The purpose of this study was to investigate the effect that Glycine max hydrolyzed with enzymes from Bacillus velezensis KMU01 has on dextran-sulfate-sodium (DSS)-induced colitis in mice. Hydrolysis improves functional health through the bioconversion of raw materials and increase in intestinal absorption rate and antioxidants. Therefore, G. max was hydrolyzed in this study using a food-derived microorganism, and its anti-inflammatory effect was observed. Enzymatically hydrolyzed G. max (EHG) was orally administered once daily for four weeks before DSS treatment. Colitis was induced in mice through the consumption of 5% (w/v) DSS in drinking water for eight days. The results showed that EHG treatment significantly alleviated DSS-induced body weight loss and decreased the disease activity index and colon length. In addition, EHG markedly reduced tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production, and increased that of IL-10. EHG improved DSS-induced histological changes and intestinal epithelial barrier integrity in mice. Moreover, we found that the abundance of 15 microorganisms changed significantly; that of Proteobacteria and Escherichia coli, which are upregulated in patients with Crohn’s disease and ulcerative colitis, decreased after EHG treatment. These results suggest that EHG has a protective effect against DSS-induced colitis and is a potential candidate for colitis treatment.

Background Moju is a traditional rice beverage local to Jeonju with an alcohol content of 1–2%. Moju is made by boiling makgeolli with several kinds of medicinal herbs, such as ginger, jujube and cinnamon. The raw materials used in Moju are well known for their physiological and functional effects. Although Moju is made with functional raw materials, the operational role of Moju has not yet been reported. Objectives The aim of this study was to identify the anti-melanogenic effects of Moju in B16F10 melanoma cells and explore the potential mechanisms. Results In this study, we investigated the antioxidant activity and anti-melanogenic effect of Moju. Moju showed no toxicity to HEK293T or B16F10 cells. The antioxidant activity of Moju was confirmed by its ability to increase radical scavenging activity. Moju decreased tyrosinase activity in a concentration-dependent manner. At the cellular level, Moju reduced melanin synthesis and the expression of proteins involved in melanin synthesis at concentrations of 100, 250, and 500 μg/mL in B16F10 cells. In addition, Moju inhibited the phosphorylation of extracellular signal-regulated kinase (ERK). Conclusions These results provide evidence that Moju has antioxidant activity and anti-melanogenic effect that occur through regulation of the ERK pathway. Although further research is needed to elucidate the specific mechanism and functional components, the ability of Moju to inhibit melanin synthesis by altering tyrosinase activation suggest that it can be used as a functional whitening ingredient.

Background Momordica charantia is a member of the Cucurbitaceae family and has traditionally been used for medical nutritional therapy to cure diabetes, and its various biological properties have been reported. However, several studies have demonstrated that M. charantia may exert toxic or adverse effects under different conditions. In this study, we prepared an M. charantia extract using ultrasound-assisted extraction, which is a green technology, and verified its anti-inflammatory effects. Objectives The aim of this study was to investigate the anti-inflammatory effects of M. charantia extract using ultrasound-assisted extraction in LPS-induced Raw264.7 macrophages and explore the potential mechanism mediated by the MAPK/NF-κB signaling pathway. Results We found that the M. charantia extract was non-toxic up to a concentration of 500 μg/mL in Raw264.7 cells. We verified that treatment with M. charantia extract significantly reduced the production of nitric oxide and proinflammatory cytokines, including TNF-α, IL-1β, IL-2, and IL-6, in LPS-stimulated RAW264.7 cells. Moreover, the anti-inflammatory cytokine IL-10 was dramatically increased by treatment with the M. charantia extract. In addition, the phosphorylation of the transcription factor NF-κB, which modulates the production of inflammatory proteins, including JNK, ERK, and p38, was reduced by downregulation of the MAPK signaling pathway. Conclusion These results indicate that the M. charantia extract collected using an industrial ultrasonic system is non-toxic and has an anti-inflammatory effect through regulation of the NF-κB and MAPK pathways, suggesting that it can act as a therapeutic candidate for the treatment of inflammatory diseases.

