Aim Tamoxifen (TAMO) is a chemotherapeutic drug used for the treatment of breast cancer. Nevertheless, there is a lack of information available in regarding its nephrotoxicity. The purpose of this work was to investigate the impact of cyanocobalamin (COB) and/or calcitriol (CAL) injections on TAMO-induced nephrotoxicity. Main methods Animals were allocated into five groups as follows: normal control group; TAMO (45 mg/kg) administered group; TAMO+COB (6 mg/kg, i.p) treated group; TAMO+CAL (0.3 μg/kg, i.p) treated group; TAMO+COB+CAL combination groups. Key findings Renal injury induced by TAMO was confirmed by the alteration in renal function parameters in the serum (urea and creatinine), as well as in the urine (creatinine clearance, total protein and albumin). These results were supported by histopathological examination. Upregulation of renal inflammatory parameters; tumor necrosis factor (TNF)-α, interleukin (IL)-6, C-reactive protein (CRP); and transforming growth factor (TGF)-β1 as well as in protein expression of nuclear factor-kappa B (NF-κB) and cleaved caspase-3 were observed to a greater extent in the TAMO-treated rats compared with the control. Renal fibrosis was also evidenced by a elevation in renal L-hydroxyproline level as well as by histomorphological collagen deposition in TAMO-treated groups compared to the control group. Administration of COB and/or CAL concurrently with TAMO significantly ameliorated the deviation in the above-studied parameters and improved the histopathological renal picture. Significance Inhibition of NF-κβ-mediated inflammation and caspase-3-induced apoptosis are possible renoprotective mechanisms of COB and/or CAL against TAMO nephrotoxicity, which was more noticeable in the TAMO group treated with the combination of the two vitamins in question.

The study aims to compare, through histological and biochemical studies, the effects of quercetin, melatonin and their combination in regulation of immuno-inflammatory mediators and heat shock protein expressions in sodium nitrite induced hypoxia in rat lungs. The results revealed that NaNO2 injection caused a significant decrease in Hb in rats, while serum levels of TNF-α, IL-6 and CRP, VEGF and HSP70 were elevated compared to the control group. Administration of melatonin, quercetin or their combination before NaNO2 injection markedly reduced these parameters. Histopathological examination of the lung tissue supported these biochemical findings. The study suggests that melatonin and/or quercetin are responsible for lung tissue protection in hypoxia by downregulation of immuno-inflammatory mediators and heat shock protein expressions. Pre-treatment of hypoxic animals with a combination of melatonin and quercetin was effective in modulating most of the studied parameters to near-normal levels.

Diabetic nephropathy (DN) is the most critical diabetic complication that leads to renal failure. The purpose of this research was to explore the anti-diabetic effectiveness of Cucumis melo var. flexuosus and/or Phoenix dactylifera fruit aqueous extracts and their mechanisms in alleviating nephropathy in hyperglycemic rats. Diabetes was promoted in rats by injection (i.p.) of a single dose of streptozotocin (STZ). C. flexuosus and P. dactylifera extracts (CFE and PDE respectively) were ingested to diabetic rats for thirty consecutive days. The data revealed that intake of either plant extract or their combination to diabetic rats, significantly diminished the serum glucose and raised the serum insulin concentration. The plant extracts significantly ameliorated the increases in relative kidney weights as well as in renal tumor necrosis factor (TNF-α), serum C –reactive protein (CRP) and renal vascular endothelial factor (VEGF). Serum kidney function markers (creatinine, uric acid and urea) were also decreased in diabetic rats treated with the plant extracts. Histopathological observation was also carried out to confirm the biochemical results. This study has proven that the current plant extracts have potential hypoglycemic effect and could attenuate diabetic inflammatory renal damage in rats. The combination of the two extracts was synergistically the effective one. This result may help in exploring a novel therapy to manage diabetes and its complications.

