Abstract The vector forces at the human-mattress interface are not only crucial for understanding the distribution of vertical and shear forces exerted on the human body during sleep but also serves as a significant input for biomechanical models of sleeping positions, whose accuracy determines the credibility of predicting musculoskeletal system loads. In this study, we introduce a novel method for calculating the interface vector forces. By recording indentations after supine and lateral positions using a vacuum mattress and 3D scanner, we utilize image registration techniques to align body pressure distribution with the mattress deformation scanning images, thereby calculating the vector force values for each unit area (36.25 mm × 36.25 mm). This method was validated through five participants attendance from two perspectives, revealing that (1) the mean summation of the vertical force components is 98.67% ± 7.21% body weight, exhibiting good consistency, and mean ratio of horizontal component force to body weight is 2.18% ± 1.77%. (2) the predicted muscle activity using the vector forces as input to the sleep position model aligns with the measured muscle activity (%MVC), with correlation coefficient over 0.7. The proposed method contributes to the vector force distribution understanding and the analysis of musculoskeletal loads during sleep, providing valuable insights for mattress design and evaluation.

Autoimmune diseases, such as rheumatoid arthritis (RA) and systemic lupus erythematous, are regulated by polymorphisms in genes contributing to the NOX2 complex. Mutations in both Ncf1 and Ncf4 affect development of arthritis in experimental models of RA, but the different regulatory pathways mediated by NOX2-derived reactive oxygen species (ROS) have not yet been clarified.Here we address the possibility that intracellular ROS, regulated by the NCF4 protein (earlier often denoted p40phox) which interacts with endosomal membranes, could play an important role in the oxidation of cysteine peptides in mononuclear phagocytic cells, thereby regulating antigen presentation and activation of arthritogenic T cells.To study the role of NCF4 we used mice with an amino acid replacing mutation (NCF4R58A), which is known to affect interaction with endosomal membranes, leading to decreased intracellular ROS production. To study the impact of NCF4 on T cell activation, we used the glucose phosphate isomerase peptide GPI325-339, which contains two cysteine residues (325-339c-c). Macrophages from mice with the NCF458A mutation efficiently presented the peptide when the two cysteines were intact and not crosslinked, leading to a strong arthritogenic T cell response. T cell priming occurred in the draining lymph nodes (LNs) within 8 days after immunization. Clodronate treatment, which depletes antigen-presenting mononuclear phagocytes, ameliorated arthritis severity, whereas treatment with FYT720, which traps activated T cells in LNs, prohibited arthritis.We conclude that NCF4-dependent intracellular ROS maintains cysteine peptides in an oxidized crosslinked state, which prevents presentation of peptides recognized by non-tolerized T cells and thereby protects against autoimmune arthritis.

Abstract Background Hepatocellular carcinoma (HCC) has a high incidence and mortality rate despite various treatment options, including 125I seed implantation. However, recurrence and radiation resistance remain challenging issues. Hsa_circ_0007895 (circEYA3)—derived from exons 2–6 of EYA3–facilitates the proliferation and progression of pancreatic ductal adenocarcinoma. However, the role of circEYA3 in HCC 125I radiation resistance remains unclear. Thus, we aimed to investigate the functions and underlying molecular mechanisms of circEYA3 in HCC under 125I and X-ray irradiation conditions. Methods CircEYA3 was identified by RNA-seq in patients with HCC before and after 125I seed implantation treatment, followed by fluorescence in situ hybridization and RNase R assays. The radiosensitivity of HCC cell lines irradiated with 125I seeds or external irradiation were evaluated using the Cell Counting Kit 8, flow cytometry, γH2A.X immunofluorescence and comet assays. RNA pull-down and RNA immunoprecipitation assays were performed to explore the interactions between circEYA3 and IGF2BP2. DTX3L mRNA was identified by RNA-seq in PLC/PRF/5 cells with overexpressed circEYA3. The corresponding in vitro results were verified using a mouse xenograft model. Results CircEYA3 decreased the radiosensitivity of HCC cells both in vitro and in vivo. Notably, using a circRNA pulldown assay and RNA-binding protein immunoprecipitation, we identified IGF2BP2 as a novel and robust interacting protein of circEYA3. Mechanistically, circEYA3 binds to IGF2BP2 and enhances its ability to stabilize DTX3L mRNA, thereby specifically alleviating radiation-induced DNA damage in HCC cells. Conclusions Our findings demonstrate that circEYA3 increases the radioresistance of HCC to 125I seeds and external irradiation via the IGF2BP2/DTX3L axis. Thus, circEYA3 might be a predictive indicator and intervention option for 125I brachytherapy or external radiotherapy in HCC.

In this work, layer-by-layer organic photovoltaics (LbL OPVs) were prepared by sequentially spin-coating PM1 and L8-BO solutions. The solvent additive 1,8-diiodooctane (DIO), which has a high boiling point, and solid additive l,3,5-trichlorobenzene (TCB), which has a high volatile, were deliberately selected to incorporate with the L8-BO solutions. The power conversion efficiency (PCE) of LbL OPVs was considerably enhanced from 17.43% to 18.50% by employing TCB as the additive, profiting by the concurrently increased short-circuit current density (JSC) of 26.74 mA cm−2 and a fill factor (FF) of 76.88%. The increased JSCs of LbL OPVs with TCB as additive were ascribed to the tilted-up absorption edge in the long wavelength range and the external quantum-efficiency spectral difference between LbL OPVs with and without TCB as an additive. The molecular arrangement of L8-BO and the PM1 domain was enhanced with TCB as an additive, which was most likely responsible for the increased charge mobilities in the layered films processed with additives. It was indicated that the dynamic film-forming process of the acceptor layers plays a vital role in achieving efficient LbL OPVs by employing additive strategy. Over 6% PCE improvement of the LbL OPVs with PM1/L8-BO as the active layers can be achieved by employing TCB as additive.

Abstract Considering the multi-functionalization of ligands, it is crucial for ligand molecular design to reveal the landscape of anchoring sites. Here, a typical triphenylphosphine (TPP) ligand was employed to explore its effect on the surface of CsPbI3 perovskite nanocrystals (PNCs). Except for the conventionally considered P-Pb coordination, an P-I supramolecular halogen bonding was also found on the NC surface. The coexistence of the above two types of bonding significantly increased the formation energy of iodine vacancy defects and improved the photoluminescence quantum yield of PNCs up to 93%. Meanwhile, the direct interaction of P and I enhanced the stability of the Pb-I octahedra and dramatically inhibited the migration of I ions. Furthermore, the introduction of additional benzene rings (2-(Diphenylphosphino)-biphenyl (DPB)) increased the delocalized properties of the PNC surface and significantly improved the charge transport of the PNCs. As a result, the DPB passivated CsPbI3 NCs based top-emitting LEDs exhibite a peak external quantum efficiency (EQE) of 22.8%, a maximum luminance of 15, 204 cd m−2, and an extremely low-efficiency roll-off of 2.6% at the current density of 500 mA cm−2.

