Abstract: International audience; Abstract In this paper,we presentMOSAIIC, an agent-based model to simulate the road traffic of a city in the context of a catastrophic event.Whether natural (cyclone, earthquake, flood) or human (industrial accident) in origin, catastrophic situations modify both infrastructures (buildings, road networks) and human behaviors, which can have a huge impact on human safety. Because the heterogeneities of human behaviors, of land-uses and of network topology have a great impact on the traffic flows, the agent-based modeling is particularly adapted to this subject. In this paper,we focus on the new traffic model itself: the way geographical data is used to build a network, the various behaviors of our agents, from the individual to the collective level.

Abstract: Thumbnail image of graphical abstract PC12 cells are used to study the signaling mechanisms underlying the neurotrophic and neuroprotective activities of pituitary adenylate cyclase-activating polypeptide (PACAP) and nerve growth factor (NGF). Previous microarray experiments indicated that serpinb1a was the most induced gene after 6 h of treatment with PACAP or NGF. This study confirmed that serpinb1a is strongly activated by PACAP and NGF in a time-dependent manner with a maximum induction (~ 50-fold over control) observed after 6 h of treatment. Co-incubation with PACAP and NGF resulted in a synergistic up-regulation of serpinb1a expression (200-fold over control), suggesting that PACAP and NGF act through complementary mechanisms. Consistently, PACAP-induced serpinb1a expression was not blocked by TrkA receptor inhibition. Nevertheless, the stimulation of serpinb1a expression by PACAP and NGF was significantly reduced in the presence of extracellular signal-regulated kinase, calcineurin, protein kinase A, p38, and PI3K inhibitors, indicating that the two trophic factors share some common pathways in the regulation of serpinb1a. Finally, functional investigations conducted with siRNA revealed that serpinb1a is not involved in the effects of PACAP and NGF on PC12 cell neuritogenesis, proliferation or body cell volume but mediates their ability to block caspases 3/7 activity and to promote PC12 cell survival. Pituitary adenylate cyclase-activating polypeptide (PACAP) and nerve growth factor (NGF) induce a strong increase in serpinb1 a expression in PC12 cells. Functional investigations revealed that this increase in serpinb1a does not affect cell proliferation or differentiation but inhibits caspase 3 activity and promotes cell survival., link_to_OA_fulltext

Abstract: International audience; Cryptosporidiosis is a common disease in children and immunodeficient individuals. In 2006, a national network was set up on the surveillance of human cryptosporidiosis in France. Since January 2015, the 41 tertiary care hospitals and the 3 private laboratories of the French National Network on the surveillance of human cryptosporidiosis have been able to declare confirmed cases of cryptosporidiosis online. Between 2015 and 2017, 210 cases of cryptosporidiosis were declared in immunodeficient patients in France; Cryptosporidium parvum and Cryptosporidium hominis represented 66% and 22% of cases, respectively. A peak was observed in autumn. Cryptosporidiosis occurred mainly in a context of solid organ transplantation (SOT) (49%) and of HIV infection (30%). In SOT recipients, cryptosporidiosis appeared more frequently in the first 6 months post transplantation. Regarding cases declared in SOT recipients, mycophenolate mofetil was used in 68%. A mortality rate of 6% was observed. Present results underline the importance of screening for cryptosporidiosis in immunocompromised patients suffering from diarrhea, especially in the course of major cell mediated immunodeficiency or even systematic screening before SOT. Exclusive Cryptosporidium free water feeding could be suggested during major cell mediated immunodeficiency.

Abstract: In this paper, we focus on deriving traffic flow macroscopic models from microscopic models containing a local perturbation such as a traffic light. At the microscopic scale, we consider a first order model of the form "follow the leader" i.e. the velocity of each vehicle depends on the distance to the vehicle in front of it. We consider a local perturbation located at the origin that slows down the vehicles. At the macroscopic scale, we obtain an explicit Hamilton-Jacobi equation left and right of the origin and a junction condition at the origin (in the sense of [ 25 ]) which keeps the memory of the local perturbation. As it turns out, the macroscopic model is equivalent to a LWR model, with a flux limiting condition at the junction. Finally, we also present qualitative properties concerning the flux limiter at the junction.

Abstract: International audience; GaN High Electron Mobility Transistors (HEMTs) are very promising for high power switching and radiofrequency operation. However, the lack of reliability feedback is one of its major drawbacks. Trapping effect especially is one of the main performance limitations of such components. Many measurement techniques exist for trapping effects characterization, especially for short time constant traps (s to several ms). However for longer time constants, self-heating may distort the measurements. This paper presents an electrical and athermal transient measurement method which has been developed to study the trapping and detrapping time constants of such components. It allows the extraction of slow transients without self-heating problems and is usable in long term electrical stress experiments. A simulation of this method with a simplified component’s model and the measurements results are presented. With this technique, we investigated especially the long time constants (τ>20 ms) over a range of temperature from 10°C to 105°C. We observed three thermally activated trap signatures on GaN devices with our method.

