Abstract Tumour detection with high selectivity and sensitivity is crucial for delineating tumour margins and identifying metastatic foci during image-guided surgery. Optical nanoprobes with preferential tumour accumulation is often limited by inefficient amplification of biological signals. Here, we report the design of a library of hydrophobic core-tunable ultra-pH-sensitive nanoprobes (HUNPs) for orthogonally amplifying tumour microenvironmental signals on multiple tumour models. We find that tuning the hydrophobicity of nanoparticle core composition with non-ionizable monomers can enhance cellular association of HUNPs by more than ten-fold, resulting in a high cellular internalization efficiency of HUNPs with up to 50% in tumours. Combining high tumour accumulation and high cell internalization efficiency, HUNPs show orthogonally amplified fluorescence signals, permitting the precise locating and delineating margins between malignant lesions and normal tissues with high contrast-to-noise ratio and resolution. Our study provides key strategies to design nanomedicines with high intracellular bioavailability for cancer detection, drug/gene delivery, and therapy.

Abstract Achieving high-luminescence organic light-emitting devices (OLEDs) with narrowband emission and high color purity is important in various optoelectronic fields. Laser displays exhibit outstanding advantages in next-generation display technologies owing to their ultimate visual experience, but this remains a great challenge. Here, we develop a novel OLED based organic single crystals. By strongly coupling the organic exciton state to an optical microcavity, we obtain polariton electroluminescent (EL) emission from the polariton OLEDs (OPLEDs) with high luminance, narrow-band emission, high color purity, high polarization as well as excellent optically pumped polariton laser. Further, we evaluate the potential for electrically pumped polariton laser through theoretical analysis and provide possible solutions. This work provides a powerful strategy with a material–device combination that paves the way for electrically driven organic single-crystal-based polariton luminescent devices and possibly lasers.

Abstract Pain is estimated to affect more than 20% of the global population, imposing incalculable health and economic burdens. Effective pain management is crucial for individuals suffering from pain. However, the current methods for pain assessment and treatment fall short of clinical needs. Benefiting from advances in neuroscience and biotechnology, the neuronal circuits and molecular mechanisms critically involved in pain modulation have been elucidated. These research achievements have incited progress in identifying new diagnostic and therapeutic targets. In this review, we first introduce fundamental knowledge about pain, setting the stage for the subsequent contents. The review next delves into the molecular mechanisms underlying pain disorders, including gene mutation, epigenetic modification, posttranslational modification, inflammasome, signaling pathways and microbiota. To better present a comprehensive view of pain research, two prominent issues, sexual dimorphism and pain comorbidities, are discussed in detail based on current findings. The status quo of pain evaluation and manipulation is summarized. A series of improved and innovative pain management strategies, such as gene therapy, monoclonal antibody, brain-computer interface and microbial intervention, are making strides towards clinical application. We highlight existing limitations and future directions for enhancing the quality of preclinical and clinical research. Efforts to decipher the complexities of pain pathology will be instrumental in translating scientific discoveries into clinical practice, thereby improving pain management from bench to bedside.

Accumulating evidence has proved the close association between alterations in gut microbiota and resistance to chemotherapeutic drugs. However, the potential roles of gut microbiota in regulating oxaliplatin sensitivity in gastric cancer (GC) have not been investigated before. We first found that antibiotic treatment diminished the therapeutic efficacy of oxaliplatin in a GC mouse model. Importantly, this effect could be transmitted to germ-free mice via fecal microbiota transplantation, indicating a potential role of gut microbiota modulation in oxaliplatin efficacy. Further, metagenomics data showed that Akkermansia muciniphila (A. muciniphila) ranked first among the bacterial species with decreased relative abundances after antibiotic treatment. Metabolically active A. muciniphila promotes oxaliplatin efficacy. As shown by metabolomics analysis, the metabolic pattern of gut microbiota was disrupted with significantly downregulated levels of pentadecanoic acid (PEA), and the use of PEA significantly promoted oxaliplatin efficacy. Mechanistically, FUBP1 positively regulated aerobic glycolysis of GC cells to hinder the therapeutic efficacy of oxaliplatin. A. muciniphila-derived PEA functioned as an inhibitory factor of glycolysis by directly antagonizing the activity of FUBP1, which potentiated GC responses to oxaliplatin. Our research suggested a key role for intestinal A. muciniphila and its metabolite PEA in promoting oxaliplatin efficacy, thus providing a new perspective for probiotic and prebiotic intervention in GC patients during chemotherapy.

