmus sp. TXH202001, isolated from a municipal wastewater treatment plant (WWTP), exhibited notable capacity for IMI removal. After an 18-day evaluation, medium IMI concentrations (50 and 100 mg/L) facilitated the growth of microalgae whereas low (5 and 20 mg/L) and high (150 mg/L) concentrations had no discernible impact. No statistically significant disparities were detected in Fv/Fm, Malonaldehyde or Superoxide dismutase across all concentrations, suggesting Scenedesmus sp. TXH202001 exhibited notable resilience and adaptability to IMI conditions. Most notably, Scenedesmus sp. TXH202001 successfully eliminated > 99 % of IMI within 18 days subjected to IMI concentrations as high as 150 mg/L, which was contingent on the environmental factor of illumination. Molecular docking was used to identify the chemical reaction sites between IMI and typical degrading enzyme CYP450. Furthermore, the study revealed that the primary path for IMI removal was biodegradation and verified that the toxicity of the degraded product was lower than parent IMI in Caenorhabditis elegans. Even though microplastics (MPs) and graphene nanomaterials (GNMs) have demonstrated individual toxicity towards aquatic organisms, the knowledge gap lies in the lack of understanding regarding their combined toxicity. The difference between the combined toxicity of MPs and GNMs, in contrast to their individual toxicities, and furthermore, the elucidation of the mechanism of this combined toxicity are scientific questions that remain to be addressed. In this study, we examined the individual and combined toxicity of three polystyrene microplastics (MPs) with different functional groups-unmodified, carboxyl-modified (COOH-), and amino-modified (NH 2 -) MPs-in combination with reduced graphene oxide (RGO) on the freshwater microalga Scenedesmus obliquus. More importantly, we explored the cellular and molecular mechanisms responsible for the observed toxicity. The results indicated that the growth inhibition toxicity of RGO, either alone or in combination with the three MPs, against S. obliquus increased gradually with higher particle concentrations. The mitigating effect of MPs-NH 2 on RGO-induced toxicity was most significant at a higher concentration, surpassing the effect of unmodified MPs. However, the MPs-COOH did not exhibit a substantial impact on the toxicity of RGO. Unmodified MPs and MPs-COOH aggravated the inhibition effects of RGO on the cell membrane integrity and oxidative stress-related biomarkers. Additionally, MPs-COOH exhibited a stronger inhibition effect on RGO-induced biomarkers compared to unmodified MPs. In contrast, the MPs-NH 2 alleviated the inhibition effect of RGO on the biomarkers. Furthermore, the presence of differently functionalized MPs did not significantly affect RGO-induced oxidative stress and photosynthesis-related gene expression in S. obliquus, indicating a limited ability to modulate RGO genotoxicity at the molecular level. Carbon supplementation strategies still have certain practical application constraints. Zn/Fe-based metal-organic frameworks (MOFs) nanoparticles that which are not toxic to Scenedesmus obliquus were successfully introduced into microalgal solutions to overcome low CO 2 solubility. The maximum specific surface area of MOFs reached 342.94 m 2 ·g -1 at a Zn/Fe molar ratio of 10/1. Under the optimal MOFs concentrations of 2.5 mg·L -1 , the conversion of inorganic carbon increased by 2.6-fold. When S. obliquuswas cultured in a MOFs-modified medium with 1.50 % CO 2 at 25 °C, the CO 2 mass transfer coefficient and mixing time reached 9.01 × 10 -3 min -1 and 55 s, respectively. The maximum chlorophyll-a content, biomass productivity, and CO 2 fixation efficiency reached 32.57 mg·L -1 , 0.240 g·L -1 ·d -1 and 21.6 %, respectively. Enriching CO 2 for ribulose-1,5-bisphosphate carboxylase/oxygenase carboxylation by MOFs may be the key to improving the photosynthetic efficiency of microalgae. Selecting the optimal microalgal strain for carbon capture and biomass production is crucial for ensuring the commercial viability of microalgae-based biorefinery processes. This study aimed to evaluate the impact of varying bicarbonate concentrations on the growth rates, inorganic carbon (IC) utilization, and biochemical composition of three freshwater and two marine microalgal species. Parachlorella kessleri, Vischeria cf. stellata, and Porphyridium purpureum achieved the highest carbon removal efficiency (>85%) and biomass production at 6 g L -1 sodium bicarbonate (NaHCO 3 ), while Phaeodactylum tricornutum showed optimal performance at 1 g L -1 NaHCO 3 . The growth and carbon removal rate of Scenedesmus quadricauda increased with increasing NaHCO 3 concentrations, although its highest carbon removal efficiency (∼70%) was lower than the other species. Varying NaHCO 3 levels significantly impacted the biochemical composition of P. kessleri, S. quadricauda, and P. purpureum but did not affect the composition of the remaining species. The fatty acid profiles of the microalgae were dominated by C16 and C18 fatty acids, with P. purpureum and P. tricornutum yielding relatively high polyunsaturated fatty acid content ranging between 14% and 30%. Furthermore, bicarbonate concentration had a species-specific effect on the fatty acid and chlorophyll-a content. Improper disposal of municipal solid waste led to the release of heavy metals into the environment through leachate accumulation, causing a range of health and environmental problems. Phycoremediation, using microalgae to remove heavy metals from contaminated water, was investigated as a promising alternative to traditional remediation methods. This study explored the potential of Scenedesmus sp. as a phycoremediation agent for heavy metal removal from landfill leachate. The study was conducted in batch, continuous, and membrane bioreactor (MBR). In the batch system, Scenedesmus sp. was added to the leachate and incubated for 15 days before the biomass was separated from the suspension. In the continuous system, Scenedesmus sp. was cultured in a flow-through system, and the leachate was continuously fed into the system with flow rates measured at 120, 150, and 180 mL/h for 27 days. The MBR system was similar to the continuous system, but it incorporated a membrane filtration step to remove suspended solids from the treated water. The peristaltic pump was calibrated to operate at five different flow rates: 0.24 L/h, 0.30 L/h, 0.36 L/h, 0.42 L/h, and 0.48 L/h for the MBR system and ran for 24 h. The results showed that Scenedesmus sp. was effective in removing heavy metals such as lead (Pb), cobalt (Co), chromium (Cr), nickel (Ni), and zinc (Zn) from landfill leachate in all three systems. The highest removal efficiency was observed for Ni, with a removal of 0.083 mg/L in the MBR and 0.068 mg/L in batch mode. The lowest removal efficiency was observed for Zn, with a removal of 0.032 mg/L in the MBR, 0.027 mg/L in continuous mode, and 0.022 mg/L in batch mode. The findings depicted that the adsorption capacity varied among the studied metal ions, with the highest capacity observed for Ni (II) and the lowest for Zn (II), reflecting differences in metal speciation, surface charge interactions, and affinity for the adsorbent material. These factors influenced the adsorption process and resulted in varying adsorption capacities for different metal ions. The study also evaluated the biomass growth of Scenedesmus sp. and found that it was significantly influenced by the initial metal concentration in the leachate. The aim of the present study was to assess the effect of hydrothermal pretreatment on the solubilization and anaerobic digestion (AD) of Scenedesmus sp. biomass. At first, the microalgae was cultivated in 5% fresh leachate (FL) to recover nutrients such as nitrogen and phosphorus. Scenedesmus sp. grown in 5% FL obtained 100%, 77% and 97% removal efficiency of ammonium nitrogen (NH 4 + - N), total Kjeldahl nitrogen (TKN) and phosphate phosphorous (PO 4 3- -P), respectively. In the following step, the hydrothermal pretreatment of Scenedesmus sp. biomass was carried out at 120, 150 and 170 °C and retention time of 0, 30 and 60 min to evaluate its solubilization and biogas production through AD in batch test. Soluble chemical oxygen demand (sCOD) increased by 260% compared to untreated microalgae at 170 °C for 60 min. In comparison to untreated microalgae, the highest increase in biogas (70%) and methane yield (100%) was observed for 150 °C and 60 min pretreated microalgae as a consequence of hydrothermal pretreatment. Hydrothermal pretreatment has shown effectiveness in enhancing biomass solubilization and increasing biogas yield. Microalgae's superior ability to fix carbon dioxide into biomass and high-value bioproducts remains underutilized in biotechnological applications due to a lack of comprehensive understanding of their carbon metabolism and energy conversion. In this work, the strain improvement technique heavy-ion beams (HIB) mutagenesis was employed on the environmentally