Abstract: The purpose of this study is to answer the following question: to what extent does the market of the professional football clubs trigger a lack of competitive balance? Based on a crossed view between the geography of sports and the sport economics, this research deals with 18 domestic leagues and the European Cups from 1975 to 2005. The results show that, in a context of commercialisation and deregulation of the activity, most of the big market teams strengthen their dominant position on a national as well as on a continental scale. Since sport experience is associated to the preservation of the uncertainty of outcome, such a report raises political issues: Does this phenomenon correspond to a reorganization of the parity which could lead to the creation of a closed league based on the North American model?; Ce travail a pour objectif de répondre à la question suivante: dans quelle mesure le potentiel local des clubs de football professionnel est-il est un facteur de segmentation sportive? Positionnée dans le champ de la géographie et de l’économie du sport, cette recherche porte sur 18 championnats domestiques et sur les Coupes européennes de 1975 à 2005. L’analyse révèle que dans un contexte de croissance des enjeux financiers et de dérégulation de l’activité, les clubs des plus grandes villes renforcent leur position dominante aussi bien à l’échelle nationale que continentale. Le spectacle sportif résidant dans le maintien de l’incertitude du résultat, un tel constat soulève des enjeux de politiques : n’assiste t-on pas à une recomposition des solidarités qui pourrait se concrétiser par la mise en place d’une ligue fermée sur le modèle nord-américain ?

Abstract: International audience; Polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are persistent organic pollutants that have been shown to affect fish life-history traits such as reproductive success, growth and survival. At the individual level, their toxicity and underlying mechanisms of action have been studied through experimental exposure. However, the number of experimental studies approaching marine environmental situations is scarce, i.e., inmost cases, individuals are exposed to either single congeners, or single types of molecules, or high concentrations, so that results can hardly be transposed to natural populations. In the present study, we evaluated the effect of chronic dietary exposure to an environmentally realistic marine mixture of PCB and PBDE congeners on zebrafish life-history traits from larval to adult stage. Exposure was conducted through diet from the first meal and throughout the life cycle of the fish. The mixture was composed so as to approach environmentally relevant marine conditions in terms of both congener composition and concentrations. Life-history traits of exposed fish were compared to those of control individuals using several replicate populations in each treatment. We found evidence of slower body growth, but to a larger asymptotic length, and delayed spawning probability in exposed fish. In addition, offspring issued from early spawning events of exposed fish exhibited a lower larval survival under starvation condition. Given their strong dependency on life-history traits, marine fish population dynamics and associated fisheries productivity for commercial species could be affected by such individual-level effects of PCBs and PBDEs on somatic growth, spawning probability and larval survival. Abstract: International audience; The Convention on the Contract for the International Carriage of Goods by Road (named ‘CMR) is one of the many major conventions for international trade. As part of international transport law, it is a uniform law convention.On the occasion of the 60th Anniversary of this major convention the organizers of the Rouen Symposium have decided to focus on the concrete difficulties raised by uniform application of the law in the field of international carriage of goods by road.These introductory remarks aim to place the issue addressed in this symposium in a broarder context. Three points will be touched: first, the utility of uniform law; following, the limits of uniform application of international conventions and the possible remedies; and eventually, the reasons why the study of the difficulties raised by uniform application of the CMR contribute to this debate.; La Convention relative au contrat de transport international de marchandises par route de 1956 (dite ‘CMR’) est l’une des importantes conventions en droit du commerce international. Elle relève plus particulièrement du droit des transports internationaux et fait partie des conventions de droit uniforme.A l’occasion du 60ème anniversaire de cette belle convention il a été fait le choix par les organisateurs de ce symposium de mettre l’accent sur les difficultés concrètes que peut poser l’application du droit uniforme dans le domaine du transport international de marchandises par route.Ces propos introductifs visent ainsi à replacer l’objet de ce colloque dans un contexte plus général. Ils aborderont ainsi trois points : tout d’abord, sera rappelée l’utilité du droit uniforme ; ensuite, seront évoqués les limites à l’application uniforme mais aussi les remèdes envisageables ; enfin, seront exposées les raisons qui justifient qu’une étude sur les difficultés de l’application uniforme de la CMR puisse contribuer à ce débat.

Abstract: International audience; An expedient synthesis of the first examples of iron-carbonyl complex-coumarin conjugates is reported. 7-Amino/7-hydroxycoumarin derivatives have been readily derivatized through an easily implemented single-step reaction involving the tricarbonyl(eta(5)-cyclohexadienyl)iron(1+) cation [(C6H7)Fe(CO)(3)](+). The scope and limitations of this N-/O-alkylation reaction were also investigated. Abstract: 26RFa, a novel RFamide neuropeptide, is the endogenous ligand of the former orphan receptor GPR103. Intracerebroventricular injection of 26RFa and its C-terminal heptapeptide, 26RFa((20-26)), stimulates food intake in rodents. To develop potent, stable ligands of GPR103 with low molecular weight, we have designed a series of aza-β(3)-containing 26RFa((20-26)) analogues for their propensity to establish intramolecular hydrogen bonds, and we have evaluated their ability to increase [Ca(2+)](i) in GPR103-transfected cells. We have identified a compound, [Cmpi(21),aza-β(3)-Hht(23)]26RFa((21-26)), which was 8-fold more potent than 26RFa((20-26)) in mobilizing [Ca(2+)](i). This pseudopeptide was more stable in serum than 26RFa((20-26)) and exerted a longer lasting orexigenic effect in mice. This study constitutes an important step toward the development of 26RFa analogues that could prove useful for the treatment of feeding disorders.

Abstract: International audience; Pituitary adenylate cyclase-activating polypeptide (PACAP) shows potent protective effects in numerous models of neurological insults. However, the use of PACAP as a clinically efficient drug is limited by its poor metabolic stability. By combining identification of enzymatic cleavage sites with targeted chemical modifications, a metabolically stable and potent PACAP38 analog was recently developed. The neuroprotective activity of this novel compound was for the first time evaluated and compared to the native peptide using a rat model of middle cerebral artery occlusion (MCAO). Our results show that as low as picomolar doses of PACAP38 and its analog strongly reduce infarct volume and improve neurological impairment induced by stroke. In particular, these peptides inhibit the expression of Bcl-2-associated death promoter, caspase 3, macrophage inflammatory protein-1α, inducible nitric oxide synthase 2, tumor necrosis factor-α mRNAs, and increase extracellular signal-regulated kinase 2, B-cell CLL/lymphoma 2 and interleukin 6 mRNA levels. These results indicate that the neuroprotective effect of PACAP after MCAO is not only due to its ability to inhibit apoptosis but also to modulate the inflammatory response. The present study highlights the potential therapeutic efficacy of very low concentrations of PACAP or its metabolically stable derivative for the treatment of stroke.

