High quality substrates are a critical requirement for the epitaxial growth of high performance Group III-V (GaSb, InP, GaAs) or II-VI (MCT) based IR detector structures. Techniques such as MBE, LPE or MOCVD used to grow complex multilayer structures where the bulk and surface characteristics of the underlying substrate strongly influences the quality of the deposited device layers and thus the performance of the detector fabricated too. And whilst for Group III-V substrates, ‘epitaxy ready’ surfaces are very much the norm with no surface pre-treatment required prior to growth, much debate exists over whether it is possible to prepare Cadmium Zinc Telluride (CZT) substrates in the same way with commercially available substrates often etched or pre-polished prior to epitaxial growth. In this work we discuss the key factors which determine the ‘epi-readiness’ of a compound semiconductor substrate and identify the challenges which are associated with polishing ternary materials when compared with binary compounds. We also discuss the role which bulk CZT crystal quality plays on chemo-mechanical polish surface quality and the importance of substrate selection and screening steps to ensure that high quality substrates are obtained. We also compare the maturity level of infrared substrate materials such as GaSb with CZT and propose how CZT substrates can be chemo-mechanically polished in a production setting with the same tool sets as used to produce volume quantities of III-V compound semiconductor substrates in epitaxy ready form, as opposed to smaller scale, bespoke finishing processes. A forward looking view is given on the potential to align II-VI and III-V substrate manufacturing technologies in a way that supports product standardization and conformity at a time when demands for IR specification substrates is forecast to increase. Not only in quantity but also size where more stringent bulk and surface quality is required if state of the art, very large format detectors are to be delivered.

Cadmium Zinc Telluride (Cd1-xZnxTe or CZT) is a ternary II-VI compound semiconductor material that has been widely used in infrared detector applications for many years. Due to its lattice spacing, CZT is the substrate of choice for stabilizing Mercury Cadmium Telluride (Hg1-xCdxTe or MCT) crystal layer growth where the lattice matching reduces stress during detector growth processes for high performance infrared detectors and focal plane (FPA) arrays used in guidance systems and a wide array of IR applications. The manufacturing of high performance MCT IR detectors requires CZT substrates of high quality for both bulk and surface conditions thus enabling high quality MCT epitaxial layer crystallinity and low defectivity. In this work, we report on results on bulk CZT material grown using the Travelling Heater Method (THM) that are suitable for infrared focal plane array (IRFPA) detector applications. This proven crystal growth process has been used to manufacture CZT substrates meeting industry requirements of IR transmission, tellurium precipitate size, dislocations and of larger single crystal area. We will present results on chemomechanical (CMP) polishing of CZT substrates of square, rectangular and state-of-the-art round geometries utilizing standard production tool sets that are identical to those used to produce epitaxy-ready surface finishes on related IR compound semiconductor materials such as GaSb and InSb. Surface quality will be assessed by various analytical and microscopy techniques to validate the suitability of this material for epitaxial growth.

Monte Carlo analyses are generally considered the standard for uncertainty analysis. While accurate, these analyses can be expensive computationally. Recently, polynomial chaos has been proposed as an alternative approach to the estimation of uncertainty distributions (Hosder et al. A non-intrusive polynomial chaos method for uncertainty propagation in CFD simulations. In: 44th AIAA aerospace sciences meeting and exhibit, Reno, Nevada, 2006; Wu et al. Uncertainty analysis for parametric roll using non-intrusive polynomial chaos. In: Proceedings of the 12th international ship stability workshop, Washington, DC, USA, 2011). This approach works by representing the function as a series of orthogonal polynomials; the weights for which can be calculated via several methods. Previous studies have demonstrated the usefulness of this technique for comparatively simple systems such as parametric roll modeled by the Mathieu equation with normally distributed parameter values (Wu et al. Uncertainty analysis for parametric roll using non-intrusive polynomial chaos. In: Proceedings of the 12th international ship stability workshop, Washington, DC, USA, 2011). In the present work, a polynomial chaos method is applied to a nonlinear computational ship dynamics model with normally distributed input parameters. Test cases were selected where parametric roll was expected to potentially occur. The resulting probability distributions are compared to the results of a Monte Carlo analysis. In general, these results demonstrate good agreement between Monte Carlo simulation and polynomial chaos in the absence of capsize with significant computation gains found with polynomial chaos. Overall, we conclude that polynomial chaos is an effective tool for reducing simulation time costs when studying parametric roll, and potentially other ship dynamics phenomena, particularly in the absence of capsize-like bifurcations.

A cryogenic apparatus is described that enables a new experiment, nEDM@SNS, with a major improvement in sensitivity compared to the existing limit in the search for a neutron Electric Dipole Moment (EDM). This apparatus uses superfluid 4He to produce a high density of Ultra-Cold Neutrons (UCN) which are contained in a suitably coated pair of measurement cells. The experiment, to be operated at the Spallation Neutron Source at Oak Ridge National Laboratory, uses polarized 3He from an Atomic Beam Source injected into the superfluid 4He and transported to the measurement cells where it serves as a co-magnetometer. The superfluid 4He is also used as an insulating medium allowing significantly higher electric fields, compared to previous experiments, to be maintained across the measurement cells. These features provide an ultimate statistical uncertainty for the EDM of 2-3× 10-28 e-cm, with anticipated systematic uncertainties below this level.

