The aim of this study was to evaluate the effect of the type of preheated resin composite on the microtensile bond strength (µTBS) between dentin and feldspathic ceramic inlays. Thirty healthy human premolars received inlay preparations for feldspathic ceramic restorations and were then divided into three treatment groups according to the luting agent (n = 10): Conventional Resin Cement (RC), Preheated Nanofilled Methacrylate-Based Resin composite (MB), and Preheated Microhybrid Silorane-Based Resin composite (SB). The composite resin was preheated to 55 °C, considering the cementation group. After cementation, the sets were cut into slabs with a bonding area of 1 mm2 to be further submitted to the µTBS test (0.5 mm/min, and10 Kgf load cell). One-way ANOVA and Tukey test (p <.5) were performed to analyze the bond strength data (MPa). Failures were observed under a stereomicroscope and representative specimens were evaluated using a Scanning Electron Microscopy (SEM). ANOVA revealed that the type of luting agent utilized was significant (p =.001). Bond strength values for the RC (7.37 ± 1.27 MPa)A and MB (6.98 ± 1.96 MPa)A treatment groups were similar and greater than the SB (4.74 ± 1.65 MPa)B. Adhesive failures between the cementing agent and ceramic were prevalent for the treatment groups. The preheated nanofilled methacrylate resin-based composite technique can promote bond strength similar to resin cement, whereas preheated silorane-based resin composite as a luting agent should be seen with caution. Thus, we concluded that the type of luting agent has influenced the µTBS of feldspathic restoration to dentin. [ABSTRACT FROM AUTHOR]

Purpose: To evaluate the bond strength between composite resin and feldspathic ceramic following repair protocols with and without hydrofluoric acid and aging by thermocycling. Materials and Methods: Forty-eight glass feldspathic ceramic blocks (8 x 8 x 6 mm) were divided into three groups on the basis of their surface repair treatment: 1. 10% hydrofluoric acid + Signum Ceramic Primer I + Signum Ceramic Primer II (control group); 2. abrasive rubber tips + Signum Ceramic Primer I + Signum Ceramic Primer II (test group); 3. Signum Ceramic Primer I + Signum Ceramic Primer II (negative control group). The treated surface of each block was built up with composite and then sectioned to produce nontrimmed bars (adhesive area = 1 mm²). Half of the bars from each group were aged by 6000 cycles of 30-s immersions in water baths at 5°C and 55°C, with a transfer time of 2 s. The other bars were immediately subjected to microtensile bond strength testing. The mean bond strength for each block was then recorded and submitted to two-way ANOVA and Tukey's test (α = 0.05). Results: The aging protocol influenced the bond strength values of all groups (p = 0.000). The non-aged groups submitted to surface treatment protocols 1 (13.1 ± 2.5 MPa) and 2 (11.5 ± 5.1 MPa) presented the highest bond strength values. Conclusions: The interface bond strength of all groups was susceptible to aging. Surface treatment protocol 2, with abrasive rubber tips and no hydrofluoric acid, appeared to be the most promising option, as the resulting bond strength values were similar to those of the control group. A hipótese nula foi que a fadiga não irá gerar alterações na microestrutura deste material. Para isto, foram confeccionados, por meio de um sistema CAD/CAM, 84 discos de zircônia (VIPI block zircon) com características finais de 12 mm de diâmetro e 1,2 mm de espessura conforme a norma ISO 6872. Os espécimes foram separados em dois grupos: Grupo I – Sinterização convencional: ciclo estabelecido pelo fabricante (2 horas a 1530 °C) e Grupo II – sinterização por energia de micro-ondas (15 min a 1450 °C). Dez amostras por grupo foram submetidas ao teste monotônico (1000 KgF – 1mm/min) para a determinação da carga para fratura, utilizada como parâmetro para os testes de fadiga (método step-stress e fadiga dinâmica). O teste de fadiga step-stress foi realizado utilizando 4 perfis de carga, ultra-suave, suave, moderado e agressivo, até que ocorresse a fratura. A fadiga dinâmica foi realizada com o ensaio dos discos até fratura sob cinco taxas de tensão (1, 0,1, 0,01, 0,001 e 0,0001 MPa/s), e a partir disso foram calculados o coeficiente de crescimento de trincas e módulo de Weibull (confiabilidade) para cada grupo. Foram realizadas análises complementares de densidade, translucidez, microscopia eletrônica de varredura (MEV) e difratometria de Raios-X (DRX) de ambos os grupos para a caracterização dos materiais. Os resultados da fadiga step-stress não mostraram diferenças no comportamento dos espécimes