adaptable microalgae Scenedesmus quadricauda. After several rounds of screening, two contrasting mutants were identified. S-#4 showed low photosynthetic activity and biomass productivity, while S-#26 exhibited adaptability to prolonged high light stress, achieving a 28.34 % increase in biomass yield compared to the wild-type strain. Integrating their photosynthetic characteristics and comparative proteomic analysis revealed that the contrasting protein regulations from central carbon metabolism mainly affects the two mutants' opposite biomass accumulation. Microalgae are increasingly recognized as promising organisms for bioremediation of organic pollutants. This study investigates the potential of enhancing the bioremediation efficiency of pyrene (PYR), a polycyclic aromatic hydrocarbon (PAH), through NaCl induced physiological and biochemical alterations in two microalgae species, Chlorella vulgaris and Scenedesmus acutus. Our findings reveal significant improvement in PYR removal when these microalgae were cultivated in the presence of 0.1% NaCl where PYR removal increased from 54 to 74% for C. vulgaris and from 26 to 75% for S. acutus. However, it was observed that NaCl induced stress had varying effects on the two species. While C. vulgaris exhibited increased PYR removal, it experienced reduced growth and biomass production, as well as lower photosynthetic efficiency when exposed to PYR and PYR + NaCl. In contrast, S. acutus displayed better growth and biomass accumulation under PYR + NaCl conditions, making it a more efficient candidate for enhancing PYR bioremediation in the presence of NaCl. In addition to assessing growth and biochemical content, we also investigated stress biomarkers, such as lipid peroxidation, polyphenol and proline contents. These findings suggest that S. acutus holds promise as an alternative microalgae species for PYR removal in the presence of NaCl, offering potential advantages in terms of bioremediation efficiency and ecological sustainability. The possibility of obtaining energy or nutritive streams and bioremediation as an add-on opens new perspectives for the massive culturing of microalgal biomass on waste waters generated by the agro-food sector. Ordinary revenue streams are fully preserved, or even boosted, if they are used in microalgal cultivation; however, the suitability of wastewaters depends on multiple nutritional and toxic factors. Here, the effect of modulating the Olive Mill Wastewater (OMW) and cattle digestate (CD) fraction in the formulation of a growth medium on biomass accumulation and productivity of selected biomass fractions and their relevance for biofuel and/or feed production were tested for the microalga Scenedesmus dimorphus and for the cyanobacterium Arthrospira platensis (Spirulina). Scenedesmus strains have been reported to have the potential to tolerate and bioremediate antibiotic pollutants through bioadsorption, bioaccumulation, and biodegradation mechanism from the wastewater medium. Hormesis effects have been observed in the Scenedesmus strains when exposed to different concentrations of antibiotic pollutants. Lower concentrations of antibiotic pollutants are known to trigger growth-stimulating effects by triggering adaptive responses such as increased metabolic activity and activating detoxifying mechanisms leading to the biotransformation pathway. The present review examines the existing body of information pertaining to biotransformation pathways tolerance, hormesis effects, and efficiency of Scenedesmus strains in removing various antibiotic pollutants. The model of thylakoid membrane system (T-M model) (Belyaeva et al. Photosynth Res 2019, 140:1-19) has been improved in order to analyze the induction data for dark-adapted samples of algal (Scenedesmus obliques) and cyanobacterial (Synechocystis sp. PCC 6803) cells. The fluorescence induction (FI) curves of Scenedesmus were measured at light exposures of 5 min, while FI and P700 redox transformations of Synechocystis were recorded in parallel for 100 s intervals. Kinetic data comprising the OJIP-SMT fluorescence induction and OABCDEF P700 + absorbance changes were used to study the processes underlying state transitions qT 2→1 and qT 1→2 associated with the increase/decrease in Chl fluorescence emission. A formula with the Hill kinetics (Ebenhöh et al. Philos Trans R Soc B 2014, 369:20130223) was introduced into the T-M model, with a new variable to imitate the flexible size of antenna Ant M (t) associated with PSII. Simulations revealed that the light-harvesting capacity of PSII increases with a corresponding decrease for that of PSI upon the qT 2→1 transition induced by plastoquinone (PQ) pool oxidation. The complete T-M model fittings were attained on Scenedesmus or Synechocystis fast waves OJIPS of FI, while SMT wave of FI was reproduced at intervals shorter than 5 min. Also the fast P700 redox transitions (OABC) for Synechocystis were fitted exactly. Reasonable sets of algal and cyanobacterial electron/proton transfer (ET/PT) parameters were found. In the case of Scenedesmus, ET/PT traits remained the same irrespective of modeling with or without qT 2→1 transitions. The global energy crisis has spurred a shift from conventional to clean and sustainable energy sources. Biomass derived from microalgae is emerging as an alternative energy source with diverse applications. Despite the numerous advantages of microalgae, large-scale biomass harvesting is not economical and convenient. Self-flocculation is considered an effective phenomenon facilitated by extracting the flocculating substances from microalgae that assist aggregation of algal cells into flocs. A novel cellulose-based bioflocculant has been synthesized from sewage water grown Chlorella sorokiniana and Scenedesmus abundans for harvesting application. The produced bioflocculant amounted to 38.5% and 19.38% of the dry weight of S. abundans and C. sorokiniana, respectively. Analysis via FTIR, XRD, and FESEM-EDX revealed the presence of cellulose hydroxyapatite (HA) in algae-derived cellulose. Harvesting efficiencies of 95.3% and 89.16% were attained for S. abundans and C. sorokiniana, respectively, at a dosage of 0.5 g/L. Furthermore, the bioflocculant was recovered, enabling its reuse with recovery efficiencies of 52% and 10% for S. abundans and C. sorokiniana, respectively. Utilizing monoalgal species for wastewater treatment is facing tremendous challenges owing to changing wastewater complexity in terms of physico-chemical characteristic, nutrient and metal concentration. The environmental conditions are also fluctuating therefore, the formation of robust system is of utmost importance for concomitant sustainable wastewater treatment and bioenergy production. In the present study, the tolerance and adaptability potential of algal consortia-1 (Chlorococcum humicola and Tetradesmus sp.) and consortia-2 (Chlorococcum humicola, Scenedesmus vacuolatus and Tetradesmus sp.) treated with municipal wastewater were examined under natural environmental conditions. The results exhibited that consortia-2 was more competent in recovering nitrate-nitrogen (82.92%), phosphorus (70.47%), and heavy metals (31-73.70%) from municipal wastewater (100%) than consortia-1. The results further depicted that total chlorophyll, carbohydrate, and protein content decreased significantly in wastewater-treated consortia-1 as compared to consortia-2. However, lipid content was increased by 4.01 and 1.17 folds in algal consortia-1 and consortia-2 compared to their respective controls. Moreover, absorption peak at 1740.6 cm -1  reflected higher biofuel-producing potential of consortia-1 as compared to consortia-2 as confirmed through FTIR spectroscopy. The results also revealed that consortia-2 showed the highest photosynthetic performance which was evident from the increment in the active photosystem-II reaction center (1.724 ± 0.068), quantum efficiency (0.633 ± 0.038), and performance index (3.752 ± 0.356). Further, a significant increase in photosynthetic parameters was observed in selected consortia at lag phase, while a noteworthy decline was observed at exponential and stationary phases in consortia-1 than consortia-2. The results also showed the maximum enhancement in ascorbic acid (2.43 folds), proline (3.34 folds), and cysteine (1.29 folds) in consortia-2, while SOD (1.75 folds), catalase (2.64 folds), and GR (1.19 folds) activity in consortia-1. Therefore, it can be concluded that due to remarkable flexibility and photosynthetic performance, consortia-2 could serve as a potential candidate for sustainable nutrient resource recovery and wastewater treatment, while consortia-1 for bio-fuel production in a natural environment. Atmospheric precipitation deposits acid-forming substances into surface water. However, the effects of water-soluble components on microalgae proliferation are poorly understood. This study analysed the growth characteristics of three microalgae bioindicators of water quality: Scenedesmus quadricauda, Chlorella vulgaris, and Scenedesmus