TiO2 is a sintering additive that can significantly improve the sintering and mechanical properties of Cr2O3–Al2O3 refractory materials with high Cr2O3 contents. In this study, TiO2-reinforced Cr2O3–Al2O3 composite refractories were prepared using a reaction-sintering process. The microstructure, phase composition, mechanical properties, and strengthening mechanism of the TiO2-reinforced Cr2O3–Al2O3 composite refractories were investigated. The results indicated that the Al2O3–Cr2O3–TiO2 composite refractory with a TiO2 content of 3.0–4.5 wt% had good compactness (porosity of 5.89%), hardness (17.21 GPa), wear resistance (wear rate of 0.4 mm3/Nm), flexural strength (101.10 MPa), and compressive strength (420.90 MPa). Malignant tumors are major causes of morbidity and mortality in China. Despite advances in surgical, radiological, chemotherapeutic, molecular targeting, and immunotherapeutic treatments, patients with malignant tumors still have poor prognoses. Low-dose-rate brachytherapy, specifically 125I seed implantation, is beneficial because of its high local delivery dose and minimal damage to surrounding tissues. Consequently, it has gained increasing acceptance as a treatment modality for various malignant tumors. In this study, we explored the fundamental principles, clinical applications, and new technologies associated with 125I radioactive seed implantation.

In this work, pseudo-bilayer planar heterojunction (PPHJ) all-polymer solar cells (APSCs) were constructed on the basis of the commonly used PY-IT and PM6 as the acceptor and donor, respectively. A nonfullerene small molecular acceptor (NF-SMA) BTP-eC9 was incorporated into the PY-IT layer as the solid additive in consideration of its similar building block to PY-IT. BTP-eC9 can serve as a photon capture reinforcer and morphology-regulating agent to realize more adequate photon capture, as well as a more orderly molecular arrangement for effective carrier transport. By incorporating 2 wt% BTP-eC9, the efficiency of PM6/PY-IT-based PPHJ-APSCs was boosted from 15.11% to 16.47%, accompanied by a synergistically enhanced short circuit current density (JSC, 23.36 vs. 24.08 mA cm−2) and fill factor (FF, 68.83% vs. 72.76%). In another all-polymer system, based on PBQx-TCl/PY-DT as the active layers, the efficiency could be boosted from 17.51% to 18.07%, enabled by the addition of 2 wt% L8-BO, which further verified the effectiveness of using an NF-SMA as a solid additive. This work demonstrates that incorporating an NF-SMA as a solid additive holds great potential for driving the development of PPHJ-APSCs.

Purpose: Whether COVID-19 reduces male fertility remains requires further investigation. This meta-analysis and systematic review evaluated the impact of COVID-19 on male fertility. Materials and Methods: The literature in PubMed, Embase, MEDLINE, Web of Science, and Cochrane Library up to January 01, 2022 was systematically searched, and a meta-analysis was conducted to investigate the effect of COVID-19 on male fertility. Totally 17 studies with a total of 1,627 patients and 1,535 control subjects were included in our meta-analysis. Results: Regarding sperm quality, COVID-19 decreased the total sperm count (p=0.012), sperm concentration (p=0.001), total motility (p=0.001), progressive sperm motility (p=0.048), and viability (p=0.031). Subgroup analyses showed that different control group populations did not change the results. It was found that during the illness stage of COVID-19, semen volume decreased, and during the recovery stage of COVID-19, sperm concentration and total motility decreased

In order to address the poor shifting quality issue of a certain model of heavy-duty vehicles, a multi-objective parameter optimization strategy based on engine coordinated control is proposed. This strategy aims to improve shifting quality by reducing the sliding friction work and impact during the shifting process. The Non-Dominated Sorting Genetic Algorithm II (NSGA-II) is employed to perform multi-objective optimization on the coordinated control parameters, which include external control torque of the engine, start of fuel cut-off timing, and duration of fuel cut-off. By comparing the performance of different parameter combinations in terms of sliding friction work and impact, the optimal parameter combination is determined. Through bench testing verification, it has been demonstrated that utilizing the optimized parameters for engine coordinated control during the torque phase of the shifting process can significantly enhance shifting quality. This strategy provides an effective solution for addressing shifting quality issues.

Opposed-piston 2-stroke diesel engines (OP2S) have lower combustion chamber surface-area-to-volume ratio, resulting in lower transfer loss but also excessive surface temperature. Investigation of combustion parameters on heat transfer, which provide a chance in improving combustion efficiency and solving surface overheating issue in OP2S at the same time, is yet to be undertaken. In this paper CFD simulation by CONVERGE software has been carried out to study heat transfer advantages in OP2S. Effect of initial swirl ratio and injection phase on combustion efficiency and heat transfer of combustion chamber surface area is investigated. Results show that OP2S has obtained peak combustion efficiency of 0.97, which is 0.02 higher than that of conventional diesel engine (4S). Also, combustion efficiency loss due to undesirable swirl ratio is only 20 % of 4S. Lowest heat transfer ratio achieved by OP2S is 0.045 of fuel energy, which is 34 % lower than that of 4s. However, heat transfer ratio of cylinder wall in OP2S is at least twice of that in 4S, and increase significantly with swirl. Unable to utilize squish, OP2S would suffer from at most 0.1 lower combustion efficiency compared to 4S when injection is retarded to TDC. Heat transfer ratio in OP2S is less sensitive to variation of injection phase, and stays at least 0.01 lower than 4S for all injection phase studied. In conclusion, OP2S achieved higher combustion efficiency while significantly cutting heat transfer loss compared to 4S at optimized swirl ratio and injection phase, proving its potential as a high-efficiency power source. Thermal load issue on cylinder wall in OP2S can be fixed by lowering swirl ratio. Reducing heat transfer loss in OP2S with delayed injection is undesirable, however, since combustion efficiency would suffer.

The HAA-GAN (Highlighting Artistic Attributes Generative Adversarial Net-work) style migration model is proposed to address the problem of poor expression of image artistic attributes and mismatch between semantic and stylistic features in images generated from Tangka image style transfer experiments. The image features extracted by the encoder are text dual-channeled feature transfer and feature refinement according to the style domain attribute refinement channel and the text self-attention semantic information matching channel. The former can highlight the artistic attributes of the images, while the latter can flexibly match the Tangka image style features to the content features according to the semantic space distribution of the content images. At the same time, adding a pooling layer based on wavelet multilevel transform to the codec structure of the generator can retain more key features of the image during the feature extraction process. Finally, in order to improve the quality of style transfer images, a multi-scale discriminative network structure is used to discriminate the image blocks, resulting in better visual effects of the generated images. The experimental results show that HAA-GAN conducted style transfer experiments on Thangka dataset and Impressionism dataset. Compared with SAFIN, DSTN and other style transfer models, the average score of FID and PSNR indicators increased by 9.06% and 12.7%, and the generation of stylized renderings achieved better results in structural detail maintenance, feature matching and artistic effect expression.

Abstract Pulmonary inflammation involves complex changes of the immune cells, in which macrophages play important roles and their function might be influenced by metabolism. Slc38a6 acts as a carrier of nutrient for macrophages (Mφ) to exert the function. In this study, pneumonia patient blood was found up-regulated SLC38A6 expression, which correlated with monocytes number and white blood cell number. The similar result was also shown in LPS induced sepsis mice. To reveal the key role of Slc38a6, we used systemic and conditional knock-out mice. Either systemic or LyzCRE specific knock-out could alleviate the severity of sepsis mice, reduce the proinflammatory cytokine TNF-α and IL-1β expression in serum and decrease the monocytes number in bronchial alveolar lavage and peritoneal lavage via flow cytometry. In order to reveal the signal of up-regulated Slc38a6, the Tlr4 signal inhibitor TAK242 and TLR4 knock-out mice were used. By blocking Tlr4 signal in macrophages via TAK242, the expression of Slc38a6 was down-regulated synchronously, and the same results were also found in Tlr4 knock-out macrophages. However, in the overexpressed Slc38a6 macrophages, blocking Tlr4 signal via TAK242, 20% of the mRNA expression of IL-1β still could be expressed, indicating that up-regulated Slc38a6 participates in IL-1β expression process. Collectively, it is the first time showed that an amino acid transporter SLC38A6 up-regulated in monocytes/macrophages promotes activation in pulmonary inflammation. SLC38A6 might be a promising target molecule for pulmonary inflammation treatment.

