This study was aimed at determining the prevalence of cryptosporidiosis in goats in central Algeria. A total of 605 samples of goat faeces were collected from several mixed farms (sheep and goats) in the regions of Médéa, Djelfa, and Ain Defla. The aim of this study was to prepare microcapsules based on a natural polymer chitosan solution (high degree of deacetylation (DDA), low molecular weight (MW), and low viscosity)/sodium alginate in the presence of a crosslinking agent (glutaraldehyde), in order to encapsulate and vectorise the active principle towards the diseased organ (colon), without being diffused into other levels of the digestive tract, to increase the therapeutic effectiveness of treatment by chemotherapy and to reduce undesirable effects. The method of preparation of the microcapsules obtained from the sodium alginate/chitosan solution/active ingredients system was examined by conventional optical microscopy. In addition, an in vitro study was carried out on the active ingredients’ release profiles, depending on the pH simulating the gastric and intestinal media for the seven systems proposed. It should be mentioned that, in the basic medium (pH(colon) = 8), the release of the active ingredients is of the utmost importance. Nevertheless, control of this release can be improved by a crosslinking agent and the coating method. The dry [sodium alginate / chitosan solution / active ingredients + crosslinking 2 %] formulation coated with non-crosslinked chitosan (Formulation 7) is the standard formula that meets all the criteria from our earlier work, with a core release rate of 67 %. On the basis of 16S rRNA gene and whole genome similarities, the isolates were almost identical and clearly grouped in the genus Pseudochrobactrum . This allocation was confirmed by the analysis of fatty acids (C19:cyclo, C18 : 1, C18 : 0, C16 : 1 and C16 : 0) and of polar lipids (major components: phosphatidylethanolamine, ornithine-lipids, phosphatidylglycerol, cardiolipin and phosphatidylcholine, plus moderate amounts of phosphatidylmonomethylethanolamine, phosphatidyldimethylethanolamine and other aminolipids). Genomic, physiological and biochemical data differentiated these isolates from previously described Pseudochrobactrum species in DNA relatedness, carbon assimilation pattern and growth temperature range. Description of the subject: Triple-negative breast cancer (TNBC), a breast cancer subtype, is characterized by the lack of both estrogen and progesterone hormonal receptors expression and by the absence of human epidermal growth factor receptor 2 overexpression. Patients with a pathological complete response (pCR) have better disease-free and overall survival compared to those with residual disease. The high level of tumor-infiltrating lymphocytes (TILs) is associated with a higher response to neoadjuvant chemotherapy (NAC) and better prognosis. Objective: Evaluation of TILs and their predictive impact in early TNBC in an Algerian population. Methods: We assessed TILs and correlated them with the pCR rate in 94 early TNBC patients treated from 2015 to 2017 who underwent breast microbiopsy, NAC, and then surgery. Results: Among 94 early TNBC patients, 53 (56.4%) achieved pCR and 41 (43.6%) had a residual disease. While some clinicopathological factors did not affect pCR, stromal TILs showed significant correlation with pCR (P < 0.0001). The presence of CD3+, CD4+, CD8+ and CD20+ TILs was also significantly correlated with pCR (P < 0.0001, P = 0.001, P = 0.0003 and P = 0.0001, respectively). Conclusion: Our data showed that TILs were significantly associated with pCR, suggesting that TILs are a predictive biomarker for pCR in early TNBC patients treated by NAC in our cohort.

Seafood rapidly deteriorates; storage by cooling in ice is a common technique for prolonging its freshness and quality. The current study compared the shelf life and quality of ice-preserved European sea bass (Dicentrarchus labrax) sourced from two different intensive fish farming systems: terrestrial ponds using heated water from a power plant and floating cages in the open sea. Using sensory analysis, the fish cultured in the heated water ponds were rejected as unacceptable for consumption after 13 days, while the fish reared in the floating cages were rejected after 15 days. Total viable counts of aerobic bacteria, pH, total volatile basic nitrogen and trimethylamine levels were consistently higher in the fish cultured in heated water ponds. Chemical, bacteriological and organoleptic acceptability demonstrated that fish rearing conditions were important for storage and needs to be taken into consideration for fish management and processing.

