Abstract In the realm of radiation therapy, a conspicuous obstacle lies in the dearth of external observation concerning radiation beams aimed at the patient. While real-time monitoring of such beams on the patient's surface during therapy holds promise, the imaging of particle beams has thus far proven to be a formidable task. Here, we show our discovery of polyester fabrics and cloths as auspicious scintillating materials, ideally suited for the visualization of radiation beams upon the patient's surface. The light output of polyester fabrics ranged from 10 to 20% of that observed in plastic scintillators. When exposed to spot scanning proton beams, clear beam spots emerged on the surface of the polyester cloths. The movement of these scanning beams was effectively captured using a CMOS camera in a light-shield-free with lights-off environment. The resulting images provided a means for evaluating spills of the proton beams. The inherent flexibility of polyester fabrics and clothing enhances their appeal for applications in the intricate landscape of radiation therapy, promising a bright future for surface beam imaging endeavors.

Abstract Peak oxygen uptake (VO2), evaluated as exercise tolerance, is a strong predictor of life prognosis regardless of health condition. Several previous studies have reported that peak VO2 is higher in those with a greater decrease in muscle oxygen saturation (SmO2) in the active muscles during incremental exercise. However, the skeletal muscle characteristics of individuals exhibiting a greater decrease in SmO2 during active muscle engagement in incremental exercise remain unclear. This study aimed to clarify the relationship among muscle strength, muscle endurance, and skeletal muscle oxygenation dynamics in active leg muscles during incremental exercise. Twenty-four healthy young men were included and categorized into the non-moderate-to-high muscular strength and endurance group (those with low leg muscle strength, endurance, or both; n = 11) and the moderate-to-high muscular strength and endurance group (those with both moderate-to-high leg muscle strength and endurance; n = 13). All participants underwent cardiopulmonary exercise testing combined with near-infrared spectroscopy to assess whole-body peak VO2 and the change in SmO2 at the lateral vastus lateralis from rest to each exercise stage as skeletal muscle oxygenation dynamics. A comparative analysis of praseodymium (Pr3+)-doped fluoride glasses, APLF [20Al(PO3)3–80LiF–PrF3], BCAYF (19BaF2–33.25CaF2–42.75AlF3–5YF3–PrF3), and BCAPLF (19BaF2–33.25CaF2–42.75AlF3–15P2O5–20LiF–PrF3), reveals the effect of the host matrix on the optical properties, particularly the luminescence behavior of Pr3+ ions. X-ray absorption near edge structure (XANES) spectroscopy verifies the presence of Pr ions in the glasses with a +3 oxidation state. Absorption spectra results suggest that the lowest energy level of the Pr3+ 4f5d configuration is influenced by the oxide/fluoride ratio of the glasses. As both interconfigurational 4f5d and intraconfigurational 4f2 transitions are observed, the energy gap between the lowest level of the 4f5d configuration and the overlapping 1S0 level of the 4f2 configuration is found to influence the predominance of UV emissions from the 4f5d and 1S0 levels as well as their decay times. Consequently, a larger 4f5d-1S0 energy gap of ∼5000 cm−1 leads to a more intense UV emission from the 4f5d level with a faster decay time. These results underscore the significance of minimizing the energy of the 4f5d level and increasing the gap between 4f5d-1S0 levels to be able to obtain fast UV emissions from Pr3+-doped fluoride glasses.

ABSTRACTCoronavirus disease 2019 (COVID-19) was extraordinarily harmful, with high rates of infection and hospitalization. This study aimed to evaluate the impact of COVID-19 vaccination status and other factors on hospitalization and disease severity, using data from Nagasaki Prefecture, Japan. Confirmed cases of COVID-19 infection with vaccination status were included and the differences in characteristics between different vaccination statuses, hospitalization or not, and patients with varying levels of disease severity were analyzed. Furthermore, logistic regression was used to calculate odds ratio (ORs) and 95% confidence intervals (CI) to evaluate the association of various factors with hospitalization and disease severity. From March 14, 2020 to August 31, 2022, 23,139 patients were unvaccinated 13,668 vaccinated the primary program with one or two doses, and 4,575 completed the booster. Vaccination reduced the risk of hospitalization with an odd ratio of 0.759 (95% CI: 0.654–0.881) and the protective effect of completed booster vaccination was more pronounced (OR: 0.261, 95% CI: 0.207–0.328). Similarly, vaccination significantly reduced the risk of disease severity (vaccinated primary program: OR: 0.191, 95% CI: 0.160–0.228; completed booster vaccination: OR: 0.129, 95% CI: 0.099–0.169). Overall, unvaccinated, male, elderly, immunocompromised, obese, and patients with other severe illness factors were all risk factors for COVID-19-related hospitalization and disease severity. Vaccination was associated with a decreased risk of hospitalization and disease severity, and highlighted the benefits of completing booster.

Pr-doped scintillators are attracting attention because of their potential applications in medical devices. In this study, we focused on Pr-doped (La, Y)2Si2O7 scintillators to grow single crystals by the micro-pulling-down method and to evaluate their optical and scintillation properties. From the powder XRD results, the crystal structure and the space group were identified to be monoclinic and P21/n, respectively. The results of optical and scintillation characterization show that the luminescence from the Pr3+ 5d1–4f transition increases with increasing Pr3+ concentration. The maximum light yield was also obtained for the sample with the highest Pr3+ concentration. The Pr-doped (La, Y)2Si2O7 single crystal shows promising properties for application in radiation detection.

The influence of nonstoichiometric engineering on the scintillation properties of Ce3+ doped yttrium aluminum perovskite (YAP:Ce) crystals was investigated. Crystals with slight yttrium excess (Y1·004AlO3:Ce), yttrium deficiency (Y0·994AlO3:Ce), and stoichiometric composition (YAlO3:Ce) were synthesized using the micro-pulling down method. Structural characterization confirmed crystallization in the pure orthorhombic Pnma space group for all compositions. Optical absorption spectroscopy revealed an extensive background in the nonstoichiometric crystals due to the increased degree of light scattering centers associated with the defects. The photoluminescence excitation and emission spectra of Ce3+ ions were unaffected by nonstoichiometry. However, the scintillation decay kinetics and light yield exhibited marked improvements in the Y1±xAlO3:Ce (x≠0) crystals. The light yield increased by 22 % and 15 % and the slower component of the decay accelerated from 171 ns to 106 and 131 ns compared to the stoichiometric crystal. Thermoluminescence glow curves revealed that nonstoichiometry significantly altered the concentrations of antisite defects and oxygen vacancies. The EPR spectroscopy investigations revealed a correlation between the Ce3+ ions concentration and the degree of nonstoichiometry. Strategic engineering of nonstoichiometry in Y1±xAlO3:Ce (x≠0) scintillators may have provided an effective approach to optimizing scintillation performance.

