Gα proteins as part of heterotrimeric G proteins are molecular switches essential for G protein-coupled receptor- mediated intracellular signaling. The role of the Gα subunits has been examined for decades with various guanine nucleotides to elucidate the activation mechanism and Gα protein-dependent signal transduction. Several approaches describe fluorescent ligands mimicking the GTP function, yet lack the efficient estimation of the proteins' GTP binding activity and the fraction of active protein. Herein, we report the development of a reliable fluorescence anisotropy-based method to determine the affinity of ligands at the GTP-binding site and to quantify the fraction of active Gαi1 protein. An advanced bacterial expression protocol was applied to produce active human Gαi1 protein, whose GTP binding capability was determined with novel fluorescently labeled guanine nucleotides acting as high-affinity Gαi1 binders compared to the commonly used BODIPY FL GTPγS. This study thus contributes a new method for future investigations of the characterization of Gαi and other Gα protein subunits, exploring their corresponding signal transduction systems and potential for biomedical applications.

The potential of designing irreversible alkyne-based inhibitors of cysteine cathepsins by isoelectronic replacement in reversibly acting potent peptide nitriles was explored. The synthesis of the dipeptide alkynes was developed with special emphasis on stereochemically homogenous products obtained in the Gilbert-Seyferth homologation for C-C triple bond formation. 23 dipeptide alkynes and 12 analogous nitriles were synthesized and investigated for their inhibition of cathepsins B, L, S and K. Numerous combinations of residues in the P1 and P2 positions as well as terminal acyl groups, allowed for the derivation of extensive structure-activity relationships, which were rationalized by computational covalent docking for selected examples. The determined inactivation constants of the alkynes at the target enzymes span a range of more than three orders of magnitude (3-10,133 M-1s-1). Notably, the selectivity profiles of alkynes do not necessarily reflect those of the nitriles. Inhibitory activity at the cellular level was demonstrated for selected compounds.

The intentional binding of radioligands to albumin gains increasing attention in the context of radiopharmaceutical cancer therapy as it can lead to an enhanced radioactivity uptake into the tumor lesions and thus, to a potentially improved therapeutic outcome. However, the influence of the radioligand’s albumin binding affinity on the time profile of tumor uptake depending on the particular albumin-binding affinity have been only partly addressed so far. Based on the previously identified Nε-4-(4-iodophenyl)butanoyl-lysine scaffold, we designed “clickable” lysine-derived albumin binders (cLABs) and determined their dissociation constants towards albumin by novel assay methods. Structure-activity relationships were derived and selected cLABs were applied for the modification of the somatostatin receptor subtype-2 ligand (Tyr³)octreotate (TATE). These novel conjugates were radiolabeled with copper-64 and subjected to a detailed in vitro and in vivo radiopharmacological characterization. Overall, the results of this study provide an incentive for further investigations of albumin binders for applications in endoradionuclide therapies.

Tissue transglutaminase (TGase 2) is proposed to be important for biomaterial–tissue interactions due to its presence and versatile functions in the extracellular environment. TGase 2 catalyzes the cross-linking of proteins through its Ca2+-dependent acyltransferase activity. Moreover, it enhances the interactions between fibronectin and integrins, which in turn mediates the adhesion, migration, and motility of the cells. TGase 2 is also a key player in the pathogenesis of fibrosis. In this study, we investigated whether TGase 2 is present at the biomaterial–tissue interface and might serve as an informative biomarker for the visualization of tissue response toward gelatin-based biomaterials. Two differently cross-linked hydrogels were used, which were obtained by the reaction of gelatin with lysine diisocyanate ethyl ester. The overall expression of TGase 2 by endothelial cells, macrophages, and granulocytes was partly influenced by contact to the hydrogels or their degradation products, although no clear correlation was evidenced. In contrast, the secretion of TGase 2 differed remarkably between the different cells, indicating that it might be involved in the cellular reaction toward gelatin-based hydrogels. The hydrogels were implanted subcutaneously in immunocompetent, hairless SKH1-Elite mice. Ex vivo immunohistochemical analysis of tissue sections over 112 days revealed enhanced expression of TGase 2 around the hydrogels, in particular at days 14 and 21 post-implantation. The incorporation of fluorescently labeled cadaverine derivatives for the detection of active TGase 2 was in accordance with the results of the expression analysis. The presence of an irreversible inhibitor of TGase 2 led to attenuated incorporation of the cadaverines, which verified the catalytic action of TGase 2. Our in vitro and ex vivo results verified TGase 2 as a potential biomarker for tissue response toward gelatin-based hydrogels. In vivo, no TGase 2 activity was detectable, which is mainly attributed to the unfavorable physicochemical properties of the cadaverine probe used.