Brucellosis is a zoonosis caused by bacteria of the genus Brucella that causes important economic losses and human suffering worldwide. Brucellosis control requires an understanding of the Brucella species circulating in livestock and humans and, although prevalent in African countries of the Mediterranean basin, data for this area are mostly restricted to isolates obtained from humans and small ruminants. Here, we report the characterization of twenty-four Brucella strains isolated from Algerian cattle. Bruce-ladder multiplex PCR and conventional biotyping showed that Algerian cattle are infected mostly by B. abortus biovar 3, and to less extent by B. abortus biovar 1 and B. melitensis biovar 3. Extended AMOS-ERY PCR showed that all Algerian B. abortus biovar 3 strains were of the subgroup 3b. Although by multi locus variable number of tandem repeats analysis (MLVA) most isolates were closer to the European counterparts, five strains displayed characteristics distinct from the European isolates and those of countries across the Sahara, including three repetitions of marker Bruce55. These five strains, plus an earlier isolate from an Algerian human patient, may represent a lineage close to clades previously described in Africa. These data provide the basis for additional molecular epidemiology studies in northern Africa and indicate that further bacteriological and molecular investigations are necessary for a complete understanding of the epidemiology of cattle brucellosis in countries north and south of the Sahara.

Highlights • The etiology of neonatal diarrhea is multifactorial and remains one of the biggest problems in ruminant livestock farming. This study represent the first report in Algeria on Cryptosporidium parvum, Escherichia coli K99, rotavirus and coronavirus in the etiology of neonatal diarrhea in lambs. • Faecal samples from 559 neonatal lambs aged less than 30 days from 30 sheepfolds were screened with pathogen-specific antigen ELISA for C. parvum, E. coli K99, rotavirus, and coronavirus. • The prevalence of C. parvum was the highest compared to other enteric pathogens. • In various combinations, mixed infections were detected only with C. parvum., The etiology of neonatal diarrhea is multifactorial and remains one of the greatest health problems in sheep livestock farming. Faecal samples from 559 neonatal lambs aged less than 30 days from 30 sheepfolds located in the north-center region of Algeria were screened with pathogen-specific antigen ELISA for Cryptosporidium parvum, Escherichia coli K99, rotavirus, and coronavirus. Of the 559 lambs, 312 (58.81 %), 155 (27.72 %), 72 (12.88 %) and 20 (3.57 %) were positives for C. parvum, E. coli K99, rotavirus and coronavirus antigens, respectively. The prevalence of C. parvum was the highest (p < 0.0001). C. parvum, E. coli K99, rotavirus and coronavirus were observed in 23 (76.66 %), 17 (56.66 %), 9 (30 %) and 3 (10 %) sheepfolds, respectively. Compared to age, the prevalence of C. parvum was highest during the second and third week of age (p < 0.001). In contrast, other pathogens were found to be more frequent in lambs aged ≤7 days (p < 0.001). The number of lambs with diarrhea was 280 (50.09 %) of which 280 (100 %), 127 (45.35 %), 52 (18.57 %) and 10 (3.57 %) were found to be infected with C. parvum, E. coli K99, rotavirus and coronavirus, respectively (p < 0.0001). In various combinations, mixed infections were detected only with C. parvum. This is the first report of C. parvum, E. coli K99, rotavirus, and coronavirus in ≤30-days old neonatal lambs in Algeria. Special attention should be given to the first colostrum feeding, hygiene of the farm, prevention and control measures for a better prevention of neonatal diarrhea in lambs.

