Summary: The 'assimilates inhibition hypothesis' posits that accumulation of nonstructural carbohydrates (NSCs) in leaves reduces leaf net photosynthetic rate, thus internally regulating photosynthesis. Experimental work provides equivocal support mostly under controlled conditions without identifying a particular NSC as involved in the regulation.We combined 3‐yr in situ leaf gas exchange observations (natural dynamics) in the upper crown of mature Betula pendula simultaneously with measurements of concentrations of sucrose, hexoses (glucose and fructose), and starch, and similar measurements during several one‐day shoot girdling (perturbation dynamics). Leaf water potential and water and nitrogen content were measured to account for their possible contribution to photosynthesis regulation.Leaf photosynthetic capacity (A/Ci) was temporally negatively correlated with NSC accumulation under both natural and perturbation states. By contrast, the strong decline in photosynthetic capacity with NSCs accumulation in mature leaves, observed most clearly with hexose, and even more tightly with its constituents, provides support for the role of assimilates in regulating photosynthesis under natural conditions. The accumulation of sugars during fruit development influences not only sweetness but also various parameters controlling fruit composition (fruit “quality”). The accumulation of organic acids and sugar in grape berry flesh cells is a key process for berry development and ripening. The present review presents an update of the research on grape berry development, anatomical structure, sugar and acid metabolism, sugar transporters, and regulatory factors.

Hydrogen produced from lignocellulosic biomass via dark fermentation is a promising alternative to fossil fuels. This study assessed fermentative H 2 production from single sugars and sugar mixtures mimicking real lignocellulose hydrolysates during a continuous process. The highest hydrogen production rate (HPR) was obtained with pentose sugars, reaching 8.80 and 8.09 L/L-d for arabinose and xylose, respectively. Glucose showed the lowest HPR (6.19 L/L-d), while in the case of sugar mixtures (glucose, cellobiose, xylose, arabinose), HPR was approximately 7.3 L/L-d and was lower than expected from calculations. Thus, cofermentation reduced the H 2 production potential. It was shown that pentose sugars are preferred for hydrogen production, but their utilization is limited by the presence of glucose, which is utilized preferentially in sugar mixtures. Xylose, arabinose and sugar mixtures showed similar H 2 yields (approximately 0.29 mol H 2 /mol C), so the efficiency of substrate conversion into H 2 was comparable for these substrates. The microbial community distribution showed that the Clostridium genus was dominant regardless of the substrate, with Clostridium beijerinckii and Clostridium guangxiense being the main bacterial species. Different substrates did not significantly affect the type of microorganisms present in the microbiome and changed only their relative abundance. Objective:Sialic acid, hexoses, hexosamines and fucose are components of glycoprotein, glycolipid and/or ganglioside. These glycoconjugates are essential components of cellular membrane and receptors, which are required for normal cellular activities. The levels of these aforementioned glycans are likely to be obstructed under biological conditions (such as oxidative stress) that leads to cellular and tissue damage. Despite the efficacy of valproate as a broad-spectrum antiepileptic drug, it administration is linked to oxidative stress and multiple organ damage. Moringa oleifera leaves have been proven to be bioactive food with diverse biochemical benefits, that include antioxidant, wound healing and tissue protective effects.Methods:In this study, female Sprague-Dawley rats were grouped into four. Group 1: control group given physiological saline; Group 2: animals given only 70% ethanol Moringa leaves extract (0.3 g/kg b.w./day); Group 3: animals that received only sodium valproate (0.5 g/kg b.w./day); Group 4: animals given similar dose of sodium valproate + Moringa extract. The treatments were administered orally for 15 days, and the animals were then fasted overnight and sacrificed. Stomach tissues collected were homogenized in ice-cold normal saline, using a glass homogenizer to make up 10% w/v tissue homogenate.Results:Analysis revealed that valproate administration resulted in elevated levels of sialic acid, hexoses, hexosamine, and fucose in the stomach tissue homogenates. Conversely, the administration of Moringa extract mitigated the adverse effect of valproate on glycan levels.Conclusion:Thus, Moringa leave extract can be a good candidate for attenuating valproate-induced toxicity on stomach tissue.