Titanium dioxide nanoparticles (TiO2-NPs) are extensively used in a wide range of applications; however, many reports have investigated their nanotoxicological effect at the molecular level either in vitro or in vivo systems. The defensive roles of quercetin (Qur) or idebenone (Id) against the hepatotoxicity induced by TiO2-NPs were evaluated in the current study. The results showed that the coadministration of Qur or Id to rats intoxicated with TiO2-NPs markedly ameliorated the elevation in hepatic malondialdehyde (MDA), serum alanine amino-transferase (ALT), glucose, tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), immunoglobin G (IgG), and C-reactive protein (CRP) levels compared to their levels in TiO2-NPs-treated rats. The aforementioned antioxidants also effectively modulated the changes in the levels of serum vascular endothelial growth factor (VEGF), nitric oxide (NO), hepatic DNA breakage, caspase-3, and inhibition of drug metabolizing enzymes (cytochrome P450s; CYP4502E12E1) in rat livers induced by TiO2-NPs toxicity. The histopathological examination of the liver section showed that TiO2-NPs caused severe degeneration of most hepatocytes with an increase in collagen in the portal region, while treatment with the antioxidants in question improved liver architecture. These outcomes supported the use of Qur and Id as protective agents against the hepatotoxicity induced by TiO2-NPs and other hepatotoxic drugs.

The aim of this study was to investigate the effective role of silymarin either alone or in a combination with vitamin E and/or curcumin against the toxic impact of carbon tetrachloride (CCl4) induced liver injury The results revealed that administration of silymarin alone or in a combination with vitamin E and/or curcumin for 21 consecutive days, 24 h after CCl4 injection to rats, markedly ameliorated DNA damaged and apoptosis markers in rat livers, proinflammatory markers including tumor necrosis factor- α (TNF- α) and C-reactive protein (CRP ) in rat livers as well as interleukin 6 (IL-6), interferon gamma (IFN-γ) and vascular endothelial growth factor (VEGF) in rat sera. These treatments also could ameliorated the alteration in cytochrome P450 2E1 (CYP2E1) activity in livers of CCl4 intoxicated rats as well as the increase in the serum alanine aminotransferase (ALT) compared with CCl4 intoxicated untreated rats. The present biochemical results are supported by histo-pathological examination. In conclusion, silymarin in a combination with vitamin E and curcumin was the most effective treatment in alleviating CCl4 induced liver damage and this may support the use of this combination as an effective treatment against liver damage induced by toxic agents.

Although titanium dioxide nanoparticles (TDO-ns) are extensively used in the food, medicine, and cosmetic industries, discussions about the possible hazards of nanomaterials are just beginning to emerge. This study aimed to detect the inflammatory stress, oxidative stress, and apoptotic cell death induced in the livers of rats exposed to TDO-ns (600 mg/kg, particle size ≤ 100 nm). Furthermore, the modulation of these toxic effects by two potent naturally occurring antioxidants, carnosine (Carno) or melatonin (Melato), was evaluated. The co-administration of carnosine or melatonin to rats intoxicated with TDO-ns significantly attenuated the increases in serum tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), C-reactive protein (CRP), immunoglobulin G (IgG), vascular endothelial growth factor (VEGF), nitric oxide (NO), and alanine aminotransferase (ALT) levels. The two agents markedly ameliorated hepatic DNA damage and the alterations in hepatic malondialdehyde (MDA), glutathione (GSH), cytochrome P450, caspase-3, total phospholipid, phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, sphingomyelin, and triglyceride (TG) levels. These results support the use of Carno or Melato as prophylactic agents against TDO-ns-induced liver damage.

Traumatic brain injury in the pediatric population can have a great economic and emotional impact on both the child's family and society.The present study aimed to compare the effects of carnosine (CAR) and/or cyclosporine A (CyA) on oxidative brain damage after closed head injury (CHI) in immature rats.Thirty-day-old rat pups were divided into five groups: non-traumatic control group, trauma group underwent CHI, trauma group injected with CAR (200 mg/kg, i.p.) following CHI for 7 d, trauma group injected with CyA (20 mg/kg, i.p.) given 15 min and 24 h after CHI, and trauma group treated with CAR and CyA. At the end of the treatment, rats were sacrificed; blood and brains were collected for assessing different biochemical parameters.Trauma significantly increased brain level of malondialdehyde, nitric oxide, glucose, calcium, inflammatory mediators. Brain DNA damage was confirmed by comet assay and the significant increase in brain caspase-3 activity. Moreover, the serum level of Fas ligand in traumatized animals was significantly elevated. Concomitant decrease in brain-reduced glutathione (GSH) and calcium-adenosine triphosphatase activity was observed in the traumatized-untreated group. Treatment of traumatized animals with CAR and/or CyA ameliorated all the biochemical changes induced by CHI with marked protective effect in the combination group.CAR and CyA exerted a synergistic neuroprotective effect against CHI through blocking the induction of lipid peroxidation, reducing inflammatory, and oxidative stress biomarkers, preserving brain GSH content, and reducing the alterations in brain apoptotic biomarkers in traumatized animals.