Novel experimental techniques are required to make the next big leap in neutron electric dipole moment experimental sensitivity, both in terms of statistics and systematic error control. The nEDM experiment at the Spallation Neutron Source (nEDM@SNS) will implement the scheme of Golub & Lamoreaux [Phys. Rep., 237, 1 (1994)]. The unique properties of combining polarized ultracold neutrons, polarized $^3$He, and superfluid $^4$He will be exploited to provide a sensitivity to $\sim 10^{-28}\,e{\rm \,\cdot\, cm}$. Our cryogenic apparatus will deploy two small ($3\,{\rm L}$) measurement cells with a high density of ultracold neutrons produced and spin analyzed in situ. The electric field strength, precession time, magnetic shielding, and detected UCN number will all be enhanced compared to previous room temperature Ramsey measurements. Our $^3$He co-magnetometer offers unique control of systematic effects, in particular the Bloch-Siegert induced false EDM. Furthermore, there will be two distinct measurement modes: free precession and dressed spin. This will provide an important self-check of our results. Following five years of "critical component demonstration," our collaboration transitioned to a "large scale integration" phase in 2018. The higher nutrient requirements of lactating dairy cows may cause hunger, and therefore the motivation to consume feed, to increase with milk yield. An experiment was conducted that aimed to establish whether dairy cows could be trained to obtain a food reward in response to lever pressing and, if so, whether high-yielding dairy cows had a greater hunger than low-yielding cows. As a measure of hunger the demand to consume feed was evaluated. In total, 7 high- and 6 low-yielding dairy cows were trained in 2 groups to use a lever to obtain 0.33kg concentrate rewards. For at least a 9-day period each cow was tested for 30min/day by increasing the number of lever presses required to receive each additional reward in a progressive ratio of 3. After the test cows were allowed to return to their quarters, where they could consume a total mixed ration (TMR). Initially high-yielding cows had lower maximum number of lever presses than low-yielding cows, possibly so that they could consume TMR after finishing the test, but from Days 6–9 the number for high-yielding cows increased until it was the same as low-yielding cows. There was a positive correlation between the maximum lever presses by high-yielding cows and their time in the test pen, which was not evident for low-yielding cows. The high-yielding cows spent more time feeding on the TMR after the test than low-yielding cows. Therefore, although the high-yielding cows were not prepared to work harder for food rewards, they did seem to adopt a more efficient feeding strategy by only spending time in the test pen if they were still receiving rewards and minimizing time without feed by eating more post-test.

The precession of (3)He spins is detected with ultra-low field NMR. The absolute strength of the NMR signal is accurately measured and agrees closely with theoretical calculations. The sensitivity is analyzed for applications to a neutron electric dipole moment (nEDM) fundamental symmetry experiment under development.

Three experiments examined the extent of behavioral interactions between housed dairy cows. In Experiment 1, 6 different cows were selected every other day from a dairy herd and allocated to 3 pairs for recording of feeding, standing, lying, and leg-raising behaviors. There was no difference in the between-pair and within-pair coefficients of variation for the behaviors, suggesting that the performance of these behaviors was independent of influences of other cows. Experiment 2 used 9 pairs of cows in a 3-period changeover design. The same behaviors were recorded as in Experiment 1, and data were used only from the last 2 periods, which ensured that cows had been together previously for at least one 48-hour period. There were again no differences in the between- and within-pair coefficients of variation when using behaviors as replicates, either for the first 2 hours, during which time no treatments had been applied, or the entire 4 hours, when some cows had been prevented from lying down and feeding. Experiment 3 investigated the consistency of between-cow associations for repeated observations of feeding, lying, and entering the milking parlor. Although there was evidence of a similar order of entry into the milking parlor between days, the number of feeding associations was similar to that predicted by a model of random behavior, and the pattern of lying associations also suggested a random pattern of associations. Across behaviors, cows were observed with specific partners more often than expected by chance. Thus although there was no evidence of interdependence of feeding, lying, or standing behavior, consistent associations were observed, particularly in parlor entry.

