The interleukin-(IL-)17 family of cytokines is composed of six members named IL-17A, IL-17B, IL-17C, IL-17D, IL-17E, and IL-17F. IL-17A is the prototype of this family, and it was the first to be discovered and targeted in the clinic. IL-17A is essential for modulating the interplay between commensal microbes and epithelial cells at our borders (i.e., skin and mucosae), and yet, for protecting us from microbial invaders, thus preserving mucosal and skin integrity. Interactions between the microbiota and cells producing IL-17A have also been implicated in the pathogenesis of immune mediated inflammatory diseases and cancer. While interactions between microbiota and IL-17B-to-F have only partially been investigated, they are by no means less relevant. The cellular source of IL-17B-to-F, their main targets, and their function in homeostasis and disease distinguish IL-17B-to-F from IL-17A. Here, we intentionally overlook IL-17A, and we focus instead on the role of the other cytokines of the IL-17 family in the interplay between microbiota and epithelial cells that may contribute to cancer pathogenesis and immune surveillance. We also underscore differences and similarities between IL-17A and IL-17B-to-F in the microbiota-immunity-cancer axis, and we highlight therapeutic strategies that directly or indirectly target IL-17 cytokines in diseases.

T-cell-based immunotherapy strategies have profoundly improved the clinical management of several solid tumors and hematological malignancies. A recently developed and promising immunotherapy approach is to redirect polyclonal MHC-unrestricted T lymphocytes toward cancer cells by bispecific antibodies (bsAbs) that engage the CD3 complex and a tumor-associated antigen (TAA). The TNF-related apoptosis-inducing ligand receptor 2 (TRAIL-R2) is an attractive immunotherapy target, frequently expressed by neoplastic cells, that we decided to exploit as a TAA. We found that a TRAIL-R2xCD3 bsAb efficiently activates T cells and specifically redirect their cytotoxicity against cancer cells of different origins in vitro, thereby demonstrating its potential as a pan-carcinoma reagent. Moreover, to mimic in vivo conditions, we assessed its ability to retarget T-cell activity in an ex vivo model of ovarian cancer patients' ascitic fluids containing both effector and target cells—albeit with a suboptimal effector-to-target ratio—with remarkable results.

Immunotherapy has become the standard-of-care in many solid tumors. Despite the significant recent achievements in the diagnosis and treatment of cancer, several issues related to patients’ selection for immunotherapy remain unsolved. Multiple lines of evidence suggest that, in this setting, the vision of a single biomarker is somewhat naïve and imprecise, given that immunotherapy does not follow the rules that we have experienced in the past for targeted therapies. On the other hand, additional immune-related biomarkers that are reliable in real-life clinical practice remain to be identified. Recently, the immune-checkpoint blockade has been approved in the US irrespective of the tumor site of origin. Further histology-agnostic approvals, coupled with with tumor-specific companion diagnostics and guidelines, are expected in this field. In addition, immune-related biomarkers can also have a significant prognostic value. In this review, we provide an overview of the role of these biomarkers and their characterization in the management of lung cancer, melanoma, colorectal cancer, gastric cancer, head and neck cancer, renal cell carcinoma, urothelial cancers, and breast cancer.

Abstract In recent years, the exploration of the therapeutic potential of Salvia has gained considerable attention, leading to a growing number of scientific studies emphasizing its pharmacological properties. Despite this, therapeutic applications of Salvia remain underexploited, requiring further investigation. Iran is a major center for sage diversity in Asia, boasting 60 Salvia species, 17 of which are unique to the area. This study aimed to comprehensively explore and compare the extracts of 102 Salvia samples belonging to 20 distinct Salvia species from Iran, providing a deeper understanding of their specific polyphenol content and, consequently, their antioxidant capabilities and potential therapeutic uses. All samples were analyzed to determine the contents of total phenolics, total flavonoids, total tannin, photosynthetic pigments, and ascorbic acid, along with their antioxidant activity. These data were then combined with the forty distinct chemical fingerprints identified by ultrafast high-pressure liquid chromatography coupled with high-resolution mass spectrometry. Multivariate data analysis was employed to find correlations and differences among the huge number of data obtained and to identify Salvia species with similar phytochemical and/or antioxidant properties. The results show that each Salvia species is characterized by a distinct class of polyphenols recognized for their antidiabetic, anti-inflammatory, cardioprotective and neuroprotective properties. Overall, our findings reveal the potential of some Salvia species for targeted therapeutic applications and provide a rational basis for the development of Salvia-derived nutraceuticals, ultimately improving the prospects for the use of Salvia in medicine.