Airway epithelial cells (AEC) are the first line of defense against airborne infectious microbes and play an important role in regulating the local immune response. However, the interplay of epithelial cells and professional immune cells during both homeostasis and infection has only been partially studied. The present study was performed to determine how bronchial epithelial cells affect the activation of monocytes. Under healthy conditions, AECs were shown to inhibit reactivity of monocytes. We hypothesized that upon infection, monocytes might be released from inhibition by AECs. We report that direct contact of monocytes with unstimulated BEAS2B epithelial cells results in inhibition of TNF secretion by activated monocytes. In addition to the known soluble modulators, we show that cell contacts between epithelial cells and monocytes or macrophages also contribute to homeostatic inhibitory actions. We find AECs to express the inhibitory molecule PD-L1 and blockade of PD-L1 results in increased secretion of pro-inflammatory cytokines from monocytes. Contrary to the inhibitory activities during homeostasis, epithelial cells infected with Respiratory Syncitial Virus (RSV) induce a significant release of inhibition. However, release of inhibition was not due to modulation of PD-L1 expression in AECs. We conclude that airway epithelial cells control the reactivity of monocytes through direct and indirect interactions; however tonic inhibition can be reverted upon stimulation of AECs with RSV and thereof derived molecular patterns. The study confirms the important role of airway epithelial cells for local immune reactions.

Developing viral vaccines through the ultraviolet (UV) inactivation of virus is promising technique since it is straightforward and economically affordable, while the resulting viruses are capable of eliciting an adequate antiviral immune response. Nodavirus (NNV) is a devastating virus that mainly affects European sea bass juveniles and larvae, causing serious economic losses in Mediterranean aquaculture. In this work, a potential vaccine consisting on UV-inactivated NNV (iNNV) was generated and administered to healthy juveniles of European sea bass to elucidate whether it triggers the immune response and improves their survival upon challenge. First, iNNV failed to replicate in cell cultures and its intraperitoneal administration to sea bass juveniles also failed to produce fish mortality and induction of the type I interferon (IFN) pathway, indicating that the NNV was efficiently inactivated. By contrast, iNNV administration induced significant serum non-specific antimicrobial activity as well as a specific antiviral activity and immunoglobulin M (IgM) titres against NNV. Interestingly, few changes were observed at transcriptional level in genes related to either innate or adaptive immunity, suggesting that iNNV could be modulating the immune response at protein or functional level. In addition, the iNNV vaccinated group showed improved survival, reaching a relative survival percentage of 57.9%. Moreover, challenged fish that had been vaccinated presented increased serum antibacterial, antiviral and IgM titres, as well as the higher transcription of mhc1a, ifn, isg15 and cd8a genes in brain, while in the headkidney the transcription of mhc1a, mhc2b and cd8a was down-regulated and mx, isg15 and tcrb was up-regulated. Redbelly tilapia (Tilapia zillii; Gervais, 1848) is one of the most valuable freshwater species in North Africa representing an important part of the continental production, especially in brackish lakes. In Algeria, T. zillii is distributed in several lakes and tributaries of some rivers in the south. Though some attempts are in progress to culture this species, many investigations covering its biology and farm management are still needed. In this sense, this is the first study attempting to evaluate some of the T. zillii immune parameters and valuable data to assess their health and well-being status. Thus, we have determined the levels of serum peroxidases as well as the alternative complement, antiprotease and bactericidal activities. Furthermore, we have also evaluated the potential impact of two acute stress factors, commonly found in fish farms, in these parameters. Although it was assessed that fish exposed to low temperatures or crowding were stressed, as indicated by their increased serum levels of cortisol and glucose, both acute stressors failed to significantly affect serum peroxidases as well as antiprotease and complement activities. However, the bactericidal activity was reduced in general but only in those exposed to crowding reached statistical significance. Further studies are needed to characterise the immune response in T. zillii as well as the effects that farming stresses may produce.