Background: Amomum villosum Lour., (Zingiberaceae) an herbaceous plant in the ginger family, has been used to treat various diseases. In a single-blind, randomized, crossover study, we assessed the postprandial blood insulin and blood glucose responses in healthy subjects (n = 40) after the Amomum villosum water extract (AVE) (5 g/person) or a placebo (5 g/person) consumption. Methods: During each treatment course, the healthy subject consumed a regular late afternoon meal, followed by fasting for 12 h, and arrived at the clinical study center the next morning. Blood insulin and blood glucose levels were assessed at 0, 30, 60, 90, and 120 min after AVE consumption. Between each treatment, the subjects accomplished one week of a washout period. Results: The AVE intake demonstrated a significant (67.26%) decline in postprandial blood glucose AUC0–120 min (incremental area under the curve from 0 to 120 min) versus the placebo (P = 0.011). Apium graveolens (celery) of the family Apiaceae contains bioactive compounds including luteolin and apigenin. The purpose of this study was to increase the extraction yield of apigenin and luteolin in celery extract using green technology and to evaluate their biological activities. The results showed that CA and β-glucosidase-assisted celery extraction transformed apiin in the celery to apigenin with an increase in luteolin concentration. The CA and β-glucosidase-treated celery extract (CAGE) improved the anti-inflammatory properties of celery extract by inhibiting the expression and production of inflammatory cytokines (IL-6, IL-8, IL-31, and TNF-α) in IL-33-stimulated mast cells (HMC-1.2 cells). Their mechanism of action was tied to the inhibition of ERK, JNK, IKKα, IκBα, and NF-κB activation by CAGE in the stimulated cells. In conclusion, CA and enzyme treatment can be considered as a useful biotechnology tool for the improvement of bioactive compounds in celery and hence improve on their bioactivity. PRACTICAL APPLICATIONS: Apium graveolens commonly called celery is an edible agricultural product cultivated throughout the world and known as a "superfood." Celery contains bioactive compounds including apigenin and luteolin that contribute to their described biological activities. However, extracting celery using normal extraction procedures such as hot water and ethanol methods yields only a small amount of apigenin and luteolin. In the present study, we introduced an eco-friendly method using citric and β-glucosidase to obtain apigenin and luteolin-rich celery extract with improved anti-inflammatory activities. The present work will spark studies on the conversion of less biologically active compounds in functional food materials to more active compounds using CA and β-glucosidase, and the development of functional food with specifically enriched bioactive substances at the industrial levels.

Schisandra chinensis (Turcz.) Baill (SC) and Lycium chinense Mill. (LC) are widely distributed in Asia, where the fruit has traditionally been used for medicinal herbs. We previously reported that the roasting process improved the antioxidant and their hangover relieving effects. In this study, we assessed the antioxidant and anti-inflammatory effects of water extract of SC, LC, and a mass ratio 1 : 1 mixture (SL), after roasting in RAW264.7 macrophage cells stimulated with lipopolysaccharide (LPS). Roasted SL (RSL) extracts showed greater enhancement potential than the others, based on the inhibition of NO (nitric oxide) and intracellular reactive oxygen species (ROS) production in RAW264.7 cells. RSL also significantly decreased the proinflammatory markers (e.g., iNOS, COX-2, TNF-α, and IL-1β) and NAD(P)H oxidase (NOX) signaling proteins (i.e., NOX (−1, −2, and −4), p22phox, p47phox, and p67phox). The inflammatory cytokine, tumor necrosis factor-alpha, interferon-1 beta levels, NF-kB, and mitogen-activated kinase activations were also significantly inhibited by RSL treatment. Based on the results of cellular levels, we compared the promotion effects of RSL extract on liver injury mediated by alcohol-induced inflammation and oxidative stress in mice. Mice were fed a Lieber–DeCarli regular liquid alcohol diet with or without SL and RSL extracts for six weeks. Alcohol intake caused liver injury, evidenced by an increase in serum alanine aminotransferase and aspartate aminotransferase activities. Consistent with the results in cell levels, RSL treatment remarkably downregulated ROS and inflammatory factors, as well as their signaling molecules, in serum and tissues. These results suggest that the roasting of SC and LC could potentially elevate the inhibition effect on alcohol-induced inflammation and oxidative stress and consequently prevent alcoholic liver damage. Also, the combination of SC and LC may provide a more synergistic effect than either alone.