Abstract Introduction Genome sequencing has utility, however, it may reveal secondary findings. While Western bioethicists have been occupied with managing secondary findings, specialists’ attention in the Arabic countries has not yet been captured. We aim to explore the attitude of the United Arab Emirates (UAE) population toward secondary findings. Method We conducted a cross-sectional study between July and December 2022. The validated questionnaire was administered in English. The questionnaire consists of six sections addressing topics such as demographics, reactions to hypothetical genetic test results, disclosure of mutations to family members, willingness to seek genetic testing, and attitudes toward consanguinity. Chi-squared and Fisher’s exact tests were used to investigate associations between categorical variables. Results We had 343 participants of which the majority were female (67%). About four-fifths (82%) were willing to know the secondary findings, whether the condition has treatment or not. The most likely action to take among the participants was to know the secondary findings, so they can make life choices (61%). Conclusion These results can construct the framework of the bioethics of disclosing secondary findings in the Arab regions.

Lutein (LT) is a naturally occurring xanthophyll carotenoid mainly concentrated in the central nervous system (CNS), but its neurotrophic role is still debated. We investigated whether LT cord blood concentrations correlate with S100B in a cohort of preterm and term healthy newborns. The success of Space missions and the efficacy of colonizing extraterrestrial environments depends on ensuring adequate nutrition for astronauts and autonomy from terrestrial resources. A balanced diet incorporating premium quality fresh foods, such as microgreens, is essential to the mental and physical well-being of mission crews. To improve the nutritional intake of astronaut meals, two levels of potassium iodide (KI; 4 µM and 8 µM) and an untreated control were assessed for iodine (I) biofortification, and overall nutraceutical profile of four microgreens: tatsoi (Brassica rapa L. subsp. narinosa), coriander (Coriandrum sativum L.), green basil, and purple basil (Ocimum basilicum L.). A dose-dependent increase in I was observed at 8 µM for all species, reaching concentrations of 200.73, 118.17, 93.97, and 82.70 mg kg−1 of dry weight, in tatsoi, coriander, purple basil, and green basil, respectively. Across species, I biofortification slightly reduced fresh yield (–7.98%) while increasing the antioxidant activity (ABTS, FRAP, and DPPH). LC–MS/MS Q extractive orbitrap analysis detected 10 phenolic acids and 23 flavonoids among microgreen species. The total concentration of phenolic acids increased (+28.5%) in purple basil at 8 µM KI, while total flavonoids in coriander increased by 23.22% and 34.46% in response to 4 and 8 µM KI, respectively. Both doses of KI increased the concentration of total polyphenols in all species by an average of 17.45%, compared to the control.

Ocimum basilicum L. is an aromatic plant rich in bioactive metabolites beneficial to human health. The agronomic biofortification of basil with Zn could provide a practical and sustainable solution to address Zn deficiency in humans. Our research appraised the effects of biofortification implemented through nutrient solutions of different Zn concentration (12.5, 25.0, 37.5, and 50 µM) on the yield, physiological indices (net CO2 assimilation rate, transpiration, stomatal conductance, and chlorophyll fluorescence), quality, and Zn concentration of basil cultivars ‘Aroma 2’ and ‘Eleonora’ grown in a floating raft system. The ABTS, DPPH, and FRAP antioxidant activities were determined by UV-VIS spectrophotometry, the concentrations of phenolic acids by mass spectrometry using a Q Extractive Orbitrap LC-MS/MS, and tissue Zn concentration by inductively coupled plasma mass spectrometry. Although increasing the concentration of Zn in the nutrient solution significantly reduced the yield, this reduction was less evident in ‘Aroma 2’. However, regardless of cultivar, the use of the maximum dose of Zn (50 µM) increased the concentration of carotenoids, polyphenols, and antioxidant activity on average by 19.76, 14.57, and 33.72%, respectively, compared to the Control. The significant positive correlation between Zn in the nutrient solution and Zn in plant tissues underscores the suitability of basil for soilless biofortification programs.

The name “Vesuvian apricot” refers to numerous varieties originating from the same production area at the foot of Vesuvius and with a long tradition of cultivation. The importance of the apricot fruit and its derivatives on human health is known thanks to the presence of several secondary metabolites, many of them being active as antioxidants. This study aims to analyze 12 vesuvian apricot accessions by means of chemical-physical (fruits weights, firmness, TSS, TA, pH, and color fruits) and nutraceutical analyses (acid organic and sugar content, antioxidant activities, and polyphenol content). All the accessions analyzed can be defined as superior-quality apricots because they had TSS values ≥ 13 and a pulp strength ≤ 1 kg/0.5 cm2. Another parameter used to express the quality of apricot fruits was TSS-F (soluble solids content-pulp hardness) which showed a value ≥ 12. The total sugar content ranged from 260.40 mg/g d.w in ‘VM’ to 744.59 mg/g d.w in ‘SC’. In all accessions analyzed the sugar content was in the following order: sucrose > glucose > fructose. The antioxidant activity showed a high variability between the different accessions. Our results show that the Vesuvian apricot accessions present a large range with different organoleptic characteristics and offer the possibility to choose according to consumer and processing preferences.