Background: We recently showed that poly(ADP-ribose)polymerase-1 (PARP-1) may play a role in allergen (ovalbumin)-induced airway eosinophilia, potentially through a specific effect on IL-5 production. We also reported that while IL-5 replenishment promotes reversal of eosinophilia in lungs of PARP-1 )/) mice, IL-4 or Immunoglobulin E replenishment do not, suggesting a potentially significant regulatory relationship between PARP-1 and IL-5. Objective: To explore the mechanism by which PARP-1 regulates IL-5 production and to determine how PARP-1 inhibition blocks allergen-induced eosinophilia. Methods: This study was conducted using a murine model of allergic airway inflammation and primary splenocytes. Results: PARP-1 knockout-associated reduction in IL-5 upon allergen exposure occurs at the mRNA level. Such an effect appears to take place after IL-4 receptor activation as PARP-1 inhibition exerted no effect on JAK1/JAK3 activation. Signal transducer and activator of transcription-6 (STAT-6) protein was severely downregulated in spleens of PARP-1 )/) mice without any effect on mRNA levels, suggesting an effect on protein integrity rather than gene transcription. Interestingly, the degradation of STAT-6 in PARP-1 )/) mice required allergen stimulation. Additionally, PARP-1 enzymatic activity appears to be required for STAT-6 integrity. The downregulation of STAT-6 coincided with mRNA and protein reduction of GATA-binding protein-3 and occupancy of its binding site on the IL-5 gene promoter. IL-4 was sufficient to induce STAT-6 downregulation in both PARP-1 )/) mice and isolated splenocytes. Such degradation may be mediated by calpain, but not by proteasomes. Conclusion: These results demonstrate a novel function of PARP-1 in regulating IL-5 expression during allergen-induced inflammation and explain the underlying mechanism by which PARP-1 inhibition results in IL-5 reduction.

Vaccination of calves with formalin-inactivated bovine respiratory syncytial virus (FI-BRSV) induces low levels of cellular immunity that may not be protective. Since inactivated and subunit vaccines formulated with CpG oligodeoxynucleotides (ODNs) have been shown to induce cellular immune responses, we studied the ability of a FI-BRSV vaccine formulated with CpG ODN to elicit cellular immunity against BRSV. Neonatal calves were immunized with FI-BRSV, FI-BRSV formulated with CpG ODN or medium and challenged with BRSV after two immunizations. Calves vaccinated with FI-BRSV formulated with CpG ODN developed increased numbers of IFN-gamma secreting cells in the peripheral blood and broncho-tracheal lymph nodes and enhanced BRSV-specific serum IgG2 in comparison to FI-BRSV immunized animals. Calves that received the FI-BRSV vaccine formulated with CpG ODN also experienced a reduction in the amount of BRSV in the lung tissue. Based on these observations, CpG ODN appears to be a suitable candidate adjuvant for inactivated BRSV vaccines.

Commercial killed bovine respiratory syncytial virus (K-BRSV) and formalin-inactivated BRSV (FI-BRSV) tend to induce Th2-type immune responses, which may not be protective and may even be detrimental during subsequent exposure to the virus. In this study we assessed the ability of CpG oligodeoxynucleotides (ODNs) to aid in the generation of effective and protective BRSV-specific immune responses. Mice were immunized subcutaneously with FI-BRSV formulated with CpG ODN, Emulsigen (Em), CpG ODN and Em, or non-CpG ODN and Em. Two additional groups were immunized with K-BRSV or K-BRSV and CpG ODN. After two vaccinations, the mice were challenged with BRSV. FI-BRSV induced Th2-biased immune responses characterized by production of serum immunoglobulin G1 (IgG1) and IgE, as well as interleukin-4 (IL-4), by in vitro-restimulated splenocytes. Formulation of FI-BRSV with CpG ODN, but not with non-CpG ODN, enhanced serum IgG2a and IFN-γ production by splenocytes, whereas serum IgE was reduced. Although the immune response induced by K-BRSV was not as strongly Th2 biased, the addition of CpG ODN to this commercial vaccine also resulted in a more Th1-type response. Furthermore, the addition of CpG ODN to the BRSV vaccine formulations resulted in enhanced neutralizing antibody responses. Significant production of IL-5, eotaxin, and eosinophilia was observed in the lungs of FI-BRSV- and K-BRSV-immunized mice. However, IL-5 and eotaxin levels, as well as the number of eosinophils, were decreased in the mice vaccinated with the CpG ODN-formulated vaccines. Finally, when formulated with CpG ODN, both FI-BRSV and K-BRSV significantly reduced virus production after challenge with BRSV.