Simple Summary: Glucose is the preferred carbon source for most cells. Yeast Snf1 (AMPK in mammals) is the main regulator of the response to low glucose availability. In the last years, much progress has been made in understanding the regulation of Snf1. We summarize three different mechanisms that control Snf1 activity: (1) phosphorylation and dephosphorylation of a critical threonine residue; (2) post-translational modification by the addition of a SUMO tag; (3) protonation and de-protonation of a polyhistidine tract. These mechanisms act independently of each other, allowing a flexible response to the availability of carbon sources. Snf1, the fungal homologue of mammalian AMP-dependent kinase (AMPK), is a key protein kinase coordinating the response of cells to a shortage of glucose. In fungi, the response is to activate respiratory gene expression and metabolism. The major regulation of Snf1 activity has been extensively investigated: In the absence of glucose, it becomes activated by phosphorylation of its threonine at position 210. This modification can be erased by phosphatases when glucose is restored. In the past decade, two additional independent mechanisms of Snf1 regulation have been elucidated. In response to glucose (or, surprisingly, also to DNA damage), Snf1 is SUMOylated by Mms21 at lysine 549. This inactivates Snf1 and leads to Snf1 degradation. More recently, glucose-induced proton export has been found to result in Snf1 inhibition via a polyhistidine tract (13 consecutive histidine residues) at the N-terminus of the Snf1 protein. Abstract Background Sugar content is an important indicator of fruit quality. Except for a few wild tomato species that accumulate sucrose in the fruits, most cultivated tomato species accumulate hexose. Although several studies have focused on wild sucrose-accumulating tomato, the sucrose accumulation mechanism is still unclear. Results Here, two homozygous inbred cherry tomato lines (‘TB0023’ and ‘TB0278’, which accumulated sucrose and hexose, respectively) were selected to analyze the sugar accumulation mechanism. Carbohydrate analysis, cytological observation, gene expression and enzyme activity analysis and proteomics methods were used in this study. The results indicated that glucose and fructose were absolutely dominant in the soluble sugar content of hexose-accumulating cherry tomato fruit, while sucrose and a certain proportion of hexose were the main forms of soluble sugar in sucrose-accumulating cherry tomato fruit. The phloem unloading pathway of the hexose-accumulating cherry tomato fruit switched from symplastic to apoplastic during fruit development, and the sucrose-accumulating cherry tomato probably had a mixed unloading pathway involving the symplastic and apoplastic. High activity of acid invertase (AI), sucrose phosphate synthase (SPS), sucrose synthase (SS) and sugar transporters LeSUT1, SlSWEET2a and SlSWEET12c were important factors for hexose accumulation in the hexose-accumulating cherry tomato fruit, while LeSUT2, SPS, SS, SlSWEET1b, SlSWEET5b, SlSWEET11b, SlSWEET7a, SlSWEET14 were responsible for solute sugar accumulation in the sucrose-accumulating cherry tomato. Conclusions This study provides detailed evidence for elucidation of the tomato sugar accumulation mechanism from the perspective of cell structure, physiology and molecular biology, providing a theoretical basis for the improvement of tomato quality and aiding the utilization of tomato genetic resources.

As a leaf vegetable, Gynura bicolor DC (G. bicolor) experiences a rapid deterioration after harvest including insufficient supply of sugar and destruction of cell membranes. In this research, four treatments were experimented on G. bicolor including the control (CK), 12% (g/g) sucrose (ST), 10 μL L−1 1-MCP (MT), and the combination of sucrose and 1-MCP (SMT). The results showed that three treated groups reduced respiratory rate, inhibited hexose consumption and promoted the decrease of starch and sucrose, which was converted into hexose including glucose and fructose to maintain cell membrane integrity. Meanwhile, the activities of AI, NI, SS-C, amylase, and corresponding gene expression levels were significantly up-regulated in three treated groups at 1 d, among which AI played a crucial role in regulating the accumulation of hexose. Furthermore, ST exerted a pronounced effect on hexose accumulation at the beginning while MT reduced hexose consumption through lowered respiratory metabolism during storage. Notably, SMT exhibited an optimum preservation effect on inhibited respiratory metabolism, maintaining cell membrane integrity, enhancing the retention of hexose, indicating that a synergistic effect of ST and MT were developed during storage. • Sucrose treatment promoted early retention of soluble sugar and hexose accumulation. • 1-MCP treatment kept a long-term effect on sugar scarcity by inhibiting respiration. • Combined treatment increased hexose accumulation optimally. • Enhanced hexose content moderated cell membrane damage. A benchmark of sugar produced from simple glycolaldehyde derivative has been achieved at ambient conditions (Science, 2004, 305, 1752), which is the most important step to produce life in nature. However, the mechanism has never been clearly understood. Here, we report a reaction mechanism which includes a two‐step synthesis of sugars by selective aldol reactions based on the density functional theory (DFT) calculations. Results show that the prochirality of attacking orientation of the enamine intermediate and steric effect in proton transfer is the main cause for stereochemical preference. Abstract Background Cellulose is not only a common component in vascular plants, but also has great economic benefits for paper, wood, and industrial products. In addition, its biosynthesis is highly regulated by carbohydrate metabolism and allocation in plant. MdFRK2, which encodes a key fructokinase (FRK) in apple, showed especially high affinity to fructose and regulated carbohydrate metabolism. Results It was observed that overexpression of MdFRK2 in apple decreased sucrose (Suc) and fructose (Fru) with augmented FRK activity in stems, and caused the alterations of many phenotypic traits that include increased cellulose content and an increase in thickness of the phloem region. To further investigate the involved mechanisms, we generated FRK2-OE poplar lines OE#1, OE#4 and OE#9 and discovered (1) that overexpression of MdFRK2 resulted in the huge increased cellulose level by shifting the fructose 6-phosphate or glucose 6-phsophate towards UDPG formation, (2) a direct metabolic pathway for the biosynthesis of cellulose is that increased cleavage of Suc into UDP-glucose (UDPG) for cellulose synthesis via the increased sucrose synthase (SUSY) activity and transcript levels of PtrSUSY1, (3) that the increased FRK activity increases the sink strength overall so there is more carbohydrate available to fuel increased cambial activity and that resulted in more secondary phloem. These results demonstrated that MdFRK2 overexpression would significantly changes the photosynthetic carbon flux from sucrose and hexose to UDPG for increased cellulose synthesis. Conclusions The present data indicated that MdFRK2 overexpression in apple and poplar changes the photosynthetic carbon flux from sucrose and hexose to UDPG for stem cellulose synthesis. A strategy is proposed to increase cellulose production by regulating sugar metabolism as a whole.

