Abstract

Abstract On June 21st, a Mw6.2 earthquake struck the Afghan‐Pakistan‐border‐region, situated within the India‐Asia collision. Thousand thirty‐nine deaths were reported, making the earthquake the deadliest of 2022. We investigate the event's rupture processes by combining seismological and geodetic observations, aiming to understand what made it that fatal. Our Interferometric Synthetic Aperture Radar‐constrained slip‐model and regional moment‐tensor inversion, confirmed through field observations, reveal a sinistral rupture with maximum slip of 1.8 m at 5 km depth on a N20°E striking, sub‐vertical fault. We suggest that not only external factors (event‐time, building stock) but fault‐specific factors made the event excessively destructive. Surface rupture was favored by the rock foliation, coinciding with the fault strike. The distribution of Peak‐Ground‐Velocity was governed by the sub‐vertical fault. Maximum slip was large compared to other events globally and might have resulted in peak‐frequencies coinciding with resonance‐frequencies of the local buildings and demonstrates the devastating impact of moderate‐size earthquakes.

Abstract

Abstract We present the result of a search for the charged-lepton-flavor violating decay τ − → μ − μ + μ − using a 424 fb −1 sample of data recorded by the Belle II experiment at the SuperKEKB e + e − collider. The selection of e + e − → τ + τ − events is based on an inclusive reconstruction of the non-signal tau decay, and on a boosted decision tree to suppress background. We observe one signal candidate, which is compatible with the expectation from background processes. We set a 90% confidence level upper limit of 1.9 × 10 −8 on the branching fraction of the τ − → μ − μ + μ − decay, which is the most stringent bound to date.

Abstract

Abstract We present a measurement of the ratio R μ = B τ − → μ − ν ¯ μ ν τ / B τ − → e − ν ¯ e ν τ $$ {R}_{\mu }=\mathcal{B}\left({\tau}^{-}\to {\mu}^{-}{\overline{\nu}}_{\mu }{\nu}_{\tau}\right)/\mathcal{B}\left({\tau}^{-}\to {e}^{-}{\overline{\nu}}_e{\nu}_{\tau}\right) $$ of branching fractions B $$ \mathcal{B} $$ of the τ lepton decaying to muons or electrons using data collected with the Belle II detector at the SuperKEKB e + e − collider. The sample has an integrated luminosity of 362 ± 2 fb −1 at a centre-of-mass energy of 10.58 GeV. Using an optimised event selection, a binned maximum likelihood fit is performed using the momentum spectra of the electron and muon candidates. The result, R μ = 0.9675 ± 0.0007 ± 0.0036, where the first uncertainty is statistical and the second is systematic, is the most precise to date. It provides a stringent test of the light-lepton universality, translating to a ratio of the couplings of the muon and electron to the W boson in τ decays of 0.9974 ± 0.0019, in agreement with the standard model expectation of unity.

Abstract

Abstract We present measurements of the branching fractions of eight B ¯ 0 $$ {\overline{B}}^0 $$ → D (*)+ K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ , B − → D (*)0 K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ decay channels. The results are based on data from SuperKEKB electron-positron collisions at the Υ(4S) resonance collected with the Belle II detector, corresponding to an integrated luminosity of 362 fb −1. The event yields are extracted from fits to the distributions of the difference between expected and observed B meson energy, and are efficiency-corrected as a function of m(K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ ) and m(D (*) K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ ) in order to avoid dependence on the decay model. These results include the first observation of B ¯ 0 $$ {\overline{B}}^0 $$ → D + K − K S 0 $$ {K}_S^0 $$ , B − → D* 0 K − K S 0 $$ {K}_S^0 $$ , and B ¯ 0 $$ {\overline{B}}^0 $$ → D* + K − K S 0 $$ {K}_S^0 $$ decays and a significant improvement in the precision of the other channels compared to previous measurements. The helicity-angle distributions and the invariant mass distributions of the K − K S ∗ 0 $$ {K}_{(S)}^{\left(\ast \right)0} $$ systems are compatible with quasi-two-body decays via a resonant transition with spin-parity J P = 1− for the K − K S 0 $$ {K}_S^0 $$ systems and J P = 1+ for the K − K* 0 systems. We also present measurements of the branching fractions of four B ¯ 0 $$ {\overline{B}}^0 $$ → D (*)+ D s − $$ {D}_s^{-} $$ , B − → D (*)0 D s − $$ {D}_s^{-} $$ decay channels with a precision compatible to the current world averages.

Abstract

Abstract Etruria contained one of the great early urban civilisations in the Italian peninsula during the first millennium BC, much studied from a cultural, humanities-based, perspective, but relatively little with scientific data, and rarely in combination. We have addressed the unusual location of twenty inhumations found in the sacred heart of the Etruscan city of Tarquinia, focusing on six of these as illustrative, contrasting with the typical contemporary cremations found in cemeteries on the edge of the city. The cultural evidence suggests that the six skeletons were also distinctive in their ritualization and memorialisation. Focusing on the six, as a representative sample, the scientific evidence of osteoarchaeology, isotopic compositions, and ancient DNA has established that these appear to show mobility, diversity and violence through an integrated bioarchaeological approach. The combination of multiple lines of evidence makes major strides towards a deeper understanding of the role of these extraordinary individuals in the life of the early city of Etruria.