The cytology of a new microsporean parasite Microsporidium epithelialis sp. n. from the intestinal epithelial cells of the freshwater oligochaete Tubifex sp. (Tubificidae) is described. The microsporean occurred together with an actinosporean of the genus Triactinomyxon, which was found between the epithelial cells. The merogonic and sporogonic stages (mature spores included) of the microsporean parasite are monokaryotic. An individual sporophorous vesicle surrounds each spore. The fixed and stained spore has an average dimension of 1.9-2.5 x 0.9-1.2 microm. The spores are oval with a characteristic surface layer, showing ornamentation-like projections, which are in close contact to the exospore. A short polar filament forming three to four coils traverses the polaroplast with two lamellar layers. The ultrastructure and other characteristic features of this microsporean parasite are distinct from those of the microsporean species described so far from oligochaetes.

The development of a sensitive, rapid and reliable nonradiometric cytotoxicity assay would significantly facilitate studies of teleost nonspecific cytotoxic cells. Such an assay would not require handling and disposal of radionuclides and it would not depend on secondary enzyme or colorimetric determinations. The requirements for this assay would consist of a one-step binding protocol which could detect early target cell membrane lesions and at very low effector:target cell ratios. In this chapter, we have developed a flow cytometry based cytotoxicity assay utilizing the uptake of propidium iodide (PI) into cells containing damaged (i.e. permeabilized) cell membranes. The basis of detection of cellular damage depended on flow discrimination of targets from effector cells by establishing 'scatter' gates from these mixtures. Teleost (catfish) anterior kidney NCC were mixed with human transformed targets (IM-9 and HL-60 cells) at effector:target cell ratios of 1, 5 and 10 and PI uptake was determined at 3 and 14 hours post-incubation. Percent specific uptake (PSU) was calculated by determining total binding capacity (TBC) (i.e. uptake of PI by cold acetone permeabilized target cells) and spontaneous binding capacity (SBC) (i.e. PI uptake by target cells incubated in media w/o effectors). This was represented by the formula PSU = [T − SBC/TBC − SBC] × 100 where T is the PI uptake of targets following addition of effector cells. Using this technique, NCC initiated target cell permeabilization as early as 30 minutes co-incubation (25:1 E:T ratio) and damaged membranes were detected in mixtures containing as few as 1:1 effector:target cell ratios. At 5:1 E:T ratios, greater than 50 PSU was determined following 14 hours co-incubation. Using these criteria, a new and sensitive cytotoxicity assay has been developed to determine NCC activity.

Inducible nitric oxide synthase (iNOS) inhibition was recently shown to exert no effect on allergen challenge in human asthma, raising serious concerns about the role of the protein in the disease. The present study investigated the role of iNOS in ovalbumin-induced eosinophilia from the perspective of its relationship with poly(ADP-ribose) polymerase-1 (PARP-1) and oxidative DNA damage. A mouse model of ovalbumin-induced eosinophilia was used to conduct the studies. iNOS-associated protein nitration and tissue damage were partially responsible for allergen-induced eosinophilia. iNOS expression was required for oxidative DNA damage and PARP-1 activation upon allergen challenge. PARP-1 was required for iNOS expression and protein nitration, and this requirement was connected to nuclear factor-kappaB. PARP-1 was an important substrate for iNOS-associated by-products after ovalbumin-challenge. PARP-1 nitration blocked its poly(ADP-ribosyl)ation activity. Interleukin-5 re-establishment in ovalbumin-exposed PARP-1(-/-) mice reversed eosinophilia and partial mucus production without a reversal of iNOS expression, concomitant protein nitration or associated DNA damage. The present results demonstrate a reciprocal relationship between inducible nitric oxide synthase and poly(ADP-ribose) polymerase-1 and suggest that expression of inducible nitric oxide synthase may be dispensable for eosinophilia after interleukin-5 production. Inducible nitric oxide synthase may be required for oxidative DNA damage and full manifestation of mucus production. Such dispensability may explain, in part, the reported ineffectiveness of inducible nitric oxide synthase inhibition in preventing allergen-induced inflammation in humans.