Abstract: AlGaN/GaN HEMTs are on their way to become a reference technology for high power and high frequency applications. However, the lack of feedback regarding the reliability of such devices can be felt by the defense, automotive and telecommunications industrialists because of its lack of maturity. This study deals with the long term reliability of power AlGaN/GaN HEMTs in RADAR operating mode. It is based upon electrical characterizations,the development of an athermal measurement technique for traps and RF stresses on an ageing bench. The sum of all the characterizations before, during and after the ageing tests as well as micro-structural analyses allows to define hypotheses regarding the physical origin of the performance drift of such components.; Les performances des HEMTs AlGaN/GaN sont en passe de devenir une technologie de référence pour les applications de forte puissance dans le domaine radiofréquence. Cependant, le manque de retour d'expérience sur la fiabilité de ces dispositifs se fait ressentir chez les industriels des domaines de la défense, de l'automobile et des télécommunications à cause de la jeunesse de cette filière. Cette étude se porte donc sur la fiabilité à long terme de transistors HEMT AlGaN/GaN de puissance en régime RADAR. Elle repose notamment sur des caractérisations électriques des composants, le développement d'une méthode athermique de caractérisation de pièges ainsi que le stress de ces transistors sur un banc de vieillissement dédié. La somme des caractérisations avant, pendant et après vieillissement ainsi que les analyses micro-structurales permet de définir des hypothèses quant à l'origine physique des dérives de performances de ces composants.

Abstract: With efficiency more than 21%, polycrystalline Cu(In,Ga)Se2 (CIGSe) semiconductors present maximum efficiency among thin film solar cells, making them a promising material to develop solar cell modules. Most efficient CIGSe cells produced till date are Ga poor cells (≈7.5% Ga) having band gap (Eg) =1.2 eV, however cells with optimum band gap (according to solar spectrum) 1.4 eV (≈18% Ga) presented much lower efficiency. This degraded performance of wide band gap CIGSe lead to scientific debates for many years suggesting various theories behind its decline in performance. Beneficial properties of Grain boundaries (GBs) are one of the main reasons for high efficiency of CIGSe and modifications in GBs could be the reason for hindered performance of Ga rich cells. In order to detect changes in chemistry of GBs, a technique able to resolve materials at atomic scale is employed in this research, known as atom probe tomography (APT). Exploring GB chemistry, we found that Ga poor cells always exhibit Cu deprived GBs which are known beneficial for cells due to their hole barrier properties, however Cu enriched GBs emerges for Ga concentration higher than 7.5%. This composition surprisingly coincides with decline in CIGSe performance and further increase in Ga concentration results in increase in Cu enriched GBs followed by degraded CIGSe performance. This suggests modifications in GBs can alter device’s performance, hence to retain GB properties some propositions and experiments are illustrated in the end.; Avec une efficacité de plus de 21%, le matériau polycristallin semiconducteur Cu(In,Ga)Se2(CIGSe) présente le maximum d’efficacité pour les cellules solaires dites à couches minces. Cette dégradation des performances des cellules à large bande interdite CIGSe conduit à des débats scientifiques depuis de nombreuses années ayant mené à diverses théories pour expliquer le déclin des performances de ces cellules avec l’augmentation de Ga. Les propriétés bénéfiques des joints de grains (GB) sont l'une des principales raisons de rendement élevé du CIGSe et les modifications de la chimie des GB pourraient être la raison de la performance limitée des cellules riches en Ga. Afin de détecter des changements dans la chimie des GB, une technique capable d’imager des matériaux à l'échelle atomique est employée dans ce travail : la sonde atomique tomographique (APT). L’exploration de la chimie des GB nous a permis de constater que les cellules pauvres en Ga présentent toujours une déplétion en Cu. Cette chimie semble bénéfique pour les cellules en raison de leurs car les GB agissent alors comme barrière pour trous, évitant ainsi les recombinaisons aux joints de grain. Cependant pour des concentrations de Ga supérieures à 7,5%, un enrichissement en Cu est observé. Cette composition coïncide étonnamment avec la baisse de la performance des cellules CIGSe. Cela suggère que des modifications dans les joints de grains peuvent altérer les performances de l'appareil. Donc, pour améliorer les propriétés des joints de grains, de nouvelles pistes sont envisagées à la fin du document.

Abstract: National audience; Nous décrivons dans cet article le modèle de données générique Information Retrieval for Omic and Clinical Sciences (IROmiCS) que nous proposons pour gérer les principaux types de données omiques (don-nées d'expression, de méthylation de l'ADN et variants génomiques). Nous décrivons également le langage de requêtes que nous avons développé qui repose sur le modèle IROmiCS et qui est dédié à l'interrogation des don-nées cliniques et omiques. Pour valider le modèle de données proposé, ainsi que le langage de requêtes associé, des données omiques expérimentales ont été intégrées dans ce modèle ainsi que des données de référence telle que les bases Gene du NCBI, Uniprot/Swissprot et la Gene Ontology. Plusieurs types de requêtes ciblant des données cliniques et des données omiques ont été réalisées sur les données intégrées. Une interface graphique facilite la visualisation des données intégrées par les cliniciens et les chercheurs. L'outil de recherche a permis de traiter des données symboliques, textuelles, numériques et chronologiques.