An O-methyltyrosine-containing azadipeptide nitrile was synthesised and investigated for its inhibitory activity towards cathepsins L, S, K, and B. Labelling with carbon-11 was accomplished by reaction of the corresponding phenolic precursor with [11 C]methyl iodide starting from cyclotron-produced [11 C]methane. Radiopharmacological evaluation of the resulting radiotracer in a mouse xenograft model derived from a mammary tumour cell line by small animal PET imaging indicates tumour targeting with complex pharmacokinetics. Radiotracer uptake in the tumour region was considerably lower under treatment with the nonradioactive reference compound and the epoxide-based irreversible cysteine cathepsin inhibitor E64. The in vivo behaviour observed for this radiotracer largely confirms that of the corresponding 18 F-fluoroethylated analogue and suggests the limited suitability of azadipeptide nitriles for the imaging of tumour-associated cysteine cathepsins despite target-mediated uptake is evidenced.

The lysyl oxidase family of enzymes (LOXs) catalyze oxidative deamination of lysine side chains on collagen and elastin to initialize cross-linking that is essential for the formation of the extracellular matrix (ECM). Elevated expression of LOXs is highly associated with diverse disease processes. To date, the inability to detect total LOX catalytic function in situ has limited the ability to fully elucidate the role of LOXs in pathobiological mechanisms. Using LOXL2 as a representative member of the LOX family, we developed an in situ activity assay by utilizing the strong reaction between hydrazide and aldehyde to label the LOX-catalyzed allysine (-CHO) residues with biotin-hydrazide. The biotinylated ECM proteins are then labeled via biotin-streptavidin interaction and detected by fluorescence microscopy. This assay detects the total LOX activity in situ for both overexpressed and endogenous LOXs in cells and tissue samples and can be used for studies of LOXs as therapeutic targets.

In a previous study, EphB4 was demonstrated to be a positive regulator of A375-melanoma growth but a negative regulator of tumor vascularization and perfusion. To distinguish between EphB4 forward and ephrinB2 reverse signaling, we used the commercially available EphB4 kinase inhibitor NVP-BHG712 (NVP), which was later identified as its regioisomer NVPiso. Since there have been reported significant differences between the inhibition profiles of NVP and NVPiso, we compared the influence of NVP and NVPiso on tumor characteristics under the same experimental conditions. Despite the different inhibitory profiles of NVP and NVPiso, the comparative study conducted here showed the same EphB4-induced effects in vivo as in the previous investigation. This confirmed the conclusion that EphB4-ephrinB2 reverse signaling is responsible for increased tumor growth as well as decreased tumor vascularization and perfusion. These results are further substantiated by microarrays showing differences between mock-transfected and EphB4-transfected (A375-EphB4) cells with respect to at least 9 angiogenesis-related proteins. Decreased expression of vascular endothelial growth factor (VEGF), angiotensin 1 (Ang-1), and protein kinase B (Akt/PKB), together with the increased expression of tissue inhibitor of metalloproteinase-1 (TIMP-1) and transforming growth factor beta-2 (TGF-β2), is consistent with the impaired vascularization of A375-EphB4 xenografts. The cell surface receptor claudin-4 (Cld-4) is upregulated in various tumours and represents an important emerging target for both diagnosis and treatment of solid tumours of epithelial origin. The C-terminal fragment of the Clostridium perfringens enterotoxin cCPE290-319 appears as a suitable ligand for targeting Cld-4. The synthesis of this 30mer peptide was attempted via several approaches, which has revealed sequential SPPS using three pseudoproline dipeptide building blocks to be the most efficient one. Labelling with fluorine-18 was achieved on solid phase using N-succinimidyl 4-[18F]fluorobenzoate ([18F]SFB) and 4-[18F]fluorobenzoyl chloride as 18F-acylating agents, which was the most advantageous when [18F]SFB was reacted with the resin-bound 30mer containing an N-terminal 6-aminohexanoic spacer. Binding to Cld-4 was demonstrated via surface plasmon resonance using a protein construct containing both extracellular loops of Cld-4. In addition, cell binding experiments were performed for 18F-labelled cCPE290-319 with the Cld-4 expressing tumour cell lines HT-29 and A431 that were complemented by fluorescence microscopy studies using the corresponding fluorescein isothiocyanate-conjugated peptide. The 30mer peptide proved to be sufficiently stable in blood plasma. Studying the in vivo behaviour of 18F-labelled cCPE290-319 in healthy mice and rats by dynamic PET imaging and radiometabolite analyses has revealed that the peptide is subject to substantial liver uptake and rapid metabolic degradation in vivo, which limits its suitability as imaging probe for tumour-associated Cld-4.