Summary Background Identifying therapeutic drugs that block the release or effects of T-helper type 2 (Th2) cytokines after allergen exposure is an important goal for the treatment of allergic inflammatory diseases including asthma. We recently showed, using a murine model of allergic airway inflammation, that poly(ADP-ribose) polymerase (PARP) plays an important role in the pathogenesis of asthma-related lung inflammation. PARP inhibition, by single injection of a novel inhibitor, thieno[2,3-c]isoquinolin-5-one (TIQ-A), before ovalbumin (OVA) challenge, prevented airway eosinophilia in C57BL/6 mice with concomitant suppression of Th2 cytokine production and mucus secretion. Objective To evaluate the efficacy of the drug when it is given after OVA challenge for its possible therapeutic potential. Methods This study was conducted using a murine model of allergic airway inflammation. Results A single injection of TIQ-A (6 mg/kg) one or 6 h post-allergen challenge conferred similar reduction in OVA challenge-induced eosinophilia. More significantly, post-allergen challenge administration of the drug exerted even better suppression on the production of IL-4, IL-5, IL-13, and IgE and prevented airway hyperresponsiveness to inhaled-methacholine. The significant decrease in IL-13 was accompanied by a complete absence of airways mucus production indicating a potential protection against allergen-induced airway remodelling. Conclusion The coincidence of the inflammation trigger and the time of drug administration appear to be important for the drug's more pronounced protection. The observed time window for efficacy, 1 or 6 h after allergen challenge may be of great clinical interest. These findings may provide a novel therapeutic strategy for the treatment of allergic airway inflammation, including asthma.

As respiratory syncytial virus (RSV) targets the mucosal surfaces of the respiratory tract, induction of both systemic and mucosal immunity will be critical for optimal protection. In this study, the ability of an intranasally delivered, formalin-inactivated bovine RSV (FI-BRSV) vaccine co-formulated with CpG oligodeoxynucleotides (ODN) and polyphosphazenes (PP) to induce systemic and mucosal immunity, as well as protection from BRSV challenge, was evaluated. Intranasal immunization of mice with FI-BRSV formulated with CpG ODN and PP resulted in both humoral and cell-mediated immunity, characterized by enhanced production of BRSV-specific serum IgG, as well as increased gamma interferon and decreased interleukin-5 production byin vitro-restimulated splenocytes. These mice also developed mucosal immune responses, as was evident from increased production of BRSV-specific IgG and IgA in lung-fragment cultures. Indeed, the increases in serum and mucosal IgG, and in particular mucosal IgA and virus-neutralizing antibodies, were the most critical differences observed between FI-BRSV formulated with both CpG ODN and PP in comparison to formulations with CpG ODN, non-CpG ODN or PP individually. Finally, FI-BRSV/CpG/PP was the only formulation that resulted in a significant reduction in viral replication upon BRSV challenge. Co-formulation of CpG ODN and PP is a promising new vaccine platform technology that may have applications in mucosal immunization in humans.

