It has been demonstrated that resveratrol can inhibit tumor proliferation and metastasis, but its application is limited due to poor water solubility and low bioavailability. In this study, we proposed to prepare folate-modified liposomes loaded with resveratrol to investigate its anti-osteosarcoma effect in vitro and in vivo.Methods: We prepared and characterized resveratrol liposomes modified with folate (denoted as, FA-Res/Lps). The effects of FA-Res/Lps on human osteosarcoma cell 143B proliferation, apoptosis, and migration were investigated by MTT, cell cloning, wound-healing assay, transwell, and flow cytometry. A xenograft tumor and lung metastasis model of osteosarcoma was constructed to study the therapeutic effects of FA-Res/Lps on the growth and metastasis of osteosarcoma in vivo.Results: The FA-Res/Lps were prepared with a particle size of 118.5 ± 0.71 and a small dispersion coefficient of 0.154 ± 0.005. We found that FA-modified liposomes significantly increased resveratrol uptake by osteosarcoma cells 143B in flow cytometric assay, resulting in FA-Res/Lps, which inhibit tumor proliferation, migration and induce apoptosis more effectively than free Res and Res/Lps. The mechanism of action may be associated with the inhibition of JAK2/STAT3 signaling. In vivo imaging demonstrated that FA-modified DiR-modified liposomes significantly increased the distribution of drugs at the tumor site, leading to significant inhibition of osteosarcoma growth and metastasis by FA-Res/Lps. Furthermore, we found that FA-Res/Lps did not cause any adverse effects on mice body weight, liver, or kidney tissues.Conclusion: Taken together, the anti-osteosarcoma effect of resveratrol is significantly enhanced when it is loaded into FA-modified liposomes. Our previous study revealed that the combination of Rapamycin liposomes (Rapa/Lps) and 5-Fluorouracil (5-FU) has anti-colorectal cancer effects. However, the therapeutic efficacy of Rapa/Lps and 5-FU in other colorectal cancer mice models is yet to be thoroughly explored. The purpose of this study was to investigate the anti-tumor effect of Rapa/Lps combined with 5-FU in vivo and in vitro.Methods: In this study, we evaluated the effect of Rapa/Lps and 5-FU on APC (Min/+) mice and AOM/DSS-induced colorectal cancer mice. The small intestine, colorectum, serum, and plasma of mice in each group were collected following sacrifice to record the number of tumors. HE staining was utilized for observing pathological damage to intestine tissues. Tube formation assay, Transwell assay, wound healing assay, Western Blot were used to explore the anti-angiogenesis effect of drugs in HUVECs.Results: As expected, Rapa/Lps and 5-FU significantly suppressed tumor formation, decreased the number of tumors, and tumor load both in two mouse models, and had no influence on mouse weight. Mechanically, the anti-tumor effect of the drug also was associated in inhibiting angiogenesis and proliferation. Furthermore, we found that Rapa/Lps obviously inhibited HUVECs tube formation and migration.Conclusion: Altogether, we revealed the Rapa/Lps synergism with 5-FU decreased colon and small intestinal tumorigenesis in AOM/DSS-treated and APC (Min/+) mice, respectively, and correlated with anti-angiogenesis.Keywords: rapamycin liposomes, 5-fluorouracil, angiogenesis, APC (Min/+) mice, AOM/DSS, colorectal cancer

Objective: To assess a safe surgical approach for intertransverse process lower thoracic intervertebral body fusion (ITIF) based on measurements from enhanced three-dimensional CT reconstruction, cadaver simulated operation, and patient operation., Methods: Enhanced three-dimensional CT image reconstruction was performed for 20 healthy volunteers on thoracic segments T8-T12. The length of the transverse process (LTP), distance between the upper and lower transverse processes (DULTP), remote distance of the transverse process (RDTP), height of the extraforaminal intervertebral space (HEIS), and oblique diameter of the intervertebral space (ODIS) were measured and recorded. The blood vessels of the intertransverse lower thoracic region were observed, and their internal diameters were measured. The rib-intervertebral space relationship for T10/11 and T11/12 was measured in 104 patients of the thoracic skeleton. Then, based on the data from the CT measurements, simulated surgery was performed on six human cadavers