Pigs are the most common amplifying hosts of the Japanese encephalitis virus (JEV). In 2016, four residents on Tsushima Island who did not own pig farms were diagnosed with JE. Therefore, a serosurvey was conducted to estimate the risk and seroprevalence of JEV after the outbreak. Sera collected from 560 Tsushima Island residents between January and September 2017 were tested for neutralizing antibodies against JEV strains JaGAr01 (genotype 3) and Muar (genotype 5). Sera collected from six wild boars between June and July 2022 were tested. The seroprevalence rates of neutralizing antibodies against JaGAr01 and Muar were 38.8% and 24.6%, respectively. High anti-JEV neutralizing antibody titers of ≥320 were identified in 16 residents, including 3 younger than 6 years with prior JEV vaccination, 2 in their 40s, and 11 older than 70. However, no anti-JEV-specific IgM was detected. Residents who engaged in outdoor activities had higher anti-JEV antibody titers. Sera from wild boars were negative for JEV RNA, but four of six samples contained neutralizing antibodies against JEV. Therefore, JEV transmission continues on Tsushima Island, even in the absence of pig farms, and wild boars might serve as the amplifying hosts.

Aluminum garnets display exceptional adaptability in incorporating mismatching elements, thereby facilitating the synthesis of novel materials with tailored properties. This study explored Ce3+-doped Tb3Al5−xScxO12 crystals (where x ranges from 0.5 to 3.0), revealing a novel approach to control luminescence and photoconversion through atomic size mismatch engineering. Raman spectroscopy confirmed the coexistence of garnet and perovskite phases, with Sc substitution significantly influencing the garnet lattice and induced A1g mode softening up to Sc concentration x = 2.0. The Sc atoms controlled sub-eutectic inclusion formation, creating efficient light scattering centers and unveiling a compositional threshold for octahedral site saturation. This modulation enabled the control of energy transfer dynamics between Ce3+ and Tb3+ ions, enhancing luminescence and mitigating quenching. The Sc admixing process regulated luminous efficacy (LE), color rendering index (CRI), and correlated color temperature (CCT), with adjustments in CRI from 68 to 84 and CCT from 3545 K to 12,958 K. The Ce3+-doped Tb3Al5−xScxO12 crystal (where x = 2.0) achieved the highest LE of 114.6 lm/W and emitted light at a CCT of 4942 K, similar to daylight white. This approach enables the design and development of functional materials with tailored optical properties applicable to lighting technology, persistent phosphors, scintillators, and storage phosphors.

Abstract High-resolution imaging of alpha particles is required in the detection of alpha radionuclides in cells or small organs for the development of radio-compounds for targeted alpha-particle therapy or other purposes. We developed an ultrahigh resolution, real time alpha-particle imaging system for observing the trajectories of alpha particles in a scintillator. The developed system is based on a magnifying unit and a cooled electron multiplying charge-coupled device (EM-CCD) camera, combined with a 100-µm-thick Ce-doped Gd3Al2Ga3O12 (GAGG) scintillator plate. Alpha particles from an Am-241 source were irradiated to the GAGG scintillator and imaged with the system. Using our system, we measured the trajectories of the alpha particles having different shapes in real time. In some of these measured trajectories, the line shapes of the alpha particles that flew in the GAGG scintillator were clearly observed. The lateral profiles of the alpha-particle trajectories were imaged with widths of ~ 2 µm. We conclude that the developed imaging system is promising for research on targeted alpha-particle therapy or other alpha particle detections that require high spatial resolution.

The 3He gas is commonly used for the detection of thermal neutrons. However, with the depletion of 3He gas, there is a need to develop new solid scintillators for thermal neutron detection. Solid scintillators containing 6Li, which have large neutron capture cross-sections and a large amount of energy released by transmutation reactions, are commonly used as alternative candidates. However, only single-crystal scintillators are currently used, and their 6Li concentration is limited by their chemical composition. In this study, we designed, grew, and evaluated a new eutectic scintillator, Rb2CeCl5/LiCl, which can improve the 6Li concentration compared with single-crystal scintillators. Rb2CeCl5, which was selected as the scintillator phase, has excellent scintillator properties (light yield: 36,000 photons/MeV, decay time: mostly 24 ns, slightly 153 ns), and is less deliquescent than other halide scintillators. The crystal grown using the vertical Bridgman method exhibited a eutectic phase composed of Rb2CeCl5 and LiCl. The eutectic crystals exhibited Ce3+ 5d-4f emissions, with a peak between 360 and 370 nm. The Rb2CeCl5 phase was identified as the luminescent phase via cathodoluminescence mapping, and 16,000 photons/neutron of the light yield and 56.1 ns of the decay time were observed. This study indicates that the Rb2CeCl5/LiCl eutectic scintillator is a promising candidate for use in thermal neutron detectors.

Abstract Background and Aim Symptoms of primary biliary cholangitis (PBC) frequently impair one's quality of life (QOL). Nonetheless, with improved treatment, the prognosis of PBC also improves. QOL plays an important role in patients with PBC. In this study, we aimed to reevaluate the transition of new symptom development in PBC and its predictive factors. Methods This retrospective multicenter study enrolled 382 patients with PBC for symptom analysis. The impact of a newly developed symptom on PBC prognosis was investigated by Kaplan–Meier analysis with propensity score matching and logistic progression analysis. Results The cumulative risk of developing a new symptom after 10 and 20 years of follow‐up was 7.6 and 28.2%, and specifically that of pruritus, which was the most common symptom, was 6.7 and 23.3%, respectively. Abstract Introduction In patients with mild cognitive impairment, pathological changes begin in the amygdala (AMG) and hippocampus (HI), especially in the parahippocampal gyrus and entorhinal cortex (ENT). These areas play an important role in olfactory detection and recognition. It is important to understand how subtle signs of olfactory disability relate to the functions of the above‐mentioned regions, as well as the orbitofrontal cortex (OFC). In this study, we evaluated brain activation using functional magnetic resonance imaging (fMRI), performed during the presentation of olfactory stimuli (classified as “normal odors” not inducing memory retrieval), and investigated the relationships of the blood oxygen level‐dependent (BOLD) signal with olfactory detection and recognition abilities in healthy elderly subjects. Methods Twenty‐four healthy elderly subjects underwent fMRI during olfaction, and raw mean BOLD signals were extracted from regions of interest, including bilateral regions (AMG, HI, parahippocampus, and ENT) and orbitofrontal subregions (frontal inferior OFC, frontal medial OFC, frontal middle OFC, and frontal superior OFC). Multiple regression and path analyses were conducted to understand the roles of these areas in olfactory detection and recognition. Results Activation of the left AMG had the greatest impact on olfactory detection and recognition, while the ENT, parahippocampus, and HI acted as a support system for AMG activation. Less activation of the right frontal medial OFC was associated with good olfactory recognition. These findings improve our understanding of the roles of limbic and prefrontal regions in olfactory awareness and identification in elderly individuals. Conclusion Functional decline of the ENT and parahippocampus crucially impacts olfactory recognition. However, AMG function may compensate for deficits through connections with frontal regions.