Alterations in the delicate balance between cell proliferation and cell death disrupt colon homeostasis and serve as determining factors in colon tumorigenesis. The two mouse strains, AKR/J (resistant) and A/J (susceptible), have been widely used as models for dimethylhydrazine-induced colon tumorigenesis. This study examined whether the differential susceptibilities of the two mouse strains to the tumorigenic effect of dimethylhydrazine were associated with intrinsic differences in the apoptotic machinery of the colon epithelial cells. While acute exposure to dimethylhydrazine caused massive apoptosis of colon epithelial cells in AKR/J mice, the effect was considerably less in A/J mice. Apoptosis in AKR/J mice occurred not only in the luminal side of the mucosa but also deep in the colonic crypts. In addition, this apoptosis appeared to involve caspase-3. The increased sensitivity of AKR/J to dimethylhydrazine was associated with a persistent expression of tumor necrosis factor (TNF) but not of its receptors. After establishing a new method for isolating primary colon epithelial cells, we determined that cells derived from A/J mice were substantially more resistant to apoptosis in response to dimethylhydrazine or to a combination of TNF, cyclohexamide, and butyrate compared to cells from AKR/J mice. These results strongly suggest that a higher intrinsic resistance to apoptosis of colon epithelial cells may be an important determinant of predisposition to colon tumorigenesis in the A/J mouse strain. © 2007 Wiley-Liss, Inc.

We recently used a murine model of allergic airway inflammation to show that poly(ADP-ribose) polymerase-1 (PARP-1) plays an important role in the pathogenesis of asthma-related lung inflammation. In this study, we show that PARP-1 inhibition, by a novel inhibitor (TIQ-A) or by gene deletion, prevented eosinophilic infiltration into the airways of OVA-challenged mice. Such impairment of eosinophil recruitment appeared to take place after IgE production. OVA challenge of wild-type mice resulted in a significant increase in IL-4, IL-5, IL-10, IL-13, and GM-CSF secretions. Although IL-4 production was moderately affected in OVA-challenged PARP-1−/− mice, the production of IL-5, IL-10, IL-13, and GM-CSF was completely inhibited in ex vivo OVA-challenged lung cells derived from these animals. A single TIQ-A injection before OVA challenge in wild-type mice mimicked the latter effects. The marked effect PARP-1 inhibition exerted on mucus production corroborated the effects observed on the Th2 response. Although PARP-1 inhibition by gene knockout increased the production of the Th1 cytokines IL-2 and IL-12, the inhibition by TIQ-A exerted no effect on these two cytokines. The failure of lung cells derived from OVA-challenged PARP-1−/− mice to synthesize GM-CSF, a key cytokine in eosinophil recruitment, was reestablished by replenishment of IL-5. Furthermore, intranasal administration of IL-5 restored the impairment of eosinophil recruitment and mucus production in OVA-challenged PARP-1−/− mice. The replenishment of either IL-4 or IgE, however, did not result in such phenotype reversals. Altogether, these results suggest that PARP-1 plays a critical role in eosinophil recruitment by specifically regulating the cascade leading to IL-5 production.

A functional relationship between the apoptotic endonuclease DNAS1L3 and the chemotherapeutic drug VP-16 was established. The lymphoma cell line, Daudi, exhibited a significant resistance to VP-16 treatment in comparison to the lymphoma/leukemia cell line, U-937. While U-937 cells degraded their DNA into internucleosomal fragments, Daudi cells failed to undergo such fragmentation in response to the drug. Activation of both caspase-3 and DNA fragmentation factor was not sufficient to trigger internucleosomal DNA fragmentation in Daudi cells. No correlation was found between expression levels of topoisomerase-II, Pgp, Bcl-2, Bax, or Bad and decreased sensitivity of Daudi cells to VP-16. Daudi cells failed to express DNAS1L3 and ectopic expression of this protein significantly sensitized the cells to VP-16. An enhancement of caspase-3 activity and collapse of mitochondrial membrane potential underlie DNAS1L3-mediated sensitization of Daudi cells to VP-16, which may be a direct result of DNAS1L3-mediated increase in PARP-1-activating DNA breaks after VP-16 treatment. Our results suggest that DNAS1L3 plays an active role in lymphoma cell sensitization to VP-16 and that its deficiency may constitute a novel mechanism of drug resistance in these cells.