Aim: The present work aimed to evaluate the effect of each of green tea (GT) and garlic (Ga) aqueous extract and their combination on pesticide Duracine induced changes in plasma levels of some indices of oxidative stress; and in protein electrophoretic patterns, in albino rats. Female Baladi goats were used to investigate the toxicological effects of 2,3,7,8 tetrachlorodibenzo-p-dioxin (TCDD) on some serum hormones andblood features. Animals were divided into two groups, group 1: served as control, group 2: animals were orallyadministeredwith three repeated doses (0.23µg/Kg body weight) of TCDD with 2 days intervalbetween dosing. Many plant-derived compounds have been used to treat microbial infections. Staphylococcus aureus a common cause of many organ infections, has generated increasing concern due to its resistance to antibacterial drugs. This work was carried out to explore the susceptibility of 6 strains (LN872136, LN872137, LN871238, LN871239, LN872140, and LN871241) of methicillin-resistant Staphylococcus aureus to aqueous extract of Lepidium sativum seeds in vitro. Various concentrations (5-20 mg/mL) were used to evaluate the effect of the extract on bacteria growth via the assessment of the microbial biomass and the inhibition zone (IZ). The results showed that the plant extract at 15 or 20 mg/mL, significantly decreased the the biomass of S aureus strains after 24 or 48 hours exposure period. Staphylococcus aureus (LN871241) showed the largest IZ at 20 mg/mL and documented by scanning electron microscope. The current work may suggest that L sativum seed extract can be candidate as a promising antimicrobial agent to treat infection with methicillin-resistant S aureus.

Despite effective chemotherapy, schistosomiasis remains the second major public health problem in the developing world, second to malaria. The present study was undertaken to explore the therapeutic effect of the micronutrients (vitamin E and selenium) and chloroform extract of Citharexylum quadrangular leaves and their mixture against the deleterious pathological impacts induced in mice livers by Schistosoma mansoni infection. Parasitological markers showed that oral ingestion of the different supplements to S. mansoni infected mice was effective in reducing the worm burden with concomitant decrease in the egg burden, granuloma count and its diameter as well as total area of infection in their livers versus untreated ones. Parasitological parameters were reflected by the improvement of the histopathological pictures of livers of infected-treated mice. The current investigation also showed that the used agents and their mixture successfully modulated liver fibrosis of infected mice which was documented by a marked decrease of liver hydroxyproline level ( as biomarker of liver fibrosis) as well as the serum levels of inflammatory fibrogenic mediators namely, fructosamine, tumor necrosis factor alpha (TNFα) and total immunoglobin E (IgE). In addition, the used agents showed ameliorative action on the elevated liver oxidative stress- nitric oxide (NO) and malondialdehyde (MDA) and the decreased antioxidant biomarkers, reduced glutathione (GSH), glutathione reductase (GR), thioredoxin reductase (TrxR) and catalase (CAT) which may have a role in liver damage and fibrosis due to S mansoni infection. In conclusion, treatment with the used micronutrients and plant extract either alone or in combination attenuated the deleterious impacts of S. mansoni infection on mice livers. The combination of all supplements was more effective as it greatly modified the inflammatory mediators and oxidative stress responsible for schistosomal liver fibrosis. Pyrethroids insectecides, permethrin(PM) and cypermethrin (CM) are used on a large scale inmany agricultural protection programs and may cause hazards for animal and human health. Theaim of the present study was to evaluate the hepatotoxicity of these pyrethroids on male albinorats in two foldes ; oxidative stress and antioxidant defence dysfunction. Rats received 1/60LD50 of PM (20.83 mg/kg b. wt., orally/ day, for 30 days) and CM (8.33 mg/kg b. wt., orally/day, for 30 days).Our results showed that serum hepatic function biomarkars, alanineaminotransferase (ALT) and aspartate aminotransferase (AST) enzymes, recorded a highlysignificant (P < 0.001) elevation in both PM and CM treated groups. Various anti-oxidants;reduced glutathione, glutathione reductase, glutathione peroxidase, catalase and superoxidedismutase enzymes, recorded highly significant (P < 0.001) reductions with both PM and CMtreatment. On the other hand, oxidative stress markers; xanthine oxidase (XO), lipidperoxidation (LPO) and advanced oxidation protein products (AOPPs) elevated in treatedanimals with PM or CM. In general, it is clear from our data that, CM is more effective thanPM. This may be expected with considering the presence of an alpha-cyno substituent in CMstructure which can potentaite its toxicity.