obliquus, adopting on-site monitoring, culture experiments simulating 96 types of water by supplementing anions and cations, and predictive modelling. The result quantified pH > 3.0 rain with dominant Ca 2+ , Mg 2+ , and K + cations, together with anions of NO 3 - and SO 4 2- . The presence of Ca 2+ of up to 0.1 mM and Mg 2+ concentrations (>0.5 mM) suppressed Scenedesmus quadricauda growth. Soluble ions, luminosity, and pH had significant impacts (p ≤ 0.01) on increased microalgae proliferation. A newly proposed microalgae growth model predicted a 10.7-fold increase in cell density six days post-incubation in the case of rainfall. The use of algae for industrial, biotechnological, and agricultural purposes is spreading globally. Scenedesmus species can play an essential role in the food industry and agriculture due to their favorable nutrient content and plant-stimulating properties. Previous research and the development of Scenedesmus-based foliar fertilizers raised several questions about the effectiveness of large-scale algal cultivation and the potential effects of algae on associative rhizobacteria. In the microbiological practice applied in agriculture, bacteria from the genus Azospirillum are one of the most studied plant growth-promoting, associative, nitrogen-fixing bacteria. Co-cultivation with Azospirillum species may be a new way of optimizing Scenedesmus culturing, but the functioning of the co-culture system still needs to be fully understood. It is known that Azospirillum brasilense can produce indole-3-acetic acid, which could stimulate algae growth as a plant hormone. However, the effect of microalgae on Azospirillum bacteria is unclear. In this study, we investigated the behavior of Azospirillum brasilense bacteria in the vicinity of Scenedesmus sp. or its supernatant using a microfluidic device consisting of physically separated but chemically coupled microchambers. Following the spatial distribution of bacteria within the device, we detected a positive chemotactic response toward the microalgae culture. To identify the metabolites responsible for this behavior, we tested the chemoeffector potential of citric acid and oxaloacetic acid, which, according to our HPLC analysis, were present in the algae supernatant in 0.074 mg/ml and 0.116 mg/ml concentrations, respectively. Carotenoids are hydrophobic pigments produced exclusively by plants, fungi, and specific microbes. Microalgae are well suited for the production of valuable carotenoids due to their rapid growth, efficient isoprenoid production pathway, and ability to store these compounds within their cells. The possible markets for bio-products range from feed additives in aquaculture and agriculture to pharmaceutical uses. The production of carotenoids in microalgae is affected by several environmental conditions, which can be utilized to enhance productivity. The current study focused on optimizing the extraction parameters (time, temperature, and extraction number) to maximize the yield of carotenoids. Additionally, the impact of various nitrogen sources (ammonia, nitrate, nitrite, and urea) on the production of lutein and loroxanthin in Scenedesmus obliquus was examined. To isolate the carotenoids, 0.20 g of biomass was added to 0.20 g of CaCO 3 and 10.0 mL of ethanol solution containing 0.01% ( w / v ) pyrogallol. Subsequently, the extraction was performed using an ultrasonic bath for a duration of 10 min at a temperature of 30 °C. This was followed by a four-hour saponification process using a 10% methanolic KOH solution. The concentration of lutein and loroxanthin was measured using HPLC-DAD at 446 nm, with a flow rate of 1.0 mL/min using a Waters YMC C 30 Carotenoid column (4.6 × 250 mm, 5 μm). The confirmation of carotenoids after their isolation using preparative chromatography was achieved using liquid chromatography-tandem mass spectrometry (LC-MS/MS) with an atmospheric pressure chemical ionization (APCI) probe and UV-vis spectroscopy. In summary, S. obliquus shows significant promise for the large-scale extraction of lutein and loroxanthin. The findings of this study provide strong support for the application of this technology to other species.

Recovering renewable chemicals from de-fatted microalgal residue derived from lipid extraction within the algal-derived biofuel sector is crucial, given the rising significance of microalgal-derived biodiesel as a potential substitute for petroleum-based liquid fuels. As a circular economy strategy, effective valorization of de-fatted biomass significantly improves the energetic and economic facets of establishing a sustainable algal-derived biofuel industry. In this scenario, this study investigates flash catalytic pyrolysis as a sustainable pathway for valorizing Scenedesmus sp. post-extraction residue (SPR), potentially yielding a bio-oil enriched with upgraded characteristics, especially renewable aromatic hydrocarbons. In the scope of this study, volatile products from catalytic and non-catalytic flash pyrolysis were characterized using a micro-furnace type temperature programmable pyrolyzer coupled with gas chromatographic separation and mass spectrometry detection (Py-GC/MS). Flash pyrolysis of SPR resulted in volatile products with elevated oxygen and nitrogen compounds with concentrations of 46.4% and 26.4%, respectively. In contrast, flash pyrolysis of lyophilized microalgal biomass resulted in lower concentrations of these compounds, with 40.9% oxygen and 17.3% nitrogen. Upgrading volatile pyrolysis products from SPR led to volatile products comprised of only hydrocarbons, while completely removing oxygen and nitrogen-containing compounds. This was achieved by utilizing a low-cost HZSM-5 catalyst within a catalytic bed at 500 °C. Catalytic experiments also indicate the potential conversion of SPR into a bio-oil rich in monocyclic aromatic hydrocarbons, primarily BETX, with toluene comprising over one-third of its composition, thus presenting a sustainable pathway for producing an aromatic hydrocarbon-rich bio-oil derived from SPR. Another significant finding was that 97.8% of the hydrocarbon fraction fell within the gasoline range (C 5 -C 12 ), and 35.5% fell within the jet fuel range (C 8 -C 16 ). This study presents the effect of ultra-violet (UV) light radiation on the process kinetics, metabolic performance, and biodegradation capability of Scenedesmus vacuolatus. The impact of the UV radiation on S. vacuolatus morphology, chlorophyll, carotenoid, carbohydrates, proteins, lipid accumulation, growth rate, substrate affinity and substrate versatility were evaluated. Thereafter, a preliminary biodegradative potential of UV-exposed S. vacuolatus on spent coolant waste (SCW) was carried out based on dehydrogenase activity (DHA) and total petroleum hydrocarbon degradation (TPH). Pronounced structural changes were observed in S. vacuolatus exposed to UV radiation for 24 h compared to the 2, 4, 6, 12 and 48 h UV exposure. Exposure of S. vacuolatus to UV radiation improved cellular chlorophyll (chla = 1.89-fold, chlb = 2.02-fold), carotenoid (1.24-fold), carbohydrates (4.62-fold), proteins (1.44-fold) and lipid accumulations (1.40-fold). In addition, the 24 h UV exposed S. vacuolatus showed a significant increase in substrate affinity (1/Ks) (0.959), specific growth rate (µ) (0.024 h -1 ) and biomass accumulation (0.513 g/L) by 1.50, 2 and 1.9-fold respectively. Moreover, enhanced DHA (55%) and TPH (100%) degradation efficiency were observed in UV-exposed S. vacuolatus. Green microalgae have emerged as beneficial feedstocks for biofuel production. A systems-level understanding of the biochemical network is needed to harness the microalgal metabolic capacity for bioproduction. Genome-scale metabolic modeling (GEM) showed immense potential in rational metabolic engineering, utilizing biochemical flux distribution analysis. Here, we report the first GEM for the green microalga, Scenedesmus obliquus (iAR632), a promising biodiesel feedstock with high lipid-storing capability. iAR632 comprises 1467 reactions, 734 metabolites, and 632 genes distributed among 7 compartments. The model was optimized under three different trophic modes of microalgal cultivation, i.e., autotrophy, mixotrophy, and heterotrophy. The robustness of the reconstructed network was confirmed by analyzing its sensitivity to the biomass components. Pathway-level flux profiles were analyzed, and significant flux space expansion was noticed majorly in reactions associated with lipid biosynthesis. In agreement with the experimental observation, iAR632 predicted about 3.8-fold increased biomass and almost 4-fold higher lipid under mixotrophy than the other trophic modes. Thus, the assessment of the condition-specific metabolic flux distribution of iAR632 suggested that mixotrophy is the preferred cultivation condition for improved microalgal growth and lipid production. Overall, the reconstructed GEM and subsequent analyses will provide a systematic framework for developing model-driven strategies to improve microalgal bioproduction.

