Cave heritage is often threatened by tourism or even scientific activities, which can lead to irreversible deterioration. We present a preventive conservation monitoring protocol to protect caves with rock art, focusing on La Garma Cave (Spain), a World Heritage Site with valuable archaeological materials and Palaeolithic paintings. This study assessed the suitability of the cave for tourist use through continuous microclimate and airborne particles monitoring, biofilm analysis, aerobiological monitoring and experimental visits. Our findings indicate several factors that make it inadvisable to adapt the cave for tourist use. Human presence and transit within the cave cause cumulative effects on the temperature of environmentally very stable and fragile sectors and significant resuspension of particles from the cave sediments. These environmental perturbations represent severe impacts as they affect the natural aerodynamic control of airborne particles and determine bacterial dispersal throughout the cave. This monitoring protocol provides part of the evidence to design strategies for sustainable cave management.

The genus Crossiella contains two species, C. equi, causing nocardioform placentitis in horses, and C. cryophila, an environmental bacterium. Apart from C. equi, which is not discussed here, environmental Crossiella is rarely reported in the literature; thus, it has not been included among “rare actinobacteria”, whose isolation frequency is very low. After C. cryophila, only five reports cover the isolation of Crossiella strains. However, the frequency of published papers on environmental Crossiella has increased significantly in recent years due to the extensive use of next-generation sequencing (NGS) and a huge cascade of data that has improved our understanding of how bacteria occur in the environment. In the last five years, Crossiella has been found in different environments (caves, soils, plant rhizospheres, building stones, etc.). The high abundance of Crossiella in cave moonmilk indicates that this genus may have an active role in moonmilk formation, as evidenced by the precipitation of calcite, witherite, and struvite in different culture media. This review provides an overview of environmental Crossiella, particularly in caves, and discusses its role in biomineralization processes and bioactive compound production.

The Cross of the Inquisition, sculpted in 1903 and raised on a column with a fluted shaft and ornamented with vegetable garlands, is located in a corner of the Plateresque façade of the Seville City Hall. The Cross was vandalized in September 2019 and the restoration concluded in September 2021. A geological and microbiological study was carried out in a few small fragments. The data are consistent with the exposure of the Cross of the Inquisition to an urban environment for more than 100 years. During that time, a lichen community colonized the Cross and the nearby City Hall façades. The lichens, bryophytes and fungi colonizing the limestone surface composed an urban community, regenerated from the remains of the original communities, after superficial cleaning of the limestone between 2008 and 2010. This biological activity was detrimental to the integrity of the limestone, as showed by the pitting and channels, which evidence the lytic activity of organisms on the stone surface. Stone consolidation was achieved with Estel 1000. Preventol RI80, a biocide able to penetrate the porous limestone and active against bacteria, fungi, lichens, and bryophytes, was applied in the restoration.

Of the several critical challenges present in environmental microbiology today, one is the assessment of the contribution of microorganisms in the carbon cycle in the Earth-climate system. Karstic subterranean ecosystems have been overlooked until recently. Covering up to 25% of the land surface and acting as a rapid CH4 sink and alternately as a CO2 source or sink, karstic subterranean ecosystems play a decisive role in the carbon cycle in terms of their contribution to the global balance of greenhouse gases. Recent data indicate that microbiota must play a significant ecological role in the biogeochemical processes that control the composition of the subterranean atmosphere, as well as in the availability of nutrients for the ecosystem. Nevertheless, there are still essential gaps in our knowledge concerning the budgets of greenhouse gases at the ecosystem scale and the possible feedback mechanisms between environmental-microclimatic conditions and the rates and type of activity of microbial communities in subterranean ecosystems. Another challenge is searching for bioactive compounds (antibiotics) used for treating human diseases. At present, there is a global health emergency and a strong need for novel biomolecules. In recent decades, great research efforts have been made to extract antibiotics from marine organisms. More recently, caves have been receiving considerable attention in search of novel antibiotics. Cave methanotrophic and heterotrophic bacteria are producers of bioactive compounds and may be potential sources of metabolites with antibacterial, antifungal or anticancer activities of interest in pharmacological and medical research, as well as enzymes with a further biotechnological use. Here we also show that bacteria isolated from mines, a still unexplored niche for scientists in search of novel compounds, can be a source of novel secondary metabolites.

The Roman Necropolis of Carmona (Sevilla, Spain) has experienced a severe deterioration since the accidental discovery in 1868 and subsequent archaeological excavations starting in 1882. To this deterioration contributed the location in a soft calcarenite rock quarry, the adverse environmental conditions and the numerous and extensive wrong archaeological and managing interventions along more than a century. The cultural, artistic and religious importance of this Necropolis converts the protection and conservation of this archaeological site in a major issue. This work present the data obtained in a multidisciplinary research were geology, geomorphology, micro-environmental and climatic monitoring, rock petrophysical characterization, description of weathering forms and biological colonization were considered in order to propose corrective measures to minimize deterioration.