Abstract The reconstruction method was proposed more than a decade ago to boost the signal of baryonic acoustic oscillations measured in galaxy redshift surveys, which is one of key probes for dark energy. After moving the observed overdensities in galaxy surveys back to their initial position, the reconstructed density field is closer to a linear Gaussian field, with higher-order information moved back into the power spectrum. We find that by jointly analysing power spectra measured from the pre- and post-reconstructed galaxy samples, higher-order information beyond the 2-point power spectrum can be efficiently extracted, which generally yields an information gain upon the analysis using the pre- or post-reconstructed galaxy sample alone. This opens a window to easily use higher-order information when constraining cosmological models.

The objective of this study was to investigate the function of the aquaporin 4 gene (LvAQP4), which had previously been characterized in Pacific white shrimp, Litopenaeus vannamei. To achieve this, double-stranded RNA (dsRNA) targeting LvAQP4 was synthesized and injected into shrimp, resulting in a significant reduction in the expression of LvAQP4. Subsequent to the evaluation of the impact of LvAQP4 transcriptional silencing on shrimp under salinity challenges, we observed that the downregulation of LvAQP4 expression in the gills resulted in the downregulation of aquaglyceroporin (LvAQP3) and unorthodox aquaporin (LvAQP11) genes, as well as ion transporter genes, such as LvNKAα, LvNKAβ, LvCAc, and LvVHA-a. Notably, the inhibition of LvAQP4 had a significant impact on the expression patterns of these osmoregulatory genes, particularly during periods of high salinity stress. Furthermore, there was a notable increase in the mortality rate of shrimp under high-salinity stress. Moreover, the suppression of LvAQP4 through RNA interference led to a significant delay in the molting rate of shrimp. Furthermore, it resulted in an increase in the expression of LvMIH and a significant reduction in key genes involved in the ecdysone signaling pathway, including LvEcR, LvRXR, and LvE75. These findings suggest that LvAQP4 may play a regulatory role in the molting process of shrimp by modulating MIH and the ecdysone signaling pathway. Moreover, this study lays the groundwork for future investigations into the functions of aquaporins in shrimp osmoregulation and growth, contributing to an enhanced understanding of these essential regulatory mechanisms.

Abstract Tumour-associated macrophages (TAMs), as one of the most abundant tumour-infiltrating immune cells, play a pivotal role in tumour antigen clearance and immune suppression. M2-like TAMs present a heightened lysosomal acidity and protease activity, limiting an effective antigen cross-presentation. How to selectively reprogram M2-like TAMs to reinvigorate anti-tumour immune responses is challenging. Here, we report a pH-gated nanoadjuvant (PGN) that selectively targets the lysosomes of M2-like TAMs in tumours rather than the corresponding organelles from macrophages in healthy tissues. Enabled by the PGN nanotechnology, M2-like TAMs are specifically switched to a M1-like phenotype with attenuated lysosomal acidity and cathepsin activity for improved antigen cross-presentation, thus eliciting adaptive immune response and sustained tumour regression in tumour-bearing female mice. Our findings provide insights into how to specifically regulate lysosomal function of TAMs for efficient cancer immunotherapy.

