【目的】调查非肝病人群中经输血传播病毒(TTV)的感染状况。【方法】在TTV ORF1 保守区设计引物, 用巢式 PCR 检测125 例医院工作人员、105 例献血员、46 例静脉透析病人、166 例性病高危人群和83 例静脉药瘾者, 共525 人血清中 TTV DNA。【结果】TTV DNA 阳性率, 医院工作人员3.2%、献血员11.4%、静脉透析病人13.0%、性病高危人群20.5 %和 静脉药瘾者39.8 %。【结论】静脉药瘾者、性病高危人群和静脉透析病人是TTV 感染的高危人群。

【目的】构建反义c-myb 重组逆转录病毒载体并建立其包装细胞株。【方法】采用RT-PCR 方法, 获取目的基因, 通过TA 克隆方法克隆入pUC19, 再反向亚克隆入逆转录病毒载体pDOR 中。采用DOTAP 法将重组逆转录病毒载体转入包 装细胞, 以NIH3T3 细胞为靶细胞测定产病毒滴度。【结果】序列测定结果与GenBank 中序列一致。c-myb 基因片段已定向 克隆入pDOR。细胞转染表明, 已形成稳定的包装细胞株PA317/ pDOR-my b, 产病毒滴度达5.2 ×104 ~ 9.5 ×104 CFU/m L。 【结论】成功构建了含有反义c-my b 的重组逆转录病毒质粒, 并建立其包装细胞株。

5. 等离子体预处理对聚氨酯 / 涤纶复合织物贴合牢度的影响.

- Abstract
The textile industry is a high energy consumption and high pollution industry, with an annual emission of over 8 billion tons. China has introduced a pollution permit system to use legal and other means to control pollution sources and improve the surrounding ecological environment. In the context of China's promotion of green, energy-saving, and environmentally-friendly measures, the current adhesives used in fabric processing in composite factories are mainly solvent based hot melt adhesives. During the operation, organic solvents are prone to volatilization, causing environmental pollution and human injury due to strong odors. And the water-based polyurethane hot melt adhesive has advantages such as good elasticity, high adhesion, and low VOC content, making it a new type of adhesive that adapts to environmental trends. Composite fabrics are mostly used to make warm winter clothing, with a soft and smooth feel. The softener is mostly water repellent amino silicone oil, which conforms to the principle of compatibility with the solvent based hot melt adhesive. The adhesion fastness can meet national standards. On the contrary, the water-based polyurethane hot melt adhesive has a low solubility in water-repellent amino silicone oil, limited diffusion, and a significant reduction in the amount of adhesive absorbed into the fabric, resulting in poor adhesion fastness. The bonding surface of the fabric was treated with plasma, and after etching, numerous concave holes were formed on the surface of the fabric, increasing the specific surface area and friction coefficient of the fabric. The resulting porosity resulted in an excellent bonding effect of the composite fabric; the introduction of hydrophilic groups such as carboxyl groups, ether bonds, and amide bonds increased the moisture absorption of the fabric, and the diffusion of waterborne polyurethane hot melt adhesive to the fabric was also correspondingly improved; the polyester fabric treated with plasma was prone to reacting with water-based polyurethane to form carbamate bonds, urea bonds, and ion bonds, forming strong cohesion and bonding force, thereby increasing the peel strength of the fabric composite. The bonding surface of the fabric was treated with plasma, and the peeling strength of the composite fabric was tested under different process parameters. The optimized plasma process was obtained as follows: a discharge voltage of 25 kV, an action time of 5.5 minutes, and an action distance of 2 mm. The results show that after plasma treatment, the friction factor, specific surface area, roughness, wettability, and capillary effect of the fabric increase, making it easier for the water-based polyurethane hot melt adhesive to adhere and accumulate on the surface of the fabric during transfer, increasing the peel strength between composite fabrics. According to the optimized process, the peel strength of the composite fabric in the warp direction was increased from 2.3 N to 18.3 N, and the peel strength in the weft direction was increased from 1.2 N to 12.6 N. After 30 washes, the composite fabric did not show any peeling or bulging, and the peeling strength still met national standards. Due to plasma etching, the breaking strength of the fabric in both warp and weft directions decreased. The breaking strength decreased slowly before the action time of 5-6 minutes, and decreased rapidly after 6 minutes. It is expected that in the future, practitioners can increase their research and development efforts on water-based polyurethane, develop water-based hot melt adhesives with stronger adhesion, simplify the fabric composite process, shorten reaction time, and achieve instant composite like solvent based hot melt adhesives, so as to improve the production efficiency. - Abstract
Real-time monitoring of glucose metabolism can not only provide information on cell growth status, but also contribute to the deep understanding of the mechanism of cell metabolism. Electrochemical sensors have become an important tool in real-time detection of glucose. However, most reported glucose sensors are based on two-dimensional electrodes that are hardly compatible with three-dimensional cell culture systems for accurate detection. Herein, a 3D glucose electrochemical sensor with high sensitivity and selectivity is prepared via immobilizing glucose oxidase (GOD) on the surface of a porous electrode which could catalyze H2O2 reduction. Then, collagen hydrogel with tumor cells (MCF-7) therein is filled into the pores of the 3D sensor, and thus an integrated platform of 3D cell culture for electrochemical detection is constructed. This platform allows real-time monitoring of the change in glucose concentration in culture medium when MCF-7 cells are cultured under different conditions, providing a new method for evaluating the cell metabolism activities. - Abstract