A reliable solution-phase synthesis of the water-soluble dipeptidic fluorogenic transglutaminase substrate Z-Glu(HMC)-Gly-OH is presented. The route started from Z-Glu-OH, which was converted into the corresponding cyclic anhydride. This building block was transformed into the regioisomeric α- and γ-dipeptides. The key step was the esterification of Z-Glu-Gly-OtBu with 4-methylumbelliferone. The final substrate compound was obtained in an acceptable yield and excellent purity without the need of purification by RP-HPLC. The advantage of this acyl donor substrate for the kinetic characterisation of inhibitors and amine-type acyl acceptor substrates is demonstrated by evaluating commercially available or literature-known irreversible inhibitors and the biogenic amines serotonin, histamine and dopamine, respectively.

Tissue transglutaminase (TGase 2) is the most abundantly expressed enzyme of the transglutaminase family and involved in a large variety of pathological processes, such as neurodegenerative diseases, disorders related to autoimmunity and inflammation as well as tumor growth, progression and metastasis. As a result, TGase 2 represents an attractive target for drug discovery and development, which requires assays that allow for the characterization of modulating agents and are appropriate for high-throughput screening. Herein, we report a fluorescence anisotropy-based approach for the determination of TGase 2's transamidase activity, following the time-dependent increase in fluorescence anisotropy due to the enzyme-catalyzed incorporation of fluorescein- and rhodamine B-conjugated cadaverines 1-3 (acyl acceptor substrates) into N,N-dimethylated casein (acyl donor substrate). These cadaverine derivatives 1-3 were obtained by solid-phase synthesis. To allow efficient conjugation of the rhodamine B moiety, different linkers providing secondary amine functions, such as sarcosyl and isonipecotyl, were introduced between the cadaverine and xanthenyl entities in compounds 2 and 3, respectively, with acyl acceptor 3 showing the most optimal substrate properties of the compounds investigated. The assay was validated for the search of both irreversible and reversible TGase 2 inhibitors using the inactivators iodoacetamide and a recently published L-lysine-derived acrylamide and the allosteric binder GTP, respectively. In addition, the fluorescence anisotropy-based method was proven to be suitable for high-throughput screening (Z' factor of 0.86) and represents a non-radioactive and highly sensitive assay for determining the active TGase 2 concentration.

The potential of papain-like cysteine proteases, such as cathepsin B, as drug discovery targets for systemic human diseases has prevailed over the past years. The development of potent and selective low-molecular cathepsin B inhibitors relies on the detailed expertise on preferred amino acid and inhibitor residues interacting with the corresponding specificity pockets of cathepsin B. Such knowledge might be obtained by mapping the active site of the protease with combinatorial libraries of peptidic substrates and peptidomimetic inhibitors. This review, for the first time, summarizes a wide spectrum of active site mapping approaches. It considers relevant X-ray crystallographic data and discloses propensities towards favorable protein-ligand interactions in case of the therapeutically relevant protease cathepsin B.