Salinity is one of the most common environmental stress factors, which has a substantial influence on the growth, metabolism, and distribution of crustaceans, while transport of ions and water are considered to play important roles in response to salinity stress. The Chinese shrimp (Fenneropenaeus chinensis) is an important shrimp species cultured in northern China and salinity conditions influence its commercial farming significantly. The study aims to elucidate the potential roles of ion-transport related genes Na+/K+-ATPase α subunit (NKAα), cytoplasmic carbonic anhydrase (CAc), and V-type H+-ATPase subunits (VHA-C, VHA-F, and VHA-G), and water channels of aquaporins (AQP3 and AQP4) in F. chinensis under high-salinity stress. The open reading frames (ORFs) of these genes were cloned, validated, and characterized based on blast against the transcriptomic data. Tissue distribution analysis revealed the predominant expression of these genes in the gills. Then, qPCR was used to examine the expression patterns of genes in the post-larvae shrimp and the gills of adult shrimp in response to hyper-osmotic challenges with two salinities (40 and 45). The small-scale linear information in galaxy samples typically lost during nonlinear growth can be restored to a certain level by the density field reconstruction, which has been demonstrated for improving the precision of the baryon acoustic oscillation (BAO) measurements. As proposed in the literature, a joint analysis of the power spectrum before and after the reconstruction enables an efficient extraction of information carried by high-order statistics. However, the statistics of the postreconstruction density field are difficult to model. In this work, we circumvent this issue by developing an accurate emulator for the prereconstructed, postreconstructed, and cross-power spectra ( ${P}_{\mathrm{pre}}$ , P _post , P _cross ) up to k = 0.5 h Mpc ^−1 based on the Dark Quest N -body simulations. The accuracy of the emulator is at the percent level; namely, the error of the emulated monopole and quadrupole of the power spectra is less than 1% and 10% of the ground truth, respectively. A fit to an example power spectrum using the emulator shows that the constraints on cosmological parameters get largely improved using ${P}_{\mathrm{pre}}$ + P _post + P _cross with ${k}_{\max }=0.25\,h\,{\mathrm{Mpc}}^{-1}$ , compared to that derived from ${P}_{\mathrm{pre}}$ alone; namely, the constraints on (Ω _m , H _0 , σ _8 ) are tightened by ∼41%–55%, and the uncertainties of the derived BAO and RSD parameters ( α _⊥ , α _∣∣ , f σ _8 ) shrink by ∼28%–54%, respectively. This highlights the complementarity among ${P}_{\mathrm{pre}}$ , P _post , and P _cross , which demonstrates the efficiency and practicability of a joint ${P}_{\mathrm{pre}}$ , P _post , and P _cross analysis for cosmological implications.

The intestinal barrier is a sum of the functions and structures consisting of the intestinal mucosal epithelium, mucus, intestinal flora, secretory immunoglobulins, and digestive juices. It is the first-line defense mechanism that resists nonspecific infections with powerful functions that include physical, endocrine, and immune defenses. Health and physiological homeostasis are greatly dependent on the sturdiness of the intestinal barrier shield, whose dysfunction can contribute to the progression of numerous types of intestinal diseases. Disorders of internal homeostasis may also induce barrier impairment and form vicious cycles during the response to diseases. Therefore, the identification of the underlying mechanisms involved in intestinal barrier function and the development of effective drugs targeting its damage have become popular research topics. Evidence has shown that multiple signaling pathways and corresponding critical molecules are extensively involved in the regulation of the barrier pathophysiological state. Ectopic expression or activation of signaling pathways plays an essential role in the process of shield destruction. Although some drugs, such as molecular or signaling inhibitors, are currently used for the treatment of intestinal diseases, their efficacy cannot meet current medical requirements. In this review, we summarize the current achievements in research on the relationships between the intestinal barrier and signaling pathways. The limitations and future perspectives are also discussed to provide new horizons for targeted therapies for restoring intestinal barrier function that have translational potential.