de cada grupo. Na fadiga dinâmica, os métodos de fadiga também não mostraram diferenças no comportamento e tiveram coeficientes de crescimento de trinca similares. O modulo de Weibull mostrou maiores valores de confiabilidade para o grupo micro-ondas (m = 7,9) do que para o grupo de sinterização convencional (m = 6,7). Os valores de densidade também foram semelhantes para os grupos estudados, já a translucidez foi menor para o grupo micro-ondas. As imagens de MEV mostraram microestruturas similares entre grupos, apenas uma diferença no tamanho dos grãos foi observada, 0,53 µm para os grãos de micro-ondas e 0,89 µm para os grãos dos espécimes sinterizados convencionalmente. Conclui-se que o processo de sinterização por micro-ondas pode ser aplicado como alternativa de sinterização de zirconia ao dia-a-dia laboratorial. The objective of this work was to evaluate the microstructure and fatigue behavior of a dental ceramic (Y-TZP) sintered by conventional method or by microwave energy. The null hypothesis was that the fatigue will not effect the microstructure of this material. 84 zirconia discs (VIPI block zircon) with final characteristics of 12 mm in diameter and 1.2 mm in thickness according to ISO 6872 were made using a CAD/CAM system. The specimens were divided in two groups: Group I - Conventional sintering: cycle established by the manufacturer (2 hours at 1530 °C) and Group II - sintering by microwave energy (15 min at 1450 °C). Ten samples per group were submitted to monotonic test (1000 KgF - 1mm/min) to determine the load to fracture, used as a parameter for fatigue tests (step-stress and dynamic fatigue). The step-stress fatigue test was performed using 4 load profiles, ultramild, mild, moderate and aggressive, until the fracture. The dynamic fatigue was performed by testing the disks to fracture under five constant stress rates (1, 0.1, 0.01, 0.001 and 0.0001 MPa/s), and from this the crack growth coefficient and Weibull module (reliability) were measured for each group. Additional analyzes of density, translucency, scanning electron microscopy (SEM) and X-ray diffractometry (XRD) for both groups were carried out to characterize the materials. The results of stepstress fatigue test did not show differences in the specimens behavior of each group. Dynamic fatigue also showed no differences in behavior and had similar crack growth coefficients. The Weibull modulus showed higher reliability values for the microwave group (m = 7.9) than for the conventional sintering group (m = 6.7). The density values were also similar for the studied groups, since the translucency was lower for the microwave group. SEM images showed similar microstructures between groups, only a difference in grain size was observed, 0.53 μm for microwave grains and 0.89 μm for the conventionally-sintered grains. It is concluded that the microwave sintering process can be applied as an alternative to zirconia sintering to daily laboratory work.

We evaluate the behavior of a device designed to automatically divert and store the first flush of harvested rainwater in cisterns. The first phase (PI) was conducted with artificial precipitation in an experimental installation seeking to identify how many millimeters of rainwater should be diverted to preserve the rainwater quality. In the second phase (PII), we designed a PVC-pipe device to store the first millimeter of rainwater, and tested it in field (a rural area in Brazil) during two real rainfall events. In the third phase (PIII), the device and a hand pump were assayed for two years using eight cisterns in a rural area where people drink the rainwater. PI results indicated that the most significant pollution of the rainwater is flushed with the first millimeter of rain, and diversion promoted the removal of 98% and 100% of the total coliforms and Escherichia coli, respectively. The bacteriological behavior was maintained in the subsequent phases. The device was able to preserve the quality of the rainwater most of the time, satisfying drinking requirements for the parameters of turbidity and color. The satisfactory performance of the device was confirmed in the field, behaving as a sanitary barrier for rainwater quality protection. [ABSTRACT FROM AUTHOR]