Until recently, alkynes were considered bioinert. Thus, they are popular reaction partners in bioorthogonal click reactions in vitro and in vivo. Despite the virtual chemical inertness of the alkyne moiety, two research groups observed the irreversible inhibition of a cysteine protease by an alkyne functionalised substrate derivative: both EKKEBUS et al. and SOMMER et al. independently described the unexpected inactivation of de-ubiquitinating enzymes by propargylated ubiquitin or ubiquitin-like modifiers bearing propargylamine in place of C-terminal glycine [1, 2]. We intended to harness that finding for the design of inhibitor-based probes for the imaging of tumour-associated cysteine proteases. Cysteine cathepsins play an important role in tumour progression. In particular, cathepsin B is involved in a variety of tumour progression-related processes and an elevated extracellular levels are linked to increased malignancy and poor prognosis [3]. Therefore, this enzyme represents a promising target for the therapy and imaging of tumours. GREENSPAN et al. reported a potent and highly selective, dipeptidyl nitrile-based cathepsin B inhibitor (N-[2-[(3-Carboxyphenyl)methoxy]-1(S)-cyanoethyl]-3-methyl-N2-(2,4-difluorobenzoyl)-L-phenylalaninamide) [4]. Based on that lead compound, cathepsin B-targeting dipeptide alkynes were designed by isoelectronic replacement of the nitrile nitrogen atom by by a methine group and consecutive variation of the 2,4-difluorobenzoyl and (3-carboxybenzyl)oxymethyl residue. Formation of the C-C triple bond by reaction of the corresponding open-chain serine-derived aldehyde with the Bestmann-Ohira reagent was accompanied by partial enantiomerisation. Therefore, the synthesis was performed via Garner’s aldehyde, which accounted for high stereochemical purity of the final compounds. The inhibitory potential was investigated against cathepsins B, S, L and K. The most potent compound exhibited irreversible inhibition of cathepsin B with an inactivation constant (kinact/KI=771 M-1s-1). Values for cathepsins L, S and K were significantly lower; no irreverisible ihibition was observed for cathepsin K. In addition, inhibition of cathepsin B activity in human glioblastoma cell lysates and living cells has been demonstrated. Based on these promising results, dipeptidyl alkynes have the potential to become a valuable tool for imaging due to the expected low activity towards other cysteine proteases. In our understanding of the many drivers of malignant progression and cancer metastasis, proteases are increasingly drawn into the spotlight as crucial agents in cancer angiogenesis, invasion, and metastasis [1]. Elevated activities of multiple members of the cathepsin family have been shown to correlate with increased metastasis and high therapy resistance [2, 3]. Especially high expression levels of extracellular cathepsin B (CTB) indicate poor prognosis in neoplastic diseases, making CTB an interesting target for activity-based molecular imaging in diagnostics as well as in therapy monitoring for personalised therapies. It is our aim to develop such a probe by combination of a polyarginine-based, activatable cell penetrating peptide (ACPP), as first described by R. Tsien, and an optimised endopeptidase substrate for CTB [4]. Substrate optimisation proofed to be challenging as two entirely opposite factors needed to be balanced – high stability against serum proteases to prevent premature cleavage of the activation sequence, while retaining efficient and specific endoproteolytic cleavability by CTB. We have generated a CTB-endoprotease substrate by C-terminally elongating the CTB carboxydipeptidase substrate Abz GIVR*AK(Dnp) OH (Abz - aminobenzoyl, Dnp – dinitrophenyl, * – cleavage site), described by Cotrin et al. in 2004, to the octapeptide Abz GIVR*AK(Dnp)GX CONH2, which could be used as activation site in the final ACPP [5]. Introduction of any amino acid other than glycine at the P4’ position resulted in hysteretic kinetics for the CTB-catalysed hydrolysis of the octapeptides, which might indicate the displacement of the occluding loop from the active site upon interaction with the substrates. Using LC-ES-MS-based analysis of serum-incubated substrates, the positions P1 and P3’ were determined to be primary determinants of serum stability. After suppression of the P3’ instability by Nα-methylation and optimisation within the positions P1-P3, we were able to increase serum half-life from < 5 min to > 1440 min under concomitant improvement of kinetic substrate efficiency towards CTB. Currently, the substrate is optimised towards CTB-specificity within the cathepsin protease family. Additionally, cell uptake studies of a fluorescently labelled ACPP using the optimised CTB-endoprotease substrate sequence are ongoing. Using this fluorescent probe, we plan to study cell uptake, in vivo stability and initial biodistribution. Furthermore, an ACPP conjugated to radiometal-chelating entities is currently prepared, which will allow for PET imaging in vivo. Small glutamate-containing peptides bearing coumarin derivatives as fluorescent leaving groups attached to the γ-carboxylic acid group of the Glu residue were synthesised and investigated with regard to their potential to act as substrates for transglutaminase 2 (TGase 2). Their synthesis was accomplished by an efficient solid-phase approach. The excellent water solubility of the compounds enabled their extensive kinetic characterisation in the context of TGase 2-catalysed hydrolysis and aminolysis. The influence of the coumarin skeleton's substitution pattern on the kinetic properties was studied. Derivatives containing 7-hydroxy-4-methylcoumarin (HMC) revealed properties superior to those of their 7-hydroxycoumarin counterparts; analogous amides are not accepted as substrates. Z-Glu(HMC)-Gly-OH, which exhibited the best substrate properties out of the investigated derivatives, was selected for representative kinetic characterisation of acyl acceptor substrates and irreversible inhibitors.