The experiment was conducted using 0.81 g Litopenaeus vannamei post-larvae as the subjects to study carbohydrate to lipid ratio in the feed on the growth, physiological and biochemhe effects of differentical indices, and body composition of Litopenaeus vannamei under a salinity of 30 mg/kg. Six isonitrogenous and isoenergetic feeds were prepared, with a protein level of 41% and carbohydrate-lipid ratios of 4.01∶1, 3.29∶1, 2.78∶1, 2.21∶1, 1.75∶1, and 1.42∶1, respectively. Each group had 3 replicates, with 30 shrimps in each replicate. The results showed that: the survival rate of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 3.29∶1 was significantly higher than those fed with ratios of 2.21∶ 1 and 1.42∶ 1 (P<0.05). The specific growth rate of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 2.78∶ 1 was significantly higher than those fed with ratios of 4.01∶1 and 3.29∶ 1 (P<0.05). The amylase activity of Litopenaeus vannamei fed with a carbohydratelipid ratio of 4.01∶ 1 was significantly higher than those fed with ratios of 1.75∶1 and 1.42∶1 (P<0.05). The hexokinase activity of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 4.01∶1 was significantly higher than the other groups. The liver glycogen content of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 3.29∶1 was significantly higher than those fed with a ratio of 1.42∶1 (P<0.05). The blood protein concentration of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 1.75∶1 was significantly higher than those fed with ratios of 1.42∶ 1 (P<0.05). The blood glucose content of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 4.01∶1 was significantly higher than those fed with a ratio of 1.42∶1 (P< 0.05). The cholesterol content of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 1.42∶1 was significantly higher than those fed with ratios of 4.01∶1, 3.29∶1, 2.78∶1, and 2.21∶1 (P<0.05). The triacylglycerol content of Litopenaeus vannamei fed with a carbohydrate-lipid ratio of 1.42∶1 was significantly higher than those fed with a ratio of 4.01∶1 and 2.21∶1 (P<0.05). Moreover, the crude fat content of Litopenaeus vannamei fed with carbohydrate to lipid ratio of 1.42∶1 was significantly higher than those fed with ratios of 4.01∶1, 3.29∶1, and 2.78∶1 (P<0.05). The results of the experiment showed that the optimal starch and fat requirements for Litopenaeus vannamei were 18.97% and 6.83%, respectively, and the optimal carbohydrate to lipid ratio was 2.78∶1. The aim of this study is to establish the IPEC-J2 cell line with MDH2 gene knockout by CRISPR/Cas9 genome editing technology and investigate whether MDH2 gene knockout confers resistance to deoxynivalenol. sgRNA sequences targeting MDH2 gene were designed and cloned into PX459 vector; the recombinant plasmid PX459-sgRNA-MDH2 was introduced into the IPEC-J2 cells by electroporation,and puromycin was added to screen KO cell colony generations. The MDH2 KO single cell colony generations were isolated by limited dilution method and validated by genotype sequencing, quantitative PCR and Western blotting to obtain MDH2-KO monoclonal cell colony generation. Finally, the cell viability was detected by CCK8 kit and apoptosis and necrosis assay kit to determine the resistance of MDH2 knockout cell colony generation to deoxynivalenol. The sequencing results showed that the MDH2 knockout vector was successfully constructed. The obtained cell colony generations were validated as MDH2 knockout monoclonal cell colony generations by RT-qPCR and immunoblotting. The CCK8 cell viability assay showed that knockout of MDH2 increased the viability of IPEC-J2 cells by 18.67%, 19.59%, 26.36%, and 27.01% at different concentrations of deoxynivalenol(4, 2, 1, and 0.5 μg/mL)for 5 d compared with wild-type cells. Flow cytometry assays showed that knockout of MDH2 reduced the mortality rate of IPEC-J2 cells by 30.33%, 15.81%, 16.00%, and 14.70% at different concentrations of deoxynivalenol respectively compared with wild-type cells for 5 d. We obtained the MDH2-KO monoclonal cell colony generation by editing the MDH2 gene in IPEC-J2 cells using the CRISPR/Cas9 system. The resistance of MDH2-KO cell colony generation to deoxynivalenol was demonstrated by cell viability and cell death assays, which provids insight into the toxic mechanism of deoxynivalenol-induced cell death and offers a new strategy for the prevention and treatment of deoxynivalenol. The hydrogen peroxide added in this process is over three times more than that