Two experiments were conducted to examine the effects of depriving dairy cows of the ability to feed and lie down for short periods, on behaviour and production. In experiment 1, cows were deprived by confining them in pairs in a pen for 2 or 4 h, and they more frequently exhibited behaviour likely to suggest discomfort - leg stamping, repositioning themselves, shifting their weight between legs and butting. After deprivation, the cows deprived for 2 h made up their lost feeding time within 24 h, but cows deprived for 4 h did not restore their feeding time within the 41-h period of observation. Lying time was not restored in either treatment within the 41-h period. Milk yield was not affected by the treatment. However, in experiment 2, when cows were deprived of feeding and lying for 4 h, during which time their hooves were trimmed (which is likely to be a painful and stressful procedure and result in some discomfort for a period post-trimming) the evidence suggested that milk yield was reduced by approximately 2 l/day for 3 days, with corresponding increases during the subsequent 2 days. Walking speed on returning to the herd was the same as before the treatment. In summary, temporary deprivation of feeding and lying for 2 and 4 h/day induced behaviours that were indicative of discomfort and frustration but had no negative effect on milk production, except when 4 h of deprivation was accompanied by foot trimming.

The Spallation Neutron Source (SNS), currently under construction at Oak Ridge National Laboratory with an anticipated start-up in early 2006, will provide the most intense pulsed beams of cold neutrons in the world. At a projected power of 1.4 MW, the time averaged fluxes and fluences of the SNS will approach those of high flux reactors. One of the flight paths on the cold, coupled moderator will be devoted to fundamental neutron physics. The fundamental neutron physics beamline is anticipated to include two beam-lines; a broad band cold beam, and a monochromatic beam of 0.89 nm neutrons for ultracold neutron (UCN) experiments. The fundamental neutron physics beamline will be operated as a user facility with experiment selection based on a peer reviewed proposal process. An initial program of five experiments in neutron decay, hadronic weak interaction and time reversal symmetry violation have been proposed.

This paper reports on the design, construction, and operational characteristics of a delay-line readout implemented on the cathode foils of a cylindrical drift chamber system. The readout was used to determine the position of an event along the length of the 1.74 m drift wires in the MEGA detectors used at the Los Alamos Meson Physics Facility. The performance of the system is interpreted by comparison to a PSPICE simulation, and to simple analytical models.

An experiment has been performed to search for the muon- and electron-number non-conserving decay mu+ to e+_gamma. The upper limit for the branching ratio to be GAMMA(mu+ to e+_gamma)/GAMMA(mu+ to e+_nu_nubar) < 1.2e-11 with 90% confidence.

A new experiment has been proposed at Los Alamos National Laboratory to measure the neutron electric dipole moment (EDM) to 4{times}10{sup {minus}28} ecm, a factor of 250 times better than the current experimental limit. Such a measure of the neutron EDM would challenge the theories of supersymmetry and time reversal violation as the origin of the observed cosmological asymmetry in the ratio of baryons to antibaryons. One possible design for this new experiment includes the use of LTC SQUIDs coupled to large ({approximately}100 cm{sup 2}) pick-up coils to measure the precision frequency of the spin-polarized {sup 3}He atoms that act as polarizer, spin analyzer, detector, and magnetometer for the ultra-cold neutrons used in the experiment. The method of directly measuring the {sup 3}He precession signal eliminates the need for very uniform magnetic fields (a major source of systematic error in these types of experiments). It is estimated that a flux of {approximately}2{times}10{sup {minus}16} Tm{sup 2} (0.1 {Phi}{sub 0}) will be coupled into the pick-up coils. To achieve the required signal-to-noise ratio one must have a flux resolution of d{Phi}{sub SQ} = 2{times}10{sup {minus}6}{Phi}{sub 0}/{radical}Hz at 10 Hz. While this is close to the sensitivity available in commercial devices, the effects of coupling to such a large pick-up coil and flux noise from other sources in the experiment still need to be understood. To determine the feasibility of using SQUIDs in such an application the authors designed and built a superconducting test cell, which simulates major features of the proposed EDM experiment, and they developed a two-SQUID readout system that will reduce SQUID noise in the experiment. They present an overview of the EDM experiment with SQUIDs, estimations of required SQUID parameters and experimental considerations. The authors also present the measured performance of a single magnetometer in the test cell as well as the performance of the two SQUID readout technique.

A design for extremely low mass, high-resolution multiwire proportional chambers (MWPC) was achieved by the MEGA collaboration in its experiment to search for the lepton family number violating decay μ→eγ. To extend the present branching ratio limit by over an order of magnitude, these MWPCs were operated in high particle fluxes. They showed minimal effects of aging, and evidenced spatial and energy resolutions for the orbiting positrons from muon decay which were consistent with our design parameters. The unique features of these chambers, their assembly into the MEGA positron spectrometer, and their performance during the experiment are described in this paper.

In the production of ultracold neutrons (UCN) by scattering cold neutrons in superfluid 4He or cold solid D2 (“superthermal” processes), the UCN production rate is proportional to the spatial density of cold neutrons in the material. For a superthermal source fed from a cold neutron guide, we show that the rate of UCN production can be increased by up to a factor of about 2.3 for realistic substances by enclosing the source in a cavity made of a material with a high cold neutron albedo.

The connection of rare decays to supersymmetric grand unification is highlighted, and a brief review of the status of rare decay experiments is given. The status of the MEGA experiment, a search for μ + → e + γ , is reported. Some ideas for a new experimental arrangement that has the potential to reach a sensitivity of 10 −14 are presented.

