This article focuses on teats and pacifiers as allergy risk of infants. At the Department of Occupational Dermatology, Karolinska Hospital, patients with suspected latex allergy are prick tested with extracts from latex gloves and with latex directly from the tree Heva brasiliensis. Less-sensitive RAST technique to identify IgE antibodies against latex was also used. Findings indicate a possible exposure of latex allergies early in life to infants, with a potential risk of developing IgE antibodies.

The fungicide chlorothalonil is used extensively under several tradenames for the protection of various horticultural and fruit crops and bananas against fungal infections. It is also used as fungicide in wood preservation and as a preservative in paints. Clinical experience has shown chlorothalonil to be a contact allergen and several cases of allergic contact dermatitis attributed to chlorothalonil have been described. 2 previous guinea pig maximization test studies have shown the sensitizing potential of chlorothalonil to be high. The sensitizing property of chlorothalonil was studied by us with the predictive test methods the local lymph node assay and the cumulative contact enhancement test. In the local lymph node assay, chlorothalonil induced a dose-dependent increase in proliferation with a maximal stimulation index of 19.2 and 27.2. In the cumulative contact enhancement test, a statistically significant dose-dependent high sensitization rate was seen with a maximal sensitization rate of 100%. In conclusion, it is evident that chlorothalonil is an extremely potent contact allergen, inducing sensitization using only topical exposure on intact skin.

The allergenicity of the preservative Euxyl K 400 and its principal allergen methyldibromo glutaronitrile (MDBGN) (1,2-dibromo-2,4-dicyanobutane) was investigated using 3 animal models; in mice, the local lymph node assay (LLNA) and in guinea pigs, the guinea pig maximization test (GPMT) and the cumulative contact enhancement test (CCET) with a dose-response protocol included. Previous attempts to define the sensitization capacity of these chemicals have given conflicting results. For comparison, the frequency and causes of positive patch test reactions to Euxyl K 400 and MDBGN were studied in patients referred to an occupational dermatology clinic. This investigation showed that Euxyl K 400 and MDBGN can give rise to contact allergy in man and that the relevant cases found mainly had similar exposure as non-occupational cases. A contact allergenic potential could be detected for MDBGN in 2 animal models, i.e., the CCET and the LLNA, and also for Euxyl K 400 in the LLNA. However, statistical analysis of the results from the GPMT with MDBGN failed to detect the sensitizing potential of this particular allergen. The results indicate that to be able to detect the allergenic potential of Euxyl K 400 and MDBGN, a predictive test method with multiple topical applications at induction is required. It is therefore important that an investigator is aware of the possibility of using various predictive test models for investigation of potential contact allergens.

The murine local lymph node assay is a method for predictive testing of contact allergenicity, but its ability to discriminate between allergens and irritants has been questioned. To explain some of the conflicting results with irritants, the proliferation induced by methyl salicylate and nonanoic acid, both considered to be non-sensitisers, was further investigated. Both substances showed a dose--response relationship and clearly positive results when tested at higher concentrations (> or = 50%) and would thus be classified as potential sensitisers according to the present criteria for a positive assay result. In the case of methyl salicylate, the use of either dimethyl formamide or methyl ethyl ketone as vehicle did not significantly influence the results. The negative results obtained for methyl salicylate in some earlier reports were probably due to testing at too low concentrations. The proliferation induced by irritants such as methyl salicylate and nonanoic acid and inter alia sodium dodecyl sulfate, Triton X-100, oxalic acid, chloroform/methanol (2:1) must be better recognized and elucidated before the assay can be generally accepted as a predictive test method.

The murine local lymph node assay is a new predictive test for identifying contact sensitizers. It measures the proliferative response in the lymph nodes during the sensitization phase. In the present study, moderate-to-extreme allergens (from human and guinea pig experience) gave clearly positive results in this assay. However, irritants tested, i.e. sodium dodecyl sulphate (SDS), chloroform/methanol, oxalic acid, triton X-100 and methylsalicylate, also gave positive results, not distinguishable from the results with low-grade/moderate allergens. Two allergens were also tested in the presence of 10% SDS. The effect on proliferation was additive in the first case and synergistic in the second. The local lymph node assay in its present design and with the criteria used for a positive response requires further validation studies and perhaps further development before it can be accepted as an alternative to guinea pig tests for allergenicity. Substances with exclusively irritating properties could falsely be classified as allergens by the method or, alternatively, the allergenicity of chemicals with both allergenic and irritating properties could be overestimated.

