Although active learning improves student outcomes in science, technology, engineering, and mathematics (STEM) programs, it may provoke anxiety in some students. We examined whether two psychological variables, social anxiety (psychological distress relating to the fear of negative evaluation by others) and academic self-efficacy (confidence in one's ability to overcome academic challenges), interact with student perceptions of evidence-based instructional practices (EBIPs) and associate with their final grades in a STEM-related course. Human anatomy and physiology students in community college courses rated various EBIPs for their perceived educational value and their capacity to elicit anxiety (N = 227). In general, practices causing students the most anxiety (e.g., cold calling) were reported by students as having the least educational value. When controlling for students' self-reported grade point averages, socially anxious students rated several EBIPs as more anxiety inducing, whereas high-efficacy students reported less anxiety surrounding other EBIPs. Furthermore, mediation analysis revealed that individual differences in academic self-efficacy at the beginning of the term explained some of the negative association between students' social anxiety levels and final grades in the course. Our results, obtained in a community college context, support a growing body of evidence that social anxiety and academic self-efficacy are linked with how students perceive and perform in an active-learning environment.

First-generation college students face a variety of barriers in higher education compared with their continuing-generation peers. Active learning practices in STEM classrooms can potentially narrow the achievement gap by increasing academic self-efficacy, or confidence in academic abilities. However, these practices can also provoke anxiety in students. Given that anxiety can impair cognitive performance, we sought to understand how first-generation students perceive active learning practices and whether these perceptions affect the anticipated benefits of active learning. As part of a larger study on pedagogical practices in anatomy and physiology courses at the community college level, we asked students to rate various active learning techniques on how much each provoked anxiety and how much each contributed to their learning. All students (N= 186) rated some techniques as more anxiety-provoking than others (e.g., cold calling); however, compared to continuing-generation students, first-generation students’ ratings tended to be higher. First-generation students anticipated doing more poorly in a course and attained lower final grades. Notably, the use of active learning practices did not improve first-generation students’ academic self-efficacy: by the end of term, academic self-efficacy decreased in non-white first-generation students whereas other students showed little change. When introducing active learning strategies, instructors may need to proactively address underrepresented minority students’ emotional reactions and ensure that all students experience success with these practices early in a course as a way to bolster academic self-efficacy.

A vital role for Clinical and Translational Science Award (CTSA) evaluators is to first identify and then articulate the necessary change processes that support the research infrastructures and achieve synergies needed to improve health through research. The use of qualitative evaluation strategies to compliment quantitative tracking measures (e.g., number of grants/publications) is an essential but under-utilized approach in CTSA evaluations. The Clinical and Translational Science Institute of Southeast Wisconsin implemented a qualitative evaluation approach using appreciative inquiry (AI) that has revealed three critical features associated with CTSA infrastructure transformation success: developing open communication, creating opportunities for proactive collaboration, and ongoing attainment of milestones at the key function group level. These findings are consistent with Bolman & Deal's four interacting hallmarks of successful organizations: structural (infrastructure), political (power distribution; organizational politics), human resource (facilitating change among humans necessary for continued success), and symbolic (visions and aspirations). Data gathered through this longitudinal AI approach illuminates how these change features progress over time as CTSA funded organizations successfully create the multiinstitutional infrastructures to connect laboratory discoveries with the diagnosis and treatment of human disease.

A number of possible mediators have been proposed to couple neuronal activity with local cerebral metabolic activity and blood flow, but the mechanisms by which these mediators act is still unclear. In order to explore these coupling mechanisms, we used the rodent whisker-barrel cortex (WBC) model to test the hypothesis that modulation of K(Ca) channels is an important step in this coupling process. Anesthetized rats were prepared for laser-Doppler flowmetry (LDF) or evoked potential recordings utilizing a thinned cranial window over WBC. Superfusion of the K(Ca) channel blockers tetraethylammonium (TEA) or iberiotoxin directly onto WBC attenuated the magnitude of the whisker evoked LDF changes. Similar effects were seen after intravenous administration of TEA. Although attenuated, neither the temporal profile of the elicited blood flow responses nor the evoked electrical activity in WBC were affected by K(Ca) blockade. These data suggest that the process of cerebral metabolism/blood flow coupling in the rodent WBC involves K(Ca) channels.