In this study, Eu:6LiBr/BaBr2 with a high Li concentration was developed as a novel neutron scintillator. Here, an Eu-doped BaBr2 phase acted as a scintillator and 6Li-containing LiBr phase acted as a neutron capturer. Eu:6LiBr/BaBr2 eutectics were fabricated via the vertical Bridgman technique in quartz ampoules. The scintillation performance of Ce:6LiBr/LaBr3 was measured using electron and neutron irradiation to evaluate its potential as a neutron scintillator. The grown Eu:6LiBr/BaBr2 eutectic exhibited a lamellar eutectic structure and optical transparency as well as a 410 nm emission due to the Eu2+ 5d–4f transition. The light yield under thermal neutron excitation was estimated to be 5600 photons/neutrons. The scintillation decay time was 550 ns.

Abstract Electronic devices have become an indispensable part of our daily lives, while their negative aspects have been reported. One disadvantage is that reading comprehension is reduced when reading from an electronic device; the cause of this deficit in performance is unclear. In this study, we investigated the cause for comprehension decline when reading on a smartphone by simultaneously measuring respiration and brain activity during reading in 34 healthy individuals. We found that, compared to reading on a paper medium, reading on a smartphone elicits fewer sighs, promotes brain overactivity in the prefrontal cortex, and results in reduced comprehension. Furthermore, reading on a smartphone affected sigh frequency but not normal breathing, suggesting that normal breathing and sigh generation are mediated by pathways differentially influenced by the visual environment. A path analysis suggests that the interactive relationship between sigh inhibition and overactivity in the prefrontal cortex causes comprehension decline. These findings provide new insight into the respiration-mediated mechanisms of cognitive function.

Abstract Background Cognitive function declines with age and has been shown to be associated with atrophy in some brain regions, including the prefrontal cortex. However, the details of the relationship between aging and cognitive dysfunction are not well understood. Methods Across a wide range of ages (24- to 85-years-old), this research measured the gray matter volume of structural magnetic resonance imaging data in 39 participants, while some brain regions were set as mediator variables to assess the cascade process between aging and cognitive dysfunction in a path analysis. Results Path analysis showed that age affected the left hippocampus, thereby directly affecting the left superior frontal gyrus. Furthermore, the gyrus directly affected higher order flexibility and maintenance abilities calculated as in the Wisconsin card sorting test, and the two abilities affected the assessment of general cognitive function. Conclusion Our finding suggests that a cascade process mediated by the left hippocampus and left superior frontal gyrus is involved in the relationship between aging and cognitive dysfunction.

The atomic positions and displacements of atoms around the Ta atom in piezoelectric materials Ca3TaGa3Si2O7 (CTGS) and Ca3TaGa1.5Al1.5Si2O7 (CTGAS) were investigated at 100 K by Ta-Lα X-ray fluorescence holography (XFH). The experimental atomic images were compared with the simulated ones using the crystal structures of CTGS and CTGAS, which were determined by single crystal X-ray diffractometry (SC-XRD). The atomic positions agreed between XFH and SC-XRD experiments. With the help of XFH simulation, the displacements of Ta, Ca, Si, and Ga atoms relative to the Ta atom were qualitatively analyzed using experimental atomic image intensities. The relative displacement of the Ca atom was increased by Al substitution, while those of the Ta and Ga atoms were decreased. The first principles calculation based on density-functional theory (DFT) was performed to understand bonding character between constituents. The evaluation of the crystal orbital Hamilton population (COHP) clarified that the Ca-O bond has strong ionic character different with the other bonds, suggesting that the positional shift of the Ca atom is responsible for the piezoelectricity in CTGS. The effect of Al substitution on piezoelectricity was also considered based on the change in the Ca-O bond.

In this study, Tl:CsI/CsCl/KCl ternary eutectic scintillators were grown using the vertical Bridgman–Stockbarger method by unidirectional solidification. The CsI and KCl phases were elongated along the growth direction, based on backscattered electron imaging (BEI) observations. The Tl:CsI scintillator phase grew as a matrix, which can be used to create optical waveguides for scintillation light. The expected Tl+ emission peak was observed at 550 nm under X-ray excitation. This emission was attributed to the self-trapped excitons perturbed by TI+ in Tl:CsI. The decay time of the Tl:CsI/CsCl/KCl eutectic was 285 ns, and its light output was estimated to be 10,800 photons/MeV.

β-Ga2O3 is a well-known semiconductor material for power devices and other applications. Recently, β-Ga2O3 has also been reported as a scintillator material with a light yield of approximately 8400 ph./MeV, scintillation decay time of 3. In this study, 45 cm diameter β-Ga2O3 single crystals were prepared via oxide crystal growth using the cold crucible (OCCC) method under various oxygen partial pressures. In the OCCC method, as in the cold crucible method, a high frequency is applied directly to the oxide materials, which are heated and melted, and the melt is held by the outermost solid material itself that is cooled by water using a copper hearth. In the OCCC method, crystal growth is performed while rotating the seed crystal, as in the Czochralski method, to increase the crystal diameter. The optical properties and radiation responses of the crystals grown under various oxygen partial pressures were evaluated.

In this study, it was demonstrated that Zn3Ta2O8 crystals containing zinc oxide with high vapor pressure can grow in melt by the shielded arc melting method. The radiation response characteristics of the crystals were evaluated. The grown crystals showed an emission peak at 410 nm, ascribed to self-trapped excitation, Ta4+–O2−. The full width at half maximum of the X-ray rocking curve was 655 arcsec. The light output under 662 keV gamma-ray irradiation of Zn3Ta2O8 was approximately 4,700 photons/MeV. The decay time was 17.3 μs? Thus, the shielded arc melting method was shown to be a quick and economic growth technique for the survey of new scintillator materials containing high-vapor-pressure starting components.