The aim of this study was to investigate the toxic impacts of titanium dioxide nanoparticles (TiO₂-NPs) on rat kidneys and the possible prophylactic role of either quercetin or idebenone. TiO₂-NPs were administered orally at either 600 mg or 1 g/kg body weight for 5 consecutive days to evaluate dose-dependent toxicity referred to the OECD guidelines for testing of chemicals. The results showed that administration of either low or high repeated doses of TiO₂-NPs to rats significantly increases serum kideney function biomarkers (urea, creatinine and uric acid) as well as increases in serum glucose and serum immuno- inflammatory biomarkers including tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), C-reactive protein (CRP), immunoglobin g (IGg), vascular endothelial growth factor (VEGF, angiogenic factor) and nitric oxide (NO) with a concomitant decrease in renal GSH content versus normal control values. The increase in these biomarkers was more evident in rats intoxicated with high TiO₂-NPs repeated doses. Oral co- administration of either quercetin or idebenone (each 200mg/Kg body weight) daily for three weeks to rats intoxicated by either of the two doses markedly ameliorated TiO₂-NPs induced alteration in the above biomarkers. The prophylactic impacts of both agents on biochemical markers were more pronounced in rats received low TiO₂-NPs repeated doses. The biochemical investigation was supported by histological examination. In conclusion, The data showed the severity in renotoxicity of TiO₂-NPs was dose-dependent and the protective effect of quercetin and idebenone may be related to their antioxidant and anti-inflammatory properties.

The present study investigates the effects of carnosine and/or cyclosporine A (CyA) treatment against traumatic brain injury (TBI) in immature rats. Traumatized rats received carnosine [(200 mg/kg/day, in pre-and post-treatment (i.p.)] for 7 consecutive days following TBI. CyA (20 mg/kg, i.p.) was administrated 15 min and 24 h after TBI. The results revealed that TBI caused sever brain injury indicated by increased nucleotide hydrolysis which was ensured by pronounced increase in ectonucleotidases, NTPDases (ATP and ADP hydrolysis) and 5'-nucleotidase (AMP hydrolysis) in traumatized rats compared with normal animals. TBI also causes elevation of glycolytic enzymatic activities as lactate dehydrogenase (LDH) and phosphoglucoisomerase (PGI) in rats’ brains. In addition TBI pronouncedly reduced the activities of antioxidant enzymes glutathione reductase (GR) and catalase (CAT) in brain tissue as compared to normal animals. Injection of carnosine and/or CyA significantly modulates the altered enzymatic activities. In conclusion, the present data may suggest the beneficial effect of carnosine and/or CyA in protection of brain tissues from disorders induced by traumatic injury. Schistosomiasis is considered the second most pre-valiant worldwide parasitic disease ranked next to malaria. It has significant economic and public health consequences in many developing countries. Several ways have been practiced in order to bring the disease under an adequate control through the breakage of the life cycle of the parasite. Snail control could be regarded as a rapid and efficient of reducing or eliminating transmission and remains among the methods of choice for schistosomiasis control. The aim of this work is to evaluate the role of Haplophyllum tuberculatum (family Rutaceae) as a plant molluscicide. The mortality rate of Biomphalaria alexandrina snails were monitored after treatment with three extracts of the plant aerial parts; petroleum ether, chloroform and ethanol. Chloroform extract that recorded the most potent effect was further evaluated through measuring the toxicity pattern against B. alexandrina snails, egg laying capacity, cercarial shedding, phenol oxidase enzyme and the levels of steroid sex hormones. Histopathological examination of hepatopancreas and ovotestis of treated snails were also done for result confirmation. Treatment of snails by chloroform extract recorded reduction in egg laying capacity, decrease in cercarial shedding, diminution in phenol oxidase enzyme, disturbance in steroid sex hormones and sever alternation of the histopathological picture of snails tissue. In conclusion, H. tuberculatum recorded molluscicidal potency against B. alexandrina snails. Further studies are needed for its environmental applications.