La asociación del alelo betaS que produce hemoglobina S (HbS) y de un alelo de beta talasemia (betatal), ocurre principalmente en poblaciones que tienen simultáneamente ascendencia africana y mediterránea, y provoca un síndrome drepanocítico denominado HbS-beta talasemia. Este síndrome presenta una gran heterogeneidad clínica y genética debida en gran parte al alelo talasémico presente. Los individuos que portan un alelo talasémico beta0 presentan, en general, un curso clínico similar a los homocigotas para el alelo betaS de la HbS. En cambio, los que portan un alelo beta+ presentan un curso clínico variable, dependiendo de la reducción en la síntesis de cadena de beta globina. En este estudio se describe el caso clínico de dos pacientes con HbS-beta talasemia que consultaron en el Centro Hemato-Oncológico Pediátrico del Hospital Pereira Rossell. Se analizó el genotipo de los dos pacientes por medio de secuenciación automática del gen de la beta globina. Se demostró la presencia del alelo talasémico beta+ IVS-1-110 G->A en uno de los pacientes y del alelo beta0 codón39 C->T en el otro paciente, por lo cual ambos son heterocigotos compuestos betaS/betatal. Se discute la relación entre los datos clínicos y paraclínicos con los resultados del diagnóstico molecular. También se discute la importancia del diagnóstico molecular en relación con la composición y estructura de la población uruguaya. Summary Association between allele betaS that produces hemoglobin S and allele (betatal) is mainly seen in African and Mediterranean populations, it causes the HbS-beta-thalassemia syndrome. This syndrome shows a wide range of clinical and genetic forms due mostly to the thalassemic allele. Carriers of thalassemic allele beta0 generally follow a similar clinical course to homocygote for talasémico betaS of the HbS. On the other hand, carriers of beta+ allele follow a variable clinical course, depending on the reduction of beta-globin chain. This study describe two clinical cases of patients with HbS-beta thalassemia of the Hematologic and Oncologic Centre of the Pereira Rossell Hospital. Genotypes of patients were analysed using automatic sequences of beta globin gen. Thalassemia allele beta+ IVS-1-110 G->A was seen in one patient and allele beta0 codon39 C->T in the other, so that both are compound heterozygous betaS/betatal. Clinical and paraclinical data is compared with molecular diagnostic findings; the importance of the molecular diagnosis in Uruguayan population is also discussed. Résumé L’association de l’allèle bS qui produit hémoglobine S (HbS) et d’un allèle de bêta tallasémie (btal), arrive surtout chez des individus noirs d’Afrique et méditerranéens, et provoque un syndrome drépanocytaire appelé HbS-bêta tallasémie. Ce syndrome présente une grande hétérogé-néité clinique et génétique dûe surtout à l’allèle tallasémi-que présent. Les individus porteurs d’un allèle tallasémque b0 présentent en général,un cours clinique semblable aux homozygotes pour l’allèle bS de la HbS. En revanche, ceux qui portent un allèle b+ ont un cours clinique variable qui dépend de la réduction à la synthèse de la chaîne de beta globine. On décrit ici le cas de deux patients avec HbS-bêta tallasémie ayant consulté le Centre Hémato- Oncologique Pédiatrique de l’Hôpital Pereira Rossell. On analyse le génotype de deux patients au moyen de séquencement automatique du gène de bêta globine. On constate la présence de l’allèle tallasémique â+IVS-1-110G->A chez l’un des patients et de l’allèle b0 codon 39C->T chez l’autre. Les deux sont donc hétérozygotes composés bS/btal. On discute ici le rapport entre les données cliniques et para cliniques et les résultats du diagnostic moléculaire. Resumo A associação do alelo betaS que produz hemoglobina S (HbS) e de um alelo de beta talassemia betatal se dá principalmente em populações com ascendência africana e mediterrânea simultáneamente e provoca uma síndrome drepanocítica denominada HbS-beta talasemia. Esta síndrome apresenta uma grande heterogeneidade clínica e genética devida principalmente ao alelo para talassemia presente. Os indivíduos que são portadores de un alelo talassémico beta0 apresentam de maneira geral uma evolução clínica semelhante aos homozigotos para o alelo betaS da HbS. Por outro lado, os portadores do alelo beta+ apresentam um quadro clínico variável, dependendo da redução na síntese da cadeia beta-globina. Neste trabalho descrevemos os casos clínicos de dois pacientes com talassemia HbS.beta que foram admitidos no Centro Hemato-Oncológico Pediátrico do Hospital Pereira Rossel. O genotipo dos pacientes foi analizado pela sequenciação automática do gen da beta globina. A presença do alelo para talassemia beta+ IVS-1-110 G->A em um dos pacientes e do alelo beta0 codon39 C->T no outro foi demonstrada sendo os dois, por tanto, heterozi-gotos compostos betaS/betatal. Discute-se a relação entre os dados clínicos e paraclínicos com os resultados do diagnóstico molecular. Também se discute a importância do diagnóstico molecular em relação a composição e estrutura da população uruguaya.