of the traditional process. After oxygen bleaching mercerization of the cotton cloth, local excessive oxidation occurs and oxygen bleaching holes are more likely to occur. In the production process of cotton cloth, the selection process of different cotton cloth specifications is different, resulting in complex reasons for holes.Therefore, it is necessary to find out the causes of holes in the production process and formulate corresponding preventive measures and technological processes. In this experiment, aldehyde cellulose produced by bleaching cotton cloth with hydrogen peroxide was oxidized by self-made weak oxidation reagent, so that metal silver atoms could be directly precipitated on the cloth surface. According to the number of silver atoms precipitated from the cloth surface and the hole opening, the color depth was formed, the intensity of hydrogen peroxide decomposition was judged, and whether the hole on cotton cloth was caused by excessive oxidation was proved from the side. Then, 1 mL of 8% silver nitrate solution, and 2 mL of mixed solution of 20% sodium bicarbonate and 20% caustic soda were added into 17 mL of deionized water. and shaken up quickly. Subsequently, 5g of cloth with holes was boiled in the solution for 2 min under100 ˚C, and then it was washed with deionized water. The color difference around the hole of that fabric was observed. The cotton cloth with holes were characterized by Fourier transform infrared spectroscopy, X-ray diffraction, scanning electron microscope (SEM) and X-ray photoelectron spectroscopy.The self-made test results show that the holes caused by mechanical and other external forces have the same color as the holes on the cloth surface, indicating that the silver content precipitated is basically the same As for holes caused by local overoxidation, the color of the opening is darker than the cloth surface, and the silver content of the hole area is obviously higher than that of other parts. During the test, it is found that the decomposition rate of hydrogen peroxide is affected by temperature, pH value and metal ions, and the symbionts and textile auxiliaries in gray cloth can inhibit the decomposition of hydrogen peroxide. Fourier transform infrared spectroscopy shows that the structure of most cotton fiber polysaccharides changes into dialdehyde cellulose after oxygen bleaching. After treatment with self-made hole breaking detection reagent, the aldehyde group and silver ions have oxidation-reduction reaction, and the aldehyde group disappears. Through characterizing the cotton cloth tested by self-made hole detection reagent, it is concluded that the black particulate matters on the cotton cloth surface are silver atoms. The hole areas are the same shade as the other cloth surface, such as burnt holes, hook holes, weak yarn strength holes and damp mildew holes. Nevertheless, compared with the main elements of the cloth surface, there are more black silver atoms in the hole than in other places, which makes the color darker than in other places. As the demand for oxygen bleached mercerized cotton cloth is gradually increasing in domestic market, it is significant to determine the causes of holes for production prevention, which will be beneficial to cost saving and energy conservation and emission reduction. For the cotton cloth with holes during the production process, the semi-finished cotton cloth can be treated through lower tension process on the machines, increase the friction between yarns by adding adhesives, or even be changed to the overflow or air cylinder with low tension for dyeing, so to reduce the probability of holes as much as possible and minimize the loss. In the future, the weaving mill will equip the magnetic metal cloth inspection machine in the gray cloth inspection. As long as the fabric has more than a certain amount of metal ions, the metal alarm device on the cloth inspection machine will raise the alarm to prevent the problematic gray cloth flowing into the next process, which guarantees the good quality. 【Objective】Continuous cropping obstacle is considered to have a close connection with the soil microbial community. The present study aimed to isolate and identify soil-borne pathogenic microorganisms in Ganoderma leucocontextum and provide a theoretical basis for continuous cropping obstacles encountered in cut-log cultivation.【Method】Overburden soil used to cultivate G. leucocontextum in the greenhouse for various periods(0, 1, 3, 5 years) were collected. Changes in culturable microbial community were investigated, and predominant microbes were isolated. Soil-borne pathogenic microorganisms of G. leucocontextum grown in continuously cropped soil were identified by plate confrontation culture. The species were further determined based on morphological and physiological features and molecular techniques. 