at the T11/12 level. An ankylosing spondylitis (AS) patient with a fracture of the T10/11 level was eventually operated on with the ITIF technique., Results: No significant difference was found between the lengths of the left and right thoracic transverse processes. The relationship of the values of the LTP and RDTP for the measured vertebrae were found to be as follows:T8 > T9 > T10 > T11 > T12. For HEIS and DULTP, T8-9 < T9-10 < T10-11 < T11-12. The results for the ODIS were as follows: T8-T9 < T9-T10 < T10-T11 < T11-T12. The blood vessel inner diameter of T11-12 was less than that of T10-11, while there was no significant difference between the diameters for T8-9 and T11-12. Almost half of the volunteer's T10/11 intervertebral spaces were covered posteriorly by the 11th rib (45.19% on left and 41.35% on right), while for most patients, the T11/12 intervertebral space was not covered by the 12th rib (98.08%). According to the cadaver experiments, intervertebral bone grafting and ipsilateral pedicle screw fixation were performed to simulate the operation. One patient with a combined AS and T10/11 fracture was then operated on with the ITIF technique and followed up for 3 years with satisfactory results., Conclusion: As verified by 3D CT reconstruction measurements, cadaver simulation surgery and patient operation with follow-up, the intertransverse process approach for some T10/T11 and almost all T11/T12 segments is a safe surgical pathway for operations such as ITIF, fracture bone grafting, clearance of focal lesions., (© 2022 The Authors. A new esterase gene est906 was identified from paper mill wastewater sediments via a function-based metagenomic approach. The gene encoded a protein of 331 amino acids, that shared 86% homology with known esterases. Based on the results of multiple sequence alignment and phylogenetic analysis, it was confirmed that Est906 contained a characteristic hexapeptide motif (G-F-S-M-G-G), which classified it as a lipolytic enzyme family V protein. Est906 displayed the highest hydrolysis activity to ρ-nitrophenyl caproate (C6), and its optimal temperature and pH were 54 °C and 9.5, respectively. Additionally, this enzyme had good stability under strong alkaline conditions (pH 10.0-11.0) in addition to moderate heat resistance and good tolerance against several metal ions and organic solvents. Furthermore, a specific nucleic acid aptamer (Apt1) bound to Est906 was obtained after five rounds of magnetic bead SELEX screening. Apt1 displayed high specific recognition and capture ability to Est906. In conclusion, this study not only identified a new esterase of family V with potential industrial application by metagenomic technology but also provided a new method to purify recombinant esterases via nucleic acid aptamers, which will facilitate the isolation and purification of target proteins in the future., Competing Interests: Declaration of competing interest We declare that we have no conflicts of interest., (Copyright © 2020 Elsevier B.V. To what extent are research results influenced by subjective decisions that scientists make as they design studies? Fifteen research teams independently designed studies to answer five original research questions related to moral judgments, negotiations, and implicit cognition. Participants from 2 separate large samples (total N > 15,000) were then randomly assigned to complete 1 version of each study. Effect sizes varied dramatically across different sets of materials designed to test the same hypothesis: Materials from different teams rendered statistically significant effects in opposite directions for 4 of 5 hypotheses, with the narrowest range in estimates being d = -0.37 to + 0.26. Meta-analysis and a Bayesian perspective on the results revealed overall support for 2 hypotheses and a lack of support for 3 hypotheses. Overall, practically none of the variability in effect sizes was attributable to the skill of the research team in designing materials, whereas considerable variability was attributable to the hypothesis being tested. In a forecasting survey, predictions of other scientists were significantly correlated with study results, both across and within hypotheses. Crowdsourced testing of research hypotheses helps reveal the true consistency of empirical support for a scientific claim. (PsycInfo Database Record (c) 2020 APA, all rights reserved).