This study aimed to explore the effective role of carnosine and /or L- arginine in down regulation of the inflammatory molecule expression caused renal damage in response to sodium nitrite (NaNO2) induced hypoxia in rats . NaNO2 was administered subcutaneously (s.c.) to rats as a single dose (60 mg/kg body weight ). L-arginine (200mg/Kg body weight) and carnosine (250 mg/ Kg body weight ) were administered (i.p.) as a single dose , 24 h before NaNO2 injection. The results revealed that pre- administration of arginine and /or carnosine to NaNO2 hypoxic rats, significantly modulated the increases in serum markers of renal function (creatinine and urea) as well as the decrease in hemoglobin (Hb) level versus hypoxic rats. The two agents each alone or in a combination, markedly down regulated the serum pro-inflammatory molecules, including tumor necrosis factor-α (TNF- α) , C-reactive protein (CRP), vascular endothelial growth factor (VEGF) and heat shock protein -70 (HSP-70) as well as interleukin-6 (IL-6) in renal tissue compared to NaNO2 hypoxic rats . Also, the two agents successfully down modulated the alteration in the serum hypoxia inducible factor 1α (HIF 1α) . The present biochemical results were also supported by histopathological examination. In conclusion, the current data revealed that although the efficacy of arginine or carnosine each alone, their combination was more effective in ameliorating the renal damage induced by inflammatory molecules in response to NaNO2 hypoxia . This may support the use of this combination as an effective drug to treat hypoxic renal damage

This study investigated the possible antidiabetic role and therapeutic crucial action of the saponin fractions of the ethanolic extract of areal parts of the medicinal plant Anabasis articulata compared to currently available antidiabetic drug gliclazide (diamicron) against diabetic complications induced tissue injury in rats. Fractionation of hydro alcoholic extract from the aerial parts of Anabasis articulata (Chenopodiaceae) led to the isolation of Four known saponins: 3-O-glucopyranosyl of(stigmasterol, s-sitosterol, sitostanol), 3-O-[s-D- the glucopyranosyl] oleanolic acid , 3-O-[s-D-glucopyranosyl -28-O-s-D - xylopyranosyl] oleanolic acid, in addition to proceric acid. The isolated compounds were identified by means of chemical methods and spectrometric analysis as Rf values, UV Mass, 1H NMR and 13CNMR spectroscopy. Animals were divided into 4 groups. Group1, control rats (not received any medication). Group 2, rats injected intraperitoneally with single dose of streptozotocin (STZ)(40 mg/kg body weight). Group 3, rats orally administered with ethanolic extract of A. articulata (400 mg/ kg B.W.) after STZ injection. Group 4, rats orally administered with gliclazide (10 mg/kg B.W.) after STZ injection. Oral administration of the plant modulated the diabetic increase in blood glucose and cortisol levels revealing the antihyperglycemic effect of this medicinal plant. It effectively increases the blood hormone insulin concentration and α- fetoprotein. It is also significantly decrease blood tumor necrosis factor α (TNF-α). The current plant also effectively decreased blood fructosamine to their normal levels as well as the consequence diabetic decrease in the hemoglobin (Hb) and albumin levels. Furthermore, ingestion of the plant effectively modulated hepatic oxidative tissue damage. Supplementation of diabetic animals with gliclazide improved diabetic induced alteration in most of the above studied markers. These results suggest that Anabasis articulata has multi-beneficial actions in controlling diabetes and consequence complications induced in pancreas and liver and may candidate as natural antidiabetic drug.