Purpose The aim of this study was to compare the effectiveness and outcomes of percutaneous ablation guided by ultrasonography (US) and computed tomography (CT) in colorectal liver oligometastases (CLOM). Methods This study included patients with CLOM treated with percutaneous ablation from January 2008 to January 2021 in this observational study. Only lesions visualized on both CT and US images were further analyzed according to whether patients’ initial ablation treatments utilized US guidance or CT guidance. The Kaplan-Meier method was used to estimate local tumor progression (LTP)–free survival after propensity score matching (PSM). The LTP-free survival and treatment-related outcomes were compared between these two groups. Results PSM identified 116 patients from each group, with 269 and 238 lesions in the USguided and CT-guided groups, respectively. US-guided ablation had a shorter average procedure time and lower cost than CT-guided ablation (27.54±12.06 minutes vs. 32.70±13.88 minutes, P=0.003; $2,175.13±618.17 vs. $2,455.49±710.25, P=0.002). For patients >60 years of age, the cumulative LTP rate at 1 year was lower in the US-guided group than in the CT-guided group (17.8% vs. 25.1%, P=0.038). For patients with perivascular liver lesions, the cumulative LTP rate at 1 year was lower in the US-guided group (14.4% vs. 28.2%, P=0.040). Conclusion For patients whose age is >60 years or who have perivascular liver lesions, USguided ablation is better than CT-guided ablation, with a shorter treatment time and lower costs when both ablation methods are feasible for patients.

It is increasing concerned that the plateau environment will potentially increase fuel consumption in engines. Despite this, the current state of research on high altitude engines is still inadequate in providing sufficient and detailed information to counteract the decrease in engine efficiency. In addition, one of the novelty of this study was that the investigated altitudes is up to 4500 m, of which the previous studies was limited. Therefore, this study aims to investigate the effect of altitude on in-cylinder spray, combustion, and soot formation processes in diesel engines. A calibrated three-dimensional (3D) computational fluid dynamics (CFD) model of a single-cylinder, four-strokes, direct injection (DI) compression ignition, intake boost research diesel engine is established. The numerical CFD model used in this study has been validated through comparison with experimental data. It effectively investigates the in-cylinder activities and provides insights into the causes behind combustion and soot emission deterioration. The simulation is operated at altitudes of 0, 1500, 3000, 4500 m with corresponding intake pressure. It has been observed that engine performance and soot emissions deteriorate as altitude increases, and a sharp drop occurs when the altitude exceeds 3000 m, which can be attributed to the dramatic decline in combustion efficiency. The extended spray leads to a decrease in air utilization, which ultimately results in a sudden drop in combustion efficiency at altitudes above 3000 m. Overall, this study has identified that the extended penetration of the spray, caused by reduced pressure and air density, leads to poor air utilization, resulting in decreased performance and increased emissions. Hence, it is recommended that optimization of the combustion chamber geometry and injection strategies of diesel engines operating at high altitudes be undertaken to improve air utilization and combustion quality. This will serve as an avenue for further research in the future.

Key points 125I brachytherapy is a treatment for locally recurrent HCC after ablation. 125I brachytherapy combined with ablation may improve tumor control. Vascular invasion, multiple lesions, and AFP are prognostic risk factors. 125I brachytherapy is a safe method with a curative effect.

PURPOSEThis study explored the clinical efficacy of transcatheter retrograde shunt occlusion with coils to prevent pulmonary oil or particle embolization prior to transarterial chemoembolization (TACE) in patients with artero-hepatic venous shunts (AHVS) secondary to hepatocellular carcinoma (HCC).METHODSFrom July 2017 to January 2021, 6 patients with advanced, unresectable HCC were found to have an AHVS by hepatic arteriography at the time of attempted TACE. The AHVS was embolized retrogradely with metal coils through a transfemoral or transjugular venous approach. After venous embolization and confirmation of the absence of the AHVS, TACE was performed using an emulsion of iodized oil and doxorubicin or drug-eluting beads. Follow-up computed tomography (CT) was performed within 1 month after the first TACE to evaluate the results and complications.RESULTSHepatic angiography after venous embolization showed that AHVS had utterly disappeared in all patients during the operation. The immediate technical success of the retrograde venous embolization was 100%. The AHVS had disappeared entirely during the follow-up period through triple-phase enhancement CT scanning. According to the modified response evaluation criteria in solid tumors, TACE in all 6 patients had a disease control response rate of 100% (6/6) with complete response in 2 patients and partial response in 4 patients. One patient died during the 6-month follow-up, and the other 5 were still alive. No complications related to pulmonary embolism occurred.CONCLUSIONRetrograde venous coil embolization of AHVS via the draining hepatic vein appears to be a safe, feasible, and effective treatment to allow TACE treatment without pulmonary embolic events. This approach appears to provide better tumor control and effectively decreases the occurrence of pulmonary embolism.

Owing to the extreme difficulty in their prevention, transverse corner cracks have been frequently observed in fine blanking steel produced on straight arc caster. In this study, the transient thermal module of Ansys was used to calculate the corner temperature distribution of a continuous casting slab. According to the actual cooling rate, a Gleeble 3500 thermal simulator was used to simulate the bending process of continuous casting, and the thermoplastic curves of test steel grades were drawn. Through the observation of fracture morphology and metallographic examination, typical precipitation detection and phase transformation temperature detection, the mechanism of corner cracks in experimental steel grades was determined and the corresponding control measures were formulated. The results showed that under the original process conditions, the corner temperature of the continuous casting slab was in the brittle temperature range of the steel grade when it entered the bending section. During the cooling process of the continuous casting slab, all Ti elements dissolved into the matrix, and the main precipitates were (Cr, Fe)XCY, while the grain boundary pinning was weak. In the high temperature brittle region, deformation-induced ferrite was produced at grain boundaries, which formed a stress concentration during the bending process which led to plastic reduction. The mixed control mode of strong cooling in the cooling stage and weak cooling in the reheating stage could not only ensure the strength of the slab shell in the vertical section, but also promoted the growth of the ferrite film induced by intergranular deformation in the strong cooling stage. It increased the maximum allowable stress of ferrite at the grain boundary. By controlling the temperature and cooling rate in the reheating stage, the growth of ferrite at the grain boundaries and the nucleation of ferrite in grains were accelerated, the stress distribution at grain boundaries and grains was uniform, and the corner cracks were eliminated.