A new bacterial lipolytic enzyme Est903 was obtained from paper mill sludge via metagenomic approach. Est903 displayed moderate similarities to two lipolytic enzymes from Rhodopirellula islandica and contained a distinctive pentapeptide motif (GFSAG) that differed from those of all known fourteen families of bacterial lipolytic enzymes. Est903 was regarded as from a new bacterial lipolytic enzyme family through multiple sequence alignment and phylogenetic analysis. The recombinant Est903 showed the highest activity for ρ-nitrophenol butyrate. The pH optimum and temperature optimum of the recombinant enzyme was 9.0 and 51 °C, respectively. Also, this enzyme displayed moderate thermostability, high activity under alkaline conditions, and good tolerance against several organic solvents. In addition, the compatibility test and washing performance analysis revealed that Est903 had good compatibility with commercial laundry detergent and high cleaning ability of oil stains. NO can be removed at the same time with SO 2 by aqueous Co(NH 3 ) 6 2+ solution. The reduction of Co(NH 3 ) 6 3+ to Co(NH 3 ) 6 2+ is catalyzed by activated carbon to regain the NO absorption ability of the scrubbing solution. Oxalic acid solution is explored to change the carbon surface to ameliorate its catalytic capability. The experimental results suggest that the best catalyst is prepared by impregnating the carbon sample in 0.7 mol l -1 oxalic acid solution for 24 h followed by being activated at 600 °C for 5 h under nitrogen atmosphere. After being treated with oxalic acid solution, the surface area and the acidity on the carbon surface increase. The experiments show that the carbon modified with oxalic acid can get a much higher NO removal efficiency than the original carbon.

A new protease gene (pro1437)was separated from an oil-polluted Mud flat metagenomic library. Pro1437 belongs to a peptidase M48 superfamily according to the results of sequence analysis, and it showed very low identities compared to other known proteases or peptidases. The error-prone PCR was used to introduce random mutations and improve the expression of pro1437. After two rounds of mutagenesis and screening, a mutant (Pro2T21) with a 6.6-fold higher activity and a 4.8-fold higher expression level than Pro1437 was obtained. Sequence analysis found three amino acid substitutions (A54V, L192H, F224L) in Pro2T21. 3D structure modelling analysis indicated A54V and L192H probably played a crucial role in the improvement of enzymatic activity and soluble expression level of Pro2T21. Furthermore, Pro2T21opti displayed a 5.8-fold higher expression level than the wild type under optimal pH 8.0 at 50°C after codon-optimization. Also, Pro2T21opti represented robust compatibility with several popular laundry detergents, and blood stains on white cloth pieces were completely washed away when endogenous protease-inactivated Tide and Pro2T21opti were used together. A total of 36 kinds of soil samples were collected from apple orchards under three kinds of management model, including clear tillage model, intercropping white clover model and intercropping small crown flower model, the mineralization of soil organic carbon and four kinds of soil enzymes activities were determined, and the relationship between the two parameters was also analyzed. The results showed that the soil organic carbon mineralization of all the three treatments was almost the same. The daily SOC mineralization rate first increased and then decreased, and finally tended to be stable. After 31 days incubation experiment, the maximum accumulative amount of SOC mineralization occurred in white clover treatment with 590 mg x kg(-1), followed by small crown flower treatment with 541 mg x kg(-1), and the minimum value was 367 mg x kg(-1) in the control treatment, and the accumulative amount of SOC mineralization decreased with increasing soil depth. Discovered by the first-order kinetics, the fitting parameter Cp value ranged from 0.252 to 2.74 g x kg(-1) and k value ranged from 0.019 to 0.051 d(-1), and the two grass treatments both showed significant difference in Cp value from the control treatment, and the soil invertase and cellulose activities showed obvious relationship with soil organic carbon mineralization.