Ciprofloxacin (CIP) and norfloxacin (NOR) belong to the organic contaminants of emerging concern (OCECs) that are frequently detected in wastewater matrices at ng/L to mg/L concentrations. This study investigates the potential of Scenedesmus obliquus in the treatment of CIP and NOR as a binary mixture from raw wastewater. Optimization of inoculum was done to find the required cell density concentration that has less inhibition and high removal. The optimum inoculum (cell density: 200 × 10 5  cells/mL and OD 680 : 1.0) has shown 75% removal with no inhibition of growth. A pilot scale study was conducted in controlled environment using high-rate algal pond to investigate the contribution of abiotic and biotic removal. Abiotic removal is negligible in comparison with the biotic contribution of removal. The order of removal efficiency is observed as COD (88%) > NOR (84.8%) > CIP (84.6%) > NH 4 + (71.7%) with biodegradation as the major removal mechanism. Biotransformed products of CIP + NOR were identified inside the Scenedesmus obliquus. During the pilot-scale study, Biomass (3.70 ± 0.07 g/L) was harvested with carbohydrates (17.85 ± 0.1%), lipids (38.36 ± 0.13%), and proteins (28.18 ± 1.63%). Lipid productivity in binary mixture was 2.6 times higher than the lipid production in control condition. Microalgae such as Chlorella pyrenoidosa, Scenedesmus obliquus and Chlorella sorokiniana were cultivated in domestic wastewater for biohydrogen production. The comparison between the microalgae was executed based on biomass productions, biochemical yields and nutrient removal efficiencies. S. obliquus showed the possibility of growing in domestic wastewater reaching maximum biomass production, lipid, protein, carbohydrate yield and nutrient removal efficiency. All the three microalgae reached high biomass production of 0.90, 0.76 and, 0.71 g/L, respectively for S. obliquus, C. sorokiniana and C. pyrenoidosa. A higher protein content (35.76%) was obtained in S. obliquus. A similar pattern of lipid yield (25.34-26.23%) and carbohydrate yield (30.32-33.21%) was recorded in all selected microalgae. Chlorophyll-a content was higher in synthetic media-grown algae compared algae grown in wastewater. The maximum nutrient removal efficiencies achieved were 85.54% of nitrate by C. sorokiniana, 95.43% of nitrite by C. pyrenoidosa, ∼100% of ammonia and 89.34% of phosphorus by C. sorokiniana. An acid pre-treatment was applied to disintegrate the biomass of microalgae, followed by dark fermentation in batch mode to produce hydrogen. During fermentation process, polysaccharides, protein and lipids were consumed. Maximum hydrogen production of 45.50 ± 0.32 mLH 2 /gVS, 38.43 ± 0.42 mLH 2 /gVS and 34.83 ± 1.82 mL/H 2 /gVS was achieved by C. pyrenoidosa, S. obliquus and C. sorokiniana respectively. Microalgae attract interest worldwide due to their potential for several applications. Scenedesmus is one of the first in vitro cultured algae due to their rapid growth and handling easiness. Within this genus, cells exhibit a highly resistant wall and propagate both auto- and heterotrophically. The main goal of the present work is to find scalable ways to produce a highly concentrated biomass of Scenedesmus rubescens in heterotrophic conditions. Scenedesmus rubescens growth was improved at the lab-scale by 3.2-fold (from 4.1 to 13 g/L of dry weight) through medium optimization by response surface methodology. Afterwards, scale-up was evaluated in 7 L stirred-tank reactor under fed-batch operation. Then, the optimized medium resulted in an overall productivity of 8.63 g/L/day and a maximum biomass concentration of 69.5 g/L. S. rubescens protein content achieved approximately 31% of dry weight, similar to the protein content of Chlorella vulgaris in heterotrophy.

Prothioconazole (PTC) is a broad-spectrum triazole fungicide with one asymmetric center and consists of two enantiomers, R -(-)-PTC and S -(+)-PTC. To address the concern of its environmental safety, the enantioselective toxic effects of PTC on Scendesmus obliquus ( S. obliquus ) were investigated. PTC racemates ( Rac -PTC) and enantiomers exhibited dose-dependent acute toxicity effects against S. obliquus at a concentration from 1 to 10 mg·L -1 . The 72 h-EC50 value of Rac -, R -(-)-, and S -(+)-PTC is 8.15, 16.53, and 7.85 mg·L -1 , respectively. The growth ratios and photosynthetic pigment contents of the R -(-)-PTC treatment groups were higher than the Rac - and S -(+)-PTC treatment groups. Both catalase (CAT) activities and esterase activities were inhibited in the Rac - and S -(+)-PTC treatment groups at high concentrations of 5 and 10 mg·L -1 , and the levels of malondialdehyde (MDA) were elevated, which exceeded the levels in algal cells for the R -(-)-PTC treatment groups. PTC could disrupt the cell morphology of S. obliquus and induce cell membrane damage, following the order of S -(+)-PTC ≈ Rac -PTC > R -(-)-PTC. The enantioselective toxic effects of PTC on S. obliquus provide essential information for its ecological risk assessment.