The development of effective tools for the sustainable supply of phyto-ingredients and natural substances with reduced environmental footprints can help mitigate the dramatic scenario of climate change. Plant cell cultures-based biorefineries can be a technological advancement to face this challenge and offer a potentially unlimited availability of natural substances, in a standardized composition and devoid of the seasonal variability of cultivated plants. Monounsaturated (MUFA) fatty acids are attracting considerable attention as supplements for biodegradable plastics, bio-additives for the cosmetic industry, and bio-lubricants. Cardoon (Cynara cardunculus L. var. altilis) callus cultures accumulate fatty acids and polyphenols and are therefore suitable for large-scale production of biochemicals and valuable compounds, as well as biofuel precursors. With the aim of boosting their potential uses, we designed a biotechnological approach to increase oleic acid content through Agrobacterium tumefaciens-mediated metabolic engineering. Bioinformatic data mining in the C. cardunculus transcriptome allowed the selection and molecular characterization of SAD (stearic acid desaturase) and FAD2.2 (fatty acid desaturase) genes, coding for key enzymes in oleic and linoleic acid formation, as targets for metabolic engineering. A total of 22 and 27 fast-growing independent CcSAD overexpressing (OE) and CcFAD2.2 RNAi knocked out (KO) transgenic lines were obtained. Further characterization of five independent transgenic lines for each construct demonstrated that, successfully, SAD overexpression increased linoleic acid content, e.g., to 42.5%, of the relative fatty acid content, in the CcSADOE6 line compared with 30.4% in the wild type (WT), whereas FAD2.2 silencing reduced linoleic acid in favor of the accumulation of its precursor, oleic acid, e.g., to almost 57% of the relative fatty acid content in the CcFAD2.2KO2 line with respect to 17.7% in the WT. Moreover, CcSADOE6 and CcFAD2.2KO2 were also characterized by a significant increase in total polyphenolic content up to about 4.7 and 4.1 mg/g DW as compared with 2.7 mg/g DW in the WT, mainly due to the accumulation of dicaffeoyl quinic and feruloyl quinic acids. These results pose the basis for the effective creation of an engineered cardoon cells-based biorefinery accumulating high levels of valuable compounds from primary and specialized metabolism to meet the industrial demand for renewable and sustainable sources of innovative bioproducts.

The floating raft constitutes a valuable system for growing herbs as it effectuates high yield and prime functional quality. However, the pressing need for advancing sustainability in food production dictates the reduction of chemical fertilizer inputs in such intensive production schemes through innovative cultivation practices. In this perspective, our work appraised the productive and qualitative responses of two “Genovese” basil genotypes (Eleonora and Italiano Classico) grown in a floating raft system with nutrient solutions of varied electrical conductivity (EC; 2 and 1 dS m−1) combined with root application of protein hydrolysate biostimulant at two dosages (0.15 and 0.3 0 ml L−1 of Trainer®). The phenolic composition, aromatic profile, and antioxidant activities (ABTS, DPPH, and FRAP) of basil were determined by UHPLC/HRMS, GC/MS, and spectrophotometry, respectively. “Eleonora” demonstrated higher number of leaves (37.04 leaves per plant), higher fresh yield (6576.81 g m−2), but lower polyphenol concentration (1440.81 μg g−1 dry weight) compared to “Italiano Classico.” The lower EC solution (1 dS m−1) increased total phenols (+32.5%), ABTS, DPPH, and FRAP antioxidant activities by 33.2, 17.1, and 15.8%, respectively, and decreased linalool relative abundance by 5.5%. Biostimulant application improved crop performance and increased total phenolic concentration in both genotypes, with the highest phenolic concentration (1767.96 μg g−1 dry weight) registered at the lowest dose. Significant response in terms of aromatic profile was detected only in “Eleonora.” Our results demonstrate that the application of protein hydrolysate may compensate for reduced strength nutrient solution by enhancing yield and functional quality attributes of “Genovese” basil for pesto.