Abstract: International audience; La technologie HEMT (High Electron Mobility Transistor) en Nitrure de Gallium est prometteuse pour le fonctionnement radiofréquence. Cependant, le manque de retour d'expérience sur cette technologie récente pose des questions sur sa fiabilité. La caractérisation de la grille (diode Schottky) de ce type de transistors permet de mettre en évidence d'éventuelles modifications physiques de grille au cours de son vieillissement. Cependant, les phénomènes de piégeage viennent perturber ces caractérisations. Des analyses avec et sans éclairage UV permettent de dissocier le comportement de la diode de celui des pièges situés au voisinage de la grille. Plusieurs caractéristiques IV de diodes sur ce type de composants sont donc mesurées sous différentes conditions d'éclairage et de température, de manière à obtenir des informations sur l'impact des phénomènes de piégeage observables sur ce type de jonctions.

Abstract: International audience; The variety of protein cross-linkers developed in recent years illustrates the current requirement for efficient reagents optimized for mass spectrometry (MS) analysis. To date, the most widely used strategy relies on commercial cross-linkers that bear an isotopically labeled tag and N-hydroxysuccinimid-ester (NHS-ester) moieties. Moreover, an enrichment step using liquid chromatography is usually performed after enzymatic digestion of the cross-linked proteins. Unfortunately, this approach suffers from several limitations. First, it requires large amounts of proteins. Second, NHS-ester cross-linkers are poorly efficient because of their fast hydrolysis in water. Finally, data analysis is complicated because of uneven fragmentation of complex isotopic cross-linked peptide mixtures. We therefore synthesized a new type of trifunctional cross-linker to overrule these limitations. This reagent, named NNP9, comprises a rigid core and bears two activated carbamate moieties and an azido group. NNP9 was used to establish intra- and intermolecular cross-links within creatine kinase, then to map the interaction surfaces between α-Synuclein (α-Syn), the aggregation of which leads to Parkinson’s disease, and the molecular chaperone Hsc70. We show that NNP9 cross-linking efficiency is significantly higher than that of NHS-ester commercial cross-linkers. The number of cross-linked peptides identified was increased, and a high quality of MS/MS spectra leading to high sequence coverage was observed. Our data demonstrate the potential of NNP9 for an efficient and straightforward characterization of protein–protein interfaces and illustrate the power of using different cross-linkers to map thoroughly the surface interfaces within protein complexes.

Abstract: With efficiency more than 21%, polycrystalline Cu(In,Ga)Se2 (CIGSe) semiconductors present maximum efficiency among thin film solar cells, making them a promising material to develop solar cell modules. Most efficient CIGSe cells produced till date are Ga poor cells (≈7.5% Ga) having band gap (Eg) =1.2 eV, however cells with optimum band gap (according to solar spectrum) 1.4 eV (≈18% Ga) presented much lower efficiency. This degraded performance of wide band gap CIGSe lead to scientific debates for many years suggesting various theories behind its decline in performance. Beneficial properties of Grain boundaries (GBs) are one of the main reasons for high efficiency of CIGSe and modifications in GBs could be the reason for hindered performance of Ga rich cells. In order to detect changes in chemistry of GBs, a technique able to resolve materials at atomic scale is employed in this research, known as atom probe tomography (APT). Exploring GB chemistry, we found that Ga poor cells always exhibit Cu deprived GBs which are known beneficial for cells due to their hole barrier properties, however Cu enriched GBs emerges for Ga concentration higher than 7.5%. This composition surprisingly coincides with decline in CIGSe performance and further increase in Ga concentration results in increase in Cu enriched GBs followed by degraded CIGSe performance. Abstract: Duplex stainless steels used in primary circuit of 2nd generation nuclear power plant endure thermal ageing at service temperatures (285°C-323°C). This leads to an increase of their hardness and to a decrease of their Charpy toughness. The evolution of these properties is due to the phase transformations which occur in ferrite. Even if it is well known that the steel composition plays a role on the mechanical properties evolution (steels with low Ni and Mo contents are less sensitive to thermal ageing), the origin of this difference of behaviour has not been clearly identified yet.In this study, the kinetics of the phase transformations in the ferrite of a duplex stainless steel with low Ni and Mo contents and duplex model alloys with specific compositions have been studied by atom probe tomography.This work showed that: i) G phase precipitation is less intense and the kinetic of the spinodal decomposition is slower for this steel than for steels with higher Ni and Mo contents, ii) the synergy between spinodal decomposition and G phase precipitation is also observed in this steel, iii) the Mo does not affect the early stages of the G phase precipitation, iv) the formation mechanism of G phase precipitates is at least a two steps mechanism, v) when the number density of G phase precipitates is low, hardening is mainly controlled by the amplitude and the mean wavelength of the spinodal decomposition, vi) when the number density of G phase precipitates is high, the hardening due to G phase precipitates may not be negligible.A kinetic Monte Carlo model has been developed to explain the synergy observed experimentally between the spinodal decomposition and the G phase precipitation. This model allowed us to show that the coarsening of G phase precipitates is driven by a diffusion mechanism along α/α’ interfaces, and not by a bulk diffusion mechanism., Les aciers inoxydables austéno-ferritiques constituants les coudes moulés du circuit primaire des centrales nucléaires de deuxième génération, vieillissent aux températures de service (285°C-323°C). Ceci se traduit par une augmentation de leur dureté et une baisse de leur résilience. L’évolution de ces propriétés est attribuée aux transformations de phases qui opèrent dans la ferrite. S'il est admis que la composition de l’acier influe sur l’évolution de ces propriétés (les aciers moins riches en Ni et Mo sont moins sensibles au vieillissement), l’origine de ces différences n’est pas clairement identifiée.Dans cette étude, la cinétique des transformations de phase de la ferrite d'un acier austéno-ferritique pauvre en Mo et Ni ainsi que de celle d'alliages modèles duplex de composition ciblée a été étudiée par sonde atomique. Ces études ont permis de montrer que : i) l'intensité de la précipitation de la phase G est plus faible et la cinétique de la décomposition spinodale plus lente pour l'acier pauvre en Ni et Mo que pour des aciers riches en Mo et Ni analysés dans le cadre de travaux antérieurs, ii) la synergie entre décomposition spinodale et phase G est également observée dans cet acier, iii) le Mo n’a pas d’influence sur les premiers stades de précipitation de la phase G, iv) le mécanisme de précipitation de la phase G a lieu en au moins deux étapes, v) lorsque la densité de phase G est faible, le durcissement est contrôlé par l'amplitude et la longueur d'onde de la décomposition spinodale et vi) le rôle de la phase G sur le durcissement n'est peut-être pas si négligeable qu'on pouvait le supposer lorsque la densité est importante.Un modèle en simulation Monte Carlo cinétique a également été développé pour expliquer la synergie qui observée expérimentalement entre la décomposition spinodale et la précipitation de phase G. Ce modèle a permis de montrer que la coalescence des particules de phase G procède via la diffusion le long des interfaces α/α’ et non par diffusion en volume.