Abstract Background Fasting mimic diet is an effect approach for gastric cancer (GC) treatment. Exploring mechanisms of glucose deprivation-mediated GC suppression is required to develop novel therapeutic regimens. Farnesyltransferase 1 (FDFT1), as a novel target in basic research, has been reported to regulate malignant progression in some types of cancer. However, biological functions of FDFT1 in GC are still unclear. This study focused on biological functions of FDFT1 in GC and the association between glucose starvation (GS) and FDFT1. Methods The data derived from the Kaplan–Meier Plotter database were collected to identify the relationship between survival time and FDFT1 expression levels of GC patients. Bioinformatic analysis was performed to explore the biological functions of FDFT1. The expression levels of targeted genes and microRNAs (miRNAs) were detected with immunohistochemistry, quantitative real-time PCR and western blot. Malignant behaviors were measured using cell counting, cell counting kit-8, 5-ethynyl-2-deoxyuridine, wound healing, invasion transwell assays in vitro and constructions of subcutaneous and lung-metastatic tumors in vivo. The glycolysis of GC cells was determined by a series of metabolites, including lactate acid, pyruvic acid, ATP production, rates of glucose uptake, extracellular acidification rate and oxygen consumption rate. Results FDFT1 was downregulated in GC and negatively correlated with pathological T stage, pathological TNM stage and cancer differentiation. High expression of FDFT1 also indicated better prognosis of GC patients. FDFT1 upregulation attenuated proliferation, migration and invasion of GC. miR-216a-5p was identified as a critical suppressor of FDFT1 expression and miR-216a-5p/FDFT1 axis regulated malignant behaviors and glycolysis of GC cells. GS suppressed malignant behaviors of GC by targeting miR-216a-5p/FDFT1 axis both in vitro and in vivo. Conclusion This study illustrated novel mechanisms by which GS effectively suppresses GC. FDFT1 may become a potential prognostic indicator and novel target of GC therapy.

Abstract Background Phosphoglucomutase 1 (PGM1) acts as an important regulator in glucose metabolism. However, the role of PGM1 in gastric cancer (GC) remains unclear. This study aims to investigate the role of PGM1 and develop novel regimens based on metabolic reprogramming in GC. Methods Correlation and enrichment analyses of PGM1 were conducted based on The Cancer Genome Atlas database. Data derived from the Kaplan–Meier Plotter database were analyzed to evaluate correlations between PGM1 expression and survival time of GC patients. Cell counting kit-8, 5-Ethynyl-2-deoxyuridine, flow cytometry assays, generation of subcutaneous tumor and lung metastasis mouse models were used to determine growth and metastasis in vitro and in vivo. Cell glycolysis was detected by a battery of glycolytic indicators, including lactate, pyruvic acid, ATP production and glucose uptake. Fatty Acid Synthase (FASN) activity and expression levels of lipid enzymes were determined to reflect on lipid metabolism. Results Correlation and enrichment analyses suggested that PGM1 was closely associated with cell viability, proliferation and metabolism. PGM1 was overexpressed in GC tissues and cell lines. High PGM1 expression served as an indicator of shorter survival for specific subpopulation of GC patients. It was also correlated with pathological tumor stage and pathological tumor node metastasis stage of GC. Under the glucose deprivation condition, knockdown of PGM1 significantly suppressed cell viability, proliferation and glycolysis, whereas lipid metabolism was enhanced. Orlistat, as a drug that was designed to inhibit FASN activity, effectively induced apoptosis and suppressed lipid metabolism in GC. However, orlistat conversely increased glycolytic levels. Orlistat exhibited more significant inhibitive effects on GC progression after knockdown of PGM1 under glucose deprivation due to combination of glycolysis and lipid metabolism both in vitro and in vivo. Conclusions Downregulation of PGM1 expression under glucose deprivation enhanced anti-cancer effects of orlistat. This combination application may serve as a novel strategy for GC treatment.

Neutrophil extracellular traps (NETs), products of neutrophil death when exposed to certain stimuli, were first proposed as a type of response to bacterial infection in infectious diseases. Since then, extensive studies have discovered its involvement in other non-infectious inflammatory diseases including thromboembolism, autoimmune diseases, and cancer. Colorectal cancer (CRC) is one of the most common malignancies in the world. NET formation is closely associated with tumorigenesis, progression, and metastasis in CRC. Therefore, the application of NETs in clinical practice as diagnostic biomarkers, therapeutic targets, and prognostic predictors has a promising prospect. In addition, therapeutics targeting NETs are significantly efficient in halting tumor progression in preclinical cancer models, which further indicates its potential clinical utility in cancer treatment. This review focuses on the stimuli of NETosis, its pro-tumorigenic activity, and prospective clinical utility primarily in but not limited to CRC.