The supplementation of bioactive compounds in astronaut’s diets is undeniable, especially in the extreme and inhospitable habitat of future space settlements. This study aims to enhance the Martian and Lunar regolith fertility (testing two commercial simulants) through the provision of organic matter (manure) as established by in situ resource utilization (ISRU) approach. In this perspective, we obtained 8 different substrates after mixing Mojave Mars Simulant (MMS-1) or Lunar Highlands Simulant (LHS-1), with four different rates of manure (0, 10, 30, and 50%, w/w) from monogastric animals. Then, we assessed how these substrates can modulate fresh yield, organic acid, carotenoid content, antioxidant activity, and phenolic profile of lettuce plants (Lactuca sativa L.). Regarding fresh biomass production, MMS-1-amended substrates recorded higher yields than LHS-1-ones; plants grown on a 70:30 MMS-1/manure mixture produced the highest foliar biomass. Moreover, we found an increase in lutein and β-carotene content by + 181 and + 263%, respectively, when applying the highest percentage of manure (50%) compared with pure simulants or less-amended mixtures. The 50:50 MMS-1/manure treatment also contained the highest amounts of individual and total organic acids, especially malate content. The highest antioxidant activity for the ABTS assay was recorded when no manure was added. The highest content of total hydroxycinnamic acids was observed when no manure was added, whereas ferulic acid content (most abundant compound) was the highest in 70:30 simulant/manure treatment, as well as in pure LHS-1 simulant. The flavonoid content was the highest in pure-simulant treatment (for most of the compounds), resulting in the highest total flavonoid and total phenol content. Our findings indicate that the addition of manure at specific rates (30%) may increase the biomass production of lettuce plants cultivated in MMS-1 simulant, while the phytochemical composition is variably affected by manure addition, depending on the stimulant. Therefore, the agronomic practice of manure amendment showed promising results; however, it must be tested with other species or in combination with other factors, such as fertilization rates and biostimulants application, to verify its applicability in space colonies for food production purposes.

The effects of climate change have a great impact on the Mediterranean regions which are experiencing an increase in drought periods with extreme temperatures. Among the various solutions reported to reduce the damage caused by extreme environmental conditions on olive plants, the application of anti-transpirant products is widespread. In an increasingly current scenario of climate change, this study was designed to evaluate the effect of kaolin on the quantitative and qualitative parameters of drupes and oil in a little-known olive cultivar known as “Racioppella”, belonging to the autochthonous germplasm of Campania (Southern Italy). To this purpose, the determination of maturation index, olive yield/plant, and bioactive components analysis (anthocyanins, carotenoids, total polyphenols, antioxidant activity, and fatty acids) were carried out. Kaolin applications showed no statistically significant differences in terms of production/plant while a significant increase in the drupe oil content was observed. Kaolin treatments resulted in increased anthocyanins (+24%) and total polyphenols (+60%) content and at the same time a significant increase in the antioxidant activity (+41%) of drupes was recorded. As far as oil is concerned, the results showed an increase in monounsaturated fatty acids, oleic and linoleic acids, and total polyphenols (+11%). On the basis of the results obtained, we can conclude that kaolin treatment can be considered as a sustainable solution to improve qualitative parameters in olive drupes and oil.

Over the years, the use of biostimulants has become increasingly widespread due to their proven efficiency in improving plant productivity and quality of fruits and mitigating the effects related to environmental stress. The aim of the present study was to evaluate the effect of three biostimulants on oil yield, production of drupes per plant, and nutraceutical components of olive drupes and oil (total polyphenols, anthocyanins, and fatty acids %) for “Racioppella” cultivar trees growing in South Italy (May–October 2021). The biostimulants used were: a tropical plants extract (A) containing amino acids, vitamins, enzymes, phytochelatins, macro- and microelements, a glycine betaine-based product (B), and a Trichoderma spp.-based biostimulant (T). The three biostimulants were compared with a control thesis (C) treated only with water. T treatment increased the polyphenols content of olive drupes by 41.04% compared to C. A and B treatments increased polyphenols content by 21.87% on average compared to C. All three biostimulants showed positive effect by increasing the amount of polyphenols in olive oil compared to C:T showed an increase of 32.19%, B 7.76%, and A 19.78%. Biostimulant application proved useful in boosting fundamental parameters that determine better drupe and oil in terms of antioxidant capacity and nutraceutical potential, other than an increased production.

Microgreens represent a new generation of food products, commonly used to garnish and embellish culinary dishes, and recently associated with an increasing interest in their nutraceutical and phytochemical profiles. Four Apiaceae species: Pimpinella anisum L. (anise), Anthriscus cerefolium L. (chervil), Carum carvi L. (caraway), and Anethum graveolens L. (dill) were assessed for fresh yield, macro- and microminerals, total chlorophylls, total ascorbic acid, carotenoids, polyphenols, and their antioxidant activity. Anise was the species yielding the most (2.53 kg m−2) and having the highest lutein content (18.4 µg g−1 dry weight (DW)). Chervil and dill were characterized by the highest total ascorbic acid content (~151 mg AA g−1 fresh weight (FW)). The phenolic profile highlighted the presence of five flavonoid derivatives and 12 phenolic acid derivatives, with quinic acid derivatives being the most abundant phenols in the species tested. In addition, anise, caraway, and dill proved to be considerably rich in total polyphenols (~11056 μg g−1 DW). Caraway and dill were characterized by the highest antioxidant activity measured by the DPPH and ABTS methods, whereas the FRAP method revealed caraway as having the highest antioxidant activity. Such results highlight the potential of Apiaceae species as an alternative to other families which are commonly used for microgreens production.