High activity of the cysteine protease cathepsin B correlates with increased metastasis, therapy resistance, and a generally poor prognosis in neoplastic diseases [1,2] Therefore, the development of substrate-based radiotracers for in vivo imaging of this protease will be beneficial towards better diagnosis and improved therapy regimens. We aim at developingsuch a probe by combination of a polyarginine-based, activatable cell penetrating peptide, as described by Tsien and others, and newly optimised endopeptidase substrates for cathepsin B [3]. A unique feature of cathepsin B within the cysteine cathepsin family is the occluding loop, a flexible element consisting of 20 amino acids (aa), which is able to cover the active site of the enzyme and thereby regulates the substrate specificity in a pH-dependent manner [1,2]. This phenomenon is mediated by two histidine residues within the occluding loop, which play a crucial role in positioning a potential substrate for carboxydipeptidolytic cleavage. Starting point for the development of endopeptidase substrates was the carboxydipeptidase substrate Abz‑GIVRAK(Dnp)‑OH (Abz - aminobenzoyl, Dnp - dinitrophenyl) described by Cotrin et al. in 2004, with the fluorophores Abz and Dnp constituting a FRET pair to enable fluorometric detection of the proteolytic cleavage [4]. Two structural changes were introduced to convert the exopeptidase substrate into an endopeptidase substrate for later use as an activator sequence in an aCPP: Firstly, C‑terminal amidation was performed to interrupt the interaction between the terminal carboxyl group and the two His residues within the occluding loop. This resulted in a reduced specificity constant (kcat/Km = 1179 mM‑1s‑1 vs. 139 mM‑1s‑1), indicating attenuated electrostatic/hydrogen bond interactions. Secondly, the peptide sequence was C‑terminally elongated to test longer substrates. Interestingly, the C‑terminal elongation resulted in a hysteretic progress of the enzymatic cleavage, likely a result of the steric displacement of the occluding loop. To evaluate the influence of size, polarity and charge of the C‑terminal aa on the enzymatic hysteresis, 14 variations of this position were synthesized and analysed for their kinetic parameters. The peptide Abz‑GIVRAK(Dnp)G‑V‑NH2 was determined to exhibit the highest specificity constant (kcat/Km = 260 mM‑1s‑1). The optimised octapeptide sequence is currently inserted as recognition site in a fluorophore-labelled aCPP for monitoring cathepsin B-mediated activation and subsequent cellular uptake, and for investigating kinetics and in vitro stability analysis in future experiments. Radiopharmacological investigations of [(18)F]NS14490 have proven that this radiotracer could be a potential PET radiotracer for imaging of alpha7 nicotinic acetylcholine receptor particularly with regard to vulnerable plaques of diseased vessels. For further optimisation of the previously automated one-pot radiosynthesis of [(18)F]NS14490 using a tosylate precursor, precursors with other leaving groups (nosylate and mosylate) were synthesized and compared with the tosylate with respect to their reactivities towards [(18)F]fluoride. The use of these different precursors resulted in comparable labelling yields of [(18)F]NS14490. A novel mosylate precursor was synthesized and evaluated, which has revealed a higher stability during a storage period of five months compared to the corresponding tosylate and nosylate.