Abstract: The goal of this work is a better understanding of the different physical processes which occurs during field evaporation of atoms. There is some considerations applied both to metalic and non-metalic case and with the atom probe tomography analysis technik. This work allows us to use different parameters to optimize the experimental condictions in atom probe to obtain the best results avaible., Ce travail met en avant de nouvelles considérations pour une meilleure compréhension de l'évaporation par effet de champ dans le cadre de la sonde atomique laser. Ces travaux portant aussi bien sur les métaux que les diélectriques permettent de proposer de nombreuses pistes afin d'optimiser les paramètres expérimentaux utilisés lors des analyses de sonde atomique.

Abstract: The proportion of permanent magnets synchronous motors used for electric and hybrid automotive traction has exponentially increased during the past decade. This evolution comes with ever-demanding low noise and vibrations requirements. Multi-physics analysis of the motor is a decisive issue for the development of the powertrain. For the exploration of potentially efficient motor solutions in first design steps, it is thus a necessity to have at disposal fast and accurate computer codes. In this respect, acoustic and vibratory aspects are modeled using finite element and analytical models in this thesis. As a result, using an electromagnetic model, the weakly-coupled magneto-vibro-acoustic analysis (iterative solving of each physic) can be performed.The thesis is divided into four parts. The first part states the art on machine vibro-acoustic emissions and focuses on noise of magnetic origins modeling. Issues of modeling are detailed. Then, models are described to a great extent in the second part. Finite element models are favorably compared to experimental measures. A third part validates analytical models in comparison to finite element analysis, by gradually complicating the geometry of a surface permanent magnets radial flux machine. Finally, a fourth part uses finite element models to study non-conventional machines such as flux switching radial flux machines and surface permanent magnets axial flux machines.; Dans un contexte automobile électrique et hybride, la part des machines synchrones à aimants permanents s'est accrue exponentiellement. Cette évolution s'accompagne d'exigences en termes d'émissions vibro-acoustiques. En termes de dimensionnement de la chaîne de traction, l'analyse multi-physique du moteur s'avère être un enjeu crucial pour son développement. La nécessité de disposer de codes informatiques de conception possédant des temps de calcul faibles pour une précision maximale se fait ressentir pour l'exploration de solutions potentiellement performantes dans les premières étapes du processus de dimensionnement. A ce titre, les aspects vibratoires et acoustiques sont modélisés analytiquement et par éléments finis dans la présente thèse. Ainsi, au moyen du modèle magnétique, l'analyse magnéto-vibro-acoustique faiblement couplée (résolution itérative des différentes physiques) peut être réalisée.La thèse est découpée en quatre parties. La première expose un état de l’art sur les émissions vibro-acoustiques des machines et notamment la modélisation du bruit d’origine magnétique. Les problématiques de la modélisation sont détaillées. Dans une deuxième partie, les modèles sont largement décrits. Les modèles éléments finis sont validés expérimentalement. Une troisième partie se propose de valider les modèles analytiques par éléments finis, en complexifiant progressivement la géométrie d’une machine à aimants montés en surface à flux radial. Interroger la temporalité du discours nous donne la possibilité de mieux appréhender la cohérence temporelle des espaces de débat associé à la crise de la grippe A/H1N1. Abstract: Politiques et représentations des épidémies. L'objectif de ce rapport est d'étudier la dynamique temporelle du discours de la presse sur la pandémie de grippe A/H1N1 et la campagne de vaccination associée (2009‐2010). Interroger la temporalité du discours nous donne la possibilité de mieux appréhender la cohérence temporelle des espaces de débat associé à la crise de la grippe A/H1N1. Abstract: International audience; SigX, one of the 19 extra-cytoplasmic function sigma factors of P. aeruginosa, was only known to be involved in transcription of the gene encoding the major outer membrane protein OprF. We conducted a comparative transcriptomic study between the wildtype H103 strain and its sigX mutant PAOSX, which revealed a total of 307 differentially expressed genes that differed by more than 2 fold. Most dysregulated genes belonged to six functional classes, including the "chaperones and heat shock proteins", "antibiotic resistance and susceptibility", "energy metabolism", "protein secretion/export apparatus", and "secreted factors", and "motility and attachment" classes. In this latter class, the large majority of the affected genes were down-regulated in the sigX mutant. In agreement with the array data, the sigX mutant was shown to demonstrate substantially reduced motility, attachment to biotic and abiotic surfaces, and biofilm formation. In addition, virulence towards the nematode Caenorhabditis elegans was reduced in the sigX mutant, suggesting that SigX is involved in virulence-related phenotypes.