• Japanese Azadinium poporum produces AZA2 as dominant toxin and several AZAs. • Twelve new AZAs were discovered from Japanese Azadinium poporum. • New AZAs detected as bi-charged ions were discovered. • AZAs detected as bi-charged ions were sulfated AZAs and some had hexoses. Lipophilic marine biotoxin azaspiracids (AZAs) are produced by dinoflagellates Azadinium and Amphidoma. Recently, several strains of Azadinium poporum were isolated from Japanese coastal waters, and detailed toxin profiles of two strains (mdd421 and HM536) among them were clarified by several detection techniques on liquid chromatography-tandem mass spectrometry (LC-MS/MS) and liquid chromatography-quadrupole time of flight mass spectrometry (LC-QTOFMS). In our present study, AZA analogues in seven strains of A. poporum from Japanese coastal waters (including two previously reported strains) were determined by these detection techniques. The dominant AZA in the seven strains was AZA2 accompanied by small amounts of several known AZAs and twelve new AZA analogues. Eight of the twelve new AZA analogues discovered in our present study were detected as bi-charged ions on the positive mode LC/MS/MS. Leaf removal (LR) around the cluster zone is a common practice for regulating grape quality. The purpose of this study was to assess the effects of cluster-zone leaf removal, applied at the pea-size stage of berry development, on berry soluble sugar, organic acid and phenolic compound, sugar unloading-related gene expression of Vitis. vinifera L. Syrah. Four different severities of leaf removal were applied: no leaf removal (Control), removing 2 leaves above the cluster (LR1), removing 4 leaves above the cluster (LR2), and removing 6 leaves above the cluster (LR3). The three leaf removal treatments (LR), especially removing 4 leaves (LR2), resulted in significantly higher reducing sugar, soluble sugar (glucose, fructose and sucrose), total anthocyanin and citric acid contents as compared to the control group during ripening for both vintages. At harvest, the LR treatments increased the transcript abundance of most sugar unloading-related genes. In addition , VvHT3, VvHT5, VvSUC11, VvSUC12, VvSS and VvcwINV were positively correlated with both reducing sugar contents and soluble sugar contents. Our results suggest that removing 4 leaves above the cluster is useful for improving the quality of Syrah (Vitis vinifera L.) grapes in cool climate regions with excessive leaves. Climate change is known to simultaneously increase berry sugars but decrease anthocyanins, leading to an imbalance between sugars and anthocyanins in grape berries. To restore the balance of sugars and anthocyanins, carbon limitation by leaf removal and exogenous abscisic acid (ABA) were separately or simultaneously applied to Vitis vinifera cv. Cabernet Sauvignon fruit cuttings to decipher their effects on berry quality with metabolite and whole-genome transcriptome analyses. Carbon limitation decreased the hexose concentration and fully blocked the accumulation of anthocyanins. However, exogenous ABA increased the anthocyanin concentration under both carbon limitation and sufficient conditions. Carbon limitation and exogenous ABA induced the profound remodeling of the whole-genome transcriptome and altered the anthocyanin concentration by regulating the transcription levels of genes involved in the anthocyanin biosynthesis pathways as well as in the genes involved in various types of hormone signaling. Moreover, two pertinent candidate genes were identified based on the co-expression network analysis between the berry metabolite and transcriptome results, including a transcriptional factor, ERF2, and a calcineurin B-like protein-interacting protein kinase gene, CIPK25. In summary, simultaneously modifying the carbon supply by leaf removal and spraying exogenous ABA could re-establish the balance between sugars and anthocyanins to improve the qualities of grape berries via whole-genome transcriptome remodeling.

However, mannose has been shown to be a special type of hexose that has immune regulatory functions. In this review, we trace the discovery process of the regulatory functions of mannose and summarize some past and recent studies showing the therapeutic functions of mannose in inflammatory diseases. We conclude that treatment with mannose can suppress inflammation by inducing regulatory T cells, suppressing effector T cells and inflammatory macrophages, and increasing anti-inflammatory gut microbiome. By summarizing all the important findings, we highlight that mannose treatment is a safe and promising novel strategy to suppress inflammatory diseases, including autoimmune disease and allergic disease. The effects of the interplay of copper(II) and manganese(II) ions on growth, morphology and itaconic acid formation was investigated in a high-producing strain of Aspergillus terreus (NRRL1960), using carbon sources metabolized either mainly via glycolysis (D-glucose, D-fructose) or primarily via the pentose phosphate shunt (D-xylose, L-arabinose). Limiting Mn2+ concentration in the culture broth is indispensable to obtain high itaconic acid yields, while in the presence of higher Mn2+ concentrations yield decreases and biomass formation is favored. However, this low yield in the presence of high Mn2+ ion concentrations can be mitigated by increasing the Cu2+ concentration in the medium when D-glucose or D-fructose is the growth substrate, whereas this effect was at best modest during growth on D-xylose or L-arabinose. A. terreus displays a high tolerance to Cu2+ which decreased when Mn2+ availability became increasingly limiting. Under such conditions biomass formation on D-glucose or D-fructose could be sustained at concentrations up to 250 mg L–1 Cu2+, while on D-xylose- or L-arabinose biomass formation was completely inhibited at 100 mg L–1. The effects of the interplay of copper(II) and manganese(II) ions