This article evaluates a patent application of the company Medivir (SE/UK) describing the synthesis of dipeptide-derived alpha alpha-ketoamides containing a propylene glycine moiety in P1 as selective inhibitors of cathepsin S for the potential treatment of various systemic human diseases such as several autoimmune diseases, MS, rheumatoid arthritis, endometriasis and chronic pain. The claims of the patent are discussed in light of recent results in the field of cathepsin S research.

The cyclic dodecapeptides tychonamide A (1) and B (2) isolated from the methanolic extract of the cyanobacterium Tychonema sp. contain the novel β-amino acid 3-amino-2,5,7-trihydroxy-8-phenyloctanoic acid (Atpoa). Compounds 1 and 2 have cytotoxic activity towards cancer cell linesin a monolayer assay with mean IC50 values of 0.9 and 3.3 μg mL–1, respectively. Compound 2 was shown to be active against tumor cell suspensions derived fromsolid tumor xenografts in a clonogenic assay (mean IC50 2.4 μg mL–1). Papain-like cysteine proteases bear an enormous potential as drug discovery targets for both infectious and systemic human diseases. The considerable progress in this field over the last two decades has also raised interest in the visualization of these enzymes in their native context, especially with regard to tumor imaging. After a short introduction to structure and general functions of human cysteine cathepsins, we highlight their importance for drug discovery and development and provide a critical update on the current state of knowledge towards their involvement in tumor progression, with a special emphasis on their role in therapy response. In accordance with a radiopharmaceutical point of view, the main focus of this review article will be the discussion of recently developed fluorescence and radiotracer-based imaging agents together with related molecular probes.

Cysteine cathepsins are lysosomal cysteine proteases which play roles in many physiological processes. Cathepsin F is predominantly expressed in macrophages. Major histocompatibility complex class II molecules (MHC-II) are expressed by antigen-presenting cell types including macrophages, B cells, and dendritic cells. The cleavage of the invariant chain is the key event in the pathway of MHC-II complexes. Cathepsin S was described as the major processing enzyme of the invariant chain, but it was shown that cathepsin F can adopt its role in cathepsin S deficient mice.[1] Low molecular weight inhibitors for cathepsin F have not been investigated so far. We have chosen the dipeptide nitrile[2] chemotype to develop covalent-reversible inhibitors for this target. An active site mapping with a library of 52 nitrile-based cathepsin inhibitors was performed at human cathepsin F to draw structure-activity relationships. With the kinetic data in hand, new compounds with optimized residues in P1, P2 and P3 position were synthesized and evaluated. With all dipeptide nitriles including the newly synthesized derivatives, a 3D activity landscape was generated to visualize similarity-activity relationships of this series of cathepsin F inhibitors. Considerable evidence for the implication of tissue transglutaminase (TGase 2) in a variety of pathological processes, such as neurodegenerative diseases, disorders related to autoimmunity and inflammation as well as tumor progression, has been revealed over the recent years. This renders TGase 2 attractive for developing agents which allow the enzyme’s targeting for both therapeutic and imaging purposes. The development of such molecules requires the establishment of reliable methods to assess the interaction with TGase 2, which can be done most conveniently in continuous kinetic assays. Several assays have been published over the last decades to determine TGase 2 activity, with only very few using the method of fluorescence anisotropy. Measurement of fluorescence anisotropy offers a better signal to noise ratio than other techniques, such as those based solely on fluorescence emission and does not need washing or separation of unbound fluorescent substance. Here, we report a fluorescence anisotropy-based approach for the determination of TGase 2’s transamidase activity, established and validated by using fluorescein- and rhodamine B-labeled cadaverines as acyl acceptor substrates. The synthesis of the cadaverine derivatives has been accomplished in a solid-phase approach. To allow efficient conjugation of the rhodamine B moiety, different linkers providing secondary amine functions have been introduced between the cadaverine and xanthenyl entities. The increase in fluorescence anisotropy resulting from covalent binding of the relatively small cadaverine derivatives to the much larger acyl donor substrate N,N-dimethylated casein was followed over time and enzyme activities were derived thereof. The assay was found to be highly reproducible and shows no background signal in the absence of the enzyme for all synthesized cadaverine derivatives. After characterization of the enzyme–substrate interaction by determination of the Michaelis constants, Km, and the maximum velocities of substrate conversion, Vmax, the assay was validated for screening of non-covalent and covalent inhibitors by using the literature-known substances GTP and iodoacetamide, respectively, as well as a recently reported L-lysine acrylamide derivative.

