Decellularized grafts have shown promising results in tissue engineering. The aim of decellularization is to create a scaffold that is devoid of immunogenic components, has natural tissue architecture, and can provide cellularity again. The aim of our study is to obtain a living/regenerative valve by coculture of HUVEC and human dermal fibroblast cells on the pulmonary heart valves of decellularized young sheep and subsequently perform their in vitro characterization to determine its suitability for clinical studies. Various characterization tests were applied to sheep pulmonary heart valves decellularized by the detergent-based method. HUVEC and dermal fibroblast cells were seeded on the scaffold, and their viability was determined by MTT analysis, adhesion by SEM, and cell infiltration by histological staining. Finally, to determine the regenerative ability of the resulting live cap, collagen type I (Col1), collagen type III (Col3), and elastin (Eln) gene expressions were analyzed by PCR. The results showed that cell proliferation increased day by day on the scaffold. In histological findings, it was observed that cellular regeneration was almost completely achieved, especially in arterial wall samples. According to PCR findings, a significant increase in Col1, Col3, and Eln gene expressions was observed in arterial wall samples. In this study, for the first time in the literature, the regeneration potential of differentiated human cells on decellularized sheep heart valves was observed, and markers related to new ECM production were analyzed. In conclusion, we suggest that the decellularized heart valves of young sheep can be used as a starting matrix in tissue engineering studies, and clinical studies are needed. [ABSTRACT FROM AUTHOR]

The aim of this study is in order to investigate the effects of explant source, explant type and MS media composition containing different concentrations of sucrose, activated carbon and Coconut milk for regeneration and browning of Laurus nobilis L. using thin cell layer (TCL) culture system. A higher rate of callus (57.15%) and shoot (2%) regeneration and a lower rate of blackening were determined in transversely cut stem TCL explants compared to leaf explants. While 1.33% callus regeneration was achieved in leaf explants; shoot regeneration could not be achieved. While more callus regeneration (35.17%) was found in explants taken from the field, more shoot regeneration (1.5%) and lower rate of browning were obtained in explants taken from in vitro. In the sugar trials, the highest callus regeneration (40.83%) was defined in MS medium containing 30 g/L sucrose supplemented with 1 mg/L BAP, and the highest shoot regeneration (2.5%) was determined in MS medium containing 45 g/L and 60 g/L sucrose supplemented with 1 mg/L BAP. When explant type, explant source and nutrient media composition are considered together; the highest callus regeneration (100%) was obtained in fieldsourced stem TCL explants cultured in medium containing 25 mL/L coconut milk and 1 mg/L BAP. The highest shoot regeneration (6.6%) was determined in in vitro stem TCL explants cultured in MS media containing 30, 45, 60 g/L sucrose and 1 mg/L BAP. The lowest percentage of browning (50%) was obtained from in vitro stem TCL explants cultured in MS medium containing 2 g/L activated carbon and 1 mg/L BAP. [ABSTRACT FROM AUTHOR]

Granül aktif karbon (GAK), gaz ve sıvı akımlardan kirleticileri uzaklaştırmak için uygulanan adsorpsiyon proseslerinde yaygın olarak kullanılmaktadır. Karbon bazlı adsorbentlerin yeniden kullanılabilirliği adsorpsiyon prosesinin teknik ve ekonomik anlamda yaygınlaşmasını sağlar. Ancak doygunluk sonrası bertarafla ilgili yüksek maliyetler ve çevresel sorunlar nedeniyle uygulama sınırlıdır. Doymuş GAK’in hizmet ömrünün uzatılması için uzun adsorpsiyon döngülerine imkan tanıyan, düşük maliyette, karbon kaybı minimum seviyede olan ve çevresel etkiyi en aza indiren farklı rejenerasyon teknikleri belirlenmiştir. Bu teknikler, iki ayrı yolla gerçekleştirilebilir: yalnızca aktif karbonda adsorbe edilen kirleticilerin desorpsiyonuna dayalı veya bu kirleticilerin ayrışmasına dayanan rejenerasyon. Genel olarak rejenerasyon metotları termal, kimyasal ve mikrobiyolojik olarak sınıflandırılmaktadır. Yeni geliştirilen rejenerasyon metotları, enerji verimliliği, seçiciliği, düşük maliyeti ve çevresel uyumluluğu açısından daha çok tercih edilmektedir. Ayrıca, yeni geliştirilen rejenerasyon metotları ile tükenmiş aktif karbonun rejenerasyonu yerinde yapılabilmektedir. Bu durum önemli avantaj sağlamaktadır. Bu çalışmada doymuş GAK'lerin rejenerasyonu ile ilgili literatür gözden geçirilmiş ve umut verici teknikler vurgulanmıştır.