Summary Introduction: Warfarin is one of the most widely used anticoagulants, yet interindividual differences in drug response, a narrow therapeutic range and a high risk of bleeding or stroke complicate its use. We aimed to determine the allele and genotype frequency of VKORC1 1173 C>T, CYP2C9*2 and CYP2C9*3 variant polymorphisms in the Egyptian population and to evaluate their influence on the interindividual differences in warfarin dosage. Methods: A total of 154 unrelated healthy adult patients and 46 warfarin-treated patients were included. SYBR Green-based real-time polymerase chain reaction (PCR) assay was used for studying VKORC1 (C1173T) and CYP2C9*3 polymorphisms. Mutagenically separated PCR assay was used to detect the CYP2C9*2 allele. Results: VKORC1 genotype frequencies were 11%, 24% and 65% for CC, CT and TT, respectively. The prevalence of CYP2C9 haplotypes was 81% (*1\*1), 3.3% (*1\*2), 9.7% (*1\*3), 4.5% (*2\*2) and 0.65% (2\*3 and *3\*3). VKORC1 TT and CYP2C9*2\*2 were associated with a significantly lower warfarin dose. VKORC1 and CYP2C9 accounted for 31.7% and 15.6% of warfarin dose variability, respectively, and together with clinical factors explained 61.3% of total variability. Conclusion: VKORC1-TT and CYP2C9 *1/*1 are the most prevalent genotypes among Egyptians. Patients with VKORC1-TT genotype required a lower warfarin dose.

To our knowledge, this is the first report describing the oxidant–antioxidant status in horses clinically affected with peritonitis. The present study was carried out to investigate the importance of oxidative stress markers and antioxidant parameters in the prediction of clinical outcomes of peritonitis in draft horses. The alleviative effects of two antioxidants, carnosine (Car) and melatonin (Mel), against titanium dioxide nanoparticles (TiO2 -NPs) toxicity-induced oxidative and inflammatory renal damage were examined in rats. Administration of these antioxidants along with TiO2 -NPs effectively reduced serum urea, uric acid, creatinine, glucose, tumor necrosis factor-α, interleukin-6, C-reactive protein, immunoglobulin G, vascular endothelial growth factor, and nitric oxide, as well as a significant amelioration of the decrease in glutathione levels in renal tissue was observed, compared to those in rats treated with TiO2 -NPs alone. The renoprotective properties of the antioxidants were confirmed by reduced intensity of renal damage as demonstrated by histological findings. In conclusion, Car and Mel play protective roles against TiO2 -NPs-induced renal inflammation and oxidative injury, likely due to their antioxidant and anti-inflammatory properties.

There are many fruit trees that could be integrated into dryland farming systems in Sub-Saharan Africa to support income and nutritional security. Fruit contains almost all known vitamins and many essential minerals. Five important fruit species that are cross-regional include: Adansonia digitata, Tamarindus indica, Zizyphus mauritiana, Sclerocarya birrea, and Mangifera indica. While these species are well integrated in the Sahel region, besides mango, they are generally absent from smallholder farms in East and Central Africa. Fruits of the species in this region are mostly harvested unsustainably from the wild communal areas. Unlike the situation in the neighboring Southern Africa region, where S. birrea is utilized extensively in the wine industry, there is virtually no use for the tree in this region, largely because of limited knowledge. Z. mauritiana use is also limited because of low quality germplasm—the hard stone clings to the flesh. An analytical framework based on five factors (site requirements, genetic variability, propagation methods, nutritional properties and utilization, and commercial potential) is used to compare knowledge status and gaps on the species in the region. While this analysis reveals the existence of considerable knowledge between and within the species, lack of improved germplasm and markets emerge as two key constraints limiting their conservation through on-farm planting. Key research and development needs identified are: (a) fostering cross-collaboration and knowledge exchange with other regions where the species are fairly well utilized, and (b) developing criteria and indicators for monitoring impacts and increased investments on the “Big Five” on livelihood of dryland communities and on biodiversity conservation.