Objective: To analyze the clinical features, risk factors and drug uses of senior hospitalized patients with chronic constipation., Methods: A total of 162 hospitalized patients aged 85 years and over at our hospital during the period of January-March 2012 conducted a questionnaire survey. There were 137 males and 25 females. And 112 cases of chronic constipation were diagnosed in accordance with the Rome III criteria. The survey included general condition, risk factors of constipation, spectrum of symptoms, associated symptoms, previous medication and medication for constipation after admission. The results were statistically analyzed by Logistic regression., Results: Their average age was (90 ± 4) years old. And the total prevalence of chronic constipation was 69.1% (112/162). Logistic regression analysis showed that less activity (OR = 10.873) and diet (OR = 4.752) were the important risk factors. Defecation effort was the most common symptom (n = 85, 75.9%). A total of 88 cases (78.6%) took lactulose alone or lactulose plus other laxatives. The reasons of using lactulose were as follows: dietary restrictions (n = 31, 35.2%), aspiration prevention (n = 21, 23.9%) and poor efficacy of other laxatives (n = 36, 40.9%)., Conclusions: Prevalence of constipation is high among senior hospitalized patients. Less activity and diet are the main risk factors. And lactulose should be a first-line therapy.

Background: Although the migration of hepatic stellate cells (HSCs) is essential for hepatic fibrotic response, the detailed mechanisms involved are poorly understood. The aim of this study was to examine the role of Rho GTPases (especially RhoA) in platelet-derived growth factor (PDGF)-BB-induced migration of HSCs., Methods: The migration of primary rat HSCs was evaluated using transwell Boyden chamber, while cytoskeletal changes were visualized by immunofluorescence staining of intracellular actins and vinculin. Quantitative real-time PCR and Western blotting analysis were used to detect the expression of Rho GTPases (RhoA, Rac1 and Cdc42) within HSCs and their activation was determined by glutathione S-transferase pull-down assay. Finally, the effects of RhoA on PDGF-BB-induced cell migration and cytoskeletal remodeling were analyzed using HSC-T6 cells stably transfected with constitutively active (CA, Q63L) or dominant negative (DN, T19N) RhoA mutants. Data were analyzed using SPSS 16.0 software. Student's t test was used to analyze differences between two groups and one-way analysis of variance (ANOVA) was used among multiple groups., Results: Rapid cytoskeletal remodeling led to a significant increase in the motility of primary rat HSCs after haptotactic (direct) and chemotactic (indirect) stimulation by PDGF-BB. PDGF-BB caused a dramatic elevation in the levels of both total and active RhoA protein. However, the levels of mRNA for Rho GTPases, including RhoA, Rac1 and Cdc42, were unaffected. Furthermore, PDGF-BB induced increased formation of stress fibers and focal adhesions in HSC-T6 cells transfected with CA-RhoA, but not in HSC-T6 transfected with DN-RhoA. Surprisingly, both CA- and DN-RhoA-transfected HSC-T6 cells showed decreased migratory potential in the absence or presence of PDGF-BB compared with controls., Conclusions: PDGF-BB induced cytoskeletal remodeling in rat HSCs and promoted their migration via regulation of intracellular RhoA. RhoA may be one of the determinants in PDGF-BB-induced HSC migration.

A series of Co-V-O (meta-CoV2O6, pyro-Co2 V2 O7, and ortho-Co3 V2 O8) catalysts were prepared by microwave oxalate co-precipitation method and characterized by (XRD), TEM, BET, FTIR, XPS, H2-TPR and conductivity measurement. The catalytic characters of the catalysts for propane oxidative dehydrogenation were investigated. The FTIR spectra of catalysts were obtained in the range of 400-1 100 cm(-1) and their major bands were assigned. The peak separation fitting of O(1s) XPS spectra was carried out and the quantity of oxygen species was calculated. The results of XRD characterization showed that pure meta-CoV2O6, pyro-Co2 V2O7, and ortho-Co3 V2O8 with nice structure were obtained. The TEM images demonstrated that the catalysts showed uniform particle with the mean particle size of 20-30 nm. The diagram of the relationship between electrical conductivity and oxygen partial pressure of Co3V2O8 and Co2 V2O7 showed dsigma/dPo2 > 0, which implied that these were p-type semiconductor, and CoV2O6 reverse showed dsigma/dPo2 < 0, which implied n-type semiconductor. 48.12%, 47.82% and 35.24% of C3 H6 selectivities were obtained for p-type semiconductor Co3 V2O8, CO2 V2O7 and n-type CoV2O6 catalysts respectively at 10% C3H6 conversion, and the results showed that p-type semiconductor catalysts Co3 V2O8 and Co2 V2O7 showed higher activity than n-type catalyst CoV2O6. The results of FTIR, XPS, H2-TPR and conductivity measurement indicated that transferring between non-stoichiometric and lattice oxygen that easily happened in Co3 V2O8 and Co2 V2O7 catalysts might promote the oxidation-reduction reaction between different valence vanadium species, and promoted the oxygen vacancy formation. Furthermore, the forming of Co-O-V bridge bond that was easy to shift between Co and V increased the mobile oxygen species of O2-, O2(2-) and O- and made the redox reaction among different valence V be realized. It is concluded that high catalytic properties of p-type semiconductor Co3 V2O8 and Co2 V2O7 can be attributed to the abundant oxygen species O- that existed in these catalysts.