We recently showed that PARP-1 plays a role in allergen (Ovalbumin)-induced airway eosinophilia, potentially through a specific effect on IL-5 production. Here we have explored the mechanism by which PARP-1 regulates IL-5 production using a murine model of allergic airway inflammation and splenocytes derived from wild type or PARP-1-/- mice. We show that PARP-1 knockout-associated reduction in IL-5 upon allergen exposure occurs at the mRNA level. Such an effect appears to take place after IL-4 receptor activation as PARP-1 inhibition exerted no effect on JAK1/JAK3 activation. STAT-6 protein was severely downregulated in spleens of PARP-1-/- mice without any effect on mRNA levels, suggesting an effect on protein integrity rather than gene transcription. Interestingly, the degradation of STAT-6 in PARP-1-/- mice required allergen stimulation. The dowregulation of STAT-6 coincided with mRNA and protein reduction of GATA-3 and occupancy of its binding site on the IL-5 gene-promoter. IL-4 was sufficient to induce STAT-6 downregulation in both PARP-1-/- mice and isolated splenocytes. Such degradation may be mediated by calpain, but not by proteasomes. Our results demonstrate a novel function of PARP-1 in regulating IL-5 expression during allergen-induced inflammation and explain the underlying mechanism by which PARP-1 inhibition results in IL-5 reduction.

The inducible nitric oxide synthase (iNOS) has been suggested to play an important role in the pathogenesis of asthma by contributing to eosinophilia and the ensuing oxidative damage after allergen exposure. IL-5 was shown to induce iNOS expression and that a reduction in iNOS expression reduces airway recruitment of eosinophils in response to allergen exposure. We recently showed that PARP-1 also plays a critical role in the pathogenesis of the disease by promoting eosinophil recruitment by participating in the cascade leading to IL-5 production. Here, we investigated the interrelationship between PARP-1 and iNOS in inducing eosinophilia. Our results show that iNOS gene deletion reduces eosinophil recruitment but to lesser degree as compared to PARP-1 gene deletion, suggesting a partial role for this protein in airway inflammation. PARP-1 gene deletion severely reduced iNOS expression in the airways of ovalbumin (allergen)-challenged mice and consequent nitration of target proteins and this phenomenon was closely associated with an impairment of p65 NF-κB nuclear translocation and phosphorylation of its inhibitor, I-κBα. IL-5-intranasal administration restored the eosinophilia in ovalbumin-challenged PARP-1−/− mice but did not reverse iNOS expression, suggesting that iNOS may be dispensable for eosinophil recruitment after IL-5 production. Altogether, our results further support the role of PARP-1 in the process of eosinophil recruitment during allergen-induced lung inflammation and provide evidence for a potential specific relationship between PARP-1 and iNOS. Identifying therapeutic drugs that block the release or effects of Th2 cytokines after allergen exposure is an important clinical goal for treatment of allergic inflammatory diseases such as asthma. An important portion of pre-clinical research is predominantly focused on prevention rather than treatment of symptoms associated with diseases. We recently showed, using a murine model of allergic airway inflammation, that poly(ADP-ribose) polymerase (PARP) plays an important role in the pathogenesis of asthma-related lung inflammation and that PARP inhibition, by a novel inhibitor (TIQ-A), prevented airway eosinophilia in ovalbumin-challenged mice. TIQ-A injection before ovalbumin challenge in C57BL/6 mice resulted in significant suppression of Th2 cytokines production including IL-4, IL-5, and IL-13 and secretion of mucus by airway goblet cells. In the present study, we show that a single injection of TIQ-A 1 or 6 hour after ovalbumin challenge conferred a marked reduction in ovalbumin-challenge-induced eosinophilia. The reduction in eosinohilia was accompanied with an even more pronounced suppression of IL-4, IL-5, and IL-13. Such suppression was closely associated with a more pronounced reduction in mucus production by airway goblet cells. The observed protection, as late as 6 hour after the allergen challenge, may be of a great clinical importance in our search towards novel therapeutic drugs in the treatment of asthma.