Immortelle grass (Helichrysum italicum (Roth) G. Don), which spreads in the Southern Marmara and Aegean regions, can be grown in arid and semi-arid regions. In addition, due to its rich essential oil and secondary metabolite content, it has an important place in modern medicine and cosmetics, including traditional treatment methods. Although the propagation of plants by shoot regeneration in vitro has been achieved in many plant species, studies on tissue culture in immortelle grass are limited. This study aims to optimize the tissue culture study in immortelle grass and provide a basis for the next in vitro, molecular, and secondary metabolite studies. In addition, it promotes the plant by optimizing the healthy and disease-free seedling production method for cultural agriculture in the region. Three different (15%, 25%, and 35%) NaOCl concentrations were tested for 10 and 20 minutes during the sterilization phase of the explants. The most successful result was obtained in the medium containing 35% NaOCl for 10 minutes. Sterilized explants were transferred to MS and Gamborg B5 nutrient media containing BAP, GA, and NAA plant growth regulators for shoot regeneration. The best regeneration in explants was obtained in MS medium containing 0.5 mg L-1 BAP, 1 mg L-1 GA, and 0.2 mg L-1 NAA. No growth was observed in trials containing Gamborg B5, and vitrification and darkening occurred in the explants. After four weeks, the shoots reaching a length of 3 cm were taken into MS and ½MS medium containing 0 MS, 0.5 mg L-1 IBA, 1 mg L-1 IBA, 1.5 mg L-1, and 2 mg L-1 IBA as a rooting medium. 100% rooting was observed in all prepared media within four weeks. As a result of micropropagation studies, the rooted plants were transferred to the acclimatization stage within three months and then moved to the pots in the greenhouse and to the field one month later. This research was carried out to determine the in vitro regeneration abilities of tulip leaf explants. In the study, both sterile leaf explants obtained by culturing sterilized onions and leaf explants obtained by sterilizing leaves obtained from the onions germinated in pots were used. For this purpose MS medium containing 2.4-D (2 mg.l-1), TDZ (2 mg.l-1), Kinetin (2 mg.l-1), NAA (2 mg.l-1), 2.4-D+Kinetin (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1+ 2 mg.l-1), İAA+TDZ (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), İAA+BAP (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), NAA+Kinetin (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), NAA+TDZ (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1) and 2.4-D+TDZ (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1) were used in the study. As a result of subcultures, 6% callus formation was detected in MS medium containing 2.4-D (2 mg.l-1) and NAA (2 mg.l-1) hormone, which is one of the subcultures of leaves formed from the in vitro germination of Purple Prince Tulip bulbs. The highest rate of regeneration was determined as 10% callus formation in the medium containing Kinetin (2 mg.l-1). As a result, while callus regeneration was achieved in 3 different media containing 2.4-D (2 mg.l-1), NAA (2 mg.l-1) and Kinetin (2 mg.l-1) hormones among subcultured media, no regeneration was achieved in the other mediums. Mineral trioksit agregat (MTA), esas olarak kalsiyum ve silikat elementlerinden oluşan biyoaktif endodontik bir simandır. 