During the study of the Mongolian medicine, we found that the Mongolian medicine contains great amount and a variety of trace elements needed for human body, but the heavy metals are very little. A method for the determination of Ca, Cu, Zn, Fe, Mg, Mn and Pb in the Mongolian medicine. (1# the morning medicine, 2# the noon medicine and 3# the evening medicine.) by ICP-AES with microwave, digestion was studied. The recoveries of standard addition are in the range of 94.63%-106.40%. The relative standard deviation is < or = 3.41, and detection limit is in the range of < or = 0.009 microg x L(-1). The results show that 1#, 2# and 3# are different kinds of medicine with some difference in the trace elements quantity, but can cure hyperlipidemia disease by their combination.

Ni3V2O8 catalyst was prepared by oxalate co-precipitation method with microwave heating in this paper. In order to study the relationship between the catalytic performance and the surface species, the catalyst was characterized by XRD, BET, H2-TPR, XPS, TEM and conductivity measurement. The surface property of Ni3V2O8 was studied by XPS and the catalytic performance of the oxidative dehydrogenation of propane to propylene was also investigated. The results of XRD showedthat pure Ni3V2O8 with nice structure was obtained. TEM experiments results demonstrated that the prepared Ni3V2O8 catalyst at 700 degrees C calcination showed uniform particle with the mean particle size of 30 nm. The surface area of the catalyst was 8.623 m2 x g(-1). The diagram of the relationship between electrical conductivity and oxygen partial pressure of Ni3V2O8 showed dsigma/dPO2, >0, implying that Ni3V2O8 catalyst was a p-type semiconductor. H2-TPR results showed that only one unsymmetrical reduction peak appeared at 663.5 degreesC within 300-900 degrees C region over Ni3V2O8 catalyst and no obvious shoulder peak was observed. It could also be found that the ratio of non complete reduction oxygen species was about 33.59% (O(-) 27.55%, O2(2-) 6.04%) from the O(1s) XPS result and more V4+ species existed on the Ni3V2O8 catalyst surface. The TPR and XPS results illustrated that the transformation of the lattice oxygen to non-complete reduction oxygen in NiV2O8 catalyst might promote the oxidation-reduction reaction between different valence vanadium and promoted the oxygen vacancy formation. This then led to abundant non-complete reduction oxygen O(-) and V4+ species formation on the surface of Ni3V2O8 catalyst. The active result of oxidative dehydrogenation of propane to propylene showed that the 60.02% propylene selectivity could be reached at 18.60% propane conversion. Compared with the reported results over the coexistent NiO and Ni3V2O8 system from the literature, pure Ni3V2O8 catalyst system in this present paper showed higher propylene selectivity than the coexistent NiO and Ni3V2O8 system under the same propane conversion condition, suggesting that the performance of propane to propene is correlated to the oxidation-reduction of V4+ / V5+ couple and non complete reduction oxygen species (O(-) or O2(2-)). This result further illustrated that NiV2O8 was active phase for oxidative dehydrogenation of propane to propylene. Combining the active and characterization results, it was found that catalytic activity was correlated to the surface non-complete reduction O(-) and V4+ species, which was beneficial to improving the propylene selectivity.