BackgroundCircular RNAs (circRNAs) regulate multiple malignant behaviors of various types of cancer. The role of circDNMT1, a newly identified circRNA, remains unknown in gastric cancer (GC). This study aimed to elucidate the underlying mechanisms of circDNMT1 in regulating GC progression.MethodsmicroRNA (miRNA) and circRNA expression was detected by quantitative real-time PCR. Western blotting was performed to measure hypoxia inducible factor-1 alpha (HIF-1α) protein expression. Sanger sequencing, gel electrophoresis and fluorescence in situ hybridization were performed to identify the presence of circDNMT1. The clinicopathological features and overall survival of patients were analyzed based on circDNMT1 expression. The proliferation, migration and invasion of GC cells were determined by cell counting kit-8, 5-ethynyl-2’-deoxyuridine, wound healing and transwell assays. Glycolysis of GC cells was detected based on the levels of glucose uptake, the lactate acid, ATP and pyruvic acid production and the extracellular acidification and oxygen consumption rates. The binding sites between miR-576-3p and circDNMT1 or HIF-1α were predicted by online bioinformatic tools and were validated using RNA pull-down and luciferase reporter assays. Xenograft models were established to determine the effects of the circDNMT1/miR-576-3p/HIF-1α axis on GC growth and metastasis in vivo.ResultscircDNMT1 was successfully identified and shown to be overexpressed in GC tissues and cell lines. The expression levels of circDNMT1 were correlated with pathological T stage, pathological TNM stage and shorter survival time of GC patients. circDNMT1 knockdown inhibited the proliferation, migration, invasion and glycolysis of GC cells. circDNMT1 functioned as an oncogenic factor by sponging miR-576-3p. HIF-1α was negatively regulated by miR-576-3p via binding its mRNA 3’ untranslated region. circDNMT1 promoted malignant behaviors and metabolic reprogramming of GC by targeting the miR-576-3p/HIF-1α axis both in vitro and in vivo.ConclusionThese findings demonstrated that circDNMT1 knockdown inhibited GC proliferation, migration, invasion and glycolysis through sponging miR-576-3p/HIF-1α axis. circDNMT1 may be a novel target for GC treatment.

Aquaporins (AQPs) are major intrinsic proteins that facilitate the transport of water, small solutes, and ions through membranes. In mammals, the classical aquaporin AQP4 plays a central role in the preservation of central nervous system (CNS) water homeostasis. Litopenaeus vannamei is a typical euryhaline crustacean, with a salinity tolerance range of 0.5–78. Compared to those reared at low-salinity, shrimps cultured at high-salinity have better taste and qualities, but slower growth, lower product yield, and higher mortality. Thus, there is an urgent need to research the osmoregulation of L. vannamei under high-salinity. Previous studies mainly focus on the osmoregulation of gill and hepatopancreas, whereas there is a lack of research on the osmoregulatory roles of CNS in L. vannamei. In the current study, we first presented another two classical AQPs from L. vannamei, of which the arthropod-specific big brain aquaporin (LvBib) was expressed as C-terminal splice variants (LvBib_L and LvBib_S), and the aquaporin-like protein (LvAqpl) was a new type of aquaporin in crustaceans. Tissue distribution revealed that LvAqp4, LvAqpl, and LvBib_L were mostly expressed in the CNS of brain and thoracic ganglion. Then, the expression pattern of aquaporins in brain and thoracic ganglion were examined under both acute and chronic high-salinity stresses. The LvAqpl was found to be down-regulated in both tissues in the two experiments; LvAqp4 was elevated in both tissues under acute stress, whereas it was decreased in the thoracic ganglion during early periods of adaption to high-salinity; LvBib_L was up-regulated in brain but down-regulated in thoracic ganglion under acute stress, whereas it showed the opposite expression pattern during chronic stress. The results suggest that these aquaporins have an important role in the CNS response to high-salinity challenge. Our study provides a base for future mechanistic studies on the role of classical aquaporins in osmoregulation.

