Abstract

This research was carried out to determine the in vitro regeneration abilities of tulip leaf explants. In the study, both sterile leaf explants obtained by culturing sterilized onions and leaf explants obtained by sterilizing leaves obtained from the onions germinated in pots were used. For this purpose MS medium containing 2.4-D (2 mg.l-1), TDZ (2 mg.l-1), Kinetin (2 mg.l-1), NAA (2 mg.l-1), 2.4-D+Kinetin (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1+ 2 mg.l-1), İAA+TDZ (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), İAA+BAP (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), NAA+Kinetin (1 mg.l-1+ 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1), NAA+TDZ (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1) and 2.4-D+TDZ (1 mg.l-1 + 1 mg.l-1 and 2 mg.l-1 + 2 mg.l-1) were used in the study. As a result of subcultures, 6% callus formation was detected in MS medium containing 2.4-D (2 mg.l-1) and NAA (2 mg.l-1) hormone, which is one of the subcultures of leaves formed from the in vitro germination of Purple Prince Tulip bulbs. The highest rate of regeneration was determined as 10% callus formation in the medium containing Kinetin (2 mg.l-1). As a result, while callus regeneration was achieved in 3 different media containing 2.4-D (2 mg.l-1), NAA (2 mg.l-1) and Kinetin (2 mg.l-1) hormones among subcultured media, no regeneration was achieved in the other mediums.