Abstract Dual‐band photomultiplication type polymer photodetectors (PM‐PPDs) are achieved with the structure of ITO/PDIN/poly[N,N′‐bis(4‐butylphenyl)‐N,N′‐bis(phenyl)benzidine] (poly‐TPD):2,2′‐((2Z,2′Z‐(12,13‐bis(2‐ethylhexyl)‐3,9‐diundecyl‐12,13‐dihydro‐[1,2,5]thiadiazolo[3,4‐e]thieno[2′′,3′′:4′,5′]thieno‐[2′,3′:4,5] pyrrolo‐[3,2‐g]thieno[2′,3′:4,5]thieno[3,2‐b]indole‐2,10‐diyl)bis‐(methanylylidene)) bis‐(5,6‐difluoro‐3‐oxo‐2,3‐dihydro‐1H‐indene‐2,1‐diylidene))‐dimalononitrile (Y6) (100:2, wt/wt)/MoO3/poly(3‐hexylthiophene) (P3HT):[6,6]‐phenyl‐C71‐butyric acid methyl ester (PC71BM) (100:2, wt/wt)/Al. Electron traps can be formed with Y6 surrounded by poly‐TPD or with PC71BM surrounded by P3HT. Trapped electrons near ITO/PDIN or Al electrode can assist hole tunneling injectionunder forward or reverse bias, respectively. The dual‐band PM‐PPDs exhibit ultraviolet spectral response under forward bias dependent on trapped electron distribution near ITO/PDIN electrode, while presenting visible spectral response under reverse bias determined by trapped electron distribution near Al electrode. The performance of dual‐band PM‐PPDs can be optimized by decreasing P3HT:PC71BM layer thickness. The optimal dual‐band PM‐PPDs exhibit external quantum efficiency (EQE) of 2800% at 350 nm under +10 V bias with full‐width at half‐maximum of 80 nm, while achieving EQE of 7500% at 430 nm under −10 V bias. The EQE values of optimal dual‐band PM‐PPDs are markedly improved by raising bias, reaching 13 000% at 350 nm under +16 V bias and 27 000% at 430 nm under −16 V bias. This work provides an effective method to prepare dual‐band PM‐PPDs by manipulating the interfacial trapped electron distribution.

Abstract Background Postoperative cognitive dysfunction (POCD) is a common complication following anesthesia and surgery. General anesthetic isoflurane has potential neurotoxicity and induces cognitive impairments, but the exact mechanism remains unclear. Astrocytes form interconnected networks in the adult brain through gap junctions (GJs), which primarily comprise connexin 43 (Cx43), and play important roles in brain homeostasis and functions such as memory. However, the role of the GJ-Cx43-mediated astrocytic network in isoflurane-induced cognitive dysfunction has not been defined. Methods 4-month-old male C57BL/6 mice were exposure to long-term isoflurane to induce cognitive impairment. To simulate an in vitro isoflurane-induced cognitive dysfunction‐like condition, primary mouse astrocytes were subjected to long-term isoflurane exposure. Cognitive function was assessed by Y-maze and fear conditioning tests. Western blot was used to determine the expression levels of different functional configurations of Cx43. The morphology of the GJs-Cx43 was evaluated by immunofluorescence staining. Levels of IL-1β and IL-6 were examined by ELISA. The ability of GJs-Cx43-mediated intercellular communication was examined by lucifer yellow dye transfer assay. Ethidium bromide uptake assays were used to measure the activity of Cx43 hemichannels. The ultrastructural morphology of astrocyte gap junctions and tripartite synapse were observed by transmission electron microscopy. Results After long-term isoflurane anesthesia, the GJs formed by Cx43 in the mouse hippocampus and primary mouse astrocytes were significantly reduced, GJs function was impaired, hemichannel activity was enhanced, the levels of IL-1β and IL-6 were increased, and mice showed significant cognitive impairment. After treatment with the novel GJ-Cx43 enhancer ZP1609, GJ-Cx43-mediated astrocytic network function was enhanced, neuroinflammation was alleviated, and ameliorated cognition dysfunction induced by long-term isoflurane exposure. However, ZP1609 enhances the astrocytic network by promoting Cx43 to form GJs without affecting hemichannel activity. Additionally, our data showed that long-term isoflurane exposure does not alter the structure of tripartite synapse. Conclusion Our results reveal a novel mechanism of the GJ-Cx43-mediated astrocytic network involved in isoflurane-induced neuroinflammation and cognitive impairments, which provides new mechanistic insight into the pathogenesis of POCD and identifies potential targets for its treatment.

Abstract The large energy loss (Eloss) is one of the main obstacles to further improve the photovoltaic performance of organic solar cells (OSCs), which is closely related to the charge transfer (CT) state. Herein, ternary donor alloy strategy is used to precisely tune the energy of CT state (ECT) and thus the Eloss for boosting the efficiency of OSCs. The elevated ECT in the ternary OSCs reduce the energy loss for charge generation (ΔECT), and promote the hybridization between localized excitation state and CT state to reduce the nonradiative energy loss (ΔEnonrad). Together with the optimal morphology, the ternary OSCs afford an impressive power conversion efficiency of 19.22% with a significantly improved open‐circuit voltage (Voc) of 0.910 V without sacrificing short‐cicuit density (Jsc) and fill factor (FF) in comparison to the binary ones. This contribution reveals that precisely tuning the ECT via donor alloy strategy is an efficient way to minimize Eloss and improve the photovoltaic performance of OSCs.

Recently, multiscale three-dimensional (3D) structures consisting of micrometer-scale structure and nanometer-scale structure have received some attention from scientists in the field of surface-enhanced Raman scattering (SERS). In this work, micrometer-scale grating structure and nanometer-scale zinc oxide nano spikes (ZnO NSs) structure are successfully introduced into the SERS substrate with silver nanoparticles (Ag NPs) as the surface plasmon. The optimized particle-in-multiscale 3D substrate (PDMS/grating/ZnO NSs/Ag NPs) presents high sensitivity with an ultralow limit of detection of 1 × 10−11 M and a high enhancement factor of 7.0 × 108 for Rhodamine 6G (R6G) as the probe molecule. It benefits from the electromagnetic field enhancement from the excellent optical capture capability of grating/ZnO NSs structure and abundant electromagnetic hot spots. The quantitative analysis ability of the SERS substrate can be indicated from the good linear correlation between the logarithmic Raman intensity and the molecular concentration. At the same time, this SERS substrate exhibits excellent homogeneity and reproducibility, which have low relative standard deviations (4.43%) of the Raman intensities at 613 cm−1 peaks for R6G as the probe molecule. In addition, this SERS substrate can realize in-situ detection of Raman signal due to its excellent light transmission and flexibility. The particle-in-multiscale 3D structure as SERS substrate exhibits the vast potential in practical applicability for qualitatively and quantitatively chemical and biomedical analysis.

The power conversion efficiencies (PCEs) of organic photovoltaics (OPVs) have reached more than 19%, along with the prosperous development of materials and device engineering. It is meaningful to make a comprehensive review of the research of OPVs for further performance improvement. In this review, some typical materials of high-performance OPVs are summarized, including representative polymer donor materials, non-fullerene acceptor materials, and interfacial modification materials, as well as their design rules for molecular engineering. From the point of view of device engineering, active layer treatment and deposition technology are introduced, which can play a critical role in adjusting the degree of molecular aggregation and vertical distribution. Meanwhile, a ternary strategy has been confirmed as an efficient method for improving the performance of OPVs, and the multiple roles of the appropriate third component in the photo-electronic conversion process are emphasized and analyzed. The challenges and perspectives concerning this region are also put forward for further developing high-performance OPVs.

BackgroundLiver hepatocellular carcinoma (LIHC), one of the most common primary malignancies, exhibits high levels of molecular and clinical heterogeneity. Increasing evidence has confirmed the important roles of some RNA helicase families in tumor development, but the function of the DEAH-box RNA helicase family in LIHC therapeutic strategies has not yet been clarified.MethodsThe LIHC dataset was downloaded from The Cancer Genome Atlas (TCGA). Consensus clustering was applied to group the patients. Least absolute shrinkage and selection operator Cox regression and univariate and multivariate Cox regression were used to develop and validate a prognostic risk model. The Tumor Immune Estimation Resource and Tumor Immune Single Cell Hub databases were used to explore the role of DEAH-box RNA helicases in LIHC immunotherapy. In vitro experiments were performed to investigate the role of DHX9 in LIHC radiosensitivity.ResultsTwelve survival-related DEAH-box RNA helicases were identified. High helicase expression levels were associated with a poor prognosis and clinical features. A prognostic model comprising six DEAH-box RNA helicases (DHX8, DHX9, DHX34, DHX35, DHX38, and DHX57) was constructed. The risk score of this model was found to be an independent prognostic indicator, and LIHC patients with different prognosis were distinguished by the model in the training and test cohorts. DNA damage repair pathways were also enriched in patients with high-risk scores. The six DEAH-box RNA helicases in the risk model were substantially related to innate immune cell infiltration and immune inhibitors. In vitro experiments showed that DHX9 knockdown improved radiosensitivity by increasing DNA damage.ConclusionThe DEAH-box RNA helicase signature can be used as a reliable prognostic biomarker for LIHC. In addition, DHX9 may be a definitive indicator and therapeutic target in radiotherapy and immunotherapy for LIHC.