Background: This study aimed to survey the attitudes toward COVID-19 vaccines and their acceptability among the Japanese public as soon as the United States Food and Drug Administration (FDA) authorized vaccines and their rollouts started around the world. Methods: An anonymous cross-sectional survey was conducted in Japan between 4 January and 5 March 2021. A questionnaire was administered to evaluate attitudes toward COVID-19 vaccines according to demographic characteristics, vaccine characteristics, and vaccine production. Results: A total of 1037 completed responses were received. More than half (63.5%) of the participants responded positively (extremely likely/likely) toward COVID-19 vaccines. The highest acceptance to be vaccinated was discovered among the youngest age group. As expected, participants who had never delayed acceptance or refused the vaccine in their history of vaccination had a significantly higher willingness to be vaccinated against COVID-19 (p < 0.001). Females (OR = 2.66, 95% CI: 1.99–3.58) and participants who had ever delayed acceptance or refuse the vaccine (OR = 3.49, 95% CI: 2.42–5.05) had higher odds of COVID-19 vaccine hesitancy. Participants with a postgraduate degree (OR = 0.64, 95% CI: 0.40–1.00) presented the highest willingness to be vaccinated against COVID-19. More than two-thirds (72.9%, 95% CI: 70.4%–75.8%) of the participants did not mind a booster dose required following primary vaccination. A total of 63.2% (95% CI: 60.0%–66.0%) of the participants only accepted a nearly 90% effective or above vaccine at preventing COVID-19. At the same, 86.4% (95% CI: 84.4%–88.4%) of the participants reported only accepting a vaccine with minor side effects. Conclusions: The moderate levels of COVID-19 vaccine acceptance found in the early phase of the pandemic demonstrate that it is important to improve the implementation of effective management for vaccine promotion and the acceptability of the vaccine to slow or delay transmission.

During the COVID-19 pandemic, infection protection/control education has become increasingly important for not only healthcare professionals but also students undertaking medical, nursing, physical therapy, occupational therapy, and other related courses. A review of the literature on infection control education reveals that the target participants often comprise healthcare workers, and very few studies of infection control education focus on students. We have developed a protocol for the systematic review of the literature on simulation-based infection prevention/control education for students undertaking medical, nursing, rehabilitation, and other related courses. The protocol for the systematic review and meta-analysis has been drafted in alignment with the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) statement. Systematic literature search will be performed for the period between 1990 (January) and 2022 (September) using the CENTRAL, MEDLINE, and Scopus databases. We will qualitatively and quantitatively examine the effects of simulation-based infection education for students in this systematic review and meta-analysis. Two investigators will independently search the databases according to the defined search strategy. The full-text of the selected articles will be screened independently keeping in mind the inclusion criteria by a pair of reviewers. Descriptive data will be extracted from each study regarding: study design, methods, participants, and outcomes. A meta-analysis will be performed if the quantitative data is suitable. Heterogeneity will be assessed using the standard χ2. Odds ratio for categorical data and weighted mean differences for continuous data and their 95% confidence intervals will be calculated and used for analysis. Where statistical pooling is not possible, the findings of the quantitative papers will be presented in narrative form. The qualitative aspect will employ narrative (descriptive) synthesis. Our review will make a valuable contribution to the domain of simulation-based infection prevention/control for students enrolled in medical and/or related courses.

Cerebral palsy (CP) causes abnormal gait pattern due to increased stretch reflex and contracture in specific muscles. Increased stretch reflexes in CP are caused by increased activation of muscle spindles due to increased sensitivity of gamma motor neurons. Furthermore, these neurological abnormalities may cause abnormal postures such as crouching and scissoring. This study aimed to construct a system for supporting treatment and orthosis development in patients with CP by simulation using a three-dimensional neuromuscular skeleton model and evaluate the validity of motor function due to CP reproduced by gait simulation. By reproducing motor function due to CP, the simulation system was able to obtain gait and muscle activity patterns similar to the actual ones. Therefore, motor function in CP can be quantitatively evaluated using this simulation system. In the model of children with CP, gait factors and joint angles of hip, knee, and ankle resembled those of the experimental results; however, the results of muscle synergy analysis were different. Hence, although gait pattern might be highly involved in the enhancement of stretch reflex and muscle contraction, muscle interaction and single muscles influence muscle control in CP.

In recent years, thermal neutron detection using scintillators has been used in a wide range of fields. Thus, the development of scintillators with a higher light yield, faster decay, and higher sensitivity for thermal neutrons is required. In this study, K2CeCl5/6LiCl and CeCl3/SrCl2/6LiCl were developed as novel eutectic scintillators for thermal neutron detection. LiCl was selected as the neutron capture phase and K2CeCl5 and CeCl3 were used as the scintillator phases. The eutectics of K2CeCl5/6LiCl and CeCl3/SrCl2/6LiCl were prepared using the Vertical Bridgman method and the phases were identified by scanning electron microscopy and powder X-ray diffraction measurements. The results of radioluminescence measurements under Ag source X-ray tube irradiation confirmed that the 5d-4f emission derived from Ce3+. The cathodoluminescence spectra and thermal neutron responses of the prepared eutectics were measured to evaluate their optical properties.

To date, 3He gas has been commonly used to detect thermal neutrons because of their high chemical stability and low sensitivity to γ-rays, owing to their low density and large neutron capture cross-section. However, the depletion of 3He gas prompts the development of a new solid scintillator for thermal neutron detection to replace 3He gas detectors. Solid scintillators containing 6Li are commonly used to detect thermal neutrons. However, they are currently used in single crystals only, and their 6Li concentration is defined by their chemical composition. In this study, 6Li-containing eutectic scintillators were developed. CeCl3 was selected as the scintillator phase because of its low density (3.9 g/cm3); high light yield (30,000 photons/MeV); and fast decay time with four components of 4.4 ns (6.6%), 23.2 ns (69.6%), 70 ns (7.5%) and >10 μs (16.3%), owing to the Ce3+ 5d-4f emission peak at approximately 360 nm. Crystals of the CeCl3, LiCl and CaCl2 ternary eutectic were fabricated by the vertical Bridgman technique. The grown eutectic crystals exhibited Ce3+ 5d-4f emission with a peak at 360 nm. The light yield was 18,000 photons/neutron, and the decay time was 10.5 ns (27.7%) and 40.1 ns (72.3%). Therefore, this work demonstrates optimization by combining a scintillator phase and Li-rich matrix phase for high Li content, fast timing, high light yield and low density.