1990' larda Torabinejad tarafından tanıtılmış, 1997' de ise Amerika Birleşik Devletleri'nde kullanılmak üzere Gıda ve İlaç İdaresi (FDA) tarafından onaylanmıştır. MTA; sement gelişimine olanak tanıyan ve periodontal ligament hücrelerinde rejenerasyon sağlayan ilk restoratif materyaldir. Biyouyumlu bir materyal olan MTA, düşük sitotoksisiteye sahiptir. Apeks dışına taştığı durumlarda minimal inflamasyona neden olur. Odontoblastik tabaka oluşumu sayesinde dentin köprüsü oluşumunu indükler. Pulpal fibroblastlardan sitokin salınımını uyarır, bu da sert doku oluşumunu stimüle ederek hücrelerin hızlı büyümesini destekler. İlk olarak retrograd dolgu materyali olarak kullanılmaya başlayan MTA; daha sonra pulpa kuafaj tedavileri, iatrojenik ve furkasyon perforasyonlarının tamiri, retine süt dişlerinin kök kanal tedavisi, apeksifikasyon sırasında bariyer oluşturulması, devital beyazlatmada bariyer gibi çeşitli klinik uygulamalarda da kullanım alanı bulmuştur. MTA’ nın biyouyumluluk, sızdırmazlık, pulpa ve periradiküler dokuların rejenerasyonunu artırma kapasitesi gibi özellikleri; dental kullanım alanlarındaki çeşitliliği ve klinik başarı oranlarını artmasını sağlamış ve bu yenilikçi yapısıyla klinisyenlerin ilgi odağı haline gelmiştir. Dişte renk değişikliğine sebep olması ve sertleşme süresindeki değişiklikler başlıca dezavantajlarıdır. Yapılan in vitro çalışmalar başarılı sonuçlar vermiş olsa da; uzun dönem başarısının desteklenmesi amacıyla kontrollü klinik çalışmalara ihtiyaç duyulmaktadır. Bu literatür derlemesinde, MTA’ nın fiziksel, kimyasal, biyolojik ve antimikrobiyal özellikleri, klinik uygulama şekilleri ve güncel formülasyonları sunulmaktadır.

Plant tissue culture techniques, which are of great importance in terms of plant breeding and cultivation, are a wide scientific area that includes various methods for plant production especially in in vitro conditions. One of the most important factors affecting the success of the techniques used in plant production in in vitro conditions is determination of explant type. Thin cell layer (TCL) culture system was first described by Tran Thanh Van in 1973. TCL culture system is a plant tissue culture technique in which structures such as tissue, organs, embryos, or whole plants are regenerated by culturing longitudinally or transversely excised explants containing thin layers of cells and tissue. This technique has been successfully used in in vitro micropropagation, synthetic seed production, cryopreservation and genetic studies. The problems encountered in the production of the lentisk trees, which are one of the important endemic plants of our country have led to the use of biotechnological methods. In this research for this purpose, the TCL (Thin Cell Layer) technique was considered to investigate of in vitro regeneration potential of expants used for production of lentisk. Firstly, the leaf, node and stem parts of the plant were cut by TCL technique and these explants had been cultured in semi-solid MS media supplemented with 2,4-D and KIN at different concentrations (0.1, 0.2, 0.5 and 1.0 mg/L). The highest callus formation percentage was 100% in transverse stem layers and longitudinal node in MS media including 1 mg/L 2,4-D and 1 mg/L KIN. The lowest callus regeneration ratios were found as 26.67% for three explant types (transverse leaf, transverse stem, longitudinal node). Due to the high rate of darkening in regenerated calli, these were transferred primarily to semi-solid media containing different antioxidants (ascorbic acid, citric acid, PVP, active charcole) and after that cultured in liquid media containing different plant growth regulators (IAA, KIN and BAP) to induced somatic embryogenesis. Later, the calli were encapsulated to prevent darkening and the nurse technique was applied with Aloe vera L. and Gossypium hirsutum L. calli as a different application. Objective: Bone grafts and even bone substitutes do not meet all of the requirements of bony reconstructions. The aim of this study was to generate bone tissue from autologous adipose tissue-derived mesenchymal stem cells (ATDMSCs) and decellularised bone allografts. Methods: A 1.5 cm bone defect developed in the middle third of the rabbit's ulna. Reconstructions were carried out using miniplate