Plant factories and high-tech greenhouses offer the opportunity to modulate plant growth, morphology and qualitative content through the management of artificial light (intensity, photoperiod and spectrum). In this study, three Light Emitting Diode (LED) lighting systems, with blue (B, 460 nm), red (R, 650 nm) and mixed red + green-yellow + blue (RGB) light were used to grow parsley microgreens to understand how light quality could change the phenotype and the profile of secondary metabolites. Plants showed altered morphological characteristics and higher amounts of secondary metabolites under RGB LEDs treatment. The results demonstrated that microgreens under red light showed the highest fresh yield, petiole length, coumaric acid content but also the highest nitrate content. Plants under RGB light showed the highest dry matter percentage and highest content of total and single polyphenols content, while blue light showed the highest ascorbic acid and ABTS antioxidant activity. Moreover, microgreens under red light showed more compact leaves with less intercellular spaces, while under blue and RGB light, the leaves displayed ticker spongy mesophyll with higher percentage of intercellular spaces. Therefore, the specific spectral band was able to modify not only the metabolic profile, but also it could modulate the differentiation of mesophyll cells. Light quality as a preharvest factor helps to shape the final parsley microgreens product as a whole, not only in terms of yield and quality, but also from a morpho-anatomical point of view.

The use of biostimulants in modern agriculture has rapidly expanded in recent years, owing to their beneficial effects on crop yield and product quality, which have come under the scope of intensive research. Accordingly, in the present study we appraised the efficacy of two plant-derived biostimulants, the legume-derived protein hydrolysates Trainer® (PH), and the tropical plant extract Auxym® (TPE) on two lettuce cultivars (green and red salanova®) in terms of morpho-physiological and biochemical traits (primary and secondary metabolites). The two cultivars differed in their acquisition capacity for nitrate and other beneficial ions, their photosynthetic and transpiration rates, and their ability to synthetize and accumulate organic acids and protective metabolites. The biostimulant effect was significant for almost all the parameters examined but it was subjected to significant cultivar × biostimulant interactions, denoting a cultivar-dependent response to biostimulant type. Notwithstanding this interaction, biostimulant application could potentially improve the yield and quality of lettuce by stimulating plant physiological processes, as indicated by the SPAD index (leaf chlorophyll index), ACO2 (assimilation rate), E (transpiration), and WUEi (intrinsic water use efficiency), and by increasing concurrently the plant mineral content (total N, K, Ca, Mg) and the biosynthesis of organic acids (malate, citrate), phenols (caffeic acid, coumaroyl quinic acid isomer 1, dicaffeoylquinic acid isomer 1), and flavonoids (quercetin-3-O-glucuronide, quercetin-3-O-glucoside). Biostimulant action may facilitate the bio-enhancement of certain lettuce cultivars that are otherwise limited by their genetic potential, for the accumulation of specific compounds beneficial to human health.

Cultivated cardoon (Cynara cardunculus L. var altilis) is a Mediterranean traditional food crop. It is adapted to xerothermic conditions and also grows in marginal lands, producing a large biomass rich in phenolic bioactive metabolites and has therefore received attention for pharmaceutical, cosmetic and innovative materials applications. Cardoon cell cultures can be used for the biotechnological production of valuable molecules in accordance with the principles of cellular agriculture. In the current study, we developed an elicitation strategy on leaf-derived cardoon calli for boosting the production of bioactive extracts for cosmetics. We tested elicitation conditions that trigger hyper-accumulation of bioactive phenolic metabolites without compromising calli growth through the application of chilling and salt stresses. We monitored changes in growth, polyphenol accumulation, and antioxidant capability, along with transcriptional variations of key chlorogenic acid and flavonoids biosynthetic genes. At moderate stress intensity and duration (14 days at 50–100 mM NaCl) salt exerted the best eliciting effect by stimulating total phenols and antioxidant power without impairing growth. Hydroalcoholic extracts from elicited cardoon calli with optimal growth and bioactive metabolite accumulation were demonstrated to lack cytotoxicity by MTT assay and were able to stimulate pro-collagen and aquaporin production in dermal cells. In conclusion, we propose a “natural” elicitation system that can be easily and safely employed to boost bioactive metabolite accumulation in cardoon cell cultures and also in pilot-scale cell culture production.