on growth, morphology and itaconic acid formation was investigated in a high-producing strain of Aspergillus terreus (NRRL1960), using carbon sources metabolized either mainly via glycolysis (D-glucose, D-fructose) or primarily via the pentose phosphate shunt (D-xylose, L-arabinose). Limiting Mn2+ concentration in the culture broth is indispensable to obtain high itaconic acid yields, while in the presence of higher Mn2+ concentrations yield decreases and biomass formation is favored. However, this low yield in the presence of high Mn2+ ion concentrations can be mitigated by increasing the Cu2+ concentration in the medium when D-glucose or D-fructose is the growth substrate, whereas this effect was at best modest during growth on D-xylose or L-arabinose. A. terreus displays a high tolerance to Cu2+ which decreased when Mn2+ availability became increasingly limiting. Under such conditions biomass formation on D-glucose or D-fructose could be sustained at concentrations up to 250 mg L–1 Cu2+, while on D-xylose- or L-arabinose biomass formation was completely inhibited at 100 mg L–1. High (>75%) specific molar itaconic acid yields always coincided with an "overflow-associated" morphology, characterized by small compact pellets (<250 μm diameter) and short chains of "yeast-like" cells that exhibit increased diameters relative to the elongated cells in growing filamentous hyphae. At low concentrations (≤1 mg L–1) of Cu2+ ions, manganese deficiency did not prevent filamentous growth. Mycelial- and cellular morphology progressively transformed into the typical overflow-associated one when external Cu2+ concentrations increased, irrespective of the available Mn2+. Plant roots depend on sucrose imported from leaves as the substrate for metabolism and growth. Sucrose and hexoses derived from it are also signalling molecules that modulate growth and development, but the importance for signalling of endogenous changes in sugar levels is poorly understood. We report that reduced activity of cytosolic invertase, which converts sucrose to hexoses, leads to pronounced metabolic, growth, and developmental defects in roots of Arabidopsis (Arabidopsis thaliana) seedlings. In addition to altered sugar and downstream metabolite levels, roots of cinv1 cinv2 mutants have reduced elongation rates, cell and meristem size, abnormal meristematic cell division patterns, and altered expression of thousands of genes of diverse functions. Provision of exogenous glucose to mutant roots repairs relatively few of the defects. The extensive transcriptional differences between mutant and wild-type roots have hallmarks of both high sucrose and low hexose signalling. We conclude that the mutant phenotype reflects both low carbon availability for metabolism and growth and complex sugar signals derived from elevated sucrose and depressed hexose levels in the cytosol of mutant roots. Such reciprocal changes in endogenous sucrose and hexose levels potentially provide rich information about sugar status that translates into flexible adjustments of growth and development. Glucose, fructose and mannose are the preferred carbon/energy sources for the yeast Saccharomyces cerevisiae. Absence of preferred energy sources activates glucose derepression, which is regulated by the kinase Snf1. Snf1 phosphorylates the transcriptional repressor Mig1, which results in its exit from the nucleus and subsequent derepression of genes. In contrast, Snf1 is inactive when preferred carbon sources are available, which leads to dephosphorylation of Mig1 and its translocation to the nucleus where Mig1 acts as a transcription repressor. Here we revisit the role of the three hexose kinases, Hxk1, Hxk2 and Glk1, in glucose de/repression. We demonstrate that all three sugar kinases initially affect Mig1 nuclear localization upon addition of glucose, fructose and mannose. This initial import of Mig1 into the nucleus was temporary; for continuous nucleocytoplasmic shuttling of Mig1, Hxk2 is required in the presence of glucose and mannose and in the presence of fructose Hxk2 or Hxk1 is required. Our data suggest that Mig1 import following exposure to preferred energy sources is controlled via two different pathways, where (1) the initial import is regulated by signals derived from metabolism and (2) continuous shuttling is regulated by the Hxk2 and Hxk1 proteins. Glomalin, one of the glycoproteins generated in the spores and hyphae of arbuscular mycorrhizal (AM) fungi, has multiple functions in plants and soil, while the role of foliar spray of easily extractable glomalin-related soil proteins (EE-GRSP) in citrus fruits is not well defined. Our study aimed to use referenced transcriptome sequencing to uncover the mechanism and the role of exogenous EE-GRSP in two late-ripening varieties of sweet orange (Citrus sinensis) fruits including Navel Lane Late (LW) and Rohde Red Valencia (XC). The 1804 and 1861 differentially expressed genes were identified in fruits of LW and XC, respectively, following foliar spray of EE-GRSP. Photosynthesis ranked second in the Kyoto Encyclopedia of Genes and Genomes (KEGG) metabolism in the LW variety, and carbon fixation in photosynthetic organizations ranked first in KEGG metabolism in the XC variety. The response to foliar spray of EE-GRSP affected the fruit starch and sucrose metabolism of KEGG, with 15 (10 up-regulated and 5 down-regulated) and 13 (2 up-regulated and 11 down-regulated) differentially expressed genes identified in the LW and XC variety, respectively. Cs5g19060 (sucrose phosphate synthase 4) was activated and reduced by EE-GRSP on XC and LW, respectively. Cs1g18220 (β-fructofuranosidase) and Cs2g12180 (glycosyl hydrolase family 9) genes were up-regulated and down-regulated in LW and XC, respectively. These results established the involvement of molecular signaling in response to foliar spray of EE-GRSP activating fruit sugar metabolism is dependent on citrus varieties.

