Abstract

Real-time monitoring of glucose metabolism can not only provide information on cell growth status, but also contribute to the deep understanding of the mechanism of cell metabolism. Electrochemical sensors have become an important tool in real-time detection of glucose. However, most reported glucose sensors are based on two-dimensional electrodes that are hardly compatible with three-dimensional cell culture systems for accurate detection. Herein, a 3D glucose electrochemical sensor with high sensitivity and selectivity is prepared via immobilizing glucose oxidase (GOD) on the surface of a porous electrode which could catalyze H2O2 reduction. Then, collagen hydrogel with tumor cells (MCF-7) therein is filled into the pores of the 3D sensor, and thus an integrated platform of 3D cell culture for electrochemical detection is constructed. This platform allows real-time monitoring of the change in glucose concentration in culture medium when MCF-7 cells are cultured under different conditions, providing a new method for evaluating the cell metabolism activities.