In Uruguay, α-thalassemia (α-thal) mutations were introduced predominantly by Mediterranean European immigrant populations and by slave trade of African populations. A patient with anemia with hypochromia and microcytosis, refractory to iron treatment and with normal hemoglobin (Hb) electrophoresis was analyzed for α-thal mutations by multiplex gap-polymerase chain reaction (gap-PCR), automated sequencing and multiplex ligation-dependent probe amplification (MLPA) analyses. Agarose gel electrophoresis of the multiplex gap-PCR showed a band of unexpected size (approximately 700 bp) in the samples from the proband and mother. Automated sequencing of the amplified fragment showed the presence of the –(α)5.2deletion (NG_000006.1: g.32867_38062del5196) [an α-thal-1 deletion of 5196 nucleotides (nts)]. The MLPA analysis of the proband’s sample also showed the presence of the –(α)5.2deletion in heterozygous state. We report here the presence of the –(α)5.2deletion, for the first time in the Americas, in a Uruguayan family with Italian ancestry, detected with a previously described multiplex gap-PCR. Harvesting rainwater is a common practice worldwide, particularly in areas with no access to a public water supply or insufficient groundwater reserves. More than two million people living in semiarid regions of Brazil consume rainwater stored in cisterns, and little information is available regarding the water quality. Despite the initial good quality of the rainwater, its harvest and storage can introduce contaminants that must be eliminated before consumption. To evaluate the influence of handling, cistern age and precipitation on the quality of harvested rainwater, we monitored seven cisterns in the semiarid Brazilian Northeast over 4 years. Microbial and physicochemical parameters were monitored once a month, and denaturant gradient gel electrophoresis (DGGE) was performed at the end of the monitoring period. Coliform bacteria were detected in 100% of samples, while Escherichia coli were observed in 73.8%. The alkalinity and conductivity were the highest for the recently built cisterns due to the dissolution of construction materials. The DGGE of the 16S r DNA did not reveal the presence of E. coli. Instead, DGGE bands sequencing indicated that species primarily affiliated with Alphaproteobacteria were present in all cisterns, indicating the presence of microbial ecosystems capable of purifying and stabilizing the stored rainwater. The article presents a study which examines the Β (beta)-globin gene cluster of two Afro-Uruguayan populations in the South and North part of Uruguay. The study collected blood samples from 46 unrelated and healthy Afro-Uruguayans, wherein the genomic DNA was removed from blood samples by routine techniques. Conclusion shows that the variations observed between both Uruguayan regions can be explained by microevolutionary events such as genetic drift and differential mixing.

Incisivos centrais bovinos (N=62), foram fixados com resina acrílica no interior de tubos de PVC, com a face vestibular do esmalte exposta. Estas foram polidas, em máquina de polimento semiautomática com lixas de granulação 600, 800, 1200 e 2500 para obtenção de uma superfície plana. Discos de cerâmica vítrea reforçada por cristais de leucita (Empress CAD, Ivoclar Vivadent) foram confeccionados em duas dimensões, 10 mm x 1 mm destinados ao teste de fadiga (n=30), e 3 mm x 1 mm para o teste de resistência de união por cisalhamento (n=32). Os discos foram polidos em máquina de polimento semiautomática com lixas de granulação 600, 800, 1200 e 2500 e cimentados sob as superfícies de esmalte, utilizando cimento resinoso fotopolimerizável (Variolink Esthetic LC, Ivoclar Vivadent). Os conjuntos obtidos foram mantidos em estufa a 37°C por 7 dias. As amostras dos grupos Controle passaram pelos testes de cisalhamento e fadiga logo após este período. As dos grupos Er:YAG, foram submetidas a aplicação do laser (3.6W - 18 Hz x 200 mJ - 2940 nm) sobre a superfície cerâmica, a fim de promover seu descolamento. Em seguida, as superfícies foram submetidas ao processo de limpeza adequado, e recimentadas sob o mesmo protocolo anterior. Para o teste de cisalhamento foi utilizado um dispositivo em forma de faca, com célula de carga de 1000 kgf (1 mm/min) e a análise estatística obtida através dos testes ANOVA e Tukey (95%). Para a classificação dos modos de falha, as amostras foram observadas em estereomicroscópio óptico. A análise do comportamento em fadiga se deu através do método Stepwise stress, com carga inicial de 300 N (5.000 ciclos), steps incrementais de 100 a 700 N (10.000 ciclos/step), seguido de steps de 50 N (10.000 ciclos/step). Para a análise estatística, os testes Kaplan-Meier e Mantel-Cox, seguidos da comparação múltipla aos pares (5%), foram empregados. Os dados obtidos no teste de cisalhamento demonstraram não haver significância estatística para o fator