Abstract

Wild watermelon is ideal rootstock for cultivated watermelon grafting because of its graft compatibility, tolerance to environmental stresses, and less effect on the fruit quality of the scion. However, it is hard to acquire the excellent wild watermelon genotype with multiple resistances through traditional breeding programs. Genetic transformation is an efficient approach for genetic improvement. In the study, we analyzed the effect of plant growth regulators on the cotyledon regeneration efficiency with the aim of establishing genetic transformation system for wild watermelon. Wild watermelon (Citrullus lanatus var. citroides) seeds were germinated under aseptic conditions. Cotyledons were cut from five day seedlings and cultured on MS medium supplemented with different plant growth regulators. The results showed that explants cultured in MS + BA (1.0 mg/L) achieved the highest regeneration rate, and 6.3 shoots were regenerated from each explant on an average. Adventitious shoots were able to elongate on MS medium with KT concentrations ranging from 0.1 to 0.3 mg/L. On an average 62% adventitious shoots were successfully rooted on MS medium supplemented with 0.05 mg/L NAA. Rooted seedlings were successfully acclimatized and gradually hardened-off to green-house conditions and subsequently cultivated in soil with a mean survival rate of 80%. The cotyledon regeneration system for wild watermelon was successfully established in the study, which is needed for genetic transformation of wild watermelon in the future.

Abstract

The present study was conducted to investigate the effects of iso-osmotic Na+, Cl- and NaCl stress on the plant growth and physiological parameters of self grafted and Figleaf gourd grafted cucumber. The results showed that, compared to non salt stress cucumber, Na+, Cl- and NaCl stress significantly decreased the plant growth. By comparison, less effect was induced by Cl-, and more effect was induced by Na+ and NaCl. Compared to self-grafted cucumber, Figleaf gourd grafting significantly increased the tolerance of cucumber to Na+ and NaCl stress. The shoot water content, Na+ content of root, K+, Ca2+ contents of stem and leaf, as well as leaf SOD activities were much higher in the Figleaf gourd grafted plants, while Na+ contents of stem and leaf were much lower than self-grafted cucumber under iso-osmotic Na+ and NaCl stress. The above results suggested that the sensitivity of cucumber to Na+ and Cl- is different. The toxicity induced by Na+ is much higher than Cl-. The improved salt tolerance of cucumber by grafting onto Figleaf gourd was achieved mainly by higher accumulation of Na+ in the root, and limited Na+ transport to the shoot. Thus, Na+ exclusion in the shoot plays a pivotal role in the salt tolerance of grafted cucumber.

Abstract

Intro: Published data is limited in describing clinical features and disease burden of invasive Escherichia coli disease (IED). This study characterized bacteremic and non-bacteremic IED in relation to clinical features and multidrug resistance (MDR) of causative E. coli isolates. Methods: EXPECT-2 (NCT04117113) was a multinational, hospital-based, observational study that prospectively enrolled IED patients aged ≥60 years between October 2019 and January 2021. IED was defined by a positive E. coli culture from blood (bacteremic IED), a sterile body site or urine (non-bacteremic IED) in the presence of requisite criteria of Systemic Inflammatory Response Syndrome (SIRS), Sequential Organ Failure Assessment (SOFA), or quick SOFA (qSOFA). In-hospital mortality and IED-related medical encounters were assessed through 28 days following IED diagnosis. MDR was defined as acquired non-susceptibility to ≥1 antibiotic in ≥3 antimicrobial classes. Findings: The full analysis set included 193 bacteremic (80.4%) and 47 non- bacteremic (19.6%) patients. Patient demographics were balanced between bacteremic and non-bacteremic IED. The rates of patients with ≥2 SIRS criteria were comparable between bacteremic (64.8%) and non-bacteremic IED (70.2%). The rates of patients with the ≥2-point change in total SOFA score were comparable between bacteremic (62.2%) and non-bacteremic IED (53.2%). The rates of sepsis and septic shock were 75.0% and 11.4%, respectively, for bacteremic, and 83.3% and 7.1%, respectively, for non-bacteremic IED. The most common IED-related medical encounter was the emergency room both in patients with bacteremic (31.5%, [41/130]) and non-bacteremic IED (35.9%, [14/39]). Of 11 patients who died, 6 (54.5%) had bacteremic IED. Of 294 E. coli isolates with antimicrobial susceptibility testing collected from 238 patients, MDR rates were 34.7% (87/251) for bacteremic and 37.2% (16/43) for non-bacteremic isolates. Conclusion: IED manifested as severe infection, with approximately one-third of E. coli isolates displaying MDR. Abstract