To prolong the shelf life of perishable food with a simple and environmentally friendly postharvest preservation technology is one of the global concerns. This study aimed to explore the application value of biological macromolecule natural cellulose nanofibers (CNFs) in extending the postharvest fruit shelf life. In this study, 0.5% (wt%) CNFs were prepared from natural wood and coated on the surface of early-ripening apple fruits. After 10 days of storage at room temperature, the results revealed that the shelf life of apple fruits with CNF coating was significantly prolonged, and the fruit appearance quality improved. The invisible network structure of CNFs in the fruit epidermis was observed under an atomic force microscope (AFM). The gas chromatography and mass spectrometry (GC-MS) analysis showed that CNFs significantly promoted the formation of epidermal wax, especially fatty alcohols, during storage. In addition, the CNFs remarkably promoted the upregulation of genes related to the synthesis of cuticular wax of apple. In conclusion, this study provides an environmentally sustainable nanomaterial for post-harvest preservation of horticultural products, and also provides a new insight into the effect of CNFs on postharvest storage of apple fruits.

PURPOSEWe aimed to explore the feasibility and clinical effectiveness of percutaneous CT-guided iodine-125 (¹²⁵I) brachytherapy combined with chemotherapy for the treatment of patients with unresectable or locally advanced pancreatic carcinoma (PC).METHODSWe retrospectively reviewed 66 patients with Stage III and IV PC who had received chemotherapy. A total of 35 (53%) patients receiving 125I brachytherapy and chemotherapy (gemcitabine + cisplatin, GP) were classified as Group A, and 31 (47%) patients who received GP chemotherapy alone were categorized as Group B. The evaluated indications were local control rate (LCR), local progression-free survival (LPFS), overall survival (OS), treatment-related complications, and the degree of symptom relief. Kaplan-Meier curves, log-rank test and Cox regression models were generated and used for further analysis to identify predictors of outcomes.RESULTSThe median follow-up time was 6.00±0.84 months. The 1-, 3-, 6-, 12- and 18-month LCRs for Group A were 100% (35/35), 89.3% (25/28), 71.4% (15/21), 37.5% (3/8) and 33.3% (1/3), respectively; and those for Group B were 87.1% (27/31), 69.6% (16/23), 41.2% (7/17), 14.3% (1/7) and 0% (0/3), respectively. The LCR differed at 1-, 3- and 6-months (P = 0.032; P = 0.009; P = 0.030; respectively). The median LPFS was 7.00±0.30 months and 5.00±0.75 months for Groups A and B (P = 0.023), respectively; however, the median OS of the groups were not significantly different (8.00±0.77 months vs. 6.00±1.04 months. P = 0.917). No life-threatening complications occurred during or after the procedures. Patients in Group A experienced better pain control and relief of abdominal distension than those in Group B.CONCLUSIONCT-guided 125I brachytherapy is a feasible, safe, and valuable treatment for patients with unresectable PC.

The application of additive manufacturing in the field of aviation hydraulics greatly improves the design freedom of hydraulic valve internal flow channels. Pressure loss in hydraulic valve internal flow channels is a primary factor that designers need to consider, and the rapid prediction of pressure loss is very helpful for flow channel design. At present, most studies only focus on how much the pressure loss in an additive manufacturing (AM) hydraulic channel is reduced compared with an original hydraulic channel, and a mathematical model of pressure loss in an AM curved channel is still lacking. In this paper, the pressure loss in a curved flow channel was firstly studied, and the main parameters affecting the pressure loss were determined using the dimensionless analysis method. Using computational fluid dynamics simulation, the relationships between the flow channel diameter, the flow channel length, the flow channel curvature radius, the fluid velocity and pressure loss were studied. According to the multiple regression analysis method, the mathematical model of pressure loss in aviation hydraulic channels was developed, and the model was solved based on the orthogonal experimental results. The pressure loss in the flow channel samples fabricated using selective laser melting was tested, and the results showed that the average error between the test results and the mathematical model calculation results was 7.72%. This model can be used to quickly predict the pressure loss in curved flow channels in the aviation hydraulic field.

Osteoarthritis (OA) is a degenerative disease of articular cartilage that is mainly characterized by chronic and mild inflammation of the joints. Recently, many studies have reported the crucial roles of long noncoding RNAs (lncRNAs) in OA as gene transcriptional regulatory factors, diagnostic biomarkers, or therapeutic targets. However, the exact mechanisms of lncRNAs in the regulation of OA progression remain unclear. In the present study, the lncRNA WDR11 divergent transcript (lncRNA WDR11-AS1) was shown to be downregulated in osteoarthritic cartilage tissues from patients, and to promote extracellular matrix (ECM) synthesis in osteoarthritic chondrocytes with knockdown and overexpression experiments. This function of lncRNA WDR11-AS1 was linked to its ability to interact with the polyadenylate-binding protein cytoplasmic 1 (PABPC1), which was screened by RNA pulldown and mass spectrometry analyses. PABPC1 was discovered to bind ECM-related mRNAs such as SOX9, and the inhibition of PABPC1 improved the mRNA stability of SOX9 to mitigate OA progression. Our results suggest that lncRNA WDR11-AS1 has a promising inhibitory effect on inflammation-induced ECM degradation in OA by directly binding PABPC1, thereby establishing lncRNA WDR11-AS1 and PABPC1 as potential therapeutic targets in the treatment of OA.

Abstract Background Immune-related genes is closely related to the occurrence and prognosis of head and neck squamous cell carcinoma (HNSCC). At the same time, immune-related genes have great potential as prognostic markers in many types of cancer. The prognosis of HNSCC is still poor currently, and it may be effective to predict the clinical outcome of HNSCC by immunogenic analysis. Methods RNASeq and clinical follow-up information were downloaded from The Cancer Genome Atlas (TCGA), the MINiML format GSE65858 chip expression data was downloaded from NCBI, and immune-related genes was downloaded from the InnateDB database. Immune-related genes in 519 HNSC patients were integrated from TCGA dataset. By using multivariate COX analysis and Lasso regression, robust immune-related gene pairs (IRGPs) that predict clinical outcomes of HNSCC were identified. Finally, a risk prognostic model related to immune gene pair was established and verified by clinical features, test sets and GEO external validation set. Results A total of 699 IRGPs were significantly correlated with the prognosis of HNSCC patients. Fourteen robust IRGPs were finally obtained by Lasso regression and a prognostic risk prediction model was constructed. Risk score of each sample were calculated based on Risk models and divided into the high-risk group (Risk-H) and low Risk group (Risk-L). Risk models were able to stratify the risk in patients with TNM Stage, Age, gender, and smoking history, and the AUC > 0.65 in training set and test set, shows that 14-IRGPs signature in patients with HNSCC has excellent classification performance. In addition, 14-IRGPs had the highest average C index compared with the prognostic characteristics and T, N, and Age of the 3 previously reported HNSCC. Conclusion This study constructed 14-IRGPs as a novel prognostic marker for predicting survival in HNSCC patients.