Due to their remarkable properties, the applications of carbon-based nanomaterials (CNMs) such as graphene and functionalized multi-walled carbon nanotubes (f-MWCNTs) are increasing. These CNMs can enter the freshwater environment via numerous routes, potentially exposing various organisms. The current study assesses the effects of graphene, f-MWCNTs, and their binary mixture on the freshwater algal species Scenedesmus obliquus. The concentration for the individual materials was kept at 1 mg L -1 , while graphene and f-MWCNTs were taken at 0.5 mg L -1 each for the combination. Both the CNMs caused a decrease in cell viability, esterase activity, and photosynthetic efficiency in the cells. The cytotoxic effects were accompanied by increased hydroxyl and superoxide radical generation, lipid peroxidation, antioxidant enzyme activity (catalase and superoxide dismutase), and mitochondrial membrane potential. Graphene was more toxic compared to f-MWCNTs. The binary mixture of the pollutants demonstrated a synergistic enhancement of the toxic potential. Oxidative stress generation played a critical role in toxicity responses, as noted by a strong correlation between the physiological parameters and the biomarkers of oxidative stress. Lignocellulose is the most abundant natural biopolymer on earth and a potential raw material for the production of fuels and chemicals. However, only some organisms such as bacteria and fungi produce enzymes that metabolize this polymer. In this work we have demonstrated the presence of cellulolytic activity in the supernatant of Scenedesmus quadricauda cultures and we identified the presence of extracellular cellulases in the genome of five Scenedesmus species. Scenedesmus is a green alga which grows in both freshwater and saltwater regions as well as in soils, showing highly flexible metabolic properties. Sequence comparison of the different identified cellulases with hydrolytic enzymes from other organisms using multisequence alignments and phylogenetic trees showed that these proteins belong to the families of glycosyl hydrolases 1, 5, 9, and 10. In addition, most of the Scenedesmus cellulases showed greater sequence similarity with those from invertebrates, fungi, bacteria, and other microalgae than with the plant homologs. Furthermore, the data obtained from the three dimensional structure showed that both, their global structure and the main amino acid residues involved in catalysis and substrate binding are well conserved. During the COVID-19 pandemic billions of face masks were used since they became a necessity in everyone's lives. But these were not disposed properly and serve as one of the most significant sources of micro and nano plastics in the environment. The effects of mask leached plastics in aquatic biota remains largely unexplored. In this work, we quantified and characterized the released microplastics from the three layers of the mask. The outer layer of the face mask released more microplastics i.e., polypropylene than middle and inner layers. We investigated and compared the acute toxic effects of the released microplastics between Scenedesmus obliquus and Chlorella sp. The results showed a decrease in cell viability, photosynthetic yield, and electron transport rate in both the algal species. This was accompanied by an increase in oxidative stress markers such reactive oxygen species (ROS) and malondialdehyde (MDA) content. There was also a significant rise of antioxidant enzymes such as superoxide dismutase (SOD) and catalase (CAT) in both the algal cells. Furthermore, morphological changes like cell aggregation and surface chemical changes in the algae were ascertained by optical microscopy and FTIR spectroscopy techniques, respectively. The tests confirmed that Scenedesmus obliquus was more sensitive than Chlorella sp. to the mask leachates. Our study clearly revealed serious environmental risk posed by the released microplastics from surgical face masks. This study aims at evaluating an innovative biotechnological process for the concomitant bioremediation and valorization of wastewater from textile digital printing technology based on a microalgae/bacteria consortium. Nutrient and colour removal were assessed in lab-scale batch and continuous experiments and the produced algae/bacteria biomass was characterized for pigment content and biomethane potential. Microbial community analysis provided insight of the complex community structure responsible for the bioremediation action. Specifically, a community dominated by Scenedesmus spp. and xenobiotic and dye degrading bacteria was naturally selected in continuous photobioreactors. Data confirm the ability of the microalgae/bacteria consortium to grow in textile wastewater while reducing the nutrient content and colour. Improvement strategies were eventually identified to foster biomass growth and process performances. Neonicotinoids, as the most widely used pesticides in the world, help improve the production of crops. Meanwhile, it also brings potential threats to surrounding environments and other organisms because of its wide use and even abuse. In this study, Scenedesmus sp. TXH isolated from a wastewater treatment plant was used to remove the neonicotinoid pesticide thiamethoxam (THIA). The removal efficiency, degradation pathway, metabolite fate of THIA and physicochemical effects on microalgae cells were studied. Meanwhile, the feasibility of using microalgal technology to remove THIA from municipal wastewater was also explored. The results showed that 5-40 mg/L of THIA slightly promoted the growth of microalgae, while 60 mg/L THIA severely inhibited microalgal growth. It was observed that malondialdehyde content and superoxide dismutase activity in 60 mg/L THIA group increased significantly (p < 0.05) in the early stage of the experiment, indicating that THIA caused oxidative damage to microalgae. Scenedesmus sp. TXH showed high-efficient degradation ability and high resistance to THIA, with 100% removal of THIA at 5, 20 and 40 mg/L groups and 97.5% removal of THIA at 60 mg/L group on day 12. THIA was mainly removed by biodegradation, accounting for 78.18%, 93.50%, 96.81% and 91.35% under 5, 20, 40 and 60 mg/L on day 12, respectively. Six degradation products were identified, and four potential degradation pathways were proposed. In practical wastewater, the removal efficiency of total dissolved nitrogen, total dissolved phosphorus, ammonia nitrogen and THIA reached 85.68%, 90.00%, 98.43% and 100%, respectively, indicating that Scenedesmus sp. Both Bisphenol A (BPA) and polystyrene nanoplastics (PSNPs) are routinely found in several consumer products such as packaging materials, flame retardants, and cosmetics. The environment is seriously endangered by nano- and microplastics. In addition to harming aquatic life, nanoplastics (NPs) also bind to other pollutants, facilitating their dispersion in the environment and possibly promoting toxicity induced by these pollutants. The toxic effects of polystyrene nanoplastics (PS-NPs) and BPA were examined in this study, as well as the combined toxic impacts of these substances on the freshwater microalgae Scenedesmus obliquus. In addition, the exopolymeric substances (EPS) secreted by algae will interact with the pollutants modifying their physicochemical behaviour and fate. This work aimed to investigate how algal EPS alters the combined effects of BPA and PSNPs on the microalgae Scenedesmus obliquus. The algae were exposed to binary mixtures of BPA (2.5, 5, and 10 mg/L) and PSNPs (1 mg/L of plain, aminated, and carboxylated PSNPs) with EPS added to the natural freshwater medium. Cell viability, hydroxyl and superoxide radical generation, cell membrane permeability, antioxidant enzyme activity (catalase and superoxide dismutase), and photosynthetic pigment content were among the parameters studied to determine the toxicity. It was observed that for all the binary mixtures, the carboxylated PSNPs were most toxic when compared to the toxicity induced by the other PSNP particles investigated. The maximum damage was observed for the mixture of 10 mg/L of BPA with carboxylated PSNPs with a cell viability of 49%. When compared to the pristine mixtures, the EPS-containing mixtures induced significantly reduced toxic effects. A considerable decrease in reactive oxygen species levels, activity of antioxidant enzymes (SOD and CAT), and cell membrane damage was noted in the EPS-containing mixtures. The contamination of the aquatic environment with microplastics has become a global environmental concern. Microplastic particles can be shredded to form smaller nanoplastics, and knowledge on their impacts on phytoplankton, especially freshwater microalgae, is still limited. To investigate this issue, the microalga Scenedesmus quadricauda was exposed to polystyrene nanoplastics (PS-NPs) of five concentrations (10, 25, 50, 100, and 200 mg/L). The growth; the contents of antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD); the chlorophyll content; and concentrations of soluble protein and soluble polysaccharide were accordingly measured. The results showed that the microalgal density increased with the increase of the polystyrene nanoplastic concentrations, and the physiological features of alga were enhanced after the stimulation of nanoplastics. Furthermore, a