Ginsenoside Rg3 is a natural active ingredient that is extracted from Korean red ginseng root. It elevates the therapeutic effect of radiotherapy and chemotherapy, but previous studies found that the application of Rg3 is heavily limited by its low bioavailability and poor absorption via oral administration. To overcome these problems, Rg3-loaded PEG-PLGA-NPs (Rg3-NPs) were prepared by the modified spontaneous emulsification solvent diffusion (SESD) method, and the physicochemical characteristics of Rg3-NPs were investigated. We treated primary glioblastoma with 50 mM Rg3-NPs for 48h. We then used gene expression arrays (Illumina) for genome-wide expression analysis and validated the results for genes of interest by means of real-time PCR. Functional annotations were then performed using the DAVID and KEGG online tools. The results showed that the Rg3-NPs are slick and uniform, the average diameter of the nanoparticles is 75-90 nm, and their entrapment efficiency is 89.7+1.7%. MTT showed that the growth of cells can be significantly inhibited by Rg3-NPs in a dose-dependent manner. FCM testing showed Rg3-NPs can be released from the conjugate nanoparticle and react with the genes in the cell nuclei, causing changes in the gene molecules. We also found that cancer cells treated with Rg3-NPs undergo cell-cycle arrest at different checkpoints. This arrest was associated with a decrease in the mRNA levels of core regulatory genes BUB1, CDC20, TTK, and CENPE, as determined by microarray analysis and verified by real-time PCR. Furthermore, Rg3-NPs induced the expression of the apoptotic and antimigratory protein p53 in cell lines. The results of the present study, together with the results of earlier studies, show that Rg3-NPs target genes involved in theprogressionoftheM-phaseofthecellcycle.Itisassociatedwithseveralimportant pathways, which include apoptosis (p53). Rg3-NPs may be a potent cell-cycle regulation drug targeting the M-phase in glioblastoma cell lines.

Abstract

In Germany, accidents are collected nationwide, including also bicycle accidents. To determine accident-prone locations, it is necessary to not only look at the number of accidents but also in relation to the absolute number of cyclists traversing that location. Thus, this study exploits a collection of bicycle accidents in combination with estimated cyclist volumes on street level in Hanover (Germany). The basis for the generated bicycle volumes is the resulting origin-destination demand for bicycle mode from an agent-based traffic simulation model. A normalization of the accidents by an absolute bicycle volume allows to estimate a risk score and to compare high frequented ways with less popular minor paths in an objective manner. This method is used to show locations with comparatively high risk for cyclists. Besides highlighting these spots on a map, e.g. for city planners, the resulting risk scores can be integrated into bicycle routing to avoid those areas for future trips.

Abstract

Abstract We present the design of a next-generation experiment, n2EDM, currently under construction at the ultracold neutron source at the Paul Scherrer Institute (PSI) with the aim of carrying out a high-precision search for an electric dipole moment of the neutron. The project builds on experience gained with the previous apparatus operated at PSI until 2017, and is expected to deliver an order of magnitude better sensitivity with provision for further substantial improvements. An overview is of the experimental method and setup is given, the sensitivity requirements for the apparatus are derived, and its technical design is described.

Introduction: Inflammatory Breast Cancer (IBC) is the most aggressive form of breast carcinoma characterized by rapid onset of inflammatory signs and its molecular fingerprint has not yet been elucidated. Objectives: The objective of this study was to detect both gene expression levels and alternate RNA splice variants specific for IBC. Abstract

VE161 wheat which has the function of promotting homoeologous pairing is a male sterile substitution line or a fertile addition line with one pair of alien chromosomes from Agropyron elongatum. The alien chromosome in VE161 wheat was detected by means of techniques of total genomic DNA in situ hybridization, RFLP and double-ditelo chromosomal analysis. The results from in situ hybridization confirmed that the alien chromosome was derived from Agropyron elongatum. The RFLP analysis showed that it belongs to fourth homoeologous group, and the results from double-ditelo chromosomal analysis gave evidence that 4B chromosome was replaced in the substitution line, so it can be concluded that, in VE161 wheat, the alien chromosome would be 4E and the replaced wheat chromosome would be 4B in the substitution line. The reason for the replaced chromosome identified as 7B in a earlier study might be due to the rearrangement occurred in the genome of wheat.

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Abstract

Proteomics has been used to investigate cross-talk between the intestinal microbiome and host biological processes. In this study, an in ovo technique and a proteomics approach was used to address how early bacterial colonization in the gastrointestinal tract (GIT) could modulate inflammatory and immune responses in young broilers. Embryos at 18 embryogenic days were inoculated with saline (S), 102 CFU of Citrobacter freundii (CF), Citrobacter species (C2), or lactic acid bacteria mixture (L) into the amnion. At 10 days posthatch, ileum samples from 12 birds per treatment were selected for tandem mass spectrometry analysis. Our further findings indicated that treatment-specific influences on early GIT microbiota resulted in different immune responses in mature broilers. Predicted functional analyses revealed activation of inflammation pathways in broilers treated in ovo with L and CF. Exposure to L enhanced functional annotation related to activation, trafficking of immune cells, and skeletal growth based-network, while CF inhibited biological functions associated with immune cell migration and inflammatory response. These results highlighted that proper immune function was dependent on specific GIT microbiota profiles, in which early-life exposure to L-based probiotic may have modulated the immune functions, whereas neonatal colonization of Enterobacteriaceae strains may have led to immune dysregulation associated with chronic inflammation.