This study investigates the prediction skill of sub-seasonal prediction models that vary based on the choice of two dynamical cores: the finite volume (FV) dynamical core on a latitude-longitude grid system and the spectral element (SE) dynamical core on a cubed-sphere grid system. Recent research showed that the SE dynamical core on a uniform grid system increases parallel scalability and removes the need for polar filters for mitigating uncertainty in climate prediction, particularly for the Arctic region. However, it still remains questionable whether the choice of dynamical cores can actually yield significant changes in prediction skill. To tackle this issue, we implemented a sub-seasonal prediction model based on the Community Atmospheric Model version 5 by incorporating the above two dynamical cores with virtually the same physics schemes. Sub-seasonal prediction skills of the SE dynamical core and FV dynamical core are verified with ERA-Interim reanalysis during the early winter (November–December) and the late winter (January–February) from 2001/2002 to 2017/2018. The prediction skills of two different dynamical cores were significantly different regardless of the similar physics scheme. In the ocean, the predictability of the SE dynamical core is similar to that of the FV dynamical core, mostly because our simulation configuration imposes the same boundary and initial conditions at the surface. Notable differences in the one-month predictability between the two cores are observed for the wintertime Arctic and mid-latitudes, particularly over North America and Eurasia continents. With a one-month lead, the SE dynamical core exhibited higher predictability over North America in late winter (r ≈ 0.45 in SE, r ≈ 0.10 in FV) whereas the FV dynamical core showed relatively higher predictability in East Asia and Eurasia in early winter (r ≈ 0.15 in SE, r ≈ 0.43 in FV). Therefore, we conclude that caution is needed when selecting the dynamical cores of sub-seasonal prediction models. Partially, these differences can be ascribed to the different manifestations of Arctic-mid-latitude linkage in the two dynamical cores; the SE dynamical core captures warmer Arctic and colder mid-latitudes relatively better than the FV dynamical core.

Recently, many studies have highlighted the importance of the ability to predict the Arctic sea ice concentration in the sub-seasonal time scales. Notably, the Arctic sea ice concentration has a potential for skillful predictions through their long-term trend memory. Based on the long-term memory of Arctic sea ice concentration, we evaluate the predictability of Arctic sea ice concentration (SIC) by applying a time-series analysis technique of the Prophet model on sub-seasonal time scales. A Prophet is a recently introduced method as a statistical approach inspired by the nature of time series forecasted at Facebook and has not been applied to the prediction of Arctic SIC before. Sub-seasonal prediction skills of Arctic SIC in the Prophet model were compared with the NCEP Climate Forecast System Reforecast (CFS-Reforecast) model as a dynamical approach and verified with the satellite observation during wintertime from 2000 to 2018 for 1 to 8 weeks lead times. The result shows that the Prophet model exhibits much better skill than the NCEP CFS-Reforecast model in the climatology prediction except for the 1 to 3 weeks lead times, as the Prophet model has mainly the ability to capture the long-term trend. In the anomaly prediction, however, the NCEP CFS-Reforecast model is superior to the Prophet model in the prediction of sub-seasonal time scales, as the NCEP CFS-Reforecast captures more effectively the sub-seasonal transition of the underlying dynamical system. Therefore, even if the Prophet model has shown a useful skill in predicting the climatological Arctic SIC, there is still a need to improve the accuracy and robustness of the predictions in an anomalous Arctic SIC. Further, we suggest that the bias correction method is needed to improve the forecast skill of Arctic SIC using the time-series analysis technique, and it will be critical to advance the field of the Arctic SIC forecasting on the sub-seasonal time scales.