Carob is a predominantly rainfed tree crop of high nutritive value and a long history of adaptation to the edaphoclimatic stress conditions of the Mediterranean. However, declining attention to the carob tree in recent decades has aggravated genetic erosion. The extant in situ germplasm varies both in terms of pod morphology and composition, reflecting the genetic and physiological divide chiefly among grafted and non-grafted material, and possibly the impact of variable agro-environments. Accordingly, the present study aimed to establish a systematic categorization of the genetic and phenotypic diversity encountered across carob germplasm identified in situ throughout Cyprus, a historical center of production and genetic diversity for the species. Linking pod morphology, primary and secondary metabolite profiles with genotyped source material originating in different agro-environments and crop seasons would provide a framework for interpreting (a) the interaction of these factors in configuring carob pod physicochemical constitution, and (b) the relative stability of phenotypic traits against environmental and seasonal variation. Microsatellite analysis discriminated 36 genotypes out of the 124 trees located in nine traditional agro-environmental zones and revealed low genetic diversity within the grafted germplasm. Two landraces were identified: “Tillyria,” which is widespread and predominant, and “Kountourka,” which is mainly localized to the northeastern peninsula of Karpasia. Morphological traits, such as seeds-to-pod weight ratio, pod width and thickness were principally under genetic control. Contrarily, compositional traits, particularly total phenolic content—including condensed tannins, in vitro antioxidant capacity and to a lesser extent gallic acid, organic acids and minerals were under agro-environmental control. Agro-environmental zone also modulated principally fructose and glucose; sucrose was modulated equally by genotype and agro-environment, while total sugars were under genetic control. Statistically significant differences between seasons were detected for all traits except for the seeds-to-pod weight ratio, pod length and width. Hierarchical cluster analysis corroborates that Cyprus may be divided into two major agro-environmental zones modulating the compositional properties of the carob pulp. The present study provides a comprehensive insight into the extant carob genetic resources of Cyprus and advances our understanding of how genetic, agro-environmental and seasonal factors interact in shaping carob pod morphology and composition.

Cynara cardunculus (Asteraceae family) is a perennial plant native to Mediterranean regions. This plant represents a source of high-value compounds, such as polyphenols and fatty acids that have several industrial applications. However, in vitro plant cell cultures can represent a valid alternative to in-field cultivation and facilitate the extraction of metabolites of commercial interest. Generally, sucrose is the main sugar used for plant cell cultures, but other carbon sources can be considered. Here, we investigated the potential use of alternative organic carbon sources, such as galactose, maltose, glucose, glycerol, fructose, lactose, and starch, for the cultivation of C. cardunculus cells. Moreover, cardoon cells were collected, and an extraction of polyphenols and oils was performed to study the effects of different carbon sources on the production of bioactive molecules. This study provided evidence that cardoon cell growth can be supported by carbon sources other than sucrose. However, the carbon source inducing optimum growth, did not necessarily induce the highest production of high-value compounds.

The negative impacts of extreme heat and drought on olive plants have driven the quest for mitigation approaches based on the use of biostimulants, which have proved to be effective in contrasting environmental stresses. The aim of our study was to evaluate the effectiveness of six biostimulants in mitigating high temperature and water stress in young olive trees in terms of vegetative and eco-physiological parameters as well as bioactive compound content. Biostimulants based on glycine betaine and macro- and micro-algae effectively protected the plants from abiotic stress by improving their eco-physiological and vegetative parameters. At the end of the growing season, olive plants were experiencing water deficit which had built up through the summer months. At this time, the glycine betaine-treated plants had a three-fold higher stomatal conductance compared with the control, while plants sprayed with the seaweed mix had a relative water content 33% higher than the control. The kaolin treatment resulted in higher total phenolics and antioxidant activities (DPPH, FRAP and ABTS) in water stress conditions and caused an increase of 238.53 and 443.49% in leaves total polyphenols content in 100% and 50% water regime, respectively. This study showed the effectiveness of biostimulants in mitigating the damage from abiotic stress on young olive trees, by improving some vegetative, eco-physiological and leaf nutraceutical parameters. Further studies are needed to test the efficiency of these biostimulants in open field conditions on olive trees in full production.

L’obiettivo di questa trattazione è estendere la metafora del “fuori luogo” alla società civile cinese e alle organizzazioni sociali per i diritti della donna e femministe. Nel primo caso il “fuori luogo” è rappresentato dall’output del tentativo di importazione di concetti puramente occidentali, nati in seno al pensiero e alla cultura occidentale; nel secondo caso il “fuori luogo” è determinato dalla ricerca di spazi di azione interstiziali, dove operare al confine tra legalità e illegalità. Nella prima parte si analizzano le differenti configurazioni della società civile cinese determinate dalla mancanza di una definizione netta dei confini spaziali tra Stato e spazio civico. Nella seconda parte viene approfondito il ruolo e la funzione delle organizzazioni sociali per i diritti della donna e femministe, le quali a seconda del grado di assoggettamento Statale dell’organizzazione cambieranno le istanze, la figura della donna e le strategie di azione.