The objective of this study was to examine the effect of late planting on sucrose metabolism in cotton bolls and the relationship to fiber properties and within-boll yield components. Two cotton lines A201 and A705 were employed in a sowing date experiment where two temperature regimes during boll maturation period were created at mean daily temperature of 26.8°C, 28.3°C for early planting and 25.2°C, 23.1°C for late planting in 2016 and 2017, respectively. Boll size, seed mass per boll, seed index and fiber length were increased, and lint percentage was decreased by late planting. Greater cell wall invertase activity and the resultant hexose concentration in fibers were observed in late planting, and thus led to decreased osmotic potential accounting for the enhanced fiber length. Similarly, late planting increased the maximum of vacuolar invertase activity in ovules occurring at 5 days post anthesis (DPA) and hexose concentrations in embryos from 10 DPA afterwards which may favor embryo cell division, and thus increase final seed size. Our data indicate that acid invertase and hexose are implicated in the formation of within-boll yield components and fiber properties as affected by the lower temperature regime due to late planting. Background and objectives: Based on the available literature, there are indications that the sugar composition present in rye flour, including pentoses present in arabinoxylan (AX), may have a significant impact on the relative proportions of fermentation products. The purpose of the study was to examine the changes in content of the most abundant sugars, organic acids, and alcohols caused by the addition of hexoses, pentoses, or hydrolyzed AX to rye flour at the beginning of the sourdough fermentation process. Findings: The HPLC procedures revealed that during rye sourdough fermentation, 1% of added pentoses act differently compared to hexoses. Available pentose residues in flour used for sourdough preparation caused higher acidification of dough, higher acetic acid biosynthesis, and a favorable ratio of lactic acid to acetic acid content. Conclusions: In order to provide appropriate amounts of arabinose and xylose residues in rye flour, AX could be used as a source of pentoses. However, in this case appropriate xylanase activity during sourdough fermentation must be provided. Significance and novelty: For the first time, the changes in fermentation products were investigated under standardized conditions during the rye sourdough bread production process in sourdough, dough after final fermentation, and baked bread. Carbohydrates, or in strictly chemical terms, polyhydroxy‐aldehydes or ketones represent enormous structural diversity in terms of the arrangement of atoms in space, resulting in hundreds of stereoisomers. Although the chemical properties of most stereoisomers may not be very different, their metabolic fate and utilization in biological systems is significantly different and known to influence the overall carbohydrate metabolism. This dictates the importance of understanding the structures of various stereochemical structures for a biochemist. At graduate level teaching, it is really a challenging task to correlate, connect, and recall all the structures. Conventional teaching methods and standard textbook illustrations seldom enable the learners and instructors to sustain the information for a prolonged period. Cucumber (Cucumis sativus L.), an important vegetable plant species, is susceptible to low temperature stress especially during the seedling stage. Vacuolar invertase (VI) plays important roles in plant responses to abiotic stress. However, the molecular and biochemical mechanisms of VI function in cucumber, have not yet been completely understood and VI responses to low temperature stress and it functions in cold tolerance in cucumber seedlings are also in need of exploration. The present study found that hexose accumulation in the roots of cucumber seedlings under low temperature stress is closely related to the observed enhancement of invertase activity. Our genome-wide search for the vacuolar invertase (VI) genes in cucumber identified the candidate VI-encoding gene CsVI1. Expression profiling of CsVI1 showed that it was mainly expressed in the young roots of cucumber seedlings. In addition, transcriptional analysis indicated that CsVI1 expression could respond to low temperature stress. Recombinant CsVI1 proteins purified from Pichia pastoris and Nicotiana benthamiana leaves could hydrolyze sucrose into hexoses. Further, overexpression of CsVI1 in cucumber plants could increase their hexose contents and improve their low temperature tolerance. Lastly, a putative cucumber invertase inhibitor was found could form a complex with CsVI1. In summary, these results confirmed that CsVI1 functions as an acid invertase involved in hexose accumulation and responds to low temperature stress in cucumber seedlings.

Abstract Background Little is known about how pre-diagnostic metabolites in blood relate to risk of prostate cancer. We aimed to investigate the prospective association between plasma metabolite concentrations and risk of prostate cancer overall, and by time to diagnosis and tumour characteristics, and risk of death from prostate cancer. Methods In a case-control study nested in the European Prospective Investigation into Cancer and Nutrition, pre-diagnostic plasma concentrations of 122 metabolites (including acylcarnitines, amino acids, biogenic amines, glycerophospholipids, hexose and sphingolipids) were measured using targeted mass spectrometry (AbsoluteIDQ p180 Kit) and compared between 1077 prostate cancer cases and 1077 matched controls. Risk of prostate cancer associated with metabolite concentrations was estimated by multi-variable conditional logistic regression, and multiple testing was accounted for by using a false discovery rate controlling procedure. The epidemiological studies have shown that antibiotic treatment increases the susceptibility to inflammatory bowel disease development. The disturbance of mucus layer integrity might be one of the possible mechanisms. The aim of the present study was to investigate the effect of antibiotic ceftriaxone treatment on glycoproteins level and its carbohydrate composition in surface mucus layer of rat intestine. The study was done on male Wistar rats (140-160 g). Ceftriaxone (300 mg/kg, i.m.) was administered once a day for 14 days. The surface mucus from terminal ileum and colon were collected on the 15th, 29th and 72nd days of the experiment. Total level of mucus glycoproteins, hexoses, hexosamines, fucose and sialic acids were measured. Ceftriaxone administration did not affect the levels of glycoproteins in rat ileum. In the colon, the levels of glycoprotein were 1.3-fold decreased (Р < 0.05) on the 72nd day of the experiment. These changes were accompanied by the 1.2-fold decrease of hexoses (Р < 0.05) and 3.1-fold (Р < 0.05) decrease of fucose level and 1.5-fold (Р < 0.05) increase of the levels of sialic acids in the surface mucus of the rat colon. Thus, ceftriaxone administration induces the long-term changes in the levels of glycoproteins and carbohydrates composition in the rat colon surface mucus. This could potentially explain the susceptibility to inflammatory bowel disea­ses development.