and screws and interpositional autogenous bone grafts according to the designs of the groups: (1) No touch, (2) cryopreserved, (3) decellularised and (4) ATDMSCs-implanted decellularised bones. Before implantation, ATDMSCs in the last group were labelled with Q-dot and identified microscopically. Results: Graft recovery and irregular callus formation were observed in the first, second and forth groups. In the first group, the organisation of Haversian systems, the structure of the lacunae and the presence of canaliculi ossiums were observed; in the second group, approximately 40% of the Haversian canals contained blood vessels, and canaliculi ossiums in the form of thin filaments were found in 90% of the microscopically examined areas; in the third group, most Haversian canals were empty, most osteocyte canals were devoid of cells, and canaliculi ossiums were absent; in the fourth group, some of the Haversian canals contained blood vessels, and there were partly lacunae containing cells due to decellularisation, whereas in approximately 50% of the examined microscope areas, the presence of canaliculi ossiums with evidence of mesenchymal stem cells differentiated into osteocytes was demonstrated by Q-dot traced cells. Conclusion: In this study, the establishment of a proper niche environment for adipose-derived mesenchymal cells promotes their development into osteogenic cells. [ABSTRACT FROM AUTHOR]

In this study, we investigate the relation between the explant size and the success of virus elimination in ‘Superior Seedless’ grape variety. Plants were tested by prior to thermotherapy and in vitro culture by using Double Antibody Sandwich-Enzyme-Linked Immunosorbent Assay (DAS-ELISA) and Reverse Transcription-Polymerase Chain Reaction (RT-PCR) and verified as infected by different viruses (Grapevine leafroll associated virus -1+3, Grapevine leafroll associated virus -4 Strains, Grapevine rupestris stem pitting associated virus). Different size of shoot tip explants (0.1-0.5-1-5-10 mm) were excised and cultured on C2 D medium supplemented with 1 mgl-1 BA. All of the explants bigger than 0.1 mm 100% regenerated while 70% of the 0.1 mm explants showed complete regeneration and turned into a whole plant. Plants regenerated from 10 mm shoot tip explants have all three viruses (GLRaV-1+3, GLRaV-4 strains and GRSPaV) as founded in mother plant. Plants regenerated from 5 mm explants have two of them (GLRaV-1+3, GLRaV-4 strains) and plants regenerated from 1 mm explants have one (GLRaV-1+3). Bu derlemede yazarlar kök hücre/progenitör hücre transplantasyonunun sinir hasarlarının iyileştirilmesinde kullanımı ile ilgili yakın zamandaki gelişmeler ve bu alandaki başlıca çalışmalar hakkında bilgi vermeyi amaçlamaktadır. Periferik sinirlerin akson rejenerasyonu ve hedef dokularını tekrar innerve etme potansiyeline sahip olmalarına rağmen ağır sinir hasarı durumlarında düzelme sınırlı kalmaktadır. Denervasyona uğramış periferik sinirin Schwann hücrelerinin rejenerasyonda yeteri kadar rol alamayışlarından dolayı denerve olmuş distal sinir dokusu ortamının dış kaynaklı doku hücreleriyle desteklenmesi yaklaşımı gerçekçi görülmüştür. Çeşitli kök hücrelerinin periferik sinir rejenerasyonundaki etkileri çalışılmıştır. Deri, kemik iliği ve yağ doku kökenli kök hücreleri Schwann hücrelerine dönüşme kapasiteleriyle en ümit verici adaylar olarak belirlenmiştir. Pluripotent indüklenmiş kök hücre denemelerine ek olarak, yakın zamanlarda yapılan çalışmalar kök hücrelerinde büyüme faktörlerinin ifadesinin uyarılması da rejenerasyonda önemli potansiyele sahiptir. Kök hücrelerinin etkili kullanılabilecek kaynak olmaları ve sinir rejenerasyonunda ümit verici faktörler olduklarının düşünülmesine rağmen bu hücre tedavi potansiyellerinin klinikte kullanılması için gerekli ideal yöntemler henüz oluşturulamamıştır.