Abstract

Background and Study Aim: Cardiovascular diseases are currently classified as civilization diseases. The number of cases since the 20th century has dramatically increased. Currently, as many as 46% of all deaths in Poland are caused by diseases of the circulatory system. The aim of the study is to assess the impact of cardiac rehabilitation according to the C model on exercise tolerance and hemodynamic indices in patients after a cardiac incident. Material and Methods: The research was carried out in the Department of Cardiac Rehabilitation in Szymbark. The study group consisted of 32 patients who, after physical examination at admission, were qualified for the preliminary assessment of physical fitness on the basis of 6MWT. The test was carried out the day after the admission, according to current standards. Subsequently, the examined persons underwent a 28-day cycle of early cardiac rehabilitation according to the C model. Results: As a result of a 4-week cardiac rehabilitation, a statistically significant improvement in physical performance was obtained, with a distance increase of 68.34 m on average. A statistically significant change in the level of fatigue according to Skala Borga after the completion of the rehabilitation cycle was demonstrated. Patients with normal body weight, overweight and obesity received a similar increase in 6MWT results due to the rehabilitation cycle. Conclusions: The results of this study indicate the urgent need to educate patients in the prevention and prevention of cardiovascular disease and to increase the availability of rehabilitation for all people after a cardiological incident.

Abstract

Abstract The synthesis of zeolite A employing kaolin waste from paper coating was studied. The kaolin waste was pre-treated at 550 to 800 ºC. For comparison purposes, a sample of zeolite A was also prepared using the IZA procedure. The materials were characterized by 27Al MAS NMR, X-ray diffraction and scanning electron microscopy using a microprobe. Pretreatment was necessary, and the best temperatures were between 600 and 700 ºC. The zeolite A formation was observed in all the prepared materials, reaching 52% crystallinity. On the other hand, the sodalite phase and amorphous materials were also formed.

Abstract

Under high light intensity, phytoplankton protect their photosystems from bleaching through non-photochemical quenching processes. The consequence of this is suppression of fluorescence emission, which must be corrected when measuring in situ yield with fluorometers. We present data from the Southern Ocean, collected over five austral summers by 19 southern elephant seals tagged with fluorometers. Conventionally, fluorescence data collected during the day (quenched) were corrected using the limit of the mixed layer, assuming that phytoplankton are uniformly mixed from the surface to this depth. However, distinct deep fluorescence maxima were measured in approximately 30% of the night (unquenched) data. To account for the evidence that chlorophyll is not uniformly mixed in the upper layer, we propose correcting from the limit of the euphotic zone, defined as the depth at which photosynthetically available radiation is ~ 1% of the surface value. Mixed layer depth exceeded euphotic depth over 80% of the time. Under these conditions, quenching was corrected from the depth of the remotely derived euphotic zone Zeu, and compared with fluorescence corrected from the depth of the density-derived mixed layer. Deep fluorescence maxima were evident in only 10% of the day data when correcting from mixed layer depth. This was doubled to 21% when correcting from Zeu, more closely matching the unquenched (night) data. Furthermore, correcting from Zeu served to conserve non-uniform chlorophyll features found between the 1% light level and mixed layer depth.

A zeólita A é uma típica zeólita sintética muito utilizada nas indústrias como trocador de íons em formulação de detergentes. Por outro lado, a diatomita, material rico em sílica (cerca de 80%), é utilizada em diversos setores industriais como agente filtrante. Neste trabalho é apresentada a síntese da zeólita A utilizando a diatomita como fonte de SiO2 e Al2O3. As relações SiO2/ Al2O3 utilizadas foram 2 e 1,5. Todas as amostras obtidas foram caracterizadas por análise de difração de raios X, adsorção de N2 e microscopia eletrônica de varredura. Foi possível sintetizar a zeólita A pura com 51% de cristalinidade com 2 h de cristalização. Maiores tempos de cristalização, independentemente da relação Si e Al levaram à formação da fase sodalita. Na síntese 2 com relação SiO2/ Al2O3 = 1,5, através do processo de separação observou-se que a formação da zeólita A ocorre a partir de aglomerados de partículas de diatomita, e tempos maiores, ao contrário de favorecer a dissolução da diatomita, favorecem a formação da fase sodalita a partir dos cristais de zeólita A.

Um estudo do processo de extração de lítio de β-espodumênio foi realizado visando a obtenção do lítio e geração de um subproduto zeolítico. A retirada de lítio foi avaliada por meio de medidas de absorção atômica do sobrenadante. As análises de difração de raios X do resíduo gerado e de precipitados indicaram o principio de formação da zeólita A, quando NaOH é utilizado no procedimento de síntese. A adição de aluminato de sódio não é favorável ao processo de extração, pois proporciona a formação compostos mais estáveis com o lítio.

Utilizou-se um resíduo sílico-aluminoso como fonte de SiO2 e Al2O3 para síntese de zeólitas. O resíduo é um subproduto do processo de extração de lítio do β-espodumênio. Os produtos sintetizados caracterizaram-se por uma mistura de fases zeolíticas e compostos presentes no resíduo, que não foram solubilizados. Quando a relação Si/Al foi ajustada para 1, foi formada uma mistura de fases zeólita A + sodalita e zeólita NaP + sodalita. A mistura de fases zeólita A + sodalita foi obtida deixando o resíduo sílico-aluminoso em agitação por 2 h a 80 ºC, antes da adição de aluminato de sódio e posterior cristalização por 4 h. Quando a mistura resíduo e NaOH ficou em estático a 100 ºC por 4 h, nesta mesma etapa, o produto formado foi uma mistura de sodalita + zeólita NaP. O aumento da razão Si/Al para 3,2, uma etapa de envelhecimento e maiores tempos de cristalização, levou a formação de uma mistura de fases NaP1 e quartzo pertencente ao resíduo.