Abstract: International audience; The understanding of interaction effects between marine energy converters represents the next step in the research process that should eventually lead to the deployment of such devices. Although some a priori considerations have been suggested recently, very few real condition studies have been carried out concerning this issue. Trials were run on 1/30th scale models of three-bladed marine current turbine prototypes in a flume tank. The present work focuses on the case where a turbine is placed at different locations in the wake of a first one. The interaction effects in terms of performance and wake of the second turbine are examined and compared to the results obtained on the case of one single turbine. Besides, a three-dimensional software, based on a vortex method is currently being developed, and will be used in the near future to model more complex layouts. The experimental study shows that the second turbine is deeply affected by the presence of an upstream device and that a compromise between individual device performance and inter-device spacing is necessary. Numerical results show good agreement with the experiment and are promising for the future modelling of turbine farms.

Abstract: We study Mixed Mode Oscillations (MMOs) in systems of two weakly coupled slow/fast oscillators. We focus on the existence and properties of a folded singularity called FSN II that allows the emergence of MMOs in the presence of a suitable global return mechanism. As FSN II corresponds to a transcritical bifurcation for a desingularized reduced system, we prove that, under certain non-degeneracy conditions, such a transcritical bifurcation exists. We then apply this result to the case of two coupled systems of FitzHugh- Nagumo type. This leads to a non trivial condition on the coupling that enables the existence of MMOs.

Abstract: L’article original est publié par The Endocrine Society; The sulfated neurosteroids pregnenolone sulfate (Δ(5)PS) and dehydroepiandrosterone sulfate (DHEAS) are known to play a role in the control of reproductive behavior. In the frog Pelophylax ridibundus, the enzyme hydroxysteroid sulfotransferase (HST), responsible for the biosynthesis of Δ(5)PS and DHEAS, is expressed in the magnocellular nucleus (Mgd) and the anterior preoptic area (Poa), two hypothalamic regions that are richly innervated by GnRH1-containing fibers. This observation suggests that GnRH1 may regulate the formation of sulfated neurosteroids to control sexual activity. Double-labeling of frog brain slices with HST and GnRH1 antibodies revealed that GnRH1-immunoreactive fibers are located in close vicinity of HST-positive neurons. The cDNAs encoding three GnRH receptors (designated riGnRHR-1, -2, and -3) were cloned from the frog brain. Reverse transcription-polymerase chain reaction analyses revealed that riGnRHR-1 is strongly expressed in the hypothalamus and the pituitary while riGnRHR-2 and -3 are primarily expressed in the brain. In situ hybridization histochemistry indicated that GnRHR-1 and GnRHR-3 mRNAs are particularly abundant in Poa and Mgd whereas the concentration of GnRHR-2 mRNA in these two nuclei is much lower. Pulse-chase experiments using tritiated Δ(5)P and DHEA as steroid precursors, and 3'-phosphoadenosine 5'-phosphosulfate (PAPS) as a sulfonate moiety donor, showed that GnRH1 stimulates in a dose-dependent manner the biosynthesis of Δ(5)PS and DHEAS in frog diencephalic explants. Since Δ(5)PS and DHEAS, like GnRH, stimulate sexual activity, our data strongly suggest that some of the behavioral effects of GnRH could be mediated via the modulation of sulfated neurosteroid production.

Abstract: International audience; High concentrations of pituitary adenylate cyclase-activating polypeptide (PACAP) and a high density of PACAP binding sites have been detected in the developing rat cerebellum. In particular, PACAP receptors are actively expressed in immature granule cells, where they activate both adenylyl cyclase and phospholipase C. The aim of the present study was to investigate the ability of PACAP to induce calcium mobilization in cerebellar granule neurons. Administration of PACAP-induced a transient, rapid, and monophasic rise of the cytosolic calcium concentration ([Ca(2+)]i), while vasoactive intestinal peptide was devoid of effect, indicating the involvement of the PAC1 receptor in the Ca(2+) response. Preincubation of granule cells with the Ca(2+) ATPase inhibitor, thapsigargin, or the d-myo-inositol 1,4,5-trisphosphate (IP3) receptor antagonist, 2-aminoethoxydiphenyl borate, markedly reduced the stimulatory effect of PACAP on [Ca(2+)]i. Furthermore, addition of the calcium chelator, EGTA, or exposure of cells to the non-selective Ca(2+) channel blocker, NiCl2, significantly attenuated the PACAP-evoked [Ca(2+)]i increase. Preincubation of granule neurons with the N-type Ca(2+) channel blocker, ω-conotoxin GVIA, decreased the PACAP-induced [Ca(2+)]i response, whereas the L-type Ca(2+) channel blocker, nifedipine, and the P- and Q-type Ca(2+) channel blocker, ω-conotoxin MVIIC, had no effect. Altogether, these findings indicate that PACAP, acting through PAC1 receptors, provokes an increase in [Ca(2+)]i in granule neurons, which is mediated by both mobilization of calcium from IP3-sensitive intracellular stores and activation of N-type Ca(2+) channel. Some of the activities of PACAP on proliferation, survival, migration, and differentiation of cerebellar granule cells could thus be mediated, at least in part, through these intracellular and/or extracellular calcium fluxes.