Polycyclic aromatic hydrocarbons, e.g., 7,12-dimethylbenz(a)anthracene (DMBA), cause various toxic effects in rat testis. To clarify the mechanism of action of DMBA in adult rat testis microsomes and mitochondria from this organ were investigated in vitro with respect to their capacity to metabolize DMBA. Qualitatively, both preparations showed DMBA-hydroxylase activities which were influenced by cytochrome P-450 inhibitors, chelators, and free-radical scavengers, suggesting that the DMBA metabolism was accounted for by different metabolic pathways in these organelles. Metabolism of DMBA was also accompanied by a pronounced covalent binding to both microsomal and mitochondrial protein, catalyzed primarily by a free-radical mechanism involving free or loosely bound iron which may involve superoxide anion shown to be generated by testis mitochondria. With microsomes covalent binding was markedly enhanced by added horseradish peroxidase but not by hydrogen peroxide whereas the mitochondrial binding was affected neither by added horseradish peroxidase nor by hydrogen peroxide. Antibodies raised against cytochrome P-450 c from rat liver inhibited the microsomal DMBA-hydroxylase but not the mitochondrial DMBA metabolism. It is concluded that the microsomal DMBA conversion and covalent binding are due to a mixture of cytochrome P-450 and free-radical-dependent metabolic pathways whereas the corresponding mitochondrial reaction is due mainly to a free-radical-dependent pathway. However, the data do not allow for a conclusion as to the quantitative importance of these pathways. It is proposed that both pathways may be important in DMBA-dependent testis toxicity but also in polycyclic aromatic hydrocarbon-dependent testis toxicity in general.

Rat adrenal cytosol was found to contain a 7,12-dimethylbenz[a]-anthracene (DMBA)-binding protein which is characterized by a pI of 7.2, a Kd-value of 3 microM and a maximal capacity of about 47 pmol/mg protein. The binding is highly specific for DMBA and is not displaced by 3-methylcholanthrene (MC), benz[a]pyrene (BP) or other polycyclic hydrocarbons. Likewise, various androgens, estrogens or glucocorticoids have no effect on the DMBA binding. It is proposed that the DMBA-binding protein may have a role in the toxic effects of DMBA or DMBA metabolites in adrenal and possibly in other DMBA-sensitive organs as well.

The metabolism of the carcinogenic and adrenocorticolytic polycyclic aromatic hydrocarbon 7,12-dimethylbenz(a)anthracene in rat adrenals was investigated. Both 7,12-dimethylbenz(a)anthracene and benzo(a)pyrene, which also is a well-known carcinogen but has no short-term effects on rat adrenals, appear to be metabolized by one common type of cytochrome P-450-dependent monooxygenase localized in the endoplasmic reticulum. Studies of the kinetic properties of this cytochrome P-450 reveal that the Km values for 7,12-dimethylbenz(a)anthracene and benzo(a)pyrene are lower than 3 microM. Identification of metabolites indicates that, with both 7,12-dimethylbenz(a)anthracene and benzo(a)pyrene, phenols and diols were formed the relative rates of formation of which were markedly influenced by the expoxide hydrase inhibitor cyclohexane oxide, suggesting that epoxides are intermediate metabolites. Added or endogenous microsomal glutathione S-transferase B had little or no effect on the distribution of metabolites. A rather selective binding of metabolites of 7,12-dimethylbenz(a)anthracene to soluble and microsomal proteins was demonstrated. The adrenal cytochrome P-450 involved in the conversion of these polycyclic aromatic hydrocarbons appears to be unrelated to those responsible for the synthesis of mineralocorticoids and glucocorticoids from cholesterol. Among androgens and estrogens, estradiol proved to be the most inhibitory steroid, suggesting a role of the hydrocarbon-metabolizing cytochrome P-450 in estrogen biosynthesis. However, no such function could be demonstrated conclusively. The metabolite patterns and the effects of nonsteroid inhibitors of liver monooxygenases, e.g., alpha-naphthoflavone, SU 9055, and ellipticine, suggest that the properties of this cytochrome P-450 resemble those of the 3-methyl-cholanthrene-inducible cytochrome P-488 from rat liver.

Cytochrome P-45011 beta has been solubilized and partially purified from bovine adrenal cortex mitochondria using chromatography on Octyl-Sepharose CL-4B or DEAE-Sepharose CL-6B. The partially purified P-450 preparations were about 90% pure as judged by SDS-polyacrylamide gel electrophoresis. In the presence of purified preparations of adrenodoxin reductase and adrenodoxin, the partially purified P-450 preparations catalyzed NADPH-supported 11 beta-hydroxylation of unconjugated and sulphoconjugated deoxycorticosterone. In presence of Triton X-100 the partially purified cytochrome P-45011 beta had a Stoke's radius of 4.5 nm, a sedimentation coefficient of 3.1 S and a partial specific volume of about 0.85 cm3/g. These results indicate that the cytochrome P-45011 beta-Triton X-100 complex has a molecular weight of about 100 000 and that P-45011 beta bound about 1.1 g of Triton X-100 per g of protein. The P-45011 beta-Triton X-100 complex was catalytically active in hydroxylation reactions supported by NADPH or the hydroxylating agent ortho-nitroiodosobenzene, suggesting that the monomer of cytochrome P-45011 beta is an active form of the protein.