30. [Polarographic determination of titanium in natural water]

Abstract

Titanium has a very sensitive catalytic polarographic wave in the system of cupferon-hexamine-sodium sulfate at pH 6.0-6.4. The catalytic current was proportional to the content of titanium. High blank of the reagents could be improved by extraction with the system of cupferron-chloroform. For determination, samples were filtered with quantitative filter paper, but it was not necessary for drinking water samples. The filtration was acidified with HCl to 0.5% (V/V) and heated in a boiling water bath for 30 min, 5 ml sample was sufficed to most analyses. The detection limit was 0.12 microgram/L. The coefficient of variation was 8% for 6 determinations at 0.2 microgram/L. Three levels of titanium were spiked to one sample, and it was found that the recoveries were 96-101%. The proposed method was applied to rain water, well-head water, well water, tap water and river water. The content of titanium of most samples was less than 2 micrograms/L.

Ammonia is a valuable chemical produced in the industry and is wide used in the manufacture of fertilizers, plastics and pharmaceuticals. On the other hand, some important tools in chemical processes are modelling, simulation and optimization, and these types of studies have attracted considerable attention of many researchers. Several models of ammonia synthesis have been developed with the purpose of design and optimization. The typical design problem in this process requires obtained an optimal reactor length with maximum economic return and many works in literature have focused on the optimization of this reactor using different optimization techniques. In this work, an alternative approach to those presented in literature for solving this boundary value problem, and determine the optimal solution, is presented. The model of the reactor along with the kinetic form a non-linear differential-algebraic system. These differential-algebraic equations are discretized using orthogonal collocation on finite elements with continuous profiles approximated by Lagrange polynomials. In this manner, the resulting algebraic collocation equations are written as equality constraints in the optimization problem. Thus, the optimization problem is solved by implementing the IPOPT solver within the GAMS optimization-modeling platform. Although IPOPT solver here implemented is a local solver that does not guarantee the global optimum, multiple initial guesses were used to solve the problem and obtain an optimal solution of the problem. The obtained results are compared with other results available in literature and indicate that the used approach is a good alternative for dealing with this type of problems.

Abstract

A population dynamics model for copepods is presented, describing the seasonal dynamics of Pseudocalanus minutus elongatus and Acartia spp. in the southern Baltic Sea (Gdańsk Deep). The copepod model was coupled with a one-dimensional physical and biological upper layer model for nutrients (total inorganic nitrogen, phosphate), phytoplankton, microzooplankton, and an early juvenile of herring as a predator. In this model, mesozooplankton (herbivorous copepods) has been introduced as an animal having definite patterns of growth in successive stages, reproduction and mortality. The populations are represented by 6 cohorts in different developmental stages, thus assuming that recruitment of the next generation occurs after a fixed period of adult life. The copepod model links trophic processes and population dynamics, and simulates individual growth within cohorts and the changes in biomass between cohorts. The simulations of annual cycles of copepods contain one complete generation of Pseudocalanus and two generations of Acartia in the whole column water, and indicate the importance of growth in the older stages of 6 cohorts of each species, to arrive at a total population biomass. The peaks of copepods' biomass are larger at the turn of June and July for Pseudocalanus and smaller in July for Acartia, lagging that of phytoplankton by ca. two mouths, due to the growth of cohorts in successive stages and egg production by females. The numerical results show that the investigated species could not be the main factor limiting the spring phytoplankton bloom in the Gdańsk Deep, because the initial development was slow for Acartia and faster for Pseudocalanus, but the main development formed after the bloom, in both cases. The phytoplankton bloom is very important in the diet of the adults of the copepods, but it is not particularly important for the youngest part of new generation (early nauplii). However, the simulated microzooplankton biomass was enough high to conclude, in our opinion, that, in this case, it was a major cause in limiting phytoplankton bloom. The model presented here is a next step in understanding how the population dynamics of a dominant species in the southern Baltic Sea interact with the environment.

Abstract

Macroinvertebrate communities are one of the most used groups in assessments of water quality, since they respond directly to the level of contamination of aquatic ecosystems. The main objective of this study was the assessment of the water quality of the Sinos River basin (Rio Grande do Sul state, Brazil) through biotic indices based on the macroinvertebrate community ("Family Biotic Index - FBI", and "Biological Monitoring Working Party Score System - BMWP"). Three lower order streams (2nd order) were selected in each one of three main regions of the basin. In each stream, the samplings were performed in three reaches (upper, middle, and lower), totalling 27 reaches. Two samplings were carried in each reach over one year (winter and summer). A total of 6,847 macroinvertebrates distributed among 54 families were sampled. The streams from the upper region were of better water quality than the lower region. The water quality did not change between the upper, middle and lower reaches of the streams. However, the upper reaches of the streams were of better water quality in all the regions of the basin. The water quality of the streams did not vary between the summer and the winter. This result demonstrated that water quality may be analysed in both studied seasons (summer and winter) using biotic indices. The analysis of the results allows us to conclude that the biotic indices used reflected the changes related to the water quality along the longitudinal gradient of the basin. Thus, aquatic macroinvertebrates were important bioindicators of the water and environmental quality of the streams of the Sinos River basin.