For a comprehensive understanding of gene expression, enzyme activity and sugar concentrations in response to short-term water deficit in apple (Greensleeves), sugar-modulated gene expression and enzyme activities were analyzed. Water stress resulted in the accumulation of sorbitol, glucose, fructose, galactose and starch, accompanied by a significant reduction in photosynthesis and sucrose concentration. In response to short-term water deficits, the activities of aldose-6-phosphate reductase (A6PR; EC 1.1.1.200), sorbitol dehydrogenase (SDH; EC 1.1.1.14), neutral invertase (NINV; EC 3.2.1.26), sucrose synthase (SUSY; EC 2.4.1.13), and fructokinase (FK; EC 2.7.1.4) were higher, whereas cell wall invertase (CWINV; EC 3.2.1.26) and hexokinase (HK; EC 2.7.1.1) activities were lower. In addition, sucrose phosphate synthase (SPS; EC 2.4.1.14) activity increased during the initial stages of dehydration and then decreased as the drought strengthened. Transcript levels of MdA6PR , MdSDH1/2 , MdNINV1/2 , MdSUSY3 , MdFK1/2/4 , MdSOT1/2 , MdSUC1-3 , MdTMT2/3 , MdvGT1 , MdpGlcT1-4 were upregulated, whereas transcript levels of MdCWINV1/2 , MdHK1/2/3/5 , and MdTMT1 were downregulated after 6 days of water stress. We examined the hypothesis that maternal nutrition and day of gestation would affect the concentrations of AAs and hexoses in bovine utero-placental fluids and maternal serum from days 16 to 50 of gestation. Forty-nine cross-bred Angus heifers were bred via artificial insemination and fed a control diet (CON = 100% of requirements for growth) or a restricted diet (RES = 60% of CON) and ovariohysterectomized on days 16, 34, or 50 of gestation; nonpregnant controls were not bred and ovariohysterectomized on day 16 of the synchronized estrous cycle. The resulting design was a completely randomized design with a 2 × 3 factorial + 1 arrangement of treatments. Maternal serum, histotroph, allantoic fluid, and amniotic fluid were collected at time of ovariohysterectomy. Samples were then analyzed for concentrations of AAs and intermediary metabolites: alanine (Ala), arginine, asparagine (Asn), aspartate (Asp), citrulline, cysteine, glutamine, glutamate (Glu), glycine (Gly), histidine, isoleucine, leucine (Leu), lysine, methionine (Met), ornithine, phenylalanine (Phe), proline (Pro), serine (Ser), threonine (Thr), tryptophan, tyrosine (Tyr), and valine (Val). The concentrations of Gly, Ser, and Thr in maternal serum were greater (P ≤ 0.05) in CON compared with RES. Furthermore, day of gestation affected (P ≤ 0.05) concentrations of Asn, Glu, Phe, Thr, and Tyr in maternal serum. Status of maternal nutrition affected the Asp concentration of histotroph where RES was greater (P = 0.02) than CON. In histotroph, Ala, Leu, Met, and Val concentrations were greater (P ≤ 0.05) on day 50 compared with day 16. Additionally, Glu and Pro concentrations in histotroph were greater (P < 0.01) on days 34 and 50 compared with day 16. A day × treatment interaction was observed for the concentration of Val in allantoic fluid where day 34 CON was greater (P = 0.05) than all other days and nutritional treatments. In addition, the concentration of Gln in amniotic fluid experienced a day × treatment interaction where day 34 RES was greater (P ≤ 0.05) than day 34 CON, which was greater (P ≤ 0.05) than day 50 CON and RES. These data support our hypothesis that day of gestation and maternal nutrition affect the concentrations of various neutral and acidic AA in beef heifer utero-placental fluids and maternal serum from days 16 to 50 of gestation. A mathematical description was developed for production of saccharides and fermentation inhibitors during the hot-compressed water pretreatment of cassava residue. Pretreatment was conducted at 150 °C, 160 °C, 170 °C, and 180 °C, and reaction times ranged from 0 to 70 min. The formation of saccharides and four main inhibitors (furfural (F), hydroxymethylfurfural (HMF), acetic acid, and formic acid) were studied. A model for predicting the concentrations of F and HMF (CF and CHMF, respectively) as functions of H+ concentration was established. Furthermore, kinetic models were built after introducing the hydrogen ion concentration index mi. Hydrogen ion concentration had a dramatic effect on the dissolution of pentosan but did not greatly affect the dissolution of hexosan or the degradation of hexose or pentose. Additionally, the activation energies for the formation of pentose or hexose were lower than the degradation energies. The coefficients of determination (R2) of the kinetic models for predicting the yield of the four compounds (pentose, hexose, furfural, and HMF) were higher than 0.923. These kinetic models provided a theoretical foundation and technical support for controlling the production of the main carbohydrates and fermentation inhibitors.