high concentration (200 mg/L) of nanoplastics increased the contents of chlorophyll, soluble protein, and polysaccharide (P < 0.05). The antioxidant enzyme activities of Scenedesmus quadricauda were significantly activated by nanoplastics. Lastly, we propose three possible algal recovery mechanisms in response to nanoplastics in which Scenedesmus quadricauda was tolerant with PS-NPs by cell wall thickening, internalization, and aggregation. In this study, an efficient microalgal strain SD07 was isolated from pond wastewater and identified as Scenedesmus sp. using the 18S rRNA gene sequence analysis. The strain SD07 was grown in a variety of concentrations (25-100%) of municipal wastewater. Scenedesmus sp. strain SD07 grown in 75% diluted wastewater produced a higher amount of biomass (1.93 ± 0.10 g L -1 ), and removal of chemical oxygen demand (COD), ammonium (NH 4 + ), total nitrogen (TN) and total phosphate (TP) by 91.36%, 88.41%, 93.26% and 96.32%, respectively from wastewater. The harvested strain SD07 biomass has protein, carbohydrate and lipid contents of 35%, 20.4% and 33%, respectively. Fatty acid profiles revealed that the strain SD07 lipids mainly consist of palmitic acid (40.5%), palmitoleic acid (19%), linoleic acid (17%) and oleic acid (13.2%). Furthermore, strain SD07 cultured in 75% diluted wastewater produced 378 mg L -1 of exopolysaccharides (EPS). The EPS was utilized as a biostimulant in the cultivation of Solanum lycopersicum under salinity stress. Bioethanol production from algal biomass is a promising alternative for sustainable biofuel production. Algae possess a high photosynthetic capacity and an adaptive ability to thrive under harsh environmental conditions. The potential properties of Scenedesmus acuminatus CCALA 436 were assessed in this research for its bioethanol efficiency, and the effects of growing the algae in wastewater and at different concentrations of mepiquat chloride were studied. Also, pre-treatment efficiencies of different concentrations of calcium oxide were carried out on microalgae biomass. Superoxide dismutase, catalase activity, glutathione, and malondialdehyde contents of microalgae were examined, and the changes in chlorophyll, photoprotective carotenoid contents, and protein concentrations were determined. The results revealed that the maximum sugar and ethanol contents of Scenedesmus acuminatus CCALA 436 were 44.7 ± 1.5% and 20.32 g/L, respectively, for 50% wastewater and mepiquat chloride (2.5 mg/L) after pre-treatment with calcium oxide (0.08%). Additionally, the levels of oxidative enzymes varied depending on the wastewater concentrations. Certain microalgal species can grow under high CO 2 concentrations providing potential for mitigating CO 2 pollution in flue gas produced by power plants. Microalga Scenedesmus obliquus strain HTB1 was isolated from the Chesapeake Bay and grow rapidly in a high level of CO 2 . However, little is known about the molecular responses of HTB1 to high CO 2 levels. Here, we investigated how HTB1 responds to 10% CO 2 using the time-resolved transcriptome analysis. Gene expression profiles indicated that HTB1 responds quickly (in 2 h) and becomes adaptive within 12 h when exposed to 10% CO 2 . Interestingly, certain genes of light-harvesting, chlorophyll synthesis and carbon fixation (i.e. rbcS) were up-regulated at 10% CO 2 , and these functional responses are consistent with the increased photosynthesis efficiency and algal biomass under 10% CO 2 . Nitrate assimilation was strongly enhanced, with amino acid biosynthesis and aminoacyl tRNA biosynthesis genes being markedly up-regulated, indicating that HTB1 actively takes up nitrogen and accelerates protein synthesis at 10% CO 2 . Carbon metabolism including fatty acid biosynthesis and TCA cycle was enhanced at 10% CO 2 , supporting the earlier observation of increased lipid content of Scenedesmus sp. under high CO 2 levels. Interestingly, key genes like RuBisCO (rbcL) and carbonic anhydrase in carboxysomes did not respond actively to 10% CO 2 , implying that exposure to 10% CO 2 has little impact on the carbon concentrating mechanisms and CO 2 fixation of the Calvin cycle. Lutein is a naturally potent antioxidant carotenoid synthesized in green microalgae with a potent ability to prevent different human chronic conditions. To date, there are no reports of the immune-stimulating effect of pure lutein isolated from Scenedesmus obliquus. Thus, we isolated the natural lutein from S. obliquus and evaluated its effectiveness as an immunostimulant against cyclophosphamide-induced brain injury. We purified all-E-(3R, 3'R, 6'R)-Lutein from S. obliquus using prep-HPLC and characterized it by 1 H- and 13 C-NMR spectroscopy. We assigned rats randomly to four experimental groups: the Control group got a vehicle for lutein dimethyl sulfoxide for ten successive days. The Cyclophosphamide group received a single i.p injection of Cyclophosphamide (200 mg/kg). Lutein groups received 50 and 100 (mg/kg) of lutein one time per day for ten successive days after the cyclophosphamide dose. Lutein administration reduced brain contents of Macrophage inflammatory protein2 (MIP2), cytokine-induced- neutrophil chemoattractant (CINC), and Matrix metalloproteinase 1 (MMP1). Besides, it lowered the contents of interleukin 1 beta (IL-1β) and interleukin 18 (IL-18), associated with low content of NLR pyrin domain protein 3 (NLRP3) and consequently caspase-1 compared to the cyclophosphamide group. In the histomorphometric analysis, lutein groups (50 and 100 mg/Kg) showed mild histopathological alterations as they significantly reduced nuclear pyknosis numbers by 65% and 69% respectively, compared to the cyclophosphamide group. This is the first study that showed the immunomodulatory roles of lutein against cyclophosphamide-induced brain injury via decreasing neuroinflammation, chemokines recruitment, and neuron degeneration with the modulation of immune markers. Nanoparticles are inevitable byproducts of modern industry. However, the environmental impacts arising from industrial applications of nanoparticles are largely under-reported. This study evaluated the ecotoxicological effects of aluminum oxide nanoparticles (Al 2 O 3 NP) and its influence on sulfacetamide (SA) biodegradation by a freshwater microalga, Scenedesmus obliquus. Although Al 2 O 3 NP showed limited toxicity effect on S. obliquus, we observed the toxicity attenuation aspect of Al 2 O 3 NP in a mixture of sulfacetamide on microalgae. The addition of 100 mg L -1 of Al 2 O 3 NP and 1 mg L -1 of SA reduced total chlorophyll by 23.3% and carotenoids by 21.6% in microalgal compared to control. The gene expression study demonstrated that ATPF0C, Lhcb1, HydA, and psbA genes responsible for ATP synthesis and the photosynthetic system were significantly downregulated, while the Tas gene, which plays a major role in biodegradation of organic xenobiotic chemicals, was significantly upregulated at 1 and 100 mg L -1 of Al 2 O 3 NP. The S. obliquus removed 16.8% of SA at 15 mg L -1 in 14 days. However, the removal was slightly enhanced (18.8%) at same concentration of SA in the presence of 50 mg L -1 Al 2 O 3 NP. This result proves the stability of sulfacetamide biodegradation capacity of S. obliquus in the presence of Al 2 O 3 NP co-contamination. The metabolic analysis showed that SA was degraded into simpler byproducts such as sulfacarbamide, sulfaguanidine, sulfanilamide, 4-(methyl sulfonyl)aniline, and N-hydroxy-benzenamine which have lower ecotoxicity than SA, demonstrating that the ecotoxicity of sulfacetamide has significantly decreased after the microalgal degradation, suggesting the environmental feasibility of microalgae-mediated wastewater technology. Chlorella vulgaris (C. vulgaris) and Scenedesmus obliquus (S. obliquus) were compared to remove toxicity under conditions of sludge extract cultivation for 30 days. The toxicity of sludge extract, the growth characteristics, photosynthetic pigment, superoxide dismutase (SOD) enzyme and catalase (CAT) enzyme activities of the two microalgae were studied by contrast. The results showed that small molecular organic matter (<500 Da) was more easily utilized by microalgae. The toxicity in the toxic group of C. vulgaris and S. obliquus on the 30 th day decreased to 56.8 ± 1.2% and 60.7 ± 2.8%, respectively. In the toxic group, the maximal SOD enzyme activity of C. vulgaris and S. obliquus were 2.02 U/mg proteins and 8.21 U/mg proteins, respectively, demonstrating that toxicity caused more oxidative damage to S. obliquus than to C. vulgaris. On the basis of determining isoelectric point of algae residue protein obtained from Scenedesmus dimorphus, this study investigated the effects of pH values, ratio of liquid to solid, extraction temperature and time on protein extraction rates, and assessed the nutritional value of protein extracted from microalgae residues. The results from orthogonal experiments revealed the optimum conditions for extracting proteins from algal residues (pH: 12; liquid-to-solid ratio: 40 mL/g; extraction temperature: 45 ℃; extraction time: 140 min). It was observed that under the optimal conditions, the protein extraction rate was 40.13%. Essential amino acids account for 44.3% of the proteins obtained from algal residues. The content of anti-nutritional factors