Many properties of nitric oxide, NO, (localization, diffusiveness, half-life, vasodilatory affects) have supported its potential role in mediating the link between local cerebral activity and blood flow. However, evidence that both supports and refutes a role for NO in functional hyperemia have been presented. The present study employed multiple nitric oxide synthase inhibitors, two anesthetic regimes and laser-Doppler flowmetry to test the hypothesis that NO is critically involved in mediating the functional hyperemic response within rodent whisker-barrel cortex (WBC). In urethane anesthetized animals, functional hyperemic responses were obtained both before and after 1 mg/kg atropine infusion, 30 mg/kg i.v. L-NAME (N-Nitro-L-arginine methylester) infusion, 30 mg/kg L-NA (N-Nitro-L-arginine) infusion or 25 mg/kg 7-NI (7-nitroindazole). L-NAME was also tested in a group of animals pretreated with halothane before urethane anesthesia. Neither the magnitude of the blood flow response nor its time course was altered by NO blockade or atropine administration when compared to pre-infusion controls in urethane anesthetized rats. In contrast, animals that were pretreated with halothane exhibited a 33% inhibition of functional hyperemia after L-NAME administration. Taken together, these data do not support a primary role for NO in rat WBC functional hyperemia and suggest that previous reports of inhibition may have been secondary to the anesthesia employed.

It has long been known that the level of neuronal activity is correlated to the level of localized blood flow. Despite the importance of functional hyperemia in the brain, the relationship between blood flow and electrical activity has not been clearly demonstrated parametrically in a single region of cerebral cortex. We investigated both the magnitude and temporal characteristics of the blood flow response in somatosensory cortex while varying the frequencies of whisker movement. The full whisker pad on one side of the rat's face was repeatedly moved for 13 s at frequencies of 1.5, 2, 3, 4, 6, 8, and 10.5 Hz, and the resulting changes in blood flow were quantified using Laser-Doppler flowmetry (LDF). The magnitude of the blood flow response increased linearly with increasing frequency while the temporal parameters of time to half maximal value and time to return halfway to baseline after stimulus termination did not vary. Baseline blood flow levels were elevated by breathing rats on a 5% CO2 mixture. No significant alteration in the LDF plateau response to whisker movement was observed compared to normal air, suggesting sustained vasodilation reserve capacity remained after CO2-induced vasodilation. These data demonstrate linear blood flow responses to presumptive linear increases in neuronal activity with sufficient vascular reserve capacity to overcome moderate CO2-induced dilation, and support the use of blood flow changes in neuroimaging studies. They provide a framework to study the neurobiological signal transduction mechanisms coupling neuronal electrical activity with regional alterations in blood flow.

In order for vacuum-assisted venous return (VAVR) to be used safely and efficiently during cardiopulmonary bypass (CPB), a full understanding of venous return is necessary. The focus of this work was to use the concepts of energy conservation and viscous energy dissipation in the development of a theoretical model of venous return utilizing vacuum assist. The effectiveness and accuracy of this model has been verified through in vitro laboratory investigations and statistical analysis. Although VAVR can provide higher flows through smaller venous cannula, vacuum assist may lead to increased levels of wall shear stress as shown in this work. The clinical implications of VAVR have yet to be investigated, but may lead to an exacerbation of the detrimental effects of CPB during cardiac surgery.

Several recent studies have demonstrated a neuromodulatory role for endocannabinoids via their ability to act as retrograde inhibitors of synaptic neurotransmission. We utilized the functional hyperemic response to controlled whisker stimulation to determine whether endogenous cannabinoids modulate synaptic transmission within the primary somatosensory cortex of rats. As previously demonstrated, whisker-stimulation resulted in a robust hyperemic response measured using laser Doppler flowmetry within the whisker barrel cortex. Administration of the CB 1 receptor antagonist, SR141716 (1 mg/kg i.v.), significantly potentiated the functional hyperemic response to whisker-stimulation, while having no effect on basal blood flow within the whisker barrel cortex. These data suggest that suppression of endogenous cannabinergic neurotransmission results in increased cortical activity in response to physiological sensory stimulation.

Dabei steht Erziehung eher im Vordergrund als Vergeltung und Strafe. Diese Haltung spiegelt sich auch im deutschen und im niederlandischen Jugendstrafrecht (wenn auch seit 1995 abgeschwacht) wider. Der Rahmen fur die moglichen Reaktionen auf Straftaten Jugendlicher wird in Deutschland durch das Jugendgerichtsgesetz und in den Niederlanden durch das Strafgesetzbuch gesteckt. For several decades, cranial windows have been used to investigate questions relating to cerebral blood flow and its regulation. In general, these techniques have utilized either `open' cranial windows for the direct observation of the intracranial vasculature, or `closed' cranial windows in which the skull and dura are removed and replaced with a clear seal, such as a coverslip. Here we describe a method of studying blood flow responses elicited by the physiological stimulus of whisker movement while using a `thinned skull' cranial window created over the rat whisker-barrel cortex. This method employing an automated whisker stimulator coupled with laser-Doppler flowmetry focused through the thinned skull cranial window, is less invasive than other cranial window techniques, and allows for the study of the effects of stimulation parameters and systemically administered compounds on whisker movement elicited blood flow responses. Automated whisker stimulation and data collection also allow for precise temporal averaging of laser-Doppler measured responses, leading to increased precision in determining the true shape of the evoked blood flow response pattern.