Recent advances in genetic engineering capabilities have enabled the development of oleochemical producing strains of Yarrowia lipolytica. Much of the metabolic engineering effort has focused on pathway engineering of the product using glucose as the feedstock; however, alternative substrates, including various other hexose and pentose sugars, glycerol, lipids, acetate, and less-refined carbon feedstocks, have not received the same attention. In this review, we discuss recent work leading to better utilization of alternative substrates. This review aims to provide a comprehensive understanding of the current state of knowledge for alternative substrate utilization, suggest potential pathways identified through homology in the absence of prior characterization, discuss recent work that either identifies, endogenous or cryptic metabolism, and describe metabolic engineering to improve alternative substrate utilization. Finally, we describe the critical questions and challenges that remain for engineering Y. lipolytica for better alternative substrate utilization.

In cattle, the detection of very early endometrial responses is considered to be hampered by the presence of only a single embryo. Therefore, we have previously developed a model of multiple embryo transfer to circumvent this hindrance. In this work, we analysed embryo–maternal interactions in the bovine uterus on day 8 of development while comparing the presence of multiple v. single embryos using embryo transfer and artificial insemination, respectively. Concentration of proteins (β-actin, NFkB, clusterin and immunoproteosome 20S β5i subunit–i20S), by western blot, and hexoses (glucose and fructose) were measured in paired samples of uterine fluid (UF) from the same animal with and without embryos in the uterus and were compared with UF obtained after artificial insemination. Prostaglandin (PG) F2α and PGE2 concentrations were also analysed in blood plasma. The four proteins analysed and hexoses were unaffected by the presence of one or more embryos in the uterus. However, blood PGF2α showed similar, significant increases with one or more embryos over cyclic animals; such changes were not observed in blood PGE2. Although multiple embryo transfer may appear to be non-physiological, we showed that the uterus, at the very early embryonic stages, does exhibit physiological reactions. Multiple embryo transfer can, therefore, be used for studies of very early embryo–maternal interactions in vivo in monotocous species.

Rapid, direct, on-site and noninvasive food analysis is strongly needed for quality control of food. To satisfy this demand, the technique of dipping probe electrospray ionization/mass spectrometry (dPESI/MS) was developed. The sample surface was pricked with a fine acupuncture needle and a sample of ∼200 pL was captured at the needle tip. After drying the sample, the needle tip was dipped into the solvent for ∼50 ms and was moved upward. A high-voltage was applied to the needle to generate electrospray when the needle reached the highest position, and mass spectra were measured with a time-of-flight mass spectrometer. For evaluation of the method, the technique was used to analyze foods such as vegetables, salmon flesh, cow’s milk, yogurt, and soy-bean milk. The detected major ions for cow’s milk and yogurt were [(Lac) n  + Ca] 2+ with n = 1–6 (where (Lac) is lactose), indicating that Ca 2+ is tightly bound by Lac molecules. [ABSTRACT FROM AUTHOR]

Premise of research. The armored scale Aulacaspis yasumatsui Takagi has invaded numerous geographic regions to devastate cultivated populations of Cycas L. and the native population of Cycas micronesica K.D. Hill on the islands of Guam and Rota. Questions remain as to how this small hemipteran insect can so rapidly kill a large cycad tree. One possibility is depletion of nonstructural carbohydrates (NSCs) until the storage pool can no longer sustain plant viability.Methodology. We infested Cycas revoluta Thunberg plants with A. yasumatsui and harvested whole plants periodically until plant mortality at week 60. Harvested plants were separated into leaves, stems, and roots, and tissue was frozen and then lyophilized. The dry mass of each organ was measured, and then fructose, glucose, maltose, starch, and sucrose were quantified. Concentration and total pool size within each organ were calculated for each NSC.Pivotal results. Concentration and total pool of all five NSCs were greatest in stems. Concentrations exhibited linear or quadratic declines with time of infestation for all NSCs and organs, with the exception of glucose in stems and maltose in leaves. The total pool of all five NSCs declined linearly or quadratically in all three organs, with the exception of maltose in leaves. Initial plant dry mass of 152 g was 23% NSC, and ending plant dry mass of 59 g was 26% NSC. Carbohydrate stoichiometry shifted in favor of sugars over starch, and sugar stoichiometry shifted in favor of hexoses over disaccharides as time of infestation progressed.Conclusions. These results are consistent with carbohydrate depletion as the mechanism that explains rapid Cycas tree mortality during chronic A. yasumatsui infestation. The conventional UV/Vis spectroscopy methods recommended by the European Pharmacopoeia (EP) for determining hexosamine, hexonic acid and methylpentose in pneumococcal polysaccharide vaccine (PPSV) hydrolates are time-consuming due to derivatization process (typically, an analysis cycle is more than 4 h) and improvements of selectivity and precision of the methods are in demand. In this study, a new approach based on hydrophilic interaction liquid chromatography and triple quadrupole mass spectrometry (HILIC-MS/MS) was optimized to overcome the drawbacks of the EP methods for simultaneous determination of methylpentose, hexose, hexosamine and hexonic acid in PPSV hydrolysates. The chromatographic, MS and sample hydrolysis conditions were systematically investigated. A zwitterionic column, Click Cys, using a gradient elution with a mobile phase of 10 mM ammonium formate (pH 4.3) in