The purpose of this study was to evaluate effects of hyperbaric oxygen (HBO) and penicillin (PCN) therapy in a murine model of streptococcal myositis. The thighs of Swiss Webster mice were inoculated with Streptococcus pyogenes. Four groups were evaluated : (1) control (N = 10), (2) HBO treatment (N = 10), (3) PCN treatment (N = 8), and (4) PCN and HBO treatment (N = 6). Mortality (day of death) and the number of colony-forming units (cfu) were measured. PCN significantly lowered cfu from control (p < 0.05). Cfu in group 4 was significantly lower than PCN alone (p = 0.006). Survival was significantly longer in the PCN group compared with the control (p < 0.01). Survival in the combined treatment group was significantly longer than PCN alone (p < 0.01). These results suggest that (1) HBO treatment alone does not decrease mortality or bacterial proliferation in vivo significantly, (2) PCN therapy alone improves outcome significantly, and (3) the combined treatment of PCN and HBO exerts at least additive effects in both decreasing bacterial counts in vivo and increasing survival in this model.

The possibility exists for improving the present experimental constraint on the tensor coupling constant in radiative pion decay by at least a factor of six. The method emphasizes relative decay probabilities rather than absolute rates.

The trigger for the MEGA detector system is based on signals from single, high-energy photons interacting in one of the three MEGA pair spectrometers. The trigger is divided into a fast and a slow stage. The first stage produces a fast output if a specific pattern of detector hits is observed in the scintillators and high-speed wire chambers of a pair spectrometer. The second, slow-stage interrogates drift chamber hit patterns and provides a veto when the pattern fails a minimal requirement for reconstruction of the hits into a pair of circular orbits. The trigger interacts with the photon detector electronics by gating limited sections of the detector during the read-out of an event. This paper describes the two stage trigger system, the photon detector electronics, and the implementation of the trigger outputs to strobe the data acquisition system. The performance of the trigger is compared to Monte Carlo simulations of the photon detector response.

This analysis of B cell development as a function of age reveals a relatively widespread distribution of progenitor B (pro-B), pre-B, and B cells in fetal tissues, and thus supports the idea of a multifocal origin of B lineage cells during embryonic development. From mid-gestation onward, the bone marrow is the major site of B cell generation in humans. A relatively constant ratio of bone marrow precursors to B cells of immature phenotype (CD24highCD10+CD20lowIgD-) is maintained from mid-gestation through the eighth decade of life. The persistence of recombinase gene activity in pro-B cells further attests the sustained production of B cells in bone marrow. Interestingly, a subpopulation of B cells with mature phenotype (CD24lowCD10-CD20highIgD+) accumulates in the bone marrow during childhood, and this becomes the predominant B cell subpopulation in adult bone marrow. This mature population of bone marrow B cells may represent a subpopulation of recirculating B cells that have undergone selection in the periphery.

We identified a novel T cell Ag in the South African clawed toad (Xenopus laevis) by a mAb designated 2B1. This Ag is present in relatively high levels on most thymocytes, approximately 65% of splenocytes, 55% of PBL, and 65% of intestinal lymphocytes, but is rarely seen on IgM+ B cells in any of these tissues. Lymphocytes bearing the 2B1 Ag proliferate in response to stimulation with Con A or PHA, whereas the 2B1- lymphocytes are reactive to LPS. Biochemical analysis indicates that this Ag is a differentially phosphorylated glycoprotein of 71 to 82 kDa. The protein core of 64 kDa bears both N- and O-linked carbohydrate side chains. The amino-terminal protein sequence of the 2B1 Ag shares significant homology with both the macrophage scavenger receptor type 1 motif and the mammalian CD5/CD6 family. The biochemical characteristics and cellular distribution of the 2B1 Ag suggest that it represents the CD5 homologue in X. laevis. While T cells constitutively express this highly conserved molecule, Xenopus B cells acquire the CD5 homologue only when they are stimulated in the presence of T cells.

Endoglin, a glycoprotein that is expressed by human endothelial cells, binds TGF-beta 1 and -beta 3 with high affinity. It was originally identified with the 44G4 mAb that was produced against a human pre-B cell line. We now report that another anti-pre-B cell mAb, 29-G8, reacts with pro-B and pre-B leukemic cells, but not with mature B and T cells, and recognizes a different epitope of endoglin. The 29-G8 mAb bound specifically to recombinant endoglin and immunoprecipitated a phosphorylated homodimeric glycoprotein with subunits of M(r) 95,000 from the 697 pre-B cell line. This new Ab removed all of the molecules identified by the prototypic 44G4 anti-endoglin Ab, but the reverse was not true. A subpopulation of 29-G8+ endoglin molecules on this pre-B cell line was unreactive with the 44G4 mAb, thus suggesting that these anti-endoglin Abs see different epitopes that may discriminate different species of endoglin molecules. Flow cytometric analysis with the 29-G8 mAb revealed two endoglin-positive subpopulations in fetal bone marrow: early B-lineage precursor cells (CD19+ and CD34+), and proerythroblasts (CD71+ and glycophorin A+). In adult bone marrow, only the proerythroblast subpopulation was observed. Stromal cells derived from fetal bone marrow also reacted strongly with the 29-G8 and 44G4 Abs, and these cells responded with enhanced proliferation after stimulation with either TGF-beta 1 or the anti-endoglin Abs. Thus, endoglin, a specialized component of the TGF-beta receptor system, may play a physiologic role in the stromal-hemopoietic cell interactions occurring during development.