The aim of this work was to study the possible cardiotoxicity of two different doses of 50 nm nano titanium dioxide (n-TiO2) and the possible modulating effects of the use of two natural antioxidants carnosine and melatonin. The results showed that TiO2- NPs produced deleterious effects on rat cardiac tissue as confirmed by the increased levels of serum myoglobin, troponin-T and CK-MB. Increased levels of serum Inflammatory markers represented by the tumor necrosis factor alpha (TNF-α) and Interleukin-6 (IL-6) was also noticed. Caspase3 and IGg were elevated compared to the control group in a dose dependant manner. treatment of the rats with Carnosine or melatonin. along with TiO2- NPs administration significantly improved most of the elevated biochemical markers. The portal provides access to numerous database projects supported by 30 scientific organizations. It includes over 800 different biological datasets spanning genomics, proteomics, model organisms, cancer data, ontology information and more. All resources available through the portal are independently administered and funded by their host organizations. The BioMart data federation technology provides a unified interface to all the available data. The latest version of the portal comes with many new databases that have been created by our ever-growing community. It also comes with better support and extensibility for data analysis and visualization tools. A new addition to our toolbox, the enrichment analysis tool is now accessible through graphical and web service interface. The BioMart community portal averages over one million requests per day. Building on this level of service and the wealth of information that has become available, the BioMart Community Portal has introduced a new, more scalable and cheaper alternative to the large data stores maintained by specialized organizations.

Anabasis articulata (Forssk) Moq. (Chenopodiaceae) is an herb, grows in Egypt, and used in folk medicine to treat diabetes, fever, and kidney infections. The protective and therapeutic effects of the ethanol extract of A. articulata aerial parts were evaluated against dimethylnitrosamine (DMN)-induced liver fibrosis, compared with the standard drug, silymarin. Hepatic hydroxyproline content, serum transforming growth factor-β1 (TGF-β1), interleukin 10 (IL-10) and fructosamine were measured as liver fibrosis markers. Hepatic malondialdehyde (MDA), nitric oxide (NO), catalase (CAT), glutathione reductase (GR) and glutathione content (GSH) were measured as oxidant/antioxidant markers. Parallel histopathological investigations were also performed. Protective and therapeutic administration of A. articulata (100 mg/kg daily for 4 weeks), markedly prevented DMN-induced loss in body and liver weights. The aim of this study was to investigate the protective role of quercetin and/or l-arginine against the cardiotoxic potency of zinc oxide nanoparticle (ZnO-NP)-induced cardiac infarction. ZnO-NPs (50 nm) were administered orally at either 600 mg or 1 g/kg body weight for 5 consecutive days. The results revealed that co-administration of quercetin and/or l-arginine (each 200 mg/kg body weight) daily for 3 weeks to rats intoxicated by either of the two doses markedly ameliorated increases in serum markers of cardiac infarction, including troponin T, creatine kinase-MB, and myoglobin, as well as increases in proinflammatory biomarkers, including tumor necrosis factor-α, interleukin-6, and C-reactive protein, compared with intoxicated, untreated rats. Each agent alone or in combination also successfully modulated the alterations in serum vascular endothelial growth factor, cardiac calcium concentration, and oxidative DNA damage as well as the increase in the apoptosis marker caspase 3 of cardiac tissue in response to ZnO-NP toxicity. In conclusion, early treatment with quercetin and l-arginine may protect cardiac tissue from infarction induced by the toxic effects of ZnO-NPs.

Praziquantel (PZQ), the drug of choice according to the World Health Organization (WHO), causes some side effects. The appearance of drug resistance against PZQ in Schistosoma mansoni infected species leads to must introduce new effective compounds and/or suppress its side effects. The aim of present work is a trial to solve this problem. Chloroform extract of Citharexylum quadrangular jacq leaves was used as a medicinal plant, which affect the Schistosoma mansoni adults. Prophylactic and therapeutic treatment of Schistosoma mansoni infected mice with the plant extract and/or the antioxidants vitamin E and selenium in combination with PZQ was studied. The study revealed that the combination of the chloroform extract together with vitamin E and selenium improved the efficiency of PZQ. These supplementations are very effective in ameliorating the oxidative insult as well as other parameters: glutathione reductase (GR), thioredoxin reductase (TrxR), catalase (CAT) and reduced glutathione (GSH), nitric oxide (NO) and lipid peroxidation (MDA), hepatic hydroxyproline content, Alanine aminotransferase (ALT), Gamma-glutamyltransferase (γ-GT), tumor necrosis factor-alpha (TNFα), interleukin- 10 (IL-10) and total immunoglobulin E (total IgE) associated with Schistosoma mansoni infection.