Specific odors can induce memories of the past, especially those associated with autobiographical and episodic memory. Odors associated with autobiographical memories have been found to elicit stronger activation in the orbitofrontal cortex, hippocampus, and parahippocampus compared with odors not linked to personal memories. Here, we examined whether continuous odor stimuli associated with autobiographical memories could activate the above olfactory areas in older adults and speculated regarding whether this odor stimulation could have a protective effect against age-related cognitive decline. Specifically, we used functional magnetic resonance imaging to investigate the relationship between blood oxygen levels in olfactory regions and odor-induced subjective memory retrieval and emotions associated with autobiographical memory in older adults. In our group of healthy older adults, the tested odors induced autobiographical memories that were accompanied by increasing levels of retrieval and the feeling of being “brought back in time.” The strength of the subjective feelings, including vividness of the memory and degree of comfort, impacted activation of the left fusiform gyrus and left posterior orbitofrontal cortex. Further, our path model suggested that the strength of memory retrieval and of the emotions induced by odor-evoked autobiographical memories directly influenced neural changes in the left fusiform gyrus, and impacted left posterior orbitofrontal cortex activation through the left fusiform response.

The technology to grow single crystals of the required shape directly from a melt has been researched extensively and developed in various industries and research fields. In this study, a micro-pulling-down method and a Mo crucible were applied to the shape-controlled crystal growth of Y3Al5O12:Ce (YAG:Ce). Three types of Mo crucibles with different die shapes were developed. Stable crystal growth of more than 50 mm in length was achieved with the same shape as the die, and scintillation light output of ~20,000 ph/MeV, which is comparable with those of the YAG:Ce crystal grown by Cz method, were obtained. The transmittance of grown crystals above 500 nm was above 70%. The standard deviation (σ) of the scintillation light output at each position of the 50-mm-long sample was found to be within ±16%.

Abstract Introduction Pathological abnormalities first appear in the medial temporal regions including entorhinal cortex and parahippocampus in patients with Alzheimer's disease. Previous studies showed that olfactory decline in elderly subjects was associated with volume reductions in the left hippocampus and left parahippocampus without cognitive impairment. The aim of this study is to investigate the link between olfaction and volume reductions in the medial temporal regions including the parahippocampus, entorhinal cortex, and hippocampal subfields. Method 27 elderly subjects and 27 young controls were measured olfaction acuity, cognitive function, and structural magnetic resonance imaging. Image processing and gray matter volumetric segmentation were performed with FreeSurfer. Volume data were analyzed with SPSS Statistics software. Results Interesting results of this study were that volume reduction in the entorhinal cortex was not directly linked with declining olfactory ability. Volume reduction in the left entorhinal cortex was correlated with volume reduction in the left parahippocampus and dentate gyrus. However, left parahippocampus volume reduction had the greatest impact on olfactory decline, and the entorhinal cortex and dentate gyrus might additionally contribute to olfactory decline. Conclusion Our results indicate that olfactory decline may be directly reflected in the medial temporal regions as reduced parahippocampus volumes, rather than as morphological changes in the entorhinal cortex and hippocampus. The parahippocampus may play an important role in the association between memory retrieval and olfactory identification.

Objective(s): To investigate the diagnostic accuracy of positron emission mammography (PEM) and positron emission tomography/computed tomography (PET/CT) for small breast tumors of less than 20 mm in size.Methods: The study was conducted on a total of 100 subjects (i.e., 50 patients with pathologically proven breast cancer and 50 normal cases of medical screening). The total number of tumors was 54 (mean size: 11±5.1 mm, range:4-20 mm). The diagnostic accuracy of PEM alone, PET/CT alone, and combined PET/CT and PEM was evaluated by two nuclear medicine physicians based on visual inspection. The two groups (i.e., tumors of ≤ 10 mm and > 10-20 mm) werecompared in terms of the diagnostic capability of the three modalities (PEM alone, PET/CT alone, and PET/CT+PEM).Results: The sensitivities of PEM alone, PET/CT alone, and combined PET/CT and PEM were 72%, 60%, and 76%, respectively. The specificities of these tests were 98%, 100%, and 98%, respectively. Furthermore, the accuracies of these diagnostic modalities were 85%, 79%, and 87%, respectively. The combined PET/CT and PEM showed significantly higher sensitivity and accuracy than PET/CT alone (P=0.005 and P=0.02, respectively). In addition, PEM demonstrated asignificantly higher sensitivity than PET/CT in the ≤ 10 mm group (P=0.03); however, no difference was observed between the two modalities in the > 10 mm group in terms of sensitivity.Conclusion: 18F-fluorodeoxyglucose PET had limited capability for the detection of small breast cancers of < 10 mm. Combined PET/CT and PEM showed higher sensitivity and accuracy, compared to PET/CT alone.

Pulmonary tumor thrombotic microangiopathy (PTTM) is a rare but fatal cancer-related disease. Owing to its non-specific findings, aggressive course, and lack of established treatment guidelines, only a few cases of antemortem diagnosis in long-term survivors have been reported. We aimed to report a case of uterine cervical cancer induced PTTM that was suspected based on pulmonary hypertension and successfully treated using combination chemotherapy despite of delayed diagnose. It is important to be aware that PTTM should be suspected when respiratory failure occurs in patients with unexplained pulmonary hypertension. Multidisciplinary treatments including molecular targeted therapies might be effective treatment options.

Emotion recognition is known to change with age, but associations between the change and brain atrophy are not well understood. In the current study atrophied brain regions associated with emotion recognition were investigated in elderly and younger participants. Group comparison showed no difference in emotion recognition score, while the score was associated with years of education, not age. We measured the gray matter volume of 18 regions of interest including the bilateral precuneus, supramarginal gyrus, orbital gyrus, straight gyrus, superior temporal sulcus, inferior frontal gyrus, insular cortex, amygdala, and hippocampus, which have been associated with social function and emotion recognition. Brain reductions were observed in elderly group except left inferior frontal gyrus, left straight gyrus, right orbital gyrus, right inferior frontal gyrus, and right supramarginal gyrus. Path analysis was performed using the following variables: age, years of education, emotion recognition score, and the 5 regions that were not different between the groups. The analysis revealed that years of education were associated with volumes of the right orbital gyrus, right inferior frontal gyrus, and right supramarginal gyrus. Furthermore, the right supramarginal gyrus volume was associated with the emotion recognition score. These results suggest that the amount of education received contributes to maintain the right supramarginal gyrus volume, and indirectly affects emotion recognition ability.

Temperature-dependent electroluminescence measurements are performed for 265-nm AlGaN-based deep-ultraviolet (DUV) light-emitting diodes (LEDs) grown on AlN substrates. The external quantum efficiency (EQE) increases as the temperature decreases from 293 K to 6 K. Using two assumptions, the internal quantum efficiency (IQE) and current injection efficiency (CIE) are unity at the peak EQE at 6 K and the light extraction efficiency is independent of current and temperature, the current and temperature dependences of the product (IQE × CIE) are derived. The temperature dependence of the EQE cannot be simply explained by the Auger recombination processes. This observation enables the CIE and IQE to be separately extracted by rate equation analysis. The room-temperature EQE of the AlGaN-based DUV LEDs is limited by the CIE and not the IQE. We propose that the relatively low CIE may originate from the nonradiative recombination process outside quantum-well layers.