El objetivo principal de este trabajo es el conocimiento de la diversidad, concentración y distribución estacional y espacial de las comunidades bacterianas y fúngicas de tres cuevas andaluzas. Las cuevas seleccionadas son representativas de cavidades de interés cultural, como la Cueva de Ardales (Ardales, Málaga) y la Cueva del Tesoro (Rincón de la Victoria, Málaga), las cuales albergan pinturas rupestres, y de interés geológico, como es el caso de la Gruta de las Maravillas (Aracena, Huelva). Estas tres cuevas mantienen distintos regímenes de visitas, por lo que podremos evaluar el impacto del turismo. Los datos proporcionados por los gestores de las cuevas sobre el número de visitas registradas en el año 2011 mostraron que la Cueva de Ardales fue visitada por 4.018 personas, la Cueva del Tesoro recibió 28.257 visitantes y la Gruta de las Maravillas 130.314. La Cueva de Ardales, también conocida como Cueva de Doña Trinidad, se localiza en el municipio de Ardales, provincia de Málaga. Fue descubierta en 1821 tras un movimiento sísmico que dejó al descubierto la boca de acceso a la cavidad. Fue habilitada y abierta a las visitas turísticas en el año 1852, y declarada Monumento Nacional en 1931, aunque su protección no se hizo efectiva. En 1992 la Consejería de Cultura de la Junta de Andalucía declaró la cavidad Bien de Interés Cultural y el Ayuntamiento de Ardales asumió su gestión. En ella se han encontrado numerosos vestigios procedentes de las ocupaciones prehistóricas y un conjunto de representaciones gráficas paleolíticas, que incluyen pinturas y grabados. En la actualidad mantiene un régimen de visitas controlado, con una media anual de 1.000 visitantes (Fernández‐Cortés y col., 2008). En esta cavidad se han realizado estudios previos sobre las condiciones microambientales y las comunidades microbianas presentes en el aire (Fernandez‐ Cortes y col., 2011), en los sedimentos y en las colonizaciones microbianas que cubren los espeleotemas (Stomeo y col., 2008).

In recent years, methane (CH4) has received increasing scientific attention because it is the most abundant non-CO2 atmospheric greenhouse gas (GHG) and controls numerous chemical reactions in the troposphere and stratosphere. However, there is much that is unknown about CH4 sources and sinks and their evolution over time. Here we show that near-surface cavities in the uppermost vadose zone are now actively removing atmospheric CH4. Through seasonal geochemical tracing of air in the atmosphere, soil and underground at diverse geographic and climatic locations in Spain, our results show that complete consumption of CH4 is favoured in the subsurface atmosphere under near vapour-saturation conditions and without significant intervention of methanotrophic bacteria. Overall, our results indicate that subterranean atmospheres may be acting as sinks for atmospheric CH4 on a daily scale. However, this terrestrial sink has not yet been considered in CH4 budget balances.

The Circular Mausoleum tomb in the Roman Necropolis of Carmona was carved on a calcarenite sequence in an ancient quarry located in the town of Carmona, Southern Spain. This rock-cut tomb, representative of Roman burial practices, currently suffers from serious deterioration. A detailed survey over several years permitted the identification of the main tomb's pathologies and damaging processes, which include loss of material (scaling, flaking, granular disintegration), surface modifications (efflorescences, crusts and deposits) and extensive biological colonization. The results obtained in this study indicated that anthropogenic changes were largely responsible and enhanced the main alteration mechanisms observed in the Circular Mausoleum. Based on the deterioration diagnosis, effective corrective actions were proposed. This study shows that any conservative intervention in the interior of the tomb should be preceded by accurate in situ measurements and laboratory analyses to ascribe the source of the deterioration damages and thus designing effective treatments.

San Roque church (Campeche, Mexico) was built at the end of the 17th century with a micritic limestone and lime mortar in baroque style. In 2005 the church exhibited heavy biodeterioration associated with the development of extensive dark green phototrophic-based biofilms. Several cyanobacteria belonging to the order Chroococcales and lichenized fungi (Toninia nordlandica, Lobaria quercizans, Lecanora subcarnea, Cystocoleus ebeneus) were predominant in the dark biofilm samples, as revealed by DNA-based molecular techniques. In 2009, a cleaning and restoration intervention was adopted; however, after few months, microbial recolonization started to be noticeable on the painted church walls, representing an early phototrophic-based recolonization. According to molecular analysis, scanning electron microscopy observations and digital image analysis of cross sections, new phototrophic-based colonization, composed of cyanobacteria and bryophytes, developed mainly beneath the restored mortars. The intrinsic properties of the mortars, the tropical climate of Campeche and the absence of a biocide treatment in the restoration protocol influenced the recolonization of the church façades and enhanced the overall rate of deterioration in a short-term period.

The Etruscan civilisation originated in the Villanovan Iron Age in the ninth century BC and was absorbed by Rome in the first century BC. Etruscan tombs, many of which are subterranean, are one of the best representations of this culture. The principal importance of these tombs, however, lies in the wall paintings and in the tradition of rich burial, which was unique in the Mediterranean Basin, with the exception of Egypt. Relatively little information is available concerning the biodeterioration of Etruscan tombs, which is caused by a colonisation that covers the paintings with white, circular to irregular aggregates of bacteria or biofilms that tend to connect each other. Thus, these colonisations sometimes cover extensive surfaces. Here we show that the colonisation of paintings in Tomba del Colle is primarily due to bacteria of the order Rhizobiales (Alphaproteobacteria), which were likely influenced by the neighbouring rhizosphere community and the availability of nutrients from root exudates.

The paintings from Tomba della Scimmia, in Tuscany, are representative of the heavy bacterial colonization experienced in most Etruscan necropolises. The tomb remained open until the late 70′s when it was closed because of severe deterioration of the walls, ceiling and paintings after decades of visits. The deterioration is the result of environmental changes and impacts suffered since its discovery in 1846. We show scanning electron microscopy and molecular studies that reveal the extent and nature of the biodeterioration. Actinobacteria, mainly Nocardia and Pseudonocardia colonize and grow on the tomb walls and this process is linked to the availability of organic matter, phyllosilicates (e.g. clay minerals) and iron oxides. Nocardia is found metabolically active in the paintings. The data confirm the specialization of the genera Nocardia and Pseudonocardia in the colonization of subterranean niches.