acetonitrile from 72% to 21% in 6 min was applied for separating the targets, which exhibited low column bleeding, easy equilibration and long-term stability. The HILIC-MS/MS method showed a high sensitivity (LOD = 0.98 μg L −1 for hexonic acid), a good repeatability (RSD of peak area less than 1.669%), accuracy (92.9%–104.2%), recovery (97.6%–99.3%) and a wide linear range. The RSD of retention time obtained from more than 3000 injections in three months was less than 1.64%. The new method was compared with the EP method for determining hexosamine in 23 serotypes of PPSV hydrolysates. The results indicated that the new HILIC-MS/MS method was highly selective, accurate, stable and extremely fast due to without need of derivatization, as compared to the conventional EP methods. Combined RB was provided 16 h per day (24 h) in all the six treatments, among which, continuous RB with only one light/dark (L/D) cycle of 16 h/8 h in treatment L/D(1) was regarded as control. Five intermittent RB treatments that were respectively L/D(2) with two L/D cycles of 8 h/4 h over a 24-h period, L/D(3) with three L/D cycles of 6 h/3 h or 4 h/2 h, L/D(4) with four L/D cycles of 4 h/2 h, L/D(6) with six L/D cycles of 3 h/1.5 h or 1 h/0.5 h, and L/D(8) with eight L/D cycles of 2 h/1 h were set up. Results indicated that shoot biomass of lettuce was increased by intermittent RB treatment L/D(2), L/D(4), L/D(6) and L/D(8) to different extents, while decreased significantly by L/D(3) compared with the control. Significantly higher hexose/sucrose ratio (in comparison with any other treatment) accompanied with the strongest combined activities of sucrose degrading enzymes as well as the weakest combined activities of sucrose synthesizing enzymes was observed in plants treated with L/D(2) treatment. Significantly higher content of soluble sugar especially fructose as well as lower contents of crude fiber and starch were observed in lettuce exposed to L/D(2) and L/D(3) compared with any other treatment. All intermittent treatments significantly enhanced the glucose content in lettuce compared with the control. Both L/D(2) and L/D(3) treatments promoted lettuce taste via improving the sweetness and crispness, whereas only L/D(2) simultaneously increased the shoot biomass (in terms of dry weight) compared with the control. Context:Olive oil is the major source of tyrosol which is a natural phenolic antioxidant. Olive oil constitutes a major component of the Mediterranean diet that is linked to a reduced incidence of chronic diseases. Objective:This study evaluates the effects of tyrosol on altered glycoprotein components in streptozotocin-induced diabetic rats. Materials and methods:Diabetes mellitus was induced in male Wistar rats by streptozotocin (40 mg/kg body weight). These rats were administered tyrosol (20 mg/kg body weight) and glibenclamide (600 μg/kg body weight) orally daily for 45 days. Plasma glucose, plasma insulin, glycoprotein components such as hexose, hexosamine, sialic acid and fucose in the plasma, liver and kidney, and histopathogy of tissues were analyzed. Results:Diabetic rats revealed significant (p < 0.05) increase in the levels of glucose, hexose, hexosamine, sialic acid and fucose (277.17, 152.45, 100.43, 79.69 and 49.29 mg/dL) in the plasma; decrease in the levels of palsma insulin (6.12 μU/mL) and sialic acid (4.36 and 5.03 mg/g) in the liver and kidney; significant (p < 0.05) increase in hexose (49.33 and 46.82 mg/g), hexosamine (22.68 and 33.20 mg/g) and fucose (31.63 and 32.44 mg/g) in the liver and kidney. Further, periodic acid-Schiff staining of tissues revealed positive-stain accumulation in diabetic rats. Tyrosol treatment showed significant (p < 0.05) effects on all the biochemical parameters and histopathology studied in streptozotocin- nduced diabetic rats. Also, thein vitrostudy revealed the antioxidant effect of tyrosol. Discussion and conclusions:Thus, tyrosol protects streptozotocin-induced diabetic rats from the altered glycoprotein components. The plant SWEET family is a sugar transporter family that plays a significant role in plant development. Here, seven loquat SWEET family members were identified by RNA-seq. These were designated as EjSWEET1, EjSWEET2a, EjSWEET2b, EjSWEET2c, EjSWEET4, EjSWEET15, and EjSWEET17. Phylogenetic and predictive functional annotation analyses suggest that the loquat SWEETs are classified as having sucrose, glucose and fructose transportation features. The in vivo responses of loquat SWEETs to exogenous sugar or NaCl was investigated by applying high concentrations of sugar or salt to 7-month-old loquat seedlings cultured in a nutrient medium. The results showed that most loquat SWEET genes can respond to exogenous applications of sucrose, glucose, fructose and salt. The response of EjSWEET1 to exogenous fructose was faster than the others, indicating that EjSWEET1 is more sensitive to exogenous fructose compared with other loquat SWEETs. EjSWEET15 can be induced by sucrose, but is suppressed by glucose. This indicates its possible role in sucrose transporting. The response of loquat SWEETs to NaCl showed broadly similar patterns compared to sugars. However, after a longer time of NaCl treatment, most loquat SWEETs are upregulated, especially EjSWEET15. D-tagatose is a naturally existing rare monosaccharide having prebiotic properties. Minimal absorption, low metabolizing energy, and unique clinical properties are the characteristics of D-tagatose. D-tagatose gained international attention by matching the purpose of alternate sweeteners that is much needed for the control of diabetes among world population. Recent efforts in understanding tagatose bioconversion have generated essential information regarding its production and application. This article reviews the evolution of D-tagatose as an important rare sugar by appreciable improvements in production results and its significant applications resulted of its unique physical, chemical, biological, and clinical properties thus considering it an appropriate product for requisite improvements in technical viability.