in algal residues was significantly reduced, and the digestibility of algal residue proteins was higher than the digestibility of algal powder. Microalgae are photosynthetic microorganisms that stand out from conventional food sources and ingredients due to their high growth rate and adaptability. In addition to being highly sustainable, significant concentrations of proteins, lipids, and pigments accumulate in their cell structures from photosynthesis. Hence, this study sought to evaluate the food potential of Scenedesmus obliquus biomasses obtained from photosynthetic cultures enriched with 3, 5, 10, 15, 20, and 25% carbon dioxide (CO 2 ) (v/v). Cultivations with 3, 5, and 10% CO 2 showed greater amino acids and proteins synthesis; the protein content reached values above 56% of the dry biomass and high protein quality, due to the presence of most essential amino acids at recommended levels for the human diet. The highest concentrations of chlorophylls were found in cultures with 15, 20, and 25% CO 2 (24.2, 23.1 and 30.8 mg g -1 , respectively), although the profiles showed higher percentages of degradation compounds. Carotenoid concentrations were three times higher in cultures with 3, 5, and 10% CO 2 (25.3, 22.7 and 18.1 mg g -1 , respectively) and all-trans-β-carotene was the major compound. Lipid synthesis was intensified at higher CO 2 enrichment; the percentages obtained were 14.8% of lipids in the culture with 15% CO 2 , 15.0% with 20% CO 2 , and 13.7% with 25% CO 2 . In addition, greater polyunsaturated fatty acids accumulation and a significant reduction in the n6/n3 ratio were also observed at the highest CO 2 concentrations. Algal biodiesel has been becoming a focus in the field of bioenergy worldwide. In this study, effects of heterogeneous sulfur (SO 4 2- , SO 3 2- and S 2- ) on growth and lipid accumulation of Scenedesmus obliquus cultured in wastewater with a C/N ratio of 30 were investigated, respectively. The results shown that SO 4 2- , the optimal sulfur source, could trigger cell growth in a concentration-dependent manner. However, SO 3 2- was superior to the others in boosting carbon uptake of cells, which was subject to NH 4 + -N concentration. Only SO 4 2- could simultaneously increase lipid content and productivity of cells with a dominant component of oleic acid (C18:1n9c) occupying approximately 40% in fatty acid profile. Additionally, the genes encoding enzymes such as CDIPT, ADPRM, DPP1, pmtA and BTA1 involved in the uppermost lipid-related pathway (glycerophospholipid metabolism) were identified facing different sulfur source regardless of the concentration changes. Comprehending the effects of synthetic nanomaterials on natural microorganisms is critical for the development of emerging nanotechnologies. Compared to artificial inactivation of microbes, the up-regulation of biological functions should be more attractive due to the possibility of discovering unexpected properties. Herein, a nanoengineering strategy was employed to tailor g-C 3 N 4 for the metabolic regulation of algae. We found that surface protonated g-C 3 N 4 (P-C 3 N 4 ) as a nanopolymeric elicitor enabled the reinforced biological activity of Microcystis aeruginosa and Scenedesmus for harmful substances removal. Metabolomics analysis suggested that synthetic nanoarchitectures induced moderate oxidative stress of algae, with up-regulated biosynthesis of extracellular polymeric substances (EPS) for resisting the physiological damage caused by toxic substances in water. The formation of oxidative . O 2 - contributed to over five-fold enhancement in the biodecomposition of harmful aniline. The interactions of microalgae and their associated microbiomes have come to the fore of applied phycological research in recent years. However, the functional mechanisms of microalgal interactions remain largely unknown. Here, we examine functional protein patterns of the microalgae Scenedesmus vacuolatus and its associated bacterial community during algal infection by the endoparasite Amoeboaphelidium protococcarum. We performed metaproteomics analyses of non-infected (NI) and aphelid-infected (AI) S. vacuolatus cultures to investigate underlying functional and physiological changes under infectious conditions. We observed an increase in bacterial protein abundance as well as a severe shift of bacterial functional patterns throughout aphelid-infection in comparison to NI treatment. Most of the bacterial proteins (about 55%) upregulated in AI were linked to metabolism and transport of amino acids, lipids, coenzymes, nucleotides and carbohydrates and to energy production. Several proteins associated with pathogenic bacterial-plant interactions showed higher protein abundance levels in AI treatment. These functional shifts indicate that associated bacteria involved in commensalistic or mutualistic interactions in NI switch to opportunistic lifestyles and facilitate pathogenic or saprotrophic traits in AI treatment. Glufosinate-ammonium, a widely used chiral herbicide, has become the focus of attention because of its toxicity toward non-target organisms and its degradation behavior in the environment. With the introduction of L-glufosinate-ammonium products, the toxicity and environmental behavior of rac-glufosinate-ammonium and L-glufosinate-ammonium have become the subject of increasing interest. The overall goal of this study was to investigate the differences in toxicity and biodegradation of rac-glufosinate-ammonium and L-glufosinate-ammonium in an aquatic organism, Scenedesmus obliquus. The toxicity of rac-glufosinate-ammonium and L-glufosinate-ammonium to S. obliquus was compared by measuring EC 50 , malondialdehyde (MDA) content, protein content and antioxidant enzyme activity. The 96-h EC 50 values of rac-glufosinate-ammonium and L-glufosinate-ammonium were 57.22 μg/mL and 25.55 μg/mL, respectively, which indicated that L-glufosinate-ammonium was more toxic to S. obliquus than rac-glufosinate-ammonium. Based on the MDA content, protein content, and antioxidant enzyme (SOD and CAT) activity, we found that L-glufosinate-ammonium could cause more serious oxidative damage than rac-glufosinate-ammonium. The residual amount of glufosinate-ammonium and its metabolites in the culture medium and S. obliquus were determined by HPLC-HRMS. Comparison of glufosinate-ammonium concentrations in algae-free and algae-containing media, showed that glufosinate-ammonium degradation in the S. obliquus system was significantly increased, and the degradation rate of L-glufosinate-ammonium was faster than that of D-glufosinate-ammonium. No enantiomerization was observed for pure L-glufosinate-ammonium treatment. Attachment effects of six carrier materials on the cultivation of high-value microalga Scenedesmus sp. LX1 in diluted swine wastewater were investigated. The results showed that when the initial algal densities were 5×10 5 cells/mL and 1×10 6 cells/mL in sterilized wastewater with 20-fold dilution, the biomass of microalgae attaching to geotextile was the highest. The contact angle and surface structure of the material affected the attachment of microalgae. Further, among the four dilutions, the highest attached biomass on geotextile was 414.47 mg/L in the sterilized wastewater at 20-fold, but the pollutants removal rate and attached biomass were higher in the non-sterile wastewater in the other three dilutions (original wastewater, 5-fold, 10-fold). Next, the microalgae were able to remove pollutants with the highest removal rates of COD, TN, NH 4 + -N and TP reaching to 86.92%, 60.75%, 71.81% and 96.13%. Small-scale photobioreactors (PBRs) in the inoculum stage were designed with internal (red or green) and external white LED light as an initial step of a larger-scale installation aimed at fulfilling the integral biorefinery concept for maximum utilization of microalgal biomass in a multifunctional laboratory. The specific growth rate of Scenedesmus obliquus (Turpin) Kützing biomass for given cultural conditions was analyzed by using MAPLE software. For the determination of total polyphenols, flavonoids, chlorophyll "a" and "b", carotenoids and lipids, UHPLC-HRMS, ISO-20776/1, ISO-10993-5 and CUPRAC tests were carried out. Under red light growing, a higher content of polyphenols was found, while the green light favoured the flavonoid accumulation in the biomass. Chlorophylls, carotenoids and lipids were in the same order of magnitude in both samples. The dichloromethane extracts obtained from the biomass of each PBR synergistically potentiated at low concentrations (0.01-0.05 mg/mL) the antibacterial activity of penicillin, fluoroquinolones or oregano essential oil against the selected food-borne pathogens ( Staphylococcus aureus , Escherichia coli and Salmonella typhimurium ) without showing any in vitro cytotoxicity. Both extracts exhibited good cupric ion-reducing antioxidant capacity at concentrations above 0.042-0.08 mg/mL. The UHPLC-HRMS analysis revealed that both extracts contained long chain fatty acids and carotenoids thus explaining their antibacterial and antioxidant potential. The applied engineering approach showed a great potential to modify microalgae metabolism for the synthesis of target compounds by S. obliquus with capacity for the development of health-promoting nutraceuticals for poultry farming.