This manuscript summarizes recent results on the development of composite luminescent materials based on the single-crystalline films and single crystals of simple and mixed garnet compounds obtained by the liquid-phase epitaxy growth method. Such composite materials can be applied as scintillating and thermoluminescent (TL) detectors for radiation monitoring of mixed ionization fluxes, as well as scintillation screens in the microimaging techniques. The film and crystal parts of composite detectors were fabricated from efficient scintillation/TL materials based on Ce3+-, Pr3+-, and Sc3+-doped Lu3Al5O12 garnets, as well as Ce3+-doped Gd3−xAxAl5−yGayO12 mixed garnets, where A = Lu or Tb; x = 0–1; y = 2–3 with significantly different scintillation decay or positions of the main peaks in their TL glow curves. This work also summarizes the results of optical study of films, crystals, and epitaxial structures of these garnet compounds using absorption, cathodoluminescence, and photoluminescence. The scintillation and TL properties of the developed materials under α- and β-particles and γ-quanta excitations were studied as well. The most efficient variants of the composite scintillation and TL detectors for monitoring of composition of mixed beams of ionizing radiation were selected based on the results of this complex study.

The aim of this study was to investigate the relationship between olfactory recognition and morphological changes in olfactory brain regions including the amygdala, hippocampus, rectus, parahippocampus, orbitofrontal cortex, and medial frontal cortex in 27 elderly subjects and 27 younger healthy controls. The specific aim of the study was to determine which brain areas are associated with the initial decline of olfaction in elderly subjects, which occurs before the onset of dementia. All subjects underwent magnetic resonance imaging to measure anatomical brain volume and cortical thickness, and subjects were assessed using tests of olfactory acuity and cognitive function measured with the Montreal Cognitive Assessment. Overall brain volume reductions were observed in elderly subjects compared with young healthy controls, but only reduction in the volume of the left hippocampus was associated with decreased olfactory ability. The parahippocampus of elderly subjects was not different from that of controls; the extent of the reduction of parahippocampus volume varied among individuals, and reduction in this region was associated with olfactory decline. Similarly, parahippocampus thinning was associated with decreased olfactory function. The path analysis showed direct and indirect effects of hippocampus and parahippocampus volume on olfactory ability and that volume reductions in these areas were not associated with cognitive function. Parahippocampus volume reduction and thinning exhibited individual variation; this may be the first appearance of pathological changes and may lead to dysfunction in the connection of olfactory memory to the neocortex. Parahippocampus change may reflect the first sign of olfactory impairment prior to pathological changes in the hippocampus, amygdala and orbitofrontal cortex.

A growing number of brain imaging studies show functional connectivity (FC) between regions during emotional and cognitive tasks in humans. However, emotions are accompanied by changes in physiological parameters such as heart rate and respiration. These changes may affect blood oxygen level-dependent signals, as well as connectivity between brain areas. This study aimed to clarify the effects of physiological noise on the connectivity between areas related to the default mode network using resting-state functional magnetic resonance imaging (rs-fMRI). Healthy adult volunteers (age range: 19–51 years, mean age: 26.9 ± 9.1 years, 8 males and 8 females) underwent rs-fMRI for 10 min using a clinical 3T scanner (MAGNETOM Trio A Tim System, Siemens) with simultaneously recorded respiration and cardiac output. Physiological noise signals were subsequently removed from the acquired fMRI data using the DRIFTER toolbox. Image processing and analysis of the FC between areas related to the default mode network were performed using DPARSF. Network-Based Statistic (NBS) analysis of the functional connectome of the DMN and DMN-related area was used to perform three groups of comparison: without physiological noise correction, with cardiac noise correction, and with cardiac and respiratory noise correction. NBS analysis identified 36 networks with significant differences in three conditions in FC matrices. Post hoc comparison showed no differences between the three conditions, indicating that all three had the same networks. Among the 36 networks, strength of FC of 8 networks was modified under physiological noise correction. Connectivity between left and right anterior medial frontal regions increased strength of connectivity. These areas are located on the medial cerebral hemisphere, close to the sagittal sinus and arteries in the cerebral hemispheres, suggesting that medial frontal areas may be sensitive to cardiac rhythm close to arteries. The other networks observed temporal regions and showed a decrease in their connectivity strength by removing physiological noise, indicating that physiological noise, especially respiration, may be sensitive to BOLD signal in the temporal regions during resting state. Temporal lobe was highly correlated with anxiety-related respiration changes (Masaoka and Homma, 2000), speech processing, and respiratory sensation. These factors may affect the rs-fMRI signaling sensitivity.

Abstract Introduction Exercise therapies during rehabilitation significantly promote recovery from various deficits after cerebral infarction, which is mediated by neuronal plasticity with distinct inputs. Although adult neurogenesis can also be modulated by neuronal activity before synaptogenesis, how distinct exercises contribute to the neurological reorganization of the injured cerebral cortex remains unclear. In the present study, we aimed to elucidate the effects of different exercise therapies on motor recovery and neuronal reorganization after photochemically induced focal cerebral infarction. Methods Here, we examined the effects of three different exercises—(a) forced lower‐intensity and (b) higher‐intensity treadmill exercises, and (c) voluntary exercise with wheel running—on motor recovery and adult neurogenesis in a rat model of focal cerebral infarction. Photochemically induced thrombosis (PIT) was used to generate focal infarction in rats that was mostly confined to their motor cortices. Results Beam walking tests showed that recovery after PIT‐induced cortical infarction differed in acute and chronic stages and was influenced by the type of exercise. Furthermore, forced low‐intensity training had more positive effects on functional recovery than other exercises or control. To evaluate the production of newly generated cells including de novo neurogenesis, we performed lineage analysis with BrdU labeling and immunofluorescence experiments. Lower‐intensity treadmill exercise increased the number of BrdU/NeuN colabeled cells, but not total BrdU‐retaining or BrdU/Sox2‐colabeled cells, in the peri‐infarct region of the ipsilateral cortex. In contrast, high‐intensity treadmill or voluntary exercises had the opposite effects. Conclusions These results suggest that neuronal maturation can be differently modulated by distinct exercises and that low‐intensity treadmill exercise could result in more potent generation of mature neurons. This also suggests the possibility that the generation of neural stem/progenitor cells and differentiation might be modulated by rehabilitation‐mediated neural plasticity.