Goal-setting and feedback techniques, previously used in a study to improve safety in the UK construction industry, were applied to a three-shift production plant, employing approximately 540 people. Critical safety behaviours were identified using accident records and ‘in-depth' interviews. Checklists of critical behaviours were developed for each of 14 departments in the factory. Forty-eight observers were trained to observe their colleagues' safety performance and complete the checklists. ‘Baseline’ measures were taken over a four- to six-week period. Participative goal setting was used to set safety improvement ‘goals’ for the critical behaviours, within each department. Performance feedback was presented graphically in each department on a weekly basis. The results indicate significant improvements in safety performance, with a corresponding reduction in the plant's accident rate. Paradoxically, a statistical inverse relationship was not obtained between safety performance and accident rates, although the importance of non-safety variables in accident causation was demonstrated.

The construction techniques used in the development of a set of large pair spectrometers for the MEGA detector are discussed. These spectrometers consist of carbon-fiber-composit cylinders on which the conversion foils are mounted, and appropriately spaced wire chambers in each layer which track the conversion leptons. Close tolerances on all dimensions are maintained. The detector has been operating well in the stopped muon beam line at LAMPF.

Virgin and memory T cells reciprocally express high levels of the RA or the RO isoforms of CD45, respectively. In an examination of T cell expression of these two CD45 isoforms during human development, the RO+RA- "memory" T cells were infrequent in the newborn blood and spleen, but comprised approximately half of the T cells in adult tissues. These anticipated findings probably reflect the immunologic naivete of the newborn. Surprisingly, however, RO+RA- T cells were relatively abundant in fetal spleen and in cord blood samples from premature births, comprising approximately 25% and 10% of the T cells in these tissues, respectively. This early peripheral wave of RO+RA- T cells was composed of polyclonal T cells in both the CD4 and CD8 subpopulations. The fetal RO+ cells of CD4+ phenotype frequently expressed the CD25-alpha chain subunits that characterize high affinity IL-2 receptors, and were able to proliferate in response to exogenous IL-2. In further contrast with their RO+ memory T cell counterparts in adults, the fetal T cells were unresponsive to mitogenic anti-CD2 and anti-CD3 antibodies. We conclude that the data suggest an embryonic population of autoreactive T cell clones with anergic characteristics.

Safety improvement in the construction industry will only be achieved if all concerned in the operation of construction sites change their behaviour. This article describes the development and effects of behaviourally-based management techniques in improving site safety. Goal-setting and feedback methods were developed and tested on six construction sites in the Northwest of England. A longitudinal research design was adopted, in which measures of safety performance were taken before, during and after the application of these methods. The measures included four categories of measurement: access to heights; site housekeeping (tidiness); scaffolding; and use of personal protective equipment. Three of these categories were used for experiments with a variety of goal-setting and feedback interventions, while the fourth was used as a control. The existence of an IgM receptor on human T cells has been suggested by T cell rosetting with IgM-coated erythrocytes. In this study we used immunofluorocytometry to demonstrate that 1) after short-term culture, a majority of the T cells can bind IgM at easily detectable levels, 2) human and mouse IgM preparations bind to human T cells but other Ig isotypes do not, 3) the IgM binding is saturable and inhibitable only by Ig of IgM isotype and 4) the Fc portion of IgM is involved in the binding to a protease-sensitive cell surface protein. Biochemical analysis of the T cell receptor for IgM reveals a cell surface protein of 60 kDa in comparison with the 58 kDa Fc mu receptors (Fc mu R) on B-lineage cells. Although Fc mu R expression is up-regulated after B cell activation, the reverse is true after T cell activation. In addition, the T cell Fc mu R is relatively resistant to phospholipase C treatment. These results indicate that T- and B-lineage cells express Fc mu R with different biologic characteristics.