Abstract

Self-grafted and pumpkin rootstock-grafted cucumber plants were subjected to the following four treatments: 1) aerated nutrient solution alone (control), 2) nutrient solution with 10 mM Ca(NO3)2 (Ca), 3) nutrient solution with 90 mM NaCl (NaCl), and 4) nutrient solution with 90 mM NaCl + 10 mM Ca(NO3)2 (NaCl+Ca). The NaCl treatment decreased the plant dry mass and content of Ca2+ and K+ but increased the Na+ content in roots and shoots. Smaller changes were observed in pumpkin rootstock-grafted plants. Supplementary Ca(NO3)2 ameliorated the negative effects of NaCl on plant dry mass, relative growth rate (RGR), as well as Ca2+, K+, and Na+ content especially for pumpkin rootstock-grafted plants. Supplementary Ca(NO3)2 distinctly stimulated the plasma membrane (PM) H+-ATPase activity which supplies the energy to remove excess Na+ from the cells. The expressions of gene encoding PM H+-ATPases (PMA) and gene encoding a PM Na+/H+ antiporter (SOS1) were up-regulated when Ca(NO3)2 was applied. The pumpkin rootstock-grafted plants had higher PM H+-ATPase activity as well as higher PMA and SOS1 expressions than the self-grafted plants under NaCl + Ca treatment. Therefore, the addition of Ca2+ in combination with pumpkin rootstock grafting is a powerful way to increase cucumber salt tolerance.

Abstract

Boundary effect analysis is related to border policy making in the cross-border ethnic area. The border effect literatures show that geographic boundaries have obvious impacts on economic, social and cultural relations in both sides of a nation border. Particularly in cross-border ethnic areas, each ethnic group has strong internal spatial structure relevance, and the boundary effect is more obvious. However, most of China's border areas are cross-border ethnic areas, each of border issues is unique. Under this perspective, we analyze the border effects of various boundaries can provide basis for formulating border management policies. For small scale of cross-border ethnic minority areas, how to formulate the boundary management policy is a good question to explore. This paper is demonstrated by a study of the impact of border management policies in Dehong Dai and Jingpo Autonomous Prefecture in Yunnan Province at the border area to Burma. The comparative method is used to analysis the border management policies in past 50 decades for the border area of Yunnan Province .This research aims to define trends within border policy and its influences to national security. This paper also examines Wendy Brown's liberal theory of border management policy. We found that it is not suitable for Sino-Burma border area. The conclusion is that the changes or instability of international economic and political situation has more influence to this cross-border ethnic area, and only innovative policy will be effective in cross-border ethnic area. So the border management policies should reflect the change of international context.

Abstract

The potential of 5-aminolevulenic acid (ALA) to enhance the salt tolerance of cucumber (Cucumis sativus L.) seedlings was investigated. ALA was applied at various concentrations (0, 1, 10, 25, 50, and 100 mg dm-3) as foliar spray or root watering. Then the seedlings were exposed to 0 or 75 mM NaCl for 5 d. NaCl stress reduced the root and leaf dry masses, leaf area, and the leaf net CO2 assimilation rate. These reductions were counteracted by exogenous ALA, and the most efficient was 50 mg dm-3 concentration via foliar spray. ALA decreased the H2O2 contents and increased the activities of ascorbate peroxidase (APX) and glutathione reductase (GR) in NaCl-treated cucumber roots and leaves and the activity of catalase (CAT) in leaves. The ALA application also up-regulated the expressions of CAT and cAPX genes in roots and leaves and the expression of GR gene in roots of the NaCl treated cucumber plants.

Cucumber plants were either self-grafted or grafted onto two salt-tolerant pumpkin rootstocks Chaojiquanwang (Cucurbita moschata Duch), and Figleaf Gourd (Cucurbita ficifolia Bouche). Plants were grown hydroponically in 0, 30, 60, or 90 mM NaCl for 16 d in greenhouse. Salinity induced a smaller decrease in plant shoot dry mass, leaf area, net photosynthetic rate, and stomatal conductance in the two rootstock-grafted plants compared to the self-grafted plants. In addition, a significant increase in intercellular CO2 concentration, as well as a significant decrease in the initial and total ribulose-1,5-bisphosphate carboxylase/oxygenase activities were observed only in the self-grafted plants under 90 mM NaCl treatment. These results suggest that the use of salt tolerant rootstock can improve cucumber photosynthetic capacity under salt stress through both stomatal and non-stomatal pathways.

39. A deep learning-based method enables the automatic and accurate assembly of chromosome-level genomes.

Abstract

The application of high-throughput chromosome conformation capture (Hi-C) technology enables the construction of chromosome-level assemblies. However, the correction of errors and the anchoring of sequences to chromosomes in the assembly remain significant challenges. In this study, we developed a deep learning-based method, AutoHiC, to address the challenges in chromosome-level genome assembly by enhancing contiguity and accuracy. Conventional Hi-C-aided scaffolding often requires manual refinement, but AutoHiC instead utilizes Hi-C data for automated workflows and iterative error correction. When trained on data from 300+ species, AutoHiC demonstrated a robust average error detection accuracy exceeding 90%. The benchmarking results confirmed its significant impact on genome contiguity and error correction. The innovative approach and comprehensive results of AutoHiC constitute a breakthrough in automated error detection, promising more accurate genome assemblies for advancing genomics research., (© The Author(s) 2024. 40. Tumor-associated macrophage clusters linked to immunotherapy in a pan-cancer census.