【Result】 With the increase of G. leucocontextum continuous cropping years, the number of culturable microorganisms in the overburden soil and population changed, the number of bacteria and actinomyces declined significantly, while the fungi number increased. Specifically, bacteria accounted increased from 25.21% to 95.29%, fungi increased from 0.69% to 4.71%. By contrast, the proportion of actinomycetes decreased. Five dominant fungal species and nine dominant bacterial species were isolated and identified from continuously cropped soil. The antagonistic test showed that the inhibition rates on the growth of G. leucocontextum mycelium of six strains reached above 80%, namely, N7, N11, N9, N12, N13 and N14.【Conclusion】The continuous cultivation of G. leucocontextum significantly affected the soil microflora in the overburden soil, and the three main microbial populations had significant differences in different cultivation years. Bacillus, Talaromyces, Rhodococcus and some Streptomyces species might be potential soil-borne pathogenic microorganisms in the continuous cropping of G. leucocontextum, leading to obstacles encountered in cut-log cultivation. 【Objective】 The purpose of the experiment was to investigate the effect of the Banqi fermentation solution (Radix isatidis, Astragalus) on the intestinal flora and growth performance of weaned lambs. 【Method】 Bacillus subtilis, B. licheniformis, B. sphericalis, Bacillus DY, mixed strains[v (B. subtilis)∶v (B. licheniformis) = 1∶1]of fermented Banqi extract were used to feed lambs, the unfermented group was fed with Banqi extract, and the control group was fed with basal diet for 28 days. The growth indexes were counted and intestinal tissue sections were prepared, and the treatment group with good performance High-throughput sequencing was performed to analyze the effect of fermented Chinese medicine on intestinal flora, and the mechanism of action of fermented Chinese medicine was initially investigated. 【Result】The fermentation Ⅰ group (B. subtilis fermented Astragalus extract) significantly increased the average daily weight gain of lambs (P<0.05), which may be directly related to the increase of intestinal villi length. The results of intestinal flora sequencing showed that the fermentation I group significantly increased the abundance and diversity of duodenal flora (P< 0.05). The fermentation I group increased the relative abundance of Actinobacteria, Fimicutes and Butyrivibrio, while decreasing the relative abundance of Proteobacteria and Cupriavidus. Lefse (LDA>3) analysis showed that the relative abundance of Actinobacteria, Tenericutes, Mogibacterium, L7A-E11 (P<0.05); The relative abundance of Elusimicrobia was significantly increased in the unfermented group (P < 0.05); And Acinetobacter was significantly enriched in the control group (P< 0.05). 【Conclusion】B. subtilis fermentation of Banqi extract can improve the length of small intestinal villi and flora diversity of weaned lambs and thus increase the average daily weight gain of lambs, which provides theoretical support for the application of fermented Chinese medicine on weaned lambs. An intelligent electronic feeding system has been an ever-increasing pattern in the breeding industry. However, the rotation eating may disrupt the diurnal eating rhythm of some pigs, leading to the health and growth disorder of pigs. This study aims to explore the effects of eating time on the behavior distribution of pig production under the intelligent electronic feeding system. A systematic analysis was also conducted to determine the diurnal proportion of effective and ineffective eating frequency, feed intake, and eating time during the electronic sow feeding (ESF) and pig performance testing station (PPTS). The results revealed that there was the general night eating syndrome of pigs under the rotation eating in the intelligent electronic feeding system, about one-fourth of which ate only at night in the ESF system. The relatively fixed eating order indicated that the herd hierarchy was also an important reason for pig eating time disorder. The ineffective eating behavior of pigs occurred mainly in the daytime, which indirectly increased the likelihood of pigs eating at night in PPTS and ESF. Furthermore, it was probable to reduce the actual number of pigs per station for the less running time of the electronic feeding system and the occurrence of eating at night, according to the operation data from the different areas. More importantly, a further investigation was also made to clarify whether the night time feeding of some pigs under the ESF and PPTS feeding patterns caused the disturbance of sleep schedules and retardation of pig growth. Twenty-six miniature pigs were selected to compare the effects of different feeding time on behavioral rhythm and performance of growing pigs. Two groups were divided into daytime feeding and night time feeding. The former was designed to mimic the natural eating habits of the pigs, while the latter was for the inconsistent feeding time. It was found that the daytime feeding increased