laser (p=0.444), porém, no fator envelhecimento o valor foi significativamente menor (p=0.031). As amostras do grupo laser apresentaram valores significativamente menores em relação ao teste de resistência a fadiga quando comparadas as do grupo controle (p=0,007), que apresentou maiores valores de falha em carga (FFL) e número de ciclos (CFF). Entretanto, o módulo de Weibull para FFL e CCL não foi estatisticamente significante entre os grupos. Com base nos resultados obtidos pode-se concluir que, a resistência de união não foi afetada enquanto sobreviência em fadiga foi significativamente reduzida em laminados cerâmicos recimentados após exposição ao laser Er:YAG. This work aimed to evaluate the adhesive bond strength and fatigue behavior of cemented ceramic laminates after their removal using Er:YAG laser. 62 samples were divided into two groups: control (C), and Er:YAG (L). Bovine central incisors (N=62) were fixed with acrylic resin inside of PVC tubes, with the buccal surface of the enamel exposed. These were polished in a semi-automatic polishing machine with #600, 800, 1200 and 2500 grit sandpaper to obtain a flat and polished surface. Discs of a leucite reinforced glass ceramic (Empress CAD, Ivoclar Vivadent) were made in two dimensions, 10 mm x 1 mm for the fatigue test (n=30), and 3 mm x 1 mm for the shear bond strength test (n=32). The discs were polished in a semiautomatic polishing machine with #600, 800, 1200 and 2500 grit sandpaper and cemented on the enamel surfaces using a light-curing resin cement (Variolink Esthetic LC, Ivoclar Vivadent). The sets obtained were stored at 37°C for 7 days. The samples from the Control groups underwent the shear and fatigue tests shortly after this period. The Er:YAG groups were submitted to laser application (3.6W - 18 Hz x 200 mJ - 2940 nm) on the ceramic surface, in order to promote its debonding. Then, the surfaces were subjected to the appropriate cleaning process, and re-cemented following the same previous protocol. For the shear bond strength test, a knife-shaped device was used, with a load cell of 1000 kgf (1 mm/min) and the statistical analysis was obtained through the ANOVA and Tukey tests (95%). For the classification of failure modes, the samples were observed in an optical stereomicroscope. The analysis of fatigue behavior was performed using the Stepwise stress method, with an initial load of 300 N (5,000 cycles), incremental steps from 100 to 700 N (10,000 cycles/step), followed by 50 N steps (10,000 cycles/step). For statistical analysis, the Kaplan-Meier and Mantel-Cox tests, followed by multiple pairwise comparisons (5%), were used. The data obtained in the shear bond strength test showed no statistical significance for the laser factor (p=0.444), however, the aging factor the value was significantly lower (p=0.031). In the fatigue test, laser group showed significantly lower values in relation to the control group (p=0.007), which showed higher values of failure under load (FFL) and number of cycles (CFF). However, the Weibull modulus for FFL and CCL was not statistically significant between groups. Based on the results obtained, it can be concluded that the bond strength was not affected while fatigue survival was significantly reduced in ceramic laminates re-cemented after exposure to Er:YAG laser.

- Abstract
Purpose: To evaluate the bond strength between composite resin and feldspathic ceramic following repair protocols with and without hydrofluoric acid and aging by thermocycling., Materials and Methods: Forty-eight glass feldspathic ceramic blocks (8 x 8 x 6 mm) were divided into three groups on the basis of their surface repair treatment: 1. 10% hydrofluoric acid + Signum Ceramic Primer I + Signum Ceramic Primer II (control group); 2. abrasive rubber tips + Signum Ceramic Primer I + Signum Ceramic Primer II (test group); 3. Signum Ceramic Primer I + Signum Ceramic Primer II (negative control group). The treated surface of each block was built up with composite and then sectioned to produce nontrimmed bars (adhesive area = 1 mm²). Half of the bars from each group were aged by 6000 cycles of 30-s immersions in water baths at 5°C and 55°C, with a transfer time of 2 s. The other bars were immediately subjected to microtensile bond strength testing. The mean bond strength for each block was then recorded and submitted to two-way ANOVA and Tukey's test (α = 0.05)., Results: The aging protocol influenced the bond strength values of all groups (p = 0.000). The non-aged groups submitted to surface treatment protocols 1 (13.1 ± 2.5 MPa) and 2 (11.5 ± 5.1 MPa) presented the highest bond strength values., Conclusions: The interface bond strength of all groups was susceptible to aging. Surface treatment protocol 2, with abrasive rubber tips and no hydrofluoric acid, appeared to be the most promising option, as the resulting bond strength values were similar to those of the control group.