Objectives: Mathematical models are useful for studying vascular and avascular tumours, because these allow for more logical experimental design and provide valuable insights into the underlying mechanisms of their growth and development. The processes of avascular tumour growth and the development of capillary networks through tumour‐induced angiogenesis have already been extensively investigated, albeit separately. Despite the clinical significance of vascular tumours, few studies have combined these approaches to develop a single comprehensive growth and development model. Materials and methods: We develop a continuum‐based mathematical model of vascular tumour growth. In the model, angiogenesis is initiated through the release of angiogenic growth factors (AGFs) by cells in the hypoxic regions of the tumour. The nutrient concentration within the tumour reflects the influence of capillary growth and invasion induced by AGF. Results and conclusions: Parametric and sensitivity studies were performed to evaluate the influence of different model parameters on tumour growth and to identify the parameters with the most influence, which include the rates of proliferation, apoptosis and necrosis, as well as the diffusion of sprout tips and the size of the region affected by angiogenesis. An optimization was performed for values of the model parameters that resulted in the best agreement with published experimental data. Fc alpha receptors (Fc alpha R) were isolated from a human monocytic cell line and used to raise four mAb with receptor specificity. The antibodies were used to identify the types of white blood cells that express Fc alpha R and the molecular heterogeneity of the receptor molecules. Nonpolymorphic epitopes, outside of the Fc alpha-binding site, were recognized only on blood cells of granulocyte and monocyte/macrophage lineages. The molecules identified, both by the antibodies and by the IgA ligand, were glycoproteins ranging in relative molecular mass from 55 to 75 kDa. However, one antibody detected a subpopulation of Fc alpha R molecules characterized by relatively restricted size heterogeneity. A complex glycosylation pattern was revealed by the resolution of discrete 32- and 36-kDa molecular species after removal of N-linked oligosaccharides and by evidence for O-linked carbohydrate moieties on at least a portion of the Fc alpha R molecules. In biosynthetic studies, all four anti-Fc alpha R antibodies and the IgA ligand bound a single 32-kDa core protein present in tunicamycin-treated cells, and the exceptional antibody again recognized molecules with relatively restricted glycosylation in the nontreated cells. These antibodies and native IgA ligands thus provide complementary reagents for definition of the complex structure and function of Fc alpha R in systemic IgA antibody responses.

The predicted amino acid sequence of the cDNA encoding the murine B-lymphocyte differentiation antigen BP-1/6C3 suggested that it is a member of the zinc-dependent metalloprotease family, possibly an aminopeptidase related to aminopeptidase N [microsomal aminopeptidase; alpha-aminoacyl-peptide hydrolase (microsomal), EC 3.4.11.2]. In the present studies, we examined the enzymatic activity of this antigen. From brush border preparations of the small intestine, a rich source of many endopeptidases and exopeptidases, the BP-1 antibody selectively removed aminopeptidase A [APA; L-alpha-aspartyl(L-alpha-glutamyl)-peptide hydrolase, EC 3.4.11.7] activity. The APA activity of a panel of cell lines correlated in linear fashion with cell-surface levels of the BP-1/6C3 antigen. APA activity was demonstrated for the BP-1/6C3 antigen immunopurified from the pre-B-cell membrane. This activity was enhanced by alkaline earth metals such as Ca2+ and was abrogated by amastatin and angiotensin, which are known competitive inhibitors of APA. The data indicate that the murine BP-1/6C3 antigen is active APA, an enzyme that catalyzes specifically the removal of unsubstituted, N-terminal glutamic acid and aspartic acid residues from peptides.

Avian homologues of mammalian gamma delta and alpha beta TCR, termed TCR1 and TCR2, have been identified in the chicken with specific mAb. A third TCR, dubbed TCR3, has been identified on a subpopulation of T cells that lack the TCR1 or TCR2 epitopes. We have now produced a mAb that identifies this TCR3 molecule. The anti-TCR3 antibody immunoprecipitates a CD3-associated heterodimer with a relative Mr of 88,000, composed of 48,000 and 40,000 disulfide-linked chains. The Mr 40,000 chains of TCR3 and TCR2 exhibited the same isoelectric points of 5.6 to 6.5 and had core proteins of 34,000. Although the Mr 48,000 chain of TCR3 and the Mr 50,000 chain of TCR2 had the same basic isoelectric point of 6.2 to 7.6, their core proteins were different in size, 31,000 vs 29,000. Immunofluorescence analysis reveals that the TCR3 was present on all of the CD3+ T cells not identified by antibodies specific for TCR1 or TCR2. Thymocytes that expressed the surface CD3/TCR3 complex at relatively low levels were predominantly CD4+ and CD8+, whereas those with higher levels of surface CD3/TCR3 were predominantly CD4+ and CD8+ singles. Mature TCR3+ cells in the periphery were also either CD4+ (80%) or CD8+ (20%). The TCR1+, TCR2+, and TCR3+ subsets of T cells were generated sequentially in the thymus and seeded to the periphery in the same order. Intrathymic development of the TCR3+ cells was selectively inhibited by embryonic treatment with the anti-TCR3 mAb. The pattern of histologic localization of TCR3+ cells in the periphery was similar to the TCR2 subset of cells except that the TCR3+ cells were rarely seen in the intestine. Cross-reactivity patterns of the anti-chicken TCR antibodies suggested that other gallinaceous species share the three types of TCR. We conclude that TCR2 and TCR3 in gallinaceous birds may represent alpha beta subfamilies of TCR that are sequentially expressed on developmentally discrete sublines of T cells.