Based on data analysis of 9649 Chinese primary NSCLC patients, we calculated the exact proportion of EGFR subtypes in NSCLC and evaluated the TMB level, PD-L1 expression level and tumor immune microenvironment among different EGFR mutation subtypes. Postoperative follow-up data for 98 patients were collected and analyzed. The results showed that several uncommon EGFR mutation subtypes have a higher proportion of TMB-high or strong positive PD-L1 expression than the total EGFR mutation group. In addition, different subtypes have different characteristics related to the immune microenvironment, such as G719 mutations being associated with more CD8+ T cell infiltration into tumors; except for EGFR 19del, CD8+ T cell infiltration into tumors of other EGFR mutation subtypes were similar to that of wildtype EGFR. Moreover, follow-up results revealed that components of the immune microenvironment have prognostic value for NSCLC patients, with different prognostic biomarkers for NSCLC patients with and without EGFR mutations. These results suggest that patients with different EGFR mutations need to be treated differently. The prognosis of NSCLC patients may be assessed through components of tumor immune microenvironment, and ICIs treatment may be considered for those with some uncommon EGFR mutation subtypes.

The dielectric permittivity of common soils is mainly controlled by water content and porosity, while the latter is closely related to the characteristics of compaction. By studying the changes in dielectric permittivity of soil samples with different soil water content and compaction levels, the influence of the controlling factors on the relationship model between soil water content and dielectric permittivity can be evaluated. In this paper, network analyzer was used to measure the dielectric permittivity of 7 groups of soil samples with gravimetric water content ranging from 8.09% to 14.52% and dry density ranging from 1.61 g/cm3 to 1.96 g/cm3. The results show that the dielectric permittivity increases with the increase of water content and dry density, and the effect of water content on permittivity is more significant for soils with higher dry density. Furthermore, when the water content is less than or equal to the optimal water content, Topp formula and the complex refractive index model (CRIM) can better predict the soil dry density. When the water content approaches the saturated state of soil, there is a deviation between the predicted value and the actual value. At last, the modified Topp formula and the complex refractive index model (CRIM) can accurately predict soil compactness. This provides an important basis for rapid detection of water content and compactness of highway subgrade soil by ground penetrating radar.

Abstract Objective The disruption of metabolic events and changes to nutrient and oxygen availability due to sustained inflammation in RA increases the demand of bioenergetic and biosynthetic processes within the damaged tissue. The current study aimed to understand the molecular mechanisms of SLC7A5 (amino acid transporter) in synoviocytes of RA patients. Methods Synovial tissues were obtained from OA and RA patients. Fibroblast-like synoviocytes (FLS) were isolated, and SLC7A5 expression was examined by using RT-qPCR, immunofluorescence, and Western blotting. RNAi and antibody blocking treatments were used to knockdown SLC7A5 expression or to block its transporter activities. mTOR activity assay and MMP expression levels were monitored in RA FLS under amino acid deprivation or nutrient-rich conditions. Results RA FLS displayed significantly upregulated expression of SLC7A5 compared to OA FLS. Cytokine IL-1β was found to play a crucial role in upregulating SLC7A5 expression via the NF-κB pathway. Intervening SLC7A5 expression with RNAi or blocking its function by monoclonal antibody ameliorated MMP3 and MMP13 protein expression. Conversely, upregulation of SLC7A5 or tryptophan supplementation enhanced mTOR-P70S6K signals which promoted the protein translation of MMP3 and MMP13 in RA FLS. Conclusion Activated NF-κB pathway upregulates SLC7A5, which enhances the mTOR-P70S6K activity and MMP3 and MMP13 expression in RA FLS.

This study was designed to explore the clinical efficacy and prognostic factors of CT-guided 125I brachytherapy for the treatment of retroperitoneal metastatic lymph nodes. Methods We retrospectively evaluated 92 patients received 125I brachytherapy for retroperitoneal metastatic lymph nodes. Neutrophil cytosolic factor 1 (Ncf1) is a major genetic factor associated with autoimmune diseases and has been identified as a key player in autoimmune mediated inflammation. We addressed the role of Ncf1 in an antigen-induced pulmonary inflammation model, and found that the Ncf1m1j mutation, causing a deficient reactive oxygen species response, alleviated disease. The Ncf1m1j mutation was associated with a reduced inflammatory cell infiltration in airways, but had limited effect on mucus secretion, antibody production and lung fibrosis. The disease remission in the Ncf1 mutated mice was reversed when functional Ncf1 was transgenically expressed in alveolar macrophages, suggesting that the cellular inflammation was depended on functional Ncf1 in alveolar macrophages. By determining cytokine and chemokine profiles in lung and serum, we found that Ncf1 deficiency allowed an increased expression of Th1 cytokines, including TNF-α, IFN-γ and IL-12. Since also epithelial cytokines were found to be regulated by Ncf1, we tested the effect of Ncf1 in IL-33 and IL-25 induced lung inflammation models. Mice with the Ncf1m1j mutation showed less sensitivity to IL-33, but not IL-25, induced lung inflammation, in a macrophage independent manner. The mice with deficient Ncf1 showed a reduced eosinophil infiltration and group 2 innate lymphoid cell (ILC2) activation. The production of IFN-γ in CD4+ T cells was increased, whereas IL-5 and IL-13 in ILC2 were decreased. Importantly, anti-IFN-γ antibody treatment of Ncf1 deficient mice increased eosinophil infiltration and rescued ILC2 activation in the lung. We conclude that Ncf1 deficiency enhances Th1 response, deactivates ILC2, and protects against pulmonitis.

BackgroundAlthough the effect of the LEP G19A (rs2167270) polymorphism on cancers is assumed, the results of its influence have been contradictory. A meta-analysis was conducted to precisely verify the relationships between LEP G19A and the development of digestion-related cancers.MethodsInvestigators systematically searched the literature in PubMed, Embase, and Web of Science and used STATA software 14.0 for the meta−analysis. The odds ratios (ORs) and 95% confidence intervals (CIs) were calculated to evaluate the associations. Subgroup analyses stratified by ethnicity, cancer type, and cancer system were further conducted to assess the relationship between the LEP G19A polymorphism and digestion-related cancers.ResultsIn the overall population, we found a significant relationship with overall cancer (allele comparison: OR = 0.921, p = 0.000; dominant comparison: OR = 0.923, p = 0.004; recessive comparison: OR = 0.842, p = 0.000; homozygote model: OR = 0.0843, p = 0.001). In a subgroup analysis conducted by ethnicity, we obtained significant results in Asians (Asian allele comparison: OR = 0.885, p = 0.000; dominant comparison: OR = 0.862, p = 0.000; homozygote model: OR = 0.824, p = 0.039; and heterozygote comparison: OR = 0.868, p = 0.000) but not in Caucasians. In a subgroup analysis conducted by cancer type and cancer system, we obtained significant results that the LEP G19A polymorphism may decrease the risk of colorectal cancer, esophageal cancer, digestive system cancer, and urinary system cancer.ConclusionsThis meta-analysis revealed that the LEP G19A polymorphism may decrease the risk of cancer.