Abstract High resolution imaging of radiation is required for such radioisotope distribution measurements as alpha particle detection in nuclear facilities or high energy physics experiments. For this purpose, we developed an ultrahigh resolution radiation imaging system using an optical fiber structure scintillator plate. We used a ~1-μm diameter fiber structured GdAlO3:Ce (GAP) /α-Al2O3 scintillator plate to reduce the light spread. The fiber structured scintillator plate was optically coupled to a tapered optical fiber plate to magnify the image and combined with a lens-based high sensitivity CCD camera. We observed the images of alpha particles with a spatial resolution of ~25 μm. For the beta particles, the images had various shapes, and the trajectories of the electrons were clearly observed in the images. For the gamma photons, the images also had various shapes, and the trajectories of the secondary electrons were observed in some of the images. These results show that combining an optical fiber structure scintillator plate with a tapered optical fiber plate and a high sensitivity CCD camera achieved ultrahigh resolution and is a promising method to observe the images of the interactions of radiation in a scintillator.

We fabricated nanosized KMgF3 single crystals via a dry pulsed laser ablation process using femtosecond laser pulses. The sizes, shapes, and crystallographic properties of the crystals were evaluated by transmission electron microscopy (TEM). Almost all of the particles were spherical with diameters of less than 100 nm, and they were not highly agglomerated. Selected-area electron diffraction and high-resolution TEM analyses showed that the particles were single crystals. Particle diameter was controlled within a wide range by adjusting the Ar ambient gas pressure. Under low gas pressures (1 and 10 Pa), relatively small particles (primarily 10 nm or less) were observed with a high number density. With increasing pressure, the mean diameter increased and the number density drastically decreased. Vacuum-ultraviolet cathodoluminescence was observed at 140–230 nm with blue shift and broadening of spectrum.

A novel single-crystal growth method was developed, using arc plasma and metal melt, for a quick survey of high melting point materials. Single crystals of Yb-doped Lu2O3, Lu0.388Hf0.612O1.806, and Lu0.18Hf0.82O1.91, with melting points of 2460, 2900, and 2840 °C, respectively, were grown by an indirect heating method using arc plasma. We refer to this indirect heating growth method as the core heating (CH) method. The CH-grown Yb1%-doped Lu2O3 sample showed a full width at half maximum of 286 arcsec in the X-ray rocking curve. This value is better than the 393 arcsec obtained for the crystal grown by the micro-pulling-down (μ-PD) method. The Yb charge transfer state (CTS) emission was observed at 350 nm in the Yb1%-doped Lu2O3 and Lu0.18Hf0.82O1.91. In the case of the μ-PD method, using a rhenium (Re) crucible, absorption due to Re contamination and a resulting reduction in the Yb CTS emission were confirmed. However, contamination did not influence the properties observed in the crystals grown by the CH method.

Ce-doped LaBr3/AEBr2 (AE = Mg, Ca, Sr, Ba) eutectics were grown using the Bridgman–Stockbarger (BS) method in quartz ampoules. The eutectics (AE = Mg and Ca) showed optical transparency like optical fiber bundles. A grown Ce-doped LaBr3/MgBr2 eutectic shows a 355 nm emission ascribed to Ce3+ 4f-5d transition under X-ray excitation. The smaller the ionic size of AE, the higher the light yield of the sample was. The light yield of Ce:LaBr3/MgBr2 was 34,300 photon/MeV, which is higher than Ce:LYSO standard. Scintillation decay time under 662 keV gamma-ray excitation was 18.8 ns.

The microstructure and mechanical properties of platinum (Pt) fibers fabricated by unidirectional solidification using the alloy-micro-pulling-down (A-μ-PD) method were investigated using a Universal Testing Machine and Electron Backscattered Diffraction (EBSD). The Pt fiber fabricated at a growth rate of 10 mm/min was composed of relatively large grains with crystal orientation along the growth direction. The crystal orientation was consistent with the easy axis of the crystal growth on the face-centered-cubic (f.c.c.) structure. On the other hand, the adjacent grains of the Pt fiber fabricated at 50 mm/min were randomly oriented owing to a faster growth rate. In tensile tests, the Pt fibers fabricated by the A-μ-PD method indicated extremely different stress−strain curves compared to the commercial Pt wire. The maximum tensile stress of the Pt fiber reached ~100 MPa, and the Pt fiber ruptured after 58% nominal strain.

Co-Cr-Mo alloy fibers of 2 mm in diameter were fabricated from the melt at 1, 2, and 5 mm/min growth rates by unidirectional solidification using an alloy-micro-pulling-down (A-µ-PD) method to control the microstructure. All elements, Co, Cr, and Mo, were distributed in stripes elongated along the growth direction due to constitutional undercooling. Both Co-Cr-Mo fibers fabricated at 2 and 5 mm/min growth rates were composed of the γ phase with a face-centered cubic structure (fcc-γ phase) and ε-phase with a hexagonal close-packed structure (hcp-ε phase), and the ratio of the fcc-γ phase in the fiber fabricated at 5 mm/min growth rate was higher than that in the fiber fabricated at 2 mm/min. The results suggest that a faster growth rate increases the ratio of the fcc-γ phase in the Co-Cr-Mo fiber fabricated by unidirectional solidification.

Autobiographical odor memory (AM-odor) accompanied by a sense of realism of a specific memory elicits strong emotions. AM-odor differs from memory triggered by other sensory modalities, possibly because olfaction involves a unique sensory process. Here, we examined the orbitofrontal cortex (OFC), using functional magnetic resonance imaging (fMRI) to determine which OFC subregions are related to AM-odor. Both AM-odor and a control odor successively increased subjective ratings of comfortableness and pleasantness. Importantly, AM-odor also increased arousal levels and the vividness of memories, and was associated with a deep and slow breathing pattern. fMRI analysis indicated robust activation in the left posterior OFC (L-POFC). Connectivity between the POFC and whole brain regions was estimated using psychophysiological interaction analysis (PPI). We detected several trends in connectivity between L-POFC and bilateral precuneus, bilateral rostral dorsal anterior cingulate cortex (rdACC), and left parahippocampus, which will be useful for targeting our hypotheses for future investigations. The slow breathing observed in AM-odor was correlated with rdACC activation. Odor associated with emotionally significant autobiographical memories was accompanied by slow and deep breathing, possibly involving rdACC processing.

Colquiriite-type Eu-doped LiCaAlF6 and LiSrAlF6 (Eu:LiCAF and Eu:LiSAF) scintillator single crystals were grown using Al metal as a starting material to improve the Eu dopant segregation. All Eu:LiCAF and Eu:LiSAF single crystals using Al metal (Eu,AlM:LiCAF and Eu,AlM:LiSAF) indicated high transparency and effective segregation coefficients (keff) of Eu2%,AlM1%:LiCAF and Eu2%,AlM1%:LiSAF single crystals were significantly improved to the value of 0.40 and 0.99, respectively. EuF3 starting material reacted with Al metal before colquiriite-type phase was generated. As a result, Eu3+ ion in EuF3 was changed to Eu2+ ion which could be incorporated into Ca2+ and Sr2+ sites easily.