Tail anomalies, such as bifurcation and trifurcation, are observed in different species of Lacertid lizards. In the present study, bilateral tail bifurcation was observed in male specimens belonging to Darevskia clarkorum and Darevskia rudis in the Eastern Black Sea Region of Turkey. In the specimen of D. clarkorum, SVL and TL are 55.37 mm and 66.43 mm, respectively. Bifurcation from the base of the tail occurred at 42.82 mm. The regenerated tail is 8.45 mm. SVL and TL are 83.56 mm and 128.93 mm, respectively in the specimen of D. rudis. Bifurcation from the base of the tail occurred at 33.38 mm. The length of the regenerated tail is 46.77 mm. Diesel Particulate Filters (DPF) are currently used in many diesel vehicles. In this study, the filtration characteristics of different DPF materials including SiC, Crd and AT were investigated under the New European Driving Cycle (NEDC) standards by designing the exhaust system for a diesel vehicle with Diesel Oxidation Catalyst (DOC) and DPF. The exhaust system model is determined by examining of the diesel vehicles. The study was performed in three dimensions with using of the AVL-FIRE software. The properties of materials which are alternatives to SiC (Silicon Carbide), the most widely used as DPF material, have been investigated. The effects of DPF material on heat transfer, pressure drop and exhaust emissions were shown in this study. As a result, it is seen that the used materials in this study have similar characteristics on the studied parameters. However, DPF materials have been found to have effects on pressure drop, CO and NOx. For DPF with SiC material, thermal losses are high at the beginning of regeneration, CO emissions are low when the AT (Aluminum Titanate) material used, and pressure drop is high for Crd (Cordierite) material. Aim: Chronic gastritis (CG), being one of the most common digestive diseases, is frequently underestimated both by patients and clinicians. However, CG developed as early as in childhood, and often accompanied by Helicobacter pylori contamination of antral mucus, has a persistent recurrent course in adult patients, complicated by mucosal transformations in the form of atrophy, metaplasia and dysplasia. CG can appear as an early stage in the morphogenesis of tumor growth. From this point of view, CG is worth noticing. Materials and Methods: A complex morphological study of antral mucosa in school children with atrophic gastritis has been performed using histochemistry, immunohistochemistry and a morphometric methods. Results: In atrophic gastritis we have revealed the decrease in the number of functionally mature cells responsible for the production of extracellular matrix and basal membrane components, the lack of which in paraepithelial localization results in the change of their special properties. In addition, sulphomucins, non-characteristic of stomach, overlapped on mucosa and in single foveolae have been found in atrophic gastritis. Conclusion: Permanent impairment in the conjugation of cell and tissue components in mucosa accompanied by the change of an epithelial layer synthetic function with the impaired physicochemical properties of gastric mucin results in intestinal metaplasia as early as in childhood. Bitkisel üretimde verimin artırılması ve biyotik-abiyotik baskı etmenleri nedeniyle oluşan ürün kayıpları biyoteknoloji destekli ıslah çalışmalarına ilgiyi artırmaktadır. Ancak bu çalışmalarda doku kültürü ve genetik mühendisliği araştırmalarının maliyeti oldukça yüksek olmakta ve bunun en büyük kısmını kullanılan kimyasal maddeler oluşturmaktadır. In vitro çalışmalarda eksplantlara besin maddesi sağlayan MS besin ortamı en çok kullanılan kimyasallardan biridir. Bu araştırmada, kallus kültürü ile bitki elde edilmesinde gerekli olan en ekonomik MS besin maddesi miktarının belirlenmesi amaçlanmıştır. Materyal olarak makarnalık Sarıçanak-98 ve ekmeklik Pehlivan buğday çeşitleri kullanılmış, kallus oluşumu için olgun embriyolar tohumlardan gevşetilerek 8 mg/l 2,4-D içeren sıvı ortamlarda kültüre alınmıştır. Bunu izleyen alt kültürlerde gelişen kalluslar, tam 4,405 g/l , yarım 2,202 g/l ve çeyrek 1,101 g/l dozlarda MS besin ortamı içeren petrilerde kültüre alınmıştır. Oluşan kallusların ağırlıkları 11 gün sonra tartılarak belirlenmiş ve 7 hafta sonunda elde edilen bitki sayıları saptanmıştır. Yapılan istatistiksel incelemeler sonucunda, kullanılan farklı MS dozlarının rejenerasyon kapasitesi ve elde edilen bitki sayısına etkisi olmadığı saptanmıştır. Kallus oluşumu aşamasında sıvı ortam ve sonraki aşamalarda çeyrek MS dozu kullanarak rejenere tam bitkiler elde edilebilmiştir. Buna göre, gen aktarım çalışmalarında sık aralıklarla yapılan alt kültürlerde MS besin ortamının % 75 oranında azaltılarak kullanılabileceği belirlenmiştir