The nitrogen content of electric arc furnace (EAF) steel is much higher than that of basic oxygen furnace (BOF) steel, which cannot meet the requirements of high-grade steel. Most denitrification processes only considered a single smelting condition, which leads to poor denitrification effect. In this study, a hot state experiment was conducted to simulate the melting process of EAF steelmaking and to explore the thermodynamic and kinetic constraints of the molten steel nitrogen reaction in the scrap melting, oxygen blowing decarburization, and rapid temperature rise stages. The experimental results showed that the nitrogen reaction in the molten pool during the scrap melting stage was a first-order nitrogen absorption reaction, and the reaction-limiting link was the diffusion of nitrogen atoms in the molten steel. When the carbon content increases to 4.5%, the bath temperature decreases to 1550 °C, and the nitrogen partial pressure decreases to 0.2 PΘ, the nitrogen saturation solubility decreased to 0.0198%, 0.0318%, and 0.0178%, respectively. At the same time, the rate constants decreased to 0.132 m/min, 0.127 m/min, and 0.141 m/min, respectively. The nitrogen reaction in the oxygen blowing decarburization stage was a secondary denitrification reaction, and the reaction-limiting link was the gas–liquid interface chemical reaction. Argon had better degassing effect. When the argon flow rate increased from 100 mL/min to 300 mL/min, the reaction constant increased by about four times. When the oxygen content of molten steel was 0.0260%, the denitrification rate constant decreased by about 2.5 times. The nitrogen content of liquid steel was higher than 0.045%, and the reaction was a secondary reaction. As the nitrogen content decreased, the reaction rate decreased, and the reaction-limiting link changed from the gas–liquid interface chemical reaction to the joint control of mass transfer and chemical reaction. The oxygen content in the molten steel can not only hinder the chemical reaction of nitrogen at the gas–liquid interface, but also reduce the mass transfer rate of nitrogen atoms in the molten steel. The results provided a theoretical basis for the optimization of nitrogen removal process and further reduction of nitrogen content in liquid steel.

Accurate state of charge (SoC) estimation is crucial for the safe and reliable running of lithium-ion batteries in electrified transportation equipment. To enhance the estimation accuracy and robustness under different ambient temperatures, H∞ and the adaptive H∞ filterings were first combined to simultaneously forecast the parameters and SoC of the battery model considering the hysteresis effect in this paper. To drop the computational complexity to the most extent, the hysteresis unit was integrated into the first-order RC battery model and the aforementioned combined algorithm was developed under a dual-time frame. Then, the battery model with the hysteresis effect is evaluated against the model without that in terms of the estimation accuracy. Subsequently, the proposed algorithm is compared with the dual H∞ algorithm based on the employed battery model. The results demonstrate the excellent performance of the utilized battery model and the proposed algorithm in terms of both the estimation accuracy and the convergence speed.

In China, the majority of patients with hepatocellular carcinoma (HCC) result from long-term infection of hepatitis B. Pathologically, HCC is characterized by rich blood supply, multicentric origins, early vascular invasion and intrahepatic metastasis. Therefore, HCC is not a local disease but a systemic disease at the beginning of its occurrence. For this reason, a comprehensive treatment strategy should be adopted in the management of HCC, including local treatments (such as surgical resection, radiofrequency ablation, microwave ablation, chemical ablation and cryoablation, etc.), organ-level treatments [such as transcatheter arterial infusion of chemotherapy and transcatheter arterial chemoembolization (TACE)], and systemic treatments (such as immunotherapy, antiviral therapy and molecular targeted therapy, etc.). This consensus sets forth the minimally-invasive and multidisciplinary comprehensive guideline of HCC, focusing on the following eight aspects (1) using hepaticarteriography, CT hepatic arteriography (CTHA), CT arterial portography (CTAP), lipiodol CT (Lp-CT), TACE-CT to find the intrahepatic lesion and make precise staging (2) TACE combined with ablation or ablation as the first choice of treatment for early stage or small HCC, while other therapies are considered only when ablation is not applicable (3) infiltrating HCC should be regarded as an independent subtype of HCC (4) minimally-invasive comprehensive treatment could be adopted in treating metastatic lymph nodes (5) multi-level subdivision of M-staging should be used for individualized treatment and predicting prognosis (6) HCC with severe hepatic decompensation is the only candidate criterion for liver transplantation (7) bio-immunotherapy, traditional Chinese medicine therapy, antiviral therapy, and psychosocial and psychopharmacological interventions should be advocated through the whole course of HCC treatment (8) implementation of multicenter randomized controlled trials of minimally-invasive therapy versus surgery for early and intermediate stage HCC is recommended.

Interface modification is an important way to get better performance from organic solar cells (OSCs). A natural biomolecular material methionine was successfully applied as the electron transport layer (ETL) to the inverted OSCs in this work. A series of optical, morphological, and electrical characterizations of thin films and devices were used to analyze the surface modification effects of methionine on zinc oxide (ZnO). The analysis results show that the surface modification of ZnO with methionine can cause significantly reduced surface defects for ZnO, optimized surface morphology of ZnO, improved compatibility between ETL and the active layer, better-matched energy levels between ETL and the acceptor, reduced interface resistance, reduced charge recombination, and enhanced charge transport and collection. The power conversion efficiency (PCE) of OSCs based on PM6:BTP-ec9 was improved to 15.34% from 14.25% by modifying ZnO with methionine. This work shows the great application potential of natural biomolecule methionine in OSCs.

Objective: Through experiments to testify a candidate novel miRNA previously discovered by us is a real miRNA and involved in cartilage development. Design: The miR-novel and the newly hairpin miRNA transcribed sequence (pre-miR-novel) was verified as a genuinely existing miRNA by northern blotting. The predicted secondary structure, sequence alignment and targets of pre-miR-novel were performed by “RNAstructure 5.3” program, LASTN2.8.0+/miRbase22 program and RNA hybird program, respective. GO/KEGG pathway analysis also were performed. The miR-novel expression in cartilage tissue during development was detected by RT-qPCR and dot blotting. The chondrocyte differentiation model was established to examine whether miR-novel is involved in cartilage development. The regulation of PRMT3 expression by novel miRNA was determined with the luciferase reporter gene assay and Western blotting after novel miRNA mimic or inhibitor transfection. Results: It’s potential role in specifically regulating rodent cartilage development and associated cellular processes. Furthermore, the expression of protein arginine N-methyltransferase 3 (PRMT3), as a predicted target of the novel miRNA, was found consistently downregulated at rat cartilage during developmental stages and RCJ3.1C5.18 (C5.18) cells during the proliferating and hypertrophic phases of the cartilage development, where the miR-novel expression was significantly up-regulated. Both the dual-luciferase reporter gene assay and the up- or down-regulation of miR-novel suggest that the later can specifically bind with the Prmt3 3′-UTR. Conclusion: Overall, this study provides the first comprehensive evidence that a genuine cartilage-specific novel miRNA directly targets PRMT3 and may regulate multitudinous cellular processes and signal transduction during cartilage development.

Accurate open-circuit voltage (OCV) is crucial for state of charge (SoC) estimation of lithium-ion batteries and, hence has become a key factor to ensure the safety and reliability of electric vehicles (EVs). In engineering, the incremental OCV (IO) testing has been widely used for OCV calibrating. Based on this, the OCV is commonly simplified by averaging the discharging and the charging OCVs, which essentially ignores the influence of the major hysteresis (MH). By a series of experiments on the LiNiMnCoO2 battery, this work first systematically investigated the influence of the MH on SoC estimation via diverse current profiles tested at various ambient temperatures. Besides, the recursive least square (RLS) and the particle filter (PF) algorithms were introduced to estimate the battery parameters and the SoC, respectively. The results report that, compared with the traditional simplified method, the discharging direction and the charging direction of the MH can enhance the estimation accuracy of the discharging process and the charging process of the battery at all the operating conditions above, respectively. By the MH-based estimation method, the maximum mean absolute estimation error can be reduced by about 70%.