Advanced analytical data on microgreens' response to different light spectra constitutes a valuable resource for designing future crop-specific spectral management systems. The current study defined variation in productivity, nutritive and functional quality (mineral–carotenoid–polyphenolic profiles and antioxidant capacity) of novel microgreens (amaranth, cress, mizuna, purslane) in response to select spectral bandwidths (red, blue, blue-red), and appraised clustering patterns configured by the genotype-light-spectrum nexus. Growth parameters dependent on primary metabolism were most favored by blue-red light's efficiency in activating the photosynthetic apparatus. Nitrate accumulation was higher under monochromatic light owing to the dependency of nitrite reductase on the light-driven activity of PSI, most efficiently promoted by blue-red light. Although mineral composition was mostly genotype-dependent, monochromatic red and blue lights tended to increase K and Na and decrease Ca and Mg concentrations. Lutein, β-carotene, and lipophilic antioxidant capacity were generally increased by blue-red light putatively due to the coupling of heightened photosynthetic activity to increased demand for protection against oxidative stress; the disparate response however of purslane highlights the importance of genotype specificity in these responses and calls for additional investigation. Analysis of polyphenols by Orbitrap LC-MS/MS revealed substantial genotypic differences. Most abundant phenolics were chlorogenic acid (x¯ = 5503 µg g−1 dw), feruloylquinic acid (x¯ = 974.1 µg g−1 dw), and caffeoyl feruloyl tartaric acid (x¯ = 993 µg g−1 dw). Hydroxycinnamic acids accounted for 79.0% of the mean total phenolic content across species, flavonol glycosides for 20.7%, and flavone glycosides for 0.3%. The general response across species was a decrease in individual polyphenolic constituents, particularly flavonol glycosides, and total polyphenols under blue-red light. The pronounced effectiveness of monochromatic blue light in eliciting synthesis of flavonoids could be linked to their capacity for absorbing shorter wavelengths thereby quenching generated photo-oxidation potential. The light-induced stimulation of the phenylpropanoid pathway by monochromatic blue light through epigenetic mechanisms or redox signaling in the photosynthetic apparatus warrants further investigation. The current work highlights how optimized genetic background combined with effective light management might facilitate the production of superior functional quality microgreens.

Tomato (Solanum lycopersicum L.) is one of the most consumed vegetables worldwide due to its low caloric intake and high fiber, minerals, and phenolic compounds, making it a high-quality functional food. However, fruit quality attributes can be affected by pre-harvest factors, especially environmental stresses. This research aimed to evaluate the influence of two shading nets (white net −30% and pearl grey net −40% shading degree) on the yield and phytochemical profile of tomato fruits grown in summer under the Mediterranean climate. Mineral and organic acid content (by ion chromatography-IC), phenolic profile (by ultra-high performance liquid chromatography-UHPLC coupled with an Orbitrap high-resolution mass spectrometry-HRMS), carotenoid content (by high-performance liquid chromatography with diode array detection-HPLC-DAD), and antioxidant activities DPPH, ABTS, and FRAP (by UV-VIS spectrophotometry) were determined. Tomato fruits grown under the pearl grey net recorded the highest values of total phenolic compounds (14,997 µg 100 g−1 of fresh weight) and antioxidant activities DPPH, ABTS, and FRAP, without affecting either fruit color or marketable yield. The reduction of solar radiation through pearl grey nets proved to be an excellent tool to increase the phytochemical quality of tomato fruits during summer cultivation in a Mediterranean environment.

Cultivated cardoon (Cynara cardunculus var. altilis L.) is a promising candidate species for the development of plant cell cultures suitable for large-scale biomass production and recovery of nutraceuticals. We set up a protocol for Agrobacterium tumefaciens-mediated transformation, which can be used for the improvement of cardoon cell cultures in a frame of biorefinery. As high lignin content determines lower saccharification yields for the biomass, we opted for a biotechnological approach, with the purpose of reducing lignin content; we generated transgenic lines overexpressing the Arabidopsis thaliana MYB4 transcription factor, a known repressor of lignin/flavonoid biosynthesis. Here, we report a comprehensive characterization, including metabolic and transcriptomic analyses of AtMYB4 overexpression cardoon lines, in comparison to wild type, underlining favorable traits for their use in biorefinery. Among these, the improved accessibility of the lignocellulosic biomass to degrading enzymes due to depletion of lignin content, the unexpected increased growth rates, and the valuable nutraceutical profiles, in particular for hydroxycinnamic/caffeoylquinic and fatty acids profiles.