Transglutaminases and polyamine transporters are promising targets for functional imaging of tumours. Therefore, our aim is to synthesise polyamine-based radiotracers that allow the in vivo imaging of the aforementioned targets by positron emission tomography (PET). Labelling with the radionuclide fluorine-18 can be accomplished via attaching a [18F]fluorobenzoyl group with the prosthetic labelling reagent N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB). To access the required non- radioactive analogues, a solid-phase synthesis was developed that enables selective fluorobenzoylation at distinct amino groups of various polyamines (e.g. cadaverine, spermidine, spermine) on the basis of a recently described synthetic concept for the selective functionalisation of polyamines. The established route can be directly applied to synthesise the 18F-labelled analogues. The mono-fluorobenzoylated polyamines were obtained by solidphase synthesis of the corresponding oxopolyamines and subsequent reduction of the amide bond with BH3-THF. By applying Dde and Boc as orthogonal protecting groups and taking advantage of the selective reaction of 2-acetyldimedone with primary amino groups in the presence of secondary amines, the selective fluorobenzoylation (FBz) of different amino groups becomes possible. Additionally, the selective mono-fluorobenzylation (FBn) of selected diamines by reaction with 4-fluorobenzaldehyde and subsequent reduction of the resulting imine using sodium triacetoxyborohydride was performed. Based on the established methodology, the following compounds among others were obtained in good yields: N-FBzcadaverine, N-FBn-cadaverine, N1-FBz- spermidine, N4-FBz-spermidine, N8-FBz-spermidine and N1-FBz-spermine. Furthermore, the naturally occurring diamine cadaverine was conjugated to different reporter groups such as biotin. The identity of the compounds was confirmed by NMR spectroscopy and mass spectrometry. The kinetic parameters towards transglutaminase 2-catalysed acyl transfer were determined for selected compounds with an in-house fluorimetric assay using the fluorogenic acyl donor Cbz–Glu(HMC)–Gly–OH.

Objectives: Lysyl oxidase (LOX, EC 1.4.3.13) and its family members LOX-like 1-4 are copper-dependent matrixmodifying enzymes [1]. The expression of LOX is elevated in many human cancers, including breast cancer and correlates with tissue hypoxia. The enzyme plays a critical role in breast cancer metastasis [2]. The goal of the current study was to target LOX with fluorescent and radiolabeled oligopeptides to visualize LOX in preclinical models of breast cancer. Methods: mRNA expression of all 5 LOX family members was analyzed by gene expression microarray analysis on samples from 176 breast cancer patients. The peptidic substrate GGGDPKGGGGG was selected to target LOX [3]. The peptide was labeled with either FITC for confocal microscopy experiments or with the positron emitter fluorine-18 for molecular imaging in vivo using positron emission tomography (PET) (Figure 1). The preclinical breast cancer models utilized were the murine breast cancer cell line EMT-6 and xenografts of MCF-7 and MDAMB-231. Results: Immunofluorescence with a LOX-specific antibody confirmed that LOX protein expression is enhanced in hypoxic EMT-6 cells. FITC-labeled oligopeptide binds to several cell compartments of EMT6 cells under hypoxic conditions. After injection of 18F-labeled oligopeptide, radioactivity uptake was visible in all three breast cancer models in vivo with SUV5min values of: 0.70±0.07 (n=3) in EMT-6, 0.57±0.01 (n=3) in MCF-7 and 0.68 (n=2) in MDA-MB-231. The following continuous washout of radioactivity led to SUV60min values of: 0.18±0.03 (n=3) in EMT-6, 0.14±0.02 (n=3) in MCF-7 and 0.13 (n=2) in MDA-MB-231. Tumor uptake was reduced by pre-dosing with the irreversible LOX inhibitor BAPN 4 h and 24 h prior to injection of the radiotracer. Conclusions: These data support further investigations towards the development of LOX-binding peptides as molecular probes for imaging of LOX expression in breast cancer.