The anther culture is one of the most important methods for producing doubled haploid plants and the efficiency of this method is influenced by several mentioned factors such as genotype and induction media. In this study, it was investigated that anther response of different F2 progenies of bread wheat (Triticum aestivum L.) hybrids using MN6 and P2 induction media. The results indicated that callus production, regeneration, green and albino plant numbers are higher on MN6 media than on P2 media for all genotypes. In addition to, all investigated parameters varied between genotypes. It can be said that the response of anthers depends mainly both the genotype and media, and the most suitable induction media for obtaining doubled haploid from our wheat hybrids was MN6 medium. However, it may be needed to develop other culture conditions for this population to utilize it in an actual breeding program. [ABSTRACT FROM AUTHOR]

In this study, it was aimed to assess effect of classical micro-suturing techniques combined with cyanoacrylate, used in nerve repair, on nerve regeneration in histopathological manner and to evaluate whether it could be safely used in peripheral nerve surgery. Adult Wistar-Albino female rats were used in this study. Animals were assigned into 4 groups for surgical processes as follows: Group 1: Control group (n=7); External circumferential neurolysis Group 2: Experiment group (n=7); Repair with 4 epineural sutures. Group 3: Experiment group (n=7); Repair with cyanoacrylate Group 4: Experiment group (n=7); combined group Histopathologically, it was seen that there was no vacuolar degeneration, foreign body type tissue reaction or fibrosis in the nerves of subjects in the control group. Moreover, it was found that there were findings of vacuolar degeneration, foreign body type tissue reaction and fibrosis in all experiments group in a similar manner. However, it was found that there was a statistically significant difference regarding grade results of vacuolar degeneration, fibrosis and foreign body type tissue reaction between control group and classical suturing, cyanoacrylate or combined groups (p<0.05). In conclusion, it has been thought that cyanoacrylate is a feasible method for peripheral nerve surgery; as it can be easily applied at conditions where surgical suturing is challenging; it reduces the trauma exposed by nerve endings; it provides easy identification of nerve endings during secondary repair, which are contaminated and could not be primarily repaired; it shortens the procedure time; it is cost-effective and; it easy to learn. Inflammation is a protective response of the host to harmful stimuli, such as microorganisms, damaged cells, chemical and physical irritants. A cascade of biochemical events propagates and matures the inflammatory response, involving the local vascular system, the immune system, and various cells within the injured tissue. Inflammation can be classified as either acute or chronic. Acute inflammation is the initial response of the body to harmful stimuli and is achieved by the increased movement of plasma and leukocytes. The classical signs of acute inflammation are pain, heat, redness, swelling, and loss of function. Prolonged inflammation, known as chronic inflammation, leads to a progressive shift in the type of cells present at the site of inflammation and is characterized by simultaneous destruction and healing of the tissue. Characteristics of the tissue and the injurious agent determine the outcome of inflammation, such as healing, fibrosis and leading to chronic inflammation. One of problems after peripheral nerve injury and surgery is scar tissue formation. Although the continuity of axons is restored, often the axonal viability and recovery is blocked by scarring. For preventing from scar formation silicone tubes are widely used. In addition, in the present study was used hyaluronic acid, which prevents adhesion. The aim of the present study is to evaluate electrophysiological results on nerve regeneration obtained after incision and reconstruction of rat sciatic nerve using only silicone tubes and silicone tubes filled with hyaluronic acid. Electrophysiological measurements were performed at 30th and 60th days after surgery. The amplitude and latency of the compound muscle action potentials and nerve conduction velocities were calculated for each rat. Although sciatic nerve conduction velocities in all operated groups not reached control values, amplitude and latency values in group with silicone tube and in group with silicone tube filled with hyaluronic acid statistically achieved control values on day 60. Silicone tubes filled with hyaluronic acid had better results compared to the silicone tubes without hyaluronic acid. Hence, the use of silicone tubes and hyaluronic acid for the reconstruction of nerve injuries seems to give good results on axonal regeneration. [ABSTRACT FROM AUTHOR]