Abstract: 9 pages; International audience; A container terminal is a complex and dynamic system. Many operations occur within the storage area: containers import, containers export and containers shifting. All these operations require the respect of many rules and even laws in order to guarantee the port safety and to prevent risks, especially for the hazardous material storage. This paper proposes an hybrid architecture, using a Multi-Agent System and a Cellular Automaton, to handle the hazardous container storage problem. It is an optimization problem since the aim is to improve the container terminal configuration, that is, the way hazardous containers are dispatched through the terminal. Simple optimization heuristic methods were tested on a terminal with four hazardous container types. We consider then containers as agents, in order to use a Multi-Agent System for the decision aid software, and a Cellular Automaton for modelling the terminal itself. This approach will improve the naive methods first implemented, and should apply to the actual data and constraints of container terminal management.

Abstract: The void prediction in LCM processes sparks off interest within the composite material industry because it is a significant issue to keep the expected mechanical properties. The liquid properties, the preform geometry and the flow conditions impact the quantity of void entrapped inside the final product. The complex geometry of the reinforcement due to the arrangement of the bundles and the fibres is a key point to understand and quantify this phenomenon. This paper deals with both simple model networks which can occur inside a fabric representing connected capillaries, so-called "Pore Doublet Model (PDM)". A first is considering two capillaries converging on a node (T-junction) and a second is representing two capillaries interconnected with a supplying principle. These configurations can affect locally the evolution of flow fronts. First, experiments of bubble formed in a T-junction device have been performed and studied. Abstract: This work aims to characterise by the means of expérimental and numerical modeling, the behavior of marine current turbine and the characteristics of the wake generated under realistic inflow conditions. For that purpose, we used a tri-bladed horizontal axis turbine of 0.7 m of diameter. Particular attention is paid to the flow characteristic effects on the efficiency of the turbine, but also on the generated wake. Tests are carried out in the Ifremer wave and current flume tank in Boulogne-sur-Mer and in the towing tank of Brest, taking into account current profils, upstream turbulence intensity level and disturbances generated by wave/current interaction. The development of a three-dimensional software based on vortex methods, taking into account the non stationary evolution of the wake generated by a turbine is implemented. The obtained numerical results are compared to the experimental ones, making the validation of tools possible in terms of efficiency and of wake evolution., Ce travail a pour objectif de caractériser par le biais de la modélisation expérimentale et numérique le comportement hydrodynamique d’une hydrolienne et les caractéristiques du sillage généré pour des conditions réalistes de fonctionnement. Les maquettes d’hydroliennes considérées sont des turbines tri-pales à axe horizontal de 0,7 m de diamètre. Une attention particulière est portée sur les effets des caractéristiques de l’écoulement sur les performances de la turbine, mais également sur le sillage géneré. Les essais sont menés au bassin d’essais d’Ifremer à Boulognesur-Mer et au canal de traction de Brest, où différents profils de courant, taux de turbulence de l’écoulement amont et conditions houle/courant ont été considérées. Les développements d’un code tridimensionnel basé sur les méthodes vortex, prenant en compte l’évolution instationnaire du sillage généré par une turbine sont présentés. Les résultats numériques obtenus sont comparés aux résultats expérimentaux, permettant de valider les outils en termes de rendement et d’évolution du sillage.

Abstract: International audience; Pituitary adenylate cyclase-activating polypeptide (PACAP) and tissue plasminogen activator (tPA) play important roles in neuronal migration and survival. However, a direct link between the neurotrophic effects of PACAP and tPA has never been investigated. In this study, we show that, in PC12 cells, PACAP induced a 9.85-fold increase in tPA gene expression through activation of the protein kinase A- and protein kinase C-dependent signaling pathways. In immature cerebellar granule neurons (CGN), PACAP stimulated tPA mRNA expression and release of proteolytically active tPA. Immunocytochemical labeling revealed the presence of tPA in the cytoplasm and processes of cultured CGN. The inhibitory effect of PACAP on CGN motility was not affected by the tPA substrate plasminogen or the tPA inhibitor plasminogen activator inhibitor-1. In contrast, plasminogen activator inhibitor-1 significantly reduced the stimulatory effect of PACAP on CGN survival. Altogether, these data indicate that tPA gene expression is activated by PACAP in both tumoral and normal neuronal cells. The present study also demonstrates that PACAP stimulates the release of tPA which promotes CGN survival by a mechanism dependent of its proteolytic activity.

Abstract: Purpose. The aim of this paper is to develop new optical bioprobes for the imaging of apoptosis.Procedure. We developed quenched near-infrared probes which become fluorescent upon cleavage by caspase-3, the key regulatory enzyme of apoptosis.Results. Probes were shown to be selectively cleaved by recombinant caspase-3. Apoptosis of cultured endothelial cells was associated with an increased fluorescent signal for the cleaved probes, which colocalized with caspase-3 and was reduced by the addition of a caspase-3 inhibitor. Flow cytometry demonstrated a similar profile between the cleaved probes and annexin V.Ex vivoexperiments showed that sections of hearts obtained from mice treated with the proapoptotic drug doxorubicin displayed an increase in the fluorescent signal for the cleaved probes, which was reduced by a caspase-3 inhibitor.Conclusion. We demonstrated the capacity of these novel probes to detect apoptosis by optical imagingin vitroandex vivo.

Abstract: International audience; The microstructure of a unidirectional glass-fiber composite material is described seeking especially for the influence of the stitching perpendicular to the reinforcement. Serial cuts are performed through the composite and the microstructure is quantified using global parameters and linear morphological analysis. A key result is that the stitching induces variations in fibers spacing within the yarns and in the matrix volume between the yarns. This can affect noticeably the flow of the resin during the manufacturing process and also the mechanical properties of the composite.