Abstract

Transcriptional heterogeneity of tumor-associated macrophages (TAMs) has been investigated in individual cancers, but the extent to which these states transcend tumor types and represent a general feature of cancer remains unclear. We performed pan-cancer single-cell RNA sequencing analysis across nine cancer types and identified distinct monocyte/TAM composition patterns. Using spatial analysis from clinical study tissues, we assessed TAM functions in shaping the tumor microenvironment (TME) and influencing immunotherapy. Two specific TAM clusters (pro-inflammatory and pro-tumor) and four TME subtypes showed distinct immunological features, genomic profiles, immunotherapy responses, and cancer prognosis. Pro-inflammatory TAMs resided in immune-enriched niches with exhausted CD8+ T cells, while pro-tumor TAMs were restricted to niches associated with a T-cell-excluded phenotype and hypoxia. We developed a machine learning model to predict immune checkpoint blockade response by integrating TAMs and clinical data. Abstract

Thousands of long noncoding RNAs (lncRNAs) have been annotated via high-throughput RNA sequencing, yet only a small fraction have been functionally investigated. Genomic knockout is the mainstream strategy for studying the biological function of protein-coding genes and lncRNAs, whereas the complexity of the lncRNA locus, especially the natural antisense lncRNAs (NAT-lncRNAs), presents great challenges. Knocking out lncRNAs often results in unintended disruptions of neighboring protein-coding genes and small RNAs, leading to ambiguity in observing phenotypes and interpreting biological function. To address this issue, we launched LncRNAway, a user-friendly web tool based on the BESST (branchpoint to 3' splicing site targeting) method, to design sgRNAs for lncRNA knockout. LncRNAway not only provides specific and effective lncRNA knockout guidelines but also integrates genotyping primers and quantitative PCR primers designing, thereby streamlining experimental procedures of lncRNA function study. Abstract

Long interspersed nuclear element-1 (LINE-1 or L1) is a retrotransposon group that constitutes 17% of the human genome and shows variable expression across cell types. However, the control of L1 expression and its function in gene regulation are incompletely understood. Here we show that L1 transcription activates long-range gene expression. Genome-wide CRISPR-Cas9 screening using a reporter driven by the L1 5' UTR in human cells identifies functionally diverse genes affecting L1 expression. Unexpectedly, altering L1 expression by knockout of regulatory genes impacts distant gene expression. L1s can physically contact their distal target genes, with these interactions becoming stronger upon L1 activation and weaker when L1 is silenced. Remarkably, L1s contact and activate genes essential for zygotic genome activation (ZGA), and L1 knockdown impairs ZGA, leading to developmental arrest in mouse embryos. These results characterize the regulation and function of L1 in long-range gene activation and reveal its importance in mammalian ZGA., (© 2024. Abstract

The impact of macroeconomic policy uncertainty (EPU) on micro-level entities has garnered increasing attention in economic circles. This study examines the influence of EPU on the efficiency of investments made by China's A-share listed companies between 2016 and 2021. Using a panel fixed effect model for analysis, the research reveals that EPU has a notable adverse effect on the investment efficiency of enterprises. Furthermore, it suggests that advancements in digital finance, strong ESG performance, and enhanced entrepreneurial confidence can mitigate this negative impact effectively. The study also highlights that enterprises with lower valuation, shareholder control, limited audit reputation, and no bank connections are more vulnerable to the impact of EPU on investment efficiency compared to those with higher valuation, manager control, strong audit reputation, and bank connections. Abstract

Aim: We investigated the effects and targets of gastrodin (GAS) for improving cognitive ability in Alzheimer's disease (AD)., Methods: The targets and mechanisms of GAS were analyzed by network pharmacology. Morris water and eight-arm radial mazes were used to detect the behaviors of 7-months-old APP/PS1 mice. The levels of IBA-1 and PPARγ were examined by histochemical staining, nerve cells were detected by Nissl staining, inflammatory cytokines were measured by ELISA, and protein expressions were monitored by Western blotting. The neurobehavioral effects of GAS on mice were detected after siRNA silencing of PPARγ. Microglia were cultured in vitro and Aβ1-42 was used to simulate the pathology of AD. After treatment with GAS, the levels of inflammatory cytokines and proteins were assayed., Results: Network pharmacological analysis revealed that PPARγ was the action target of GAS. By stimulating PPARγ, GAS inhibited NF-κB signaling activation and decreased neuroinflammation and microglial activation, thereby ameliorating the cognitive ability of AD mice. After silencing PPARγ, GAS could not further improve such cognitive ability. Cellular-level results demonstrated that GAS inhibited microglial injury, reduced tissue inflammation, and activated PPARγ., Conclusions: GAS can regulate microglia-mediated inflammatory response by stimulating PPARγ and inhibiting NF-κB activation, representing a mechanism whereby it improves the cognitive behavior of AD.