Abstract Iodine‐125 (125I) brachytherapy, a promising form of radiotherapy, is increasingly applied in the clinical treatment of a wide range of solid tumors. However, the extremely hypoxic microenvironment in solid tumors can cause hypoxia‐induced radioresistance to 125I brachytherapy, resulting in therapeutic inefficacy. In this study, the aim is to sensitize hypoxic areas in solid tumors using ultrasound‐activated oxygen microbubbles for 125I brachytherapy. A modified emulsion freeze‐drying method is developed to prepare microbubbles that can be lyophilized for storage and easily reconstituted in situ before administration. The filling gas of the microbubbles is modified by the addition of sulfur hexafluoride to oxygen such that the obtained O2/SF6 microbubbles (OS MBs) achieve a much longer half‐life (>3×) than that of oxygen microbubbles. The OS MBs are tested in nasopharyngeal carcinoma (CNE2) tumor‐bearing mice and oxygen delivery by the OS MBs induced by ultrasound irradiation relieve hypoxia instantly. The post‐treatment results of brachytherapy combined with the ultrasound‐triggered OS MBs show a greatly improved therapeutic efficacy compared with brachytherapy alone, illustrating ultrasound‐mediated oxygen delivery with the developed OS MBs as a promising strategy to improve the therapeutic outcome of 125I brachytherapy in hypoxic tumors.

Purpose:125I seeds were effective in the treatment of non-small cell lung cancer in previous research. However, the exact signaling pathway-mediated apoptosis mechanism is still unclear. The present study analyzed the effects and potential mechanisms of 125I seed on the growth and migration of A549 cells.Methods: Lung cancer A549 cells were irradiated with 125I seed for various times. MTT, invasion assay, and flow cytometry were used to detect the proliferation, invasion, and apoptosis of treated cells, respectively. A Nimblegen genome-wide expression profile chip was used to evaluate gene expression changes in 125I seed-treated A549 cells. Validation studies were performed using phosphorylated protein chip technology, Western blot, nude mouse tumor xenograft assay, and immunohistochemical experiments. All statistical analyses were performed using unpaired Student's t tests and Kruskal-Wallis test.Results: Irradiation with 125I seed inhibited A549 cell proliferation and invasion and induced apoptosis (primarily early apoptosis). Irradiation with 125I seed also caused the downregulation of p38MAPK, degradation of mouse double-minute 2 homolog (MDM2), and higher expression of p53, which eventually resulted in non-small cell lung cancer cell apoptosis.Conclusion:125I seed irradiation activated the p38MAPK/MDM2/p53 signaling pathway and promoted non-small cell lung cancer cell apoptosis. Future clinical studies targeting this signal may provide a new potential therapeutic approach for non-small cell lung cancer.

Surgery remains the first choice of cure for early stage lung cancer. However, many patients are diagnosed at advanced stage, and thus miss the opportunity to undergo surgery. As such patients derive limited benefits from chemotherapy or radiotherapy, alternatives focusing on local control have emerged, including iodine‐125 seed implantation. The Interstitial Brachytherapy Society, Committee of Minimally Invasive Therapy in Oncology, Chinese Anti‐Cancer Association organized a group of multidisciplinary experts to develop guidelines for this treatment modality. These guidelines aim to standardize iodine‐125 seed implantation procedures, inclusion criteria, and outcome assessment to prevent and manage procedure‐related complications.

The high temperature brittleness range of medium carbon microalloyed steel under an actual continuous casting process was determined by the high temperature tensile test. The test results revealed that only a third of the brittle temperature range from 650–825 °C was due to intergranular ferrite in the experimental steel. In addition, it was found that the plastic recovery was fast and stable when the temperature was lower than 725 °C (the lowest plastic temperature). Bending/straightening operation in this temperature range was conducive to controlling the generation of corner cracks. In order to keep the corner temperature at the low temperature end of the plastic curve when the slab was bent/straightened, the cooling water scheme of the secondary cooling zone of the continuous caster was formulated by numerical calculation. By appropriately increasing the cooling water flow in the foot roll and the secondary cooling zones 1–5, the corner temperature of slab during bending operation was 600–700 °C, avoiding the brittle temperature range. The industrial test was then carried out. The results showed that after using the optimized water volume, the corner grains of the slab were uniform and the microstructure was mainly pearlite + ferrite. In addition, the abnormally large grain size was reduced, and a large amount of ferrite was generated inside the grain, which avoided stress concentration at the corner of the slab during bending/straightening operation, and basically eliminated the corner crack of the slab.

Abstract Background The phenotypes of osteoarthritis (OA) consist of cartilage extracellular matrix (ECM) metabolism disorder and the breakdown of cartilage homeostasis, which are induced by pro-inflammatory factors and oxidative stress. Selenoproteins regulated by selenocysteine insertion sequence binding protein 2 (SBP2) are highly effective antioxidants, but their regulatory mechanisms, particularly the involvement of miRNAs, are not fully understood. Methods To explore whether miR-181a-5p and SBP2 are involved in OA pathogenesis, we established an IL-1β model using the chondrocyte SW1353 cell line. Next, we up- or down-regulated SBP2 and miRNA-181a-5p expression in the cells. Finally, we measured the expression of miRNA-181a-5p, SBP2 and three selenoproteins in OA cartilage and peripheral blood. Results The results showed that IL-1β increased hsa-miR-181a-5p and decreased SBP2 in a time- and dose-dependent manner. GPX1 and GPX4, which encode crucial glutathione peroxidase antioxidant enzymes, were up-regulated along with SBP2 and miR-181a-5p. Furthermore, SBP2 showed a significant negative correlation with miR-181a-5p during induced ATDC5 cell differentiation. There was lower GPX1 and GPX4 mRNA expression and SBP2 protein expression in damaged cartilage than in smooth cartilage from the same OA sample, and hsa-miR-181a-5p expression on the contrary. Similar results were observed in peripheral blood. In conclusion, we have reported a novel pathway in which pro-inflammatory factors, miRNA, SBP2 and selenoproteins are associated with oxidation resistance in cartilage. Conclusion Overall, this study provides the first comprehensive evidence that pro-inflammatory factors cause changes in the cartilage antioxidant network and describes the discovery of novel mediators of cartilage oxidative stress and OA pathophysiology. Our data suggest that miR-181a-5p may be used to develop novel early-stage diagnostic and therapeutic strategies for OA.

Background: Currently, hepatocellular carcinoma (HCC) patients with refractory ascites (RA) have a very poor prognosis, and there are no effective treatments recommended by the guidelines. A treatment strategy that utilizes a transjugular intrahepatic portosystemic shunt (TIPS) combined with subsequent antitumor treatment is explored in this study for its feasibility and clinical value. Methods: One month after TIPS, the ascites grade and Child-Pugh scores and stages were reassessed to compare changes in the preoperative indicators. Results: A total of 68 patients from 3 centers were enrolled. After TIPS, the following results were obtained: a complete response (CR), partial response (PR), or absent RA response (AR) of 38 [55.9%], 21 [30.9%], and 9 [13.2%], respectively. The control of RA was 86.8%. The median Child–Pugh scores prior to TIPS and one month after TIPS were 8 (IQR 7–9) and 7 (IQR 6–8), respectively. The down, unchanged, and elevated Child–Pugh stages were 26 [38.2%], 36 [53.0%], and 6 [8.8%], respectively. The postoperative Child–Pugh scores were significantly lower than the preoperative (p