Euryhaline crustaceans cope with external salinity changes by mechanisms of osmoregulation. In the current study, we first cloned and confirmed the ORF sequences of the ion-transportation-related genes Na+/K+-ATPase α subunit (NKAα), cytoplasmic carbonic anhydrase (CAc), and V-type H+-ATPase G subunit (VHA-G), and water channels of aquaporins (AQP3, AQP4, and AQP11) from kuruma shrimp (Marsupenaeus japonicus). Further tissue expression patterns showed a higher expression of MjAQP4, MjCAc, MjNKAα, and MjVHA-G in the gills, as well as a higher expression of MjAQP3 and MjAQP11 in the intestine and muscle, respectively. Then, qPCR analysis was used to assess the mRNA expression levels of those osmoregulatory genes in both post-larvae and adult shrimp when they were exposed to acute salinity stress or salinity acclimation. The results revealed significantly decreased expression levels of MjAQP3, MjAQP11, MjNKAα, and MjCAc, and higher expression levels of MjAQP4 and MjVHA-G when the post-larvae shrimp were directly subjected to 10‰ or 50‰ salinity. Moreover, similar expression patterns were also observed in the post-larvae shrimp during the accommodation to 10‰ or 50‰ salinity. As to the adult shrimp, significantly higher expression levels of those genes were observed in the gills after exposure to 10‰ salinity, whereas only the expression levels of MjAQP3, MjAQP11, and MjNKAα were up-regulated in the gills at 40‰ salinity. In contrast, the expression of MjVHA-G was significantly decreased at 40‰ salinity. Finally, during the acclimation to 10‰ salinity, the expression levels of MjAQP3, MjAQP11, and MjNKAα were also significantly elevated, while the expression of MjCAc was significantly decreased in the gills. In addition, the expression levels of MjAQP3, MjAQP4, MjCAc, and MjVHA-G were significantly decreased in the gills during the acclimation to 55‰ salinity. The findings of the study suggest that the examined genes are critical for the adaptation of aquatic crustaceans to changing environmental salinity. Our study lays as the foundation for further research on osmoregulation mechanisms in M. japonicus.

Background: This study aimed to analyze and compare the short-term and long-term outcomes of proximal gastrectomy (PG) and total gastrectomy (TG) in patients with locally advanced proximal gastric cancer (GC) following neoadjuvant chemotherapy (NACT). Method: A multicenter retrospective cohort study and propensity score matching (PSM) were employed. The authors examined 367 patients with proximal GC who received NACT followed by PG (n =164) or TG (n=203) at two Chinese medical institutions between December 2009 and December 2022. Clinical and pathological parameters, postoperative complications, and 5-year overall survival (OS) and recurrence-free survival (RFS) were compared between the two groups. The dissection status and metastasis rate of each lymph node station were assessed. Results: After PSM, 80 patients were enrolled in both TG and PG group, and baseline characteristics were comparable between the groups (all P> 0.05). The TG group had a higher total number of lymph nodes retrieved (P<0.001) and longer operative time (P =0.007) compared to the PG group. The incidence of Clavien-Dindo grade II or higher postoperative complications was similar between the TG group (21.3%, 17/80) and the PG group (17.5%, 14/80) (P =0.689). The 5-year OS rates were 68.4 for the PG group and 66.0% for the TG group (P= 0.881), while the 5-year RFS rates were 64.8 and 61.9%, respectively (P= 0.571), with no statistically significant differences. Metastasis rates at lymph node stations #4d, #5, #6, and #12a were notably low in the TG group, with values of 2.74, 0.67, 1.33, and 1.74%, respectively. Conclusion: For proximal GC patients following NACT, PG maintains comparable curative potential and oncological efficacy to TG, making it a safe option. [ABSTRACT FROM AUTHOR]