Transmission electron microscopy (TEM), specific heat, and thermal conductivity measurements were carried out for PrRh4.8B2 single crystals. PrRh4.8B2 takes an interesting layered crystal structure, where PrRh3B2 blocks are separated by metal Rh honeycomb layers. The existence of the Rh layers enhances the ferrimagnetic transition temperature. From area-selective picosecond thermoreflectance, the cross-plane thermal conductivity of PrRh4.8B2 is estimated to be 1.39 Wm−1 K−1, much lower than other layered borides like AlB2. Judging from the fine crystal structures obtained by TEM, phonon conduction is considered to be depressed due to random dispersion of Rh vacancy sites and two-dimensional nature of PrRh4.8B2.

Ca3Nb(Ga1−xAlx)3Si2O14 (CNGAS) single crystals with various Al concentrations were grown by a micro-pulling-down (µ-PD) method and their crystal structures, chemical compositions, crystallinities were investigated. CNGAS crystals with x = 0.2, 0.4 and 0.6 indicated a single phase of langasite-type structure without any secondary phases. In contrast, the crystals with x = 0.8 and 1 included some secondary phases in addition to the langasite-type phase. Lattice parameters, a- and c-axes lengths, of the langasite-type phase systematically decreased with an increase of Al concentration. The results of chemical composition analysis revealed that the actual Al concentrations in as-grown crystals were almost consistent with the nominal compositions. In addition, there was no large segregation of each cation along the growth direction.

Previously, studying the development, especially of corticospinal neurons, it was concluded that the main compensatory mechanism after unilateral brain injury in rat at the neonatal stage was due in part to non-lesioned ipsilateral corticospinal neurons that escaped selection by axonal elimination or neuronal apoptosis. However, previous results suggesting compensatory mechanism in neonate brain were not correlated with high functional recovery. Therefore, what is the difference among neonate and adult in the context of functional recovery and potential mechanism(s) therein? Here, we utilized a brain unilateral decortication mouse model and compared motor functional recovery mechanism post-neonatal brain hemisuction (NBH) with adult brain hemisuction (ABH). Three analyses were performed: (1) Quantitative behavioral analysis of forelimb movements using ladder walking test; (2) neuroanatomical retrograde tracing analysis of unlesioned side corticospinal neurons; and (3) differential global gene expressions profiling in unlesioned-side neocortex (rostral from bregma) in NBH and ABH on a 8 × 60 K mouse whole genome Agilent DNA chip. Behavioral data confirmed higher recovery ability in NBH over ABH is related to non-lesional frontal neocortex including rostral caudal forelimb area. A first inventory of differentially expressed genes genome-wide in the NBH and ABH mouse model is provided as a resource for the scientific community.

Interest in applications of stabilized zirconia (PSZ) as dental material has recently increased. In a corrosive environment such as a living body, PSZ is subjected to a cyclic load and needs higher fatigue strength. In our previous paper, appreciable degradation occurred in the fatigue lifetime of yttria-doped tetragonal zirconia polycrystals (TZP). In this study, PSZ specimens were annealed for 100 h at temperatures of 200, 400 and 600°C in a low vacuum atmosphere to improve the fatigue resistance. Both static and cyclic fatigue tests were conducted on the annealed specimens under static four-point bending and under cyclic reversed plane bending, respectively. As a comparison, specimens without annealing were tested in the same way. The test results indicated that the lifetime appreciably increases in cycling fatigue under a low applied load in specimens annealed at the low temperatures of 200 and 400°C from that of specimens without annealing. It was supposed that the occurrence of fine microcracks accompanied by the phase transformation from tetragonal to monoclinic caused enlargement of the process zone in the wake field of a crack in cyclic loading and contributed to the increase of crack resistance.

Severe fever with thrombocytopenia syndrome (SFTS) is a tick-borne infectious disease with a high case fatality rate, and is caused by the SFTS virus (SFTSV). SFTS is endemic to China, South Korea, and Japan. The viral RNA level in sera of patients with SFTS is known to be strongly associated with outcomes. Virological SFTS diagnosis with high sensitivity and specificity are required in disease endemic areas.We generated novel monoclonal antibodies (MAbs) against the SFTSV nucleocapsid (N) protein and developed a sandwich antigen (Ag)-capture enzyme-linked immunosorbent assay (ELISA) for the detection of N protein of SFTSV using MAb and polyclonal antibody as capture and detection antibodies, respectively. The Ag-capture system was capable of detecting at least 350-1220 TCID50/100 μl/well from the culture supernatants of various SFTSV strains. The efficacy of the Ag-capture ELISA in SFTS diagnosis was evaluated using serum samples collected from patients suspected of having SFTS in Japan. SUMMARY Brain ischemia, also termed cerebral ischemia, is a condition in which there is insufficient blood flow to the brain to meet metabolic demand, leading to tissue death (cerebral infarction) due to poor oxygen supply (cerebral hypoxia). Our group is interested in the protective effects of neuropeptides for alleviating brain ischemia, as well as the underlying mechanisms of their action. The present study was initiated to investigate molecular responses at the level of gene expression in ischemic brain tissue. To achieve this, we used a mouse permanent middle cerebral artery occlusion (PMCAO) model in combination with high-throughput DNA microarray analysis on an Agilent microarray platform. Briefly, the right (ipsilateral) and left (contralateral) hemispheres of PMCAO model mice were dissected at two time points, 6 and 24 hours post-ischemia. Total RNA from the ischemic (ipsilateral) hemisphere was subjected to DNA microarray analysis on a mouse whole genome 4x44K DNA chip using a dye-swap approach. Functional categorization using the gene ontology (GO, MGD/AMIGO) of numerous changed genes revealed expression pattern changes in the major categories of cellular process, biological regulation, regulation of biological process, metabolic process and response to stimulus. Reverse-transcriptase PCR (RT-PCR) analysis on randomly selected highly up- or downregulated genes validated, in general, the microarray data. Using two time points for this analysis, major and minor trends in gene expression and/or functions were observed in relation to early- and late-response genes and differentially regulated genes that were further classified into specific pathways or disease states. We also examined the expression of these genes in the contralateral hemisphere, which suggested the presence of bilateral effects and/or differential regulation. This study provides the first ischemia-related transcriptome analysis of the mouse brain, laying a strong foundation for studies designed to elucidate the mechanisms regulating ischemia and to explore the neuroprotective effects of agents such as target neuropeptides.