Objective: To investigate whether the insertion/deletion(I/D) polymorphism in the angiotensin converting enzyme(ACE) gene is associated with essential hypertension in Xinjiang Kazakh isolated population., Methods: The study covered 201 hypertensives and 151 normotensive controls in Xinjiang Barlikun Kazakh population. The I/D polymorphism of ACE gene was determined by polymerase chain reaction., Results: The frequencies of D and I in the hypertensive group (0.44 and 0.56, respectively) were not significantly different from the controls(0.39 and 0.61, respectively, P=0.16). The frequencies of ACE genotypes of DD, ID, and II were 0.18, 0.52, 0.30 in hypertensives respectively and 0.17, 0.43, 0.40 in control group respectively. There was no significant difference in genotypes between hypertensive group and normotensive group (P=0.14)., Conclusion: The results suggested that the I/D polymorphism of ACE gene might not be associated with hypertension in the Kazakh population of Xinjiang Barlikun area.

Aim: To explore the possible mechanism why drinking Maotai liquor dose not cause hepatic fibrosis., Methods: After being fed with Maotai for 56 days consecutively, the male SD rats were decollated for detecting the biological indexes, and the livers were harvested to examine the liver indexes and the level of hepatic metallothioneins (MT). Hepatic stellate cells (HSC) proliferation and collagen generation were also observed., Results: Hepatic MT contents were 216.0 ng.g(-1)+/-10.8 ng.g(-1) in the rats of Maotai group and 10.0 ng.g(-1)+/-2.8 ng.g(-1) in the normal control group, which was increased obviously in Maotain group (P<0.05). In the rats with grade CCL(2) poisoning induced by Maotai, hepatic MT content was 304.8 ng.g(-1)+/-12.1 ng.g(-1) whereas in the controls with grade CCL(4) poisoning, it was 126.4 ng.g(-1)+/-4.8 ng.g(-1) (P<0.05). MDA was 102.0 nmol.g(-1)+/-3.4 nmol.g(-1) in Maotai group and 150.8 nmol.g(-1)+/-6.7 nmol.g(-1) in the control group (P<0.05). When both of the groups were suffering from grade CCL(4) poisoning, hepatic MT contents was negatively correlated with MDA (r=-0.8023, n=20, P<0.01). The 570 nmA values of each tube with HSC regeneration at concentrations of 0, 10, 50, 100, and 200 g.L(-1) of Maotai were 0.818, 0.742, 0.736, 0.72, 0.682, and 0.604, respectively. From the concentration of 10 g.L(-1), Maotai began to show obvious inhibitory effects against HSC, and the inhibition was concentration-dependent (P<0.05, P<0.01). Type I collagen contents in HSC were 61.4, 59.9, 50.1, 49.2, 48.7, 34.4 microg.g(-1) at concentrations of 0, 10, 50, 100, and 200 g.L(-1) of Maotai. At the concentration of 100-200 g.L(-1), Maotai had obvious inhibitory effect against the secretion of type I collagen (P<0.05). Gene expression analysis was conducted on cells with Maotai concentrations of 0, 50, 100g.L(-1) respectively and the ash values of beta-actin gene expression were 0.88, 0.74, and 0.59, respectively,suggesting that at the concentration of 100g.L(-1), Maotai could obviously inhibit gene expression of type I procollagen (P<0.05), but the effect was not obvious at the concentration of 50 g.L(-1) (P>0.05). At the concentration of 10 g.L(-1), HSC growth in vitro inhibition rates were 16.4+/-2.3 in Maotai group and -8.4+/-2.3 in the control group (P<0.05)., Conclusion: Maotai liquor can increase metallothioneins in the liver and inhibit the activation of HSC and the synthesis of collagen in many aspects, which might be the mechanism that Maotai liquor interferes in the hepatic fibrosis.