by 135.68 g/d (P<0.1) in the daily feed intake of growing pigs, but there was no significant difference in the daily gain between the two groups. The eating behavior of the growing pigs in the daytime feeding group was mostly eating, drinking, and scratching during the daytime, as well as lying at night time. By contrast, the eating behavior of growing pigs in the night time feeding group was mainly concentrated at the feeding time of 19:00 and the alternating time of daytime and night time at 7:00. The daytime feeding increased by 44.28, 14.22, and 61.95 min/d (P<0.05) in the duration of feeding, drinking, and lying, respectively, whereas, decreased by 48.75, 57.40, and 20.30 min/d (P<0.05) in the duration of standing, walking, and scratching, respectively. Therefore, the night time feeding disrupted the normal circadian eating, drinking, and lying behavior rhythm of some growing pigs, particularly for the occurrence of bad mood signals, such as standing and scratching. Consequently, the intelligent electronic feeding system destroyed part of the eating rhythm, leading to the growth difference in individual pigs. Some recommendations were also given to arrange the number of pigs in each feeding station for the optimal running time of the intelligent electronic feeding system during pig breeding. Thermodynamic properties in an alloy system play an important role in the materials science and engineering. Therefore, theoretical calculations via using thermodynamic model with the flexibility and reliability to deal with complexity are very useful and have scientific meaning . The modified M-MIVM was deduced from physical principles,and it can reflect the contributions from both the molecular configuration micro-state number (entropy) and molecular interactions ( enthalpy) towards excessive Gibbs free energy. The proposed equations are more consistent with the practical solid solutions. A significant advantage of this model lies in it can use either the relevant binary infinite dilution activity coefficients orbinary activity to describe and predict the thermodynamic properties of the binary or multi-component solid solutions. The thermody namic properties of solid solution alloys such as Au-Ni,Co-Ni, Cr-Ni, Cu-Ni, Fe-Ni, Ni-Pd, Ni-Pt, Ni-V, Co-Fe, Cr-Fe, Co-Fe-Ni, Cr-Fe-Ni and C-Co-Fe-Ni systems were predicted by using the proposed model. The results show that the activity values predicted by the modified M-MIVM are in good agreement with the experimental data except for Cr-Ni and Ni-V binary discontinuous solid solutions,and the total average relative error of the 13 systems is 4.75 %, and the prediction effect of the new model is better than that of MIVM, which can meet the requirement of engineering calculation. The modified M-MIV M has certain accuracy, stability, and applicability in the application of Ni-based solid alloy, and can provide more reliable thermodynamic data for Ni-based alloy design. To explore the predictive value and prevention strategy of nomogram model for incision infection after transumbilical single hole laparoscopic cholecystectomy. The clinical data of 94 patients with after transumbilical single hole laparoscopic cholecystectomy who were admitted to Hefei First People's Hospital from October 2018 to May 2021 were retrospectively analyzed. Patients were divided into infection group (n=12 cases) and non-infection group (n=82 cases) according to whether incision infection occurred after surgery. The medical records of the two groups were consulted, and univariate and multivariate Logistic regression analysis was performed on the possible influencing factors of postoperative incision infection. R software was introduced to establish the prediction model of incision infection after transumbilical single hole laparoscopic cholecystectomy and draw the line graph model to obtain the predicted risk value. The ROC curve was drawn and the predictive efficacy of nomogram model for postoperative incision infect ion was analyzed. Among 94 cases of transumbilical single hole laparoscopic cholecystectomy, 12 cases had postoperative incision infection, and the infection rate was 12.77%. Multivariate Logistic regression analysis showed that age, operation time, hospital stay and gallbladder rupture were the influencing factors of incision infection after transumbilical single hole laparoscopic cholecystectomy (P<0.05). The nomogram model shows that the age score was 63 scores, the hospital stay score was 37.6 scores, the operation time score was 71.5 scores, the gallbladder rupture score was 50 scores, and the predicted risk value was 2.221, the area under ROC curve is 0.832. The incision infection rate after transumbilical single port laparoscopic cholecystectomy is high, and they are affected by many factors. Therefore, the prediction model constructed based on the above factors can achieve high prediction efficiency, and which is worthy of popularization and application. [ABSTRACT FROM AUTHOR]