Abstract: La problématique de nos travaux de recherche se place dans le contexte de la RI textuelle sur le Web. Nous proposons en ce sens des méthodes de RI basées sur l'exploitation de connaissances. Nos expérimentations sont réalisées dans le cadre du projet CISMeF qui indexe un grand nombre de documents en focntion d'une ressource Termino-Ontologique du domaine médical. Nous avons développé le prototype KnowQUE (Knowledge Based QUery Expansion) pour corriger, préciser et enrichir les requêtes des utilisateurs. Ses modules exploitent les traitement linguistiques, la fouille de données et les mécanismes de raisonnement associés aux logiques de description., This thesis deals with the problemn of Information Retrieval on the Internet. We propose several methods of query expansion founded on knowledge exploitation. Experimentations are done in the context of the CISMeF project which indexes several documents according to a termino-ontological resource of the medical domain. We developed the KnowQUE tool (Knowledge Based QUery Expansion) tfor correcting, precising and enriching users' queries. All its modules exploit natural language processing, data mining and reasoning services associated to description logics.

Abstract: International audience; Le catalogage des fonds musicaux anciens conservés en Haute-Normandie est une entreprise de grande envergure commandée par le Ministère de la culture et réalisée par le département de musicologie de l'université de Rouen. Il s'inscrit logiquement dans le programme d'inventaire des fonds musicaux en France, dont l'objectif est le recensement et la mise en valeur des sources musicales anciennes. Le patrimoine musical haut-normand est localisé principalement à Rouen, à la Bibliothèque municipale, pour laquelle est prévue la parution de cinq volumes. Trois autres volumes décriront les collections musicales des bibliothèques de Dieppe, du Havre et de l'Eure.Ce premier volume propose un inventaire précis du fonds du Théâtre des Arts de Rouen. Actuellement conservé à la Bibliothèque municipale de Rouen, cet ensemble découle directement de l'intense activité lyrique qui rythme quotidiennement la vie musicale des Rouennais depuis l'inauguration de la première salle en 1776 jusqu'à la destruction par les bombardements de la seconde en 1944. Le fonds s'inscrit dans une période plus large puisqu'il commence à se constituer dès les années 1750 et qu'il continue à être exploité jusqu'aux années 1990.Composé intégralement de matériels d'exécution (partitions et parties séparées), ce fonds est un témoin important de la vie musicale rouennaise et havraise (villes entre lesquelles existaient des échanges entre les théâtres), éclairant notamment les phénomènes de circulation de la musique et de réception des œuvres en province entre la fin de l'Ancien Régime et la Seconde Guerre mondiale.L'opéra est le genre le plus représenté dans le fonds du Théâtre des Arts, plus particulièrement l'opéra-comique auquel les Rouennais sont très attachés. Les autres genres de la scène théâtrale sont toutefois représentés : le vaudeville, le ballet, la symphonie et le quatuor. Par ailleurs, se distinguent dans ce fonds de nombreuses pièces rares voire uniques, manuscrites ou imprimées.L'introduction de ce catalogue est divisée en quatre parties :1. Histoire du fonds du Théâtre ;2. Concordance des cotes avec le catalogue de 1924 ;3. Description des notices ;4. Description de l'index et des tables. Elle est suivie de 874 notices très détaillées et richement illustrées, rédigées selon les principes de description des sources musicales établis par le RISM et la Bibliothèque nationale de France. La troisième partie du volume est constituée d'un index et de tables qui font de cet ouvrage un véritable outil de recherche. Abstract: The aim of this work is to contribute in improving fibrous preforms impregnation for Liquid Composite Molding (LCM) processes. The void prediction in LCM sparks off interest within the Composite Material elaboration because it represents a significant issue to keep the expected mechanical properties of the final product. The liquid properties, the preform geometry and the flow conditions impact the void or bubble entrapped inside and outside the yarns. Nevertheless, due to the complex geometry of the reinforcement, experimental characterization of bubble formation remains delicate. Thus, our study deals with two simple model networks representing connected pores so called "Pore Doublet Model". A first is considering two capillaries converging on a node (T-junction) and a second is representing two capillaries interconnected with a supplying principle. Abstract: Intracerebral-intraventricular hemorrhages (ICH/IVH) in very preterm neonates are responsible for high mortality and subsequent disabilities. In humans, tissue plasminogen activator (t-PA) initiates fibrinolysis and activates endoluminal-endothelial receptors; dysfunction of the t-PA inhibitor (PAI-1) results in recurrent hemorrhages. We used PAI-1 knockout (PAI-1) mice to examine the role of t-PA in age-dependent intracranial hemorrhages as a possible model of preterm ICH/IVH. Intracortical injection of 2 μL of phosphate-buffered saline produced a small traumatic injury and a high rate of hemorrhage in PAI-1 pups at postnatal day 3 (P3) or P5, whereas it had no effect in wild-type neonates. This resulted in white matter and cortical lesions, ventricle enlargement, hyperlocomotion, and altered cortical levels of serotonin and dopamine in the adult PAI mice. N-methyl-D-aspartate receptor blockers, plasmin- and matrix metalloproteinases inhibitors reduced hemorrhage and tissue lesions. In contrast to P3 to P5, no significant hemorrhages were induced in P10 PAI-1 pups and there were no behavioral or neurochemical alterations in adulthood. These data suggest that microvascular immaturity up to P5 in mice is a determinant factor required for t-PA-dependent vascular rupture. Neonatal PAI-1 mice could be a useful ICH/IVH model for studying the ontogenic window of vascular immaturity and vascular protection against later neurodisabilities.