Various products are used to mitigate the negative effects of abiotic stress in olive trees. The aim of the research was to examine an anti-transpirant product (Vapor Gard®, V) and a kaolin- based product (Manisol, K) effect on the growth of two-year-old olive tree seedlings under high temperature. The study was conducted in a greenhouse on trees of a native cultivar of Campania (cv. Salella) grown in pot during the growing season from May to September 2020. The experimental design included two products: di-1-p-menthene (product V) and kaolin (product K), applied five times at 20 day intervals compared with a control. The following biometric, physiological, and nutraceutical parameters were evaluated: stomatal conductance, chlorophyll a fluorescence, Soil Plant Analysis Development (SPAD) index, relative water content (RWC), shoots growth, total leaf area per plant, trunk cross-sectional area, dry matter partitioning, total polyphenols, and antioxidant activity. The results obtained showed that the application of di-1-p-menthene (V) was able to induce a significant improvement of shoots growth (+37.22%) and trunk cross-sectional area (+46.60%) and a reduction of the stomatal conductance and an increase of leaf RWC values. Application with kaolin had positive effects on the total polyphenol content, with an increase over the control of 240.33% and higher antioxidant activity values. Further studies are necessary to determine the effect of these products on the biometric, physiological and nutraceutical parameters of mature olive trees cultivated in open field conditions.

Treatment of relapsed/refractory multiple myeloma (RRMM) is more complex today due to the availability of novel therapeutic options, mostly applied as combination regimens. immunotherapy options have especially increased substantially, likewise the understanding that patient-, disease- and treatment-related factors should be considered at all stages of the disease. RRMM is based on definitions of the international myeloma working group (IMWG) and includes biochemical progression, such as paraprotein increase, or symptomatic relapse with CRAB criteria (hypercalcemia, renal impairment, anemia, bone lesions). When choosing RRMM-treatment, the biochemical markers for progression and severity of the disease, dynamic of disease relapse, type and number of prior therapy lines, including toxicity and underlying health status, need to be considered, and shared decision making should be pursued. Objectively characterizing health status via geriatric assessment (GA) at each multiple myeloma (MM) treatment decision point has been shown to be a better estimate than via age and comorbidities alone. The well-established national comprehensive cancer network, IMWG, European myeloma network and other national treatment algorithms consider these issues. Ideally, GA-based clinical trials should be supported in the future to choose wisely and efficaciously from available intervention and treatment options in often-older MM adults in order to further improve morbidity and mortality.

Neglected and underutilized species (NUS) offer largely unexplored opportunities for providing nutritious plant food, while making agro-ecosystems more diverse and resilient to climate change. The aim of this work was to explore the potential of two typical Mediterranean underutilized species, purslane and borage, as novel vegetable product (microgreens). Micro-scale production of edible plants is spreading due to the simplicity of their management, rapid cycle, harvest index, and phytochemical value of the edible product. Microgreens, therefore, represent an opportunity to link NUS, nutrition, and agricultural and dietary diversification. By analyzing yield, antioxidants activities, mineral composition, and main phenolic acids and flavonoids, our work indicated that the two species provide interesting results when compared with those reported for crops and horticultural species. Specifically, purslane should be considered highly nutritional due to the amount of phenolic compounds and ascorbic acid, and to potential good β-carotene bioavailability. Borage microgreens have a very high fresh yield and a more composite and balanced phenolic profile. In conclusion, our work provided evidence for implementing new ways to expand the NUS market-chains and for developing added-value food products.

While imparting gastronomic novelty and sensory delight, microgreens also constitute rudimentary leafy greens packed with nutrients and phytochemicals. As such, they comprise an upcoming class of functional foods. However, apart from bioactive secondary metabolites, microgreens also accumulate antinutritive agents such as nitrate, especially under conducive protected cultivation conditions. The current work examined nutrient deprivation before harvest (DBH), applied by replacing nutrient solution with osmotic water for six and twelve days, as a strategy for reducing microgreen nitrate levels in different species (lettuce, mustard, and rocket). The three species were sown on a peat-based substrate, cultivated in a controlled climate chamber, and harvested 18 days after sowing, when the first two true leaves emerged. DBH impact on major constituents of the secondary metabolome, mineral content, colorimetric, and yield traits was appraised. Nitrate and mineral content were determined through ion chromatography, phenolic composition through UHPLC-Q-Orbitrap HRMS, and carotenoid composition through HPLC-DAD. Nutrient deprivation was effective in reducing nitrate content; however, effective treatment duration differed between species and decline was more precipitous in nitrate hyperaccumulating species such as rocket. Quercetin and kaempferol glycosides were the flavonol glycosides most abundant in brassicaceous microgreens, whereas lettuce microgreens were steeped in caffeoyl quinic acid. DBH interacted with species as it increased the total phenolic content of lettuce, decreased that of rocket, but did not affect mustard. Further research to link changes in phenolic composition to the sensory and in vivo bioactive profile of microgreens is warranted. Notably, brief (≤6 days) DBH can be applied across species with moderate or no impact on the phenolic, carotenoid, and mineral composition of microgreens. Brief DBH applications also have limited impact on microgreens’ yield and colorimetric traits hence on the commercial value of the product. They can therefore be applied for reducing microgreen nitrate levels without significantly impacting key secondary metabolic constituents and their potential bioactive role.