Cerium is the most abundant rare earth metal in the earth's crust, and it has deleterious effects on aquatic ecosystems from fertilizer runoff. Scenedesmus obliquus is an oil-rich microalga that grows rapidly and is sensitive to many kinds of toxins. Given that microalgae are useful indicators of eutrophication and toxic stress, it was found that lower concentrations of cerium (0.50-5.00 mg·L -1 ) stimulated algal growth and increased chlorophyll a content, whereas higher concentrations (above 50.00 mg·L -1 ) had an inhibitory effect on algal growth and chlorophyll a content. The algal growth rate and chlorophyll a content peaked at a cerium concentration of 5.00 mg·L -1 . Both the donor and acceptor sides of photosystem II (PSII) reaction centers were sensitive to cerium-induced stress. Specifically, high concentrations of cerium damaged the oxygen evolving complex and PSII reaction center and suppressed electron transport at the donor and receptor side of the reaction center, influencing the absorption, transfer, and application of light energy in S. obliquus XJ002. In addition, we established a simple method to quantify the intracellular lipid content of S. obliquus XJ002, and the optimum staining conditions for Nile red were as follows: volume percentage of dimethyl sulfoxide was 2%, the concentration of Nile red was 2.0 µg·mL -1 , and the staining time of Nile red was 5 min. The addition of cerium resulted in a significant increase in the total lipid content of XJ002. When the concentration of cerium was 50 mg·L -1 , the total lipid content was 16.26% higher than the control group. Background: The three-phase extraction process of olive oil produces highly contaminated wastewater (OMW). The elimination of this toxic by-product is an important environmental issue that requires the development of an appropriate management solution. The cultivation of microalgae using OMW as growth medium was therefore studied using single (the culture medium was formed by 0% to 80% ultrafiltered olive mill wastewater (OMUF) or OMW added to BG11) and two-stage strategies (microalgae were firstly cultivated in the BG11 medium. In the second stage, 40% and 80% of OMUF and OMW were added to the culture). In this work, biodegradation of OMW and subsequent extraction of lipid and antioxidant molecules was investigated as an ecofriendly method for the bioremediation and valorization of OMW., Results: For two-stage cultivation, OMUF and OMW stress enhanced the intracellular amount of polyphenol accumulated in Scenedesmus sp. and exhibited the highest 2, 2-diphenyl-1- picrylhydrazyl radical (DPPH) and 2,2'-azino-bis (3-ethylbenzoline-6-sulfonate) radical (ABTS) scavenging ability compared with single-stage cultivation. Moreover, the lipid profile is dominated by polyunsaturated acids. In the single-stage cultivation, the Ch a, Ch b, carotenoid, carbohydrate and lipid content of 2.57, 7.4, 1.69, 368, and 644 g kg -1 were observed in 40% OMUF added culture, respectively, along with high biomass productivity and 58% of polyphenol removal. Moreover, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay showed that the biomass of Scenedesmus sp. cultured on 40% OMUF did not show any toxic effect, making it an efficient strategy., Conclusion: The results indicate that Scenedesmus sp. is a promising microalga for the biotreatment of OMW and the extraction of bioactive metabolites. © 2021 Society of Chemical Industry., (© 2021 Society of Chemical Industry.)

Photooxidative stress-inducible water-soluble astaxanthin-binding proteins, designated as AstaP, were identified in two Scenedesmaceae strains, Coelastrella astaxanthina Ki-4 and Scenedesmus obtusus Oki-4N; both strains were isolated under high light conditions. These AstaPs are classified as a novel family of carotenoprotein and are useful for providing valuable astaxanthin in water-soluble form; however, the distribution of AstaP orthologs in other microalgae remains unknown. Here, we examined the distribution of AstaP orthologs in the family Scenedesmaceae with two model microalgae, Chlamydomonas reinhardtii and Chlorella variabilis . The expression of AstaP orthologs under photooxidative stress conditions was detected in cell extracts of Scenedesmaceae strains, but not in model algal strains. Aqueous orange proteins produced by Scenedesmaceae strains were shown to bind astaxanthin. The protein from Scenedesmus costatus SAG 46.88 was purified. It was named ScosAstaP and found to bind astaxanthin. The deduced amino acid sequence from a gene encoding ScosAstaP showed 62% identity to Ki-4 AstaP. The expression of the genes encoding AstaP orthologs was shown to be inducible under photooxidative stress conditions; however, the production amounts of AstaP orthologs were estimated to be approximately 5 to 10 times lower than that of Ki-4 and Oki-4N.

Microalgae have received significant attention as potential next-generation microbiologic cell factories for biofuels. However, the production of microalgal biofuels is not yet sufficiently cost-effective for commercial applications. To screen higher lipid-producing strains, heavy carbon ion beams are applied to induce a genetic mutant. An RNA-seq technology is used to identify the pathways and genes of importance related to photosynthesis and biofuel production. The deep elucidation of photosynthesis and the fatty acid metabolism pathway involved in lipid yield is valuable information for further optimization studies. This study provided the photosynthetic efficiency and transcriptome profiling of a unicellular microalgae, Scenedesmus obliqnus mutant SO120G, with enhanced lipid production induced by heavy carbon ion beams. The lipid yield (52.5 mg L -1 ) of SO120G mutant were enhanced 2.4 fold compared with that of the wild strain under the nitrogen deficient condition. In addition, the biomass and growth rate were 57% and 25% higher, respectively, in SO120G than in the wild type, likely owing to an improved maximum quantum efficiency (F v /F m ) of photosynthesis. As for the major pigment compositions, the content of chlorophyll a and carotenoids was higher in SO120G than in the wild type. The transcriptome data confirmed that a total of 2077 genes with a change of at least twofold were recognized as differential expression genes (DEGs), of which 1060 genes were up-regulated and 1017 genes were down-regulated. Most of the DEGs involved in lipid biosynthesis were up-regulated with the mutant SO120G. The expression of the gene involved in the fatty acid biosynthesis and photosynthesis of SO120G was upregulated, while that related to starch metabolism decreased compared with that of the wild strain. This work demonstrated that heavy-ion irradiation is an promising strategy for quality improvement. In addition, the mutant SO120G was shown to be a potential algal strain for enhanced lipid production. Transcriptome sequencing and annotation of the mutant suggested the possible genes responsible for lipid biosynthesis and photosynthesis, and identified the putative target genes for future genetic manipulation and biotechnological applications.

Essential trace element selenium in association with selenoproteins, which is found in almost all organisms except higher plants and fungi, is involved in various biological functions. Advancement in the field of whole genome sequencing and data analyzing bioinformatic tools led to the accumulation of genome information of organisms. However, selenoproteins are unique and it needs specialized genomics tool for its identification as well as characterization. In this study, the presence of selenoprotein T (SelT) from Scenedesmus quadricauda was shown for the first time with experimental evidence and compared with SelT of marine microalgae Nannochloropsis oceanica. Along with SelT, all the associated machineries required to synthesize the selenoproteins were also identified. Also, the present study tried to explicate the evolutionary relatedness of SelT of these two organisms with other known bacteria and eukaryotes. Transcript level analysis in S. quadricauda under endoplasmic reticulum stress showed a 1.2 ± 0.28-fold increase in SelT expression. Microalgae efficiently convert sunlight into lipids and carbohydrates, offering bio-based alternatives for energy and chemical production. Improving algal productivity and robustness against abiotic stress requires a systems level characterization enabled by functional genomics. Here, we characterize a halotolerant microalga Scenedesmus sp. NREL 46B-D3 demonstrating peak growth near 25 °C that reaches 30 g/m 2 /day and the highest biomass accumulation capacity post cell division reported to date for a halotolerant strain. Functional genomics analysis revealed that genes involved in lipid production, ion channels and antiporters are expanded and expressed. Exposure to temperature stress shifts fatty acid metabolism and increases amino acids synthesis. Co-expression analysis shows that many fatty acid biosynthesis genes are overexpressed with specific transcription factors under cold stress. These and other genes involved in the metabolic and regulatory response to temperature stress can be further explored for strain improvement.
