Abstract

Oxygen-, nitrogen-, and sulfur-containing heterocycles exhibit remarkable biological and pharmaceutical activities that are often found in naturally occurring products, pharmaceutical molecules, and synthetic compounds. Therefore, the synthesis of heterocycle building blocks has gained huge attention in the past decade. Compared with the modification of heterocyclic compounds, the construction of heterocyclic compounds from scratch is more attractive. In this Perspective, we describe our recent advances in the rational design and strategic application of the de novosynthesis of various synthetically and biologically important heterocycles. The organization of the Perspective is categorized in the following order: (a) type of heterocycle, (b) ring size of heterocycle, (c) number of heteroatoms, and (d) fused heterocycles. Moreover, some significant and representative synthetic methodologies and their synthetic applications and brief reaction mechanisms have also been described. We hope that this Perspective will help to provide a guideline for researchers who are interested in this fertile area.

Abstract

A palladium‐catalyzed cascade cyclization/alkenylation for the assembly of synthetically valuable isoxazolyl vinylsilane derivative has been accomplished. Easily accessible ynone oximes, and available vinylsilane agents were used as the reaction starting materials This protocol features broad substrate scope, good functional group tolerance, and good step‐ and atom‐economy. Remarkably, this approach provides a new approach for the construction of structurally diverse isoxazolyl‐containing vinylsilanes with high molecular complexity, showing a promising application in synthetic and pharmaceutical chemistry. 23. Palladium-catalyzed vinylation of indoles with vinyl ethers viadeoxygenative coupling

Abstract

Indole is a promising heteroarene in many natural products and pharmaceuticals; therefore, various synthetic methods for indole functionalizations have arisen in recent years. Herein, we report a Pd-catalyzed deoxygenative coupling for the N-vinylation of indoles by employing vinyl ethers. The vinylated indoles could be readily prepared in good to excellent yields with N1-regioselectivity regardless of the electronic characteristics and substitution patterns of indole substrates. Some indole-based pharmaceutical molecules, such as gramine, evodiamine, rutaecarpine, and melatonine, are also successfully vinylated. Moreover, carbazoles and indazoles are shown to participate. Additionally, vinylated indole can readily transform into structurally interesting indole derivatives.

Abstract

A novel and reliable palladium‐catalyzed oxylallylation of alkynone oxime ethers with fluorine‐containing alkenes was accomplished. Using the bulk industrial chemical 3‐bromo‐3,3‐difluoroprop‐1‐ene as the coupling partner, this synthetic methodology offers the first example for the assembly of structurally diverse gem‐difluorinated isoxazole derivatives in moderate to good yields with high atom‐ and step‐economy and excellent functional group compatibility. More importantly, this strategy allows for the direct combination of the isoxazole motifs and gem‐difluoroalkene unit, which is not easy to obtain through a general synthetic strategy. Abstract

A reliable and efficient palladium‐catalyzed cascade cyclization/alkylation of oxime ethers with unactivated alkenes is described, affording a whole variety of structurally diverse isoxazole derivatives in moderate to good yields with excellent functional group compatibility. Ionic liquid [Aeim]Br not only acts as an environmentally friendly solvent but also acts as an accelerating agent to provide excess bromine source to eliminate bromomethane from oxime ethers. More importantly, the use of "chain‐walking" strategy provides a novel methodology in organic synthesis to rapid generation of molecular complexity from readily available starting materials.