In a study of T cell ontogeny using monoclonal antibodies specific for chicken T cell receptors (TcR) and associated cell surface molecules, we found a subset of lymphocytes that express cytoplasmic CD3 epitopes in the absence of cell surface CD3/TcR complexes. Approximately half of these cells, which were present in the spleen, bursa and intestine of young chick embryos, expressed surface CD8. None expressed CD4, TcR 1 (gamma/delta), TcR 2 (alpha/beta) or TcR 3 (a third CD3-associated heterodimer in the chicken). These cytoplasmic CD3+CD8 +/- cells, tentatively named TcR 0 cells to denote their lack of surface TcR, appeared first in the spleen of 8-day embryos, 4 days before TcR expression begins in the thymus, and reached a peak frequency of approximately 10% of the splenic cell in 14-day embryos. The TcR 0 cells were also present in adult birds, where they comprised only about 1% of the CD3+ spleen cells and approximately 40% of the lymphocytes in the intestinal epithelium. We conclude that the avian TcR 0 cells represent a thymus-independent lineage of lymphocytes which, like natural killer cells in mammals, may play an important role in body defense.

A panel of murine mAb against chicken TCR and associated molecules was used to study the ontogeny of T cells. The intrathymic maturation of the TCR-gamma delta, (TCR-1) and TCR-alpha beta (TCR-2) sublineages was the focus of these studies employing immunoperoxidase staining of tissue sections and immunofluorescence analysis of cell suspensions. The first CD3+ cells appeared in the thymus on embryonic day 9 (E9) when the CD3 Ag was restricted to the cytoplasm. In tissue sections, both TCR-1+ and TCR-2+ cells were observed on E12, whereas only the TCR-1 cells were identifiable by surface immunofluorescence. On the next day, when a discrete thymic medullary region was first recognizable, the TCR-1 cells were present in both cortex and medulla. Two days later (E15), TCR-1 cells were found in the spleen. Surface TCR-2+ cells did not appear until E14, began to migrate in to the medulla on E17, and appeared in the spleen on E19. The first TCR-1 cells thus move quickly through this maturational pathway, whereas TCR-2 cells undergo a prolonged developmental period in the cortex. While most TCR-1+ cells were CD4-CD8-, a minor subpopulation (5 to 15%) were CD4-CD8+, and less than 1% were CD4+CD8+. In contrast, immature TCR-2+ thymocytes in the cortex were predominantly CD4+CD8+, whereas cells expressing a higher density of the CD3/TCR-2 complex were either CD4+CD8- or CD4-CD8+ and were localized in the thymic medulla. In the medulla of the mature thymus, the TCR-1+ cells preferentially occupy the cortico-medullary junction and form small aggregates around vessels. TCR-2+ cells were less frequent in these areas of TCR-1 accumulation. The thymic ontogeny and, by implication, the selection of the receptor repertoire thus differs substantially for these two TCR isotypes.

Cows are often temporarily deprived of the opportunity to lie down while waiting for veterinary or reproductive procedures. Sixty cows were deprived of the opportunity to lie down for 0, 2, or 4 h by confining them in pairs in a small indoor pen. Behavior was recorded during deprivation and for 40 h afterwards. In the first 2 and 4 h of the experiment, cows that were not deprived chose to lie down for 70 and 142 min, respectively. When cows were discouraged from lying, they regularly stomped their legs, repositioned themselves, but never lay down. In the 4-h treatment, both stomping and repositioning increased after the first hour. Butting and weight shifting (displacing weight from one side of the body to the other) increased during deprivation, indicating restlessness. Cows deprived for 4 h sniffed and rubbed their heads against the housing more than cows deprived for 0 or 2 h. Time spent feeding and standing without ruminating increased with the duration of deprivation, especially during the early stages; standing ruminating also increased in the final stages. After deprivation, feeding time decreased, which compensated for the increase during deprivation. By 40 h after deprivation, the lying-deprived cows had recovered approximately 40% of their lost lying time. Milk yield was not affected by lying deprivation. It is concluded that cows experience discomfort during short periods of lying deprivation, after which they recover some, but not all, of the lost lying time by rescheduling feeding and standing time.

An international collaboration has undertaken a project to search for the electric dipole moment (EDM) of the neutron with a sensitivity of 10−28 e⋅cm. The search should span a region of sensitivity where effects are expected from supersysmmetric models and from models of time reversal violation that might explain electroweak baryogenesis. The technique utilizes the special properties for producing and storing ultra‐cold neutrons in a bottle filled with liquid 4He and doped with a small amount of 3He. The technique will be discussed and compared to that of competing experiments. The extensive R&D program undertaken by the collaboration will be reviewed. The status of the DOE project will be explained.

We describe a neutron radiography technique that can be used to map the distribution of $^{3}\mathrm{H}\mathrm{e}$ impurities in liquid $^{4}\mathrm{H}\mathrm{e}$, providing direct and quantitative access to underlying transport processes. Images reflecting finite normal- and superfluid-component $^{4}\mathrm{H}\mathrm{e}$ velocity fields are presented.