A rapid protocol using shoot tip explants for micropropagation of Diyarbakır watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai] types Sürme, Beyazkış and Karakış was achieved. Shoot tips from 5-day-old in vitro germinated seedlings were cultured on shoot regeneration medium for 3 weeks. The effects of the different concentrations of benzyl adenine (BA) and carbohydrate types on shoot proliferation were examined. The results confirmed that a range of 0.5-1.0 mg/L of BA was almost equally effective in promoting the shoot length of cultures in the 3 genotypes. Sucrose, in the presence of BA, was superior to other carbohydrates for the 3 genotypes studied in terms of the number of proliferated shoots and the average shoot length obtained. The highest percentage of rooting was achieved when medium supplemented with 1.0 mg/L of indole-3-butyric acid (IBA) was used for the 3 genotypes. The highest frequency of acclimatized plantlets for the 3 genotypes was 85%, 85%, and 90%, respectively, in sterile compost, when the shoots of Beyazkış, Karakış, and Sürme were rooted in the IBA-supplemented rooting treatments. The Sürme genotype produced a significantly higher yield and mean fruit weight than the Beyazkış and Karakış genotypes in both direct-seeded and transplanted watermelons. The main soluble sugar of the experimental types was separated, identified, and quantified using high performance liquid chromatography (HPLC). Fructose was found to be the most abundant sugar and was highly detected in Beyazkış for the seeded watermelon and in Karakış for the transplanted watermelon. This demonstrates that in vitro propagation can be used to produce high quality diploid Diyarbakır watermelon for use in breeding lines. [ABSTRACT FROM AUTHOR]

Motor vehicles have a great contribution to air pollution, which make the emission control technologies compulsory to be developed. The high efficiency of diesel engines, their low operating costs, high durability and reliability have provided them with a leadership role in the heavy-duty vehicle market. The most important pollutants formed at the exhaust gases of diesel engines are particulate matters (PM), nitrogen oxides (NOx), hydrocarbons (HC), and carbon monoxides (CO). The control of diesel vehicles' exhaust emission has become more critical because of the spreading use of diesel engines. This encourages the researches on engine technologies, fuels, and the use of post combustion devices, an example of which is particulate filters. Diesel particle filters (DPFs) are one of the most technically feasible solutions to reduce PM. A diesel particulate filter is a device designed to remove diesel particulate matter emissions from the exhaust gas of a diesel engine. The soot collected by DPFs needs to be removed to avoid excessive fuel penalty and damage to the engine and the filter. The removal of the soot, termed "regeneration," is a rather complex process. In vitro morphogenesis of Cucumis melo var. flexuosus (L.) Naudin was studied by direct organogenesis. Induction of adventitious buds was obtained from distal and proximal parts of the cotyledon incubated on Murashige and Skoog (1962) medium containing different concentrations of 6-benzyladenine (BA) (0.0, 0.5, 1.0, and 2.0 mg L-1) and indole-3-acetic acid (IAA) (0.0, 0.1, and 0.5 mg L-1). The best organogenesis was obtained from the proximal part of the cotyledon on medium containing 0.5 mg L-1 BA and 0.5 mg L-1 IAA (88%). This concentration was used for histological analysis. Less regeneration was obtained on media containing 1.0 mg L-1 BA (75%) or 1.0 mg L-1 BA and 0.1 mg L-1 IAA (60%). Plant recovery was much greater from proximal rather than distal explants. A callus formed on almost every explant on media supplemented with BA. Histological analyses showed that the formation of early shoot apical meristems was observed in 14-day-old tissues and meristematic structures were seen in 17-day-old tissues. Two different types of explant (proximal and flamingo-bill) from Emphasis seedlings, a hybrid cultivar of bottle gourd (Lagenaria siceraria (Molina) Stand.), germinated under dark and light conditions were cultured on 9 regeneration MS media containing various combinations of BA (0, 1.0, and 2.0 mg l-1) and IAA (0, 0.25, and 0.5 mg l-1). Comparison of the explant types showed that the flamingo-bill type explant had better shoot formation than did the proximal explant. The MS medium containing 1 mg l-1 of BA was optimal for shoot formation capacity when flamingo-bill type explants germinated in the dark (44%) and light (36%) were used. Histological analysis showed that explant cell division began after 3 days in regeneration medium and formation of primordium was observed in the tissues in culture between days 5 and 7. Differentiation of meristimatic structures was first observed after 9 days and development completed after 9-12 days in the culture. [ABSTRACT FROM AUTHOR]