Abstract

Long interspersed element-1 (LINE-1 or L1) comprises 17% of the human genome, continuously generates genetic variations, and causes disease in certain cases. However, the regulation and function of L1 remain poorly understood. Here, we uncover that L1 can enrich RNA polymerase IIs (RNA Pol IIs), express L1 chimeric transcripts, and create contact domain boundaries in human cells. This impact of L1 is restricted by a nuclear matrix protein scaffold attachment factor B (SAFB) that recognizes transcriptionally active L1s by binding L1 transcripts to inhibit RNA Pol II enrichment. Acute inhibition of RNA Pol II transcription abolishes the domain boundaries associated with L1 chimeric transcripts, indicating a transcription-dependent mechanism. Deleting L1 impairs domain boundary formation, and L1 insertions during evolution have introduced species-specific domain boundaries. 47. The accuracy of different mismatch negativity amplitude representations in predicting the levels of consciousness in patients with disorders of consciousness.

Abstract

Introduction: The mismatch negativity (MMN) index has been used to evaluate consciousness levels in patients with disorders of consciousness (DoC). Indeed, MMN has been validated for the diagnosis of vegetative state/unresponsive wakefulness syndrome ( VS /UWS) and minimally conscious state (MCS). In this study, we evaluated the accuracy of different MMN amplitude representations in predicting levels of consciousness., Methods: Task-state electroencephalography (EEG) data were obtained from 67 patients with DoC (35 VS and 32 MCS). We performed a microstate analysis of the task-state EEG and used four different representations (the peak amplitude of MMN at electrode Fz (Peak), the average amplitude within a time window -25- 25 ms entered on the latency of peak MMN component (Avg for peak ± 25 ms), the average amplitude of averaged difference wave for 100-250 ms (Avg for 100-250 ms), and the average amplitude difference between the standard stimulus ("S") and the deviant stimulus ("D") at the time corresponding to Microstate 1 (MS1) (Avg for MS1) of the MMN amplitude to predict the levels of consciousness., Results: The results showed that among the four microstates clustered, MS1 showed statistical significance in terms of time proportion during the 100-250 ms period. Our results confirmed the activation patterns of MMN through functional connectivity analysis. Among the four MMN amplitude representations, the microstate-based representation showed the highest accuracy in distinguishing different levels of consciousness in patients with DoC (AUC = 0.89)., Conclusion: We discovered a prediction model based on microstate calculation of MMN amplitude can accurately distinguish between MCS and VS states. Aim: We aimed to assess the effects of cerebral glucagon-like peptide-1 receptor (GLP-1R) activation on the glymphatic system and whether this effect was therapeutic for traumatic brain injury (TBI)., Methods: Immunofluorescence was employed to evaluate glymphatic system function. The blood-brain barrier (BBB) permeability, microvascular basement membrane, and tight junction expression were assessed using Evans blue extravasation, immunofluorescence, and western blot. Immunohistochemistry was performed to assess axonal damage. Neuronal apoptosis was evaluated using Nissl staining, terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining, and western blot. Cognitive function was assessed using behavioral tests., Results: Cerebral GLP-1R activation restored glymphatic transport following TBI, alleviating BBB disruption and neuronal apoptosis, thereby improving cognitive function following TBI. Glymphatic function suppression by treatment using aquaporin 4 inhibitor TGN-020 abolished the protective effect of the GLP-1R agonist against cognitive impairment., Conclusion: Cerebral GLP-1R activation can effectively ameliorate neuropathological changes and cognitive impairment following TBI; the underlying mechanism could involve the repair of the glymphatic system damaged by TBI., (© 2023 The Authors. 49. Cancer cell-intrinsic PD-1: Its role in malignant progression and immunotherapy.

Abstract

Programmed cell death protein-1 (PD-1), also called CD279, is coded by the PDCD1 gene and is constitutively expressed on the surface of immune cells. As a receptor and immune checkpoint, PD-1 can bind to programmed death ligand-1/programmed death ligand-2 (PD-L1/PD-L2) in tumor cells, leading to tumor immune evasion. Anti-PD-1 and anti-PD-L1 are important components in tumor immune therapy. PD-1 is also expressed as an intrinsic variant (iPD-1) in cancer cells where it plays important roles in malignant progression as proposed by recent studies. However, iPD-1 has received much less attention compared to PD-1 expressed on immune cells although there is an unmet medical need for fully elucidating the mechanisms of actions to achieve the best response in tumor immunotherapy. iPD-1 suppresses tumorigenesis in non-small cell lung cancer (NSCLC) and colon cancer, whereas it promotes tumorigenesis in melanoma, hepatocellular carcinoma (HCC), pancreatic ductal adenocarcinoma (PDAC), thyroid cancer (TC), glioblastoma (GBM), and triple-negative breast cancer (TNBC). In this review, we focus on the role of iPD-1 in tumorigenesis and development and its molecular mechanisms. Advancements in comparative genomics research have led to a growing interest in studying species evolution and genetic diversity. To facilitate this research, OrthoVenn3 has been developed as a powerful, web-based tool that enables users to efficiently identify and annotate orthologous clusters and infer phylogenetic relationships across a range of species. The latest upgrade of OrthoVenn includes several important new features, including enhanced orthologous cluster identification accuracy, improved visualization capabilities for numerous sets of data, and wrapped phylogenetic analysis. Furthermore, OrthoVenn3 now provides gene family contraction and expansion analysis to support researchers better understanding the evolutionary history of gene families, as well as collinearity analysis to detect conserved and variable genomic structures.