The gene for one of the activating members of the paired Ig-like receptor family, Pira6, was isolated from a genomic library and sequenced. The first of 9 exons in the approximately 8.2 kb Pira6 gene encodes the 5' untranslated region, the translation initiation site, and approximately half of the signal sequence. The second exon encodes the rest of the signal sequence, exons 3-8 each encode a single Ig-like extracellular domain, and exon 9 encodes the transmembrane region, cytoplasmic tail and 3' UTR with four polyadenylation signals and six mRNA instability sequences. A soluble form of PIR-A6 may be generated by alternative splicing. The exonic sequences account for approximately 42% of the Pira6 gene and approximately 34% for the single inhibitory Pirb gene, thus defining Pira and Pirb as genes with relatively short intronic sequences. Extensive sequence homology was found between Pira6 and Pirb from approximately 2 kb upstream of the ATG initiation site to the beginning of intron 8. The Pir genes appear to be distributed in three regions of the proximal end of chromosome 7 based on the present data and an analysis of currently available mouse genomic sequence databases. One region contains a single Pir gene which is almost identical to Pira6, and the other two contain multiple Pir genes in opposite transcriptional orientations. Potential binding sites for hemopoiesis-specific and ubiquitous transcription factors were identified upstream of the Pira6 transcription start sites that reside within the initiator consensus sequence motif. These results provide important clues to the coordinate regulation observed for PIR-A and PIR-B expression during hematopoiesis.

The MEGA experiment, which searched for the muon- and electron-number violating decay ${\ensuremath{\mu}}^{+}\ensuremath{\rightarrow}{e}^{+}\ensuremath{\gamma},$ is described. The spectrometer system, the calibrations, the data taking procedures, the data analysis, and the sensitivity of the experiment are discussed. The most stringent upper limit on the branching ratio, $\mathcal{B}({\ensuremath{\mu}}^{+}\ensuremath{\rightarrow}{e}^{+}\ensuremath{\gamma})l1.2\ifmmode\times\else\texttimes\fi{}{10}^{\ensuremath{-}11}$ with $90%$ confidence, is derived from a likelihood analysis.

We have measured the mass diffusion coefficient D of 3He in superfluid 4He at temperatures lower than were previously possible. The experimental technique utilizes scintillation light produced when neutron react with 3He nuclei, and allows measurement of the 3He density integrated along the trajectory of a well-defined neutron beam. By measuring the change in 3He density near a heater as a function of applied heat current, we are able to infer values of D with 20% accuracy. At temperatures below 0.7 K and for concentrations of order 10^{-4} we find D=(2.0+2.4-1.2)T^-(6.5 -/+ 1.2) cm^2/s, in agreement with a theoretical approximation., 8 pages, 5 figures. The potential of the paired Ig-like receptors of activating (PIR-A) and inhibitory (PIR-B) types for modifying an IgE antibody-mediated allergic response was evaluated in mouse bone marrow-derived mast cells. Although mast cells produced both PIR-A and PIR-B, PIR-B was found to be preferentially expressed on the cell surface, where it was constitutively tyrosine phosphorylated and associated with intracellular SHP-1 protein tyrosine phosphatase. PIR-B coligation with the IgE receptor (FcepsilonRI) inhibited IgE-mediated mast cell activation and release of serotonin. Surprisingly, the inhibitory activity of PIR-B was unimpaired in SHP-1-deficient mast cells. A third functional tyrosine-based inhibitory motif, one that fails to bind the SHP-1, SHP-2, and SHIP phosphatases, was identified in parallel studies of FcepsilonRI-bearing rat basophilic leukemia (RBL) cells transfected with constructs having mutations in the PIR-B cytoplasmic region. These results define the preferential expression of the PIR-B molecules on mast cells and an inhibitory potential that can be mediated via a SHP-1-independent pathway.

To explore the phylogenetic history of the murine paired Ig-like receptors of activating (PIR-A) and inhibitory (PIR-B) types, we isolated PIR homologues from a rat splenocyte cDNA library. The rat (ra) PIR-A and raPIR-B cDNA sequences predict transmembrane proteins with six highly conserved extracellular Ig-like domains and distinctive membrane proximal, transmembrane, and cytoplasmic regions. The raPIR-B cytoplasmic region contains prototypic inhibitory motifs, whereas raPIR-A features a charged transmembrane region and a short cytoplasmic tail. Southern blot analysis predicts the presence of multiple Pira genes and a single Pirb gene in the rat genome. Although raPIR-A and raPIR-B are coordinately expressed by myeloid cells, analysis of mRNA detected unpaired expression of raPIR-A by B cells and raPIR-B by NK cells. Collectively, these findings indicate that the structural hallmarks of the Pir gene family are conserved in rats and mice, yet suggest divergence of PIR regulatory elements during rodent speciation.