The schistosomicidal properties of Nigella sativa seeds were tested in vitro against Schistosoma mansoni miracidia, cercariae, and adult worms. Results indicate its strong biocidal effects against all stages of the parasite and also showed an inhibitory effect on egg-laying of adult female worms. In the present work we also studied the effects of crushed seeds on some antioxidant enzymes; which have a role in protection of adult worms against host oxidant killing; as well as some enzymes of glucose metabolism; which have a crucial role in the survival of adult worms inside their hosts. The data revealed that the used drug induce an oxidative stress against adult worms which indicated by a decrease in the activities of both antioxidant enzymes, superoxide dismutase, glutathione peroxidase, and glutathione reductase and enzymes of glucose metabolism, hexokinase and glucose-6-phosphate dehydrogenase. Disturbing of such enzymes of adult worms using N. sativa seeds could in turn render the parasite vulnerable to damage by the host and may play a role in the antischistosomal potency of the used drug.

Carnosine is a naturally occurring dipeptide (beta-alanyl-L-histidine) found in muscles, brain and other tissues. This study was designed to test the ability of carnosine to offset metabolic disturbances induced by Schistosoma mansoni parasitism. Results indicate that parasitic infection caused elevation of liver weight/body weight in S. mansoni-infected hamsters, induced lipid peroxidation and reduced glycogen levels. Moreover, adenylate energy charge (AEC) and ATP/ADP and ATP/AMP concentration ratios were markedly lower in infected hamsters. Administration of carnosine (10 mg/day) for 15 days concurrent with infection effectively reduced worm burden and egg count. Administration of carnosine 2 and 4 weeks post-exposure only partially ameliorated the S. mansoni effects on metabolism. Carnosine treatment also normalized most of the parameters measured, including glycogen repletion, the antioxidant status and AEC. These finding support the use of carnosine for possible intervention in schistosomiasis.

Background/Objective Bladder cancer is the commonest type of malignant tumors as a result of schistosomaisis which is a major healthy problem in many subtropical developing countries. The aim of this study is to comparatively elucidate the underlying biochemical tumor markers in schistosomal bladder cancer versus non-schistosomal bladder cancer when compared to normal healthy ones. Methods This work was performed on tissue specimens from total 25 patients and serum samples from total 30 patients versus ten healthy individuals served as control. The investigated parameters in serum are: xanthine oxidase (XO), fructosamine, lactate dehydrogense (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), total proteins, essential and non- essential amino acids profile, hydroxyproline, total immunoglobulin E (IgE) and tumor necrosis factor alpha (TNF-α). In addition, the current investigation also extended to study some markers in tumor bladder tissues including, pyruvate kinase enzyme (PK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Results Results showed that biharzial bladder cancer patients recored more significant elevation in serum XO, fructosamine, LDH, AST, ALT, hydroxyproline, IgE and TNF-α than in bladder cancer patients when compared to control ones. While, in tissues there were significant increase in PK, LDH, AST & ALT activities of schistosomal bladder cancer than in bladder cancer as compared to control healthy patients. Conclusions It could be concluded that, bilharzial and non-bilharzial bladder cancer showed distinct biochemical profile of tumor development and progression which can be taken into consideration in diagnosis of bladder cancer.

The aim of this study was to investigate the effective role of silymarin either alone or in combination with chlorogenic acid and/or melatonin against the toxic impact of carbon tetrachloride (CCl4) induced cardiac infarction. CCl4 (l.2ml/kg body weight) was administered as a single dose intraperitoneally. The results revealed that the administration of silymarin alone or in combination with chlorogenic acid (CGA) and/or melatonin for 21 consecutive days, 24h after CCl4 injection to rats, markedly ameliorated the increases in serum markers of cardiac infarction, including troponin T and creatine kinase-MB (CK-MB), as well as increases in the pro-inflammatory biomarkers, including interleukin-6 (IL-6), interferon-γ (IFN-γ) in serum and tumor necrosis factor-α (TNF-α) and C-reactive protein in cardiac tissue compared to CCl4 intoxicated rats. The used agents also successfully modulated the alteration in vascular endothelial growth factor (VEGF) in serum and the oxidative DNA damage and the increase in the apoptosis marker caspase 3 in cardiac tissue in response to CCl4 toxicity. The present biochemical results are supported by histo-pathological examination. The current results proved that treatment with silymarin in combination with CGA and melatonin was the most effective one in ameliorating the toxicity of CCl4 induced cardiac damage and this may support the use of this combination as an effective drug to treat cardiac damage induced by toxic agents.
